Exposure to airborne particulate matter has adverse effects on human health and ecosystem. Mutagenic activity of airborne particulate organic matter extracts in three time periods from total suspended particles (TSP) and particles less than 10 μm (PM10) was evaluated in an area under the influence of a petrochemical industry located in the town of Triunfo, Brazil. The extracts were investigated using the Salmonella/microsome assay, with the microsuspension method. The extracts were obtained by sonication extracted using dichloromethane (DCM) solvent. The fractions were tested for mutagenicity with the Salmonella typhimurium strains TA98 (with and without metabolic activation), TA98NR and TA98/1,8DNP6; or YG1021 and YG1024. A positive frameshift mutagenic response was observed for the environmental samples during the different periods. The responses according to percentage of extractable organic matter (EOM%), EOM/m3, revertants/μg (rev/μg) and revertants/m3 (rev/m3) were lower for TSP than for PM10 extracts. The highest rev/m3 values were observed in PM10 extract samples collected in winter, July 2005, in the presence (13.79 rev/m3) or absence (6.87 rev/m3) of S9 fraction. Similarly in the first (1995) or second period (2000) the highest values for TSP were observed in winter, but with lower activity (3.00 and 0.89 rev/m3 respectively). The responses observed for the nitrosensitive strains suggest the contribution of nitro, amino and/or hydroxylamino derivatives of PAHs to the total mutagenicity of matter extracted from airborne particles. The Salmonella/microsome assay was a sensitive method to define areas contaminated by genotoxic compounds, even in samples with TSP or PM10 values that are acceptable according to legal environmental quality standards, favoring environmental control measures with an effective response seen in the population's improved quality of life.  相似文献

Since 1992, the International Atomic Energy Agency (IAEA) has been promoting studies of air pollution using a standard design of air sampler that provides separation on filters into two size fractions with cutoffs of 2.5 and 10 Μm (approximately). These are the size ranges presently considered to have the most important health consequences. Such filter samples are highly amenable to analysis using nuclear and related techniques. After reviewing some of the health effects of airborne particulate matter and current air quality standards and guidelines, this article provides an overview of current and recent IAEA programs in this area, which involve collaborative activities with participants in more than 40 countries.  相似文献

Summary The presence of heavy metals and Polynuclear Aromatic Hydrocarbons (PAH) has been evidenced in airborne particulate matters samples collected in the atmosphere of a non-industrialized middle size Italian town (150.000 inhabitants). The PAH concentrations, higher in winter than in other seasons, was inversely correlated with some meteorological variables such as temperature (p<0.01) and wind speed (p<0.05). The positive correlation found (p<0.01) between lead and total PAH seems to indicate traffic as the most important source of pollution.All samples were mutagenic in the Ames test and the highest mutagenicity levels were obtained in winter samples. The positive correlation found between Benzo(a)Pyrene {B(a)P} concentration and the number of revertants/m3 air, suggests that B(a)P could be an index of mutagenic potential of the matrix considered.The airborne particulate matter extracts obtained by organic solvents were able to inhibit the phagocytosis of mice alveolar macrophages, giving further proof of the possibility of interaction between air-particulate chemical compounds and important biological functions.  相似文献

Particulate air pollution is an important environmental health risk. In the present study, we have investigated the ability of chemically characterized water and organic-soluble extracts of PM(10) from two different regions of Mexico City to induce micronuclei in a human epithelial cell line. We also evaluated the association between the chemical characteristics of the PM and its genotoxicity. The airborne particulate samples were collected from an industrial and a residential region; a Hi-Vol air sampler was used to collect PM(10) on glass fiber filters. PM mass was determined by gravimetric analysis of the filters. One section of each PM(10) filter was agitated either with deionized water to extract water-soluble compounds or with dichloromethane to prepare organic-soluble compounds. The chemical composition of the extracts was determined by ion and gas chromatography and atomic adsorption spectroscopy. A549-human alveolar epithelial cells were exposed to different concentrations of PM(10) extracts and the cytokinesis blocked micronucleus assay was performed to measure DNA damage. Even though the industrial region had a higher PM concentration, higher amounts of metals and PAHs were found in the residential area. Both industrial and residential extracts induced a significant concentration-related increase in the micronuclei frequency. The PM(10) water-soluble industrial extract induced significantly more micronuclei than the one of the residential region; inversely, the organic residential extract induced more micronuclei than the one from the industrial region. The association between the induction of micronuclei and the chemical components obtained by the comparative analysis of standardized regression coefficients showed that cadmium and PAHs were significantly associated with micronuclei induction. Data indicate that water-soluble metals and the organic-soluble fraction of PM(10) are both important in the production of micronuclei. Effects observed, point to the risk of PM exposure and shows the need of integrative studies.  相似文献

