Tradescantia pallida cv. purpurea, a popular garden plant in Brazil, was used for the Tradescantia micronucleus (Trad-MCN) assay. In situ monitoring of the genotoxicity of air pollutants was carried out by sentinel approach, using the plant grown in the field or using the plants in pots which were carried to the monitoring sites. Two highly polluted sites, in S?o Paulo city (Cerqueira Cesar and Congonhas) and two rural sites (the cities of Pirassununga, 200 km and Caucaia do Alto, 50 km from S?o Paulo, respectively) were chosen for this study, in order to determine the gradient difference of the air pollution levels. Sentinel plants in Congonhas site presented the highest frequency of micronuclei (4.4%), in comparison with 2.2 and 2.3% found in plants from Pirassununga and Cerqueira Cesar sites, respectively (Kruskal-Wallis; P<0.020). Significant increases (F test; P<0.0001) in the frequency of micronuclei were observed in plants exposed in the polluted urban sites (Cerqueira Cesar: 5.7%; Congonhas: 7.1% and Caucaia do Alto: 2.3%). The increase in the frequency of micronuclei observed indicates the potential risk of mutagenicity in presence of high concentrations of pollutants.  相似文献

The Tradescantia-micronucleus (Trad-MCN) bioassay was used to determine the clastogenicity of wastewater samples collected from the Arena canal which contains effluent from the industrial district Benito Juarez of the city of Queretaro, Mexico. Fifteen wastewater samples which were collected, in most cases, at bi-weekly intervals beginning in September 1986 through February 1988, after a 3-fold dilution were used to treat Tradescantia plant cuttings. The clastogenicity expressed in terms of micronucleus frequencies of treated groups (30 h of treatment without recovery time) was significantly (0.01) higher than that of the tapwater control groups. The Trad-MCN bioassay was also used for in situ monitoring of air pollutants for the clastogenicity at 3 sites near the industrial and residential areas (Flores Magon, Conalep and Bellas Artes) of the city of Queretaro. Fourteen monitoring trips were made to each of the 3 sites at monthly intervals beginning in May 1988 through June 1990. Seasonal variation of micronucleus frequencies was exhibited with the peak clastogenicities shown in May and June 1988, June 1989 and April 1990 at the three sites. Micronucleus frequencies of all the exposed groups at the Conalep site, a predominantly industrial area, were markedly higher than that of the laboratory control groups throughout the 2-year period.  相似文献

We have taken soil samples from two sites in Prague, the capital of the Czech Republic, that are heavily polluted by motor vehicles. As a negative control, soil samples from a recreational site in Prague with no motor vehicle traffic were used. Soil samples from these sites were extracted with water or 5% dimethylsulphoxide (DMSO) for 24 h and cuttings of Tradescantia clone 4430 were immersed for 12 h at 25 degrees C in the extraction solutions. As a positive control Tradescantia plants have been treated with the promutagenic arylamine o-phenylenediamine at the same treatment conditions. None of the tested soil extractions significantly increased the frequency of somatic mutations in the stamen hair assay. By contrast, a 5% DMSO soil extract from one of the tested sites (entrance of the Letná tunnel) significantly increased the frequency of micronuclei (MNC) in the pollen mother tetrad cells. A repetition of the treatment 14 days later also resulted in an increase in the frequency of MNC, however the increase was not statistically significant. Three well known plant bioassays, the Allium root chromosome aberration (AL-RAA) assay, the Tradescantia micronucleus (Trad-MCN) assay, and the Tradescantia stamen hair (Trad-SHM) mutation assay were validated in 1991 by the International Programme on Chemical Safety (IPCS) under the auspices of the World Health Organization, and the United Nations Environment Programme (UNEP). These plant bioassays have proven to be efficient tests for chemical screening and especially for in situ monitoring for genotoxicity of environmental pollutants. As a result of this validation study, standard protocols of these three plant bioassays were used by some of the 11 participating countries in the IPCS to carry on genotoxicity tests on air, water and soil as a follow up activity. In the city of Queretaro, Mexico, wastewater coming from both industrial and domestic sources and without any treatment is used to irrigate the farm crops, polluting the soil. Potentially the pollutants could reach the food chain. For the above reason, soil irrigated with wastewater was sampled and monitored for the presence of genotoxic agents using the above three bioassays. Extracts from soil samples were made using distilled water and organic solvents by shaking the sample for about 12 h under a relatively low temperature (15-20 degrees C). Plant cuttings of Tradescantia or the roots of Allium were treated by submerging them in the extracts. Three replicates of each sample were analyzed in each of the three bioassays. Extracts using DMSO, ethanol and distilled water tested positive in the three bioassays and there were no differences for the genotoxicity of the extracts with the different solvents.  相似文献

