Nonstructural carbohydrate status and activities of ADP-glucose pyrophosphorylase (EC 2.7.7.27, ADPG pyrophosphorylase) and sucrose phosphate synthase (EC 2.4.1.14, SPS) were determined during ageing of tobacco ( Nicotiana tabacum L., cvs KY 14 and Speight G28) leaves sampled from control plants and from plants that had the apical meristem and subsequent axillary growth removed (detopped plants). Over the 30-day period shoot growth increased much more for control compared to detopped plants, but the increase in root growth was similar for both treatments. Dry matter and leaf area of the individual leaf used for enzyme and metabolite analysis were constant over time for controls but increased 5-fold for detopped plants. Ageing of control leaves was indicated by a progressive loss of chlorophyll and ribulose 1, 5-bisphosphate carboxylase (EC 4.1.1.39, Rubisco) activity; loss of these components was diminished for detopped plants. In contrast to chlorophyll and Rubisco activity, activities of ADPG pyrophosphorylase and SPS remained relatively constant over time for controls. Thus, under normal ageing conditions, changes in activities of ADPG pyrophosphorylase and SPS were not closely associated with changes in the standard senescence indicators chlorophyll and Rubisco activity. The activities of ADPG pyrophosphorylase and SPS were enhanced, relative to controls, within 6 days after applying the detopping treatment and activities remained high for the duration of the 30-day period. Detopping also led to increased concentrations of starch and sucrose, but the increases were not well correlated with changes in enzyme activities. The data indicated that the leaves of detopped plants functioned as both source leaves, with enhanced ability to synthesize carbohydrate, and sink leaves, with enhanced growth. Therefore, activities of ADPG pyrophosphorylase and SPS were more responsive to changes within an individual leaf than to changes in whole plant growth.  相似文献

To better understand the genetic controls of leaf senescence, a tobacco (Nicotiana tabacum L. cv. SR1) mRNA that is up-regulated during senescence was isolated by the cDNA-amplified restriction fragment polymorphism method and the cDNA was cloned. The mRNA coded for the early light-induced protein (ELIP), a member of the chlorophyll a/b-binding protein family that has been implicated in assembly or repair of the photosynthetic machinery during early chloroplast development and abiotic stress. A protein antigenically recognized by antibodies to ELIP appeared during senescence with kinetics similar to those of its mRNA. The mRNA, designated ELIP-TOB, was detected earlier when senescence was enhanced by leaf detachment and treatment with 1-amino-cyclopropane-1-carboxylic acid, and was detected later when senescence was retarded by benzyladenine. However, no ELIP-TOB mRNA was seen in the dark even though senescence was accelerated under these conditions. Furthermore, water stress and anaerobiosis stimulated the appearance of ELIP-TOB mRNA before losses of chlorophyll could be detected. We discuss the conditions that may lead to the up-regulation of ELIP-TOB during senescence and speculate as to the role of the gene product in this terminal phase of leaf development. Two of the major challenges in functional genomics are to identify genes that play a key role in biological processes, and to elucidate the biological role of the large numbers of genes whose function is poorly characterized or still completely unknown. In this study, a combination of large-scale expressed sequence tag sequencing, high-throughput gene silencing and visual phenotyping was used to identify genes in which partial inhibition of expression leads to marked phenotypic changes, mostly on leaves. Three normalized tobacco (Nicotiana tabacum) cDNA libraries were prepared directly in a binary vector using different tissues of tobacco as an RNA source, randomly sequenced and clustered. The Agrobacterium-tobacco leaf disc transformation system was used to generate sets of antisense or co-suppression transgenic tobacco plants for over 20 000 randomly chosen clones, each representing an independent cluster. After transfer to the glasshouse, transgenic plants were scored visually after 10-14 days for changes in growth, leaf form and chlorosis or necrosis. Putative hits were validated by repeating the transformation. This procedure is more stringent than the analysis of knockout mutants, because it requires that even a partial decrease in expression generates a phenotype. This procedure identified 88 validated gene/phenotype relations. These included several previously characterized gene/phenotype relationships, demonstrating the validity of the approach. For about one-third, a function could be inferred, but a loss-of-function phenotype had not been described previously. Strikingly, almost one-half of the validated genes were poorly annotated, or had no known function. For 77 of these tobacco sequences, a single or small number of potential orthologues were identified in Arabidopsis. The genes for which orthologues were identified in Arabidopsis included about one-half of the genes whose function was completely unknown. Comparison with published gene/phenotype relations for Arabidopsis knockout mutants revealed surprisingly little overlap with the present study. Our results indicate that partial gene silencing identifies novel gene/phenotype relationships, which are distinct from those uncovered by knockout screens. They also show that it is possible to perform these analyses in a crop species in which full genome sequence information is lacking, and subsequently to transfer the information to a reference species in which functional studies can be performed more effectively.  相似文献

