Chromosome numbers were determined for 63 Anthurium species. Thirty-eight of these were newly determined. Generally the present work confirmed existing chromosome counts when these were available for comparison. The most common somatic chromosome number found was 30, but counts ranged from 2n = 20 to 90. In a few instances conflicting counts were obtained. B chromosomes were found frequently in Sect. Cardiolonchium and varied in number from one to three. Four polyploid series were evident from all available counts: 20-40, 24-30-48-84, 28-56 and 30-60-90-ca 124. Most species were part of the polyploid complex based on 30. Although species were not observed with n = 5, 6 or 7, movement among the basic numbers was considered to have occurred at this level. The relationship among these basic numbers and n = 15 (x2) is still obscure.  相似文献

Cytological features are presented for the first time for 12 species and one natural hybrid from 16 Brazilian populations of the genus Xyris , including members of two sections ( Xyris and Nematopus ). The results are compared with a review of all available chromosome numbers previously reported for the genus. All analysed species show semi-reticulate interphase nuclei and prophase to prometaphase chromosomes with heterogeneous condensing behaviour. Chromosome numbers varied between 2 n = 26 and 2 n ≈ 108. In Brazil especially, the karyological differentiation of Xyris seems to be generally characterized by dysploid and polyploid increase of the chromosome number, possibly after interspecific hybridization. Infections of two heterologous insect cell lines derived from Malacosoma disstria (Md108) and Choristoneura fumiferana (Cf70) by the Lambdina fiscellaria lugubrosa nucleopolyhedrovirus (LafiNPV-W) were characterized. Cytopathic effects characteristic of LafiNPV-W infection, including rounding of cells, nuclear hypertrophy, and occlusion body (OB) production, were observed in both cell lines. Budded virus titers were slightly higher in Md108 cells than Cf70 cells (5.8?×?107 versus 3.1?×?107 TCID50 units mL?1). Viral replication kinetics and cytopathic effects induced by LafiNPV-W infection were very similar in both cell lines. Actin rearrangements and redistribution of heterochromatin and euchromatin were observed within 24 h post-inoculation (hpi), and large quantities of nucleocapsids and virions were observed by electron microscopy at 48 hpi in both cell lines. Cf70 cultures produced OBs with numerous embedded virions, while OBs in Md108 cultures contained few virions or were empty with nucleocapsids packed in the nucleoplasm between OBs. In bioassays against second instar L. fiscellaria lugubrosa, OBs derived from LafiNPV-W-infected Md108 cells induced significantly lower levels of mortality than OBs derived from LafiNPV-W-infected Cf70 cells or from infected L. fiscellaria fiscellaria larvae.  相似文献

Chromosome numbers were determined for 86 Anthurium species. Fifty-one of these were newly determined with counts ranging from 2n = 24 to 66 and 30 being the most common. All known Anthurium chromosome numbers were summarized, and 43 taxonomic changes were made in the previous reports to reflect current taxonomy. In terms of somatic chromosome numbers, the numbers form four polyploid series of 20–40–60, 24–30–48–84, 28–56 and 30–60–90–ca. 124. Paleoaneuploidy, polyploidy and B-chromosomes are basic features of the genus, but subsequent recent aneuploidy is not. The exact nature of chromosome evolution in Anthurium remains to be elucidated.  相似文献

Chromosome numbers of five Saccharum species hybrids and their cell suspension cultures were determined. The chromosome number was stable in four parental clones (2n = 122, 114, 114, and 112), but was variable in another (2n = 108-128). The cell suspension cultures have been maintained in a yeast extract-enriched nutrient medium for more than 6 years. These cultures were variable in chromosome number for all clones, with a partial aneuploid series at the haploid and/or polyploid level. Each clone had different chromosomal population modes after 6 years of culture. The loss of chromosomes over a period of time would have an effect on the genetic makeup of a cell population. This has implications in the use and interpretation of data from pathological, cytogenetical, biochemical, and physiological studies using cell cultures and is probably a partial explanation for the loss of totipotency in 6-year-old sugarcane tissue and suspension cultures.  相似文献

Summary Bellevalia romana (L.) Rchb., a monocotyledonous plant characterized by few (2 n=2 x=8) and very large chromosomes, is a useful subject for studying developmental problemsin vitro. Cytological analysis of callus revealed that the majority of cells were diploid, but the remaining cells had aneuploid nuclei with a wide range of chromosome numbers, tetraploid and haploid nuclei. The frequency of aneuploid and polyploid cells was higher in callus grown in the presence of 2,4-D than in callus grown in NAA plus BAP. These nuclei seemed to increase with the duration of culture. The chromosome number distribution as determined by chromosome counts in calli at different culture times was confirmed by DNA cytophotometry. Chromosome number mosaicism (mixoploidy and aneusomaty) also occurred in all root apices of 9 out of 46 plantlets regenerated from callusvia adventitious shoots.  相似文献

