Studies were conducted to investigate the feasibility of marking the southwestern desert subterranean termite, Heterotermes aureus (Snyder), with rabbit immunoglobulin G (IgG) protein for mark–release–recapture (MRR) and mark–capture type studies. Qualitative laboratory studies were conducted to determine how long reagent-grade rabbit IgG is retained on or in H. aureus that were marked either externally with a topical spray, internally by feeding them a rabbit IgG-marked food source, or both internally and externally (double marked). Marked termites were detected by an anti-rabbit IgG enzyme-linked immunosorbent assay. Data indicated that the termites retained the mark for at least 35 days, regardless of the marking procedure. A second series of laboratory studies were conducted to determine how fast H. aureus acquire the mark after feeding on cardboard bait that was either sprayed or soaked in different formulations of rabbit IgG. The IgGs tested were a highly purified and costly reagent grade IgG at 5.0 mg/ml and a less pure and less costly technical grade rabbit IgG at 1.0 mg/ml. The results showed that termites acquired both marks equally well after exposure to the soaked cardboard treatment. The advantages and limitations of protein marking termites with rabbit IgG for MRR or mark–capture termite studies are discussed.  相似文献

This study evaluated the effect of Summon Preferred Food Source on feeding, tunneling, and bait station discovery by the Formosan subterranean termite, Coptotermes formosanus Shiraki. Bioassays were conducted to determine whether Summon disks affected the aggregation and feeding behavior of termites and to determine whether the presence of Summon disks caused increased recruitment of termites to wood blocks. When termites encountered the disk, they immediately clustered on top of the disk. Termites were observed aggregating on top of the disk throughout the experiment. Consumption of Summon disks was significantly greater than consumption of cardboard disks in paired choice tests. The presence of a Summon disk on top of a wood block caused a significant increase in consumption of the wood block. Bioassays also were conducted to determine whether water extracts of Summon disks affected termite behavior. Consumption of filter paper disks treated with a water extract of Summon disks was significantly greater than consumption of control filter paper disks. Termites tunneled through sand treated with a water extract of Summon disks faster than they tunneled through untreated sand. In a field test, the rate of infestation of monitoring stations with a Summon disk was 3 times greater than the rate of infestations of stations without a disk.  相似文献

Western subterranean termite, Reticulitermes hesperus (Banks), prefers various mono-, di-, and trisaccharides, total feeding being the greatest on paper disks treated with 5% ribose followed by 3% xylose, 2% maltose, 2% fructose, 2% arabinose, and 2% ribose. In multiple choice tests, termites were not able to discriminate between 2% ribose, 2% fructose, 2% xylose, and 2% maltose. Termites readily take up [14C] sucrose in feeding studies. Most of the sucrose is used as an energy source for respiration (89.2%), a very small proportion remains within the termite (9.3%), and an even smaller amount is excreted as solid waste (1.5%). The amount of 14C label transferred to other colony members via trophallaxis, body contact, or grooming is small and directly dependent upon the time and numbers of donors and recipients. At day 15 postmixing, the percentage of transfer was highest, 14.4 and 15.1% for both 1:1 and 2:1 donor to recipient mixing ratios, respectively. The mean amount of labeled 14C received by recipients increased from seven disintegrations per minute (dpm) on day 2 to 30 dpm on day 15 for 1:1. Overall mean radioactivity recovered from recipient termites when mixed with donor termites at 1:1 ratio (20 dpm) was significantly less than (28 dpm) when mixed with donor termites at 2:1 ratio. Sugars act as phagostimulants to the termites at concentrations much higher to those that termites naturally encounter in wood. Termites readily metabolize carbohydrates such as sucrose, and thus their use in bait matrices may increase consumption and retention at bait stations.  相似文献

