Three distinct types of mitochondria as related to shape, position, and size in the femoral muscle of the cockroach, Leucophae maderae, are described. The elongated mitochondria are interposed between the myofibrils and are oriented parallel to the long axis of the fiber. Surface indentations on these mitochondria for surrounding structures indicate their permanent position. The oval mitochondria are situated under the sarcolemma. These organelles have a parallel orientation to the underlying muscle fiber but no alignment with respect to the “sarcomeric repeat” and thereby suggest that this type is mobile. The Y-shaped mitochondria are observed in the intermyofibrillar sarcoplasm. Their paired processes are on each side of the Z-disc and imply that the Y-shaped mitochondria are a sessile type. The cristae in all three categories of mitochondria display a tightly packed and complex arrangement.  相似文献

Summary The fibers of drum and trunk muscles of the Tigerfish, Therapon jarbua, differ greatly in diameter. The myofibrils of the trunk muscles are irregularly oriented, while those of the drum muscles are rolled into spiral or concentric bands. Both muscle types possess the sarcomere structure typical of cross-striated musculature. However, the myofibrils of the drum muscles differ greatly in sarcomere length and width from those in the trunk musculature. The trunk muscles contain few mitochondria, whereas in the drum muscles mitochondria are abundant. The sarcoplasmic reticulum (SR) of the drum muscles takes the form of elongated tubes in both the A and the I region; that of the trunk musculature consists of small vesicles. Of the two muscle types, the drum muscle contains more SR. With respect to the form of the T system, the trunk musculature is of the Z type and the drum muscles of the A-I type. The drum muscle displays a considerably greater number of motor endplates; these lack typical junctional folds and have mitochondria with very few cristae. No fat could be demonstrated in either the drum or the trunk muscles. However, the concentration of glycogen is higher in the drum muscle than in the musculature of the trunk.This work was accomplished with support from the Deutsche Forschungsgemeinschaft and is gratefully dedicated to Prof. R. Cellular functions involve the temporal and spatial interplay of ions, metabolites, macromolecules, and organelles. To define the mechanisms responsible for completing cellular functions, we used methods that can yield both temporal and spatial information on multiple physiological parameters and chemical components in the same cell. We demonstrated that the combined use of selected fluorescent probes, fluorescence microscopy, and imaging methods can yield information on at least five separate cellular parameters and components in the same living cell. Furthermore, the temporal and spatial dynamics of each of the parameters and/or components can be correlated with one or more of the others. Five parameters were investigated by spectrally isolating defined regions of the ultraviolet, visible, and near-infrared spectrum based on five distinct fluorescent probes. The parameters included nuclei (Hoechst 33342), mitochondria (diIC1-[5] ), endosomes (lissamine rhodamine B-dextran), actin (fluorescein), and the cell volume Cy7-dextran). Nonmotile, confluent Swiss 3T3 cells did not show any detectable polarity of cell shape, or distribution of nuclei, endosomes, or mitochondria. These cells also organized a large percentage of the actin into stress fibers. In contrast, cells migrating into an in vitro wound exhibited at least two stages of reorganization of organelles and cytoplasm. During the first 3 h after wounding, the cells along the edge of the wound assumed a polarized shape, carried the nuclei in the rear of the cells, excluded endosomes and mitochondria from the lamellipodia, and lost most of the highly organized stress fibers. The cell showed a dramatic change between 3 and 7 h after producing the wound. The cells became highly elongated and motile; both the endosomes and the mitochondria penetrated into the lamellipodia, while the nuclei remained in the rear and the actin remained in less organized structures. Defining the temporal and spatial dynamics and interplay of ions, contractile proteins, lipids, regulatory proteins, metabolites, and organelles should lead to an understanding of the molecular basis of cell migration, as well as other cellular functions.  相似文献

1. An improved type of ground glass homogenizer for soft tissues has been described which brings about a high degree of cell disruption and liberation of nuclei without causing appreciable damage to mitochondria. The gentleness and effectiveness of the new homogenizer in respect to isolation of mitochondria have been ascertained by comparing the ATP-ase activities of mitochondria isolated in 0.25 M sucrose solution without pH adjustment using a previous type of homogenizer with those of mitochondria isolated under the same conditions with the aid of the new homogenizer. In these experiments sucrose of 0.25 molarity without pH adjustment has been used in order to maintain the mitochondria in a rather sensitive state so as to make slightly deleterious effects of homogenization readily apparent. 2. A new method is described for the isolation of morphologically intact mitochondria and cell nuclei from the same homogenate. In this procedure the pH of the homogenate in 0.44 M sucrose is maintained at 6.0-6.2 with citric acid during the homogenization. An alternative method employing 0.44 M sucrose plus 0.005 M CaCl(2) is given for the isolation of nuclei from tumor cells. However, the latter method does not produce unaltered mitochondria. 3. The alpha-ketoglutarate, malate, succinate, and hexanoate oxidases of the "intact" mitochondria isolated in 0.44 M sucrose adjusted to pH 6.0-6.2 with very dilute citric acid as described in this paper have been investigated, and it has been shown that the mitochondria compare favorably to those isolated in 0.25 M sucrose by a previously described method. 4. Mitochondria have been found to contain an enzyme which causes nuclei to lose their ability to form gels in dilute alkali. This enzyme is released from the mitochondria when the latter are disrupted. 5. Some properties of nuclei isolated by the new method have been briefly discussed.  相似文献

