Gari was examined for its post-processing microbial content. Aerobic mesophilic bacteria and fungi were isolated from all samples. The total viable bacterial counts ranged from 2.0 X 10(2) to 8.0 X 10(4) cfu/g. Fungal counts ranged from 1.0 X 10(2) to 1.5 X 10(4) cfu/g. The total viable counts of fresh samples were much lower than those of market and packaged samples. Bacillus, Micrococcus and Proteus spp. were the bacteria isolated, Aspergillus niger, Aspergillus flavus and Penicillium spp. the fungi. Food borne parasites and pathogens such as Staph. aureus and Clostridium perfringens were not found. The gari samples were quite stable, having a shelf life of 3-6 months. The water activities of the samples ranged from 0.52 to 0.68. Based on the microbial counts of the samples, the critical upper limit for the safety of gari was set at 10(4) cfu/g dry sample.  相似文献

Feed samples collected from different poultry farms and feed mills situated in Andaman and Nicobar islands in India were assessed for microflora and aflatoxin B1 contamination. The bacterial counts ranged from 1.0 times 107 to 8.8 times 107 cfu/g of the feeds, while counts of fungi ranged from 1.0 times 103 to 8.7 times 103 cfu/g. The mycoflora comprised mainly of Aspergillus spp., A. flavus being most dominant. Aflatoxin B1 was detected by monoclonal antibody-based enzyme linked immunosorbent assay technique and the content in different feed samples ranged from 5.5 to 90 ng/g.  相似文献

The amylase-producing ability of the intestinal microflora in cultured specimens of ayu, carp, channel catfish, Japanese eel and tilapia was determined. Mean viable counts of aerobes and anaerobes ranged from 1·1×106 to 3·7×108 cfu g−1 and from 1·3×103 to 1·6×108 cfu g−1, respectively. Aeromonas spp. and Bacteroidaceae were predominant in four to five fish species. Of 206 strains examined, 65 (31·6%) produced ≥0·01 U amylase ml−1. The percentage of producers differed among families and genera of bacteria and fish species. While 56% of the anaerobes produced amylase, only 20% of the aerobes did. More than 50% of Aeromonas , Bacteroidaceae and Clostridium strains produced amylase efficiently while Acinetobacter , coryneforms, Enterobacteriaceae, Moraxella , Plesiomonas and Streptococcus strains did not. High amylase production (≥0·05 U ml−1) was found in 12 strains, 11 from Aeromonas and one Pseudomonas . The percentage of high amylase producers in Japanese eel was lower than the other four fish (2–30%). These results strongly suggest that the amylase produced by the intestinal microflora play an important role in the digestion of starch in freshwater fish to some extent.  相似文献

Polyclonal antisera made in rabbits against whole washed cells of Vibrio pelagius and Aeromonas caviae were used for detection of these bacterial species in the rearing water and gastrointestinal tract of healthy turbot ( Scophthalmus maximus ) larvae exposed to V. pelagius and/or Aer. caviae . The results demonstrated that this method is suitable for detection of V. pelagius and Aer. caviae in water samples and larvae at population levels higher than 103 ml−1 and 103 larva−1. Populations of aerobic heterotrophic bacteria present in the gastrointestinal tract of turbot larvae, estimated using the dilution plate technique, increased from approximately 4 × 102 bacteria larva−1 on day 3 post-hatching to approximately 105 bacteria fish−1 16 days post-hatching. Sixteen days after hatching, Vibrio spp. accounted for approximately 3 × 104 cfu larva−1 exposed to V. pelagius on days 2, 5 and 8 post-hatching. However, only 103 of the Vibrio spp. belonged to V. pelagius . When larvae were exposed to Aer. caviae on day 2 post-hatching, the gut microbiota of 5-day old larvae was mainly colonized by Aeromonas spp. (104 larva−1), of which 9 × 103 belonged to Aer. caviae . Later in the experiment, at the time when high mortality occurred, 9 × 105 Aer. caviae were detected. Introduction of V. pelagius to the rearing water seemed to improve larval survival compared with fish exposed to Aer. caviae and with the control group. It was therefore concluded that it is beneficial with regard to larval survival to introduce bacteria ( V. pelagius ) to the rearing water.  相似文献

