Summary Mitochondrial DNA has been isolated from paired lines of pearl millet maintainer and cytoplasmic male sterile plants. Summary In the selfed progeny of a spontaneously produced triploid interchange heterozygote four different double trisomic plants were observed. In all the plants the frequency of alternate orientation of multivalents was lower compared to their respective types in the sib single trisomic plants. The frequency of alternate co-orientation of the interchange complex in these trisomics was also reduced compared to that of parental euploid disomic interchange heterozygotes. It is suggested that the presence of extra chromosomes influences the orientation behaviour of higher associations in different trisomics. 相似文献

Summary The somatic karyotype of pearl millet Pennisetum americanum (L.) Leeke. (2n = 14) has been studied in several cultivars, but few cytological markers have been discovered which could help in the easy identification of the chromosomes. Analysis of pachytene bivalents permits such identification but is feasible only in a few cultivars. Recently, several lines having telocentric chromosomes have been produced and classified but their potentialities as cytogenetic tools have yet to be explored. Some African populations of pearl millet carry B-chromosomes in their karyotype. Cytogenetics of B-chromosomes has been reported in great detail. Bs undergo spontaneous changes to produce deficient- and iso-chromosomes. The main effect of B-chromosomes is on chiasma frequency which is exerted by the relative amounts of chiasma promoting euchromatin and the chiasma depressing heterochromatin in the Bs. Haploid plants occur occasionally and sometimes show a low degree of seed set, offering a possibility of establishing homozygous inbred lines. Cytogenetics of several spontaneous and induced autotetraploids have been reported. In general quadrivalent formation between the seven sets of four homologues was random. Seed set of the autotetraploids could be improved by selection; improved seed fertility was found to be associated with increased chiasma frequency, increased quadrivalent frequency and regular distribution of chromosomes at anaphase I. Genes controlling morphological characters of plant phenotype segregate independent of those controlling fertility and in pearl millet polyploidy per se is not limiting to plant vigour. Primary trisomics represent the best studied among the aneuploids of pearl millet. All the seven primary trisomics have been identified and described. Some were used in assigning genes to specific chromosomes but in general trisomies have poor vigour and fertility, and show low frequency of transmission. Apart from B-chromosomes, cytogenetics of interchanges has been the best studied aspect of pearl millet. The frequency of co-orientation of an interchange complex at metaphase I, which determines the fertility or sterility of the interchange heterozygote, is influenced by the genetic background and thus is theoretically amenable for selection leading to improved fertility of the heterozygote. Interchange tester-stocks have been assembled which can be used to identify the chromosomes involved in any newly obtained interchange. A complex interchange line involving all the chromosomes of the complement has also been produced, but the ring-of-fourteen produces total male and female sterility.Genotypic control of mitosis and meiosis has been reported, with reference to chromosome numerical mosaicism, multiploid sporocytes, desynapsis and chromosome fragmentation, and male sterility. Pearl millet being a largely outbreeding species, forced inbreeding was mainly found to result in loss of morphological vigour and reduction in mean chiasma frequency per PMC. Interspecific hybrids between pearl millet and several related species have been cytologically investigated and homology of the seven chromosomes of pearl millet with seven of the fourteen chromosomes of P. purpureum has been demonstrated. Cytogenetic evidence from haploids, autopolyploids and interspecific hybrids has indications to suggest that the haploid number of x = 7 is derived from x = 5, but the evidence is inconclusive and needs critical evaluation. 相似文献

The plastids in the cells of the tapetum in anther of Oenothera are involved in the development of male sterility (mst). We combined nuclear homozygosity for each of the two mst genes with the four different plastomes of Oenothera and demonstrated that in both cases the sterile anther phenotype is independent of the plastome. A pot experiment was carried out with pearl millet (Pennisetum americanum [L.] Leeke) growing in a sandy soil in which the upper (topsoil) and lower (subsoil) parts of the pots were separated by a perlite layer to prevent capillary water movement. Using microtensiometers a study was made to establish whether it was possible to measure hydraulic lift by which the upper part of the soil was rewetted when water was supplied exclusively to the lower part of the soil.Hydraulic lift occurred during the first seven days of the period of measurement, with a maximum water release to the soil of 2.7 Vol. % during one night (equivalent to 10.8 mL water in the top 10 cm of the soil profile). This magnitude was obtained at very high root length densities, so that water release from the roots would be expected to be much smaller under field conditions.Hydraulic lift ceased when the soil matric potential in the topsoil dropped below-10 kPa at the end of the light period and could not be re-established, neither by extending the dark period, nor after rewatering the topsoil. The disappearance of hydraulic lift could be explained in part through osmotic adaptation of plant roots and, thus prevention of water release from the roots in the topsoil. It is concluded that hydraulic lift may affect nutrient uptake from drying topsoil by extending the time period favourable for uptake from the topsoil. 相似文献

