A self-review of long-standing studies of induced androgenesis in fish. The data are provided on the induction of diploid androgenesis in teleost fish via blocking the first cleavage division and the development of the method of dispermic androgenesis in sturgeon fishes. Androgenetic nucleocytoplasmic hybrids were obtained using the method of dispermic androgenesis and eggs of previously produced true interspecific hybrids. Specific morphological features of androgenetic hybrids were described and dispermic androgenesis was realized using cryopreserved sperm. Results are described of experimental crossing for obtaining of triple hybrids of the Russian sturgeon Acipenser gueldenstaedtii, the great sturgeon A. huso, and the starred sturgeon A. stellatus and of comparative analysis of some quantitative morphological characters in these hybrids, in the progeny of parental species and of reciprocal hybrids of starred sturgeon and great sturgeon. By most investigated characters, in triple hybrids, the states of the paternal species (Russian sturgeon) are inherited, by some other characters-of great sturgeon-the state of starred sturgeon is not inherited. Therefore, on the basis of external appearance, they may be identified as the hybrid of Russian sturgeon with great sturgeon. The observed segregation by the form of gill rakers into paternal, maternal, or intermediate variants leads to a conclusion of nonmonogenous inheritance of a wide form and a conical form of gill rakers in sturgeons. On the whole, the obtained results clearly demonstrate how, due to effects of matrocliny and partocliny, and different combination of dominant and recessive alleles of the genes determining the diagnostic characters, the hybrid individuals may acquire a different extent of similarity and differences from the parental species. The investigated triple hybrids are an interesting example of mutual leveling of divergent traits in the course of combination of different genomes within limits of one closely related Atlantic group of sturgeons. As a result, a certain averaged habitus is formed which might take place also in case of formation of polyploidy species in the process of evolution.  相似文献

Comparative analysis of several morphological characters is conducted in experimentally obtained hybrids between Siberian sturgeon Acipenser baerii and beluga A. huso, in parental species, and in gynogenetic progeny of Siberian sturgeon. The gynogenetic individuals are similar to Siberian sturgeon by all characters used for differentiation between Siberian sturgeon and beluga that supports gynogenetic origin of these individuals. Based on the data obtained on the hybrid specimens, the features of inheritance of the studied characters in sturgeons and their variation in postlarval ontogeny are discussed.  相似文献

Dispermic androgenesis was used to produce, for the first time, an androgenetic progeny of the Siberian sturgeon (Acipenser baerii) and the androgenetic nucleo-cytoplasmic hybrids (Siberian sturgeon, A. baerii × Russian sturgeon, A. gueldenstaedtii) using cryopreserved sperm. Microsatellite DNA analysis confirmed exclusively paternal inheritance in the androgenetic progeny of Siberian sturgeon. Heterozygotes for certain microsatellite loci were detected among the androgenetic hybrids, thereby confirming a dispermic nature of androgenesis. According to the data of comparative morphological analysis, the obtained androgenetic hybrid, by the age of 15 months old, was completely identical to the paternal species. Both a female and a male were detected in the androgenetic sturgeon progenies, which is of interest for producing bisexual progenies via androgenesis. The data of this study confirm the feasibility of dispermic androgenesis using cryopreserved sperm to preserve and recover the gene pools of endangered sturgeon species.  相似文献

Overexploitation of wild populations due to the high economic value of caviar has driven sturgeons to near extinction. The high prices commanded by caviar on world markets have made it a magnet for illegal and fraudulent caviar trade, often involving low‐value farmed caviar being sold as top‐quality caviar. We present a new molecular approach for the identification of pure sturgeon species and hybrids that are among the most commercialized species in Europe and North America. Our test is based on the discovery of species‐specific single nucleotide polymorphisms (SNPs) in the ribosomal protein S7, supplemented with the Vimentin gene and the mitochondrial D‐loop. Test validations performed in 702 specimens of target and nontarget sturgeon species demonstrated a 100% identification success for Acipenser naccarii, A. fulvescens, A. stellatus, A. sinensis and A. transmontanus. In addition to species identification, our approach allows the identification of Bester and AL hybrids, two of the most economically important hybrids in the world, with 80% and 100% success, respectively. Moreover, the approach has the potential to identify many other existing sturgeon hybrids. The development of a standardized sturgeon identification tool will directly benefit trade law enforcement, providing the tools to monitor and regulate the legal trade of caviar and protect sturgeon stocks from illicit producers and traders, hence contributing to safeguarding this group of heavily threatened species.  相似文献

