An investigation of chloride accumulation and distribution in unifoliate leaves of Phaseolus vulgaris following supra-acute exposures to gaseous hydrogen chloride (HCl) was conducted. Plants which were 8-22 days old (post-seeding) were exposed for 20 min to HCl gas in concentrations ranging from 6.0-54.2 mg m-3 in different combinations of age and concentration. Aqueous extracts of treated unifoliates contained more chloride than was present in control leaves. In both 8 and 12-day-old leaves, the amount of chloride accumulated was proportional to the exposure concentration of HCl, but 12-day-old plants accumulated less chloride than 8-day-old plants. The number of leaves with macroscopic injury symptoms (epidermal glazing and interveinal necrosis) was also related to chloride content. Precipitation of chloride ions by silver salts was employed to examine the ultrastructural distribution of chloride. Deposits of AgCl were present in cell walls, ground plasm, vacuoles, and chloroplasts of both control and HCl-treated leaf tissues. In leaves sampled immediately after treatment, chloroplastic AgCl precipitates were less than in controls, but as sampling was delayed from the HCl treatment, AgCl deposits in chloroplasts regained control levels. The frequency and distribution of AgCl deposits indicated that chloride passed through the leaf cuticle and migrated through the apopolast. Chloride also entered the cytoplasm of cells and appeared to be sequestered in vacuoles of treated cells.  相似文献

Phaseolus vulgaris plants decapitated above the primary leaves accumulate high cytokinin activity. The major cytokinin in these leaves was identified by Sephadex LH 20 chromatography, sensitivity to -glucosidase and permanganate oxidation, and by combined gas chromatography-mass spectrometry as 6-(4-O--D-glucosyl-3-methylbutylamino) purine, (dihydrozeatin-O--D-glucoside). Cell expansion in dicotyledonous leaves is strongly stimulated by bright white light (WL), at least in part as a result of light-induced acidification of the cell walls. It has been proposed that photosynthetic reactions are required for light-stimulated transport processes across plasma membranes of leaf cells, including proton excretion. The involvement of photosynthesis in growth and wall acidification of primary leaves of bean has been tested by inhibiting photosynthesis in two ways: by reducing chlorophyll content of intact plants with tentoxin (TX) and by treating leaf discs with 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU). Exposure to bright WL stimulated growth of intact leaves of TX-treated plants. Discs excised from green as well as from TX-or DCMU-treated leaves also responded by growing faster in WL, as long as exogenous sucrose was supplied to the photosynthetically inhibited tissues. The WL caused acidification of the epidermal surface of intact TX-leaves, but acidification of the incubation medium by mesophyll cells only occurred when photosynthesis was not inhibited. It is concluded that light-stimulated cell enlargement of bean leaves, and the necessary acidification of epidermal cell walls, are mediated by a pigment other than chlorophyll. Cotyledons of resting kidney beans ( Phaseolus vulgaris , L., cv. "Processor") contain high activities of two alkaline peptidases, an aminopeptidase (EC 3.4.11) acting on Leu-Tyr and Leu-Gly-Gly and a dipeptidase (EC 3.4.13) hydrolysing Ala-Gly together with low activities of neutral naphthyiamidases (marker substrate Leu-β-NA) and of acid carboxypeptidases (EC 3.4.16; marker substrate Z-Phe-Ala). The intracellular localisation of these peptidases and that of α-mannosidase (EC 3.2.1.24) was studied by subcellular fractionations in different media. In density gradient centrifugations in non-aqueous glycerol-potassium iodide media the alkaline peptidases remained mainly in the application zone suggesting localisation in the cytosol. The carboxypeptidase and α-mannosidase activities banded mainly in the protein body zone, but about 15–30% of each activity was found in the cell wall zone. Results obtained by short centrifugation in glycerol or high-density sucrose solutions (65/70%) and by the isolation of essentially pure cell wall fractions confirmed these assignments. The results are in accordance with previous suggestions that the abundant alkaline peptidases may play a role in the mobilization of reserve proteins in germinated seeds by hydrolysing peptides which are produced initially within the protein bodies by acid proteinases and carboxypeptidases and which subsequently leak out or are transported from the autolyzing protein bodies to the cytosol.  相似文献

