细脚拟青霉田间分离菌株间的异核现象 总被引:3,自引:0,他引:3
本文报道细脚拟青霉(paecilomyces tenuipes)不同田间分离菌株单孢子后代间的异核现象。用来自荣园、菜地、水稻田三种生境的四个菌株(803、2801、1401和3101)的单孢子培养后代,在加有酵母膏和麦芽糖的改良萨氏培养基上进行配接实验,仅在803与1401两菌株间能形成异核体,其频率为13.5%。在配接实验中两亲和菌落交界处长出白色致密的菌丝簇组成的实线可推测为异核体。从来自菌丝簇组线的每个单菌丝尖端培养物中分离出30个以上的单孢子,井分别移接到 PDA 平板上。将来源于单孢子的菌落与两亲本菌落进行比较,有三个单菌丝尖端培养物(C_3、B_3、F_3)重现了两亲本类型或出现了新的菌落类型,从而异核现象得到证实。 相似文献

用 Giemsa 染色观察了细脚拟青霉(Paecilomyces tenuipes)803和1401菌株间的菌丝联结现象,观察了菌丝、孢子的细胞核数目。孢子均是单核,菌丝细胞有1—2个核,多数为单核,菌丝尖端有1—6个核。毒力测定结果表明:能形成异核体的亲和单孢子菌株组合803-17和1401-9、803-3和1401-8的混合培养物毒力与未经培养的相同菌株组合的菌丝体混合物毒力显著不同。在讨论申提出这种差异的原因是:在 Sm 液体培养中,混合接种803-17和1401-9、803-3和1410-8,经11天的混合培养后,混合培养物中形成了与核体亲本菌丝(803-17、1401-9、803-3、1401-8)毒力显著不同的异核菌丝体(803-17×1401-9)、(803-3×1401-8)。 相似文献

本文观察了细脚拟青霉人工培养的几种固体培养基及其组合。认为大麦、小麦以及组合培养基中的麸皮+玉米面+谷糠(2:1:1)较合适。通常该菌采收期以30—40天为好,但氨基酸含量40—50天最高,60天以后下降。以天然冬虫夏草cordyceps sinensis作对照,此菌栽培产物通过分析表明,两者主要化学组分相似,所含氨基酸种类亦相同,但是多种氨基酸却高出冬虫夏草。 相似文献

对斜链拟青霉胞外多糖进行体外清除自由基活性研究。利用醇沉法从斜链拟青霉发酵液中醇沉获得斜链拟青霉胞外多糖,经DEAE-纤维素-52柱层析后得到1个主峰P3和2个小峰P1与P2,对主峰P3进行Sephadex G-100柱层析,表明P3为纯化物; 应用DPPH-酶标法和化学发光法分别对P3进行清除DPPH·、·OH和O-2·能力测试。结果表明:P3对DPPH·、·OH和O-2·均具有明显的清除能力,且与浓度呈量效关系,IC50分别为0.083、0.121和0.214 mg/mL。 相似文献

用二苯基苦基苯肼自由基酶标仪法,对业热带蔷薇科常见的50种木本植物鲜叶的自由基清除活性进行了比较,发现不同属、不同种树木鲜叶的80%甲醇提取物的自由基清除活性有很大差异,其中苹果属5种植物在相当于鲜叶浓度为0.5mg/mL于37℃下孵育20min时,对0.5mmol·L-1DPPH自由基平均清除率达62.4%,而绣线菊属3种植物的平均自由基清除率仅11.6%.尖嘴林檎、棣棠、木瓜、三叶海棠、湖北海棠、木香花、小果蔷薇和黄山花楸等鲜叶有较强的自由基清除活性,它们在浓度为0.5mg/mL时的自由基清除率分别可达85.0%、75.8%、70.7%、69.6%、64.5%、62.5%、61.8%和61.3%,显示其有较大的开发潜力. 相似文献

Bioassay-guided fractionation of the marine-derived endophytic fungus Paecilomyces variotii resulted in the isolation of two new butenolides, namely, butyrolactone IX (1) and aspulvinone O (7), together with eight known related congeners, butyrolactones I, IV, V, and VI (2–5), aspernolide A (6), and aspulvinones H, C, and D (8–10). Their structures were elucidated on the basis of detailed spectroscopic analysis and by comparison with literature data. All of the isolated butenolides were tested for their activity against DPPH radicals and the results showed that butyrolactones (1–6) possessed potent activity with IC50 values ranging from 38.0 to 186.3 μM, while aspulvinones (7–10) exhibited significant activities with IC50 values ranging from 11.6 to 29.4 μM, which are stronger than that of the positive control BHT (with IC50 117.7 μM). 以生长在长江滩地三种类型(江滩、洲滩、湖滩)上、三种水淹状况下2个品系的人工林杨树木材(欧美杨无性系72杨、美洲黑杨无性系69杨)木材为研究对象,用二苯基苦基苯肼自由基(DPPH)酶标仪法,对其80%甲醇提取物的自由基清除活性进行了比较,发现相同立地上杨树边材和心材提取物的自由基清除活性存在显著差异;且边材提取物的自由基清除活性普遍较高.不同立地上杨树心材提取物的自由基清除活性差异不显著;但边材提取物的自由基清除活性存在较大的差异,其中水淹深度为1 m和3 m的杨树边材提取物自由基清除活性差异达到极显著水平.相关性分析表明,心材提取物的IC50与水淹深度呈显著的负相关,边材提取物的IC50与水淹深度呈极显著的正相关. 相似文献

