Activities of phosphatases were found in the fractions whichwere solubilized from cell walls of both pea and cowpea seedlingswith 0.5 M NaCl. These phosphatases hydrolyzed triphosphonucleotidesin the order: UTP=CTP>GTP>ATP; and UTP=GTP>CTP=ATP,respectively. The activities of a pyrophosphatase and a p-nitrophenylphosphatasewere also detected in these fractions. The suppressor in thespore germination fluid of a pea pathogen,Mycosphaerella pinodes,inhibited all of these phosphatase activities in the fractionsolubilized from pea cell walls, but it rather enhanced onlythe activity of the ATPase among those phosphatases from thecowpea cell wall. Hydrolysis of ATP by a cell wall fractionof pea was also markedly inhibited by the suppressor, whilehydrolysis of ATP by similar fractions from cowpea, kidney beanand soybean were rather enhanced by the suppressor, as wellas by the elicitor. Thus, the cell wall-bound ATPases respondedto the suppressor species-specifically. These cell wall-boundATPases seemed to be different from the plasma membrane ATPasesin several respects. The results suggest that plants recognizethe fungal signals not only on their plasma membranes but alsoon their cell walls and, moreover that putative receptors forthe fungal signals might be located close to cell wall-boundATPases or might even be these ATPases themselves. Cellular membrane fractions, including endoplasmic reticum (ER),Golgi-enriched membrane, plasma membrane and tonoplasts, wereisolated from Vigna radiata seedlings. Each of these membranefractions was associated with specific ATPases which were highlydependent on Mg2+. ATPases of ER, Golgi-enriched membrane andplasma membrane were sensitive to vanadate but the tonoplastATPase was not. ATPases were mostly dependent on Cl1, but aslight stimulation by K+ was observed in the case of ATPasesof Golgi-enriched membrane and plasma membrane. KNO3 inhibitedtonoplast ATPase but stimulated the other ATPases. ER ATPasecan be distinguished from other ATPases by the following characteristics:specific inhibition by KNO2 and Triton X-100, stimulation bylow concentrations of diethylstilbestrol and 4,4'-diisothiocyanostilbene-2,2'-disulfonicacid, and high sensitivity to heat. The ATPases showed typicalMichaelis-Menten kinetics and had Km values of 0.5 to 0.6 ITIMMg2+-ATP for ER, Golgienriched-membrane and tonoplast ATPases,and 2.27 msi Mg2+-ATP for plasma membrane ATPase. ATPases ofGolgi-enriched membranes and plasma membranes had similar properties,but they were still distinguishable by the differences in theirKm values and their responses to Triton X-100. Two membrane fractions were obtained from 16%/26% and 34%/40%interfaces following discontinuous sucrose density gradientcentrifugation of a 10,000–80,000xg pellet from mung bean(Phaseolus mungo L.) roots. The ATPases in the fractions differedfrom each other in their sensitivity toward various inhibitors,activation with salts, dependence of activity on pH, and Kmfor ATP.Mg2+. Judging from their sensitivity toward inhibitors,the ATPases in the low and high density membranes are consideredmainly of tonoplast and plasma membrane origin, respectively.Both ATPases were activated by gramicidin D and nigericin. ATP-inducedquenching of quinacrine fluorescence in both fractions requiredMg2+ and permeant anions such as Cl and quenching wascollapsed by carbonylcyanide p-trifluoromethoxyphenyl hydrazone.The sensitivities of quenching to the inhibitors were essentiallythe same as those of ATPase activity in the membranes. The light-induced H+ efflux observed at acidic pH in Cyanidiumcells was shown to be an active H+ transport depending on theintracellular ATP produced by cyclic photo-phosphorylation.Triton X-100 was found to act as an effective uncoupler in intactCyanidium cells without collapsing the pH gradient across theplasma membrane. Triton X-100 at 0.015% significantly reducedthe intracellular ATP levels, stimulated the p-BQ, Hill reactionand completely inhibited the light-induced H+ efflux. Inhibitionof the H+ efflux by Triton X-100 correlated well with the depressionof the apparent rale of light-induced ATP synthesis as wellas the decrease in the intracellular ATP level in light. The light-induced H+ efflux was completely inhibited by diethylstilbestrol,a specific inhibitor of plasma membrane ATPase, without anychanges in the intracellular ATP level, thereby suggesting theparticipation of the plasma membrane ATPase in the light-inducedH+ efflux. 