In this study, a quinone profiling method was applied to clarify the differences in community structure between suspended and sessile microorganisms in rivers. The compositions of microbial quinone of 6 sites for 4 rivers were analyzed. Ubiquinone (UQ)-8, UQ-10, menaquinone (MK)-7, and plastoquinone (PQ)-9 were observed in all samples of suspended and sessile microorganisms for the sites investigated. The dominant quinone species in suspended microorganisms was ubiquinone, and that in sessile microorganism was photosynthetic quinones (namely PQ-9 and vitamin K1). This indicated that aerobic bacteria were abundant in the suspended microorganisms, and photosynthetic microorganisms such as micro-algae and cyanobacteria dominated in the sessile microorganisms. The quinone concentration in the river waters tested, which reflects the concentration of suspended microorganisms, ranged from 0.045 to 1.813 nmol/L. The microbial diversities of suspended and sessile microorganisms calculated based on the composition of all quinones were in the range from 3.4 to 7.5, which was lower than those for activated sludge and soils. Moreover, the diversity of heterotrophic bacteria for sessile microorganisms in the rivers was higher than that for the suspended microorganisms. 相似文献

昆虫体外共生菌是指能在体外与宿主发生互利共生关系的微生物。体外共生菌虽然不如肠道微生物那样普遍存在于昆虫中,但也在宿主生长发育过程中扮演着重要的角色。昆虫体外共生菌一般寄生于昆虫体表或体内特异器官(如储菌器),在特定时期转移到植物组织中。体外共生菌产生的挥发物能作为宿主定位寄主植物的信号物质,能为宿主提供生长发育所需的营养物质,还参与了宿主体外免疫。对昆虫体外共生菌的研究,不仅能进一步揭示昆虫与微生物之间的互作关系,丰富昆虫共生菌的研究,还能从共生菌的角度探索害虫引诱剂和昆虫免疫豁免机制。本文对昆虫体外共生菌寄生方式、传播途径、对宿主的影响等研究成果进行了综述,旨在为害虫综合防控提供新思路。 相似文献

天山北坡甜菜内生菌分离鉴定及其动态变化 总被引:3,自引:0,他引:3
对新疆昌吉和石河子两地种植的甜菜内生菌进行了分离、鉴定和分析,结果表明甜菜内生菌多属于细菌,其中假单胞菌 (Pseudomonas sp. )和芽孢菌类(Bacillus sp.)的分离频率分别在33.2%~59.2%和12.7%~28.1%,是甜菜植株中的优势内生菌群.16S rDNA 和 ITS 序列同源性比较和系统发育分析表明内生菌具有丰富的多样性.根中内生菌的多样性高于茎、叶,昌吉地区种植的甜菜中分离出的内生菌种类较多.从感病品种及生长不良甜菜植株中分离出的内生菌种类比较丰富.通过回接分离及利用扫描电镜观察内生菌在植物体内分布发现,内生菌能够定殖于甜菜块根. 相似文献

【目的】揭示恶臭假单胞菌(Pseudomonas putida) Y-9在氨氧化过程中主动调节胞外和胞内pH稳态机制。【方法】在初始pH为7.19和9.40的硝化培养基中培养Y-9生长48 h,利用代谢组学对比分析Y-9氨氧化过程中的显著差异代谢产物并预测解离常数(pKa);结合转录组学对比分析Y-9氨氧化过程中的显著差异调控基因。【结果】Y-9在初始pH为7.19的相对酸性条件下,产生麦芽糖醇提高胞外pH;通过上调脱氨酶、脱亚胺酶和阳离子转运相关基因在相对酸性环境中的表达来维持细胞内pH稳定性。在初始pH为9.40的碱性条件下,产5-氨基戊酸3和草氨酸等有机酸及酸性物质降低胞外pH;通过调控NADH脱氢酶、细胞色素、ATP合酶和氨基酸转运相关基因的表达来维持细胞内酸度,应对碱性环境。【结论】本研究结果首先发现了Y-9具有稳定胞外pH的能力,探讨了其胞内pH稳态机制,拓展了对微生物与环境相互作用的认知,为进一步认识微生物脱氮过程中系统pH稳定机理提供了理论依据。 相似文献