烤烟冠层光谱参数与叶片叶绿素含量的相关分析   总被引:4,自引:0,他引:4  
为了明确烤烟冠层光谱参数与叶片叶绿素含量的相关性,测定了不同氮肥施用量条件下烤烟冠层光谱特征和烤烟鲜烟叶片叶绿素a(Chl-a)、叶绿素b(Chl-b)、类胡萝卜素(Cars)含量,并对光谱参数与叶绿素含量进行了相关分析和回归分析。结果表明:随着氮肥施用量增加,团棵期和旺长期鲜烟叶片的Chl-a、Chl-b和Cars含量均增加,可见光波段反射率降低、近红外波段反射率增加;而打顶期叶片的3种色素含量和光谱特征的变化规律不明显。可见光460~670nm范围内,460nm反射率与叶片叶绿素含量呈显著正相关,其他波段反射率与叶片叶绿素的含量呈显著负相关;近红外780~1260nm范围内,所有波段与叶片叶绿素含量的都呈显著正相关,1480nm反射率与叶片叶绿素含量呈显著负相关。反映Chl-a、Chl-b、Cars含量与光谱参数——比值植被指数(ratio vegetation index,RVI)定量关系的最佳回归方程分别为幂函数、幂函数和指数函数:Chl-a=0.250RVI(730,550)1.511,Chl-b=0.049RVI(730,550)1.841,Cars=0.0998e0.379RVI(730,550)。  相似文献

Peptidases in the extracellular space might affect the integrity of recombinant proteins expressed in, and secreted from, plant cells. To identify extracellular peptidases, we recovered the leaf intercellular fluid from Nicotiana tabacum plants by an infiltration-centrifugation method. The activity of various peptidases was detected by an in vitro assay in the presence of specific inhibitors, using BSA and human serum gamma-globulin as substrates. Peptidases were detected by 1- and 2-D zymography in a polyacrylamide gel containing gelatin as substrate. Proteolytic activity was observed over a wide range of molecular masses equal to, or higher than, 45 kDa. To identify the peptidases, the extracellular proteins were digested with trypsin and analyzed by LC and MS. Seventeen peptides showing identity or similarity to predicted plant aspartic, cysteine, and serine peptidases have been identified. The extracellular localization of a cysteine peptidase aleurain homolog was also shown.  相似文献

Green fluorescent protein (GFP) allows the direct visualization of gene expression and sub cellular localization of fusion proteins in living cells. Many GFP variants have been developed to solve stability and emission problems. In this report the localization of different GFP fusion proteins, targeted to vacuoles, was studied in Nicotiana tabacum cv SR1. Even if a strong emission variant of the plant adapted GFP was used, no fluorescence was detected in differentiated tissues of N. tabacum with few exceptions. This model plant does not appear a good experimental system for the use of GFPs as vacuolar markers compared to Arabidopsis thaliana. In spite of this, our observations have evidenced a peculiar pattern of separated vacuoles in guard cells, providing new elements in the understanding of the vacuolar system organization.  相似文献

A new method for isolation of quantities of mature pollen protoplasts in Nicotiana tabacum has been established. The first step was to germinate mature pollen in Brewbaker and Kwack medium containing 20% sucrose. When most of the pollen grains had just germinated short pollen tubes, they were transferred to an enzymatic solution for the second step. The enzymatic solution contained 1% pectinase, 1% cellulase, 0.5% potassium dextran sulfate, 1 mol/L mannitol, 0.4 mol/L sorbitol in Dx medium with or without 15% Ficoll. The enzymes firstly degraded the pollen tube wall and then the intine. As a result, intact pollen protoplasts were released with the isolation rate up to 50%-70%. Factors affecting pollen protoplast isolation during the germination and maceration of pollen grains were studied. The suceees depended on two key points:pollen germination duration and osmotieum concentration. The optimal germination duration was 30 rain at 30℃. When it was too long, long pollen tubes formed and subsequently, large number of subprotoplasts instead of whole protoplasts were yielded, as the case reported by previous investigators. The optimal concentration of mannitol and sorbitol in enzyme solution was as high as 1.4 mol/L in total. Lowering of the osmoticum concentration resulted in decrease of percentage of pollen protoplasts.  相似文献

