Salt Tolerance in the Triticeae: Leymus sabulosus   总被引:1,自引:0,他引:1  
Elymus dahuhcus, Leymus giganteus, L. angustus, L. sabulosusand, to a lesser extent, L. triticoides, were found to tolerate200 mol m–3 NaCl in solution culture. Elymus dahuricusdiffered from the Leymus species in its ion-uptake characteristics,showing a greater uptake of Cl and Na and a greater loss ofK from the shoots. In a more detailed experiment on Leymus sabulosusit was found that transpiration rates altered rapidly in responseto changes in external salinity whereas the accumulation ofNa and Cl in the leaves exhibited a lag of several days. Insalt stressed L. sabulosus Cl partially replaced the high levelsof nitrate found in the leaves of control plants. Glycinebetainelevels increased in the leaves from 8.0 mol m–3 plantsap in the controls to 28 mol m–3 plant sap at 250 molm–3 NaCl. For decades frequent mass mortalities of Lesser Flamingos (Phoeniconaiasminor Geoffroy) have been observed at alkaline-saline KenyanRift Valley lakes. To estimate the potential influence of toxiccyanobacteria on these mass deaths, the phytoplankton communitieswere investigated in Lakes Bogoria, Nakuru and Elmenteita. Cyanobacterialtoxins were analyzed both in the phytoplankton from the threelakes and in isolated monocyanobacterial strains of Arthrospirafusiformis, Anabaenopsis abijatae, Spirulina subsalsa and Phormidiumterebriformis. Lake Bogoria was dominated by the cyanobacteriumA. fusiformis. In L. Nakuru and L. Elmenteita the phytoplanktonmainly consisted of A. fusiformis, A. abijatae and Anabaenopsisarnoldii, and in L. Nakuru an unknown Anabaena sp. was alsofound. Furthermore, this is the first time A. abijatae and theunknown Anabaena sp. have been found in Kenyan lakes. Phytoplanktonwet weight biomass was found to be high, reaching 777 mg L–1in L. Bogoria, 104 mg L–1 in L. Nakuru and 202 mg L–1in L. Elmenteita. Using HPLC, the cyanobacterial hepatotoxinsmicrocystin-LR, -RR -YR, -LF and -LA and the neurotoxin anatoxin-awere detected in phytoplankton samples from L. Bogoria and L.Nakuru. Total microcystin concentrations amounted to 155 µgmicrocystin-LR equivalents g–1 DW in L. Bogoria, and 4593µg microcystin-LR equivalents g–1 DW in L. Nakuru,with anatoxin-a concentrations at 9 µg g–1 DW inL. Bogoria and 223 µg g–1 DW in L. Nakuru. In L.Elmenteita phytoplankton, no cyanobacterial toxins were found.A. fusiformis was identified as one source of the toxins. Theisolated strain of A. fusiformis from L. Bogoria was found toproduce both microcystin-YR (15.0 µg g–1 DW) andanatoxin-a (10.4 µg g–1 DW), whilst the A. fusiformisstrain from L. Nakuru was found to produce anatoxin-a (0.14µg g–1 DW). Since A. fusiformis mass developmentsare characteristic of alkaline-saline lakes, health risks towildlife, especially the Arthrospira-consuming Lesser Flamingo,may be expected.  相似文献

This study was conducted with Lolium temulentum, Festuca pratensis,and the two hybrids L. multiflorum x F. pratensis ‘Elmet’and L. perenne x F. pratensis ‘Prior’. In a comparisonof various durations (7–42 d) of pretreatment at 4 or7 °C the highest yield of microspore-derived callus of L.temulentum was obtained after pretreatment of spikes at 7 °Cfor 28 d, conditions which also proved optimal for panicle pretreatmentwith F. pratensis. For ‘Elmet’, durations of 21–42d were optimal, and for ‘Prior’ the responses tendedto decline with increasing duration. The chemotactic behaviour of Laminaria digitata spermatozoidsupon stimulation with sex pheromone was studied using videomicroscopyand high speed cinematography. The cells show phobic (‘shock-’)reorientation reactions when perceiving decreasing stimulusconcentrations in a pheromone gradient and are able to adaptto constant concentrations. The phobic responses are executedby regulation of flagellar activity: drastic changes in thedirection of cell movement are induced by rapid deflexions ofthe posterior flagellum. DNA-DNA hybridizations between single-copy nuclear DNA fromLaminaria digitata and total DNA fromL. saccharina, L. Hyperborea, L. rodriguezii, L. ochroleuca andChorda filum, respectively, show that these species ofLaminaria are genotypically closely related.Chorda filum is only distantly related withL. digitata. Based on the thermal elution patterns of the DNA hybrids, as quantified by ΔTm(e) values, it is hypothesized that all five species ofLaminaria evolved at about the same time from their most recent common ancestor some 15–19 Ma ago. This phylogenetic hypothesis is discussed in relation to the history of modern laminarialean distribution patterns. Zinc-induced Vacuolation in Root Meristematic Cells of Cereals   总被引:1,自引:0,他引:1  