Acid phosphatase (ACP) activity in common bean grown with or without 1.5 mM of phosphate has been examined. Leaves and root nodules responded to the absence of an exogenous phosphate source with an increase in ACP activity. Increases in enzyme activity were not associated with the synthesis of new isoforms of the enzyme. We partially purified and characterized the ACPs, which consisted of three proteins, one of leaf and two of nodule. Proteins of leaf migrated at 72 and 51 kDa in SDS-PAGE, whereas that of nodule migrated at 72, 49, 41 and 34 kDa. Enzymes of both organs had a pH optimum of 5.6, and were relatively heat stable. The enzymes exhibit a broad substrate selectivity, with maximal activity obtained with alpha-naphthyl-phosphate, ribulose 1,5-bisphosphate and p-nitrophenyl-phosphate (p-NPP). Potent inhibition by Zn2+, Hg2+, Cu2+, Pb2+, Al3+ and (MoO4)2- was observed.  相似文献

The major cytokinins in stems of decapitated, disbudded bean plants have been identified by enzymic degradation, Sephadex LH20 and reversed phase high performance liquid chromatography, and by combined gas chromatography-mass spectrometry as 6-(4-hydroxy-3-methylbut-trans-2-enylamino)-9--D-ribofuranosylpurine (zeatin riboside), 6-(4-hydroxy-3-methylbutylamino)-9--D-ribofuranosylpurine (dihydrozeatin riboside), and the 5-phosphates of these compounds (zeatin ribotide and dihydrozeatin ribotide). Minor cytokinins in this tissue were tentatively identified as dihydrozeatin-O--D-glucoside and zeatin ribotide-O--D-glucoside. [8-14C-]Dihydrozeatin appeared to be rapidly metabolized to dihydrozeatin ribotide when supplied to segments of stems from decapitated plants. Although determinations of the ABA content of lateral buds ofPhaseolus vulgaris revealed no difference between decapitatedand intact control plants in the first 12 h following decapitation,a relative decrease in the ABA content of lateral buds of decapitatedplants was detectable 24 h following decapitation. Shoot decapitationwas also observed to result in a decrease in the ABA contentof stem tissue. The application of IAA to the stem of decapitatedplants prevented these changes and increased the ABA contentof stem tissue relative to that of intact plants. The levelsof IAA and ABA were also determined in the stem tissue fromthe nodes of intact bean plants. The possible interdependenceof these two plant hormones was further investigated by a studyof [2–14ClABA metabolism. A recent controversial report suggests that the nocturnal inhibitor of Rubisco, 2-carboxy-D-arabinitol 1-phosphate (CAIP), does not bind to Rubisco in vivo and therefore that CA1P has no physiological relevance to photosynthetic regulation. It is now proved that a direct rapid assay can be used to distinguish between Rubisco-bound and free CA1P, as postulated in the controversial report. Application of this direct assay demonstrates that CA1P is bound to Rubisco in vivo in dark-adapted leaves. Furthermore, CA1P is shown to be in the chloroplasts of mesophyll cells. Thus, CA1P does play a physiological role in the regulation of Rubisco.  相似文献

