Wood-derived naturally acetylated galactoglucomannans (AcGGM) can be recovered even in ton-scale at mechanical pulp mills using spruce as raw material. These cell wall polysaccharides have a great potential as hydrocolloids and bioactive polymers in food and pharmaceutical applications, or as starting material for production of functional polymers. The immunostimulatory activity of both AcGGM and its deacetylated form (GGM) was now in vitro tested. The biological response of both AcGGM and GGM in the lymphocyte transformation test was dose-dependent. The direct mitogenic as well as comitogenic activities of the AcGGM were comparable to those of the immunogenic corn cob xylan used as control, and GGM showed significantly higher biological responses also at lower doses. In contrast to GGM, AcGGM possessed also DPPH radical-scavenging activity. The results suggested that the spruce AcGGM and GGM are potentially important as additives with immuno-potentiating and antioxidant properties in food products and pharmaceutical formulations.  相似文献

Acetylated galactoglucomannans (GGMs) are the main hemicellulose type in most softwood species and can be utilized as, for example, bioactive polymers, hydrocolloids, papermaking chemicals, or coating polymers. Acetylation of spruce GGM using acetic anhydride with pyridine as catalyst under different conditions was conducted to obtain different degrees of acetylation on a laboratory scale, whereas, as a classic method, it can be potentially transferred to the industrial scale. The effects of the amount of catalyst and acetic anhydride, reaction time, temperature and pretreatment by acetic acid were investigated. A fully acetylated product was obtained by refluxing GGM for two hours. The structures of the acetylated GGMs were determined by SEC-MALLS/RI, 1H and 13C NMR and FTIR spectroscopy. NMR studies also indicated migration of acetyl groups from O-2 or O-3 to O-6 after a heating treatment in a water bath. The thermal stability of the products was investigated by DSC-TGA.  相似文献

Galactoglucomannans fromDactylium dendroides, strain NRRL 2575, and from the galactose oxidase-producing mold originally described asPolyporus circinatus were structurally characterized. The purified galactoglucomannans were homogenous according to cellulose acetate paper and polyacrylamide gel electrophoresis experiments. The major monosaccharide constituents wered-mannose,d-galactose, andd-glucose. Methylation analysis, Smith degradation, graded acid hydrolysis, and1H-NMR and13C-NMR spectroscopy were employed to determine the polysaccharide structures. The immunologic and chemical data showed that the polysaccharides were structurally different.  相似文献

A study was made to explore the rheological properties of the water-solublemucilage of a green laver, Monostroma nitidium. The apparentviscosity of the water-soluble mucilage decreased with increasing shear rate,with increasing solution temperature (from 4 to 70 °C), withincreasing NaCl concentration from (0.5% to 2.0%), and decreasedslightly with a shearing time for 60 min. The decreases in the apparentviscosity were more pronounced for higher-concentration water-solublemucilage. However, it increased with increasing solution pH from pH 3 to 9.  相似文献

Several hemicellulolytic microorganisms were screened for their capability of liberating acetyl side groups from native softwood galactoglucomannan. All the microorganisms tested were found to produce an extracellular acetyl glucomannan esterase(s). The highest activity was detected in Schizophyllum commune culture filtrate. However, the enzyme produced by Aspergillus oryzae was most efficient in long-term hydrolysis. Acting alone, the purified esterase of A. oryzae was able to liberate most of the acetic acid from galactoglucomannan. The addition of other galactoglucomannan-degrading enzymes did not affect the action of esterase. On the other hand, the addition of esterase clearly enhanced the action of mannanase and -galactosidase. The purified acetyl esterase of Trichoderma reesei was able to liberate acetic acid from short oligomers of glucomannan, whereas the acetyl xylan esterase of T. reesei was unable to act on glucomannan oligomers of any size. Solubilities and rheological properties of dissolved fractions of cashew gum from Anacardium occidentale were investigated. A fraction of gum (64.2%) dissolved in water at 30 °C. A further 13.4% dissolved when the remaining suspension was heated. The fraction that dissolved at 30 °C produced greater solution viscosity than the other fraction or the whole gum. When the pH was raised above 5.5, the solution viscosity dropped sharply.  相似文献

Synovial fluid is the joint lubricant and shock absorber [Semin. Arthritis Rheum. 32 (2002), 10-37] as well as the source of nutrition for articular cartilage. The purpose of the present paper is to provide a comprehensive review of the rheological properties of synovial fluid as they relate to its chemical composition. Given its importance in the rheology of synovial fluid, an overview of the structure and rheology of HA (hyaluronic acid) is presented first. The rheology of synovial fluids is discussed in detail, with a focus on the possible diagnosis of joint pathology based on the observed differences in rheological parameters and trends. The deterioration of viscoelastic properties of synovial fluid in pathological states due to effects of HA concentration and molecular weight is further described. Recent findings pertaining to the composition and rheology of periprosthetic fluid, the fluid that bathes prosthetic joints in vivo are reported.  相似文献