H. SUGITA, J. KAWASAKI, J. KUMAZAWA AND Y. DEGUCHI. 1996. The amylase-producing ability of intestinal bacteria in one marine crab and seven fish species was determined. Mean total viable counts ranged from 1.3 × 105 to 1.5 × 108 cfu g−1, and Vibrionaceae were predominant in all specimens. Of 1585 strains examined, 341 (21.5%) produced ≥0.01 U amylase ml−1. Percentage of producers (≥0.01 U ml−1) differed among genera/families. High abilities (≥0.05 U ml−1) were found in 1.4-3.6% of Enterobacteriaceae, Pseudomonas and Vibrionaceae strains. On the other hand, percentage of producers varied with animal species. These results reveal that the amylase producers were widely distributed in the digestive tracts of coastal animals including crabs and fish, irrespective of their food habitats.  相似文献

The sludge from hospital waste treatment facilities is a potential source of infectious organisms. The average numbers of micro-organisms in the sludge of hospital wastewater in Taiwan were as follows: total count 8·1 × 107 cfu g−1 (dry weight of sludge), and 1·4 × 106, 3·6 × 105, 1·6 × 105, 2·2 × 105 and 5·5 × 104 cfu g−1 (dry weight of sludge) for total coliforms, faecal coliforms, faecal streptococci, Pseudomonas aeruginosa and Salmonella spp., respectively . Salmonella spp. were detected in 37% (10 of 27) of the sludges from hospital wastewaters. Therefore, the treatment of such sludge to reduce pathogenic micro-organisms should be considered.  相似文献

Abstract A method for extraction of microbial populations from wood samples was worked out which gave good recovery of both aerobic and anaerobic microorganisms in agar shake dilution and plating enumerations. This method was applied to the quantification of microbial populations in three European white firs ( Abies alba Mill.) which were afflicted with the European fir disease. Low numbers of aerobic microorganisms (102−104 colony- forming units (cfu) per g fresh tissue) were detected in sapwood irrespective of the degree of affliction. Anaerobic bacteria were usually 1–2 orders of magnitude less frequent. Wetwood of highly diseased firs contained significantly higher numbers of aerobic microorganisms (105−107), whereas the number of anaerobes was not enhanced significantly. Among the prevalent aerobic microorganisms in wetwood were Protaminobacter, Pseudomonas strains, and a yeast. In anaerobic counts from wetwood, Klebsiella and Vibrio strains predominated. The sapwood contained Bacillus, Beijerinckia, Staphylococcus , and Clostridium spp. High numbers of aerobic microorganisms were also detected in the roots and lower stem of a diseased vine plant ( Vitis vinifera L.). The importance of microbial populations in wetwood formation and disease expression is discussed.  相似文献

Abstract There was a progressive increase in the size of the aerobic heterotrophic bacterial populations along the gastro-intestinal tract of farmed Dover sole. Moreover, higher counts were recorded in juvenile than in adult animals. Thus, in juvenile fish, 5.2 × 105, 8.0 × 105 and 9.8 × 106 aerobic heterotrophs/g were recovered from the stomach/foregut, midgut and hindgut/rectum, respectively. In adult fish, comparative samples revealed the presence of only 3.0 × 104, 7.0 × 104 and 2.3 × 105 bacteria/g, respectively. There bacteria were equated with Acinetobacter, Alcaligenes , Enterobacteriaceae representatives, Flavobacterium, Micrococcus, Photobacterium, Staphylococcus and Vibrio . Of the compounds tested, many isolates, particularly those recovered from the hindgut/rectum, degraded p -nitrophenyl- β - N -acetylglucosaminide, chitin and collagen. Consequently, it is likely that such organisms may contribute to nutritional processes within Dover sole.  相似文献

