Hydrolysis of plant seed gums by microwave irradiation   总被引:5,自引:0,他引:5  
Under microwave irradiation (MW), the seed gums, guar and Ipomoea quamoclit were hydrolyzed to constituent monosaccharides and oligosaccharides in very mild conditions and short reaction time. Under MW both the seed gums could be completely hydrolyzed using very dilute acid (0.00625N H2SO4) within two minutes. Hydrolysis occurs in 2 min and 20 s even in absence of acid under the MW irradiation. Thus hydrolytic fragmentation under MW provides an efficient tool in structural elucidation of polysaccharides.  相似文献

Seeds from 300 species representing 139 genera in 31 plant families have been surveyed for water-soluble gum (mucilage). Four families, Leguminosae, Plantaginaceae, Cruciferae, and Convolvulaceae, have species the seeds of which contain more than 18% gum. The legumes were studied most intensively. Of 163 legume species in 45 genera, 36 contained more than 18% gum, and 54 contained 10 to 18%. Gums from 20 legumes have been analyzed for component sugars; these are all galactomannans. Their specific optical rotation increases regularly as the mannose content decreases. In the legume seed, gum often comprises most of the endosperm. These galactomannan gums can be separated by a milling process to provide commercial-grade gums useful for paper additives and for sizing, thickening, or stabilizing agents.  相似文献

The silver-nanoparticle (AgNP) containing paper was successfully prepared. The AgNP is deposited by the in situ reduction of silver nitrate on the acrylamide grafted bagasse paper sheets in the presence of citrate molecules as stabilizing agent. In the present paper, grafting of acrylamide onto bagasse paper sheets using potassium persulfate was carried out under the influence of microwave radiations (MWR). The modified paper sheets were characterized by Fourier transform infrared spectroscopy (FTIR), UV-spectroscopy, and scanning electron microscopy (SEM). Antimicrobial activities of the prepared paper sheets were also investigated against G+ve bacterium Staphylococcus aureus, G-ve bacterium Pseudomomas aeruginosa, and yeast Candida albicans, which are model microorganisms for testing bactericidal properties. The AgNP containing paper sheets exhibited antibacterial activity.  相似文献

The hydrolysis of colominic acid under microwave irradiation was studied and compared with traditional heating methods. The microwave irradiation has several advantages over the heating method in the hydrolysis of colominic acid: (a) products with higher degrees of polymerization are obtained, (b) less lactone byproducts are observed, and (c) the hydrolytic rate is much faster. These advantages are probably due to the microwave effect. Oligosialic acids as the products of the acid hydrolysis of polysialic acid with conventional heating methods were fully lactonized, especially under the conditions of higher temperature and stronger acid.  相似文献

Using a catalytic amount of potassium persulfate (1.48 x 10(-4)M), eight different seed gums were fully hydrolyzed on alumina support under microwave irradiation. The hydrolysis time varied between 1.33 and 2.33 min depending upon the seed gum structure. The used solid support could be easily separated from the hydrolyzates and recycled. However, under microwave field in an aqueous medium, the same amount of persulfate was unable to hydrolyze the seed gums. Solid-supported microwave hydrolysis has been compared with the microwave-enhanced aqueous hydrolysis (using K2S2O8 or 0.1N H2SO4) and also with the conventional hydrolysis procedures.  相似文献

The effect of inorganic salts on the hydrolysis of starch in a microwave field was investigated and it was found that some inorganic salts can effectively accelerate the acid hydrolysis of starch. The yield of D-glucose reached 111 wt% (equal to the theoretical yield).  相似文献

The effect of inorganic salts such as sodium chloride on the hydrolysis of chitosan in a microwave field was investigated. While it is known that microwave heating is a convenient way to obtain a wide range of products of different molecular weights only by changing the reaction time and/or the radiation power, the addition of some inorganic salts was shown to effectively accelerate the degradation of chitosan under microwave irradiation. The molecular weight of the degraded chitosan obtained by microwave irradiation was considerably lower than that obtained by traditional heating. Moreover, the molecular weight of degraded chitosan obtained by microwave irradiation assisted under the conditions of added salt was considerably lower than that obtained by microwave irradiation without added salt. Furthermore, the effect of ionic strength of the added salts was not linked with the change of molecular weight. FTIR spectral analyses demonstrated that a significantly shorter time was required to obtain a satisfactory molecular weight by the microwave irradiation-assisted inorganic salt method than by microwave irradiation without inorganic salts and conventional technology.  相似文献