A mathematical model accounting for key parameters as microbial propagation, metabolite formation, dispersion, microbial chemotaxis and water flooding has been proposed to simulate the transport of microorganisms and their metabolites in a cylindrical pore with oil adhered to its inside surface. The model focuses on the transport and the concentration distributions of microorganisms and their metabolites in the cylindrical pore, especially the concentrations that on the oil-water interface. Results from the present model indicate that microorganisms and their metabolites assembled on the oil-water interface during the water flooding process, and the concentration gradients of microorganisms and their metabolites from the pore center region up to the oil-water interface in radial direction of the cylindrical pore were consequently formed. Equilibrium concentrations of microorganisms and their metabolites in the cylindrical pore were obtained when water flow rate within a certain scope, and there existed a critical water flow rate at which the maximum equilibrium concentration of microorganisms on the oil-water interface was developed. Investigations carried out in this study may provide better understanding on the transport mechanism of microorganisms in porous media. 相似文献

近年来抗生素耐药性问题日趋严重,患癌人数也在逐年增加,亟需开发新型药物。嗜盐微生物作为一类特殊的极端环境微生物,具有代谢多样性丰富、营养需求较低和能适应恶劣条件等特点,是发现新型药物的希望。目前,国内外学者已从嗜盐微生物中分离出了多种代谢产物和酶,具有明显的抗菌和/或抗肿瘤等活性。文中综述了嗜盐微生物及其相关产物在抗菌、抗炎、抗肿瘤、抗氧化、生物医学材料以及药物载体等生物医学方面的作用,尤其对近年来在嗜盐微生物中发现的新型抗菌和抗肿瘤物质以及嗜盐微生物特有的代谢产物四氢嘧啶等进行了总结,并对其后续在生物医药领域的开发和产业化应用进行了展望。 相似文献

Cold environments, including polar and alpine regions, are colonized by a wide diversity of microorganisms able to thrive at low temperatures. There is evidence of a wide range of metabolic activities in alpine cold ecosystems. Like polar microorganisms, alpine microorganisms play a key ecological role in their natural habitats for nutrient cycling, litter degradation, and many other processes. A number of studies have demonstrated the capacity of alpine microorganisms to degrade efficiently a wide range of hydrocarbons, including phenol, phenol-related compounds and petroleum hydrocarbons, and the feasibility of low-temperature bioremediation of European alpine soils by stimulating the degradation capacity of indigenous microorganisms has also been shown. 相似文献

Recent advances in technology have highlighted the vast and previously unknown genetic information that is contained in extant marine microorganisms, from new protein families to novel metabolic processes. Scale insects, like other plant sap-consumers, are host to symbiotic microorganisms which provide them with the substances missing from their diet. In contrast to most scale insects, Kermes quercus (Linnaeus) was regarded as asymbiotic. Our histological and ultrastructural observations show that in the body of the feeding stages of K. quercus collected in two locations (Warsaw and Cracow), numerous yeast-like microorganisms occur. These microorganisms were localized in the cytoplasm of fat body cells. The yeast-like microorganisms were observed neither in other organs of the host insect nor in the eggs. These microorganisms did not cause any damage to the structure of the ovaries and the course of oogenesis of the host insect. The females infected by them produced about 1300 larvae. The lack of these microorganisms in the cytoplasm of eggs indicates that they are not transmitted transovarially from mother to offspring. Molecular analyses indicated that the microorganisms which reside in the body of K. quercus are closely related to the entomopathogenic fungi Cordyceps and Ophiocordyceps, which belong to the Sordariomycetes class within the Ascomycota. The role of yeast-like microorganisms to their host insects remains unknown; however, it has been suggested that they may represent newly acquired symbionts. 相似文献

A membrane filtration/bioluminescence system was developed for the differentiation and quantitation of mixed populations of microorganisms. Samples containing microorganisms were filtered through two membrane filters of descending pore size. The microorganisms retained on the filter contain ATP that can be extracted and measured on the filter via the firefly luciferase-luciferin bioluminescence assay. Results, obtained in less than 20 min, show a good correlation (r greater than or equal to 0.95) between the light produced and the number of organisms in the sample. Using these techniques, Escherichia coli can be separated from yeast or mold and measured in samples containing both microorganisms. When lysostaphin is used to selectively lyse Staphylococci on the filter, the specific quantification of these bacteria among other microorganisms can also be accomplished. The filtration/bioluminescence technique offers the potential of being a rapid and sensitive method to differentiate and detect microorganisms, by selective sizing or lysing, in a variety of samples. 相似文献

