The octopamine-sensitive adenylate cyclase associated with haemocytes of the American cockroach, Periplaneta americana, has been used as a model system with which to study desensitization of the octopamine receptor. Preincubation of the haemocytes with octopamine results in a large decrease in subsequent maximal stimulation of cyclic AMP production by octopamine with little change in affinity of the receptor for the agonist. This effect of preincubation is dependent upon the concentration of octopamine in the preincubation media and on the duration of exposure. The attenuation appears to be a receptor-mediated event rather than an artifact of the preincubation. Octopamine receptor agonists (octopamine, synephrine, N-demethylchlordimeform) induce desensitization while biogenic amines with poor octopamine receptor affinity (dopamine, serotonin, norepinephrine) are without affect. In contrast, the octopamine receptor antagonist, phentolamine, appears to enhance subsequent stimulation by octopamine. The attenuation of octopamine stimulation of adenylate cyclase is conserved in broken-cell preparations with no alteration of responses to NaF or forskolin. Incubation of the cells with dibutyryl cyclic AMP or forskolin does not induce desensitization. The data indicate that the OA receptors coupled to AC in cockroach haemocytes undergo an homologous desensitization in response to exposure to agonists.  相似文献

We measured the reduction in locomotion of unrestrained pond snails, Lymnaea stagnalis, subsequent to transdermal application of two selective octopamine antagonists, epinastine and phentolamine. After 3 h in fresh standard snail water following treatment with 4 mM epinastine or 3.5 mM phentolamine, the snails’ speed was reduced to 25 and 56% of the controls (P < 0.001 and P = 0.02, respectively). The snails’ speed decreased as the drug concentration increased. In the isolated CNS, 0.5 mM octopamine increased the firing rate of the pedal A cluster motoneurons, which innervate the cilia of the foot. In normal saline the increase was 26% and in a high magnesium/low calcium saline 22% (P < 0.05 and 0.01, respectively). We conclude that octopamine is likely to modulate snail locomotion, partially through effects on pedal motoneurons.  相似文献

Early instar nymphs of the mayfly, Cloeon sp. were kept on an algal or a detrital diet at 20 ± 1 °C in the laboratory. A control group was not given any food. Nymphs kept on algae showed significantly (P = 0.001) higher growth both in terms of length and biomass than for those kept on detritus. None of the nymphs given algae or detritus died during the experiment, and 40 per cent of those fed algae and 20 per cent fed detritus reached maturity by the termination of the experiment. Food retention time as well as gill ventilation rate of nymphs feeding on algae were significantly (P = 0.001) higher than those feeding on detritus. It is probable that these two factors largely enabled the nymphs to survive as well as to grow and reach maturity, though relatively slowly, solely on a detrital diet. However, algae was clearly shown to be a superior food resource for Cloeon sp. nymphs.  相似文献

Insect feeding trials were carried out to determine the effects of incorporating a range of plant derived proteins into artificial diets fed to leafhopper and planthopper pests of rice. The lectins Galanthus nivalis agglutinin (GNA) and wheat germ agglutinin (WGA), and the enzyme soy bean lipoxygenase (LPO) were shown to exhibit significant antimetabolic effects towards first and third instar nymphs of rice brown planthopper (Nilaparvata lugens Stål) when incorporated into artificial diet at 0.1% (w/v), 0.1% (w/v) and 0.08% (w/v) levels respectively. The lectin GNA was also shown to exhibit a significant antimetabolic effect towards third instar nymphs of the rice green leafhopper (Nephotettix cinciteps Uhler). A number of inert proteins, lectins, protein inhibitors and enzymes also tested showed relatively little or no effect towards both insects.  相似文献