Partly depolymerized hemicelluloses isolated from wood chips of spruce and aspen employing microwave treatment were resolved using size-exclusion chromatography (SEC) into oligo- and polysaccharide fractions containing components with a narrow range of sizes, as determined by MALDI mass spectroscopy. The degree of substitution with acetyl moieties (DS) was also calculated on the basis of the MALDI-MS spectra obtained prior to and following deacetylation. For spruce hemicelluloses, the low molecular mass fraction contained small arabino-4-O-methylglucuronoxylan oligosaccharides, with DP values ranging from 4 to approximately 20, separated primarily on the basis of their charge density. The fraction eluted last consisted of an O-acetyl-(galacto)glucomannan polysaccharide of peak-average DP value (DP(p)) 14. The degree of substitution with acetyl groups (DS) decreased with decreasing DP, a value DS of 0.39 being obtained for the fraction with DP(p) 12. For the aspen hemicelluloses, the SEC fractions eluted first contained an acidic O-acetyl-4-O-methylglucuronoxylan polysaccharide with DP ranging from 10 to approximately 28 and an average DS of approximately 0.75. The fractions eluted last consisted of oligosaccharide mixtures composed primarily of small neutral O-acetyl-xylooligosaccharides (DP(p) 6, DS 0.41), together with minor quantities of an O-acetyl-glucomannan.  相似文献

Hydraulic and osmotic properties of roots of 2-year-old Norwayspruce seedlings (Plcea abiea (L.) Karst) were investigatedusing different techniques (steady flow, pressure probe, andstop flow technique). Root pressures were measured using theroot pressure probe. Compared to roots of herbaceous plantsor deciduous trees, excised root systems of spruce did not developappreciable root pressure (-0.001 to 0.004 MPa or -10 to 40cm of water column). When hydrostatic pressure gradients wereused to drive water flows across the roots, hydraulic conductivities(Lpr) were determined in two types of experiments: (i) rootpressure relaxations (using the root pressure probe) and (ii)steady flow experiments (pneumatic pressures applied to theroot system or xylem or partial vacuum applied to the xylem).Root Lpr ranged between 0.2 and 810–8m s–1 MPa–1(on average) depending on the conditions. In steady flow experiments,Lpr depended on the pressure applied (or on the flow acrossthe roots) and equalled (0.190.12) to (1.21.7)10–8m s–1 MPa–1 at pressures between 0.2 and 0.4 MPaand (1.51.3)10–8 m s–1 MPa–1 at appliedpressures between 0.8 and 1.0 MPa. When pressures or vacuumwere applied to the xylem, Lpr values were similar. The hydraulicconductivity measured during pressure relaxations (transientwater flows) was similar to that obtained at high pressures(and water flows). Although there was a considerable scatterin the data, there was a tendency of the hydraulic conductivityof the roots to decrease with increasing size of the root system.When osmotic gradients were used to drive water flows, Lpr valuesobtained with the root pressure probe were much smaller thanthose measured in the presence of hydrostatic gradients. Onaverage, a root Lpr=0.01710–8 was found for osmotic andLpr=6.410–8 m s–1 MPa–1 in correspondinghydrostatic experiments, i.e. the two values differed by a factorwhich was as large as 380. The same hydraulic conductivity asthat obtained in osmotic experiments using the pressure probewas obtained by the 'stop flow techniquel. In this technique,the suction created by an osmoticum applied to the root wasbalanced by a vacuum applied to the xylem. Lpr values of rootsystems did not change significantly when measured for up to5 d. In osmotic experiments with different solutes (Na2S04,K2S04, Ca(NO3)2, mannitol), no passive uptake of solutes couldbe detected, i.e. the solute permeability was very low whichwas different from earlier findings on roots of herbs. Rheological properties are important for the design and modelling of handling and treating fluids. In the present study, the viscosity of liquid manure (about 10% total solids) was measured at different shear rates (2.38-238 s(-1)). The effect of temperature on the viscosity at different shear rates was also studied. The results showed that manure has non-Newtonian flow properties, because the viscosity strongly depended on the applied shear rate. The results showed also that manure behaves like real plastic materials. The power-law model of the shear stress and the rate of shear showed that the magnitude of the consistency coefficient decreased while increasing the temperature, with high values of the determination coefficient. Moreover, the results showed that the Arrhenius-type model fitted the temperature effect on manure viscosity very well (R2 at least 0.95) with calculated activation energy of 17.0+/-0.3 kJ mol(-1).  相似文献

Water-soluble cellulose acetate sulfate derivatives (CAS) have been prepared through chemical reaction involving sulfuric acid as a catalyst. These CAS have been obtained from cellulosic materials of different origins (pure cellulose, wheat bran, maize bran) and their rheological behavior in salt-free aqueous solution has been estimated in dilute and semi-dilute regime using dynamic viscoelastic and viscosity measurements. Influence of concentration, temperature of solubilization and temperature of measurement has been investigated. Weak gel-like properties were exhibited at elevated concentration (typically above 7-8 g/L). These systems also exhibited thixotropic properties: the structure was partly broken down upon shearing and recovered at rest. They also displayed thermoreversibility with large hysteresis, the melting temperature being approximately 15 degrees C higher than the temperature at which gelation took place. These overall observations clearly indicate that these distinctive properties arise from intermolecular association of the macromolecular chains of the cellulose derivative.  相似文献

Galactoglucomannans were isolated by selective precipitation from aqueous and alkaline extracts of endosperm and hulls of Cercis canadensis, a member of the family Fabaceae. Their monosaccharide composition (Man: Gal: Glu) was as follows: 10.4: 0.9: 1 (polysaccharide from the endosperm) and 4.5: 0.9: 1 (polysaccharide from the hulls). The identity of IR spectra was indicative of the commonness of their structure. Analysis of the structure of the galactoglucomannan from endosperm by 13C NMR spectroscopy showed that its main chain consisted of 1,4-beta-D-manno- and 1,4-beta-D-glucopyranose. Part of mannose residues in the chain were substituted at C6 with single residues of alpha-D-galactopyranose. Galactoglucomannans are located in different part of the seed and implement different functions.  相似文献
