Besides mosquitoes, ticks are regarded as the primary source of vector-borne infectious diseases. Indeed, a wide variety of severe infectious human diseases, including those involving viruses, are transmitted by ticks in many parts of the world. To date, there are no published reports on the use of next-generation sequencing for studying viral diversity in ticks or discovering new viruses in these arthropods from China. Here, Ion-torrent sequencing was used to investigate the presence of viruses in three Rhipicephalus spp. tick pools (NY-11, NY-13, and MM-13) collected from the Menglian district of Yunnan, China. The sequencing run resulted in 3,641,088, 3,106,733, and 3,871,851 reads in each tick pool after trimming. Reads and assembled contiguous sequences (contigs) were subject to basic local alignment search tool analysis against the GenBank database. Large numbers of reads and contigs related to known viral sequences corresponding to a broad range of viral families were identified. Some of the sequences originated from viruses that have not been described previously in ticks. Our findings will facilitate better understanding of the tick virome, and add to our current knowledge of disease-causing viruses in ticks living under natural conditions.  相似文献

Recently, 3-year-old seedlings of Cedrus deodara in Nanjing, China developed root rot, crown rot, chlorosis and defoliation, often resulting in death. Two species of Phytophthora were isolated. One was identified as A2 mating type of P. cinnamomi Rands while the other failed to produce sex organs in single or dual cultures and was tentatively assigned to P. drechsleh Tucker. Their pathogenicity was confirmed by re-inoculation and re-isolation.  相似文献

云南会泽铅锌矿渣中节杆菌的生理特征和系统发育   总被引:1,自引:0,他引:1  
对分离自铅锌矿渣中一种具有奶油色不扩散菌落形态的优势细菌进行了系统发育和部分生理特征研究。在可培养的细菌类型中,该种菌落的出现频率在50%左右。利用60个菌株16SrRNA基因约500bp的序列和其相关种构建的系统树表明,这些菌落属于节杆菌属内多个系统发育地位有差异的类群。各类群代表菌株BS11、BS20和AS19约1440bp左右的16SrRNA基因序列分析的结果表明,BS11菌株同Arthrobacter histidinolovorans和Arthrobacter nicotinovorans关系密切,BS20菌株同Arthrobacter chlorophenolicus关系密切,而AS19同Arthrobacter aurescens和Arthrobacter ilicis关系密切。在考察的39个碳源中,3个代表菌株都能利用其中的15个,不能利用其中的12个,而对另外12个的利用存在差异。此外,它们对Zn2+、Pb2+、Cd2+、Cu2+和Co2+等5种重金属的抗性水平都较高,但对链霉素、氨苄青霉素、卡那霉素和利福平4种抗生素较敏感。它们是潜在的新物种。  相似文献

Many Phytophthora species are pathogens on fruit trees and may cause destructive diseases. In the current study, we examined six Phytophthora isolates recovered from rivers in Bulgaria, representatives of the following three species: Phytophthora chlamydospora, Ppseudocryptogea and Psyringae. Morphological traits, cardinal temperatures and growth rates of the isolates were described. We found considerable variation in the size of sporangia and significant difference in the mycelial growth rates of the two P. pseudocryptogea isolates, along with multiple polymorphic sites in the ITS region of one of them. In the cases of the other two Phytophthora species, no such differences were found between the isolates. Both Pchlamydospora isolates had a lower optimum growth temperature compared with the reported in the literature for this species. In pathogenicity tests against leaves and fruits of apple, pear, cherry, apricot and plum, the isolates proved to be capable of causing infections with varying severity. Pchlamydospora showed to be the most aggressive towards the leaves, while Ppseudocryptogea isolates induced the highest percentage of decay on the fruits of all tested tree species, which may suggest partial organ or tissue specificity. The demonstrated infection capacity of the described isolates points out the investigated Phytophthora species as a potential threat for the orchards in Bulgaria, if favourable conditions are available.  相似文献

Caragana species are woody legumes widely distributed in the arid regions of China. These plants form root nodules but their nodule bacteria have not been clearly classified. A total of 112 symbiotic bacterial isolates were obtained from four Caragana species grown in Liaoning Province and were characterized with ARDRA, sodium dodecyl sulfate-polyacrylamide gel electrophoresis of proteins, BOX-PCR and sequencing of the 16S rRNA gene. Most of them were classified as Mesorhizobium belonging to 11 putative species. Three isolates were identified as Rhizobium etli and Burkholderia spp. This study offers new information about the Caragana-rhizobia association and resource for selection of inoculants used in sustainable agriculture and for further studies on the Caragana rhizobia.  相似文献