The weight loss observed in consumers of extracts of Citrus aurantium (bitter orange) has been tentatively attributed to the lipolytic and thermogenic effects of the alkaloids abundant in the unripe fruit. Synephrine, octopamine, tyramine, and other alkaloids have been repeatedly identified and quantified in Citrus members of the Rutaceae family or in their extracts incorporated in dietary supplements for weight management. However, there are only scarce reports on their lipolytic action. This study aimed at comparing the acute lipolytic activity of synephrine, octopamine, tyramine, and N-methyltyramine in rat and human adipocytes. Maximal response to the prototypical β-adrenergic agonist isoprenaline was taken as reference in both species. In rat, octopamine was slightly more active than synephrine while tyramine and N-methyl tyramine did not stimulate—and even inhibited—lipolysis. In human adipocytes, none of these amines stimulated lipolysis when tested up to 10 μg/ml. At higher doses (≥100 μg/ml), tyramine and N-methyl tyramine induced only 20% of the maximal lipolysis and exhibited antilipolytic properties. Synephrine and octopamine were partially stimulatory at high doses. Since synephrine is more abundant than octopamine in C. aurantium, it should be the main responsible for the putative lipolytic action of the extracts claimed to mitigate obesity. Noteworthy, their common isopropyl derivative, isopropylnorsynephrine (also named isopropyloctopamine or betaphrine), was clearly lipolytic: active at 1 μg/ml and reproducing more than 60% of isoprenaline maximal effect in human adipocytes. This compound, not detected in C. aurantium, and which has few reported adverse effects to date, might be useful for in vivo triglyceride breakdown.  相似文献

The effects of increased levels of dopamine (feeding flies with dopamine precursor, l-dihydroxyphenylalanine) and octopamine (feeding flies with octopamine) on ecdysone 20-monooxygenase activity in young (2 days old) wild type females (the strain wt) of Drosophila virilis have been studied. l-dihydroxyphenylalanine and octopamine feeding increases ecdysone 20-monooxygenase activity by a factor of 1.6 and 1.7, respectively. Ecdysone 20-monooxygenase activity in the young (1 day old) octopamineless females of the strain Tβh nM18 , in females of the strain P845 (precursor of Tβh nM18 strain) and in wild type females (Canton S) of Drosophila melanogaster have been measured. The absence of octopamine leads to a considerable decrease in the enzyme activity. We have also studied the effects of juvenile hormone application on ecdysone 20-monooxygenase activity in 2-day-old wt females of D. virilis and demonstrated that an increase in juvenile hormone titre leads to an increase in the enzyme activity. We discuss the supposition that ecdysone 20-monooxygenase occupies a key position in the regulation of 20-hydroxyecdysone titre under the conditions that lead to changes in juvenile hormone titre and biogenic amine levels.  相似文献

Competitive interactions in arthropod predators are well-known, but positive interactions have received less attention. The two-spotted stinkbugPerillus bioculatus often feeds gregariously on leaf beetle larvae and caterpillar prey. Consequences of prey sharing amongP. bioculatus conspecifics of dissimilar size (instar) was studied using Colorado potato beetle (CPB) prey. Rearing second-instar (N2) nymphs ofP. bioculatus with an N5 conspecific facilitated early feeding on L4 CPB larvae (a difficult prey to handle by N2 nymphs but not by N5’s), thus increasing survival and accelerating development. One in every 20 cases ofP. bioculatus foraging in the field was accounted for by pairs or small groups of mostly feeding individuals. CPB egg masses and L4’s represented a disproportionate number of cases of aggregated feeding byP. bioculatus, compared to feeding singly. Small CPB larvae decreased in the diet of aggregated stinkbugs compared to L4 larvae and egg masses, suggesting that sharing these prey may be favorable or unavoidable. In a field test measuring residence/survival of N2’s limited to L4 prey, the N2’s rate of residence/survival increased significantly when large nymphs acting as food providers were also present. The function of communal feeding inP. bioculatus is discussed, as well as the potential for greater impact on prey density that may be expected from tolerance to opportunistic feeding by conspecifics in slightly gregarious predators.  相似文献

This study compares the relative abilities of rist-instar nymphs of the german cockroach (Blattella germanica L.) and the brown-banded cockroach (Supella longipalpa F.) to survive prolonged periods of food and/or water deprivation.S. longipalpa nymphs survived significantly longer thanB. germanica nymphs when drinking water was absent, but the advantage ofS. longipalpa overB. germanica disappeared if food was also absent. These results are consistent with the hypothesis thatS. longipalpa is adapted to drier habitats than isB. germanica, and suggest thatS. longipalpa nymphs may be more capable of producing and utilizing extra metabolic water from food thanB. germanica nymphs. Evidence from the literature suggests that this species difference applies mainly to juveniles, since adults ofB. germanica andS. longipalpa do not differ in ability to withstand water deprivation, regardless of food availability.  相似文献

