The phylogenetic relationships of 27 north-eastern Atlantic and Mediterranean blennioids are analysed based on a total of 1001 bp from a combined fragment of the 12S and 16S mitochondrial rDNA. The most relevant results with implications in current blenniid taxonomy are: (1) Lipophrys pholis and Lipophrys (=  Paralipophrys ) trigloides are included in a well-supported clade that by the rule of precedence must be named Lipophrys ; (2) the sister species of this clade are not the remaining species of the genus Lipophrys but instead a monotypic genus comprising Coryphoblennius galerita ; (3) the smaller species of Lipophrys were recovered in another well-supported and independent clade, which we propose to be recognized as Microlipophrys ; (4) although some authors included the genera Salaria and Lipophrys in a single group we have never recovered such a relationship. Instead, Salaria is more closely related to the genera Scartella and Parablennius ; (5) the genus Parablennius , which was never recovered as a monophyletic clade, is very diverse and may include several distinct lineages; (6) the relative position of Aidablennius sphynx casts some doubts on the currently recognized relationships between the different blenniid tribes. Meristic, morphological, behavioural and ecological characters support our results and are also discussed. The Mediterranean turbot Psetta maxima consists of two main genetically distinct lineages (western Mediterranean and 'eastern secluded Mediterranean' basins) as investigated by mitochondrial DNA analysis. Within the latter lineage, most haplotypes from the Sea of Azov were endemic and more than half of them derived from a single ancestral haplotype shared among all the eastern Mediterranean areas. There was no relation between morphotype variation in bony tubercles and mitochondrial genealogy.  相似文献

The phytochemical system of mustard-oil glucosides (glucosinolates) accompanied by the hydrolytic enzyme myrosinase (beta-thioglucosidase), the latter usually compartmented in special myrosin cells, characterizes plants in 16 families of angiosperms. Traditional classifications place these taxa in many separate orders and thus imply multiple convergences in the origin of this chemical defense system. DNA sequencing of the chloroplast rbcL gene for representatives of all 16 families and several putative relatives, with phylogenetic analyses by parsimony and maximum likelihood methods, demonstrated instead a single major clade of mustard-oil plants and one phylogenetic outlier. In a further independent test, DNA sequencing of the nuclear 18S ribosomal RNA gene for all these exemplars has yielded the same result, a major mustard-oil clade of 15 families (Akaniaceae, Bataceae, Brassicaceae, Bretschneideraceae, Capparaceae, Caricaceae, Gyrostemonaceae, Koeberliniaceae, Limnanthaceae, Moringaceae, Pentadiplandraceae, Resedaceae, Salvadoraceae, Tovariaceae, and Tropaeolaceae) and one outlier, the genus Drypetes, traditionally placed in Euphorbiaceae. Concatenating the two gene sequences (for a total of 3254 nucleotides) in a data set for 33 taxa, we obtain robust support for this finding of parallel origins of glucosinolate biosynthesis. From likely cyanogenic ancestors, the "mustard oil bomb" was invented twice.  相似文献

The phylogenetic relationships among threePseudorasbora fishes (Cyprinidae, Sarcocheiichthyinae) occurring in Japan (P. parva, P. pumila pumila andP. pumila subsp. sensu Nakamura [1963]) were inferred from nucleotide sequences of the mitochondrial 16S rRNA gene. The sequences. of 1240 bp, were determined and compared for 22 specimens from 2–8 populations for each taxon, with a singlePungtungia herzi specimen as an outgroup. A total of 171 sites (13.8%) were variable among the specimens, but only 0–2 sites within each population. The phylogenetic relationships estimated by neighbor-joining, maximum-parsimony and maximum-likelihood methods confirmed a sister relationship between the twoP. pumila subspecies, with a high level of confidence. However, their genetic distinction from each other (4.1±0.4SD % sequence difference on average) was at a level similar to that between them andP. parva (5.9±0.5%). The geographic distribution of the twoP. pumila subspecies, which are separated by the Fossa Magna region, suggests that the genetic divergence of the two subspecies originated from a vicariant process separating the freshwater ichthyofaunas of eastern and western Honshu.Pseudorasbora parva populations were divided into two genetic groups (1.8±0.2% sequence difference), one group comprising continental and part of the Japanese populations, and the other the remaining Japanese populations. This suggests that at least two genetically divergent lineages had been originally distributed in Japan, but a strong possibility remains that the present situation has resulted from artificial transplantation.  相似文献

