Studies have been conducted to ascertain the preventive effect of pyruvate against diabetes induced damage to DNA and associated morphogenetic changes in the mouse lens. Such changes were characterized by DNA nicks as well as by gross morphological changes in the nuclei, evident respectively by TUNEL and Hoechst staining procedures. Morphogenetic changes were also apparent by abnormal diferentiation of the germinal epithelial cells and errors in their migratory pathway. These changes were prevented by simultaneous administration of pyruvate to the diabetic animals. The preventive effect of this agent is attributable to its property of scavenging oxy-radicals generated by high levels of the sugars.  相似文献

Hypoxia during the expansion of adipocytes is known to contribute both to the secretion of multiple inflammation-related adipokines as well as to obesity. We therefore investigated the nature of protein changes occurring in adipocytes during hypoxia by observation of the intracellular proteins that are expressed in 3T3-L1 adipocytes. Lysates were utilized for quantitative proteome analysis using isobaric tags for relative and absolute quantitation (iTRAQ) combined with peptide separation by multi-dimensional liquid chromatography. Antioxidants and elongation factors, as well as glycolytic enzymes were increased in hypoxic adipocytes. These changes were supported by similar changes suggested by real-time PCR. The proteins showing changes are all potential targets for revering the mechanism behind the phenomenon of induction of obese adipocytes by hypoxia. This study can therefore aid in defining the proteomic changes that occur in adipocytes in response to oxygen stress, and can further characterize adipocyte metabolism and adaptation to low oxygen conditions.  相似文献

Calsequestrin, a high-capacity, intermediate-affinity, calcium-binding protein present in the lumen of sarcoplasmic reticulum, undergoes extensive calcium-induced conformational changes at neutral pH that cause distinct intrinsic fluorescence changes. The results reported in this work indicate that pH has a marked effect on these calcium-induced intrinsic fluorescence changes, as well as on calorimetric changes produced by the addition of Ca(2+) to calsequestrin. The addition of Ca(2+) at neutral pH produced a marked and cooperative increase in calsequestrin intrinsic fluorescence. In contrast, at pH 6.0 calsequestrin's intrinsic fluorescence was not affected by the addition of Ca(2+), and the same intrinsic fluorescence as that measured in millimolar calcium at neutral pH was obtained. The magnitude and the cooperativity of the calcium-induced intrinsic fluorescence changes decreased as either [H+] or [K+] increased. The evolution of heat production, determined by microcalorimetry, observed upon increasing the molar ratio of Ca(2+) to calsequestrin in 0.15 M KCl, decreased markedly as the pH decreased from pH 8.0 to pH 6.0, indicating that pH modifies the total heat content changes produced by Ca(2+). We propose that protons bind to calsequestrin and induce protein conformational changes that are responsible for the observed proton-induced intrinsic fluorescence and calorimetric changes.  相似文献

The shoot systems of trees undergo characteristic changes withtime, as they increase in size and complexity. Certain of thesechanges, such as reduced annual growth increment, loss of apicaldominance, and changes in the geotropic responses, are easilyreversible, by pruning or by grafting shoots from older treeson to young stocks. These changes are referred to as ‘ageing’. This paper describes the changes occurring in Pinus sylvestrisand Larix leptolepis during ageing and reports an investigationinto the possible mechanisms controlling these changes. Pruningexperiments suggest that ageing is due to increased competitionfor nutrients between the various constituent shoots as thebranch systems increase in complexity, but the distributionof available nutrients is modified by apical dominance. Theseconclusions were supported by the results of experiments involvingthe injection of radioactive phosphorus (22P), in associationwith the pruning treatments.  相似文献

The effect of caldesmon on the conformational changes of F-actin caused by myosin subfragment 1 (S-1) binding was studied, using the polarized microfluorimetry method. It was demonstrated that the polarized fluorescence of rhodaminil-phalloin specifically bound to F-actin of pure actin filaments as well as of tropomyosin-containing actin filaments changes as a result of binding to S-1. The nature of these changes depends on the presence of caldesmon in the filaments. Caldesmon was supposed to modify the conformational changes in F-actin induced by S-1.  相似文献