1The data in this paper are included in the Ph. A pathogenic fungus of pea, Mycosphaerella pinodes, secretesa so-called "suppressor" in its pycnospore germination fluid.The suppressor blocks the defense responses and induces localsusceptibility (accessibility) in pea plants to agents thatare not pathogenic in pea. The suppressor nonspecifically inhibitsthe ATPase activity in plasma membranes prepared from pea, soybean,kidney bean, cowpea and barley plants. However, cytochemicalstudies by electron microscopy indicate that the suppressorspecifically inhibits the ATPase in pea cell membranes, butnot in those of four other plant species tested. That is, thespecificity of the suppressor appears at the cell and/or tissuelevel, but is not evident in vitro. Furthermore, the inhibitoryeffect of the suppressor is temporary because the ATPase activityrecovers 9 h after the treatment. A similar effect was observedafter inoculation with M. pinodes but not with a nonpathogenof pea, M. ligulicola. The role of the suppressor in host-parasitespecificity is discussed. 1. Homogenates of neural lobes of bovine pituitary glands were fractionated by differential and density-gradient ultracentrifugation and the distribution of adenosine triphosphatase (ATPase) activity was studied. It was shown that all the activity was membrane-bound. 2. On the basis of ionic requirements the ATPase activity was grouped into three categories: (a) Mg2+-dependent, (b) Ca2+-dependent and (c) Mg2++Na++K+-dependent (ouabain-sensitive) ATPases. The activity in the absence of bivalent cations was negligible. The ratio between the activities of the three ATPases varied between the different subcellular fractions. 3. Preincubation of the subcellular fractions with deoxycholate increased the activity of the Mg2++Na++K+-dependent enzyme, whereas the Mg2+- and Ca2+-activated ATPases were either unaffected or slightly inhibited. Triton X-100 solubilized the Mg2+- and Ca2+-ATPases; however, the activity of the Mg2++Na++K+-ATPase was abolished by the concentration of Triton X-100 used. 4. All the subfractions displayed unspecific nucleotide triphosphatase activity towards GTP, ITP and UTP. These substrates inhibited the hydrolysis of ATP by all three ATPases. ADP also inhibited the ATPases. 5. Polyacrylamide-gel electrophoresis of extracts containing the Mg2+- and Ca2+-dependent ATPase activity solubilized by Triton X-100 revealed the presence of two enzymes; one activated by either Mg2+ or Ca2+ and the other activated only by Ca2+. 6. In sucrose density gradients the distribution of vasopressin was different from that of all three types of ATPases. It is therefore suggested that the neurosecretory granules do not possess ATPase activity.  相似文献

Envelope membranes were isolated from potato tuber amyloplastby a discontinuous sucrose density gradient and high speed centrifugation.These membranes catalyzed the transfer of [14C]glucose fromUDP-[14C]glucose to endogenous sterol acceptors and, in turn,catalyzed the esterification of steryl glucosides with fattyacids from an endogenous acyl donor. The synthesis of sterylglucosides was stimulated in the presence of Triton. X-100,while formation of acyl steryl glucosides was inhibited by thedetergent. However, in the presence of an added sterol acceptorand Triton X-100, the inhibition of acyl steryl glucoside synthesiswas overcome by the addition of phosphatidylethanolamine. Theenzyme involved in steryl glucoside formation was solubilizedby treatment of the envelope membranes with 0.3% Triton X-100.The solubilized enzyme had an almost absolute requirement forsterol acceptors. The microsomal fraction obtained from guard cell protoplastswas assayed for ATPase activity at pH 6.5. Triton X-100 didnot affect this stimulation of activity