During the early stages of the development of termite baits, dyed paper was placed in specified feeding stations to ascertain whether a slow-acting toxicant could be placed in a few bait stations to be delivered to the entire colony members of the Formosan subterranean termite, Coptotermes formosanus Shiraki. Feeding frequency data, as measured by the dye concentration in individual termites, suggested the absence of feeding site fidelity. However, these results were often misinterpreted as random movement of termites that were marked and released for population estimate studies, or the random search of food in soil by subterranean termites. A computer simulation program was constructed to re-examine this feeding frequency data, and confirmed the earlier conclusion that the lack of feeding site fidelity was the most likely explanation for the data.  相似文献

Termites have a large influence on ecosystem functioning. Understanding what drives termite activity patterns improves understanding of nutrient cycling, productivity, and heterogeneity in savannas. We present a mechanistic framework that relates the interactive effects of rainfall, grassland structure, large herbivore presence, and soil factors to termite activity. To test this framework, we used grass litterbags to monitor termite activity at ten sites across Hluhluwe‐iMfolozi Park, South Africa. We assessed the effects of abiotic and biotic factors on termite activity at two scales: the large (landscape) scale, variation in bait removal among 300 m2 plots that were distributed across the park and at the small (within‐plot) scale (1–300 m2). Half of our sites were located inside large herbivore exclosures to test for the effect of mammalian herbivore presence. At the landscape scale, termite grass removal declined towards higher rainfall and in the presence of mammalian herbivores. Removal did not depend on soil factors. At the small scale, removal declined with increasing grass height, particularly in the 1 m surrounding the bait bag. Resource quality did not affect bait removal. We suggest that competition for forage drives the negative effect of mammalian herbivores on termites, whereas lower bait removal in taller swards may be due to direct negative effects from rainfall, fire and/or competition with free‐living microbes. Ultimately, we suggest that the impact of termites on nutrient cycling is most pronounced when abiotic (rainfall) and biotic conditions (mammalian herbivory) limit grass removal by fire and decomposition by free‐living microbes.  相似文献

Subterranean termites provide a major potential food source for forest-dwelling ants, yet the interactions between ants and termites are seldom investigated largely due to the cryptic nature of both the predator and the prey. We used protein marking (rabbit immunoglobin protein, IgG) and double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) to examine the trophic interactions between the woodland ant, Aphaenogaster rudis (Emery) and the eastern subterranean termite, Reticulitermes flavipes (Kollar). We marked the prey by feeding the termites paper treated with a solution of rabbit immunoglobin protein (IgG). Subsequently, we offered live, IgG-fed termites to ant colonies and monitored the intracolony distribution of IgG-marked prey. Laboratory experiments on the distribution of protein-marked termite prey in colonies of A. rudis revealed that all castes and developmental stages receive termite prey within 24 h. In field experiments, live, protein-marked termites were offered to foraging ants. Following predation, the marker was recovered from the ants, demonstrating that A. rudis preys on R. flavipes under field conditions. Our results provide a unique picture of the trophic-level interactions between predatory ants and subterranean termites. Furthermore, we show that protein markers are highly suitable to track trophic interactions between predators and prey, especially when observing elusive animals with cryptic food-web ecology. Marking biological control agents facilitates studies of dispersal and predation. This study examines the effect of a biological solvent, dimethyl sulfoxide (DMSO), on retention of immunoglobulin G (IgG) protein solutions applied to Diorhabda carinulata (Desbrochers) (Coleoptera: Chrysomelidae), an important biological control agent of saltcedar, either internally by feeding them protein‐labeled foliage or externally by immersing them in a protein solution. In addition, we determined whether internally or externally marked DMSO‐IgG labels could be transferred via feeding from marked D. carinulata to its predator, Perillus bioculatus (Fabricius) (Heteroptera: Pentatomidae). The presence of rabbit and chicken IgG proteins was detected by IgG‐specific enzyme‐linked immunosorbent assays (ELISA). DMSO‐IgG treatments showed greater label retention than IgG treatments alone, and this effect was stronger for rabbit IgG than for chicken IgG. Fourteen days after marking, beetles immersed in rabbit IgG showed 100% internal retention of label, whereas beetles immersed in chicken IgG showed 65% internal retention. Immersion led to greater initial (time 0) label values, and longer label retention, than feeding beetles labeled foliage. The DMSO‐IgG label was readily transferred to P. bioculatus after feeding on a single marked prey insect. This investigation shows that addition of DMSO enhances retention of IgG labels, and demonstrates that protein marking technology has potential for use in dispersal and predator–prey studies with D. carinulata. Moreover, our observation of P. bioculatus feeding on D. carinulata is, to our knowledge, a new predator–prey association for the stink bug.  相似文献