Eukaryotic cells have highly organized, interconnected intracellular compartments. The nuclear surface and cytoplasmic cytoskeletal filaments represent compartments involved in such an association. Intermediate filaments are the major cytoskeletal elements in this association. Desmin is a muscle-specific structural protein and one of the earliest known muscle-specific genes to be expressed during cardiac and skeletal muscle development. Desmin filaments have been shown to be associated with the nuclear surface in the myogenic cell line C2C12. Previous studies have revealed that mice lacking desmin develop imperfect muscle, exhibiting the loss of nuclear shape and positioning. In the present work, we have analyzed the association between desmin filaments and the outer nuclear surface in nuclei isolated from pectoral skeletal muscle of chick embryos and in primary chick myogenic cell cultures by using immunofluorescence microscopy, negative staining, immunogold, and transmission electron microscopy. We show that desmin filaments remain firmly attached to the outer nuclear surface after the isolation of nuclei. Furthermore, positive localization of desmin persists after gentle washing of the nuclei with high ionic strength solutions. These data suggest that desmin intermediate filaments are stably and firmly connected to the outer nuclear surface in skeletal muscles cells in vivo and in vitro.  相似文献

The conidiophore vesicle is composed of a peripheral regionwhich contains many nuclei and mitochondria and a central regionwhich is densely packed with glycogen granules but containsvery few organelles. The phialides, which arise as sphericalprotuberances from the surface of the vesicle, are bounded bya much thinner wall than that of the vesicle. Nuclei migrateinto the phialides at a comparatively early stage in their development.Septa are present at the proximal ends of mature phialides andalthough pores are present in these septa it is suggested thatthey are too small to permit the ingress of such organellesas nuclei and mitochondria from the vesicle. The developmentof adjacent phialides is well synchronized but the developmentof the vesicle as a whole is not precisely co-ordinated. Eachmature phialide contains a single nucleus, as do the conidiosporeswhich they produce. The first conidiospore produced by a phialidearises as a spherical protrusion from the tapered, thickenedapex of the phialide. The conidiospores are delimited from thephialide by the formation of a cross wall in a manner reminiscentof the process of septation in hyphae. When first formed theconidiospores have a cylindrical shape and they only assumetheir characteristic spherical shape at a later stage in development.  相似文献

We report the preparation and investigation of hierarchically organized host-guest structures, presenting successive ordering from the molecular up to macroscopic scale, thus supporting the relationship between the molecular arrangements and the macroscopic properties. Size, shape and surface composition of the host which is zeolite L play a decisive role. Its base and coat have distinctively different chemical properties. The guests, organic dye molecules or complexes, are well oriented inside the channels and can be organized into distinctive patterns. Zeolite L crystals containing oriented fluorophores in their parallel nanochannels possess remarkable fluorescent properties and they can be arranged in nearly any desired manner by means of self-organization methods. This makes them ideal host-guest structures for the exploitation of energy transfer and energy funneling processes. Size, shape and surface composition of the objects but also the properties of the surface on which they should be organized play a decisive role. We present a simple model of an artificial antenna based on supramolecular organization of dyes in nanochannels of the host, and we explain why zeolite L can be considered as an ideal host for this purpose. The preparation of different dye-zeolite L materials is described, and F?rster energy transfer experiments carried out with them. Further, increasing supramolecular organization is discussed: the first unidirectional antenna system on a macroscopic level, organization of crystals and communication of the crystals interior with the environment. Additionally, we explain spectroscopy on monolayers of dye-zeolite L micro-crystals. The materials are shown to be new building blocks for optical, electro-optical and sensing devices.  相似文献

The tubular fibers of the claw-closer muscle of the scorpion have a central core containing nuclei and mitochondria. The myofibrils have the shape of thin lamellae (1 µ) extending radially from the core to the surface membrane (20 µ). The thick myofilaments are organized in a hexagonal array with orbits of 10–13 thin myofilaments. The ratio of thick-to-thin filaments is 1:5. Transverse tubular system (TS) openings are located between lamellated myofibrils. In each sarcomere two TS's are found, one on each side of the H band. The TS is composed of a transverse tubule and tubular pockets (TP). The TP's form diadic contact with the terminal cisternae of the sarcoplasmic reticulum. The TS can be traced from the cell membrane down to the cell core. The surface area of the TS was calculated to be six times that of the outer surface membrane.  相似文献