of ATPase by K+ up to5 mM, but the detergent abolished the stimulatory effect ofK+ at higher concentrations. The ATPase activity was inhibitedby N,N-dicyclohexylcarbodiimide and ABA. Irradiation with redlight enhanced the ATPase activity more than did irradiationwith far-red light. ABA and irradiation with red or far-redlight were effective only in the presence of K+. These resultssuggest the possibility that the ATPase activity is modulatedonly indirectly by light and ABA. Soluble and wall-bound acid phosphatases isolated from rape seed pollen showed similar properties except for the pH optimum curve which was elevated for the cell wall enzyme. About 50 % of the phosphatase activity of washed pollen wall preparations could be solubilized with Triton X-100, compared with only ca 20% for the corresponding preparation from lily pollen. A comparison of the wall-bound acid phosphatase of rape seed and lily pollen showed a marked difference in specificity towards fructose-6-phosphate and glucose-6-phosphate. A Mg2+-dependent alkaline pyrophosphatase was obtained from rape seed pollen but this activity could not be detected in cell wall preparations.  相似文献

A pulse-chase technique involving the in vivo feeding of L-[1-3H]arabinoseto suspension-cultured rose (Rosa) cells at 4 d and 9 d aftersubculture (fast- and slow-growing, respectively) was used tocreate a population of [3H]xyloglucan molecules and to followtheir subsequent fate. The weight-average relative molecu larmass (Mw) of [3H]xyloglucan freshly deposited in the cell wallwas 160 000 and 240 000 in the fast- and slow-growing cells,respectively. The wall-bound [3H]xyloglucan of both culturesunderwent a decrease in Mw of 40 000 during the first 2 d afterthe pulse-labelling. At the same time, 20–30% of the initially-deposited[3H]xyloglucan disappeared from the cell wall, and a similaramount appeared in solution in the culture medium. Its failureto remain bound to the cell wall and its low Mw (39 000) indicatedthat this soluble extracellular ( was derived from partial degradationof segments of wall-bound xyloglucan that were not directlyhydrogen-bonded to microfibrils (‘loose ends’ and‘tethers’). Nitrate reductase was solubilized and purified from Anabaenacylindrica by Triton X-100 treatment of particulate preparationsfollowed by adsorption on calcium phosphate gel. Natural 15N abundances of various tissues from twelve speciesof leguminous plants and those of nodule fractions from threespecies grown in Japan were determined by mass spectrometry.The 15N concentrations of non-nodular tissues of nodulated plantswere close to the value of atmospheric N2. The nodules of soybean,mung bean, cowpea, kidney bean, adzuki bean, sword bean, whiteclover, Leucaena leucocephala, and horsegram showed highly elevated15N concentrations, and those of pea had a slightly enriched15N concentration compared to other tissue, while the nodulesof peanut and lucerne had 15N values similar to those of theirshoot tissues. A plasma membrane fraction was isolated from the hypocotylsof cowpea {Vigna unguiculata) by a combination of differentialcentrifugation and sucrose density gradient centrifugation.The ATPase activity of this fraction was dependent on divalentcations (Mn2+>Mg2+>Co2+>Ca2+>Fe2+>Zn2+>Ni2+)but was not further stimulated by monovalent cations (K+ and/orNa+). The pH optimum for the activation of ATPase by Mg2+ was7.0. This fraction hydrolyzed ATP or UTP as a substrate andthe ATPase activity obeyed a Michaelis-Menten type of kinetics.The Km for MgATP ranged from 0.65 to 1.1 mM. The ATPase activitywas inhibited by inhibitors such as N, N'- dicyclohexylcarbodiimide,diethylstilbestrol and triphenyltin chloride, all of which arereported to block proton (H+) transport in plant cells, butwas insensitive to those of mitochondrial ATPase such as oligomycinand sodium azide. The ATPase activity was not stimulated bytreatment with ionophores (e.g., carbonyl cyanide p-trifluoromethoxyphenylhydrazone,3,5-di-ter-butyl-4-hydroxybenzilidenemalononitrile and valinomycin+KCl)which would be expected to dissipate the electrochemical potentialdifference