Higher-level snake relationships are inferred from sequence analyses of one nuclear gene (C-mos) and three mitochondrial genes (12S rRNA, 16S rRNA and cytochrome b). Extant snakes belong to two lineages: the fossorial Scolecophidia, which feed on small prey on a frequent basis, and the ecologically diverse Alethinophidia ('typical' snakes), which feed on large prey on an infrequent basis. The vast majority of Alethinophidia, if not all of them, belong to two clades, corresponding to two distinct prey neutralization modes: unimodal constriction for the Henophidia (locomotor and feeding systems coupled) and injection of toxic saliva, in addition (or not) to diverse alternate modes of constriction, for the Caenophidia (locomotor and feeding systems uncoupled). Within Alethinophidia, non-macrostomatan (small gape) Aniliidae (genus Anilius) and macrostomatan (large gape) Tropidophiidae (genera Trachyboa and Tropidophis), both from the Neotropics, are closest relatives. Although our data are insufficient to robustly infer the ancestral mode of life of snakes, we find evidence of plasticity in the basic ecological and trophic modes of snakes. Consequently, the macrostomatan condition should not be treated a priori as a derived character state devoid of homoplasy.  相似文献

The Order Stolidobranchiata comprises the families Pyuridae, Styelidae and Molgulidae. Early molecular data was consistent with monophyly of the Stolidobranchiata and also the Molgulidae. Internal phylogeny and relationships between Styelidae and Pyuridae were inconclusive however. In order to clarify these points we used mitochondrial and nuclear sequences from 31 species of Styelidae and 25 of Pyuridae. Phylogenetic trees recovered the Pyuridae as a monophyletic clade, and their genera appeared as monophyletic with the exception of Pyura. The Styelidae, on the other hand, appeared as a paraphyletic group split into several clades. One of them was formed by solitary oviparous species, of which the Pyuridae were a sister group. A second clade included the colonial genera Botryllus, Botrylloides and Symplegma. The remaining colonial and solitary genera formed several poorly resolved clades. One of the more species genus, Polycarpa, was shown to be polyphyletic, and the species Styela plicata grouped into two genetically distant clades suggesting the existence of two cryptic species. The internal phylogeny of Styelidae has bearings on the origin of coloniality in this family. We suggest to abandon the traditional division of colonial forms into social and compound species and use instead the categories of aggregated colonies that do not have common vascular systems, and integrated colonies, that do possess such systems. Our molecular results indicate that there have been several independent acquisitions of coloniality in the Styelidae, and that viviparity may be a pre-adaptation for a colonial life-style.  相似文献

To study the phylogeny of Hynobiidae, we amplified DNA fragments of 470 bp 16S ribosomal RNA (16S rRNA) gene on mitochondrial DNA from Ranodon sibiricus and Ranodon tsinpaensis. PCR products were cloned into PMD18 T vector after purification. These sequences were determined and deposited in the GenBank (accession numbers: AY373459 for Ranodon sibiricus, AY372534 for Ranodon tsinpaensis). By comparing the nucleotide differences of 16S ribosomal RNA sequences among Liua shihi, Pseudohynobius flavomaculatus and Batrachuperus genus from GenBank database, we analyzed the divergences and base substitution among these sequences with the MEGA software. The molecular results support that B. tibetanus, B. pinchonii and B. karlschmidti are classified into three valid species. Liua shihi has closer phylogenetic relationships to Ranodon tsinpaensis than to other species. More our results reveal that Pseudohynobius flavomaculatus is not a synonym of Ranodon tsinpaensis. 文章测定了中国分布的蜜蜂属(Apis)5种蜜蜂22个样本的线粒体基因ND2、CO2、16S rRNA以及核基因ITPR的序列,对序列的碱基组成和蜜蜂种间的遗传距离进行了分析。结合下载的蜜蜂属其他4个种的相关序列,采用最大简约法、邻接法和最大似然法重建了蜜蜂属系统发育关系。系统发育分析结果支持蜜蜂属划分为3个类群,即小蜜蜂类群(包括小蜜蜂和黑小蜜蜂)、大蜜蜂类群(包括大蜜蜂和黑大蜜蜂)和穴居蜜蜂类群(西方蜜蜂、东方蜜蜂、沙巴蜂、苏拉威西蜂、绿努蜂),且小蜜蜂类群较早分化。结果还显示,我国海南岛的大蜜蜂和大陆的大蜜蜂之间可能存在较大的遗传分歧。  相似文献

