Xenarthran osteoderms are integumentary bones with high fossilization potential presenting a high degree of morphological and histological diversity. Here, new data on the osteoderms histology of two glyptodonts, Panochthus and Neuryurus are presented. The poor spatial organization of the mineralized fibers and a large trabecular area in the middle zone identified in Neuryurus indicate a different bone pattern than the one found in Panochthus, which is mainly characterized by a middle zone with less spongiosa. Through the Bone Profiler program, the degree of compactness of the specimens was obtained, with about 70% for Neuryurus sp. and approximately 90% for Panochthus sp., showing the difference in bone pattern. These values confirm the visible difference in the histological patterns of these taxa, especially in the middle zone. This work demonstrates the microstructural variation studied in osteoderms and shows the importance of paleohistology as a starting point for a better understanding of extinct taxa.  相似文献

In the tropical rain forest in southeastern Yunnan, China, the members of Angiopteris Hoffm. and Archangiopteris Christ et Gies. are often growing side by side in great abundance. In January 1978 the second writer made a botantical trip there and collected in one locality ample herbarium material of an Angiopteris, which is identical with A. sparsisora Ching (ined.), based upon type specimens gathered by Mr. S. K. Wu from the same locality in 1962. To our great surprise, however, this distinct taxon reveals a number of important morphological characteristics intermediate between Angiopteris and Archangiopteris as diagnosed above. The present species resembles Archangiopteris in small build with creeping dorsiventral rhizome, unigeniculate, thin, smooth stipe in pinnate to bipinnatifid fronds, sparingly clad in linear-lanceolate peltately affixed scales with variously fimbriate margin, lamina 1—2 pinnate or subbipinnate, pinnules distinctly and shortly stipitate, base cuneate or broadly cuneate, sori of different length separated from each other by broad spaces, and rather far away (2—3 mm) from the margin, paraphyses few, rudimentary and short, but, on the other hand, it also similar to Angiopteris in usually bipinnate fronds, the petiole of pinnae with swollen base (blackish upon drying), short (2—4 mm long) linear sori consisting of 5—16 pairs of sporangia and globulax spores with densely tubucular sporoderms. All the above characteristics combined clearly indicate the species is in all likelihood a natural hybrid between Angiopteris and Archangiopteris, pending cytological investigation and the reason for presently placing it in Angiopteris rather than in Archangiopteris is because of the fact that in overall impression the newtaxon appears more like the former than the latter.  相似文献

Summary Four new species of hystrignathid nematodes are described from the midgut caeca of Passalus unicornus (Coleoptera: Passalidae) from St. Lucia, West Indies. Artigasia horridospina n.sp. is differentiated from A. dispar van Waerebeke, 1973 by virtue of fewer rows of spines in the oesophageal region and in possessing lateral alae, and from A. insignia Travassos & Kloss, 1958 in the extent of the oesophageal spiens and in having the lateral alae commence more anteriorly. The first annule is also much shallower than that of A. insignia. Longior semialata n.sp. is differentiated from L. longicollis Travassos & Kloss, 1958 on body length, b ratio, egg size and the starting point of the lateral alae. Mentecle magnifica n.sp. differs from M. parvus Travassos & Kloss, 1958 in having the spines of the oesophageal region extending much further down the body and in having ridged eggs. It is also longer and relatively thinner. Paraxyo ensicrinatus n.sp. differs from P. monodelpho Travassos & Kloss, 1958 in having a proportionately longer tail. The spines in the oesophageal region also extend considerably further posteriorly than in P. monodelpho.Two types of male associated with the above nematodes are described and illustrated, but not assigned to females. A carotenoid-accumulating bacterium isolated from soil, identified as a Sphingomonas sp., grew at 0.18 h(-1) and produced 1.7 mg carotenoids g(-1) dry cell, among which beta-carotene (29% of total carotenoids) and nostoxanthin (36%). A mutant strain, obtained by treatment with ethyl methanesulfonate, accumulated up to 3.5 mg carotenoids g(-1) dry cell. Accumulation of beta-carotene by this strain depended on the oxygenation of the growth medium, with maximal accumulation (89%) occurring under limiting conditions. Beta-carotene accumulation could be further enhanced by incubating the cells in the presence of glycerol (either not or only slowly assimilated) and yeast extract resulting in an accumulation of 5.7 mg beta-carotene g(-1) dry cell wt. The strain used lactose as carbon source with similar biomass and carotenoid production, providing a viable alternative use for cheese whey ultra-filtrate.  相似文献