of H+ or the membrane potential difference. Membrane ghosts were prepared from protoplasts of the greenalga Mougeotia, and the Ca2+-sensitivity of microtubules onthe ghosts was examined. Microtubules on the protoplast ghosts were not depolymerizedby 3 min treatment with 1 mM Ca2+. As the treatment was prolonged,some depolymerization of microtubules became evident, but evenafter 10 min about 50% of the ghosts showed no depolymerization.Ca2+ introduced into intact protoplasts seemed to be ineffectivein depolymerizing microtubules; abundant microtubules were presenton membrane ghosts prepared from protoplasts which had beentreated with 2x10–5M Ca2+-ionophore A23187 [GenBank] plus 1 mM Ca2+for 20 or 30 min. Neither 3 min treatment with 0.2% Triton X-100 nor with 1 mMCa2+ solution containing 5 min MgSO4 and 100 mM KCl caused depolymerisationof microtubules on protoplast ghosts. However, when given successively,these treatments caused complete depolymerization of microtubules. These results suggest that Mougeotia microtubules are stableto Ca2+ and that the stability is conferred by a microtubule-associatedfactor which can easily be removed by Triton X-100 treatment. The major isoenzymes of -mannosidase (EC 3.2.1.24 [EC] ) and ß-galactosidase(ECf 3.2.1.23 [EC] ) have been separated from cotyledons of gardenpea, Pisum sativum L. (Vicieae), chick pea, Cicer arietinumL. (Cicereae), and cowpea, Vigna unguiculata (L.) Walp. (Phaseoleae).Some of their properties have been determined, including pHoptima, Km values for p-nitrophenyl glycosidc substrates, andthe effects of several inhibitors. Swainsonine, an indolizidinealkaloid, was the most effective inhibitor of mannosidase 1,with I30 values of 5.6 x 10–8 M (cowpea), 1x 10–7M (chick pea) and 2.9 x 19–7 M (pea). The most effectiveinhibitor of ß-galactosidase 2 from all sources wasD-galactonic acid-1,4-lactonwe (-lactone), with Ki values rangingbetween 3.0 and 3.9x 10–3 M. An inhibitor of the E. coliß-galactosidose, p-aminophenyl thio-ß-D-galactopyranoside,did not inhibit any of the legume ß-galctosidases;rather it enhanced the activites of the enzymes from chick peaand cowpea cotyledons. Etiolated hull and seed tissues frompea pods developing in darkness contained similar acid glycosidaseactivities to normal green tissues, thus the chloroplast isan unlikely location for ß-galactosidase 2. The majorß-galactosidasesdetected with an indigogenic substrate (5-bromo-4-chloro-3-indoxyl-ß-D-galactopyranoside)following gel electrophoresis of extracts from pea hull, seedcoats and cotyledons appeared to be different from ß-galactosidase2. Healthy pea plants contain a substance, tentatively called "endogenoussuppressor", which specifically suppresses the accumulationof pisatin in pea plants that is induced by treatment with CuCl2or an elicitor from Mycosphaerella pinodes. This suppressorelicits the accumulation of phytoalexins in other legumes, suchas kidney bean, soybean and cowpea. The endogenous suppressorfunctions to delay the accumulation of pisatin, the activationof phenylalanine ammonialyase (PAL) and the accumulation ofmRNAs for PAL and chalcone synthase induced by the elicitorfrom M. pinodes. The substance specifically induces susceptibilityto nonpathogens, such as Mycosphaerella ligulicola and M. melonis,in pea out of four species of legume tested, but the effectis not cultivar-specific. Thus, the endogenous suppressor inhealthy pea plants suppresses a series of self-defense reactionsand induces susceptibility in pea plants in a species-specificmanner, being similar to the exogenous fungal suppressor fromthe pea pathogen, M.pinodes. Guard cell protoplasts (GCP) were prepared from leaves of Commelinacommunis L. and phosphoenolpyruvate carboxylase (PEPc) activityrecorded after injection of the protoplasts directly into theassay medium. The GCP were lysed immediately by the presenceof Triton X-100 and a lowered osmotic concentration in the assaycuvette enabling PEPc activity to be measured with ‘nascent’enzyme. There was no light activation of the enzyme with KmPEP (about 3.7 mol m–3) and Vmax being similar in light-ordark-treated