In the absence of Zn, vacuolar volume fractions of root meristematiccells of Secale cereale L. cv. K2, Triticum aestivum L. cv.Chinese Spring and Oryza sativa L. cv. IR34 were 5.64 x 10–2,2.17 x 10–2 and 1.63 x 10–2 µm3 vacuole µm–3tissue, respectively. A 4-d exposure to a subtoxic concentrationof zine (0.2 µg Zn cm–3) induced a 2.93-fold anda 6.78-fold increase in the total vacuolar volume fraction inOryza and Triticum, respectively, whereas no significant increasewas observed for Secale. It is proposed that this Zn-inducedvacuolation represents a compartmentalization mechanism. Theinitial total vacuolar volume fraction in Secale was greaterthan that for Oryza and Triticum and this may enable compartmentalizationof the metal soon after the onset of treatment so reducing itscytotoxic effects. Although Littorina littorea (L.) exhibits a relatively consistentpattern of vertical distribution throughout the North Atlantic,ranging from the mid-intertidal to the shallow subtidal zone,its horizontal distribution and abundance are highly variable.In this study, we first described the snail's horizontal distributionpatterns on Appledore Island, ME, USA and then asked whetherwave exposure, rugosity, predator density (e.g. Carcinus maenasand Cancer borealis), the percentage of the substrate coveredby Ascophyllum nodosum, Chondrus crispus, Polysiphonia spp.,and ephemeral green algae, or the percentage of uncovered substrate(bare rock) were correlated with L. littorea abundance in theintertidal zone (0.6 to 0.0 m relative to Mean Lower LowWater [MLLW]) and the shallow subtidal zone (–1.5 to –3.0 mMLLW) at nine study sites. Intertidal densities of L. littoreawere highly variable across sites, ranging from 0 to >600 m–2.In this zone, L. littorea density showed a significant positivecorrelation with rugosity and percentage of bare rock. Densitieswere very low in the subtidal zone (range: 0–19 m–2)and varied little among sites. Exploratory multiple regressionanalysis suggested that L. littorea density was positively correlatedwith the density of C. maenas in the shallow subtidal zone.Additionally, snails in the subtidal zone had thicker shellsthan snails of the same size in the intertidal zone. Our resultssuggest that structural elements of the habitat, such as rugosityand percentage of uncovered substrate, are among the most importantpredictors of L. littorea abundance on moderately protected,rocky intertidal shores. The response of the germination of seeds of Barbarea vema (Mill.)Aschers, Brassica chinensis L., Brassica juncea (L.) Czern.& Coss., Brassica oleracea L. var. gongylodes L., Camelinasaliva (L.) Crantz, Eruca saliva Mill., Lepidium sativum L.,Nasturtium officinale R. Br., and Rorippa palustris (L.) Besserto white fluorescent light of different photon flux densitiesapplied for different daily durations in a diurnal alternatingtemperature regime of 20 °C/30 °C (16 h/8 h) was quantifiedby linear relations between probit percentage germination andthe logarithm of photon dose, the product of photon flux densityand duration. The low energy reaction, in which increasing dosepromotes germination, was detected in all the seed populationsbut in Barbarea vema and Brassica Juncea the lowest photon doseapplied (10–5–2 and 10–5 7 mol m–2 d–1,respectively) was sufficient to saturate the response. Comparisons,where possible, between photoperiods demonstrated reciprocity,i.e. germination was proportional to photon dose irrespectiveof photoperiod, for the low energy reaction in Brassica oleracea(1 min d–1 to 1 h d–1), Camelina saliva (1 min d–1to 8 h d–1), Eruca saliva (1 min d–1 to 24 h d–1),Lepidium sativum (I min d–1 to 8 h d–1) and Rorippapalustris (1 min d–1 to 8 h d–1), but not in Brassicachinensis and Nasturtium officinale. The high irradiance reaction,in which increasing dose inhibits germination, was detectedin Barbarea vema, Brassica chinensis, Brassica juncea, Brassicaoleracea, and Camelina saliva. The minimum dose at which inhibitionwas detected was lO–0–3 mol m–2 d–1.These results are discussed in the context of devising optimallight regimes for laboratory tests intended to maximize germination The response of germination to photon dose was also quantifiedwith 3 x 10–4 M GA2, co-applied (Brassica chinensis, Camelinasaliva, and Lepidium sativum) and with 2 x 10–2 M potassiumnitrate co-applied (Brassica chinensis). In the latter casepotassium nitrate had no effect in the dark and inhibited germinationin the light, but GA2, promoted germination substantially inall three species. Variation amongst seeds in the minimum photondose required to stimulate germination was not affected by co-applicationof GA2, in Brassica chinensis and Camelina saliva, whereas seedsof Lepidium salivum showed a narrower distribution of sensitivitiesto the low energy reaction in the presence of GA2 Barbarea vema (Mill.) High-frequency Organogenesis from Direct Seed Culture in Lathyrus   总被引:2,自引:0,他引:2  
