对甘蓝根肿病菌生物学特性研究表明,该菌休眠孢子萌发的最适温度24℃,最适pH值6.0-6.7,致死温度45℃,肿根腐烂处理可以显著提高萌发率,光对休眠孢子萌发有明显抑制作用。该菌休眠孢子在感病寄主的根分泌物溶液中萌发率最高,达75%,耐病甘蓝品种及番茄的根分泌物均能刺激休眠孢子萌发。通过电镜观察,根肿病菌休眠孢子为近球形,孢壁有乳状突起,直径2.1-3.1μm(平均直径2.5μm)。游动孢子为近球形或椭圆形,大小为1.6-3.6μm。同侧着生不等长尾鞭式双鞭毛。  相似文献

为明确香蕉根系分泌物对枯萎病菌及其生防枯草芽孢杆菌的生物效应,采用离位溶液培养法收集抗枯萎病香蕉品种(南天黄)和感枯萎病香蕉品种(桂蕉6号)的根系分泌物,研究根系分泌物对土壤微生物种群数量、香蕉枯萎病菌和枯草芽孢杆菌生长的影响。结果表明: 抗病品种根系分泌物能显著减少土壤真菌的数量,抑制枯萎病菌孢子的萌发;而感病品种根系分泌物能显著促进病菌菌丝生长和孢子的萌发,两个品种根系分泌物均能显著促进枯草芽孢杆菌的生长和生物膜形成。经抗(感)病香蕉品种根系分泌物处理,病菌菌丝生长速率分别为11.28和12.28 mm·d-1,病菌孢子的萌发率分别为34.6%和79.5%;枯草芽孢杆菌培养12 h后菌体生长量的OD600分别为1.27和1.14,生物膜形成量在静置培养72 h后OD570分别达1.11和1.30,两个品种处理间的差异均达显著水平。枯草芽孢杆菌在香蕉感病品种根际中定殖的菌量显著高于抗病品种。通过对两个品种根系分泌物中可溶性总糖、游离氨基酸和有机酸的含量和组成分析,发现抗病品种根系分泌物中有机酸和游离氨基酸的含量明显高于感病品种,在各组成成分中,以乙酸和脯氨酸在抗(感)病香蕉品种根系分泌物中含量比值较高,分别达3.7和2.4倍。综上所述,抗病品种根系分泌物能抑制病菌生长,感病品种根系分泌物则会显著促进病菌生长,而两个品种根系分泌物均能显著促进枯草芽孢杆菌的生长和生物膜的形成。  相似文献

以沈阳地区芹菜斑枯病菌的纯培养菌株为试材,通过对菌丝生长、孢子萌发的营养及环境条件的多因子试验,明确了芹菜斑枯病菌菌丝生长以在CDAMS—VI培养基上最好,其次是CSA、CCSA和PDA,菌丝生长及产孢对碳源、氮源种类有明显的选择性。菌丝生长的最适pH值为4~5,最适温度为21℃,致死温度为39℃(处理40min),光照明显抑制菌丝生长。病菌分生孢子在水和芹菜汁中不能萌发,在水琼脂上萌发最好;孢子萌发的适宜pH值为4.6~6.6,适温为17—25℃,致死温度为42℃(处理20min),RH100%最适宜孢子萌发。光照和变温处理可促进分生孢子器的形成,病菌在PDA上从孢子萌发到分生孢子器形成并释放出分生孢子需要16~18d。  相似文献

本文研究碳源、氮源、温度、湿度、pH值和光照等对蝉拟青霉LB菌株生长、产孢和孢子萌发的影响.结果表明,适合该菌株菌落生长和产孢的最佳碳源是可溶性淀粉和蔗糖,最佳氮源为蛋白胨;菌丝生长和孢子萌发的最适温度范围是25℃~27℃,产生分生孢子的最适温度是25℃;分生孢子萌发所需湿度范围是RH 90%~100%,当RH低于90%时很难萌发;在pH值4~10的范围内该菌能生长和产孢,菌丝生长最适pH为6,产生分生孢子和孢子萌发最适pH范围为6-7;光照处理对该菌产孢有一定的影响;分生孢子的致死条件为55℃ 10min.生物学特性显示,蝉拟青霉LB菌株是一株对营养要求不高、对环境适应能力较强的昆虫病原真菌.  相似文献

