改进的SDS-CTAB法提取濒危植物连香树总DNA   总被引:17,自引:0,他引:17  
对珍稀濒危植物连香树(Cercidiphyllum japonicum)的6种总DNA提取方法进行了对比试验,结果表明改进的SDS-CTAB法更适合于连香树总DNA提取。该方法提取的DNA经紫外消光值检测,其A260/A280为1.8532,优于CTAB法(1.4872)、SDS法(1.3552)、PVP法(1.5079)、尿素法(1.1858)和高盐低pH法(1.4534)。琼脂糖凝胶电泳和PCR扩增结果也得出同样的结论。  相似文献

实验以日本龟蜡蚧CeroplastesjaponicusGreen,白蜡绵粉蚧PhenacoccusfraxinusTang ,朝鲜球蚧DidesmococcuskoreanusBorchseniush和瘤大球坚蚧EulecaniumgiganteaShinji等 4种蚧虫为材料 ,分别用十二烷基硫酸钠 (SDS)法、十六烷基三乙基溴化铵 (CTAB)法、醋酸钾 (KAc)法和氯化钠 (NaCl)法等 4种方法 ,对单只蚧虫进行基因组DNA提取 ,用 0 8%琼脂糖凝胶电泳检测所提DNA。结果表明 ,4种方法都可以提取到基因组DNA ,但是比较而言 ,CTAB法和NaCl法所提取的DNA质量明显优于SDS法和KAc法 ,并适用于PCR。因此认为 ,CTAB法和NaCl法是实验室提取单只蚧虫基因组DNA更有效而实用的方法。  相似文献

The Kato-Katz (KK) method is a well-known method of fecal examination for helminthiases. Its diagnostic sensitivity was found very high for clonorchiasis. The present study evaluated the correlation of Clonorchis sinensis egg counts by the KK method with those by direct smear and formalin-ether (FE) technique. The egg counts obtained by the KK method (Y) were correlated with the counts by direct smear (X) with the equation of Y = 659.4 + 0.266X (r2 = 0.738), but not with those by the FE method. The present study demonstrated that the KK method and direct smear were useful for both qualitative and quantitative diagnosis of clonorchiasis, especially in the field.  相似文献

A colorimetric method for the rapid determination of the quantitative content of microbial mass in B. pertussis suspensions has been developed. The method is based on the indirect determination of carbon in microbial suspensions by its oxidation with the mixture consisting of potassium bichromate in concentrated sulfuric acid and the subsequent colorimetric analysis of the products of this reaction. The method ensures sufficient accuracy, the determination procedure is simple, takes not more than 2 hours and requires no complex reagents. The results thus obtained are well comparable with those obtained by the classical gravimetric method. The new method permits the determination of microbial mass in B. pertussis suspensions with a minimum concentration of 0.5 mg/ml. The method is recommended for the determination of dry microbial mass in B. pertussis suspensions.  相似文献

A simplification of Brown’s fluorometry for monkey urinary estrogens has been attained. The method requires only a one-step extraction of estrogens instead of the complicated three-step extractions in the original Brown’s method. The procedure made it possible to shorten the time from the six to seven hours required in the original method to three hours. The present method is satisfactory in precision, sensitivity, accuracy, and specificity. Although the values of urinary estrogens obtained by the simplified method are lower than those obtained by the original method, the use of the present method is validated, since the correlation coefficient between the two assay procedures is excellent (γ=0.90) and also the application of the present method to the measurement of daily changes in urinary estrogens throughout the menstrual cycle has been successful. The simplified assay is expected to be useful in the routine assessment of urinary estrogens in Japanese monkeys. This work was supported by a grant from the Ford Foundation (No. 670-0558).  相似文献

Coagulase-negative staphylococci (CNS) species identification is still difficult for most clinical laboratories. The scheme proposed by Kloos and Schleifer and modified by Bannerman is the reference method used for the identification of staphylococcal species and subspecies; however, this method is relatively laborious for routine use since it requires the utilization of a large number of biochemical tests. The objective of the present study was to compare four methods, i.e., the reference method, the API Staph system (bioMérieux) and two methods modified from the reference method in our laboratory (simplified method and disk method), in the identification of 100 CNS strains. Compared to the reference method, the simplified method and disk method correctly identified 100 and 99% of the CNS species, respectively, while this rate was 84% for the API Staph system. Inaccurate identification by the API Staph method was observed for Staphylococcus epidermidis (2.2%), S. hominis (25%), S. haemolyticus (37.5%), and S. warneri (47.1%). The simplified method using the simple identification scheme proposed in the present study was found to be efficient for all strains tested, with 100% sensitivity and specificity and proved to be available alternative for the identification of staphylococci, offering, higher reliability and lower cost than the currently available commercial systems. This method would be very useful in clinical microbiology laboratory, especially in places with limited resources.  相似文献