Carboxymethyl chitosan was prepared by reacting chitosan with chloroacetic acid in water under microwave irradiation. The effect of the reaction conditions was investigated and optimal conditions were identified. The influence of mass ratio of chloroacetic acid to chitosan, microwave power and pH on the degree of substitution or intrinsic viscosity were further studied. Fast methods for the removal of permanent amide exo-cyclic protective groups widely used in phosphoramidite-method DNA synthesis are desirable for many genomics and proteomics applications. In this communication, we present a method for the deprotection of a range of N-acyl deoxyribonucleosides (T, dA Bz, dC Bz, dC Ac, dG ibu, dG PAC) and synthetic oligodeoxyribonucleotides, ranging in length from 5-mer to 50-mer. Oligodeoxyribonucleotides were synthesized using standard amide protecting groups (dA Bz, dC Bz, dG ibu) and phosphoramidite chemistry on cis-diol solid phase support. This deprotection method utilizes 29% aqueous ammonia solution at 170 degrees C for 5 minutes under monomode microwave irradiation at a 20-nmole reaction scale. Reaction products were analyzed by TLC, RP-HPLC, CE, ESI-MS, real-time PCR, agarose gel electrophoresis, and by DNA uracil glycosylase (UDG) and phosphodiesterase I (PDE) enzymatic digestions.  相似文献

High-intensity ultrasound (HIU) and microwave (MW) irradiation, having emerged as effective promoters of organic reactions, were exploited for the synthesis of bile acids derivatives. Esterification, amidation, hydrolysis, oxidation, and reduction were investigated. Compared to conventional methods, both techniques proved much more efficient, increasing product yields and dramatically cutting down reaction times. Scaled-up studies are now under way.  相似文献

The process of fragmentation can greatly influence plant–animal interactions. To assess the degree to which it affects the balance between two interactions of opposite sign, namely seed dispersal and post-dispersal seed predation, we selected 16 patches of chestnut forest in O Courel and El Bierzo, northwestern Spain. We assessed the effect of fragmentation over two different seed dispersal–predation systems using Helleborus foetidus and Ilex aquifolium as model species. In the first case, field experiments consisted of seed-offering trays with selective exclusion of rodents and ants in a two-way orthogonal design. In the second experiment, we placed experimental branches and trays on the floor to assess seed dispersal and predation. The interactions between several fragment traits and the relative contribution of rodents, ants and birds to seed removal were analyzed by means of generalized linear mixed models. Results show that for H. foetidus, differences in seed dispersal–predation were accounted for by patch shape, which affected mainly the dispersal phase. Major seed dispersal took place in patches with a smaller edge to core ratio and high plant cover (abandoned patches), whilst the latter also showed maximum seed predation. For I. aquifolium, fragmentation effects were significant only for seed predation, which was increased in abandoned patches. This shows that the effects of habitat fragmentation can emerge at different phases depending on specific traits of the interacting animals. It also highlights the importance of traditional land-use practices in species interactions.  相似文献