Culture conditions were developed for inducing a high frequencyof direct shoot morphogenesis and whole plant regeneration incultures of intact seedlings of Lathyrus cicera L., L. ochrus(L.) DC., L. sativus L., and L. tingitanus L. The procedureof shoot regeneration involved culturing of whole, mature seedson MS medium containing cytokinins, or thidiazuron (TDZ), asubstituted phenylurea with cytokinin-like activity. Differentiationof shoots occurred without an intervening callus phase fromthe cotyledonary node and surrounding tissues of the intactseedlings developed from seeds germinated on media containingkinetin, BAP or TDZ. An average of 19·0, 15·8,28·8 and 43·0 shoots were regenerated at optimalconcentrations of 50·0, 50·0 and 80·0 µMBAP in L. cicera, L. tingitanus, L. ochrus and L. sativus, respectively.TDZ enhanced the shoot formation at the concentration of 10µM to an average of 33·1 and 33·7 shootsper seedling in L. cicera and L. tingitanus and at 50 µM,to 57·4 shoots per seedling in L. sativus. Regeneratedshoots developed roots on a modified MS medium containing 2·5µM NAA and the surviving plantlets were transferred tosoil. Histological studies revealed de novo formation of shoot budsfrom the epidermal or subepidermal cells of the basal and nodalregion of multiple epicotyls. Several meristematic centres consistingof actively-dividing cells developed in the subepidermal celllayer of the nodal tissue and adjacent areas within 7 d of seedculture on a cytokinin- or TDZ-supplemented medium. This paper describes the macroflora and macrofauna associated with two bull kelp species,Laminaria hyperborea andL. digitata, at the island of Helgoland, North Sea. During a study period of seven months (March–September 1987), 29 macroflora species and 125 macrofauna species were found. The dominant taxonomic groups were Polychaeta (25 species), Bryozoa (17), Amphipoda (14), Hydrozoa (10) and Ascidiae (8). The species maximum was in July. In general,L. hyperborea was preferred as a substrate for settlement toL. digitata. Composition of the communities associated with kelp changed during the season according to exposure to wave action, and according to location on the kelp thallus. The rhizoid community of both kelps bore more species at exposed locations. Wave-exposedL. digitata lacked obvious faunal settlement on both phylloid and cauloid. Phylloid and cauloid ofL. hyperborea were chosen as an attractive substrate at both sheltered and wave-exposed locations, showing an association of encrusting bryozoan and hydrozoan colonies.  相似文献

The various pathways of pollen development were investigatedin cultured anthers of Lolium temulentum, Festuca pratensisand the L. multiflorum x F. pratensis hybrid ‘Elmet’.In all three, development from the vegetative cell was the predominantpathway of pollen callus development. However, there were characteristicdifferences in the behaviour of the generative cell. In L. temulentumit remained attached to the pollen wall and degenerated, whereasin F. pratensis it divided. In ‘Elmet’ it detachedfrom the pollen wall and remained undivided. Both polarizedand unpolarized partitioned calluses were observed. Developmentof the fusion product of the vegetative and generative nucleiwere also observed in anthers of L. temulentum. Anomalous grainswere not found to be major source of pollen calluses. Sections of anthers of L. temulentum were used to investigatethe origin of S pollen grains, the small pale-staining grainswhich denote pollen dimorphism. Such grains form out of contactwith the tapetum and are therefore determined before or duringmeiosis (i.e. before harvest of anthers for culture). Sectionswere also used to demonstrate the influence of the durationof pretreatment on the development of the middle layer of theanther wall. Sporophytes of Laminaria digitata (L.) Lamour. were assayed for their content of accumulated iodine, which ranged from 0.4% of dry weight in adult plants up to 4.7% for young plantlets. Sporophyte tissue from Laminaria saccharina (L.) Lamour. and L. digitata took up iodide according to Michaelis-Menten kinetics. Hydrogen peroxide and various substances known to interfere with oxidative metabolism were shown to either inhibit or enhance the uptake of iodide, confirming that apoplastic oxidations play a key role in iodide uptake in Laminaria. Consistently, iodide uptake was triggered in L. saccharina protoplasts by incubation in the presence of hydrogen peroxide. Similarly, the uptake of