The orientation responses of the rice weevil, Sitophilus oryzae (L.) (Coleoptera: Curculionidae) as influenced by the olfactory stimulants emanating from either resistant or susceptible pearl millet, Pennisetum americanum, kernels were studied using a Y-tube olfactometer. The rice weevils were more attracted towards odours from millet grains than to blank controls and to susceptible entries more than to resistant entries, in general. However, there were exceptions. Odours from some resistant entries caused statistically the same, or even more positive orientations than those from some susceptible entries, and this suggests that the relative resistance of an entry and its olfactory attractiveness may not be strongly related, or that the resistance to infestation might be influenced by factors other than attractiveness. The partial damage of the millet kernel pericarp by abrasion slightly increased the orientational response of the weevils to odours from some susceptible and some resistant entries alike. 相似文献

Summary Reciprocal differences for male sterility, dwarfism and morphological traits have been studied in intra- and interspecific crosses of five Epilobium species. Male sterility occurred in two interspecific hybrids with E. montanum as the male parent while dwarfism has been found to varying degrees in three interspecific crosses with E. watsonni. In contrast to transient differences in plant height and leaf morphology in reciprocal hybrids of the cross between E. hirsutum and E. parviflorum, male sterility and dwarfism persistently occur as reciprocally different traits which may be influenced by determinants of the cytoplasm. The molecular characterization of the plastid DNA of the parental lines and the F1 hybrids indicate that the plastome of male sterile and dwarf plants is identical to that of the female parents. Furthermore, in spite of these developmental disturbances, the expression of plastid genes coding for polypeptides of thylakoid-membrane complexes is unchanged. Thus, it seems unlikely that the genetic compartement of the plastids is responsible for the expression of the male sterile or the dwarfed phenotype. 相似文献

Summary Embryogenic calli were derived from cultured segments of immature inflorescences of Pennisetum americanum (pearl millet). The original explants as well as the embryogenic calli and the plants regenerated via somatic embryogenesis were examined cytogenetically. Embryogenic calli were predominantly diploid (2n=14) after one month and six months in culture (92% and 76%, respectively). Tetraploid and aneuploid cells were observed in the original explant (2.5% and 1.2%) as well as in one (4.0% and 4.0%) and six-month-old calli (10.0% and 14.0%). Plants were regenerated from calli that had been in continuous culture for two, four and six months. Of the 101 regenerants, 100 were diploid and 1 was tetraploid. The tetraploid was an albino as were three of the diploid regenerants. Examination of 30 of the regenerants in meiotic diakinesis, anaphase I, anaphase II and quartet stages revealed no cytogenetic differences between control and regenerated plants. Gel electrophoresis for total protein content and alcohol dehydrogenase and malate dehydrogenase activity also did not reveal any differences between the controls and regenerants. The results of this study show that a slight shift toward aneuploidy and polyploidy may occur in embryogenic cultures, but there also is a strong selection in favor of plant regeneration from cytogenetically normal cells. 相似文献

Summary Outcrossing rates were estimated in three populations of the gynodioecious species Plantago coronopus by means of electrophoresis of adult plants and their natural progenies. A multilocus estimation procedure was used. Heterogeneity among the pollen-pool allele frequencies did not exist either in space of in time. Differences between populations in mean outcrossing rates were large (range: 0.34–0.93), probably caused by differences in densities of flowering plants. In addition, there was considerable variability between individuals, which was at least partly caused by the presence of male sterility. Population density may, via its influence on outcrossing rates, be a factor influencing the maintenance of male sterile plants in the population. The level of outcrossing in hermaphrodites was not low enough to explain the maintenance of male steriles. Outcrossing rates in two populations, established via progeny analysis, were much lower than calculated with the fixation index, possibly indicating heterozygote advantage in these natural populations.Grassland Species Research Group Publication no. 134 相似文献

