The natural presence of Beauveria bassiana was determined (Balsamo) Vuillemin from insects with mycotic symptoms collected in leaves and in the stalks of sugar cane (Saccharum sp. hybrid) in Pinar del Río, Ciudad de la Habana, Habana, Matanzas, Villa Clara, Cienfuegos and Camagüey. The results obtained demonstrate the natural presence of the entomopathogen fungus in larvae and chrysalises of Diatraea saccharalis (Fabricius) and they suggest the basis for the strategy of biological control against this borer.  相似文献

Leptocereus scopulophilus is newly described as endemic to the limestone summits and rocky hillsides of Somorrostro Hill, 25 km southeast of La Habana, Cuba. Since being discovered in 1923, it has been confused withLeptocereus leonii, from which it can be easily distinguished by its erect, ascending, branches which are greater in diameter and often 4–5 ribbed, by the more prominent and less crenate ribs bearing more widely spaced areoles and longer spines, and by its larger flowers with exserted stigma lobes and nearly tubular, green, spiny receptacles. The fruits also differ in being larger and in bearing clusters of persistent spines at maturity. Areces-Mallea, Alberto E. Elevated esterase Estbeta1 was purified from larvae of newly isolated strains of the mosquito Culex quinquefasciatus from Colombia (COL) and Trinidad (TRI) with resistance to organophosphate (OP) insecticides. Insecticide interactions were compared with those of elevated Estbeta1(2) from the OP-resistant Habana strain and the non-elevated Estbeta1(3) from the susceptible PelSS strain. On the basis of insecticide binding efficiency, all elevated Estbeta1 esterases were readily distinguishable. Differences between the EcoRI restriction fragment patterns of the amplified estbeta1 gene in COL and TRI strains compared with each other, and between amplified estbeta1(1), estbeta1(2) and the non-amplified estbeta1(3), suggest differences in their nucleotide sequence. Considering their variable insecticide binding efficiencies, these genetic differences would imply that, in contrast to estalpha2 and estbeta2, amplification of estbeta1 has occurred several times independently. Generally, the elevated Estbeta1s were more reactive with insecticides than the non-elevated Estbeta1(3). This supports the hypothesis that the elevated esterase-based mechanism confers resistance through amplification of alleles coding for esterases which have a greater specificity for the insecticides they sequester than the esterases coded by their non-amplified counterparts.  相似文献

The molecular basis of familial hypercholesterolemia (FH) in three families of Spanish descent from La Habana was investigated by the candidate gene approach. The Arg3500Gln mutation of apolipoprotein B-100 was not found. Identification of low density lipoprotein receptor (LDLR) gene haplotypes segregating with FH guided the characterisation of three point mutations by automated sequencing. One, a Val408Met missense mutation, a founder mutation in Afrikaner FH patients, was recurrent, being associated with a distinct DNA haplotype. The other two, Glu256Lys and Val776Met missense mutations, were novel and modified highly conserved residues. These mutations were absent in normolipidemic subjects and were associated in heterozygous carriers with twice the cholesterol levels observed in noncarriers. Noticeably, cardiovascular complications were rarely observed in older heterozygotes, even in those with the Afrikaner FH-2 mutation. These findings confirm the molecular heterogeneity of LDLR gene mutations causing FH and the variability of their expression across different populations.  相似文献

We aimed to estimate the prevalence of Candida albicans and Trichomonas vaginalis in immunocompetent pregnant women living in Havana City, Cuba, with or without symptoms of vaginitis, using a sample of 640 women from 6 Gyneco-obstetrics hospitals, which represents 2.5% of total yearly pregnant women. Diagnosis was made using a new latex agglutination kit (Newvagin C-Kure, La Habana, Cuba). Clinical sensitivity and specificity of this assay were validated against culture method, with 467 and 489 clinical specimens for Candida albicans and Trichomonas vaginalis, respectively. Results showed that the kit clinical sensitivity was 100% for Candida albicans and 86.7% for Trichomonas vaginalis compared with a clinical specificity of 93.3% for Candida albicans and 95.1% for Trichomonas vaginalis by culture. The prevalence of candidiasis was determined to be 42.3% (95% confidence interval [CI] 3.8%); the prevalence of trichomoniasis was 9.84% (95% CI 2.3%). In our sample, 48.7% of the women tested negative with respect to both candidiasis and trichomoniasis. Only 6.41% of the cases yielded inconclusive results. The test has high sensitivity, and our results indicate a relatively high prevalence of both infections. However, a significant difference (P < .001) was also observed in candidiasis and trichomoniasis prevalence among hospitals corresponding to the quantity of women with clinical vaginitis. No difference was observed between diabetics and nondiabetics, probably due to the special care of diabetic pregnant women. We conclude that the method is useful for this kind of vaginitis prevalence study and that candidiasis and trichomoniasis prevalences in pregnant women of Havana are 38.5% to 46.2 % (95% CI) and 7.5% to 12.1% (95% CI), respectively.  相似文献