A taxonomic study was conducted on 16 bacterial strains isolated from wild Adélie penguins (Pygoscelis adeliae) from Seymour (Marambio) Island and James Ross Island. An initial screening by repetitive sequence-based PCR fingerprinting divided the strains studied into four coherent groups. Phylogenetic analysis based on 16S rRNA gene sequences assigned all groups to the genus Corynebacterium and showed that Corynebacterium glyciniphilum and Corynebacterium terpenotabidum were the closest species with 16S rRNA gene sequence similarities between 95.4 % and 96.5 %. Further examination of the strains studied with ribotyping, MALDI-TOF mass spectrometry, comprehensive biotyping and calculation of average nucleotide identity and digital DNA–DNA hybridisation values confirmed the separation of the four groups from each other and from the other Corynebacterium species. Chemotaxonomically, the four strains P5828T, P5850T, P6136T, P7210T representing the studied groups were characterised by C16:0 and C18:1 ω9c as the major fatty acids, by the presence of meso-diaminopimelic acid in the peptidoglycan, the presence of corynemycolic acids and a quinone system with the predominant menaquinone MK-9(H2). The results of this study show that the strains studied represent four new species of the genus Corynebacterium, for which the names Corynebacterium antarcticum sp. nov. (type strain P5850T = CCM 8835T = LMG 30620T), Corynebacterium marambiense sp. nov. (type strain P5828T = CCM 8864T = LMG 31626T), Corynebacterium meridianum sp. nov. (type strain P6136T = CCM 8863T = LMG 31628T) and Corynebacterium pygosceleis sp. nov. (type strain P7210T = CCM 8836T = LMG 30621T) are proposed.  相似文献

A novel group of hyperthermophilic rod-shaped motile methanogens was isolated from a hydrothermally heated deep sea sediment (Guaymas Basin, Gulf of California) and from a shallow marine hydrothermal system (Kolbeinsey ridge, Iceland). The grew between 84 and 110°C (opt: 98°C) and from 0.2% to 4% NaCl (opt. 2%) and pH 5.5 to 7 (opt: 6.5). The isolates were obligate chemolithoautotrophes using H2/CO2 as energy and carbon sources. In the presence of sulfur, H2S was formed and cells tended to lyse. The cell wall consisted of a new type of pseudomurein containing ornithin in addition to lysine and no N-acetylglucosamine. The pseudomurein layer was covered by a detergent-sensitive protein surface layer. The core lipid consisted exclusively of phytanyl diether. The GC content of the DNA was 60 mol%. By 16S rRNA comparisons the new organisms were not related to any of the three methanogenic lineages. Based on the physiological and molecular properties of the new isolates, we describe here a new genus, which we name Methanopyrus (the methane fire). Worldwide glaciers are annually retreating due to global overheating and this phenomenon determines the potential lost of microbial diversity represented by psychrophilic microbial population sharing these peculiar habitats. In this context, yeast strains, all unable to grow above 20°C, consisting of 42 strains from Antarctic soil and 14 strains isolated from Alpine Glacier, were isolated and grouped together based on similar morphological and physiological characteristics. Sequences of the D1/D2 and ITS regions of the ribosomal DNA confirmed the previous analyses and demonstrated that the strains belong to unknown species. Three new species are proposed: Mrakia robertii sp. nov. (type strain CBS 8912), Mrakia blollopis sp. nov. (type strain CBS 8921) and a related anamorphic species Mrakiella niccombsii sp. nov. (type strain CBS 8917). Phylogenetic analysis of the ITS region revealed that the new proposed species were closely related to each other within the Mrakia clade in the order Cystofilobasidiales, class Tremellomycetes. The Mrakia clade now contains 8 sub-clades. Teliospores were observed in all strains except CBS 8918 and for the Mrakiella niccombsii strains.  相似文献

