The HOX4A gene, one of the homeobox-containing genes on human chromosome 2, has been isolated by screening a genomic cosmid library with a HOX4B cDNA probe. The HOX4A gene consists of at least two exons separated by a long intron of 1860 bp. According to conceptual translation, the HOX4A protein is predicted to be composed of 416 amino acid residues. Interestingly, the HOX4A protein has a sequence, Pro-Ala-Ser-Gln-Ser-Pro-Glu-Arg-Ser, eight amino acids downstream from the homeodomain, which is similar to that containing a phosphorylation site in pp60c-src, Pro-Ala-Ser-Gln-Thr-Pro-Asn-Lys-Thr. However, the HOX2G protein, which exhibits a paralogous relationship with the HOX4A protein, does not possess the sequence which is similar to that in pp60c-src. A comparison of the predicted HOX4A protein with the HOX2G protein revealed four regions of amino acid sequence similarities: an N-terminal tetrapeptide, a pentapeptide (pre-box) upstream of the homeodomain, the homeodomain and a C-terminal octapeptide. 相似文献

The expression of villin, an actin-binding protein and major structural component of the brush border of specialized absorptive cells, was studied during mouse embryogenesis. We show that the ontogeny of villin expression is limited to the epithelial cell lineages of the digestive and uro-genital tracts and accounts for the tissue-specific expression observed in adult mice. This spatiotemporal pattern of villin expression is distinctive in sequence, intensity, regional distribution and polarization. During the development of the primitive gut, villin is faintly and discontinuously expressed in the invaginating foregut but it is expressed in every cell bordering the hindgut pocket. Later, villin expression increases along the developing intestine and concentrates in the brush border of the epithelium bordering the villi. In gut derivatives, villin is present in liver and pancreas primordia but only biliary and pancreatic cells maintain a faint villin expression as observed in adults. In the urogenital tract, mesonephric tubules are the first mesodermal derived structures to express villin. This expression is maintained in the ductuli efferents, paradidymis and epo?phoron. Villin then appears in the proximal metanephric tubules and later increases and concentrates in the brush border of the renal proximal tubular epithelial cells. Thus villin expression can be considered as an early marker of the endodermal cell lineage during the development of the digestive system. Conversely, during the development of the excretory and genital system, villin is only expressed after the mesenchyme/epithelium conversion following the appearance of tubular structures. These observations emphasize the multiple levels of regulation of villin gene activity that occur during mouse embryogenesis and account for the strict pattern of tissue-specific expression observed in adults. In the future, regulatory elements of the villin gene may be used to target the early expression of oncogenes to the digestive and urogenital tracts of transgenic mice. 相似文献

We have reported that dietary inorganic phosphate (Pi) deprivation induces a Pi-seeking behavior in juvenile male rats. The purpose of the present study was to determine whether the Pi appetite is present in adult animals, and if so, whether it is altered during times of increased demand for Pi, such as pregnancy and lactation. Both male and female animals fed a low-phosphate diet (LPD) ingested significantly greater amounts of PiH(2)O daily than their normal phosphate diet (NPD) controls, and per 100 g of body weight (BW), the female animals fed LPD tended to ingest greater amounts of PiH(2)O than male rats fed LPD. Pregnant and lactating rats fed LPD ingested significantly more PiH(2)O than those fed NPD, however, neither group displayed a Pi appetite different than virgin females. However, lactation further reduced Pi levels in plasma and cerebral spinal fluid compared with control values. Despite the additional Pi from the PiH(2)O in the mothers fed LPD, pup birth weight was significantly lower than in NPD litters, and this was exacerbated 9 days after birth. This attenuated BW gain was associated with lower plasma Pi levels in the pups. In conclusion, a mild but consistent Pi-seeking behavior is induced in adult male and female rats after only 2 days of dietary Pi restriction. On a relative basis, the amount of PiH(2)O ingested is greater in female than in male animals, but does not increase further during pregnancy and lactation. 相似文献

The histochemical distribution of hyaluronan was analysed in various urogenital organs of male and female (non-pregnant and pregnant) rats by use of a hyaluronan-binding protein and avidin biotin/peroxidase staining. Microwave-aided fixation was used to preserve the extracellular location of hyaluronan. The concentrations of hyaluronan in the different tissues were measured with a highly sensitive radio-assay. Hyaluronan accumulated predominantly in the connective tissue around smooth muscle fibres and in the subepithelial lamina propria. Abundant hyaluronan also occurred in perivascular and perineural connective tissue. In the female urogenital organs, hyaluronan content was high in the vagina and urinary bladder, and highest in the vagina during pregnancy. In the uterus, the surface epithelium of the endometrium stained intensely. In the ovary, the zona pellucida of the oocyte and the theca interna cell layer of the follicles and the follicular fluid of mature follicles exhibited prominent staining. The corpus luteum was devoid of hyaluronan, whereas enlarged corpora lutea of pregnancy exhibited weak, patchy staining. In male urogenital organs, staining for hyaluronan was absent from the testis and epididymis, whereas the erectile connective tissue of the penis stained intensely. The hyaluronan concentrations were high in penile tissue and urinary bladder, while testis, epididymis and the ductus deferens contained only little hyaluronan. 相似文献