The Lijang river which passes through the Guilin mountains, and Guilin city is a world renowned scenic spot on the southwest border of China. The river and its tributaries receive water from the mountain tops and springs underground. The river water was clean two decades ago before the development of industrial establishments and extra heavy tourism. Deforestation over the mountain tops on the upper stream and its tributaries in the last decades has created serious erosion and increased sedimentation in the river. In the present study, the Tradescantia micronucleus (Trad-MCN) and Tradescantia stamen hair mutation (Trad-SHM) assays were used to evaluate the genetoxicity of water samples collected from 60 different sites along the river. Results indicate that most of the water samples from the tributaries were highly mutagenic, and that pollutants had accumulated in the main river in the Guilin city area from the industrial effluent and city sewage. Both the Trad-MCN and Trad-SHM assays were highly effective for the detection of mutagens in the water samples.  相似文献

Tradescantia albiflora (Kunth), a trailing ground species naturally occurring in deep shade in rainforests, has an unusual photosynthetic acclimation profile for growth irradiance. Although capable of increasing its capacity for electron transport, photophosphorylation and carbon fixation when grown in full sunlight, Tradescantia has constant chlorophyll alb ratios, photosystem reaction centre stoichiometry and pigment-protein composition at all growth irradiances (Chow et al. 1991. Physiol. Plant. 81: 175–182). To gain an insight into the compensatory strategies which allow Tradescantia to grow in both high and low lights, plants were grown under shade cloth (100 to 1.4% relative growth irradiance) and leaf and chloroplast attributes were compared. While shade Tradescantia chloroplasts had three times more chlorophyll per chloroplast and twice the length of thylakoid membranes compared to plants grown in full sunlight, the ratios of appressed to nonappressed thylakoid membranes were constant. The average net surface charge density of destacked thylakoids was the same for plants grown at moderate and low-irradiance, consistent with their similar stacking profiles. Tradescantia plants grown in direct sunlight had 10-times more fresh and dry weight per plant compared to plants grown in shade, despite a lower photosynthetic capacity on a leaf area basis with partial photoinhibition. We conclude that having a light-harvesting apparatus permanently locked into the "shade-plant mode " does not necessarily prevent a plant from thriving in high light. Analyses of leaf growth at different irradiances provide a partial explanation of the manner in which Tradescantia compensates for very low photosynthetic capacity per unit leaf in sunlight.  相似文献

 30种2~3年生园林植物分别盆栽于不同污染强度的工业生产区、交通繁忙区和相对清洁区,在第一生长季结束后对各试验区植物叶片气体交换特征参数进行测定,评价了大气污染对不同植物种类的可能影响效果。结果表明:工业污染区和交通繁忙区均有较高的SO2浓度,交通繁忙区NOx浓度明显高于其它试验区。根据综合污染指数,3个试验区污染强度为交通繁忙区>工业污染区>相对清洁区。生长在污染区的大多数植物的净光合速率(Pn)、蒸腾速率(E)和气孔导度(gs) 均出现不同程度的下降,下降幅度因植物种类不同而有较大差异。植物Pn、E与gs之间存在显著线性相关关系,表明多数植物能够感应污染胁迫和调节气孔开张度,是植物对胁迫环境的一种适应策略。但是污染胁迫下Pn、E与gs线性相关的显著程度被削弱,表现出污染胁迫下不同植物种类Pn和E的变化与gs变化的不协同性和复杂性。根据Pn下降程度可将植物分组为轻度敏感(11种)、中度敏感(11种)和高度敏感植物(8种)。   相似文献