iodide was enhanced in L. digitata gametophytes by addition of haloperoxidase, suggesting that this enzyme catalyses the oxidation of iodide by hydrogen peroxide and plays a key role in iodine uptake. Oxidative stress resulted in a marked efflux of the intracellular iodine. In both influx and efflux experiments, a marked proportion (10–30%) of the tracer was not accounted for, indicating volatilisation of iodine. Quercus coccifera L. and Q. ilex L. leaves, collected in winter,when the plants were in full turgor, were treated with polyethyleneglycol (PEG) with a water potential of –48 and –65bar, at 15 °C and at 30 °C, for 2 days. The responseof each species differs with temperature as far as the plastidpigment, anthocyanin and soluble sugar content is concerned.Thus Q. ilex is affected more at 15 °C rather than at 30°C, while in Q. coccifera, apart from a small increase inanthocyanins, no significant change was observed at 15 °C.On the contrary, at 30 °C significant changes were observedin all studied parameters. Chloroplast pigments decreased generallyon the first day and their content either dropped further orincreased to reach the control values on the second day. Alltreatments resulted in an increase in anthocyanin content. Solublesugar content raised in Q. ilex at 15 °C and decreased inboth species at 30 °C. The stomatal response of seedlings grown in 360 or 720 µmolmol–1 to irradiance and leaf-to-air vapour pressure deficit(VPD) at both 360 and 720 µmol mol–1 to CO2 wasmeasured to determine how environmental factors interact withCO2 enrichment to affect stomatal conductance. Seedlings offour species with different conductances and life histories,Cercis canadensis (L.), Quercus rubra (L.), Populus deltoides(Bartr. ex Marsh.) P. nigra (L.), and Pinus taeda (L.), weremeasured in hopes of identifying general responses. Conductanceof seedlings grown at 360 and 720 µmol mol–1 CO2were similar and responded in the same manner to measurementCO2 concentration, irradiance and VPD. Conductance was lowerfor all species when measured at 720 than when measured at 360µmol mol–1 CO2 at both VPDs ({small tilde}1.5 and{small tilde}2.5 kPa) and all measured irradiances greater thanzero (100, 300, 600,>1600 µmol m–2 S–2)The average decrease in conductance due to measurement in elevatedCO2 concentration was 32% for Cercis, 29% for Quercus, 26% forPopulus, and 11% for Pinus. For alt species, the absolute decreasein conductance due to measurement in CO2 enrichment decreasedas irradiance decreased or VPD increased. The proportional decreasedue to measurement in CO2 enrichment decreased in three of eightcases: from 0.46 to 0.10 in Populus and from 0.18 to 0.07 inPinus as irradiance decreased from>1600 to 100 µmolm–2 s–1 and from 0.35 to 0.24 in Cercis as VPD increasedfrom 1.3 to 2.6 kPa. Aspects of growth and development were evaluated in the fast-developingannual Triticum aestivum L. ‘Chinese Spring’, theslow-developing perennial Lophopyrum elongatum Löve, theiramphiploid, and chromosome addition and substitution lines ofL. elongatum into ‘Chinese Spring’. Relative growthrates (RGR) of shoots of L. elongatum and the amphiploid werelower than those of ‘Chinese Spring’ (34 and 13%respectively) and main stem development was also slower. Therewas no difference in shoot RGR of any of the chromosome additionor substitution lines and that of ‘Chinese Spring’when assessed between Haun stages 2.0 and 5.0. In contrast,several aspects of plant development were observed to differin the chromosome addition and substitution lines. SubstitutingE genome chromosomes (with the exceptions of 3E and 4E) forD genome chromosomes, or adding E genome chromosomes, slowedthe rate of main stem development, at least up to Haun stage5.0. Despite these differences in the rate of main stem development,the appearance of adventitious roots commenced at approximatelyHaun stage 2.0 in all genotypes. Lateral root primordium development has been examined in primaryroots of Vicia faba L., Pisum sativum L., Zea mays L. and Phaseolusvulgaris L. Following their initiation from an estimated minimumnumber of 77–162, 20–57, 17 and 12 cells respectivelyin Vicia, Phaseolus, Pisum and Zea, the primordia rapidly increasedin cell number to emerge as secondary roots about 2.8–3.6days later depending on the species being examined. Cell doublingtimes were estimated directly from cell numbers at differenttimes following primordium inception and were found to increasewith increase in primordium size in each of the species investigated. The number of primordia formed per cm of root growth per daywas greatest in Zea and least in Pisum. A comparison of thedata obtained for Vicia with that in the literature led