Esterase amplification is the major organophosphorus (OP) insecticide resistance mechanism in Culex mosquitoes. The amplified Estα2 1\ Estβ2 1 esterases are found in > 90% of resistant populations worldwide, whereas amplified DNAs (amplicons) containing Estβ1s are much rarer. Individuals with the Estβ1 amplicons appear to be at a selective disadvantage in competition with those carrying the Estα2 1\ Estβ2 1 amplicons. To test the hypothesis that this is because Estβ1 is less able to bind insecticide than the common amplified esterases, Estβ12 was purified from the multi-resistant Habana strain of Culex quinquefasciatus , from Cuba. In its native form Estβ1 is a monomeric enzyme of 66 kDa, with a pI of 4.8. The bimolecular rate constants for interaction of Estβ12 with several OP insecticides were similar to those for the commonly elevated esterases Estα21 and Estβ21, and much higher than for the electrophoretically identical non-elevated Estβ13 and Estα3. Hence the apparent selective advantage of the Estα2 1\ Estβ2 1 amplicon is not due to its greater efficiency of insecticide binding, as OP insecticides are significantly better inhibitors of all the amplified esterases than of their non-amplified counterparts and therefore should be equally effective at conferring resistance.  相似文献

Four-locus recombination frequencies are summarized into two-locus pair frequencies and three-locus frequencies, and further, into two-locus frequencies such that higher-order frequencies are linear functions of lower-order frequencies. Frequencies of gene combinations are defined according to their position on the same or distinct gametes, and linear functions of these provide the measures of linkage disequilibria. These concepts are utilized to derive the transitional behavior of the gene combinations frequencies and the linkage disequilibria in a finite monoecious population with random union of gametes for up to four loci. The transitions of lower-order disequilibria in a higher-order (more loci) setting involve the higher-order disequilibria which must be taken into account in arriving at the final (fixation) frequencies. The methods allow different initial conditions. Since corresponding data functions of the gene combination frequencies provide unbiased estimates of the parameters, estimators follow naturally.  相似文献

The results of study of micronuclei (MN) frequencies among the participants of Ukrainian school biological olympiads are presented. Totally 266 persons have been inspected. The distribution of MN frequencies correspond to the Poisson's distribution with lambda = 2.5. The average frequency of micronuclei in males was 2.4 +/- 0.15%, in females it was 2.7 +/- 0.14%. The difference of the average MN frequencies for these two groups was statistically insignificant. The individual micronuclei frequencies varied from 0 to 8.3%, the average MN frequency in the general group was 2.5 +/- 0.11%, (limits 2-5%). The micronuclei frequencies in different age groups of males and females were compared. Significantly higher MN frequencies in females than in males at the age of sixteen were detected. The age-related changes of micronuclei frequencies (14-18 age) were different for females compared to males.  相似文献