Protaspis is an enigmatic genus of marine phagotrophic biflagellates that have been tentatively classified with several different groups of eukaryotes, including dinoflagellates, euglenids, and cercomonads. This uncertainty led us to investigate the phylogenetic position of Protaspis grandis n. sp. with ultrastructural and small subunit (SSU) rDNA sequence data. Our results demonstrated that the cells were dorsoventrally flattened, shaped like elongated ovals with parallel lateral sides, 32.5-55.0 mum long and 20.0-35.0 mum wide. Moreover, two heterodynamic flagella emerged through funnels that were positioned subapically, each within a depression and separated by a distinctive protrusion. A complex multilayered wall surrounded the cell. Like dinoflagellates and euglenids, the nucleus contained permanently condensed chromosomes and a large nucleolus throughout the cell cycle. Pseudopodia containing numerous mitochondria with tubular cristae emerged from a ventral furrow through a longitudinal slit that was positioned posterior to the protrusion and flagellar apparatus. Batteries of extrusomes were present within the cytoplasm and had ejection sites through pores in the cell wall. The SSU rDNA phylogeny demonstrated a very close relationship between the benthic P. grandis n. sp. and the planktonic Cryothecomonas longipes. These ultrastructural and molecular phylogenetic data for Protaspis indicated that the current taxonomy of Protaspis and Crythecomonas is in need of re-evaluation. The composition and identity of Protaspis is reviewed and suggestions for future taxonomic changes are presented. Problems within the genus Cryothecomonas are highlighted as well, and the missing data needed to resolve ambiguities between the two genera are clarified.  相似文献

A novel α-amylase, AmyM, was purified from the culture supernatant of Corallococcus sp. strain EGB. AmyM is a maltohexaose-forming exoamylase with an apparent molecular mass of 43 kDa. Based on the results of matrix-assisted laser desorption ionization–time of flight mass spectrometry and peptide mass fingerprinting of AmyM and by comparison to the genome sequence of Corallococcus coralloides DSM 2259, the AmyM gene was identified and cloned into Escherichia coli. amyM encodes a secretory amylase with a predicted signal peptide of 23 amino acid residues, which showed no significant identity with known and functionally verified amylases. amyM was expressed in E. coli BL21(DE3) cells with a hexahistidine tag. The signal peptide efficiently induced the secretion of mature AmyM in E. coli. Recombinant AmyM (rAmyM) was purified by Ni-nitrilotriacetic acid (NTA) affinity chromatography, with a specific activity of up to 14,000 U/mg. rAmyM was optimally active at 50°C in Tris-HCl buffer (50 mM; pH 7.0) and stable at temperatures of <50°C. rAmyM was stable over a wide range of pH values (from pH 5.0 to 10.0) and highly tolerant to high concentrations of salts, detergents, and various organic solvents. Its activity toward starch was independent of calcium ions. The Km and Vmax of recombinant AmyM for soluble starch were 6.61 mg ml−1 and 44,301.5 μmol min−1 mg−1, respectively. End product analysis showed that maltohexaose accounted for 59.4% of the maltooligosaccharides produced. These characteristics indicate that AmyM has great potential in industrial applications.  相似文献

Pseudorhabdosynochus cupatus (Young, 1969) is characterised by small lamellosquamodiscs made up of central telescopic rings and peripheral rows of rodlets, and a sclerotised vagina in the shape of a thin-walled coiled tube with three accessory chambers. Two new species of Pseudorhabdosynochus are herein described from epinepheline fish off New Caledonia, South Pacific, and are considered part of the 'P. cupatus group'. P. cyathus n. sp. from Epinephelus howlandi has lamellosquamodiscs made up of four telescopic rings and four rows of separate rodlets; there are dorsal tegumental scales. This species was found only in young hosts; older hosts harbour P. venus Hinsinger & Justine, 2006. P. calathus n. sp. from Epinephelus rivulatus has lamellosquamodiscs made up of four telescopic rings and five rows of separate rodlets; the tegument is smooth. The three species, P. cupatus, P. calathus and P. cyathus, are strictly species specific; although morphologically very similar, they can be distinguished by features of the tegumental scales and measurements of the vagina and haptoral hard parts. P. melanesiensis (Laird, 1958) from E. merra is close to the 'P. cupatus group' according to the morphology of its sclerotised vagina but is distinguished by its squamodiscs which lack telescopic rings. Rare specimens found in E. merra are tentatively attributed to P. coioidesis Bu et al., 1999. A paratype of P. coioidesis is figured for comparison. E. merra, E. fasciatus, E. rivulatus and E. howlandi each have a Pseudorhabdosynochus species of the P. cupatus lineage: these are the dominant species in each host. In addition, E. merra, E. fasciatus and E. howlandi harbour a rare species of Pseudorhabdosynochus, respectively P. cf. coioidesis, P. caledonicus Justine, 2005 and P. venus; these rare species are morphologically unrelated to each other and to the 'P. cupatus group'.  相似文献