Summary In a research program to transfer apomixis from Pennisetum squamulatum Fresen to pearl millet, P. americanum L. Leeke, a polyhaploid plant (2n=21) was discovered in the uniform open-pollinated progeny of an apomictic interspecific hybrid (2n = 41) between pearl millet and P. squamulatum. The polyhaploid was shorter, less vigorous and was smaller morphologically than its maternal parent. It probably originated by parthenogenetic development of a reduced gametophyte in the apomictic interspecific hybrid. The most common metaphase I chromosome association in the polyhaploid was 4 bivalents plus 13 univalents. Irregular chromosome distribution, tripolar spindles, bridges and fragments were observed at anaphase I and telophase I. The polyhaploid was male-sterile and partially female- fertile having multiple aposporous embryo sacs in 95% of the ovules. Seed set was 3% when open-pollinated and 33% when pollinated with pearl millet pollen. Low seed set was due to competition among multiple embryos developing in the same ovule. Seventeen progeny from seed produced under open-pollination on the polyhaploid each had 2n=21 chromosomes and were morphologically uniform and identical to the female parent. The expression of obligate apomixis in the polyhaploid conditioned by the P. squamulatum genome between the simplex and duplex condition indicates that apomictic reproduction is possible in nonpolyploid plants. 相似文献

Summary Nucleocytoplasmic determination of male sterility in Thymus vulgaris L. has been assumed in all papers attempting to explain the remarkably high frequencies of male steriles found in natural populations of this species. This paper provides strong evidence that both nuclear and cytoplasmic genes are involved in the determination of male sterility of this species, giving a complex inheritance. Interpopulation and intrapopulation crosses have shown that the ratio of females versus hermaphrodites among offsprings varied widely from 10 to 11. Furthermore, interpopulation crosses consistently yielded a higher frequency of females than intrapopulation crosses. Nucleocytoplasmic inheritance was demonstrated by an absence of male fertiles in backcrosses and asymmetrical segregation in reciprocal crosses. Molecular analysis of the mitochondrial DNA of some of the parents used in crosses suggested the involvement of different cytoplasms in the inheritance of male steriliy. 相似文献

ontogenic processes in the fertile and sterile microsporangia are compared, using samples collected from Chitou Experimental
Forest and Yeou-Shoei-Keng Clonal Orchard of the National Taiwan University, Nantou, Taiwan. The development of male strobili
occurred from August to the end of March. Microsporogenesis starts with the formation of the archesporium and ends with the
maturation of 2-celled pollen grains within the dehiscing microsporangium. Before meiosis, there was no significant difference
in ultrastructure between the fertile and sterile microsporangia. Asynchronous pollen development with various tetrad forms
may occur in the same microsporangium of either fertile or sterile strobili. However, a callose wall was observable in the
fertile dyad and tetrad, but not in the sterile one. After dissolution of the callose wall, the fertile microspores were released
into the locule, while some sterile microspores still retained as tetrads or dyads with intertwining of exine walls in the
proximal faces. As a result, there was no well developed lamellated endexine and no granulate ectexine or intine in the sterile
microspores. Eventually, the intracellular structures in sterile microspores were dramatically collapsed before anthesis.
The present study shows that the abortion in pollen development is possibly attributed to the absence of the callose wall.
The importance of this structure to the male sterility of T. cryptomerioides is discussed.

Summary Unlike tuberosum, andigena potato germ plasm exhibits a high degree of genetic variation in morphological, biochemical and reproductive traits. Sixty-five percent of the 565 genotypes comprising 145 accessions of Solanum tuberosum ssp. andigena obtained from Argentina, Bolivia, Chile, Colombia and Peru remain totally vegetative and never develop any floral bud when cultivated in northern India. In 18% of genotypes, the floral buds develop but they drop off from the plants prematurely. Thus, 83% of genotypes do not develop mature flowers. The frequency of such genotypes is maximum in the Bolivian genotypes. Whereas 17% of genotypes produce mature flowers, only 2% develop berries. The highest proportion of floral bud formation and their subsequent development and differentiation into mature flowers occur in Peruvian and Colombian genotypes. Partial to high male sterility occurs in 93% of the flowering genotypes; their pollen sterility ranges from 15% to 91%. Seven percent of the flowering genotypes are completely pollen sterile. The male sterility is expressed variously, ranging from structural to sporogenous types. The floral bud formation, its development and retention to maturity, pollen and ovule functionability and fruit development are under the control of a large number of genes, most of which are unlinked and independent. Summary Male sterility genes isolated in four inbred lines of pearl millet were found allelic. The differences between male fertile and male sterile phenotypes is mainly due to a single gene. Presence of a dominant gene (Ms) resulted in male fertility and double recessiveness (ms ms) in male sterility. However, genic male sterility (GMS) in Pennisetum is not a simply inherited case of monogenic recessive condition but is influenced by cytoplasmic and several nuclear factors. In a male sterile, the stage at which the male sterility gene is expressed during the development of the male gametophyte resulting in breakdown of the cells is influenced by cytoplasmic and other nuclear factors. Two types of cytoplasm, C-1 and C-2, are recognized. Presence of any two recessive male sterility alleles in C-1 led to breakdown of male development before differentiation of an archesporium in the anther (Arc-type); in C-2 cytoplasm, degeneration started during meiosis with fusion of meiocytes and syncyte formation (Syn-type), or at post-meiotic stages terminating in abortion of microspores before first pollen mitosis (PGM type). The triggering of activity of recessive male sterility genes in C-2 cytoplasm appeared to be regulated by two nuclear factors, R
1 and R
2 with duplicate gene action. Recessiveness for both the R factors in C-2 cytoplasm resulted in PGM-type expression. The action of R
1 and R