AIMS: The study aimed to provide characterization of a potential new species of Coxiella, identified following a series of outbreaks of disease in Australian native freshwater crayfish. METHODS AND RESULTS: PCR primers designed for amplification of Coxiella burnetii genes including 16S rDNA, com1 and sodB were used to amplify homologues in the Coxiella-like crayfish pathogen. Products were then cloned and sequenced. The organism demonstrated a high degree of sequence homology in the highly conserved 16S rDNA (96%) and sodB (99%) genes, as well as the Coxiella sp. specific com1 (100%) gene. Regions flanking the sodB coding sequence demonstrated homology to C. burnetii antioxidant AhpC/Tsa family protein and dihydrodipicolinate reductase gene. CONCLUSIONS: The degree of homology between the genes selected and flanking regions suggested the two organisms were sufficiently closely related to belong to the same genus. SIGNIFICANCE AND IMPACT OF THE STUDY: This study provided evidence for a potential new species in the currently monospecific genus Coxiella, with the only described member being C. burnetii, a category B biological warfare agent.  相似文献

Some novel members of extremely halophilic archaea, strains AJ 11, AJ 12 and AJ 13, were isolated from the Aularz Lake located in the Altun Mountain National Nature Reserve of Xinjiang, Uygur Autonomous Region in China. Partial DNA fragments encoding a bacteriorho-dopsin (BR), as well as for 16S rRNA of isolated strains, were amplified by PCR and their DNA sequences were determined subsequently. On the basis of homology and phylogenetic analysis of the 16S rDNA, we thought that the isolated strains forming a microbiological population are the members of the genus Natrinema. The results of genetic analysis, such as GC content, transition/transver-sion (Ti/Tv) rate ratios and synonymous substitution rates (Ks) indicate that the br fragments, with a high level of genetic divergence, are faced with both purifying selection and bias mutation pressure. We assessed the mitochondrial DNA sequence divergence of a 718 bp fragment of the control region and 1007 bp of the cytochrome b gene between two allopatric morphologically different subspecies of bluethroat ( Luscinia svecica ). None of the 17 total haplotypes was shared between L. s. namnetum and L. s. svecica . However, the mean distances between subspecies were very low for both fragments (0.00168 ± 0.00001 (mean ± SE) for the control region; 0.00306 ± 0.00016 for the cytochrome b gene). Only one substitution made the two subspecies genetically differentiated, highlighting their recent divergence. Interestingly, the control region was not more variable than the cytochrome b gene.  相似文献

With about 60,000 described species, Curculionoidea represent the most species-rich superfamily in the animal kingdom. The immense diversity apparently creates difficulties in the reconstruction of the phylogenetic relationships. Independent morphological studies have led to very different classifications. This study is based on molecular data from two independent molecular sources, the 16S and 18S rDNA. Sensitivity analyses were conducted for the sequence alignment (gap costs were varied) as well as the phylogenetic reconstruction algorithms and some of their parameters. The higher-level relationships reconstructed within Curculionoidea are sensitive to alignment and reconstruction method. Nemonychidae or Oxycorynidae+Belidae were found to be sister to all remaining Curculionoidea in many analyses. The 16S rDNA sequence data (obtained from 157 species) corroborate many tribes and genera as monophyletic. It is observed that the phylogenetic reconstruction of genera with specific genetic features such as polyploidy and parthenogenetic reproduction is difficult in weevils. The curculionid subfamily Lixinae appears monophyletic. A new monophylum consisting of Entiminae, Hyperinae, Cyclominae, Myllorhinus plus possibly the Cossoninae is distinguished and we call it Entiminae s.l. For most other subfamilies and families homoplasy concealed the phylogenetic signal (due to saturation of the 16S sequences), or the species sampling was insufficient, although our sampling scheme was rather broad. We observed that although data from one source can easily be misleading (16S) or hardly informative (18S), the combination of the two independent data sets can result in useful information for such a speciose group of organisms. Our study represents the most thorough analysis of molecular sequence data of the Curculionoidea to date and although the phylogenetic results appear less stable than expected, they reflect the information content of these sequence data realistically and thus contribute to the total knowledge about the phylogeny of the Curculionoidea.  相似文献