A method has been developed for the analysis of (14)CO(2) evolution from the mineralization of (14)C-labelled organic compounds in soil samples. The new method is less space demanding and substantially cuts down laborious manual work compared to the traditional incubation bottle method used. Furthermore, the use of scintillation cocktail is largely reduced with the new method. In the new method, (14)CO(2) is trapped in filter paper held in the lid of a 20 ml glass vial by surface tension. The trapping solution used is Ca(OH)(2), which fixates CO(2) in the filter paper and the analysis of trapped (14)CO(2) is done using the Cyclone trade mark Storage Phosphor system. The lids are placed in a 32 well holder and exposed to a phosphor screen prior to scanning in a Cyclone trade mark scanner. The new filter method has been tested and compared to results obtained using the traditional method. The results show good agreement but due to a smaller capacity for CO(2) with the filter method compared to the traditional method, the interval between sampling has to be shorter using the filter method when the CO(2) development is high. The detection limits for the filter method is higher compared to the traditional method. With the filter method, the level of radioactivity has to exceed 300 dpm before detection is possible, while the same limit for the traditional method is around 30 dpm. On the other hand, the gas trapping faster and the efficiency is higher with the filter method.  相似文献

A method that uses an automated spectrophotometer to read microtiter plates in a modification of the diphenylamine method of DNA determination in tissue extracts, is presented. The micromethod saves technician time, and reduces exposure to acetic acid fumes. The reproducibility of the method is superior to the manual method. A spectrophotometric method has been developed for the quantitative determination of antioxidant capacity. The assay is based on the reduction of Mo(VI) to Mo(V) by the sample analyte and the subsequent formation of a green phosphate/Mo(V) complex at acidic pH. The method has been optimized and characterized with respect to linearity interval, repetitivity and reproducibility, and molar absorption coefficients for the quantitation of several antioxidants, including vitamin E. The phosphomolybdenum method, in combination with hexane monophasic extraction, has also been adapted for the specific determination of vitamin E in seeds. The results obtained with the proposed method were validated by comparison with a standard HPLC method. The phosphomolybdenum method is routinely applied in our laboratory to evaluate the total antioxidant capacity of plant extracts and to determine vitamin E in a variety of grains and seeds, including corn and soybean.  相似文献

Matrix vesicles (MVs) are extracellular organelles involved in the initial steps of mineralization. MVs are isolated by two methods. The first isolation method of MVs starts with collagenase digestion of osseous tissues, followed by two differential centrifugations. The second isolation method does not use proteases but rather starts with differential centrifugation, followed by a fractionation on a sucrose gradient. The first method results in a homogeneous population of MVs with higher cholesterol/lipid content, alkaline phosphatase activity, and mineral formation rate as compared with MVs isolated by the second method. The second method leads to higher protein diversity as compared with MVs isolated according to the first method. Due to their distinct protein composition, lipid-to-protein and cholesterol-to-phospholipid ratios, and differences in rates of mineral formation, both types of isolated MVs are crucial for proteomic analysis and for understanding the regulation of mineralization process at the molecular level.  相似文献

ET-AAS and HG-AAS methods of selenium determination were compared and evaluated. The ET-AAS method has been followed with deuterium background correction and Zeeman background correction respectively. The following validation parameters were determined: accuracy (under repeatability conditions), trueness, calibration curve and linearity, limit of detection, limit of determination and combined standard uncertainty of the method. The HG-AAS method and the ET-AAS method with Zeeman correction were more suitable for the determination of low selenium concentrations in vegetables. The results were calculated by the standard addition method because of the strong matrix effect.  相似文献