This study was designed to determine the clastogenicity of particulate matter (aerodynamic diameter smaller than 10 microm) in the urban polluted air in the city of S?o Paulo. The Tradescantia-micronucleus (Trad-MCN) assay was used throughout this study to evaluate the clastogenicity of the extracts of the particulate matter. Tradescantia pallida (Rose) Hunt. cv. purpurea, an indigenous cultivar, was used in the Trad-MCN assay. The efficacy of this plant material for the Trad-MCN assay was validated with dose-response studies using formaldehyde and beta radiation. Dose-response curves were established with these known mutagens. The extracts of the PM10 particles at concentrations between 5 and 50 ppm induced a dose-related increase in MCN frequencies. The results indicate that T. pallida is equally sensitive to mutagens as the standard Tradescantia clone 4430 or 03 and the particulate matter in the urban air are clastogenic to the chromosomes of this plant. Inhalation of these particles by urban dwellers may affect their health by inducing similar genetic damage.  相似文献

Pectin methylesterase (PME) is one of a number of enzymes released by the fungus Aspergillus niger that are involved in the degradation of specific plant cell-wall structures. PME is a glycoprotein with three potential sites for N-linked glycosylation. The glycosylation may affect the hydrolytic activity or the substrate specificity of PME. In this work, we investigate first the structures and the attachment sites of the glycans present on recombinant wild-type PME. Further, a series of PME mutants was created in which the three potential N-linked glycosylation sites were eliminated in all possible combinations. The glycosylation of the mutants and their activities were then studied. Mass spectrometric techniques tailored for carbohydrate analysis were applied to both characterize the glycan structures and to determine the specific sites of attachment. High mannose structures with variable numbers of mannose were found on the wild-type, as well as the mutant forms. Studies using the mutants suggest that glycosylation does not strongly influence the activity. Whether it may affect the substrate specify of the enzyme is unknown, and that aspect will be explored in future work.  相似文献

In response to contamination from the recent Fukushima nuclear accident, we conducted radionuclide analysis on bamboos sampled from six sites within a 25 to 980 km radius of the Fukushima Daiichi nuclear power plant. Maximum activity concentrations of radiocesium (134)Cs and (137)Cs in samples from Fukushima city, 65 km away from the Fukushima Daiichi plant, were in excess of 71 and 79 kBq/kg, dry weight (DW), respectively. In Kashiwa city, 195 km away from the Fukushima Daiichi, the sample concentrations were in excess of 3.4 and 4.3 kBq/kg DW, respectively. In Toyohashi city, 440 km away from the Fukushima Daiichi, the concentrations were below the measurable limits of up to 4.5 Bq/kg DW. In the radiocesium contaminated samples, the radiocesium activity was higher in mature and fallen leaves than in young leaves, branches and culms.  相似文献