Summary Embryo development was examined in reciprocal crosses of Phaseolus vulgaris cv. Great Northern and P. coccineus cv. Scarlet Runner. The formation of abnormal (shrunken and underdeveloped) embryos constituted the primary crossing barrier between the two species when P. coccineus was the female parent. Plants of P. coccineus X P. vulgaris were obtained by embryo culture. Although the P. vulgaris X P. coccineus cross resulted in normal seed development, the fertility of the resulting hybrids was much lower (27%) than that of the reciprocal hybrids (81%). Three classes of F2 embryos, normal, shrunken, and underdeveloped were formed on reciprocal F1s and the frequencies did not differ between reciprocal populations. Thus, the interactions between embryo and endosperm and/or maternal parent rather than cytoplasmic-nuclear effects seem to be important in the determination of the extent of embryo growth. The examination of pollen fertility of F2 plants and the development of F2 and F3 embryos suggests that the formation of abnormal embryos and reduced male fertility are independent events. The P. vulgarisP. coccineus crosses may be useful in studying the possible involvement of interspecific differences in hormonal metabolism in the development of hybrid embryos.  相似文献

The aim of this research was to investigate the nature of heterogeneity in stomatal conductance and, in particular, to determine whether the characteristic 'patchy' pattern of water infiltration is reflected in measurements on individual stomata. Silicone rubber replicas were made of primary leaves of glasshouse-grown Phaseolus vulgaris L. plants, and the leaves were then infiltrated with water at controlled sub-atmospheric gas pressures according to their estimated or measured stomatal conductance. Seven leaves examined in detail all showed patchy infiltration, and the mean sire of infiltrated areas was negatively correlated with the prevailing stomatal conductance. In four of the leaves, a one millimetre wide transect across the leaf was selected for further detailed study. Measurements of mean peristomatal groove distance (PGD) and stomatal frequency were made along the transect and related to the state of infiltration. Analysis of variance indicated that, in all four cases, variation in PGD among patches was highly significant, but there was no significant difference between patches of different infiltration categories. Thus, local (patch-level) variation in stomatal aperture appeared to bear no relation to the infiltration status of the patches. The dominant source of stomatal variability was between individual pores in the same locality, which accounted for 82% or more of the total variability. Taking into account variation in stomatal frequency, correlations between predicted stomatal conductance and the extent of infiltration were significant in only one out of the seven leaves studied. Possible reasons for these results are discussed. It is suggested that the infiltration method misrepresents the underlying state of the stomata as being either open or closed, when there is little evidence for this from measurements of stomatal dimensions. For these unstressed plants under relatively stable conditions, it is concluded that the 'unit of variability' in stomatal heterogeneity may rest at the individual pore ('micro') scale, rather than at the areolar patch ('macro') scale, or above.  相似文献

Abstract

Callus production and plant regeneration from different explants of Phaseolus vulgaris L. cv. Giza are reported. Calli cultures were induced from leaf, hypocotyl, embryo and root explants. Rapid growth of callus was achieved by leaf explants cultured on MS salts, B5 vitamins and supplemented with 2,4— dichlorophenoxyacetic acid (2, 4—D)+0.5 mg/l kinetin (kin). Addition of casein hydrolysate at 2 g/l to maintenance medium enhanced callus growth and hindered the early appearance of necrotic parts. This report also provides a detailed method for production of multiple shoots directly from the wounded edges of immature cotyledon explant via organogenesis on 1 mg/l benzyladenine (BA) or indirectly on 0.5 mg/l naphthaleneacetic acid (NAA)+2 mg/l BA. The regeneration of bean plants through the two ways described here (direct or indirect) may be of use in genetic improvement of bean.  相似文献

Summary Restriction fragment length polymorphism (RFLP) was determined among P. vulgaris genotypes and Phaseolus species using 19 probes. The incidence of polymorphism was high (70–86%) between species, but relatively low (22–26%) between genotypes of P. vulgaris. Suitable probes were identified for the analysis of P. vulgaris and P. coccineus hybrids. The segregation pattern in F2 populations was Mendelian for two probes (LHB and VEE20) and non-Mendelian for GS-g, CHS, and CHI. Statistical analyses indicated gametic selection with preferential transmission of the P. vulgaris alleles, which may account for the selective recovery of P. vulgaris progeny types observed earlier. The available hybrids of P. vulgaris and P. coccineus and the high degree of interspecific RFLP will facilitate the construction of a linkage map for Phaseolus.  相似文献

