The great majority of angiosperm species form a group in which either every cell in the root epidermis produces a root hair, or the cells that produce these hairs are randomly distributed. We describe, for the first time, pattern in the root epidermal cells of a species within this group. The seedling root of Echium plantagineum L. (Boraginaceae) has an epidermis in which almost every cell produces a root hair, but these are of two types, short hairs (up to 200 micro m) and long hairs (>200 micro m), which are in separate cell files, with the cells bearing long hairs usually separated by one or two files of cells bearing short hairs; the epidermal cells with the long root hairs are longer than the epidermal cells with the short root hairs. The long root hairs are initiated and develop earlier than the short root hairs. Transverse sections of the region of the root which contains only developing long root hairs show that the hair cells are located above anticlinal walls between underlying cortical cells. We regard the distribution of root epidermal cells in E. plantagineum as a sub-type of this group. We discuss the possible evolution, from this sub-type, of another group that is characterised by hair cells and non-hair cells occurring in separate files.  相似文献

Various concentrations of salt (NaCI) were shown to have an influence on the differentiation of tissues in the root and stem ofBacopa monniera (L) Wettst. Higher concentrations induced drastic changes in roots grown on salt-supplemented media; epidermal and cortical cells experienced changes in shape, size, and orientation and/or were got disintegrated. A low concentration of salt induced a profuse development of root hairs which gradually disappeared at higher concentrations. Air spaces in the stem cortex were enlarged and xylem cell walls in the vascular ring were thickened.  相似文献

Extracts fromshiitake (Lentinula edodes) mycelial culture broth, by an organic solvent ethyl acetate, inhibited the proliferation of cultured cells. At lower concentrations (1.25–15 μg/ml), this inhibition, measured by the MTT assay, was dose- and cell line-dependent. Inhibition of tumor cells, such as Caski, SiHa, HeLa, HP-1 and A375, byL. edodes-436 extracts was stronger than inhibition of normal cells (3T3). At 20 μg/ml, the extracts induced changes in cell shape, DNA-fragmentation and the activation of caspase-3. The extracts also inhibited the binding of E2F protein to its promoter. The results suggest that extracts ofL. edodes culture broth contain substances that have the ability to induce apoptosis in the cultured cells.  相似文献

Summary The sub-cellular localization of some nitrogen compounds within the leaf cavities ofAzolla filiculoides Lam. was obtained by means of electron spectroscopic imaging (ESI) and electron energy loss spectroscopy (EELS). The analyses were performed on ultrathin unstained sections of differentAzolla leaf cavities which contain epidermal hairs,Anabaena azollae Strasb. and bacteria. Net nitrogen distributions were visualized by image analysis, and nitrogen peaks were evidenced in spectra recorded in the same areas. Different distributions of nitrogen compounds were observed within the leaf cavities along the stem, in particular inside the epidermal hairs ofAzolla and the vegetative cells and heterocysts ofA. azollae.  相似文献

利用光学显微镜和扫描电子显微镜,观察了新疆委陵菜属5组不同花柱组10种4变种植物叶表皮的微形态特征,测量统计叶表皮毛的类型、表皮细胞的形状及大小、气孔器的分布及类型、气孔的形状、大小、密度及指数、气孔外拱盖形态及其纹饰等指标。结果显示:新疆委陵菜属10种4变种植物叶的下表皮均有气孔器的分布,形状为长椭圆形、椭圆形、宽椭圆形和近圆形;气孔器的类型多为无规则四细胞型、无规则型、围绕型和辐射型;表皮毛的类型为针状毛、带状柔毛和腺毛;表皮细胞的形状分为不规则形和多边形2种类型。研究表明,新疆委陵菜属植物表皮毛特征、叶片表皮细胞的形状、垂周壁式样、气孔器的形状类型、气孔密度指数及外围蜡质纹饰等存在差异,对属以下等级的划分有重要价值,可作为物种分类及鉴别的依据,同时也为本属一些分类群间的系统关系的探讨提供佐证。  相似文献