为确定萼花臂尾轮虫休眠卵的最适形成条件 ,或为提高特定培养条件下形成的休眠卵的萌发率 ,对不同温度、食物、光照和 pH条件下萼花臂尾轮虫形成的休眠卵进行了萌发研究。发现不同培养条件下形成的休眠卵在不同萌发条件下的萌发历时均为实验开始后的第 1~ 7天 ,萌发率高峰均出现在第 1~ 3天。在萌发温度为15℃~ 30℃范围内 ,2 0℃下形成的休眠卵在 2 0℃的萌发温度下累积萌发率最高 ,为 5 2 5 0± 6 89% ;母体以小球藻为食物时 ,其所产休眠卵的平均累积萌发率为 2 7 92 % ,显著大于以斜生栅藻或蛋白核小球藻和斜生栅藻所组成的混合藻为食物时 ;休眠卵形成和萌发时的光照条件对其累积萌发率无显著的影响 ;pH6 5和 7 5条件下形成的休眠卵的累积萌发率较高 ,分别为 32 5 0± 7 0 7%和 38 75± 15 5 3%。本研究和已报道的研究结果表明 :不同培养条件下形成的休眠卵在不同条件下萌发时均呈同步萌发型式 ;2 0℃和pH7 5分别是武汉东湖萼花臂尾轮虫休眠卵形成的最适温度和最适 pH条件。  相似文献

用组织培养法和光学显微镜技术初步研究光照和温度影响扇蕨孢子萌发的结果表明,扇蕨孢子发芽是需光型,具有明显的光休眠现象。光照是孢子萌发的主要影响因子,25℃下,孢子萌发率达(85.1±5.1)%。相同温度下孢子即使在黑暗中培养50d也不能萌发,转入光照下后萌发率可达(82.4±6.6)%。孢子在光下的最适发芽温度为21.6-26.5℃,7d开始萌发,6-7周完全萌发,温度升高或下降均降低孢子萌发率。  相似文献

通过对诺氏虫疠霉生物学特性的研究,明确了该菌孢子萌发,生长的适温范围为15℃-25℃,萌发最适湿度为100%,最适pH为9-10。紫外线对诺氏虫疠霉分生孢子有强烈杀伤作用,照射10分钟,萌发率降低41.9%,不同光照处理间菌落生长差异显,全光照可完全抑制菌落生长,明暗交替最地菌落生长和产孢。  相似文献

在宁夏灌区,春小麦白粉病菌(Erysiphe graminis f.sp.tritici)的分生孢子在-3-29℃温度范围都可萌发。温度达33℃时不能萌发,最适温度为10—15℃。分生孢子虽然在0—100%的湿度下都可以萌发。但以饱和湿度最适。紫外光对分生孢子有强烈的杀伤作用。直射阳光对分生孢子的萌发有抑制作用;在散射光和黑暗条件下,孢子的萌发率没有显着差异。  相似文献

Attempts at management of diseases caused by protozoan plant parasitic Phytomyxea have often been ineffective. The dormant life stage is characterised by long-lived highly robust resting spores that are largely impervious to chemical treatment and environmental stress. This review explores some life stage weaknesses and highlights possible control measures associated with resting spore germination and zoospore taxis. With phytomyxid pathogens of agricultural importance, zoospore release from resting spores is stimulated by plant root exudates. On germination, the zoospores are attracted to host roots by chemoattractant components of root exudates. Both the relatively metabolically inactive resting spore and motile zoospore need to sense the chemical environment to determine the suitability of these germination stimulants or attractants respectively, before they can initiate an appropriate response. Blocking such sensing could inhibit resting spore germination or zoospore taxis. Conversely, the short life span and the vulnerability of zoospores to the environment require them to infect their host within a few hours after release. Identifying a mechanism or conditions that could synchronise resting spore germination in the absence of host plants could lead to diminished pathogen populations in the field.  相似文献