Establishing safe exposure levels for microwave irradiation is important since new, more powerful emitters are developed and the potential for accidental exposure is increasing. Analysis of the behavior of exposed laboratory animals has proven to be an accurate and repeatable metric for assessing the effects of microwave irradiation. Determining the specific absorption rate (SAR) at which an animal will cease an ongoing behavior has proven useful in the development of safe exposure levels for humans. Behaviors that have been used are simple tasks, and the point at which behavior changes significantly or ceases has often been referred to as “work stoppage.” The tasks have been used to evaluate the overwhelming effects of heating produced by microwave irradiation. Both whole‐body exposures and partial‐body exposures with hotspots have been evaluated. Recent studies have suggested that microwave effects on specific cognitive aspects of behavior such as attention, learning, memory, discrimination, and time perception may occur at SAR levels far below the SARs needed to cause work stoppage. We studied nociceptive responses to subcutaneous injections of formalin and electrical stimulation of the limbs in control mice and in mice whose acupuncture points (AP) were subjected to low-intensity microwave irradiation. In the latter animals, nociceptive reactions were significantly weaker than those in the control mice. The analgesic effect depended on what AP was selected and irradiated and on the duration and timing of microwave irradiation. In different experimental series, the duration of a formalin injection-induced nociceptive behavioral reaction decreased by 23.3–59.6%. The threshold of vocalization responses to stimulation on an electrified floor increased by 25.8±28.0%. The results demonstrate that a technique of analgesia by influencing the AP with microwave irradiation of a nonthermal intensity is rather effective.  相似文献

The pulse flows of populations of sensomotor cortical neurons are studied in unanesthetized non-immobilized rabbits before, during, and after 1-min exposure to microwave electromagnetic irradiation (wavelength 37.5 cm, power density 0.2–0.3, 0.4, 0.5, and 40 mW/cm2) by analyzing the burst activity detected at time thresholds of 5, 10, and 20 ms. These exposures change the number of spike bursts. The changes in pulse flows are recorded both during irradiation and during the first minute after its cessation. The directions of shifts and their dynamics are determined by both the irradiation intensity and the types of bursts themselves.  相似文献

Selective enzymic esterification of free fatty acids, obtained from blackcurrant oil by chemical saponification, with n-butanol using four immobilized lipases under microwave irradiation and under classical heating was studied. A positive effect of microwave irradiation on chemical yields of the products of the enzymic reactions and specificity of lipases were observed in comparison with a controlled heating in an incubator equipped with shaking (classical heating) applied during the identical enzyme-mediated processes. The maximum quantity of -linolenic acid (30%) was obtained with Lipozyme used as biocatalyst of the reaction under microwave irradiation. The maximum quantity of butyl -linolenate (20%) was obtained by a Pseudomonas cepacia lipase catalyzed esterification under classical heating.  相似文献

In experiments on 159 sexually mature dogs with an average weight of 6.5 + 0.71 kg, a study was made of the dynamics of physiological indices (rectal temperature, respiration and heart activity) of heat stress and various (%) effects of damage depending on power density (500, 300, 100 mV/cm2) and duration of microwave irradiation (2400 MHz). On the basis of the data obtained, it was established that the relationship between power density and duration of microwave irradiation with an equally probable (0.1%) effect of damage may be interpolated (100 < power density < 500) by an exponential equation of the type y = 1416 x-0.8156, where y = power density, mV/cm2, and x is the time of irradiation in minutes. The probability characteristic of adaptation possibilities of functionally critical values of damage not exceeding the indeterminate probability is obtained.  相似文献

Summary Formaldehyde is the most commonly used fixative in pathology laboratories. However, due to time pressures, this fixative is often not optimally exploited. the majority of biopsies are only partly fixed when histoprocessing is started, with adverse effects. This paper reports how formaldehyde fixation is improved, by using 1.5 min of microwave irradiation of tissue previously soaked for four hours in the fixation solution. It is argued that this beneficial effect of microwave irradiation can be attributed to the acceleration of the reaction of formaldehyde to the tissue. Formation of free formaldehyde, by the dehydration of methylene glycol present in the tissue when the irradiation starts, is also enhanced. Five different formaldehyde-containing fixatives were evaluated, using five different working protocols. Spleen was taken as a suitable tissue for these tests. The technique described leads to uniform microscopical results. It is a simple method and is suitable for use in routine laboratories.  相似文献

Summary Suspensions of E. coli were exposed to microwave irradiation at constant temperatures. No cell death was observed at 35°C, whereas, at 45, 47, and 50°C, the death rates of E. coli exposed to microwave irradiation were higher than those obtained in conventional heat sterilization at the same temperatures.  相似文献