This report describes the early cytological events in the infection byRhizobium leguminosarum biovartrifolii of the root hairs ofTrifolium repens seedlings kept alive on agar medium in glass slide culture experiment. The infection threads bearing rhizobia were formed as soon as the epidermal cells began to emerge as root hairs. On the top of some of these infected emerging root hairs, there were smoky, cell-debris-like bodies, which appeared to be derived from the cell wall dug by rhizobia. Similar bodies were also observed in longer root hairs. None of the root hair cells along the length of the roots which contained infection threads were curled or distorted. A substantial number of pink-colored nodules were later formed on the roots with non-curled infected root hairs.  相似文献

Summary Microtubule (MT) arrays in stomatal complexes ofLolium have been studied using cryosectioning and immunofluorescence microscopy. This in situ analysis reveals that the arrangement of MTs in pairs of guard cells (GCs) or subsidiary cells (SCs) within a complex is very similar, indicating that MT deployment is closely coordinated during development. In premitotic guard mother cells (GMCs), MTs of the transverse interphase MT band (IMB) are reorganized into a longitudinal array via a transitory array in which the MTs appear to radiate from the cell edges towards the centre of the walls. Following the longitudinal division of GMCs, cortical MTs are reinstated in the GCs at the edge of the periclinal and ventral walls. The MTs become organized into arrays which radiate across the periclinal walls, initially from along the length of the ventral wall and later only from the pore site. As the GCs elongate, the organization of MTs and the patterns of wall expansion differ on the internal and external periclinal walls. A final reorientation of MTs from transverse to longitudinal is associated with the elongation and constriction of GCs to produce mature complexes. During cytokinesis in the subsidiary mother cells (SMCs), MTs appear around the reforming nucleus in the daughter epidermal cells but appear in the cortex of the SC once division is complete. In the root epidermis of Hydrocharis morsus-ranae L. distinctive trichoblasts, which later grow out as root hairs, are formed by the unequal division of protodermal (immature epidermal) cells. The trichoblast is the more proximal product of this division. Trichoblasts differ from adjacent epidermal cells in manner of growth, in size, amount of cytoplasm, degree of succinic dehydrogenase and cytochrome oxidase activity, and in the structure of their plastids. Plastids in the trichoblasts gradually become colorless and of less complex structure with increasing distance from the root tip, in contrast to those in adjacent epidermal cells. The trichoblasts do not divide, but they elongate to a considerable extent in the most distal 3000 μ of the root tip and less extensively in the next 3000 μ. By contrast the sister cells and their products divide, but the individual products do not become markedly longer than the mother cell until situated more than 3000 μ from the root tip, when they undergo extensive elongation. The trichoblasts are thus characterized by delayed maturation and inhibition of cytokinesis; it is suggested that delayed maturation is a necessary prerequisite for differentiation of root hairs in this and other species.  相似文献

Summary Anabaena azollae is associated with two types of multicellular epidermal trichomes inAzolla leaf cavities, the simple and branched hairs. The observation of transfer cell ultrastructure in some hair cells led to speculation that the cavity hairs might participate in metabolite exchange between the symbionts. The developmental ontogeny of cavity trichomes is described here, using transmission electron microscopy, with a goal of improving our understanding of possible functions of these structures in the symbiosis. The observations have established that all cells of simple and branched hairs develop the structural characteristics of transfer cells, but not simultaneously. Rather, there is an acropetal succession of transfer cell ultrastructure beginning in terminal cells, moving to body cells where present, and ending in stalk cells. The transfer cell stage is followed immediately by senescence in all hair cells. The timing of transfer cell differentiation, considered together with information from other studies, suggests that branched hairs may be involved in exchange of fixed nitrogen between the symbionts, while simple hairs may participate in exchange of fixed carbon fromAzolla toAnabaena. The root hairs of plants are tubular projections of root epidermal cells and are suitable for investigating the control of cellular morphogenesis. In wild-typeArabidopsis thaliana (L.) Heynh, growing root hairs were found to exhibit cellular expansion limited to the apical end of the cell, a polarized distribution of organelles in the cytoplasm, and vesicles of several types located near the growing tip. Therhd3 mutant produces short and wavy root hairs with an average volume less than one-third of the wild-type hairs, indicating abnormal cell expansion. The mutant hairs display a striking reduction in vacuole size and a corresponding increase in the relative proportion of cytoplasm throughout hair development. Bead-labeling experiments and ultrastructural analyses indicate that the wavy-hair phenotype of the mutant is caused by asymmetric tip growth, possibly due to abnormally distributed vesicles in cortical areas flanking the hair tips. It is suggested that a major effect of therhd3 mutation is to inhibit vacuole enlargement which normally accompanies root hair cell expansion.  相似文献