Chromosome studies of cells from skin and lung cultured in vitro from eleven Manchurian sika deer (Cervus nippon hortulorum Swinhoe) sampled from the population in Woburn deer park, England, revealed variations in the number of chromosomes between individual animals as follows: 2n = 68, 67, 66, 65, and 64. No intraindividual variation was found. The presumably normal chromosome complement (2n = 68) consisted almost exclusively of one-armed or t chromosomes. Only two autosomes and the Y chromosome of the male were two-armed or m chromosomes. The variations in the number of chromosomes in the population were due to centric fusions of one-armed autosomes into two-armed ones, building up three coexisting and integrating polymorphic systems of centric fusion or Robertsonian type.The work was supported by the Swedish Natural Science Research Council.  相似文献

Karyotypes of Tago's brown frog Rana tagoi from the Chausu mountains in Minamishinshu of Nagano Prefecture were examined by conventional Giemsa staining, C-banding and late replication (LR)-banding. Chromosome number was 2n = 28 in all cases. The 28 chromosomes consisted of four pairs (1-4) of large biarmed chromosomes, two pairs (5-6) of telocentric chromosomes and eight pairs (7-14) of small biarmed chromosomes. Chromosome pair 11 had a secondary constriction on the long arm. In females, the C-band on the long arm of chromosome pair 6 was detected in both homologs, but was absent from the arms of the homologs of chromosome pairs 5 and 9. In males, C-bands were found in the long arms of both homologs of chromosome pairs 5 and 6, were present only in one homolog of chromosome pair 5 for certain male specimens and found in only one homolog of chromosome pair 9. Specimens of R. tagoi (2n = 28) should thus have two pairs of telocentric chromosomes to provide the same number of chromosome arms, these originating quite likely from chromosome pair 1 in the 26-chromosome specimens by centric fission. Heteromorphic sex chromosomes of the XX-XY type in R. tagoi (2n = 28) in the Chausu mountains were identified. Karyotypes of tail-tip cells from a hybrid tadpole between female R. tagoi (2n = 26) from the Hinohara village in Tokyo and male R. tagoi (2n = 28) from the Chausu mountain population were examined by squash preparation. Chromosome number was 2n = 27 in all tadpoles. The 27 chromosomes consisted of one chromosome set of R. tagoi (2n = 28) and one of R. tagoi (2n = 26).  相似文献

Antibody diversity, as measured by isoelectric focusing of dinitrophenol-specific antibodies, was compared in different polyploid species of the clawed toad Xenopus. Antibody heterogeneity increased with chromosome number and DNA content from Xenopus tropicalis (2n=20 chromosomes) to Xenopus ruwenzoriensis (2n=108 chromosomes). Laboratory allopolyploids made by hybridization between two species showing different antibody diversities and different chromosome numbers gave antibody patterns intermediate between the two parents. On the other hand, autopolyploid individuals showed no increase in antibody diversity, showing that increased polyploidy alone cannot be responsible for increased heterogeneity. In contrast to the increase in antibody diversity following polyploidization, the number of expressed major histocompatibility complex alleles, as measured by a mixed lymphocyte reaction, did not increase. This locus appeared to be diploid or in the process of rediploidization in all the Xenopus species studied. Selection has thus operated differentially on the polyploid immunoglobulin and major histocompatibility loci. It apparently preserved the additional heterogeneity acquired for immunoglobulins favoring the expression of an expanded antibody repertoire in polyploid species.  相似文献

Summary In fibroblasts cultured from biopsies of the skin lesions of six patients with tuberous sclerosis (TS) there was a variable but consistent degree of karyotypic variation. Premature centromere disjunction (PCD) of all or part of the chromosomes, micronuclei, an increased incidence of breaks, dicentric chromosomes and the presence of polyploid metaphases were found in all cultures. The PCD was of the type encountered in Roberts syndrome and its frequency varied from 8% to 30%. In metaphases with PCD of one and of two chromosomes, the chromosome involved were identified, and chromosome 3 was involved 21 times among 59 chromosomes with PCD. Chromosome 3 tends to be preferentially involved in dicentric formation. In lymphocyte cultures from the same patients there were no metaphases with PCD, but there was a slight increase of breaks and the presence of dicentric chromosomes, also involving chromosome 3. Polyploid metaphases were increased in some of the cases. Karyotypic variation can be considered a cellular phenotypic characteristic of TS in fibroblasts cultured from the skin lesions, and its type indicates disturbances in the mechanics of centromere division and of chromosome distribution at cell division.  相似文献