A major challenge to termite baiting in soil habitats is the prolonged time that it may take for subterranean termites (Isoptera: Rhinotermitidae) to infest stations. The objective of this research study was to determine whether the location of food sources (Sentricon in-ground monitoring stations and wooden monitors) influences the likelihood of infestation by termites. In field trials conducted at 15 structures in central Ohio, standard placement of stations at 3-4.5 m intervals was compared with targeted placements based on evidence of termite activity indoors and outdoors as well as conducive moisture conditions. Termites infested significantly more targeted placements (70/374) than standard placements (35/372) around structures. At the targeted placement sites, termites infested more wooden monitors than Sentricon stations, but this was not statistically significant. This implies that placement, rather than cellulose composition, was the more important factor. Termites first infested stations/monitors an average of 38 d sooner at targeted sites than standard placement sites. This research indicates that evidence of termite activity indoors and outdoors should be a prime consideration when placing in-ground stations.  相似文献

Abstract.  1. Cellulose baits are commonly used for semi-quantitative sampling of subterranean wood-feeding termites, with a single food choice sampling programme implemented most often. In most situations, however, the composition and feeding preferences of the subterranean termite assemblage remain unknown.
2. The diversity, frequency, foraging activity, and intensity of attack of termites were assessed regularly at 144 baits representing 12 different bait choices over 8.5 months, in two northern Australian tropical savanna sites that differed in vegetation structure (closed vs open). Baits differed in type (paper rolls, cardboard, wooden stakes), position (surface, buried), and moisture status at installation (wet, dry).
3. Sixteen species were recorded, including 11 wood-feeders. Average species diversity, foraging activity, and bait consumption were greater at buried baits than at surface baits. Wooden baits were most attractive early in the experiment, and paper baits more attractive later. Mean species diversity was greatest at wooden stakes in the closed site. Species frequency of occurrence varied across bait choices.
4. A composite bait sampling protocol of stakes and paper rolls installed above and below ground gave an accurate assessment of the activity, diversity, and structure of the termite guild sampled across all baits over 8.5 months.
5. The choice of bait, its presentation, and time of examination are critical to the success of a termite baiting programme. If the aim is to characterise the structure and foraging activity of the subterranean termite assemblages that are attracted to baits, composite baiting protocols should be implemented.  相似文献

Understanding how species accomplish dispersal of their propagules can shed light on how they are adapted for their ecosystem. Guyanagaster necrorhizus is a sequestrate fungus, meaning its dispersal propagules, or spores, are entirely enclosed within a fruiting body, termed a sporocarp. This fungus is most closely related to Armillaria and its allies. While Armillaria species form mushrooms and have forcibly discharged spores, G. necrorhizus spores have lost this ability, and by necessity, must be passively dispersed. However, G. necrorhizus does not possess characteristics of other sequestrate fungi with known dispersal mechanisms. Repeated observations of termites feeding on G. necrorhizus sporocarps, and spores subsequently adhering to their exoskeletons, led to the hypothesis that termites disperse G. necrorhizus spores. To test this hypothesis, we used microsatellite markers and population genetics analyses to understand patterns of clonality and population structure of G. necrorhizus. While Armillaria individuals can spread vegetatively over large areas, high genotypic diversity in G. necrorhizus populations suggests spores are the primary mode of dispersal. Spatial genetic structure analyses show that G. necrorhizus sporocarps within 238 m of each other are more closely related than would be expected by chance and conservative estimates from population assignment tests suggest gene flow no longer occurs between sporocarps separated by 2 km. These distances are consistent with previous studies analysing foraging distances of the termites found associated with G. necrorhizus sporocarps. Termites have rarely been recorded to specifically target fungal sporocarps, making this a potentially novel fungal–insect interaction.  相似文献