Of the many nitroarenes, dinitropyrenes (DNPs) have the potential to revert Salmonella typhimurium his- mutants. This study was conducted to investigate the potential mutagens present in airborne particulate matter collected in Santiago, Chile. 5 organic substances extracted with dichloromethane showed mutagenic rates of from 38.9 to 287 revertants per m3 of air for S. typhimurium his- strain TA98 without S9 mix. 4 of the samples had greatly reduced mutagenicity for strain TA98/1,8DNP6 but not for strain TA98NR. The 1-nitropyrene (1-NP) content accounted for 0.06-0.15 microgram per g of particulate, as determined by high-performance liquid chromatography (HPLC), but the contribution of the compound to mutagenicity was less than 1% of the total activity. On the other hand, by using two columns in the HPLC, DNPs of 1,6- and 1,8-isomers were detected in the samples pooled after the determination of 1-NP, and the amount of the derivatives was about 0.2 microgram per g of particulate matter.  相似文献

Wood combustion produces compounds that are mutagenic in the Salmonella/microsome assay. As combustion products can be emitted in the home and the use of wood as a residential energy source is growing, an impact on human health might be of concern. In this study experiments were carried out to determine the contribution of wood combustion in stoves and fire places to indoor mutagenic activity under normal living conditions. Airborne particles from living rooms which were heated by stoves, or by fire places, and from outdoors were collected simultaneously. In each room two samples were collected during two consecutive weeks: one week the room was heated by central heating, the other week by wood combustion. Sampling took place in a total of 24 homes. Methanol extracts of the samples were tested in the Salmonella/mammalian microsome assay. Results show that mutagenic activity of outdoor air exceeds indoor mutagenicity. At the same time a correlation is found between in- and out-door mutagenicity, both with and without S9. However, a large difference is found between the ratio -S9/+S9 of in- and out-door mutagenic activity. Systematic differences in the ratio -S9/+S9 between control and experimental conditions are not observed. The use of wood stoves caused an increase of indoor mutagenicity in 8 out of 12 homes. It could be concluded that the use of an open fire consistently leads to an increase of mutagenic activity. This increase was caused by wood combustion products.  相似文献

City smog was collected in a heavily industrialized area and investigated for its ability to induce cytogenetic effects in human lymphocytes in vitro. Total extract of city smog was found to produce sister-chromatid exchanges and chromosomal aberrations in a dose-dependent manner. In addition cell-cycle delay was observed at higher concentrations of city smog extract. Results of cytogenetic testing are discussed with respect to cell-cycle kinetics.  相似文献

Aims: To assess antibiotic resistance in airborne bacteria associated with coarse particulate matter (PM10) in an urban area, with specific considerations about the Staphylococcus genus. Methods and Results: Disc diffusion test was performed on 243 microbial strains, isolated from PM10 in winter and summer and belonging to families Pseudomonadaceae and Enterobacteriaceae and genera Acinetobacter, Enterococcus and Staphylococcus. Staphylococci resistances were the most heterogeneous, being distributed among almost all tested antibiotics. Staphylococcus isolates resistant to some selected antibiotics were further investigated for the presence of the corresponding genetic determinants. Only tetK, which mediates the tetracycline resistance through the action of an efflux protein, was found in almost all resistant isolates. Conclusions: The lack of specific genetic determinants makes their transmission among staphylococci less likely. This may reduce the theoretical risk associated with the inhalation of airborne micro‐organisms. Significance and Impact of Study: Although the spreading of antibiotic resistant micro‐organisms is of particular concern in clinical settings, the origin of antibiotic resistance genes can be traced in natural environments. As behaviour, viability and transport of bacteria in the atmospheric compartment suffer from a lack of information, the evaluation of the actual risk posed by airborne micro‐organisms to human health is still challenging.  相似文献