Plant DNA is distinguished from the DNA of all other organisms by its high content of 5-methylcytosine (5mC). 5mC levels may amount to 30% of total cytosines, distributed between the sequences CG and CXG. The results presented here show that the methylation status of CXG sequences could be influenced by culturing tobacco tissues on subtoxic concentrations of ethionine. The hypomethylating effect of ethionine, evaluated as the capability of MspI or HpaII to cleave the DNA, proved to be rather specific for CCG and differed from that or 5-azacytidine which did not discriminate between CG and CXG sequences.  相似文献

Leaves of tobacco (Nicotiana tabacum) are covered with glandular trichomes that produce sucrose esters and diterpenoids in varying quantities, depending on cultivar type. The bicyclic diterpene Z‐abienol is the major labdanoid present in some oriental tobacco cultivars, where it constitutes a precursor of important flavours and aromas. We describe here the identification and characterization of two genes governing the biosynthesis of Z‐abienol in N. tabacum. As for other angiosperm labdanoid diterpenes, the biosynthesis of Z‐abienol proceeds in two steps. NtCPS2 encodes a class‐II terpene synthase that synthesizes 8‐hydroxy‐copalyl diphosphate, and NtABS encodes a kaurene synthase‐like (KSL) protein that uses 8‐hydroxy‐copalyl diphosphate to produce Z‐abienol. Phylogenetic analysis indicates that NtABS belongs to a distinct clade of KSL proteins that comprises the recently identified tomato (Solanum habrochaites) santalene and bergamotene synthase. RT‐PCR results show that both genes are preferentially expressed in trichomes. Moreover, microscopy of NtCPS2 promoter‐GUS fusion transgenics demonstrated a high specificity of expression to trichome glandular cells. Ectopic expression of both genes, but not of either one alone, driven by a trichome‐specific promoter in transgenic Nicotiana sylvestris conferred Z‐abienol formation to this species, which does not normally produce it. Furthermore, sequence analysis of over 100 tobacco cultivars revealed polymorphisms in NtCPS2 that lead to a prematurely truncated protein in cultivars lacking Z‐abienol, thus establishing NtCPS2 as a major gene controlling Z‐abienol biosynthesis in tobacco. These results offer new perspectives for tobacco breeding and the metabolic engineering of labdanoid diterpenes, as well as for structure–function relationship studies of terpene synthases.  相似文献

哺乳动物肝细胞中cyp2e1基因所编码的蛋白CYP2E1在代谢异型有机物方面起着重要作用,转cyp2e1基因植物可以代谢多种小分子有机污染物;但cyp2e1基因在植物体内的表达调控和代谢机理尚不完全清楚。文中将含有cyp2e1基因的质粒pSLD50-6和对照gus基因的质粒pKH200转入根癌农杆菌GV3101,利用根癌农杆菌转基因技术将cyp2e1基因和对照gus基因成功转入烟草,分别获得了转cyp2e1和gus基因再生植株。选取PCR鉴定的再生植株进行荧光定量PCR(qRT-PCR)分析,结果表明:在转录水平上,转cyp2e1基因烟草中,乙醇处理后cyp2e1基因的表达明显下降,苯和甲苯处理后cyp2e1基因的表达量稍有下降;而丙酮、甲醛处理和缺氧条件下cyp2e1基因的表达有不同程度的升高。此外,苯处理后,转cyp2e1基因烟草中NADPH-P450氧化还原酶和细胞色素b5酶的基因活性显著提高,说明烟草中NADPH-P450氧化还原酶和细胞色素b5酶与CYP2E1酶的解毒过程有关,可能起到哺乳动物体内的NADPH-P450氧化还原酶和细胞色素b5的功能,参与CYP2E1酶催化过程的电子传递链。  相似文献