用二苯基苦基苯肼自由基酶标仪法,对常见的60余种裸子植物鲜叶的自由基清除活性进行了比较.发现不同科属,不同树种鲜叶的80%甲醇提取物的自由基清除活性有很大差异,其中罗汉松科罗汉松属树木鲜叶在浓度在0.5mg/mL,于37℃下孵育20min时的自由基清除率平均可达55.9%,而柏科侧柏属平均仅为7.1%;罗汉松,孔雀柏、小叶罗汉松、湿地松、杉木和刺柏等鲜叶有很强的自由基清除活性,它们的自由基清除率分别可达61.1%,58.4,57.6%,57.1%,51.8%和50.4%,表明这些树种有较大的开发潜力。 相似文献

研究用二苯基苦基苯肼自由基酶标仪法,对常见的三十余种单子叶木本植物鲜叶的自由基清除活性进行了比较,发现不同科属、不同种植物鲜叶的80%甲醇提取物对DPPH自由基的清除活性有很大差异,其中供试的棕榈科7种植物鲜叶提取物,在相当于鲜叶浓度为2.5mg/ml于37℃下孵育20min时,对0.5mmol/L DPPH自由基清除率平均可达40.0%,而龙舌兰科5种植物平均仅为7.2%;刺葵、棕竹、筋头竹、蒲葵和棕榈等鲜叶有较强的自由基清除活性,它们在相当于鲜叶浓度为2.5mg/ml时的自由基清除率分别可达83.1%、79.2%、64.9%、60.5%和51.3%。这些树种有一定的开发潜力。 相似文献

AIMS: In the present study, two different optimization techniques were used to determine the suitable operating parameters for exo-biopolymer production in submerged mycelial cultures of two entomopathogenic fungi Paecilomyces japonica and Paecilomyces tenuipes. METHODS AND RESULTS: First, the rotating simplex method, a nonstatistical optimization technique, was employed to obtain the best combination of physical parameters (viz. pH, agitation intensity, aeration rate) for maximum exo-biopolymer production by P. japonica in a batch bioreactor. The optimal combination was determined to be a pH of 8.06, an aeration of 3 vvm, without any impeller agitation, producing a 17-time increase in exopolymer production (34.5 g l(-1)) when compared with that achieved in unoptimized flask cultures. Second, the uniform design method, a statistical optimization technique, was employed to determine the best operating parameters for submerged culture of P. tenuipes. The optimal combination for mycelial growth was determined to be a pH of 4.88, an aeration of 2 vvm and an agitation of 350 rpm, while a pH of 4, an aeration of 2 vvm and an agitation of 150 rpm was best for exo-biopolymer production. CONCLUSIONS: The exo-biopolymer production in P. japonica optimized by the rotating simplex method was strikingly improved (max. 34.5 g l(-1)), and the exo-biopolymer production in P. tenuipes optimized by the uniform design method was also significantly increased (max. 3.4 g l(-1)). SIGNIFICANCE AND IMPACT OF THE STUDY: The successful application of these two different optimization techniques in this study implies that these methods are worthy of applying to other fermentation systems for the production of bioactive mycelial biomass and exo-biopolymers in liquid culture of higher fungi. 相似文献

选择不同来源的12株球孢白僵菌,分别测定菌丝体2种溶剂提取物在不同抗氧化模型中的抗氧化作用。结果表明,不同溶剂提取物对二苯基苦味酰基苯肼自由基(DPPH)和羟自由基(.OH)均有清除作用,但是清除自由基的活性不同,乙酸乙酯提取物对.OH的清除作用最为显著,活性较高,最高清除率达到71.54%。甲醇提取物活性均较低,30 min时甲醇提取物对DPPH和.OH清除率最高分别为36%和38.96%,12株白僵菌间的清除自由基活性有较大的差异。 相似文献

The objectives of this study were to determine phenolic content and antioxidant activities of chloroform, acetone, methanol and hot water extracts of Bauhinia vahlii leaves. The hot water extract afforded the highest yield (6.3%) while the lowest yield was obtained from the chloroform extract (2.1%). The methanol extract contains higher levels of total phenolics (48.7 ± 0.7 g GAE/100 g extract), tannins (21.7 ± 0.7 g GAE/100 g extract) and flavonoids (10.3 ± 0.2 RE/100 g extract). The extracts were subjected to assess their antioxidant potential using various in vitro systems such as DPPH, ABTS+, FRAP, OH, β-carotene linoleic acid bleaching system, phosphomolybdenum reduction and Fe2+ chelation. It is concluded that the methanolic extract of B. vahlii leaves have strong antioxidant potential. Further study is necessary for isolation and characterization of the active antioxidants, which may serve as a potential source of natural antioxidants. 相似文献