The ultrastructure of locust muscles with different function is examined: the pleuroaxillary flight steering muscle is compared with a typical flight (power muscle) and a typical leg muscle, in particular with respect to sarcomere length, tracheation, mitochondria, and sarcoplasmatic reticulum. The pleuroaxillary muscle exhibits some features characteristic of flight muscles but most of the ultrastructure resembles that of leg muscles. This is in agreement with the innervation of this muscle by an octopaminergic neuron, which also innervates leg muscles but no other flight muscles. It also supports the hypothesis that octopaminergic neurons are important metabolic regulators and that the above muscle types exhibit important differences in energy metabolism.  相似文献

Large quantities of intact generative cells and their protoplasts were isolated from pollen protoplasts of four liliaceous plants, and their structural features were investigated. The generative cells, liberated from the vegetative cell cytoplasm of the pollen protoplasts, were initially spindle-shaped with two long, oppositely oriented extensions, and were surrounded by two cell membranes, one on each side of a wall of uniform thickness. The generative nuclei, stained with 4′,6-diamidino-2-phenylindole (DAPI), showed ellipsoidal and highly condensed chromatin, whereas the generative cell cytoplasm, whose quantity was widely different from species to species, showed no fluorescence, suggesting the absence of plastid and mitochondria! DNA, although many mitochondria were present. The isolated generative cells, which were spindle-shaped at first, became spherical in shape in vitro. Immunocytochemistry and transmission electron microscopy revealed that this change was associated with the depolymerization of an axial array of microtubules present in generative cells in situ. These results are discussed in relation to the function of the generative cell within the bicellular pollen of angiosperms.  相似文献

Striated visceral muscle cells are scattered singly or in small groups at the base of the intestinal cells of the mid-gut of Drosophila melanogaster larvae. Fibers less than 1 μ in diameter, designated as small, contain a single myofibril, few, if any, dyads and few mitochondria. Fibers of somewhat larger diameter contain dyads and more mitochondria. Both types of fiber have a perforate Z band which appears as discontinuous bodies in longitudinal sections and as a perforate sheet of dense rims and clear perforations in transverse sections. The Z rims contain filaments, 30 to 50 Å, oriented in the transverse direction. The number and arrangement of myofilaments and the ultrastructure of the Z band are consistent with the function of these muscles.  相似文献

The Euterpe edulis embryo consists of a prominent single cotyledon, a very short radicle-hypocotyl axis and an epicotyl. The epicotyl is obliquely angled with respect to the cotyledon; consequently it corresponds to one of the two categories recognized for palm seeds by DeMason (1988 ). Parenchyma, protoderm and procambium can be distinguished on the basis of position and shape of their cells, which are highly vacuolated with one central vacuole and the cytoplasm restricted to a thin parietal layer. Initial cells from both apical meristems are also vacuolated but they have small vacuoles distributed around the nuclei. Silica occurs in cell walls of some protodermal cells. Raphides, silica bodies and tannins all occur occasionally in vacuoles, especially in the basal cotyledon region. Most embryo cells lack storage reserves and exhibit an active state, with numerous mitochondria, RER cisternae and Golgi apparatus, indicating a strategy of continuous development without the interposition, at maturity, of a dry state. The endosperm consists of living cells with very large nuclei and thickened cell walls. Similar to the endosperm of other studied palm species, their cells exhibit a quiescent appearance with lipid, protein, minerals (in the cytoplasm) and mannans (in the cell walls) as the insoluble storage reserves.  © 2004 The Linnean Society of London, Botanical Journal of the Linnean Society , 2004, 145 , 445–453.  相似文献

It is shown that locust adult endocuticle consists of a daily alternation of two types of chitin-protein architecture: (i) non-lamellate day layers with microfibrils oriented in a preferred direction, traversed by pore canals whose shape resembles an untwisted ribbon, (ii) lamellate night layers with helicoidally oriented microfibrils traversed by pore canals shaped like regularly twisted ribbons. Uncoupling the circadian clock which normally controls the timing of these two types leads to growth of cuticles which are organized like one or the other throughout. We can thus experimentally change the architecture of the microfibrils which in turn changes the pore canals.  相似文献