Animals, including insects, have the ability to self-select an optimal diet from a choice of two or more incomplete diets that lack an essential nutrient. This paper demonstrates that nymphs of the cockroach Rhyparobia madera also have this ability. The nymphs chose a protein:carbohydrate (P:C) ratio of approximately 25:75 when faced with a choice between one cube of protein (casein) and another of carbohydrate (sucrose). This self-selected ratio was shown to promote growth as well or better than other diets tested. When given a wide range of P:C choices, the R. madera nymphs consistently selected a P:C ratio of approximately 25:75, suggesting that they have the ability to diet-balance. Finally, injections of various serotonergic drugs into self-selecting nymphs influenced their choice of diets. Serotonin promoted a decrease in carbohydrate feeding, while injection of the antagonist -methyltryptophan caused the nymphs to overfeed on carbohydrate. The results suggest that serotonin may help alter the carbohydrate feeding response in cockroaches.  相似文献

Octopamine and 5-hydroxytryptamine (5-HT) were previously shown to affect phagocytosis in cockroach hemocytes through unidentified receptor-mediated events. In the present study, we examined the ability of 5-HT and octopamine to enhance inositol trisphosphate (IP3) production using hemocyte membranes of the American cockroach, Periplaneta americana. Octopamine enhanced IP3 production with a maximal peak at 100 nM. Similarly, 5-HT enhanced IP3 production with a maximal effect at 10 nM. The effects of 5-HT and octopamine are not additive, suggesting that both are working through the same receptor. Phentolamine, a general octopamine antagonist, blocked the effects of octopamine and 5-HT, while a mammalian 5-HT2 antagonist that blocks 5-HT-sensitive receptors in insect peripheral tissue, ketanserin, did not. A pharmacological profile indicates that the receptor is similar to an octopamine1-type. Octopamine at 1 μM increased phagocytosis in cockroach hemocytes exposed to Staphylococcus aureus in vitro, and this effect was mimicked by IP3 (10 μM). The octopamine-treated hemocytes were shown to increase IP3 production in the latter stage of phagocytosis. Adult cockroaches exposed to an LD50 dose of S. aureus in conjunction with either 0.1 mM octopamine or the octopamine1 agonist, clonidine, had higher survival rates compared to saline-treated cockroaches. Correspondingly, the octopamine1 antagonist, chlorpromazine, partially blocked the octopamine-mediated increase in cockroach survival. © 1994 Wiley-Liss, Inc.  相似文献

1. A possible adaptive benefit of coprophagy was investigated in nymphs of the German cockroach Blattella germanica (L.). 2. Newly ecdysed first instars, given no source of food other than conspecific faeces, survived significantly longer than first instars deprived of faeces. The faeces of adult males and females may be of different quality, however, because nymphs given female faeces were more likely to moult into the second stadium than nymphs given male faeces. 3. In contrast to first instars, second instars provided adult faeces survived only slightly longer than starved counterparts. Faecal feeding is therefore stage‐specific, as is the benefit derived from it. 4. The relationship between the nutrient composition of faeces and the survival of nymphs was also examined. First instars fed the faeces of adults that had been maintained on a high (50%) protein diet, died more slowly than first instars fed the faeces of adults that had been maintained on medium (22.5%) and low (5%) protein diets. Chemical analysis of faeces showed that the concentration of protein in adult faeces increased with the level of protein in the diet. Moreover, food choice assays showed that first instars, unlike adults, ingested more of the high‐protein diets. 5. These data support the idea that coprophagy is a stage‐specific adaptive behaviour that permits first instars to moult into the second stadium with minimal foraging.  相似文献

A unique group of immunoglobulin-binding proteins (IGBPs), produced by ixodid male ticks during the latter half of their prolonged feeding period, improves the feeding performance of co-feeding females. As a follow-up to this observation, we investigated whether male tick feeding also affects the feeding of other developmental stages. Immature stages of Rhipicephalus sanguineus (Latreeille) and Amblyomma americanum (L.) were fed on rabbits in the presence or absence of conspecific males. The mean weight of larvae and nymphs of both species that fed around males and detached from the host on the first day of dropping was significantly higher than when the immature ticks fed on rabbits in the absence of males. However, larvae of both species and nymphs of R. sanguineus that fed slower and detached on the second day of dropping did not show significant differences in weight. A similar pattern was observed for A. americanum nymphs although, unlike R. sanguineus, the presence of males also influenced the feeding performance of the nymphs that fed slowly and detached on the second day of drop-off. The improved feeding performance demonstrated by immature ticks in the presence of males may be due to immunomodulatory saliva proteins, such as immunoglobulin-binding proteins (IGBPs) that are introduced into the co-feeding site. The results are considered in relation to the distribution of ixodid tick species on their natural hosts.  相似文献