为研究中国南方汉族人群核苷酸修复基因hMTH1遗传多态性,应用聚合酶链反应-单链构象多态性技术检测172名健康人外周血白细胞hMTH1基因启动子及全部5个外显子多态性,并进行DNA测序。结果发现hMTH1基因启动子及外显子1序列保守,未见突变;外显子2第73位碱基存在T→C杂合型突变,基因型TT和TC频率分别为93.02%、6.98%,等位基因T和C频率分别为96.51%、3.49%;外显子3第45位遗传密码存在T→C杂合型突变,基因型TT和TC频率分别为95.35%、4.65%,等位基因T和C频率分别为97.67%、2.33%,该多态性为首次发现;外显子4第83位遗传密码存在G→A杂合型突变,基因型GG和GA频率分别为89.53%、10.47%,等位基因G和A频率分别为94.77%、5.23%;外显子5第119位氨基酸遗传密码存在C→T杂合型突变,基因型CC和CT频率分别为95.93%、4.07%,等位基因C和T频率分别为97.97%、2.03%。Abstract: In order to study the genetic polymorphisms of nucleotide repair gene hMTH1 in southern Chinese Han population, the polymorphisms of the gene’s promoter and its five exons among peripheral blood lymphocytes of 172 Chinese Han people were analyzed with polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and DNA sequencing. The sequences of the promoter and exon 1 of hMTH1 gene were conserved. A T to C polymorphism was detected at the 73th base in exon2. The genotype frequencies of TT and TC were 93.02% and 6.98%, respectively. The allelic frequencies of T and C were 96.51% and 3.49%, respectively. A T to C polymorphism was detected at codon 45 in exon3, which was first reported. The genotype frequencies of TT and TC were 95.35% and 4.65%, respectively. The allelic frequencies of T and C were 97.67% and 2.33%, respectively. A G to A polymorphism was detected at codon 83 in exon4. The genotype frequencies of GG and GA were 89.53% and 10.47%, respectively. The allelic frequencies of G and A were 94.77% and 5.23%, respectively. A C to T polymorphism was detected at codon 119 in exon5. The genotype frequencies of CC and CT were 95.93% and 4.07%, respectively. In this study, we analyzed the amino acid pairs affected by mutations in two spike proteins from human coronavirus strains 229E and OC43 by means of random analysis in order to gain some insight into the possible mutations in the spike protein from SARS-CoV. The results demonstrate that the randomly unpredictable amino acid pairs are more sensitive to the mutations. The larger is the difference between actual and predicted frequencies, the higher is the chance of mutation occurring. The effect induced by mutations is to reduce the difference between actual and predicted frequencies. The amino acid pairs whose actual frequencies are larger than their predicted frequencies are more likely to be targeted by mutations, whereas the amino acid pairs whose actual frequencies are smaller than their predicted frequencies are more likely to be formed after mutations. These findings are identical to our several recent studies, i.e. the mutations represent a process of degeneration inducing human diseases.  相似文献

Periods of reentrant activation and effective refractory periods are correlated with dominant frequency or reciprocal of cycle periods during ventricular fibrillation (VF). In the present study, we used an analysis technique based on Wigner transforms to quantify time-varying dominant frequencies in electrocardiograms (ECGs) during VF. We estimated dominant frequencies within orthogonal ECGs recorded in 10 dogs during trials of 10 s of VF and in 9 dogs during trials of 30 s of VF. In four additional dogs, we compared dominant frequencies during 10 s of VF before and after administration of amiodarone. Our results showed the following. 1) There was substantial frequency variation or modulation within the ECGs during 10 and 30 s of VF, the average variation being +/-15% from the mean frequency. Amiodarone decreased mean frequencies (P < 0.05) as expected; however, amiodarone also decreased the variation in frequencies (P < 0.05). 2) During 30 s of VF, the dominant frequencies increased continuously from 7.3 to 8.1 Hz (P < 0.05). The increase in frequency was almost linear with a rate of 0.022 Hz/s (r(2) = 0.93, P < 0.0005). 3) Modulation of frequencies during the first and the last one-half of 30 s of VF was not different. Average (in time) mean frequencies and modulation of frequencies were similar in all three ECGs. 4) Although the averages were similar, during any VF episode, dominant frequencies in ECGs recorded from different locations on the body surface were similar to each other at some times and markedly different from each other at other times. We conclude that during VF, 1) frequencies in ECGs vary considerably and continuously, and amiodarone decreases this variation; 2) mean frequencies increase linearly during first 30 s; 3) the variability in frequency does not change during 30 s; and 4) at any given time, the frequencies within spatially different body surface ECGs can be either similar or markedly different.  相似文献