Abstract A method was developed for direct extraction, purification and amplification of DNA from forest soil. Eighty-two % of the DNA in Pseudomonas aeruginosa UG2Lr introduced into soil was recovered. The detection limit for the strain was approximately 800 cfu g−1 of dry soil based on the polymerase chain reaction (PCR). Survival of κ-carrageenan-encapsulated and unencapsulated UG2Lr was monitored by antibiotic selective and bioluminescence-based nonselective plating and PCR-amplification of a tnsA fragment. After freeze-thaw treatment of soil samples, the unencapsulated UG2Lr declined from an initial population density of 1 × 109 cfu g−1 of dry soil to below the detection threshold of both selective (14 cfu g−1 of dry soil) and nonselective (1 × 103 cfu g−1 of dry soil) plating. However, presence of nonculturable UG2Lr cells in the soil was revealed by PCR and resuscitation of the bacteria. Population density of the encapsulated UG2Lr increased from 2.7 × 106 to 2.9 × 108 cfu g−1 of dry soil after a 3-week incubation at 22°C and declined to 6.3 × 106 cfu g−1 of dry soil after the freeze-thaw treatment.  相似文献

SUMMARY: Experiments are described in which minced chicken meat, packed anaerobically, was irradiated at room temperature and in the frozen state with a wide range of doses of 4 MeV cathode rays. Sterility was achieved in 14 out of 15 samples which had received 2 × 106 rads or more. Doses of 0·5 and 1·0 × 106 rads allowed survival of a few bacteria/g, usually spore formers. Bacterial counts indicated an approximately logarithmic decrease in numbers at lower doses, while freezing reduced the bactericidal effect.
The storage life at 5° was prolonged only slightly by doses of 5 × 104 and 10 × 104 rads, and highly variable results were obtained with 17·5 × 104 rads. A dose of 25 × 104 rads, however, increased the storage life very considerably. The types of bacteria present initially, and after irradiation with low doses and storage at 5°, were studied. After storage for 12 days or more various types of nonsporing Gram-positive rods were predominant in almost all samples, both control and irradiated. Streptococci were also important where irradiation with 17·5 × 104 and 25 × 104 rads was followed by long storage.  相似文献

Abstract The sulfur cycle in a microbial mat was studied by determining viable counts of sulfate-reducing bacteria, chemolithoautotrophic sulfur bacteria and anoxygenic phototrophic bacteria. All three functional groups of sulfur bacteria revealed a maximum population density in the uppermost 5 mm of the mat: 1.1 × 108 cells of sulfate reducers cm−3 sediment, 2.0 × 109 cells of chemolithoautotrophs cm−3 sediment, and 4.0 × 107 cells of anoxygenic phototrophs cm−3 sediment. Bacterial dynamics were studied by sulfate reduction rate measurements, both under anoxic conditions (dark incubation) and oxic conditions (incubation in the light), and determination of the vertical distribution of the potential rate of thiosulfate consumption under oxic conditions. Sulfate reduction rates in the top 5 mm of the sediment were 566 nmol cm−3 d−1 in the absence of oxygen, and 123 nmol cm−3 d−1 in the presence of oxygen. In the latter case, the maximum rate was found in the 5–10-mm depth horizon (361 nmol cm−3 d−1). Biological consumption of amended thiosulfate was rapid and decreased with depth, while in the presence of molybdate, thiosulfate consumption decreased to 10–30% of the original rate.  相似文献

Bacteria were separated from raw meat homogenate by a simple three-stage process. Centrifugation (10 s at 2000 g) removed coarse particles; stirring with the cation exchange resin Bio-Rex 70 removed smaller particles and filtration through 0.22 μm membranes removed soluble materials. By this process 70—80% of the microbial populations of meat homogenates were consistently isolated on the filters. A linear relationship was found between log10 microbial ATP and log10 colony count of meat over the range 105—109 cfu/g. The value of ATP/cfu for meat samples was within the range previously reported for pure cultures. These data indicated that ATP extracted from the filters originated from bacteria in the meat samples. Several samples can be analysed simultaneously in an elapsed time of 20—25 min. The variability associated with estimates of both colony counts and ATP levels has been determined.  相似文献