The occurrence of an epidemic outbreak of a powdery mildew disease on mulberry in Yunnan province, China, is reported. Its symptoms are characteristic for powdery mildews and visible as white pathches covering the abaxial surfaces of leaves leading to chlorosis and necrosis. The pathogen is morphologically barely distinguishable from Phyllactinia moricola. However, it exhibits several new morphological characteristics which 2–3 conidia could be formed in short chains at the apex of the conidiophores and the conidia could produce two germ tubes in any position. Phylogenetic analyses of ITS sequences show that the pathogen has a close genetic relationship with P. moricola and Ph. broussonetiae‐kaempferi, two species on hosts belonging to family Moraceae. However, the ITS differences between Japanese sequences and the Chinese sequence derived from mulberry are greater than expected for a single species and suggest a cryptic species in China, but the present data are not sufficient for a final conclusion. Therefore, the Morus powdery mildew in Yunnan can currently only be classified as Phyllactinia sp. Morphological features, including conidial germination pattern of this powdery mildew are described in detail, and the local climatic conditions of the disease are analysed, which will provide the base for finding an effective method, including bio‐control, to control the disease under local conditions.  相似文献

The genus Phytophthora belongs to the oomycetes and is composed of plant pathogens. Currently, there are no strategies to mutate specific genes for members of this genus. Whole genome sequences are available or being prepared for Phytophthora sojae, P. ramorum, P. infestans, and P. capsici and the development of molecular biological techniques for functional genomics is encouraged. This article describes the adaptation of the reverse-genetic strategy of targeting induced local lesions in genomes (TILLING) to isolate gene-specific mutants in Phytophthora spp. A genomic library of 2,400 ethylnitrosourea (ENU) mutants of P. sojae was created and screened for induced point mutations in the genes encoding a necrosisinducing protein (PsojNIP) and a Phytophthora-specific phospholipase D (PsPXTM-PLD). Mutations were detected in single individuals and included silent, missense, and nonsense changes. Homozygous mutant isolates carrying a potentially deleterious missense mutation in PsojNIP and a premature stop codon in PsPXTM-PLD were identified. No phenotypic effect has yet been found for the homozygous mutant of PsojNIP. For those of PsPXTM-PLD, a reduction in growth rate and an appressed mycelial growth was observed. This demonstrates the feasibility of target-selected gene disruption for Phytophthora spp. and adds an important tool for functional genomic investigation.  相似文献

Potential vectors of Leishmania braziliensis Vianna were assessed at four study sites in the mountainous Valle del Cauca, western Colombia, from March to June 1989. In an active focus of transmission at 1450 m altitude, a coffee plantation at Versalles, there were high densities of antropophilic phlebotomines: Lutzomyia columbiana (Ritorcelli & Van Ty) and Lu.townsendi (Ortiz), both in the verrucarum species group, and of Lu.pia (Fairchild & Hertig). At a comparable altitude in a forest reserve at Yotoco where leishmaniasis is unknown, Lu.pia was the prevalent species and Lu.townsendi was absent. In two localities at 1150 m altitude, there were plentiful Lu.lichyi (Floch & Abonnenc) plus both species in the verrucarum group, but Lu.pia was absent. One of these localities, a coffee plantation at Villa Hermosa where a leishmaniasis outbreak occurred in 1986, was compared with a leishmaniasis-free, partly wooded nature reserve at Mateguadua. No natural infections of Leishmania were found in a total of 1896 wild-caught female phlebotomines belonging to at least seven species. It remains unclear why Leishmaniasis transmission is associated with coffee plantations in this part of Colombia. Laboratory-bred Lu.lichyi females were invariably autogenous, and blood-seeking females of this species were always parous. Parity rates in wild-caught females of other species were 55% Lu.pia, 24% Lu.columbiana and 14% Lu.townsendi. Female Lutzomyia infected artificially with Le.braziliensis promastigotes developed peripylarian infections. Higher proportions of Lu.townsendi (96%) and Lu.columbiana (78%) became infected but these species developed lower rates of stomodaeal infections (P less than 0.1) than Lu.lichyi (37%) or Lu.pia (44%).(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A total of 18 paralogs of xyloglucan-specific endoglucanases (EGLs) from the glycosyl hydrolase family 12 were identified and characterized in Phytophthora sojae and Phytophthora ramorum. These genes encode predicted extracellular enzymes, with sizes ranging from 189 to 435 amino acid residues, that would be capable of hydrolyzing the xyloglucan component of the host cell wall. In two cases, four and six functional copies of these genes were found in tight succession within a region of 5 and 18 kb, respectively. The overall gene copy number and relative organization appeared well conserved between P. sojae and P. ramorum, with apparent synteny in this region of their respective genomes. Phylogenetic analyses of Phytophthora endoglucanases of family 12 and other known members of EGL 12, revealed a close relatedness with a fairly conserved gene sub-family containing, among others, sequences from the fungi Emericella desertorum and Aspergillus aculeatus. This is the first report of family 12 EGLs present in plant pathogenic eukaryotes.  相似文献