Taro Colocasia esculenta (L. Schott) and rice (Oryza sativa L.) form a major part of the staple diet of pacific islanders. Pest constraints hamper the sustainability of taro and rice production in the Pacific region. Insect feeding trials were conducted in vitro to determine the effects of plant lectins against planthopper pests of taro and rice. Lectins were incorporated into artificial diet at 0.1% (w/v) level. The lectins Galanthus nivalis agglutinin (GNA) and concanavalin A (Con A) showed significant antimetabolic effects towards third instar nymphs of taro planthopper (Tarophagous proserpina Kirkaldy) whilst Pisum sativum agglutinin (PSA) showed no significant effects toward the insect. Psophocarpus tetragonolobus agglutinin (PTA) showed significant antimetabolic effects towards third instar nymphs of rice brown planthopper (Nilaparvata lugens Stål). PTA also reduced honeydew excretion levels of rice brown planthopper, over a 24-hour period, demonstrating antifeedant properties of the protein.  相似文献

In the courtship behavior of the German cockroach, the male presents tergal glands to the female and feeds her with glandular secretions to place her in the appropriate precopulatory position. The phagostimulant activity of the secretions was quantitatively examined using the polyethylene glycol film method. The methanol extract of the glands on the eighth tergite induced a potent feeding response in 6-day-old virgin females (EC 50 = 0.0037 male equivalent/40 g PEG spot). However, there was no temporal relation between the feeding response and the sexual receptivity of the females. Moreover, besides virgin females, the extract induced a feeding response in gravid or mated females, males, and the last-instar nymphs. These results strongly suggest that the secretions function as a dietary feeding stimulant in principle but as a courtship pheromone in the context of courtship behavior where the stimulants are offered as a nuptial gift.  相似文献

Incubation of bovine luteal cells with the alternative pathway catecholamines octopamine, synephrine and deoxyadrenaline at concentrations of 10(-6) to 10(-3) M enhanced the production of progesterone (P less than 0.05). Tryamine did not alter basal progesterone production (P greater than 0.05). Addition of noradrenaline and adrenaline at concentrations of 10(-4) to 10(-7) M significantly elevated the production of progesterone (P less than 0.05). The steroidogenic response to noradrenaline and adrenaline was greater than that for octopamine, synephrine and deoxyadrenaline (P less than 0.05). Response to both primary (10(-6) M) and alternative (10(-4) M) pathway catecholamines was inhibited by propranolol (10(-5) M, P less than 0.05) but not phentolamine (10(-5) M, P greater than 0.05). These results demonstrate that octopamine, synephrine and deoxyadrenaline can affect steroidogenesis by bovine luteal cells, and their action is mediated by beta-adrenergic receptors.  相似文献

Summary Juvenile hormone production by the corpora allata of the adult female cockroach, Diploptera punctata, can be modulated by treatment with the biogenic amine, octopamine. Endogenous octopamine has been identified within the CA, using HPLC and electrochemical detection. Treatment with octopamine results in a sinusoidal, dose-dependent inhibition of JH biosynthesis by CA from day 2 virgin females, with maximal inhibition occurring at 10-10 M and 10-4 M. In day 4 and day 8 mated female corpora allata octopamine inhibited JH biosynthesis at 5·10-5 M. Although the elevation of either cAMP or cGMP within the CA is known to be associated with an inhibition of JH biosynthesis, treatment with high concentrations of octopamine results in an increase in the level of cAMP but not cGMP. This effect is both dose- and time-dependent.Octopamine treatment also initiates changes in the passive membrane responses of the CA. Superfusion of CA with octopamine results in a pronounced hyperpolarization of CA cells and an increase in the electrotonic potential (indicative of the degree of electrical coupling between CA cells). This effect could be blocked by the octopamine receptor blocker phentolamine. 1. Dicyphus hesperus Knight (Heteroptera: Miridae) nymphs were fed from egg hatch to the adult stage on Ephestia kuehniella eggs provided either alone or in combination with tomato leaves or with a supplementary water source. 2. Only 6% of individuals completed nymphal development on a diet of eggs alone. In contrast, a high proportion of nymphs completed development on a diet of eggs when either tomato leaves (97%) or a supplementary water source (88%) were provided. 3. The development times of nymphs given access to leaves were significantly shorter than those of nymphs given access only to supplementary water. 4. Adult female D. hesperus that were given access to tomato leaves prior to feeding trials consumed significantly more eggs in a 4‐h period than females that were dehydrated before trials. Dehydrated females that were allowed access to water for 3 h before trials consumed an intermediate number of eggs. 5. Plant feeding or access to some other water source is required for prey feeding, growth, and development in D. hesperus, and acquisition of water is proposed as a primary function of plant feeding. In addition, D. hesperus derives nutrients from plant feeding that increase the rate of nymphal development, although nymphs cannot complete development when provided only with tomato leaves. 6. Three simple models are presented of feeding behaviour in predatory Heteroptera where the amount of plant feeding either decreases, increases, or is constant as a function of the amount of prey feeding. The models are discussed with reference to the results and the probable multifunctional nature of plant feeding in predatory Heteroptera.  相似文献