Because the epidermis ofV. faba L. leaves easily can be peeled into strips of one cell layer, we developed a simple method ofin situ hybridization using epidermal peels as a substitute for paraffin, resin and cryosections. Our method sufficiently detected the expression of broad bean aquaporin 1 in guard cells. RT-PCR revealed higher expression of aquaporins (AQPs) in guard cells compared to other leaf cell types; this indicates the importance of AQP for bulk water flow across guard cell membranes and, therefore, for stomatal movements.  相似文献

Summary In order to study developmental changes in microtubule organization attending the formation of a longitudinally oriented preprophase band, the guard mother cells ofAvena were examined using a new procedure for anti-tubulin immunocytochemistry on large epidermal segments. We found that the interphase band (IMB) of transverse cortical microtubules present in these cells following asymmetric division is replaced after subsidiary cell formation by mesh-like to radial microtubules that extend throughout the cytoplasm. Many of the Mts are also grouped in bundles. Gradually, this intermediate array is succeeded by longitudinal elements of the PPB. Thus, preprophase band formation is accompanied by a 90° shift in Mt orientation, with a radial arrangement serving as an intermediate stage. The micrographs are most consistent with the rearrangement of intact Mts, although changes in Mt assembly are possible as well. Summary Migration of bilayered epidermal cell sheets out of explants of tadpole tails (Xenopus laevis) were investigated with time-lapse cinemicrography using reflection-contrast optics. Cell-sheet formation begins beneath the explant in a region where it is closely attached to the coverslip. A single basal cell extends a lamellipodium through the outer (surface) epidermal layer and starts moving in a direction free of attached cells. This cell remains connected to the following basal cell, which the also extends a lamellipodium onto the glass. The cell sheet develops as increasingly more adjacent basal cells start to migrate. Surface cells do not actively locomote but they remain attached to the basal cells and to adjacent surface cells. Thus, they are transported as an intact cell layer, and consequently the in situ arrangement of the tadpole epidermis is largely preserved in the cell sheet, i.e., basal cells adhere to the substratum and are covered by outer cells (surface cells) which face the culture medium. Basal cells extend lamellae beneath the rear end of the preceding cell, which is slightly fifted off the substratum. The direction of locomotion is determined by the frontal cells. Cell-sheet enlargement and locomotion cease when all the epidermal cells facing the coverslip have left the explant, and the cell sheet and epidermis covering the explant form a continuous layer.  相似文献

在光学显微镜下,对安徽省贝母属(Fritillaria L.)8种1变种,其中包括安徽贝母(F. anhuiensis)和宁国贝母(F. ningguoensis)各两个居群的植物叶片进行了解剖结构观察,结果发现:叶肉细胞均含有淀粉粒,近上表皮的叶肉细胞多为方形或圆柱状,较大,排列较紧密,近下表皮的叶肉细胞形状不规则,多有分枝,细胞间隙大;叶均无表皮毛等附属物,除铜陵黄花贝母(F. monantha var. tonglingensis)的下表皮细胞类似平行四边形外,其它种叶表皮细胞表面观为长方形或长条形,细胞长轴与叶脉平行,垂周壁为平直、浅波状或深波状;气孔器为Allium型,仅分布于下表皮,椭圆形,长轴与叶脉平行,单个随机分布,部分种保卫细胞两极有“T”形加厚。叶表皮细胞的形状,垂周壁式样,气孔器的类型等特征较为稳定,可以为贝母属内种的划分及系统演化关系的探讨提供实验证据。  相似文献