从施氏鲟(Acipenser schrencki)、小体鲟(Acipenser ruthenus)、西伯利亚鲟(Acipenser baeri)、哲罗鲑(Hucho taimen)和金鳟(Oncorhynchus mykiss)五种鱼类卵黄中分离、纯化Ig,并对其分子结构进行了初步研究。结果表明:五种鱼类卵中Ig的分子量分别为施氏鲟524kD、小体鲟468kD、西伯利亚鲟475kD、哲罗鲑为490kD,金鳟498kD,其Ig重链的相对分子量相同,约为97kD。轻链的相对分子量各不相同,施氏鲟为27kD,小体鲟28.5kD,西伯利亚鲟30kD,哲罗鲑28.5kD和金鳟16kD。其中西伯利亚鲟、施氏鲟和小体鲟Ig等电点约为5.85,哲罗鲑和金鳟的Ig等电点约为6.55,其蛋白质结构上具有一定的相似性,且均为糖蛋白  相似文献

Predation is a common cause of early life stage mortality in fishes, with reduced risk as individuals grow and become too large to be consumed by gape-limited predatory fishes. Large-bodied species, such as sturgeon, may reach this size-refuge within the first year. However, there is limited understanding of what this size threshold is despite the value of this information for conservation management. We conducted laboratory-based predation experiments on juvenile green sturgeon, Acipenser medirostris, to estimate vulnerability to predation during outmigration from their natal reaches in California to the Pacific Ocean. Two highly abundant and non-native predatory fish species (largemouth bass, Micropterus salmoides, and striped bass, Morone saxatilis) were captured in the wild to be tested with developing juvenile green sturgeon from the UC Davis Green Sturgeon Broodstock Program. Experimental tanks, each containing five predators, received thirty prey for 24-hr exposures. Between sturgeon prey trials, predators were exposed to alternative prey species to confirm predators were exhibiting normal feeding behaviors. In addition to green sturgeon mortality data, trials were video recorded and predatory behaviors were quantified. Overall, these predator species displayed much lower rates of predation on juvenile green sturgeon than alternate prey. Predation decreased with green sturgeon size, and predation risk diminished to zero once sturgeon reached a length threshold of roughly 20–22 cm total length, or between 38% and 58% of predator total length. Behavioral analyses showed low motivation to feed on green sturgeon, with both predators attempting predation less frequently as sturgeon grew. Results of this study imply that optimizing growth rates for larval and juvenile sturgeon would shorten the time in which they are vulnerable to predation. Future experiments should assess predation risk of juvenile green sturgeon by additional predator species common to the Sacramento-San Joaquin watershed.  相似文献

The eastern part of the Black Sea and its tributaries are suitable habitats for several sturgeon species, among which Acipenser gueldenstaedtii, A. stellatus, A. nudiventris, A. persicus, A. sturio, and H. huso are well documented. However, different threats have led these species to a dramatic decline, all of them are currently listed as Critically Endangered, and some Locally Extinct, in that area. We tested 94 wild sturgeon samples from the Black Sea and Rioni River by analyzing the mitochondrial Control Region and nuclear markers for hybrid identification. The data analyses (1) assessed mitochondrial diversity among samples, (2) identified their species, as well as (3) indicated instances of hybridization. The data collected, besides confirming a sharp decrease of catches of Beluga and Stellate sturgeon in recent years, also revealed four juvenile hybrids between Russian and Stellate sturgeon, providing the first evidence of natural interspecific hybridization in the Rioni. The present communication raises concerns about the status of sturgeon species in this area and underlines the urgent need for conservation programs to restore self-sustaining populations.