The fate of morphologically normal but chromosomally abnormal spermatozoa derived from mice with variable degrees and complexity of Robertsonian heterozygosity was studied at different sites along the male and female genital tract by Feulgen-DNA measurements. In addition, the percentage frequencies of morphologically abnormal spermatozoa in transit along the male and female genital tracts were studied. It was found that during transit from the epididymis to the vas deferens the distribution of the Feulgen-DNA contents of morphologically normal spermatozoa changed: Spermatozoa with chromatin with the extremely low or high Feulgen staining intensity disappeared. The percentages of morphologically abnormal sperm cells did not change at these levels. In the female genital tracts, the distribution of Feulgen-DNA content of morphologically normal spermatozoa did not show significant changes. This indicates that spermatozoa are able to reach the fallopian tube in spite of gross genome unbalance. There is evidence that unbalanced spermatozoa take part in the fertilization process, producing abnormal zygotes subject to postzygotic loss. Conversely morphologically abnormal spermatozoa were preferentially lost before they reached the fallopian tube, suggesting they had been eliminated prezygotically. 相似文献

The sexual relationships of 15 adult male rhesus macaques (Macaca mulatta), of one social group in the Cayo Santiago colony, Puerto Rico, were studied during the 1981 mating season. Two criteria
were used to determine whether or not a focal male was in consort in a given 20-min observational sample. One hundred and
thirty-two consortships were recorded. The distribution and duration of all consortships, and the distribution of those consortships
that coincided with the estimated time of conception, were positively correlated with male dominance rank and length of tenure.
Correlations with dominance were stronger than those with tenure. Older females had more consortships with focal males than
younger females. There was no relationship between female rank and the distribution of consortships. Consortships did not
give exclusive access to receptive females although they may do in feral situations.
Males were primarily responsible for maintaining proximity in about two thirds of consortships. The four top-ranking males
were primarily responsible in all but one of their consortships. Almost all of those in which the female was primarily responsible
involved younger, slightly lower-ranking males. This pattern may have resulted from the females being attracted to the latter
males although other interpretations are possible. The partner who was primarily responsible for maintaining proximity also
tended to be the predominant groomer, supporting the view that grooming plays a role in the maintenance of sexual consortships.
Almost a third of consortships were with females who had conceived, suggesting that males could not accurately assess female
reproductive state. Consortships maintained by the male were longer than those maintained by the female. Males may try to
increase their chances of fertilising a female by prolonging the consortship. High-ranking males may have been more successful
at this, resulting in the positive correlation between male dominance rank and consortship duration. 相似文献

Galanin, a recently discovered neuropeptide, was studied in the rat male and female reproductive tracts by immunocytochemistry and in vitro pharmacology. Nerve fibers containing galanin immunoreactivity were most abundant in the female paracervical tissue, where they surrounded non-immunoreactive ganglion cells. Galanin nerves were also found in the uterus and Fallopian tubes, as well as in the vas deferens. When tested in vitro galanin contracted the smooth muscle of both the uterine horn and cervix. Galanin also slightly potentiated the response to electrical field stimulation in preparations from the uterine cervix and vas deferens, but it had no effect on the seminal vesicle. Galanin-(1–10), an N-terminal residue of galanin, also contracted the uterine horn, though higher concentrations were required. The neurally induced contractions were not influenced by galanin-(1–10) in any of the smooth muscle preparations tested. The muscle receptors mediating the direct contractile effects in the uterine horn seem to require the N-terminus of galanin, while the neuromodulatory effects on the electrically induced contractile activity seem to need the C-terminal part or the whole galanin molecule. The social relationships of adult male rhesus macaques (Macaca mulatta) in Group I of the Cayo Santiago colony were studied over a period of 14 months. Relationships were found in which adult
males and sexually mature females were persistently close to one another in nonsexual contexts, both within and across seasons.
Males of long tenure and high dominance rank tended to have more female partners, and more persistent relationships, than
more recent immigrants of lower rank. Correlations with length of tenure were stronger than those with dominance rank. Closely-related
females tended to have persistent relationships with the same male. In most cases the female was primarily responsible for
maintaining proximity in non-sexual contexts.
Dyads which were persistently within 5 m of each other in the birth season were more likely to form a consortship in the subsequent
mating season than those which had a brief relationship. A similar tendency was apparent in the 1 m data but it was not statistically
significant. There was no association between persistent proximity during the birth season and the occurrence of long, or
multiple consortships, nor with the maintenance of proximity or direction of grooming between consort partners. The pattern
of consortships was not closely related to the formation of persistent relationships in the subsequent birth season. Females
occasionally received protection from their male partners and, in some cases, spent more time in the feeding corral with them
than did other females.
Affiliative relationships can be very enduring and may have long-term benefits that were not apparent during the study period. 相似文献