protoplasts. Illumination of the GCP in the presenceof CO2-free air and KCI, a treatment which is known to swellGCP, did not change the kinetics. PEPc activity at saturating PEP was very sensitive to malateinhibition, 20 mmol m–3 (the I50 value) inhibiting activityby about 50%. Inhibition was similar in light- or dark-treatedprotoplasts. Malate inhibition was, however, much less (I50= 500 mmol m–3) if the enzyme source was a protoplastextract kept in the absence of glycerol. Inclusion of 20% glycerolin the extraction medium maintained the enzyme in the malate-sensitiveform as occurred in the in vivo assays. The high apparent KmPEP and the high sensitivity to malate inhibition of GCP PEPcare features unlike those observed with PEPc from leaf tissuesof C4 and CAM plants and from GCP extracts. PEPc activity increased slightly in the presence of KCI in theassay medium up to about 10 mol m–3 and thereafter activityslowly declined as KCI concentrations increased further. Latent nitrate reductase activity (NRA) was detected in corn (Zea mays L., Golden Jubilee) root microsome fractions. Microsome-associated NRA was stimulated up to 20-fold by Triton X-100 (octylphenoxy polyethoxyethanol) whereas soluble NRA was only increased up to 1.2-fold. Microsome-associated NRA represented up to 19% of the total root NRA. Analysis of microsomal fractions by aqueous two-phase partitioning showed that the membrane-associated NRA was localized in the second upper phase (U2). Analysis with marker enzymes indicated that the U2 fraction was plasma membrane (PM). The PM-associated NRA was not removed by washing vesicles with up to 1.0 M NACl but was solubilized from the PM with 0.05% Triton X-100. In contrast, vanadate-sensitive ATPase activity was not solubilized from the PM by treatment with 0.1% Triton X-100. Treatment of a partially purified preparation of cell walls of Escherichia coli with Triton X-100 at 23 C resulted in a solubilization of 15 to 25% of the protein. Examination of the Triton-insoluble material by electron microscopy indicated that the characteristic morphology of the cell wall was not affected by the Triton extraction. Contaminating fragments of the cytoplasmic membrane were removed by Triton X-100, including the fragments of the cytoplasmic membrane which were normally observed attached to the cell wall. Treatment of a partially purified cytoplasmic membrane fraction with Triton X-100 resulted in the solubilization of 60 to 80% of the protein of this fraction. Comparison of the Triton-soluble and Triton-insoluble proteins from the cell wall and cytoplasmic membrane fractions by polyacrylamide gel electrophoresis after removal of the Triton by gel filtration in acidified dimethyl formamide indicated that the detergent specifically solubilized proteins of the cytoplasmic membrane. The proteins solubilized from the cell wall fraction were qualitatively identical to those solubilized from the cytoplasmic membrane fraction, but were present in different proportions, suggesting that the fragments of cytoplasmic membrane which are attached to the cell wall are different in composition from the remainder of the cytoplasmic membrane of the cell. Treatment of unfractionated envelope preparations with Triton X-100 resulted in the solubilization of 40% of the protein, and only proteins of the cytoplasmic membrane were solubilized. Extraction with Triton thus provides a rapid and specific means of separating the proteins of the cell wall and cytoplasmic membrane of E. coli.  相似文献

The electron transfer activity of an aa3-type two-subunit cytochromec oxidase of Nitrobacter agilis was inhibited by DCCD. Althoughthe activity of the purified cytochrome c oxidase dissolvedin 1% Triton X- 100 was not affected by DCCD even at a ratioof 1,000 mol DCCD per mol cytochrome aa3, the activity of theenzyme dissolved in 0.02% Tween 20 or 0.02% Triton X-100 wasinhibited by 60% or more at a ratio of 1,000 mol DCCD per molcytochrome aa3. The results of SDS polyacrylamide gel electrophoresisof the enzyme incubated with DCCD suggested that subunit IImight be a binding site for DCCD.