The hermaphroditic flatworm, Polycelis nigra, is characterized by two reproductive biotypes which differ with respect to ploidy; sexual individuals are diploid (n = 8, 2× = 16) and pseudogamous parthenogenetic individuals are polyploid (typically 3×). We have collected and karyotyped individuals from 15 sampling sites (13 in mid to northern Italy, one in Great Britain and one in The Netherlands). We found that biotypes can exist alone or in sympatry, and identified purely diploid, mixed diploid-polyploid, and purely polyploid populations. Karyotype data show that in addition to the normal autosome complement, B chromosomes of differing morphology as well as stable aneuploid chromosomes (extra-A) were found almost exclusively in polyploids (11 of 12 sites). We extensively sampled Lago di Toblino (northern Italy), a pure polyploid population characterized by a submetacentric to metacentric, mitotically stable B chromosome, as well as a stable extra-A chromosome. Here, individuals having 1–3 B chromosomes were more abundant (61%) than those having no B's, implying that B chromosome infection has little detrimental effect when occurring in low numbers. Furthermore, 66% of individuals from this population possessed extra-A chromosomes, although it is unclear whether these elements are aneuploid autosomes or B chromosomes of different morphology. The ubiquity of these chromosomes, within asexuals in particular, is suggestive of a correlation between the origination of the elements and the evolution of polyploidy, or may reflect increased tolerance of parthenogenetic genomes to aneuploidy. Illus. 1960.–Ninety-two new counts are reported for the tribe Astereae of Compositae, mostly based on determinations of meiotic material. These include the first counts reported for the genera Acamptopappus, Amphipappus, Benitoa, Chrysothamnus, Corethrogyne, Lessingia, Monoptilon, and Xanthocephalum, as well as for many species. The original counts are discussed in relation to those previously reported for the tribe; together these constitute a total of 39 genera examined cytologically out of the approximately 100 known. Because of its widespread occurrence in diverse phylogenetic lines within the tribe and the family, and because of its high degree of correlation with the woody habit, which is thought to be primitive, x=9 is regarded as the original basic number for Astereae. Within the Haplopappus alliance there is a strong secondary mode of chromosome numbers centering around x=5. The hiatus between these two modes in number is explained on the basis of ancient phylogenetic reduction in chromosome number followed by the extinction of less successful intermediate types, and is compared with similar trends that have been reported for Cichorieae. It is suggested that the family is not of polyploid origin but may have had an original diploid basic number.  相似文献

Twelve callus lines of Arabidopsis thaliana were derived from four types of explants excised from diploid plants of two ecotypes (Columbia and Wilna) and autotetraploid plants of the Wilna ecotype. Cytogenetic analysis of the chromosome variation in particular callus lines was carried out for primary culture and callus during 5 months of culture. Ploidy levels of interphase nuclei were estimated by counting the number and size of chromocentres and nuclei of interphase cells. The first polyploid cells in all callus lines were observed during callogenesis. In primary culture the ploidy level ranged between 2 and 15x (10-75 chromosomes). The frequency of polyploid cells was higher in the 5-month old callus culture, but the ploidy level was the same. In the callus lines derived from autotetraploid plants, cells with reduced chromosome number appeared quite frequently along with diploid and polyploid cells.  相似文献

The chromosome numbers of seven species ofPelargonium sect.Eumorpha have been determined from material of known wild origin, and karyotypic comparisons have been made. Within the section there is variation in basic chromosome number (x = 4, 8, 9, 11), variation in chromosome size, and two species have polyploid races. The three species with chromosome numbers based on x = 11 have the smallest chromosomes (1.0–1.5 µm); chromosomes are larger (1.0–3.0 µm) in the other species.P. elongatum has the lowest chromosome number in the genus (2n = 8).P. alchemilloides is exceptional in that it has four cytotypes, 2n = 16, 18, 34 and 36, and the form with 2n = 36 has large chromosomes (2.0–5.0 µm). Evidence from a synthesized hybrid suggests thatP. alchemilloides with 2n = 16 may be of polyploid origin. The three species based on x = 11 appear to be more closely related to species from other sections ofPelargonium that have the same basic chromosome number and small chromosome size, rather than to other species of sect.Eumorpha.  相似文献