A new species of the isoetalean leaf genus Lepidophylloides is described. This Guadalupian species from the Paraná Basin, Lepidophylloides corumbataensis, found as silicified microphylls in the Corumbataí Formation in the municipality of Piracicaba, São Paulo state, in southern Brazil, is the first species of the genus to be formally described for Brazil, and possibly the first for Gondwanaland. This is also the first time that surviving palisade tissue has been found in a species of Lepidophylloides. The new species is compared to known species and shown to be distinct from all previously described from Euramerican and Cathaysian floras. The organization of the xylem in arch-like or crescent-shaped bundles suggests a relation to Cathaysian species. The close association with Lycopodiopsis derbyi stems (which have true leaf scars and must have suffered leaf abscission) suggests that they are different organs from the same species.  相似文献

We describe 2 new species of leucochloridiid-like brachylaimoid digeneans parasitizing a variety of birds in the Area de Conservación Guanacaste, Costa Rica, each of which we assign to a new genus. According to Pojmanska's (Pojmanska, T. 2002a. Superfamily Brachylaimoidea Joyeux & Foley, 1930. In Keys to the Trematoda, D. I. Gibson, A. Jones, and R. A. Bray [eds.]. CAB International and The Natural History Museum, London, U.K., p. 31-36.) key for the Brachylaimoidea, we are unable to place either species in any family. One species most closely resembles members of Leucochloridium by having well-developed suckers, lacking an esophagus, and having cecal shoulders, gonads at the posterior end, and the genital pore at posterior end of body but differs by having symmetrical testes, a posttesticular ovary, and a terminal genital pore; thus, we propose the genus Bakkeius for it. The second new genus resembles members of Michajlovia by having ventral genital pores but differs by having extracecal uterine loops in the forebody, a cirrus sac containing the pars prostatica and seminal vesicle, and gland cells surrounding the genital pore; thus, we propose Pojmanskia for it. These new genera must currently be treated as incertae sedis according to Pojmanska (op. cit.); however, we feel that future phylogenetic analyses will require emendation of the family diagnosis for Leucochloridiidae to include those taxa with terminal and ventral genital pores and with preovarian testes.  相似文献

A solventogenic strain of Clostridium sp. BOH3 produces extracellular α-amylase (7.15 U/mg protein) in reinforced clostridial medium supplemented with sugarcane bagasse hydrolysate (1 % w/v) and a small amount of starch (0.1 % w/v), which is essential for the expression of α-amylase. In the presence of α-amylase, BOH3 utilizes starch directly without any pretreatment and produces butanol almost equivalent (~90 %) to the production of butanol from glucose. α-Amylase can be purified from culture supernatant by using one-step weak anion exchange chromatography with a yield of 43 %. In peptide fingerprinting analysis, this enzyme shows homology with α-amylase produced by Clostridium acetobutylicum ATCC824. However, the molecular weight is 54 kDa, which is smaller than α-amylase of ATCC824 (84 kDa). This enzyme has optimum temperature at 45–50 °C and optimum pH at 4.5–5.5. Under this condition, the enzyme activity is 91.32 U/mg protein, and its K m and V max values are 1.71?±?0.02 mg/ml and 96.13?±?0.15 μmol/min/mg protein, respectively. Activity of this α-amylase can be enhanced (>1.5 times) by addition of Ca2+ and Co2+ and its activity can be maintained at an acidic pH (pH 3–5) for about 24 h. These unique characteristics suggest that this enzyme can be used for saccharification of starch for production of biofuel in one pot.  相似文献

Multiple alignments of primary structures of many kinds of prenyltransferases that participate in the most fundamental prenyl-chain backbone synthesizing process in isoprenoid biosynthesis showed seven conserved regions in the primary structures of (E)-prenyl diphosphate synthases. However, no information has been available about the structures of (Z)-prenyl diphosphate synthases until our recent isolation of the gene for the undecaprenyl diphosphate synthase of Micrococcus luteus B-P 26.

The amino acid sequence of the (Z)-prenyl diphosphate synthase is totally different from those of (E)-prenyl chain elongating enzymes. Protein data base searches for sequences similar to that of the undecaprenyl diphosphate synthase yielded many unknown proteins which have not yet been characterized. Two of the proteins have recently been identified as the undecaprenyl diphosphate synthase of Escherichia coli and the dehydrodolichyl diphosphate synthase of Saccharomyces cerevisiae, indicating that there are three highly conserved regions in the primary structure of (Z)-prenyl chain elongating enzymes.  相似文献