We have isolated a hyperrecombinogenic Nicotiana tabacum mutant. The mutation, Hyrec, is dominant and segregates in a Mendelian fashion. In the mutant, the level of mitotic recombination between homologous chromosomes is increased by more than three orders of magnitude. Recombination between extrachromosomal substrates is increased six- to ninefold, and intrachromosomal recombination is not affected. Hyrec plants were found to perform non-homologous end joining as efficiently as the wild type, ruling out the possibility that the increase in homologous recombination is due to a defect in end joining. In addition, Hyrec plants show significant resistance to gamma-irradiation, whereas UV resistance is not different from the wild type. This suggests that homologous recombination can be strongly up-regulated in plants. Moreover, Hyrec constitutes a novel type of mutation: no similar mutant was reported in plants and hyperrecombinogenic mutants from other organisms usually show sensitivity to DNA damaging agents. We discuss the insight that this mutant provides into understanding the mechanisms of recombination plus the potential application for gene targeting in plants.  相似文献

Age-related DNA changes in soybean ( Glycine max L. cv. Ransom) cotyledon nuclei were investigated by Feulgen cytophotometry and cloned DNA probes. The amount of nuclear DNA in 17-day-old senescing cotyledons was 23% lower than that of 5-day-old young cotyledons. In order to understand the nature of senescence-related DNA loss, fragments of repetitive DNA from young cotyledons were cloned into Escherichia coli HB101 cells by DNA recombination. The cloned DNA probed changes in specific repeated nucleotide sequences in senescing cotyledons. The colony hybridization between cloned DNA and [32P]-labeled total DNA from 5- and 17-day cotyledons indicated loss of specific repeated sequences. The selected sequences of repeated DNA further showed a loss in their copy numbers. The study suggested that some repetitive DNA sequences were degraded selectively in the genome of senescing cotyledons.  相似文献

The cytokinin complex in tobacco leaves of various maturities was characterized by radioimmunoassay and mass spectrometry. Zeatin was the major base, whereas zeatin riboside was identified as the main riboside. in leaves of all maturities studied. Relative to upper younger leaves, the basal yellow leaves had reduced levels of both cytokinin bases and ribosides. Exogenous applications of dihydrozeatin and zeatin to detached tobacco leaves in amounts sufficient to delay senescence, elevated cytokinin base and riboside levels 2–5 fold. Presenescent and senescent leaves of intact plants showed quantitatively similar changes in cytokinin content. which therefore appear to be of significance in control of senescence. When supplied exogenously, the principal cytokinin bases found to occur in tobacco leaves (zeatin and dihydrozeatin) were markedly more effective than auxins and gibberellic acid in retarding senescence. Localised application of cytokinins to leaf blades of detopped plants was much less effective than application to intact plants. The cytokinin induced senescence retardation in tobacco leaves was independent of effects on directed metabolite transport. Evidence that endogenous levels of active cytokinins in intact tobacco leaves are involved in control of sequential leaf senescence is discussed.  相似文献

The relationship between cellular growth and aluminium (Al) uptake was examined by applying brefeldin A, a vesicle transport inhibitor, to cultured tobacco (Nicotiana tabacum L. cv. BY-2) cells. Cultured cells almost completely lost the capacity for Al uptake when pre-incubated for 1–3 h in a minimal medium. Pre-incubation also diminished subsequent growth in a culture medium. However, competency for Al uptake (20 μm, pH 4.5) was sustained in a dose-dependent and reversible manner when cells were treated with brefeldin A (10 μm), an inhibitor of Golgi-mediated secretion, prior to and during the incubation in minimal medium.  Sperm cells released from in vivo-in vitro grown pollen tubes of tobacco are associated in pairs and initially enclosed by the plasma membrane of the pollen tube. When the sperm cells are placed together, using glass microinjector needles, in an enzymatic solution, up to half undergo cellular fusion with subsequent nuclear fusion. The frequency of sperm cell fusion decreases with time during the elongation of the pollen tube, suggesting that mechanisms inhibiting self-fusion of sperm cells may develop as the pollen tube elongates through the style toward the ovule. This tendency may play an important role in inhibiting fusion of the two sperm cells inside the calcium-rich synergid where the male germ unit dissociates and sperm cells are transported to their target cells - the egg and central cell. Sperm cells are released from pollen tubes of tobacco as linked cells, associated with the vegetative nucleus in an assemblage known as the male germ unit (MGU). Using light microscopy, the MGU assemblage appears to be ensheathed by cytoplasmic material of the pollen tube, which may stabilize their association. Following their release, the shape of the sperm cells and vegetative nucleus changes from an ellipsoidal to a more spheroidal morphology. When most of the cytoplasmic material is dispersed, a boundary remains around the two sperm cells. Using scanning electron microscopy, the cytoplasmic material surrounding the MGU appears filamentous, sometimes twisted and rope-like.