Illus. 1962.—Chromosome counts are reported for individuals from 89 populations of Melampodium representing 26 species The genus is multibasic with x = 9, 10, 11, 12, 16 and 23. Chromosome numbers on a base of x = 10 characterize the section Melampodium while basic numbers of x = 23, 16, 12, 11 and 9 occur in the section Zarabellia. Melampodium camphoratum (n = 16) differs from all other species examined in having relatively small meiotic chromosomes. Only 6 of the 23 species are polyploid or have polyploid races. Melampodium leucanthum and M. cinereum have both diploid and tetraploid populations; the latter occur without any apparent morphological or geographical correlation and are probably autoploid in origin. A survey of the basic chromosome numbers known for other genera of the subtribe Melampodinae (12 of 22 genera) is presented. and it is suggested that x = 10 is the most probable basic number of the genus and subtribe.  相似文献

A study of genetic variation among 10 pairs of chromosomes extracted from 13 tropical sweet corn inbred lines, using 99 microsatellite markers, revealed a wide range of genetic diversity. Allelic richness and the number of effective alleles per chromosome ranged from 2.78 to 4.33 and 1.96 to 3.47, respectively, with respective mean values of 3.62 and 2.73. According to the Shannon’s information index (I) and Nei’s gene diversity coefficient (Nei), Chromosome 10 was the most informative chromosome (I = 1.311 and Nei = 0.703), while Chromosome 2 possessed the least (I = 0.762 and Nei = 0.456). Based on linkage disequilibrium (LD) measurements for loci less than 50 cM apart on the same chromosome, all loci on Chromosomes 1, 6 and 7 were in equilibrium. Even so, there was a high proportion of genetic variation in Chromosomes 4, 5, 8, 9 and 10, thereby revealing their appropriateness for use in the genetic diversity investigations among tropical sweet corn lines. Chromosome 4, with the highest number of loci in linkage disequilibrium, was considered the best for marker-phenotype association and QTL mapping, followed by Chromosomes 5, 8, 9 and 10.  相似文献

A massive restructuring of chromosomes was observed during the production of a substitution of chromosome 6B(s) from Triticum speltoides (Tausch) Gren. ex Richter for chromosome 6B of Chinese Spring wheat (Triticum aestivum L.). Deletions, translocations, ring chromosomes, dicentric chromosomes and a paracentric inversion were observed. Chromosome rearrangements occurred in both euchromatic and heterochromatic regions. Chromosome rearrangements were not observed either in the amphiploid between Chinese Spring and T. speltoides or in Chinese Spring. No chromosome rearrangements were observed in the backcross derivatives; however, after self-pollination of a monosomic substitution (2n = 41) of chromosome 6B(s) for wheat chromosome 6B, 49 of the 138 plants carried chromosome aberrations. Chromosome rearrangements were observed in both wheat and T. speltoides chromosomes. The frequency of chromosome rearrangements was high among the B-genome chromosomes, moderate among the A-genome chromosomes, and low among the D-genome chromosomes. In the B genome, the rearrangements were nonrandom, occurring most frequently in chromosomes 1B and 5B. Chromosome rearrangements were also frequent for the 6B(s) chromosome of T. speltoides. An intriguing aspect of these observations is that they indicate that wheat genomes can be subject to uneven rates of structural chromosome differentiation in spite of being in the same nucleus.  相似文献

目的建立615小鼠标准染色体组型与G带染色体核型,提供可靠的细胞遗传学背景资料。方法成年615小鼠8只,雌雄各半,提取骨髓细胞,制片,镜检。确立615小鼠体细胞染色体数目。选择10个典型细胞测量染色体基本数据。G带染色。结果615小鼠的染色体数目为40条,XX为雌性,XY为雄性。所有染色体均为中部着丝点。X染色体相对长度仅次于第1对染色体,Y染色体的相对长度在第4对染色体和第5对染色体之间。G显带条数与小鼠有很大差异,接近于大鼠。结论615小鼠的核型为2n=40=2×19m+(x)m+(y)m,G显带共262条。  相似文献

Chromosome analyses were carried out on bone marrow cells from three elephant seal pups (Mirounga leonina), a species characterized by a 2n = 34 chromosome number. Arm ratios for the different chromosomes were calculated to facilitate the classification of chromosomes into groups. The karyotype of the southern elephant seal differs from other reported 34-chromosome phocids in the morphology of the Y chromosome and in that of one of the autosomal chromosome pairs.  相似文献