Clubroot, caused by Plasmodiophora brassicae, is a severe soil-borne disease that restricts the production of cruciferous crops worldwide. A better understanding of biotic and abiotic factors regulating germination of P. brassicae resting spores in the soil is significant for developing novel control methods. Previous studies reported that root exudates can trigger P. brassicae resting spore germination, thus enabling a targeted attack of P. brassicae on host plant roots. However, we found that native root exudates collected under sterile conditions from host or non-host plants cannot stimulate the germination of sterile spores, indicating that root exudates may not be direct stimulation factors. Instead, our studies demonstrate that soil bacteria are essential for triggering germination. Through 16s rRNA amplicon sequencing analysis, we found that certain carbon sources and nitrate can reshape the initial microbial community to an inducing community leading to the germination of P. brassicae resting spores. The stimulating communities significantly differed in composition and abundance of bacterial taxa compared to the non-stimulating ones. Several enriched bacterial taxa in stimulating community were significantly correlated with spore germination rates and may be involved as stimulation factors. Based on our findings, a multi-factorial ‘pathobiome’ model comprising abiotic and biotic factors is proposed to represent the putative plant-microbiome-pathogen interactions associated with breaking spore dormancy of P. brassicae in soil. This study presents novel views on P. brassicae pathogenicity and lays the foundation for novel sustainable control strategies of clubroot.  相似文献

内生真菌与丛枝菌根(AM)真菌是构成草原生态系统的重要组成部分.内生真菌会抑制其宿主植物的AM真菌侵染率.本研究以感染2种香柱菌属内生真菌[Epichloё gansuensis(Eg)和E. sibirica(Es)]的天然禾草羽茅为供试材料,进行体外纯培养的内生真菌培养滤液、感染内生真菌的羽茅叶片(包括鲜叶和枯叶)浸提液,以及根系分泌物对摩西球囊霉(Gm)和幼套球囊霉(Ge)2种AM真菌孢子萌发影响试验.结果表明: 香柱菌属内生真菌的培养滤液会显著抑制2种AM真菌孢子的萌发,而感染香柱菌属内生真菌的羽茅根系分泌物只对Ge孢子萌发有显著抑制作用,且上述抑制作用与内生真菌种类无关;鲜叶浸提液对Gm和Ge的孢子萌发率均无显著影响,而枯叶浸提液对Ge的孢子萌发有显著抑制作用.在自然生态系统中,香柱菌属内生真菌通常存在于宿主植物体内,可能通过影响宿主植物的根系分泌物来影响AM真菌孢子的萌发.  相似文献

 The effect of root exudates from onions differing in P status on spore germination and hyphal growth of arbuscular mycorrhizal fungi was investigated. Onion (Allium cepa) was grown in solution culture at different phosphorus concentrations (0, 0.1, 1.0, 8.0 and 24.0 mg P l–1) and root exudates were collected. When spores of the arbuscular mycorrhizal fungus, Gigaspora margarita were incubated with these root exudates, spore germination was only slightly affected but hyphal growth was greatly affected, particularly with exudates from P-deficient plants. This suggests that the P nutrition of host plants influences the composition of root exudates and thereby the hyphal growth of arbuscular mycorrhizal fungi. Accepted: 25 June 1995  相似文献