The glutamine synthetase (GS) isozymes in the plant fraction of nodule extracts from 62 cultivars of Phaseolus vulgaris L. and one cultivar of Phaseolus lunatus L. were analyzed by polyacrylamide gel electrophoresis. All P. vulgaris nodule extracts displayed two GS activity bands: a nodule-specific band (GSn1) and a band (GSn2) similar to the single band (GSr) present in root extracts. In nodule extracts of P. lunatus, the GSn1 band was detected, but the GSn2 band was barely detectable. In contrast to P. vulgaris, the GSn2 band and the GSr band of P. lunatus appeared to be different. The electrophoretic mobility of the GSn1 band in P. vulgaris was governed by both the plant cultivar and the development stage of the nodule. In nodule extracts of P. vulgaris and P. lunatus, the zone of GSn1 activity coincided with six to nine distinct protein bands as revealed after treatment of gels, which had previously been stained for GS activity, with Coomassie blue. All these protein bands were shown to consist of polypeptides of identical molecular weight (approximately 47,000 daltons) by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Our results indicate that P. vulgaris continuously generates isozymes of GSn1 of increasing electrophoretic mobility during the course of nodule development.  相似文献

Summary The development of increased activities of ribulosediphosphate carboxylase (EC 4.1.1.39) and of phosphoribulokinase (EC 2.7.1.19) in greening bean leaves was completely inhibited by D-threo chloramphenicol but unaffected by L-threo chloramphenicol. This indicates that these enzymes are synthesized by the ribosomes of the developing plastids. A different mechanism appears to be responsible for the development of activity of NADP-dependent triosephosphate dehydrogenase (EC 1.2.1.13) where the D-threo isomer gave 45% inhibition and the L-threo isomer gave 18% inhibition. Thus both specific (D-threo isomer) and unspecific (both isomers) inhibition occurred. It is suggested that the development of NADP-dependent triosephosphate dehydrogenase activity may result from the allosteric activation, in the plastids, of the NAD-dependent enzyme (Müller et al., 1969) which has been synthesized by cytoplasmic ribosomes. Neither isomer inhibited the development of five other enzymes of the photosynthetic carbon cycle namely ribosephosphate isomerase (EC 5.3.1.6), phosphoglycerate kinase (EC 2.7.2.3), triosephosphate isomerase (EC 5.3.1.1), tructosediphosphate aldolase (EC 4.1.2.13) and transketolase (EC 2.2.1.1), but there was a significant stimulation of the activity of transketolase by D-threo chloramphenicol.  相似文献

Chill-stressed or aged bean (Phaseolus vulgaris cv. 194) plants showed relative increase in the campesterol content of the primary leaves. Sitosterol/stigmasterol ratio declined significantly 24 h following transfer of chilled plants to warmer conditions. Sitosterol/stigmasterol varied markedly according to plant age, showing a decline during the leaf greening phase but an increase by the senescent (leaf-yellowing) phase.  相似文献

Leaf area expansion, dry weight, and water relations of Phaseolus vulgaris L. and P. acutifolius Gray were compared during a drying cycle in the greenhouse to understand the characteristics which contribute to the superior drought tolerance of P. acutifolius. Stomates of P. acutifolius closed at a much higher water potential than those of P. vulgaris, delaying dehydration of leaf tissue. P. acutifolius had a more deeply penetrating root system, which also contributes to its drought tolerance. Root-shoot ratios did not differ between the two species either under well watered or water stressed conditions. Leaf osmotic potential was also similar in the two species, with no apparent osmotic adjustment during water stress. These results indicate that P. acutifolius postpones dehydration and suggest that sensitive stomates and a deeply penetrating root system are characteristics which, if incorporated into cultivated beans, might increase their drought tolerance.  相似文献