Summary Double-stranded RNA (dsRNA) was isolated from rice Oryza sativa ssp. japonica, but not from other subspecies. The dsRNA has been found in all of the examined cytoplasmic male-sterile (CMS) lines of BT (Chinsurah Boro II)-type rice, but was not detected in their companionate maintainer lines. It is uniquely and positivley correlated with the CMS trait in BT-type rice. Recently, the dsRNA was also found in a nuclear malesterile (NMS) rice, Nongken 58s, but was not found in its normal Nongken 58. The molecular weight of this dsRNA was estimated to be about 18 kb. Electron microscopic analysis reveals that it is linear snapped. The double strandedness of the RNA molecules was characterized by CF-11 cellulose column chromatography and nuclease treatments. It bound to CF-11 cellulose in the presence of 15% ethanol. It was sensitive to RNase A at low salt concentrations, but insensitive to DNase I, SI nuclease, and RNase A at high salt concentrations. The dsRNA was detected in both mitochondrial and cytoplasmic fractions. Dot-blot hybridization reveals that there is no sequence homology between this dsRNA and mtDNA, but there is homology between this dsRNA and nuclear genomic DNA. Background
We previously cloned the Ssp411 gene. We found that the Ssp411 protein is predominantly expressed in elongated spermatids in the rat testis in a stage-dependent manner. Although our findings strongly suggested that Ssp411 might play an important role in mammalian spermatogenesis, this hypothesis has not been studied.Methods
We first used real-time PCR, Western blotting and immunohistochemistry to confirm that the expression pattern of Ssp411 in several murine tissues is similar to its expression pattern in corresponding rat tissues. To better understand the roles of Ssp411 in male reproduction in vivo, we identified and characterized an Ssp411 expression-disrupted murine strain (Ssp411PB/PB) that was generated by piggyBac (PB) transposon insertion. We studied Ssp411-interacting proteins using proteome microarray, co-IP and GST pull-down assay.Results
Both Ssp411 mRNA and protein were detected exclusively in spermatids after step 9 during spermiogenesis in testis. Phenotypic analysis suggested that only Ssp411PB/PB males are sterile. These males have smaller testes, reduced sperm counts, decreased sperm motility and deformed spermatozoa. Microscopy analysis indicated that the manchette, a structurally reshaped sperm head, is aberrant in Ssp411PB/PB spermatids. The results of proteome microarray analysis and GST pull-down assays suggested that Ssp411 participates the ubiquitin-proteasome system by interacting with PSMC3. This has been reported to be manchette-associated and important for the head shaping of spermatids.Conclusions
Our study suggested that Ssp411 is required for spermiogenesis. It seems to play a role in sperm head shaping. The lack of Ssp411 causes sperm deformation and results in male infertility.General significance
Ssp411PB/PB mouse strain is an animal model of idiopathic oligoasthenoteratozoospermia (iOAT), and the gene may represent a therapeutic target for iOAT patients. 相似文献18.

Summary Forty-three percent of the progeny from 4x × 2x crosses [Group Tuberosum cultivar x diploid hybrid (Groups Phureja or Stenotomum x Group Tuberosum haploid)] were male sterile. In contrast only four percent of the progeny from the reciprocal crosses were male sterile. Male sterility among the former progeny is presumed to result from the interaction of Tuberosum cytoplasm with dominant genes from the cultivated diploids, Groups Phureja and Stenotomum, an interaction known to occur in crosses of Tuberosum haploid x cultivated diploid species (2n = 2x = 24). The frequency of fertile progeny from the 4x × 2x crosses (57%) was significantly higher than that from the 2x × 2x crosses (Tuberosum haploid x cultivated diploid), (28%). The frequency of male fertility among progeny from different cultivars in 4x × 2x crosses varied from 31–82 percent. The difference between cultivars strongly suggests that some cultivars may have dominant male fertility restorer genes. 相似文献

A plant with a mutant phenotype was observed in a Longittee cultivar. The plant was late in maturity, had white-translucent anthers, and was male sterile. The inheritance of this mutant was studied in a cross involving the mutant and the mother parent and their F1, F2, F3 and BC1F1 generations. The results suggested that the sterile character was genetic and due to a recessive gene. 相似文献