DNA treated with thiophosphamide was studied for changes in its biological activity in transformation system of Bac. subtilis. DNA alkylation by this three-functional alkylating agent is shown to be followed by the reduction of transforming activity, changes in the competitive ability as well as by a decrease of the cotransfer coefficient for linked markers of transforming DNA. The observed changes in the transforming and cotransforming activities may be explained by variations in the DNA structure.  相似文献

Rotaviruses are non-enveloped, triple-shelled particles that cause enteritis in animals and humans. The interactions among the different viral proteins located in the three concentric layers make the rotavirus particle an excellent model for physico-chemical and biological studies of viral assemblage. SA11-4S rotaviruses subjected to high pressure were inactivated by more than five log units. After pressure treatment, the particles were recovered with slight structural changes when compared to the control. Electron microscopy suggested subtle changes in the viral outer layer in some pressurised particles. Fluorescence spectroscopy showed that much more dramatic changes were produced by urea denaturation than by pressure. Based on the fluorescence spectrum, the genome resistance to ribonuclease, and the absence of changes in hydrodynamic properties, there was little or no disruption of the capsid under pressure. On the other hand, hemagglutination assays indicated that the main component affected by pressure was the spike protein VP4, thus accounting for changes in interaction with host cells and greatly reduced infectivity. The changes leading to inactivation did not cause removal of VP4 from the outer capsid, as verified by size-exclusion chromatography. Antibodies raised against pressurised material were as effective as antibodies raised against the intact virus, based on their neutralisation titre in plaque reduction assays, enzyme-linked immunosorbent assays and direct interaction with the particle, as measured by gel-filtration chromatography. Therefore, the new conformation of the pressurised particle did not result in loss of immunogenicity. We propose that pressure alters the receptor-binding protein VP4 by triggering changes similar to those produced when the virus interacts with target cells. As the changes in VP4 conformation caused by pressure occur prior to virus exposure to target cells, it leads to non-infectious particles and may lead to the exposure of previously occult epitopes, important for vaccine development.  相似文献

Raman analysis of Na+,K(+)-ATPase structural changes induced by cation binding reveals a slight decrease ( < 10%) of the alpha-helical content upon E1-E2 transition. Pronounced conformational changes of the enzyme are unlikely as the character of the environment of tyrosine residues remains unaltered. Leptin treatment prevents the effects of fasting on reproductive processes in a variety of species. The mechanisms that underlie these effects have not been elucidated. Progress in this area of research might be facilitated by viewing reproductive processes in relation to mechanisms that maintain fuel homeostasis. Reproduction, food intake, and fuel partitioning can be viewed as homeostatic responses controlled by a sensory system that monitors metabolic signals. These signals are generated by changes in intracellular metabolic fuel availability and oxidation rather than by changes in the amount of body fat or by changes in any aspect of body composition. Leptin might be viewed as either a mediator or as a modulator of the intracellular metabolic signal. Consistent with its purported action as a mediator of the metabolic signal, leptin synthesis and secretion are influenced acutely by changes in metabolic fuel availability, and these changes might lead to changes in reproductive function. The effects of leptin treatment on reproduction are blocked by treatments that inhibit intracellular fuel oxidation. Metabolic signals that inhibit reproduction in leptin-treated animals might act via neural pathways that are independent of leptin's action. Alternatively, both leptin and metabolic inhibitors might interact at the level of intracellular fuel oxidation. In keeping with the possibility that leptin modulates the metabolic signal, leptin treatment increases fuel availability, uptake, and oxidation in particular tissues. Leptin might affect reproduction indirectly by altering fuel oxidation or other peripheral processes such as gastric emptying. Reproductive processes are among the most energetically expensive in the female repertoire. Because leptin increases energy expenditure while simultaneously inhibiting energy intake, it may have limited use as a long-term treatment for infertility.  相似文献