The occurrence of mycobacteria was studied in aerobic brook sediments from 39 drainage areas in Finland. The culturable counts of mycobacteria were related to climatic conditions, characteristics of the drainage area, chemical characteristics of the sediment and water, culturable counts of other heterotrophic bacteria, and microbial respiration rate in the sediment. The counts of mycobacteria varied from 1·1 × 102 to 1·5 × 104 cfu g−1 dry weight of sediment. They correlated positively with the proportion of the drainage area consisting of peatland, total content of C, content of Pb, microbial respiration rate in the sediment, and chemical oxygen demand of the water. The correlations of the mycobacterial counts with pH of sediment and alkalinity of water were negative. The results of the present sediment study and of the forest soil study published earlier strongly suggest that an increase in acidity increases the counts of mycobacteria and decreases the counts and activity of other heterotrophic bacteria. Mycobacterial counts were more than 100 times higher (per dry weight) in forest soils with pH 3·5–4·3 than in sediments with pH 4·5–6·3.  相似文献

Abstract This paper describes the microbial ecosystem found on the leaves of Atriplex halimus , a salt-excreting plant in the central Negev highlands of Israel. Because of the regular nightly occurence of dew at this location, these leaves undergo a diurnal wetting so that phylloplane microorganisms experience large fluctuations in salinity and water activity, as well as tolerate repeated desiccation. During the dry season, in the late spring and summer, a significant amount of salts and organic material coats the leaf surface. During dew events the salt concentration at the leaf surface was calculated to be > 0.4 M. Direct counts of the respiring bacteria on the leaf surface ranged from 1.06×104 to 5.06×105 per cm2. Using a variety of media it was shown that there was limited bacterial diversity which could be cultured, with greater than 90% of the isolates being orange colored Gram-negative rods. Viable counts ranged from 0.32 to 2.32×104 bacteria per cm2 of A. halimus leaf surface. No bacteria capable of nucleating ice were recovered in these studies. The dominant orange pigmented bacterium, identified as a halotolerant Pseudomonas sp., grew optimally at 30°C and at 5% NaCl and was capable of growth in media containing up to 20% NaCl. This bacterium could grow on a variety of organic compounds, including some associated with plant materials. The leaf bacteria were desiccation-tolerant when on the leaf surface or when directly washed off the leaves, but much less so when in isolatd culture. A major component of the tolerance to desiccation is probably related to the compounds on the leaf surface.  相似文献

Groundnut cake ('kulikuli') purchased from four major markets in Ibadan, Oyo State, Nigeria were analysed for aflatoxin B, and associated rnycoflora. In all but two of the samples aflatoxin B, concentrations were between 20 pg/kg and 455 pg/ kg. Mould counts were low (1·0 × 1024·40 × 102 colonies/g). Eight mould species were isolated. Of these Aspergillus niger, Paecilomyces oarioti, Aspergillus flavrs and Fusarium moniliforme dominated in decreasing sequential order. The results obtained show that groundnut cake on sale in Ibadan markets is unacceptable for animal feed rations and human consumption and there is a need for some form of quality control and decontamination before usage.  相似文献

Abstract The fate of a Bacillus amyloliquefaciens with the recombinant plasmid pSB20 sprayed on the phyllosphere of grass, and of a Tn 5 marked Pseudomonas syringae sprayed on the phyllosphere of bush beans was studied in planted soil microcosms. B. amyloliquefaciens showed a decline from 1.5×108 to 3.1×102 cfu g−1 on the phylloplane of grass in the course of the experiment. B. amyloliquefaciens was easy to follow by selective cultivation due to the complete absence of bacterial background growth. Southern blot hybridization of Hin dIII digested genomic DNA showed plasmid restriction patterns identical with pSB20 indicating high plasmid stability. In total DNA extracts from phyllosphere bacteria the recombinant plasmid was detectable by Southern blot hybridization up to 6×104 cfu g−1 (wet weight). Counts of hybridizing colonies showed that P. syringae established on the phyllosphere of bush beans at between 5×103 and 4×106 cfu g−1 fresh weight. During senescence of the bean plants the strain was no longer detectable by selective cultivation and subsequent colony hybridization. In contrast, Tn5 marked DNA was detected after PCR amplification over the whole period of the experiment.  相似文献