研究旨在分析玫瑰高原鳅(Triplophysa rosa)肠道微生物的结构组成和多样性,探索其肠道微生物的潜在功能。提取了5尾玫瑰高原鳅的肠道总DNA,运用Illumina Miseq平台对肠道微生物16S rRNA的V3—V4区进行了测序,统计样品肠道微生物的操作分类单元(Operational Taxonomic Units, OTUs)数量,分析物种组成、丰度及Alpha多样性,并预测肠道微生物的功能。结果显示,玫瑰高原鳅的肠道微生物有19门、31纲、87目、146科、253属、 320种, 451个OTUs。在门水平上,优势菌群为变形菌门(Proteobacteria)、放线菌门(Actinobacteria)和拟杆菌门(Bacteroidetes);在属水平上,优势菌群为气单胞菌属(Aeromonas)、爱德华菌属(Edwardsiella)、邻单胞菌属(Plesiomonas)和希瓦氏菌属(Shewanella)。功能预测表明,肠道微生物编码的大多数基因与新陈代谢相关,其中"碳水化合物运输和代谢"和"氨基酸转运与代谢"功能类群的相对丰度较高。玫瑰高原鳅肠道内微生物组成复杂,... 相似文献

目前我国油田开发主要处于高含水后期,微生物驱提高石油采收率技术(MEOR)以低成本、环境友好等独特的优势引起了石油工业界的重视。实际上,经过半个多世纪的发展,MEOR已经成为提高采收率的重要前沿技术。高压是油藏的主要环境特征,在影响油藏微生物生存与活性等方面具有重要作用。本文从油藏及其微生物的主要特征、微生物对高压环境的适应机制以及高压下微生物降解烃的代谢特征等方面进行了综述。介绍了对油藏微生物资源、群落结构、微生物在油水相中分布的认识,微生物乳化原油机制,以及微生物在油藏厌氧环境中协同代谢、受温度和压力影响的特点,并列举了MEOR的矿场应用。在高压适应机制上,微生物主要通过改变和调整细胞膜结构、增加胞内脂质组分和表达胞内特殊酶等作用来实现对压力的适应;在高压下烃降解微生物代谢速率低于常压,而且耐压菌和嗜压菌具有不同的烃降解效率。 相似文献

Many animals, plants and protists contain non-parasitic microorganisms and these endosymbioses are widely assumed to be mutualistic. Most of the microorganisms possess metabolic capabilities, such as the ability to fix nitrogen, photosynthesize or degrade cellulose, that their partners utilize. The emergence of drug-resistant microorganisms is an important medical and social problem. Drug-resistant microorganisms are thought to grow selectively in the presence of antibiotics. Most clinically isolated drug-resistant microorganisms have mutations in the target genes for the drugs. While any of the many mutagens in the environment may cause such genetic mutations, no reports have yet described whether these mutagens can confer drug resistance to clinically important microorganisms. We investigated how environmental mutagens might be implicated in acquired resistance to antibiotics in clinically important microorganisms, which causes human diseases. We selected mutagens found in the environment, in cigarette smoke, or in drugs, and then exposed Pseudomonas aeruginosa to them. After exposure, the incidence of rifampicin- and ciprofloxacin-resistant P. aeruginosa strains markedly increased, and we found mutations in genes for the antibiotic-target molecule. These mutations were similar to those found in drug-resistant microorganisms isolated from clinical samples. Our findings show that environmental mutagens, and an anticancer drug, are capable of inducing drug-resistant P. aeruginosa similar to strains found in clinical settings. 相似文献

An in situ method for cultivating microorganisms using a double encapsulation technique 总被引:1,自引:0,他引:1
The lack of cultured microorganisms represents a bottleneck for advancement in microbiology. The development of novel culturing techniques is, therefore, a crucial step in our understanding of microbial diversity in general, and the role of such diversity in the environment, in particular. This study presents an innovative method for cultivating microorganisms by encapsulating them within agar spheres, which are then encased in a polysulfonic polymeric membrane and incubated in a simulated or natural environment. This method stimulates growth of the entrapped microorganisms by allowing them access to essential nutrients and cues from the environment. It allows for the discovery of microorganisms from dilutions that are 10–100-fold greater than possible with conventional plating techniques. Analysis of microorganisms grown in such spheres incubated in and on a number of different substrates yielded numerous novel ribotypes. For example, spheres incubated on the mucus surface of a Fungiid coral yielded numerous ribotypes, with only 50% sharing similarity (85–96%) to previously identified microorganisms. This suggests that many of the species represent novel ribotypes. Hence, the technique reported here advances our ability to retrieve and successfully culture microorganisms and provides an innovative tool to access unknown microbial diversity. 相似文献