BackgroundAtherosclerosis is a progressive disease characterized by the accumulation of lipids and fibrous plaque in the arteries. Its etiology is very complicated and its risk factors primarily include genetic defects, smoking, hyperlipidemia, hypertension, lack of exercise, and infection. Recent studies suggest that fine particulate matter (PM2.5) air pollution may also contribute to the development of atherosclerosis.Scope of reviewThe present review integrates current experimental evidence with mechanistic pathways whereby PM2.5 exposure can promote the development of atherosclerosis.Major conclusionsPM2.5-mediated enhancement of atherosclerosis is likely due to its pro-oxidant and pro-inflammatory effects, involving multiple organs, different cell types, and various molecular mediators.General significanceStudies about the effects of PM2.5inhalation on atherosclerosis may yield a better understanding of the link between air pollution and major cardiovascular diseases, and provide useful information for policy makers to determine acceptable levels of PM2.5 air quality. The health effects of airborne fine particles are the subject of government regulation and scientific debate. The aerodynamics of airborne particulate matter, the deposition patterns in the human lung, and the available experimental and epidemiological data on health effects lead us to focus on airborne particulate matter with an aerodynamic mean diameter less than 2.5 microm (PM(2.5)) as the fraction of the particles with the largest impact in health. In this article we present a novel hypothesis to explain the continuous production of reactive oxygen species produced by PM(2.5) when it is deposited in the lung. We find PM(2.5) contains abundant persistent free radicals, typically 10(16) to 10(17) unpaired spins/gram, and that these radicals are stable for several months. These radicals are consistent with the stability and electron paramagnetic resonance spectral characteristics of semiquinone radicals. Catalytic redox cycling by semiquinone radicals is well documented in the literature and we had studied in detail its role on the health effects of cigarette smoke particulate matter. We believe that we have for the first time shown that the same, or similar radicals, are not confined to cigarette smoke particulate matter but are also present in PM(2.5). We hypothesize that these semiquinone radicals undergo redox cycling, thereby reducing oxygen and generating reactive oxygen species while consuming tissue-reducing equivalents, such as NAD(P)H and ascorbate. These reactive oxygen species generated by particles cause oxidative stress at sites of deposition and produce deleterious effects observed in the lung.  相似文献

近年来,大气颗粒物成为我国城市大气的主要污染物,其中细颗粒物(PM2.5)粒径小、沉降困难,对环境的危害已成为亟待解决的问题。森林植被可显著消减空气颗粒物,有效改善空气环境质量。本文概述了植被对颗粒物的移除过程和方法,探讨了大气颗粒物与森林植被的相互关系。从单叶、单木及群落3个尺度,结合气象因素讨论了植被对移除大气颗粒物的影响,分析了颗粒物的后续再悬浮过程及对植被的危害。最后,从植被吸附颗粒物的能力测定和评价、本土高吸附PM2.5能力植被的筛选及综合研究不同植被配置结构的吸附效应等方面提出了植被吸附颗粒污染物,尤其是细颗粒物的研究重点与趋势。  相似文献

减少道路环境的颗粒物污染对通勤者的健康至关重要。道路绿地在交通排放和邻近区域之间形成屏障,但这种植被屏障是否可以有效消减局部颗粒物污染仍然需要深入研究和探讨。结合现有研究成果,阐述了植被对颗粒物污染的影响途径;分析了街道峡谷和开放道路两种典型城市道路环境中,植被组成及群落结构设计的关键特征对颗粒物分布和扩散的影响;总结了有利于消减颗粒物浓度的植被单株性状和叶片微形态;探讨了影响颗粒物浓度的其他因子的耦合作用;针对不同道路环境提出了有效的植被屏障设计建议,并指出了植被群落设计和叶片微形态方面的研究趋势,以期优化道路植被规划,改善路域空气质量。  相似文献

The research aims to assess the performance of the lichen Parmotrema reticulatum as an air pollution biomonitor of four heavy metals, namely, chromium (Cr), copper (Cu), lead (Pb) and zinc (Zn). Four contrasting land use sites within the greater Auckland region of New Zealand are used. One site is located within the relatively clean air shed of conservation land, the others within residential, commercial, and industrial areas, characterised by increasingly polluted air sheds, respectively. Three groups of lichens are monitored over a two-year period using active and passive biomonitoring methods to assess ‘on-thallus’ and ‘in-thallus’ concentrations of heavy metals. Seasonal transplants are used to quantify heavy metals accumulated by the lichen during each season. Long-term transplants are used to measure how fast lichens accumulate heavy metals and to better understand how and when heavy metals within the lichen thallus achieve equilibrium with air pollutant concentrations over time. The results show that the lichens continuously accumulates pollutants from the air until equilibrium is reached, thus transplanted lichens are useful for monitoring air pollution concentrations over time. Since pollutant concentration in the transplanted lichen at equilibrium stabilises, at this point the lichen ceases to be useful for monitoring temporal trends in air pollution, but may useful for spatial air pollution monitoring. The industrial location has the highest total accumulation for all four heavy metals, followed by the commercial and residential locations, respectively. Overall, the results show that the lichen P. reticulatum may be successfully used to monitor spatial and temporal pollution patterns caused by even very low concentrations of Cr, Cu, Pb and Zn.  相似文献

Turbidity measurement, as an estimate of the concentration of a particulate subcellular membrane fraction, is an effective alternative to protein assay.