Rates of catalase synthesis and degradation were measured in the scutellum of the germinating maize seedling by the technique of Price, Sterling, Tarantola, Hartley & Rechcigl [J. Biol. Chem. (1963) 237, 3468-3475] by using the porphyrinogenic drug 2-allyl-2-isopropylacetamide to inhibit catalase synthesis. Results indicate that developmental changes in catalase activity are determined by changes in the rate of enzyme degradation as well as by changes in the rates of its synthesis.  相似文献

The role of the proton gradient and external pH in the motility and chemotaxis of Bacillus subtilis was investigated. Presence of a substantial proton gradient is not necessary for motility or chemotaxis, as long as the electrical potential is sufficient to maintain motility. Changes in the proton gradient do, however, lead to changes in swimming behavior, and these changes are mediated by two processes. One is sensitive to external pH and probably operates through a pH receptor. The second is sensitive to changes in the proton gradient. When the level of the protonmotive force is high enough to maintain motiligy, changes in the components of the protonmotive force are sensed by the bacteria and lead to behavioral changes, but changes in the protonmotive force are not necessary for chemotaxis.  相似文献

We report here for the first time, in chick retina, Muller cell localization of glutamine synthetase (GS) activity by an immunohistochemical technique, in agreement with previous reports of glial localization of this enzyme in rat brain and retina. Age-dependent changes in the endogenous enzyme activity as well as cortisol-induced changes in GS activity, both in ovo and in vitro, measured biochemically, reflect the changes observed by staining.  相似文献

The lipopolysaccharide (LPS) ofPseudomonas aeruginosa (ATCC 9027) exhibited gross changes in both the amount of LPS produced per cell and the composition of the LPS in response to changes in magnesium ion concentrations. Compositional variation in the LPS was detected under both batch and continuous culture conditions, and was particularly apparent as changes in the levels of heptose in the molecule. Compositional changes in the molecule were also reflected as functional alterations in the LPS. However, functionally, these alterations were counterbalanced by increased production of the lipopolysaccharide. We propose that LPS composition changed in accordance with environmental factors, and that cells responded to these changes by increasing or decreasing the amount of LPS produced.  相似文献

Cyclical changes in microfossil (mainly foraminiferal) assemblages were analysed for sixteen boreholes from three stratigraphical levels in the Lower and Middle Miocene of the Central Paratethys. The following characteristics of assemblages were quantified and used for interpretation of cyclical changes in assemblages: (1) abundance of foraminifers, calcareous nannoplankton, sponge spicules and diatoms; (2) similarity, diversity and epifauna/infauna ratio of benthonic foraminiferal assemblages; (3) planktic/benthic ratio of foraminiferal assemblages. The palaeoecology (mainly palaeobathymetry) fluctuations were interpreted from the species composition of assemblages. Values of the mentioned quantitative characteristics as well as palaeoecological interpretations may be influenced by postmortem transport and resedimentation of foraminiferal tests. Therefore, prior to the interpretation of cyclical changes of quantitative characteristics, the studied sections were classified on the basis of the intensity of taphonomical changes in foraminiferal assemblages. Three different categories of sections were obtained. For every category, those quantitative characteristics of foraminiferal assemblages were chosen which reflect cyclical changes most efficiently: (1) cyclical changes of abundance of foraminifers, calcareous nannoplankton and sponge spicules, as well as the fluctuations in palaeobathymetry for sections dominated by indigenous foraminifers; (2) percentage of indigenous, suspension-transported, bedload-transported and reworked foraminiferal tests and changes in the abundance of indigenous tests for sections dominated by transported tests; (3) different modes of test preservation used for the identification of a cycle boundary for sections with only transported or reworked tests. The distinguished cycles were interpreted predominantly as manifestations of relative sea-level changes. If comparable data exist from other Paratethys regions, the determined cycles can be correlated with the other basins.  相似文献