The distribution of microbial populations that decomposed sugar, cellulose and lignin-related substrates was examined in a beech Fagus grandifolia Ehrh. and maple Acer saccharum Marsh. dominated woodlot developed on glacial till. The topography of the woodlot, characterized by rises, depressions and more extensive level areas about 1 m in diameter with a 0.5 m vertical maximum, produced a mosaic of decomposer habitats designated as high, level and low sites.
In general, populations of sugar, cellulose and lignin decomposing organisms (based on ten estimates made from April to October) were two to four times higher in litter and soil samples from low sites than those from high sites. Sugar decomposing bacteria in litter were most abundant at all topographic sites. 135 × 106 g−1 dry litter at high sites, 396 × 106 g−1 at level sites and 456 × 106 g−1 at low sites; lignolytic fungi were least abundant, 391 × 102 g−1 dry litter at high sites. 700 × 106 g−1 at level sites and 954 × 102 g−1 at low sites. Numbers of microbial decomposers in the topographic sites were correlated with organic matter content. Distribution of fungal genera did not appear to be related to topographic site. Most populations examined showed two numerical peaks, one in late May or June and one in late September or October. It is suspected that these peaks were influenced by the coincident timing of favourable physical conditions and priming by soluble nutrients leached from litter.  相似文献

Abstract Viable counts and activities of sulfate-reducing bacteria were determined in the oral cavities of 12 healthy volunteers. Of these, 10 harboured viable sulfate-reducing bacteria populations. Six separate sites were sampled: the posterior tongue, anterior tongue, mid buccal mucosa, vestibular mucosa, supragingival plaque and subgingival plaque. Sulfate-reducing bacteria occurred in all areas, with the highest incidence in supragingival plaque. Viable counts and sulfate-reducing activities in each of the regions varied from 0 to 108 cfu (g wet weight)−1 and from 0 to 50 nmol (g wet weight) −1 h−1, respectively. As sulfate-reducing bacteria can be detected in the oral cavity, they may potentially be involved in terminal oxidative processes carried out by the microflora of the mouth.  相似文献

Flowering stems of four rose cultivars (Sonia, Madelon, Jacaranda and Frisco) were placed in aqueous suspensions of bacteria at 104 and 108 colony-forming units (cfu) ml-1 for 24 h at 5C, then stored dry or held in water for 24 h at 8C and subsequently placed in vase-water at 20C. The effects of these treatments on vase-water uptake were similar to the effects on flower opening. In Sonia and Madelon roses flower opening was negatively affected both by 108 cfu ml-1 of bacteria and by dry storage. No effect was found at 104 cfu ml-1, but this concentration had a detrimental effect on flower opening when combined with dry storage. Although flower development in Jacaranda roses was not affected by the bacteria treatments it was inhibited by dry storage. This inhibition was progressively greater when the stems had previously been pulse-treated with a larger number of bacteria. Flower opening in Frisco roses was not affected by even the highest concentration of bacteria, nor by the period of dry storage. It is concluded that placing flowers in water containing bacteria (up to 108 cfu ml-1) may not always have a negative effect on flower development in cut rose flowers but, together with the effects of dry storage, the presence of even a low number of exogenous bacteria (104 cfu ml-1) inhibits the development in several cultivars. Such bacterial counts are nearly always found in samples of water used for standing roses during distribution to the consumers.  相似文献

The structure of six Irish kefir samples was studied in the electron microscope, and the microbial composition and fermentation kinetics during growth in 10% reconstituted skim milk at 21°C. The microbial composition of the six samples was similar; at the end of the fermentation the counts of lactococci, leuconostocs, lactobacilli, acetic acid bacteria and yeasts were 109, 108, 5 × 106, 105 and 106 ml−1 respectively; the levels of acetic acid bacteria and lactobacilli showed some intersample differences. Lactate was the major metabolite followed in order by ethanol, acetate and acetoin. The final concentrations of L-lactate produced (66–90 mmol kg−1) were 10-fold higher than those of D-lactate. Acetate and ethanol concentrations varied from 4 to 14 and 2 to 40 mmol kg−'1 respectively. The rates of citrate utilization and concentration of acetoin produced during growth differed between samples. Scanning electron microscopy showed not only variation between the interior and exterior of the sample but also large variation between different sections of the interiors and exteriors of the same sample. Long and short, and straight and curved rods and yeasts were seen in all samples, the curved rods observed in the interior, but lactococci were seen on the surface of only one sample.