Gut contents of shovelnose and pallid sturgeon from the lower and middle Mississippi River were obtained by colonic flushing, a safe and easily implemented alternative to gastric lavage. Diets of both species were dominated numerically by immature Trichoptera, Ephemeroptera, and Diptera. Primary prey, based on volume, for shovelnose sturgeon were Trichoptera, and for pallid sturgeon were various fishes. Geographic and seasonal nuances in diet were observed for both species, but the general dichotomy of shovelnose sturgeon as browser on invertebrates and pallid sturgeon as predator on fishes did not change. Data indicate that both species require hard substrates for feeding. Data demonstrate that colonic flushing is an effective technique for describing diet and inferring ecological and behavioral information about sturgeon.  相似文献

Extensive habitat modifications within the Mississippi and Missouri rivers have presumably interfered with the reproductive isolating mechanisms between the endangered pallid sturgeon, Scaphirhynchus albus, and the sympatric shovelnose sturgeon, Scaphirhynchus platorynchus, causing hybridization between these two species. Several character indices were developed to assist fisheries biologists in identifying specimens of S. albus, S. platorynchus, and their putative hybrids. The indices have numerous assumptions, including that pure strains of both parental species are within the sample analyzed and that hybrids are morphologically intermediate relative to their parents. If these indices have produced inaccurate identifications, then previous work on Scaphirhynchus studies in the Mississippi and Missouri rivers are questionable, including status surveys, captive propagation efforts, or the harvesting of tissues for genetic studies. In this study we tested indices by examining progeny of ‘known’ pallid, shovelnose, and hybrid sturgeon propagated, raised, and preserved at hatcheries. These 60 specimens [78–600 mm standard length (SL)] were propagated with breeding stock from the upper Missouri River drainage, where hybridization between these two species presumably does not occur. Existing indices did not correctly identify small (<250 mm SL) or a combination of small and large (>250 mm SL) sizes of S. albus, S. platorynchus, and their hybrids. Indices worked fairly well in identifying large S. platorynchus, but not in differentiating large S. albus from hybrids. We used principal components analysis (PCA) as an alternative approach to character indices. No a priori knowledge of the identity of the specimen is required with this multivariate technique, which avoids potential circular reasoning present in indices. We were able to completely or almost completely separate both sturgeon species and their hybrids by extracting principal components from a correlation matrix of 13 meristic characters in a standard PCA and extracting size‐corrected principal components from a covariance matrix of 51 morphometric variables using a sheared PCA. Additionally, we demonstrated that first generation hybrids were intermediate with respect to their parental species. Multivariate analyses with a reduced character set of six meristic and 12 morphometric variables also led to accurate and reliable specimen identification. Head spines and numerous qualitative characters are also extremely useful in differentiating between Scaphirhynchus species and their hybrids. In addition to all morphometric characters, some meristic characters and the degree of head spine fusion vary significantly with SL of sturgeons. Recording appropriate data from released specimens, including photovouchers, and making this information available is essential for researchers to have any scientific or legal basis for genetic or any other studies involving these sturgeons.  相似文献

The state of natural spawning of sturgeons in the Lower Volga River was investigated through genetic monitoring of juveniles (larvae, fingerlings, subadults) captured at seven traditionally used monitoring locations. Sampling was performed during the period 2017–2019. In total, 460 individuals were caught and genotyped using a set of six microsatellite loci (Afug41, Afug51, Afug135, An20, AoxD161, AoxD165) and mtDNA control region sequencing. The observed species-specific microsatellite alleles revealed the presence of juvenile sturgeons originating from natural spawning also included some hybrids between Acipenser stellatus and A. ruthenus. Thus, the majority of sterlet breeding takes place in natural spawning grounds but also sterlet hybridizes with a small number of stellate sturgeons. Juveniles of other sturgeon species were not identified, and it is concluded that natural spawning activities of Russian sturgeon, beluga and ship sturgeon were insignificant or even absent in the Lower Volga during the observation period.  相似文献