Dual channel segmentation of the EEG signal has been developed. The purpose was to divide the signals into segments, according to information common for the two channels. The criterion for segmentation was based on the changes in the cross-spectrum of the two signals. It has been shown theoretically, as well as by simulation studies and by analysis of real EEG data that this method is sensitive to changes common for both channels, whereas segmentation does not occur as a result of changes in each channel separately.  相似文献

Abstract. Periodic activities of Dictyostelium discoideum can be observed in cell suspension as two types of oscillations in the light-scattering properties, spike-shaped and sinusoidal. Responses of suspended cells to applied chemoattractants are also reflected by transient changes in light scattering. Alterations in the light-scattering properties are due to structural changes such as changes in cell shape and/or changes in the size of cell aggregates. Therefore, changes in the aggregation state during autonomous oscillations and during attractant-induced responses were investigated. In order to be able to withdraw multiple samples and larger sample volumes from optically monitored cell suspensions, a photometer comprising glass fiber optics immersable in a cell suspension was constructed. Samples were fixed with formaldehyde and photographed. The aggregation state of the samples was quantified by counting the number of particles (cells and cell aggregates) per volume. Folic acid elicited in suspensions of undifferentiated cells a transient decrease in the number of particles per volume as did cAMP in suspensions of preaggregation cells. Periodic changes in the number of particles per volume occurred synchronously with spike-shaped and sinusoidal oscillations. The relative amplitude of the oscillations in particle number was larger during sinusoids than during spikes. Photographs showed periodic changes in the aggregate size during sinusoidal oscillations. In each cycle, the cell-aggregation phase was followed by a phase of partial disaggregation. The recurring loosening of cell-cell contacts may be relevant for sorting out the different cell types. The potential role of contact site as synchronizer and as constituent of an oscillator is discussed.  相似文献

Dendritic spines are multifunctional integrative units of the nervous system and are highly diverse and dynamic in nature. Both internal and external stimuli influence dendritic spine density and morphology on the order of minutes. It is clear that the structural plasticity of dendritic spines is related to changes in synaptic efficacy, learning and memory and other cognitive processes. However, it is currently unclear whether structural changes in dendritic spines are primary instigators of changes in specific behaviors, a consequence of behavioral changes, or both. In this review, we first examine the basic structure and function of dendritic spines in the brain, as well as laboratory methods to characterize and quantify morphological changes in dendritic spines. We then discuss the existing literature on the temporal and functional relationship between changes in dendritic spines in specific brain regions and changes in specific behaviors mediated by those regions. Although technological advancements have allowed us to better understand the functional relevance of structural changes in dendritic spines that are influenced by environmental stimuli, the role of spine dynamics as an underlying driver or consequence of behavior still remains elusive. We conclude that while it is likely that structural changes in dendritic spines are both instigators and results of behavioral changes, improved research tools and methods are needed to experimentally and directly manipulate spine dynamics in order to more empirically delineate the relationship between spine structure and behavior.  相似文献

Hemopexin (Hx) functions as a major heme scavenging protein in blood plasma and as such circulates without heme bound. In recent work, we have demonstrated that Hx binds metal ions in vitro in a manner that varies from one metal ion to another and that changes with heme binding. The structural consequences of metal ion binding to the form of Hx that dominates in plasma have now been evaluated by monitoring metal ion-linked changes in tertiary structure of the protein as reflected by changes in the near-UV CD spectrum and the ultraviolet absorption spectrum as a function of temperature. As part of this analysis we have developed thermally induced difference absorption maps (TIDAMs) to afford efficient visualization of temperature-dependent changes in the UV spectrum of Hx that are induced by binding of metal ions. The results are interpreted in terms of recent models proposed for metal ion binding sites on Hx and have implications for the possible modulation of heme binding to Hx by metal ions in vivo.  相似文献