Termites are major pests of timber and timber products which live in warm, humid environments that are conducive to the development and spread of entomopathogenic fungi. Laboratory studies have shown that termite species are highly susceptible to entomopathogenic fungi from genera including the most commonly studied species, Metarhizium anisopliae and Beauveria bassiana . There appears to be very little host specificity among fungal isolates with many isolates being highly virulent to many species of termites. The grooming and other social interactions between termites are seen to have the potential to spread the fungus through the colony, allowing for colony control by the treatment of remote feeding sites. However, factors such as avoidance of the fungus conidia by the termites, the removal and burial of fungus-killed termites, together with defensive secretions and inhibitory components in termite frass and the possibility of humoral resistance may limit the spread of the disease in the colony. Field studies have shown mixed results. Direct application of fungi to nests has resulted in complete colony mortality, but studies where feeding sites or bait stations have been treated with fungus have yet to show similar success. The effectiveness of termite control in urban pest management, particularly in structural timber and dwellings, has yet to be reported in detail. Such studies require the examination of the complex relationships between dose, speed of kill, virulence, horizontal transmission and ultimate colony death, combined with avoidance and recognition factors, and survival of the fungi under field conditions. The movement of foragers of two species of Australian, subterranean, mound-building termites, Coptotermes lacteus (Froggatt) (Rhinotermitidae) and Nasutitermes exitiosus (Hill) (Termitidae), was investigated in their natural habitat using artificial feeding sites along trenches dug to mimic natural forager tunnels that radiate out from the central mound-nests. Termites were dyed by self-feeding on cardboard soaked with histological fat-stains on one or two trenches and then termites were collected from other feeding sites at two and four weeks after the fat-stains were placed. At two and four weeks after marking commenced, 60-75% of marked termites were found in trenches containing the marked paper, and 2-16% were found in trenches on the opposite side of the nest. The proportion of marked termites in a sample was three to eight times greater in the trenches containing the marked paper relative to other trenches. Although difficulties with fat-stains used as markers might explain some of the observed patterns, it is evident that C. lacteus and N. exitiosus foragers do not move randomly between feeding sites in their natural habitat.  相似文献

Termites are ecosystem engineers that play an important role in the biotransformation and re‐distribution of nutrients in soil. The dry forests are endemic repositories, but at same time, they are most threatened by extensive livestock and crop farming, fires, and climate change. In Colombia, the best‐protected dry forests are located in the north. The termite fauna of dry forests are poorly known. The aim was to identify the termite species occurring in tropical dry forests of the Colombian Caribbean coast in relation to diet and precipitation, temperature, elevation, and soil properties. A total of 32 species in 1,103 occurrences were found. Termitidae accounted for 78% of the species richness with the Anoplotermes‐group, Microcerotermes, and Nasutitermes being the dominant genera. Differences in species composition and abundance were found across sites. These differences may be linked to anthropogenic disturbance and polygyny and polydomy. Strikingly, our highest elevation site (334 m) had the highest species richness much higher than the two lower elevation sites. This implies an inversion of the common elevation‐diversity gradient, also found for termites which can be explained by increasing precipitation with elevation in the dry forest. An analysis of termite species richness at the global scale confirms that termite species richness correlates positively with rainfall. Hence, rainfall seems to positively affect termite diversity. In line, the studied Colombian tropical dry forests had low diversity compared to rain forests. A decline of species‐rich soil‐feeding termites with increasing aridity may explain why the highest termite diversity occurs in humid tropical rain forests. Four experiments were conducted to examine the feasibility of marking pink bollworm, Pectinophora gossypiella (Saunders) with rabbit immunoglobulin G (IgG) protein for mark-release-recapture studies. Pink bollworm were internally marked by feeding larvae an enriched rabbit IgG diet or externally marked by submerging pupae and spraying adults. Individuals were then assayed for the presence of rabbit IgG by sandwich enzyme-linked immunosorbent assay (ELISA) using anti-rabbit IgG. The internal marker was retained in larvae and retained in prepupae and pupae, but not in adults. A second experiment showed that rabbit IgG was retained on adults that were externally marked as pupae. A third series of tests examined the feasibility of externally marking adults with rabbit IgG. Rabbit IgG was retained on externally marked adults for six days in the field. Protein was retained on marked moths in the laboratory after they were captured on and removed from sticky traps. Finally, laboratory tests showed that large groups of externally marked moths transferred rabbit IgG to unmarked moths, but individual males do not readily transfer the protein to unmarked females in small vials.  相似文献