A Major Role for Bacteriophage T4 DNA Polymerase in Frameshift Mutagenesis   总被引:6,自引:2,他引:4  
T4 DNA polymerase strongly influences the frequency and specificity of frameshift mutagenesis. Fifteen of 19 temperature-sensitive alleles of the DNA polymerase gene substantially influenced the reversion frequencies of frameshift mutations measured in the T4 rII genes. Most polymerase mutants increased frameshift frequencies, but a few alleles (previously noted as antimutators for base substitution mutations) decreased the frequencies of certain frameshifts while increasing the frequencies of others. The various patterns of enhanced or decreased frameshift mutation frequencies suggest that T4 DNA polymerase is likely to play a variety of roles in the metabolic events leading to frameshift mutation. A detailed genetic study of the specificity of the mutator properties of three DNA polymerase alleles (tsL56, tsL98 and tsL88) demonstrated that each produces a distinctive frameshift spectrum. Differences in frameshift frequencies at similar DNA sequences within the rII genes, the influence of mutant polymerase alleles on these frequencies, and the presence or absence of the dinucleotide sequence associated with initiation of Okazaki pieces at the frameshift site has led us to suggest that the discontinuities associated with discontinuous DNA replication may contribute to spontaneous frameshift mutation frequencies in T4.  相似文献

The relative frequencies of the *A allele of the APOA1 gene at -75 bp (M1-) and the C or T +83/+ 84 bp allele (M2-) varied significantly between populations. We found the frequencies of M1- and M2- to be 0.22 and 0.067, respectively, in 150 healthy Omanis. These frequencies were compared to frequencies found in other world populations.  相似文献

It has been recently suggested that mechanical loads applied at frequencies close to the natural frequencies of bone could enhance bone apposition due to the resonance phenomenon. Other applications of bone modal analysis are also suggested. For the above-mentioned applications, it is important to understand how patient-specific bone shape and density distribution influence the natural frequencies of bones. We used finite element models to study the effects of bone shape and density distribution on the natural frequencies of the femur in free boundary conditions. A statistical shape and appearance model that describes shape and density distribution independently was created, based on a training set of 27 femora. The natural frequencies were then calculated for different shape modes varied around the mean shape while keeping the mean density distribution, for different appearance modes around the mean density distribution while keeping the mean bone shape, and for the 27 training femora. Single shape or appearance modes could cause up to 15% variations in the natural frequencies with certain modes having the greatest impact. For the actual femora, shape and density distribution changed the natural frequencies by up to 38%. First appearance mode that describes the general cortical bone thickness and trabecular bone density had one of the strongest impacts. The first appearance mode could therefore provide a sensitive measure of general bone health and disease progression. Since shape and density could cause large variations in the calculated natural frequencies, patient-specific FE models are needed for accurate estimation of bone natural frequencies.  相似文献

为研究中国汉族群体CKMM基因NcoI 酶切位点的遗传多态性以及该位点的具体多态形式, 采用PCR-RFLP技术, 对306例无血缘关系的健康中国北方汉人的染色体进行检测,并对3种基因型的扩增产物进行基因测序。用卡方检验对所得等位基因频率、基因型频率与其他种族进行比较。结果NcoI 位点多态性测序结果为:A/G颠换; 等位基因频率是A=86%,G=14%;基因型频率为:A/A=74%, A/G=24% , G/G=2%;经卡方检验符合Hardy-Weinberg遗传平衡定律;认为中国汉族群体CKMM 基因NcoI酶切位点具有遗传多态性。其基因型频率和等位基因频率在男女间没有显著性差异,与欧美人群相比有极显著差异,而与韩国人相比没有显著性差异。  相似文献

We have studied whether there is natural genetic variation for mutation frequencies, and whether any such variation is environment-related. Mutation frequencies differed significantly between wild strains of the fungus Sordaria fimicola isolated from a harsher or a milder microscale environment in "Evolution Canyon," Israel. Strains from the harsher, drier, south-facing slope had higher frequencies of new spontaneous mutations and of accumulated mutations than strains from the milder, lusher, north-facing slope. Collective total mutation frequencies over many loci for ascospore pigmentation were 2.3, 3.5 and 4.4% for three strains from the south-facing slope, and 0.9, 1.1, 1.2, 1.3 and 1.3% for five strains from the north-facing slope. Some of this between-slope difference was inherited through two generations of selfing, with average spontaneous mutation frequencies of 1.9% for south-facing slope strains and 0.8% for north-facing slope strains. The remainder was caused by different frequencies of mutations arising in the original environments. There was also significant heritable genetic variation in mutation frequencies within slopes. Similar between-slope differences were found for ascospore germination-resistance to acriflavine, with much higher frequencies in strains from the south-facing slope. Such inherited variation provides a basis for natural selection for optimum mutation rates in each environment.  相似文献

A biophysical mechanism of interaction between the man and environment is proposed, which treats these components as two correlated oscillation contours with discrete resonance frequencies.