Results from analyses of beta-glucosidase (EC 3.2.1.21) and phosphatase (EC 3.1.3.1;EC 3.1.3.2) activities indicated that presence of a Trichoderma isolate reduced development of Phytophthora cinnamomi. It was also observed that P. cinnamomi was more competitive in coinoculated cultures than in cultures where Trichoderma was added on day 3. Analysis of trehalase (EC 3.2.1.28) activity indicated that Trichoderma either utilized portions of the P. cinnamomi mycelium as substrate or the action of P. cinnamomi released additional nutrients not normally available to Trichoderma. 【背景】辣椒疫霉是一种毁灭性的土传病害,当前主要使用化学合成杀菌剂防治,但容易导致环境污染和食品安全等问题。【目的】筛选可拮抗辣椒疫霉的候选菌株,探究分离菌株拮抗辣椒疫霉的生理生化作用机制。【方法】综合应用形态学、核糖体RNA (rRNA)基因非转录区ITS序列相似性方法鉴定分离菌株,通过对峙实验筛选抑菌效果较高的拮抗菌株,基于比色法测定分离菌株发酵液粗提物对辣椒疫霉菌丝脂质过氧化、纤维素酶、β-葡萄糖苷酶(β-GC)和多聚半乳糖醛酸酶(PG)活性的影响。【结果】从腐木和土壤样品中分离得到11株木霉,分属于绿色木霉(Trichodermavirens)、哈茨木霉(Trichoderma harzianum)、钩状木霉(Trichoderma hamatum)和棘孢木霉(Trichoderma asperellum) 4个种。11株木霉对辣椒疫霉均有一定的抑制作用,抑制率达到90%以上的菌株包括:绿色木霉Tv-1(92.68%)、Tv-2 (95.12%),哈茨木霉Thz-2 (92.68%),钩状木霉Tha-1 (90.24%)。以4株高效木霉的发酵液粗提物处理辣椒疫霉菌丝5 d后,因脂质过氧化产生的丙二醛含量显著增加,分别达到1.20、1.48、2.69和3.16 nmol/g,显著高于对照处理的0.77 nmol/g;与对照组相比,β-GC、PG酶活性显著下降,分别降低了12.28%-64.91%、7.2%-15.5%;同时纤维素酶活性呈上升趋势,最显著组为2.647 U/mL,相对于对照组增加了0.831U/mL。【结论】分离得到4株明显抑制辣椒疫霉菌生长的高效木霉菌,主要通过破坏细胞壁结构、降低致病因子酶活力和增强脂质过氧化等方式起拮抗作用,可为辣椒疫病的生物防治提供理论依据和技术支持。  相似文献

Samples of tomato, lettuce and cucumber submitted for diagnosis to the Plant Protection Centre at the Norwegian Crop Research Institute and samples of soil, water and cucumber collected from greenhouses employing hydroponic cultures were examined for the occurrence of Pythium spp. and Phytophthora spp. Two species of Phytophthora and 16 species of Pythium were identified. Phytophthora cryptogea was found on tomato and lettuce. Phytophthora nicotianae was found on tomato fruit. Phytophthora was not found on cucumbers. Pythium irregulare and Pythium group F were the two most commonly found Pythium species in hydroponically cultivated cucumbers. A pathogenicity test with 56 isolates was performed on cucumber seedlings. The most aggressive species were Pythium aphanidermatum, P. irregulare, Pythium paroecandrum and Pythium ultimum.  相似文献

我国间座壳属真菌种类繁多,但对其分类研究滞后。通过对云南省间座壳属标本采集,基于形态学和核糖体内部转录间隔区、β微管蛋白基因、翻译延伸因子基因、钙调蛋白基因和组蛋白基因的DNA序列数据比较进行物种鉴定,描述了分别采集自八角黄皮和可可患病叶片上的间座壳属2个新种——八角黄皮间座壳和可可间座壳,并讨论了其与间座壳属物种的形态和系统发育关系。  相似文献

Two new species, Elatostema planinerve W. T. Wang & Y. G. Wei sp. nov. and E. longicuspe W. T. Wang & Y. G. Wei sp. nov. are described and illustrated from the Guizhou province, China. The former resembles E. bracteosum but differs in leaves, pistillate bracts and pistillate bracteoles; the latter is similar to E. tenuicornutum, but differs in stem surface, leaf shape, venation, cystoliths, staminate inflorescence bracts, bracteolate and staminate flowers.  相似文献

Lagerstroemia menglaensis C. H. Gu, M. C. Ji & D. D. Ma, a new species of Lythraceae from Xishuangbanna, Yunnan, China is described and illustrated. The morphological characteristics of the new species and two morphologically similar species (L. guilinensis and L. venusta) are compared, and a key to distinguish between them is provided.  相似文献