Some quantitative characters of three-summer old reciprocal hybrids of beluga Acipenser huso and starred sturgeon A. stellatus,as well as of juveniles of their parental species, are comparatively analyzed. The found maternal and paternal effects in manifestation of some characteristics in hybrids are considered in connection with elucidation of inheritance type of morphological characters in acipenserids. The paternal effect observed by some characters in hybrids beluga × starred sturgeon is explained by prevalence of dominant alleles in the genome of starred sturgeon in comparison with beluga. The maternal effect is elucidated by a few characteristics related to parameters of the head, first of all to the volume of the brain region of skull. Evidently, in this case, the matrocliny depends on the fact that information on the number of segments of embryo and their position is determined by the maternal organism during oogenesis.  相似文献

Microsatellite variation from 13 disomic loci is reported for a total of 208 individuals of the genus Scaphirhynchus. This includes 105 individuals of the pallid sturgeon (Scaphirhynchus albus) from the lower Mississippi River, 11 pallid sturgeon from the Upper Mississippi and Missouri rivers, 65 shovelnose sturgeon (S. platorynchus) from the lower Mississippi River, six Alabama sturgeon (S. suttkusi), and 21 individuals of sturgeon identified as intermediate between S. albus and S. platorynchus. Results indicate that all five of the above population/species units are significantly differentiated from one another based on pairwise FST estimates. Locus Spl‐7 was diagnostic for the Alabama sturgeon and serves to further differentiate this allopatric species from other Scaphirhynchus. Classification of genotypes with and without a priori designations failed to clearly delineate the species and intermediates in the latter case but was successful for the species but not in the intermediates in the former case. The presence of six unique alleles in five of the 21 morphologically ‘intermediate’ sturgeon examined requires additional evaluation but suggests that these individuals are possibly not the result of hybridization. We hope that raising these important issues will bring all stakeholders to the table to establish a concerted effort needed for both morphological and molecular analyses to adequately address the question of hybridization and the origin of the morphological variation in these fishes.  相似文献

The method of polymerase chain reaction with random primers (RAPD PCR) was used to identify the progeny of the crosses between three sturgeon species, Amur sturgeon (Acipenser schrenckii Brandt, 1869), Siberian sturgeon (A. baerii Brandt, 1869), and sterlet (A. ruthenus Linnaeus, 1758). Using ten primers, genetic variation in 70 yearlings, produced in seven individual crosses: Acipenser schrenckii × A. schrenckii, A. baerii × A. baerii, A. ruthenus × A. ruthenus, A. schrenckii × A. baerii, A. baerii × A. schrenckii, A. schrenckii × A. ruthenus, and A. ruthenus × A. schrenckii was described and evaluated. It was demonstrated that the samples composed of hybrids from individual crosses were more variable than the samples of parental species. On the other hand, pooled samples of hybrids from two cross directions were genetically less variable than the pooled samples of their parents. The three main features of the hybrid RAPD profiles identified included: (1) preservation of marker DNA fragments of both parents in one genome; (2) presence of specific DNA fragments, absent from both parents; and (3) dependence of the frequency of some DNA fragments from the cross direction. Multidimensional scaling clearly distinguishes in the space of three coordinates the individuals of original species and the hybrid progeny with differentiation in the groups of direct and backcross hybrids. Analysis of relationships (UPGMA and NJ) pointed to substantial differentiation between the species, as well as between the species and hybrid progeny. Close genetic relationships between direct and backcross hybrids were demonstrated. Multilocus RAPD markers in association with statistical methods are considered to be the useful tool for discrimination of interspecific hybrids of sturgeon. The histological and ultra-structure of the pituitary in diploid red crucian carp (Carassius auratus red var.), triploid crucian carp and allotetraploid hybrids within and after the breeding season were comparatively studied. The result showed that there were six endocrine cell types in the pituitary of these three kinds of fishes, and there was an obvious difference in cell size among different ploidy level fishes. As for the same type of pituitary cells, the cell size was increased gradually with the increasing ploidy level. In the breeding season, the allotetraploid hybrids had higher proportion of gonadotropin cells (GTH) than triploids, and the trploids had higher proportion of GTH than diploids. The results were related to the earlier sexual maturity of allotetraploid hybrids and sterility of triploid crucian carp. On the other hand, among the three kinds of fishes, the proportion of somatotropin (STH) cells in triploids crucian carp was the highest, whereas that in allotetraploid hybrids was the lowest. The results might be connected with the faster growth rate of triploids and slower growth rate of allotetraploid hybrids. In addition, in GTH cells of meso-adenohypophysis after the breeding season, there were many endocrine particles in triploids, while those endocrine particles were released from the cells in allotetraploids and diploids. This result showed that the sterility of triploid crucian carp might be related to the hormone which was not released from the GTH cells. In a word, the present study indicated that the differences in the structure of pituitary among different ploidy level fishes contributed to their difference in the growth rate and gonadal development.  相似文献