to theconclusion that although the number of primordia produced percm of root growth was independent of the rate of primary elongation,the number produced per day increased in a linear fashion withincrease in the rate at which the primary lengthened. In soyabeans (Glycine max (L.) Merr.), glutamine synthetase(GS) activity was greater at 28 °C than at 35 °C. Thereverse was true for foliar-N loss. In field-grown plants at29 °C, the GS activity was higher and foliar-N loss waslower in soyabeans than in Amaranthus palmeri (S.) Wats. Methioninesulphoximine, a GS inhibitor, and 6-diazo-5-oxo-L-norleucine,a glutamate synthase inhibitor, significantly increased foliar-Nloss from soyabeans. The data suggest that conditions conduciveto decreased GS or glutamate synthase activity may result inincreased foliar-N loss. Ion chromatographic methods determined organic acids and mainnutrient minerals in the apoplastic solution from leaves ofseveral Fagaceae (Quercus ilex L., Quercus cerris L., Quercusvirgiliana (Ten.) Ten, and Fagus sylvatica L.). The anions oforganic acids found in high amounts (250 to 650 µM) werequinate, malate, and oxalate. Lactate, pyruvate, formate andacetate were detected in relatively low amounts with concentrationsbetween 20 and 200 µM. The total concentration of organicacids in the apoplastic sap ranged between 1.5 and 2 mM. Thetotal concentration of inorganic cations (K+, Mg2+, NH4+, Ca2+,Na+) and anions (C1, NO3, SO2–4 and PO3–4)in the apoplastic sap varied between 5 and 10 mM, and 0.35 and1.8 mM, respectively. We conclude that the concentration oforganic acid ions in the leaf apoplast depends mainly on theexchange with the leaf cells and is influenced by the electrochemicalgradient between the symplast and the apoplast in relation tothe water potential of the leaf. The determination of formateand acetate in the apoplastic compartment of leaves lend weightto the argument that the production of these acids by treesis a important emission source to the atmosphere. The relationship between germinability and fluorochromasia (FCR)has been studied in pollen of eight genera, Secale, Iris, Carex,Eleocharis, Cytisus, Digitalis, Plantago and Lonicera. The FCRtests two properties of the pollen, (a) the integrity of theplasmalemma of the vegetative cell and (b) the presence of anesterase capable of cleaving the fluorogenic ester, fluoresceindiacetate. In general, the correlations between FCR and germinabilitywere found to be very highly significant. This is interpretedas meaning that the primary determinant of pollen viabilityin short-term storage is the state of the vegetative cell membranes.It is suggested that in the partly dehydrated grain at the timeof dispersal the membranes are largely dissociated and do notform an osmotic barrier, but that normal properties are recoveredduring controlled hydration which would normally take placeon the stigma. According to this view, the decay of apparentviability is related to the progressive loss of the capacityof the vegetative cell membranes to regain a normal structureon rehydration. The genera investigated varied in their longevityin storage in low and high humidity conditions. After low-humiditystorage, most showed some enhancement of germination with 1h exposure to a humid atmosphere before transfer to the germinationmedium. In acidic soils, monomeric aluminium (Al3+) can reach levelsthat are toxic to plants, thus preventing many species fromgrowing there. Organic acids chelate Al and render it non-toxic.It has been shown that exudation of organic acids by Al-tolerantcrops increases their tolerance to Al. We have extended thisobservation to wild plants by comparing the ability of ten herbsto exude organic acids in response to elevated Al levels. Wehypothesized that exudation of organic acids was related tothe ability of plants to grow on Al-rich soils. Two grasseswere grown in rhizotrons in soils with 41 and 63 µM reactiveAl. Organic acids were sampled from root tips connected to anintact plant-root system.Deschampsia flexuosa (L.) Trin. exudedmore malic acid when grown in the soil with the highest Al content.Five forbs and five grasses were also exposed to three Al levels(0, 25 and 75 µM) in a hydroponic system.Rumex acetosellaL. and Viscaria vulgaris Bernh. increased exudation of oxalicacid and Galium saxatile auct. non L. and Veronica officinalisL. increased exudation of citric acid in response to elevatedAl. The distribution of the forbs in the field as describedby soil pH was negatively related to the amount of organic acidsexuded in response to Al. In contrast, none of the grasses exudedhigher amounts of organic acids with increasing Al concentrationin the hydroponic experiment.