蓝花丹(Plumbago auriculata Lam.)原产南非,由于其存在自交不亲和现象,导致自然结实率极低。本文研究了蓝花丹种子的形态结构、吸水特性及最适萌发温度,根据形态观测以及发芽率等指标筛选出优质种子,并对优质种子不同的干燥方法、贮藏温度等进行探讨。结果表明:蓝花丹种子呈长椭圆状,长8.00~13.80 mm,长宽比约为5.3:1。种皮薄而柔软,有网状褶皱。外种皮表面有附着物,大面积分布有腺体,利于种子传播。胚芽靠近合点,萌发率较高。种子播种前的浸种时间应保持在11 h以上,最适发芽温度为25~30℃。当种皮颜色为深褐色,种子长度大于11 mm且发芽率大于75%时,可判断为优质种子,发生此性状时即为最佳采种时间。种子适宜的干燥方式为烘箱内30℃干燥1 d,干燥种子最佳贮藏温度为-86℃,且贮藏时间越短发芽率越高,而新鲜种子在此条件下不宜贮藏超过15 d。  相似文献

Bacteriol. 85:207-217. 1963.-It was possible to isolate refractile spores of Bacillus megaterium, from a calcium dipicolinate germination solution, that were activated and would germinate spontaneously in distilled water. Some of the characteristics of the initial phases of bacterial spore germination were determined by studying these unstable activated spores. Activated spores of B. megaterium were resistant to stains and possessed a heat resistance intermediate between that of dormant and of germinated spores. The spontaneous germination of activated spores was inhibited by copper, iron, silver, or mercury salts, saturated o-phenanthroline, or solutions having a low pH value, but not by many common inhibitors. These inhibitions could be partially or completely reversed by the addition of sodium dipicolinate. The activated spores could be deactivated and made similar to dormant spores by treatment with acid. Analyses of the exudates from the variously treated spore suspensions revealed that whatever inhibited the germination of activated spores also inhibited the release of spore material. The composition of the germination exudates was different than that of extracts of dormant spores. Although heavy suspensions of activated spores gradually became swollen and dark when suspended in solutions of o-phenanthroline or at pH 4, the materials released resembled those found in extracts of dormant spores rather than those of normal germination exudates.  相似文献

Spores of the arbuscular mycorrhizal fungi (AMF) Glomus geosporum and Glomus constrictum were harvested from single-spore-derived pot cultures with either Plantago lanceolata or Hieracium pilosella as host plants. PCR-denaturing gradient gel electrophoresis analysis revealed that the bacterial communities associated with the spores depended more on AMF than host plant identity. The composition of the bacterial populations linked to the spores could be predominantly influenced by a specific spore wall composition or AMF exudate rather than by specific root exudates. The majority of the bacterial sequences that were common to both G. geosporum and G. constrictum spores were affiliated with taxonomic groups known to degrade biopolymers (Cellvibrio, Chondromyces, Flexibacter, Lysobacter, and Pseudomonas). Scanning electron microscopy of G. geosporum spores revealed that these bacteria are possibly feeding on the outer hyaline spore layer. The process of maturation and eventual germination of AMF spores might then benefit from the activity of the surface microorganisms degrading the outer hyaline wall layer.  相似文献

通过扫描电镜和透射电镜观察了禾谷多粘菌PolymyxagraminisLed.休眠孢子堆的超微结构。休眠孢子堆仅分布于寄主根表皮细胞中。休眠孢子堆形状不一,有的呈球状,有的呈律状,少则由几十个,多则由数百个紧密排列的休眠孢子组成。休眠孢子彼此通过刺突连接,细胞壁分4层,第三层局部区域结构松散,可能与初生游动孢子萌发孔有关。成熟休眠孢子细胞质丰富,细胞质膜内侧含有大量脂质粒,细胞质中央含一个细胞核,围围分布线粒体、内质网、高尔基体、液泡等细胞器。成熟的休眠孢子在越夏前大多数已释放初生游动孢子,只剩下空壳。表面凹陷是已释放游动孢子的休眠孢子一个特征。本文还讨论了禾谷多粘菌休眠孢子在病害流行学中的作用。  相似文献