Chinese sturgeon Acipenser sinensis belongs to the family Acipenseridae, an ancient species of actinopterygian fishes. In order to advance molecular research on its reproduction, ontogenetic development, we were seeking for genomic information in the NCBI expressed sequence tag (EST database). We found 3384 indentified cDNA sequences which were assembled into 861 unigenes. Blast analysis revealed 301 unigenes shared high similarity with genes in the public databases, and these were classified into three groups: 202 known genes, 81 putative genes and 8 unknown genes. The remainder (560 genes) had no significant match to any protein sequence. Further, 255 unigenes and 333 unmatched unigenes were annotated with Gene Ontology (GO), which could be classified into cellular component, molecular function, and biological process. Among the known genes, the hormone genes pomc A (proopiomelanocortin), pomc B, GtH alpha I subunit (gonadotropin hormone), GtH alpha II subunit and GH (growth hormone) were present in this library. Comparison of the Chinese sturgeon proteins (GH, GtH alpha subunit and POMC) to proteins of other species showed higher levels of homology among sturgeon species. We performed five hormone related genes including GnRHRI (gonadotropin-releasing hormone receptor I), cpH (carboxypeptidase H), ppiB (peptidylprolyl isomerase B), stmn3 (stathmin-like 3), 7B2 (neuroendocrine protein 7B2), and four novel genes (contig 192, 177, 170 and 168) a semi-quantitative RT-PCR on different tissues from Chinese sturgeon.  相似文献

The karyology of species of sturgeon from the Russian Far East demonstrates that the karyotype of the Sakhalin sturgeon (Acipenser mikadoi) includes 262 ± 4 chromosomes with 80 biarmed chromosomes and the number of chromosome arms (NF) 342 ± 4, the karyotype of the Amur sturgeon (A. schrenckii) includes 266 ± 4 chromosomes with 92 biarmed chromosomes and NF 358 ± 4, and the karyotype of the kaluga (A. dauricus) consists of 268 ± 4 chromosomes with 100 biarmed chromosomes and NF 368 ± 4. These results prove that all western Pacific sturgeon species are from a tetraploid origin, based on a recent ploidy scale. This suggests that at least three polyploidization events have occurred during the evolution of Acipenseridae. However, if polyploid species originated by hybridization between diploid species, there may have been more polyploidization events in this group of fishes.  相似文献

Natural hybrids between the boreal species Hexagrammos octogrammus and two temperate species Hexagrammos agrammus and Hexagrammos otakii were observed frequently in southern Hokkaido, Japan. Previous studies revealed that H. octogrammus is a maternal ancestor of both hybrids; the hybrids are all fertile females and they frequently breed with paternal species. Although such rampant hybridization occurs, species boundaries have been maintained in the hybrid zone. Possible explanations for the absence of introgressions, despite the frequent backcrossing, might include clonal reproduction: parthenogenesis, gynogenesis and hybridogenesis. The natural hybrids produced haploid eggs that contained only the H. octogrammus genome (maternal ancestor) with discarded paternal genome and generated F1‐hybrid type offspring by fertilization with the haploid sperm of H. agrammus or H. otakii (paternal ancestor). This reproductive mode was found in an artificial backcross hybrid between the natural hybrid and a male of the paternal ancestor. These findings indicate that the natural hybrids adopt hybridogenesis with high possibility and produce successive generations through hybridogenesis by backcrossing with the paternal ancestor. These hybrids of Hexagrammos represent the first hybridogenetic system found from marine fishes that widely inhabit the North Pacific Ocean. In contrast with other hybridogenetic systems, these Hexagrammos hybrids coexist with all three ancestral species in the hybrid zone. The coexistence mechanism is also discussed.  相似文献