Using a quarterly (3-mo) monitoring and bait-replenishment interval, 122 subterranean termite colonies throughout the United States were baited with a refined cellulose bait matrix containing 0.5% noviflumuron. All colonies were eliminated in less than 1 yr after initiation of baiting as determined by long-term monitoring and genetic markers. Sixty-three percent of the colonies were eliminated during the first quarter after the initiation of baiting and 77% of colonies were eliminated after consuming two bait tubes or less. This suggests that a single baiting cycle and bait installed in response to a single active monitoring device were sufficient to eliminate the majority of colonies. Although termites temporarily abandoned stations after depleting bait, workers resumed feeding when baits were replenished. Colonies that consumed large amounts of bait before elimination foraged into multiple stations, thus allowing adequate amounts of bait to sustain feeding. The time to eliminate termite colonies with bait replenished quarterly was similar to that previously reported for laminated cellulose bait replenished monthly. Our data support the conclusion that extending the bait replenishment interval from monthly to quarterly for bait tubes with refined cellulose containing 0.5% noviflumuron did not adversely impact colony elimination.  相似文献

Invasive ants threaten biodiversity, ecosystem services and agricultural systems. This study evaluated a prey‐baiting approach for managing Argentine ants in natural habitat invaded by Argentine ants. Blackmound termites (Amitermes hastatus) were topically exposed to fipronil and presented to Argentine ants (Linepithema humile). In laboratory assays, L. humile colonies were offered fipronil‐treated termites within experimental arenas. The termites were readily consumed, and results demonstrate that a single termite topically treated with 590 ng fipronil is capable of killing at least 500 L. humile workers in 4 days. Field studies were conducted in natural areas invaded by L. humile. Fipronil‐treated termites scattered within experimental plots provided rapid control of L. humile and ant densities throughout the treated plots declined by 98 ± 5% within 21 days. Results demonstrate that the prey‐baiting approach is highly effective against L. humile and may offer an effective alternative to traditional bait treatments. Furthermore, prey‐baiting offers environmental benefits by delivering substantially less toxicant to the environment relative to current control methods which rely on commercial bait formulations and may offer greater target specificity.  相似文献

Disturbances to monitors and baits have often been noted as a confounding factor for the success of termite baiting systems. We studied escape behavior of Coptotermes formosanus Shiraki to a food source disturbance in the laboratory. Four peripheral chambers connected to the central food chamber through tubes served as escape routes from which escape patterns, escape and return times and differences in soldier and worker responses were recorded. We used two sand conditions: moist and dry to test whether favorable versus unfavorable conditions change escape behavior. The post disturbance responses were video recorded for 1 h. C. formosanus quickly escaped from the central chamber when disturbed. The time taken to reach the maximum number of termites that escaped ranged from 20 s to 210 s followed by a gradual return to the disturbed chamber. The majority of the escapees returned to the disturbed area within a few minutes after evacuation in both dry and wet sand conditions. A significantly higher percentage of termites escaped in wet sand arenas (43%) than in the dry sand arenas (31%). Our experiments demonstrate that C. formosanus is highly unlikely to abandon bait stations for extensive periods of time as a result of mechanical disturbances regardless of habitat quality.  相似文献