Summary Ubiquitin was localized by immunofluorescence microscopy during post-mating histolysis of fibrillar flight muscle in female fire ants, Solenopsis spp. Normal muscles, as well as histolysing muscles from artificially inseminated and haemolymph-injected females contained ubiquitin in association with nuclei, Z-lines, myofilaments and mitochondria. However, the density of the ubiquitin immunoreaction was markedly increased in the nuclei, Z-lines and mitochondria of degenerating tissues 6, 12 and 24 h post treatment. At these times the heaviest immunoreactivity for ubiquitin was seen in association with the nuclei, Z-lines and mitochondria. Immuno-controls, incubated in the absence of the primary antibody, showed no similar immunostaining. When insemination was preceded by the injection of actinomycin D, muscle degradation was significantly depressed after a 24-h period. Also, ubiquitin immunofluorescence was markedly reduced in tissues pre-treated with actinomycin D. These observations suggest that insemination increases the ubiquitination of specific myofibrillar proteins destined for degradation.  相似文献

To follow the intracellular distribution of calcium in the breast muscles of developing chickens, Ca45 was injected into the albumen of predeveloped eggs. Since the embryos were grown in a radioactive medium, a complete exchange of the isotope for its non-radioactive counterpart in muscles was accomplished. Subcellular particulates of the muscle cells were separated by the method of differential centrifugation. Analysis of the separated fractions showed that in the muscles of the 13-day embryo, when the nuclear-myofibrillar ratio is high, 65 per cent of the muscle calcium is in the nuclei. With the increased synthesis of myofibrils, the nuclear-myofibrillar ratio decreases with a concomitant fall in radioactivity. Thus, calcium was not associated with the developing myofibrils. At the time of hatching, when myofibrils perform physiological work, the highest level of calcium is in the mitochondria. This suggests that the mitochondria play a key role in the physiological activities of calcium in the cell. The microsomal fraction reaches a maximal level of calcium when the adult composition of muscle is attained. Results of investigations on dystrophic muscles show changes in the calcium distribution of the fractions as early as the 3rd week of embryonic development, which are interpreted to indicate an alteration in the protein metabolism of the cell, or an early destruction of muscle tissue. Further, alterations in the calcium content of fractions which seem to regulate the movements of this ion in the cell are discussed. A new technique for homogenizing tissues from embryos of different ages is presented.  相似文献

The retinula of the compound eye of the worker honey-bee has been examined with the electron microscope. The rhabdom lies on the ommatidial axis; it is usually cylindrical in shape, about 3 to 4 µ in diameter, and lacks an axial cavity. Cross-sections show it to be four parted, although it is formed from eight retinular cells (Figs. 2, 3). Each quadrant of the rhabdom consists of a closely packed parallel array of tubules with long axes perpendicular to the axis of the rhabdom. The tubules in adjacent quadrants of the rhabdom are mutually perpendicular. At the distal end of the ommatidium these tubules are seen to be microvilli of the retinular cells. Immediately surrounding the rhabdom, the cytoplasm of the retinular cells contains a membranous endoplasmic reticulum which is oriented approximately radially with respect to the axis of the ommatidium. Farther away from the rhabdom the cytoplasm contains numerous mitochondria.  相似文献

The actin-like protein with a molecular weight of 42 kDa was obtained from the preparation of freshly isolated mitochondria of the rat liver using the method of immobilized DNAse affinity chromatography. The inhibitory ability of the isolated protein with respect to pancreatic DNAse I was the same as that of muscular actin. The native structure of the mitochondria protein is confirmed by the data of spectral analysis and its ability to globular-fibrillar transformation with an increased ionic strength of the solution. The polymerization ability as well as a stimulating effect of the actin-like protein of mitochondria on the ATPase activity of myosin is much less pronounced as compared to actin of skeletal muscles.  相似文献

Various aspects of the ultrastructure of the dividing nuclei in the large radiosensitive amoeba Pelomyxa illinoisensis are demonstrated. Evidence of nuclear envelope breakdown is presented, and membrane fragments are traced throughout metaphase to envelope reconstruction in anaphase and telophase. Annuli in the nuclear envelope and its fragments are shown throughout mitosis. During metaphase and anaphase some 15 to 20 mitochondria are aligned at each end of the spindle, and are called polar mitochondria. The radioresistant amoebae Pelomyxa carolinensis and Amoeba proteus do not have polar mitochondria, and Pelomyxa illinoisensis is unique in this regard. The shape of the P. illinoisensis interphase nucleoli differs from that in the two radioresistant species, and certain aspects of nucleolar dissolution in the prophase vary. Helical coils in the interphase nucleoplasm are similar to those in the radioresistant amoebae. A "blister" phase in the flatly shaped telophase nuclei of P. illinoisensis is described which is interpreted to be the result of a rapid nuclear expansion leading to the formation of the normal spherical interphase nuclei.  相似文献