The sturgeon genus Scaphirhynchus consists of threerecognized species. Pallid and shovelnose sturgeon (S. albusand S. platorynchus, respectively) are sympatric in theMissouri and lower Mississippi Rivers of the central United States. TheAlabama sturgeon (S. suttkusi) is endemic to the nearby MobileRiver drainage and is isolated geographically from the other twospecies. Pallid sturgeon and the extremely rare Alabama sturgeon arelisted as endangered under the US Endangered Species Act (ESA).In contrast, shovelnose sturgeon are relatively common and are notlisted. Despite these taxonomies and morphological evidence, somebiologists have questioned the genetic and taxonomic distinctions of thethree species, thus raising doubts concerning the validity of protectingpallid and Alabama sturgeon under the ESA. To investigate thesequestions, we compared a 436 base-pair sequence of the mitochondrial DNA(mtDNA) control region among the three species. We observed 16 mtDNAhaplotypes defined by 27 single base-pair substitutions (transitions)and one single base-pair insertion/deletion (indel) among 78individuals examined. The maximum sequence divergence among thosehaplotypes (2.06%) was less than values usually observed betweenfish species. However, Alabama sturgeon (n = 3) weredistinguished from the other two taxa (n = 75) by aunique base-pair substitution and haplotype, and pallid and shovelnosesturgeon at their northern range of natural sympatry (upper MissouriRiver) did not share any haplotypes. On the other hand, only frequencydifferences among shared haplotypes distinguished (P < 0.01)pallid and shovelnose sturgeon at their southern range of naturalsympatry (Atchafalaya River), and genetic distances between northern andsouthern localities for each species were nearly as large as thedistances between species. These latter results are consistent withseveral hypotheses, including reports (based on morphology) of putativenatural hybrids in the Atchafalaya River but not in the upper MissouriRiver. Overall, these mtDNA results indicate significant reproductiveisolation between pallid and shovelnose sturgeon in areas of naturalsympatry, and recent evolutionary divergence of Alabama sturgeon. ThesemtDNA results provide the first molecular genetic evidence fordistinguishing the three Scaphirhynchus species and, coupledwith morphological and biogeographic data, indicate that pallid andAlabama sturgeon should be evaluated as distinct species under theESA.  相似文献

Taste preferences towards 20 free amino acids (L-isomers, 0.1–0.0001 M) were determined in juveniles of Persian sturgeon Acipenser persicus. It was found that most amino acids (16) had a positive effect on extraoral gustatory reception (increased the frequency of catching artificial pellets by fishes). The most efficient amino acids were the following: threonine, histidine, arginine, asparagine, phenylalanine, cysteine, glutamine, and glycine. Fifteen amino acids were efficient for intraoral gustatory receptors: serine, arginine, cysteine, histidine, alanine, and some others. The presence of these amino acids in pellets increased the consumption. Amino acids decreasing the frequency of catching or consumption of pellets were not found. Highly significant positive correlation was found between the amino acid ranges mediated by the extraoral and intraoral gustatory reception. The fishes demonstrated maximum sensitivity to aspartic acid, 0.01 and 0.001 M, respectively, for intraoral and extraoral gustatory systems. The threshold concentrations of arginine and glycine were higher for the intraoral gustatory system (0.01 M) than for the extraoral one (0.1 M). The comparison of Persian sturgeon with sturgeon fishes (Russian sturgeon A. gueldenstaedtii, Siberian sturgeon A, baerii, and starred sturgeon A. stellatus) studied earlier confirmed high species specificity of intraoral taste preferences in representatives of Acipenser genus. Species specificity of extraoral taste ranges was less pronounced.  相似文献