Summary The p-dimethylamino-benzaldehyde method of Bomstein and Evans, the ninhydrin method of Marrelli and the new p-dimethyl-amino-benzaldehyde method of Kornfeld were evaluated for analysis of penicillin-V acylase and cephalosporin-V acylase in fermentation broths. 水稻是我国主要粮食作物,每年都会因虫害造成大量的经济损失,为了挽回害虫造成的损失,必须对害虫进行防治。田间节肢动物群落调查是评价害虫防治效果的重要依据,取样方式对节肢动物群落调查的准确性具有重要的影响。另外,对转基因作物对稻田生物多样性安全性进行评价时,取样方式对多样性评价的准确性也具有重要的影响。本文采用吸虫器法、盆拍法和马氏网诱集法3种取样方式进行稻田节肢动物调查,并评估不同取样方式的采集效率。得到的结果有:1.采集到的节肢动物物种数:马氏网诱集法吸虫器法盆拍法;2.采集的节肢动物数量:盆拍法吸虫器法马氏网诱集法;3.吸虫器取样法在调查叶蝉科、秆蝇科、茧蜂科、姬蜂科、金小蜂科、缘腹细蜂科、蕈蚋科时,取样效率较高;4.盆拍取样法在调查叶蝉科、瘿蚊科、微蛛亚科、跳蛛科、狼蛛科、猫蛛科、弹尾虫目、飞虱科时,取样效率较高;5.马氏网诱集法在调查缟蝇科和毛蠓科时取样效率较高。马氏网诱集法善于采集具有飞行能力的节肢动物;吸虫器法对不同习性的节肢动物采集效果均较高;盆拍法适合采集活动于水稻基部的节肢动物。  相似文献

The best method that was developed to extract protein from Chlorella was by sonic disruption of the cells for 3 cycles of 1 min. at 70W on ice in the presence of 1% (w/v) SDS. The best method to quantify the amount of protein in algal extracts is the staining method with the dye bicinchoninic acid. The developed method is reliable for the tested fresh-water micro-alga Chlorella, is also acceptable for the fresh-water micro-alga Chlamydomonas, but is not reliable for the marine micro-algae Dunaliella, Rhodomonas and Synechococcus. The extraction method was the most critical factor factor for determination of proteins in micro-algae and needs to be optimized for each algal species.  相似文献

Upon plotting of areas against optical densities in immunocytochemically stained tissue sections, hyperbolic curves were obtained which could be reduced to two straight lines, one representing variations in stained structures, and the other variations in background. The slopes of the stained structure lines reflected staining intensity independently of total area of stained structure in a section. The ratio of slopes of the stained structure and background lines reflected immunocytochemical sensitivity. A comparison of the peroxidase-antiperoxidase (PAP) method with the avidin-biotin complex (ABC) method showed that at usual antibody dilutions the PAP method was much more sensitive than the ABC method, while at impractically high antibody dilutions it was moderately more sensitive. Once sufficient dilutions of antibodies were reached, staining intensities dropped sharply with the PAP method. On the other hand, the dilution curves were flat with the ABC method. The ABC method consequently appeared unsuitable for estimating variations in concentration of antigen or for distinguishing high or low concentrations of antigen. The ABC method provided a stain for myelin even in the absence of any antibodies.  相似文献

The objective of this study was to validate two indirect methods for estimating the extent of menstrual blood loss against a reference method to determine which method would be most appropriate for use in a population of young adult women. Thirty-two women aged 18 to 29 years (mean +/- SD; 22.4 +/- 2.8) were recruited by poster in Dunedin (New Zealand). Data are presented for 29 women. A recall method and a record method for estimating extent of menstrual loss were validated against a weighed reference method. Spearman rank correlation coefficients between blood loss assessed by Weighed Menstrual Loss and Menstrual Record was rs = 0.47 (p = 0.012), and between Weighed Menstrual Loss and Menstrual Recall, was rs = 0.61 (p = 0.001). The Record method correctly classified 66% of participants into the same tertile, grossly misclassifying 14%. The Recall method correctly classified 59% of participants, grossly misclassifying 7%. Reference method menstrual loss calculated for surrogate categories demonstrated a significant difference between the second and third tertiles for the Record method, and between the first and third tertiles for the Recall method. The Menstrual Recall method can differentiate between low and high levels of menstrual blood loss in young adult women, is quick to complete and analyse, and has a low participant burden.  相似文献