Tests have shown plant bioassays to be excellent for mutagenicity studies. Most studies with plant bioassays, however, have been carried out either in the laboratory, or if, in situ, as monitors of atmospheric contaminants. The primary purpose of this study was to assess the utility of in situ plant mutagenicity bioassays in monitoring water contaminants. The assay systems tested were the Tradescantia stamen hair and micronucleus assays for the detection of gene mutations and chromosomal aberrations respectively, and the Vicia faba bioassay system which detects chromosomal aberrations in root tips. The assays were used to test the effluent from a pulp and paper mill located on the north shore of Lake Superior. Assays were performed in a creek containing raw effluent and in the bay of Lake Superior into which the creek emptied. All in situ treatments were carried out for 24 h. The effluent from the creek was heavy with pulp and debris which coated the plant cuttings and the Vicia faba seedlings and may have restricted the uptake from the effluent. In the creek, at test sites 11.5 km from the source, the effluent was toxic to the Vicia faba roots as evidenced by a reduction in the mitotic index. The data for the Tradescantia stamen hair assay in the creek were equivocal. The cuttings from the creek test sites and the air and water control sites appeared to have undergone a physiological delay. Within a day or two after the return to the laboratory, that is 6-8 days after testing, flowering almost ceased and did not fully resume until about day 35. This reduction in flowering was particularly severe with the cuttings from the effluent and air control sites, making it very difficult to interpret the results. In contrast, the Tradescantia micronucleus and Vicia faba chromosomal aberration data were unequivocal; each produced positive responses at both test sites relative to the air and water controls. The results obtained for the bay sites with all 3 assays were in agreement. In that section of the bay visibly contaminated by the creek effluent, increases in stamen hair mutants, micronuclei, and chromosome aberrations were measured. Changes in the lipid peroxidation and oxidatively modified protein levels as well as the antioxidant defense system in the blood of white stork (Ciconia ciconia) chicks were analyzed during their development in nests in polluted and control environments. The control, relatively pure, environment was the village of K?opot, with no industrial plants within a radius of 150 km. Blood samples were also collected in two polluted areas, including (1) the suburban village of Czarnowo, located 20 km from the city of Zielona Góra (southwestern Poland) and (2) an area near the town of G?ogów, where a large copper smelter is situated. The effects of various ecological threats from different environments tended to initiate oxidative stress and decrease total antioxidant capacity. The initiation of lipid peroxidation and increases in the levels of carbonyl derivatives of oxidative amino acids were observed in the plasma of chicks from polluted areas. Plasma catalase and the selenium-dependent glutathione peroxidase activities of erythrocytes were significantly higher in chicks from polluted areas than in those from control areas. Oxidative stress and components of the antioxidant defense system, especially glutathione peroxidase activity, can be used as indicators of oxidative stress, which was found to be greater in the polluted areas (near the G?ogów copper smelter). Research into oxidative stress biomarkers can aid assessments of the conditions of birds, and points to a positive association with miscellaneous environmental loads.  相似文献

This study evaluated the impact of pollution on growth responses inDatura innoxia. Coal-smoke emissions were produced by the Badarpur Thermal Power Plant in Delhi, India. At the polluted site, the size of roots and leaves as well as the number of branches and leaves per plant increased, but shoot lengths and leaf areas were lower, compared with control plants. The net photosynthesis rate, stomatal resistance, and the amount of pigments (chlorophyll a, b, and carotenoids) were less in pollution-affected plants, while stomatal conductance and intercellular CO2 concentration were higher in these plants. Explants from both sites (polluted and non-polluted), grown in vitro on various combinations of auxin (2,4-D, NAA) and cytokinin (BAP, KN), showed the maximum response on a medium containing NAA (0.1 mg L-1) with BAP (5.0 mg L-1). Hyoscyamine content was higher in all parts (root, stem, leaf, and regener-ants) of the polluted plants.  相似文献

Industrial barrens replacing coniferous forests around the Severonikel smelter in the Kola Peninsula, northwestern Russia, have recently expanded over 3000 ha or more. Total concentrations of metal contaminants in the upper soil layers approach 3000–5000 μg/g, and maximum hourly concentrations of sulphur dioxide in ambient air exceed 1000 μg/m3. To monitor possibilities for vegetation recovery in the denuded landscapes continuously affected by industrial emissions, we conducted several experiments with 4- to 15-year-old (1–25 cm tall) seedlings of Betula pubescens ssp. czerepanovi (mountain birch), replanted to two barren sites. Specifically, we investigated the effects of wind-sheltering, watering, and fertilization on seedling performance in the polluted sites. Sheltered and watered seedlings had more symmetrical leaves than control seedlings, suggesting less environmental stress. Consistently, sheltering and (to a lesser extent) watering improved the survival of seedlings compared with controls. The beneficial effects of watering and sheltering were most pronounced the first 2–4 weeks following planting and were greatest in the most polluted site. We conclude that the revegetation of industrial barrens can be significantly promoted by inexpensive treatments such as wind sheltering and watering, even under current emissions.  相似文献