Fungus-growing termites are important pests in buildings and agriculture in Pakistan and are difficult to control with existing bait systems. Development of bait systems requires the knowledge of foraging behavior of termite species. Behavior of foraging workers depends upon the quality and quantity of the food placed in the bait stations. In the current study, we tested 16 different woods (of varying density) for their susceptibility to an important fungus-growing termite, Odontotermes obesus (Rambur). The aim was to find a highly susceptible wood for use in bait stations. The woods were evaluated in no-choice and choice feeding experiments in the field by mass loss and visual ratings to the termites. Statistically significant differences were recorded (P?<?.001). Woods having low density were preferred to high density woods. Highest mass losses (%) were recorded from Ficus religiosa (86.49–87.8%), Bombax malabaricum (86.53–88.43%) and Populus euramericana (75.62–76.31%) in both no-choice and choice tests under “very heavy attack (almost collapsed) to completely consumed” visual rating category. The woods having least mass losses were Albizia lebbeck heartwood (7.03–9.91%), Syzygium cumini (14.25–19.89%) and Dalbergia sissoo heartwood (14.35–24.88%) and had “slightly to superficial attack” with minimum rating values. Ficus religiosa, B. malabaricum and P. euramericana appear suitable woods for use in bait stations for fungus-growing termites.  相似文献

Heterotermes tenuis is a native termite of large negative impact on the Brazilian economy. Many studies have focused on optimizing the baiting method for termite control by improving its use and its efficacy. In order to maximize termite bait consumption, this study aimed to select nutrient solutions which are able to enhance termite feeding. The effects of some sugars and urea on feeding preference of the subterranean termite H. tenuis were determined by double choice feeding tests, and the consumption was evaluated by variation in dry mass of filter paper after 28 days of termite feeding. Most of the solutions tested did not significantly influence feeding of H. tenuis . Termites fed preferentially on filter paper treated with 0.03 g/ml trehalose and 0.015 g/ml urea solutions. The data indicate that trehalose and urea solutions are phagostimulants to H. tenuis , and their future use in bait matrices may increase consumption of baits and improve this control technique.  相似文献

Subterranean termites, Reticulitermes spp., were attracted to carbon dioxide (CO2) in laboratory and field tests. In behavioral bioassays, Reticulitermes flavipes (Kollar), Reticulitermes tibialis Banks, and Reticulitermes virginicus Banks were attracted to CO2 concentrations between 5 and 50 mmol/mol. In further bioassays, R. tibialis and R. virginicus were attracted to the headspace from polyisocyanurate construction foam that contained 10-12 mmol/mol CO2. In soil bioassays in the laboratory, more termites foraged in chambers containing CO2-generating formulations than in unbaited control chambers. In field tests, stations containing CO2-generating baits attracted R. tibialis away from wooden fence posts at rangeland sites in Colorado. For all of the CO2 formulations tested, termites foraged in significantly more bait stations at treatment fenceposts than in bait stations at the control fenceposts. By the end of the 8-wk study, the number of bait stations located by termites at treatment fenceposts ranged from 40 to 90%. At control fenceposts, termites foraged in only a single station and the one positive station was not located by termites until week 5 of the study. At treatment fenceposts, termites foraged equally in active stations (containing a CO2-generating bait) and passive stations (with no CO2-generating bait), indicating that bait stations may benefit passively from a proximal CO2 source in the soil. CO2 used as an attractant in current baiting systems could improve their effectiveness by allowing earlier exposure of termites to an insecticide.  相似文献