The in vivo activation of T cells by a variety of antigens can be inhibited by the administration of anti-I-A antibodies (Ab) at the time of antigen priming. This inhibition can partially be explained by the temporary loss of Ia molecules from Ia-bearing antigen-presenting cells (APC) in the spleen. In this study, the effects of i.p. injected monoclonal Ab specific for I-A glycoproteins of different H-2 haplotypes on Ia antigen expression and APC function of spleen cells and epidermal Langerhans cells were compared. It was found that anti-I-A Ab quickly bound to both spleen cell and Langerhans cell Ia antigens. Although spleen cell Ia antigens were modulated and thus temporarily disappeared, Ia antigen expression by epidermal Langerhans cells was not modulated. In functional studies, the capacity of spleen cells and epidermal cells from anti-I-A Ab treated vs control animals to function as APC for antigen-specific, I-A- or I-E-restricted T cell clones was tested. A single injection of anti-I-A Ab completely abolished the APC function of spleen cells as shown in several inbred mouse strains, F1 animals, and with the use of several different Ab and T cell clones. In contrast, Langerhans cell-dependent APC function of epidermal cells remained completely unaltered. Even multiple injections of high doses of Ab never caused any inhibition of Langerhans cell function. Experiments with anti-I-Ak or anti-I-Ad Ab in an (H-2k X H-2d)F1 animal showed abrogation of APC function of spleen cells, but again not of Langerhans cells. Thus in vivo anti-I-A Ab administration appears to differentially affect Ia antigen expression and APC function from spleen and epidermis: Ia antigens are modulated from spleen cells but not from epidermis, and APC function disappears in the spleen but not in the epidermis. The abrogation of splenic but not of Langerhans cell APC function with anti-I-A Ab will facilitate the dissection of the relative contributions of Langerhans cells as compared with other APC in the generation of cutaneous immune responses.  相似文献

We report a novel feature of leaf variegation. As is often the case in tropical forest floor herbs, Schismatoglottis calyptrata leaves feature structural variegation. Examination of leaf anatomy in S. calyptrata revealed a novel feature of structural variegation, which was generated by variation in the spatial arrangement of the adaxial-most tip of the palisade cells. In fully green leaf parts, contact between the adaxial-most tip of the palisade cells and the cone-shaped base of the outer epidermis cells was tight, and palisade cells were arranged radially around each epidermal cell. In dull, grayish-green leaf parts, the contact was loose, and no particular spatial arrangement of palisade cells relative to epidermal cells was observed. This feature of structural variation could be disadvantageous for photosynthesis efficiency in view of the hypothesis that, for rainforest herbs, cone-shaped epidermal cells may function as lenses. However, the high frequency of leaf variegation of S. calyptrata in natural habits suggests that this structural variegation plays an unknown advantageous role.  相似文献

The diversity of non-glandular and glandular hairs of Cannabis sativa L. (marihuana) are described by scanning electron microscopy. The non-glandular hairs are of two major types, as distinguished by size differences and locations, and all of them are highly silicified. The presence of silica as well as cystoliths of calcium carbonate help in the identification of marihuana even in its ash residues. X-ray microanalyses of Cannabis hairs are compared with those of Humulus lupulus and Lantana camera, whose hairs have been considered to resemble those of marihuana. Glandular hairs are found to be of two major categories. One group consists of glands whose heads are generally made up of eight cells and the other group whose heads are generally made up of two cells but never more than four cells. All glands of both categories are stalked. Some glands of the first category are massively stalked and these are restricted solely to anthers and bracts of staminate and pistillate plants. The massive stalk is considered to be made up of epidermal and hypodermal cells that have grown in response to some stimulation during anthesis. Fine details of the shoot system of Cannabis, such as cuticular ridges on epidermal cells, warty protuberances on non-glandular hairs, and surface views of glands in developing stages are also reported. Glandular hairs on the bracts of Humulus lupulus resemble those of Cannabis.  相似文献