The genetic impact of the 1986 accident at the Ukraine Chernobyl Nuclear Power Plant (NPP) on populations of living organisms has yet to be fully assessed. Monitoring of the genotoxicity of polluted soils is a key element in the disaster management program. We used Arabidopsis thaliana and Nicotiana tabacum plants transgenic for a reporter gene revealing homologous recombination to study the genetic effects of chronic low-dose radiation stemming from the soil in inhabited areas of Ukraine where contamination by the accident ranges from 1 to 40 Ci/km2. We noted a significant dose-dependent increase of homologous recombination in plants cultivated in the affected inhabited areas, proving the persistently high genotoxicity of the radioactively contaminated soils.  相似文献

Inflorescences of Tradescantia clone 4430 were exposed to different concentrations of radon (Rn) gas (0.85, 12.10, 36.50 and 98.16 kBq/m3) from plants placed in an acrylic chamber that received radon from a container with pitchblende (containing uranium mineral). The exposure time was 24 h, afterwards the plants were left for 6 h in water and constantly aerated. Positive control plants were irradiated with gamma rays (0.8 Gy) and negative control plants received ambient air only (the background measurement had a mean of 0.38 kBq/m3). Micronuclei (MCN) in the tetrads induced by alpha particles emitted from Rn were tabulated and a linear concentration response was obtained. The potency of radon to induce MCN from the slope of the regression line was 0.13 MCN/kBq/m3 of alpha-radiation. Radon could reach the anthers by diffusion through the aerial spaces within the buds.  相似文献

In order to assess the impact of SO2, CO2, NO x , and CO on non-enzymatic foliar defense mechanism around oil refineries, the present study was undertaken in Batman, Turkey during the periods February–April–June 2002. For this purpose, the four common evergreen plants at all sites, Pinus nigra Arn. subsp. pallasiana (Lamb.) Halmboe, Ligustrum vulgare L., Thuja orientalis L., and Nerium oleander L. were selected as test plants. Four study sites were selected at the Batman Refinery area (AR). Four control sites were also selected at a distance of about 5 km from the refinery (OR). Responses to oil refinery pollution of plants, heavy metals, chlorophyll (chl), proline, carotenoids, lipid peroxidation, SH-compounds, and ascorbic acid were measured. In addition, ambient pollutant gas concentrations were measured. The pollutant gas concentrations were high in the immediate vicinity of oil refinery. The levels of total chl, SH-compounds, ascorbic acid, and chla/chlb ratio were found to decrease, and the amount of proline, carotenoids, lipid peroxidation, and reduced ascorbate/oxidized ascorbate ratio were found to increase in comparison to plants growing at the control sites. A comparison of four evergreen plant species indicated that L. vulgare was the most resistance to refinery induced air pollution and the magnitude of foliar injury symptoms was greatest in P. nigra and T. orientalis.  相似文献

The five species of Tradescantia section Cymbispatha studied, including one species T. poelliae D. R. Hunt, have chromosome numbers of In = 12, 14, 16, 22, 28, 30 and 36 and karyotypes of acrocentric, metacentric or telocentric chromosomes, or mixtures of both acrocentric and metacentric chromosomes. The numbers of major chromosome arms of these cytotypes give a nombre fondamentaP series of 14, 28, 42 and 56 which, in combination with meiotic analyses, indicates plants which, in genetical terms at least, are diploid, tetraploid, hexaploid and octoploid. This series has evolved from a 2 n = 14 acrocentric or telocentric karyotype by a combination of Robertsonian fusion and polyploidy. Pseudo-iso-chromosomes are sometimes formed in this evolutionary development and can persist as stable members of normal complements.  相似文献