Selection pressure by natural enemies on phytophagous insect larvae is intense and has frequently triggered the evolution of chemical defence as an effective counterstrategy. In the chrysomelid subfamily Galerucinae, glandular structures and defensive fluids have been described for the tribe Sermylini Wilcox, 1965. Previous morphological and ultrastructural studies raised doubts that these defensive devices in Sermylini can be traced back to a common origin. The taxonomy of the Galerucinae cannot clear these doubts because the phylogeny of this taxon is a matter of current debate. We therefore investigated the phylogeny of the Galerucinae based on approximately 1740 bp of the mitochondrial 12S and 16S rRNA and the nuclear elongation factor 1 alpha genes. Our data support the hypothesized close relationship between the subfamilies Galerucinae and Alticinae, yet, by contrast to other recent analyses, the two groups are mostly resolved as monophyletic sister groups or, in some analyses, with the Galerucinae nested paraphyletically within the Alticinae. Within the subfamily Galerucinae, only the tribe Galerucini formed a monophyletic taxon, except for one species, Cerochroa brachialis Stal, 1858. In none of our analyses were the Sermylini recovered as a monophyletic tribe. However, our data support monophyly of each of the three groups within the Sermylini that have morphologically distinguishable larval defensive openings. We conclude that the defensive structures in larvae of Sermylini have no common origin, but evolved independently. The phylogenetic relationships of the West Mediterranean lineages of the subtribe Scaritina have been investigated by studying the mitochondrial cytochrome oxidase gene (COI) and the mitotic and meiotic chromosomes, including the localization of rDNA genes by fluorescence in situ hybridization. These sources of data are congruent and suggest the following conclusions: (1) the subgenus Parallelomorphus (genus Scarites ) is a monophyletic group according to molecular and karyotypic data, in agreement with its geographical distribution and morphological characters; (2) in the same genus, the subgenus Scallophorites seems to be monophyletic but includes two well-separated lineages – one represented by Scarites buparius (Forster, 1771) and Scarites occidentalis (Bedel, 1895) (both with multiple sex chromosomes), and one represented by Scarites hespericus (Dejean, 1831); (3) the nominal subgenus Scarites s.s . represented by Scarites eurytus (Fischer, 1828), is cladistically more closely related to subgenus Scallophorites than to subgenus Parallelomorphus ; (4) Distichus planus (Bonelli, 1813) belongs to a distinct clade that is well separated from Scarites, and shows more plesiomorphic states of COI sequence and chromosome number. In addition, the presence of two autapomorphies, such as rDNA sites located in the X chromosome, and an A + T rich intergenic spacer of about 120 bp between the stop codon of the COI gene and the tRNAleu, agree with the ranking of Distichus as a separate genus from Scarites .  相似文献

We used sequence variation within 297 bp of control region mitochondrial DNA (mtDNA) amplified from 53 lesser long-nosed bats, Leptonycteris curasoae (Phyllostomidae: Glossophaginae) captured at 13 locations in south-western United States and Mexico and one site in Venezuela to infer population structure and possible migration routes of this endangered nectar- and fruit-eating species. Phylogenetic analysis using maximum parsimony and UPGMA confirmed species and subspecies distinctions within Leptonycteris and revealed two clades exhibiting 3% sequence divergence within the Mexican subspecies, L. c. yerbabuenae . Even though many roosts contained L. c. yerbabuenae from both clades, weak population structure was detected both by a correlation between genetic differentiation, F st, and geographical distance and by a cladistic estimate of the number of migration events required to align bat sequences with geographical location on maximum parsimony, as compared to random, trees. Three results suggest that L. c. yerbabuenae are more likely to migrate between sites along the Pacific coast of Mexico or along the foothills of the Sierra Madre Occidental than between these regions. (1) Seventeen of 20 bats which shared an identical sequence were captured up to 1800 km apart but within the same putative migration corridor. (2) Residuals from a regression of F st on distance were greater between than within these regions. (3) Fewer migration events were required to align bats with these two groups than expected from random assignment. We recommend analysing independent genetic data and monitoring bat visitation to roost sites during migration to confirm these postulated movements.  相似文献