Several insect tissues were examined for their ability to take up octopamine in the presence and absence of sodium ions. The cockroach Malpighian tubules, ovary, and ventral nerve cord showed the highest level of sodium-dependent uptake. The adult firefly lantern exhibited substantial sodium-independent uptake. Some of these tissues were also examined for their ability to metabolize octopamine by N-acetylation. Measurable N-acetyltransferase activity was present in the cockroach ventral nerve cord, tobacco hornworm CNS, and firefly light organ. N-Acetylation is proposed to be the major metabolic pathway for octopamine in the cockroach (Periplaneta americana) nervous system. Several classes of compounds, including octopamine receptor agonists, tricyclic antidepressants, amphetamines, chloroethylbenzylamines, and some experimental insecticides, were tested for their ability to inhibit octopamine uptake and metabolism. The sodium-insensitive component of uptake was not inhibited by most compounds tested, but the sodium-sensitive component was strongly inhibited by xylamine, N-ethyl-N-chloroethyl-o-bromobenzylamine, and their aziridinium ions (60-100%). These compounds also effectively inhibited N-acetyl-transferase (IC50 values at or below 1 microM). Other good inhibitors of N-acetyltransferase included desipramine, synephrine, and an experimental insecticide, CGA 132427. Formamidine pesticides had limited effect on both processes, and neither action seems likely to be involved in their octopaminergic actions in vivo. Cocaine was unique in stimulating N-acetyltransferase activity. When inhibition of sodium-sensitive uptake is compared with inhibition of N-acetyltransferase in the cockroach ventral nerve cord, two groups of inhibitors are discernible. Type 1 compounds inhibit uptake without an effect on N-acetyltransferase, whereas type 2 compounds inhibit both processes. These results suggest a functional linkage between the uptake and acetylation of octopamine.  相似文献

Various biogenic amines including octopamine, dopamine and serotonin, and their precursors and metabolites in haemolymph and the central nervous system from American cockroaches (Periplaneta americana L.) were measured using electrochemical detection.2. Octopamine was found in similar high relative abundances in haemolymph and the central nervous system.3. The amount of octopamine was much higher than that of tyramine and synephrine in haemolymph and thoracic nerve cord, whereas tyramine was at the highest level followed by octopamine and synephrine in the brain.4. Insects were stressed by vibrating at 100 or 1000 Hz, visually by flashing light at 4 Hz for 15 min or by immersing the insect in water at 60°C for 30 sec, which resulted in the elevation of octopamine, tyramine, synephrine and tyrosine levels in thoracic nerve cord.  相似文献

In subsocial xylophagous cockroaches it is thought that parental feeding is important for the survival and growth of the altricial offspring, but the details of parental feeding in these groups are poorly known. We observed stomodeal (oral) trophallaxis between parents and the 2nd or 3rd instars of the wood‐feeding cockroach Salganea esakii Roth, and here report basic features of trophallaxis in young families. Both the female and male parents fed young nymphs with stomodeal food, and there was no difference in the frequency of the behavior between parental sexes. Up to three nymphs could be fed simultaneously during a single trophallactic event. Adults occasionally rejected contact with nymphs by blocking them with their forelegs. Nymphs utilized trophallactic food from parents more frequently than feeding independently on wood pieces or fecal pellets. Trophallaxis between sibling nymphs was never observed. These results suggest that the 2nd and 3rd instar nymphs rely on the stomodeal substances provided by their parents, and that provisioning is an essential component of subsocial behavior in biparental wood‐feeding cockroaches.  相似文献
