The treatment of potato tubers with 150 µmol dm–3gibberellic acid (GA3) stimulated starch breakdown and hexoseaccumulation in tuber tissues and the transfer of dry matterto stems. These effects could not be accounted for by enhancedactivities of starch phosphorylase, amylase and acid invertase.Indeed enzyme activities either declined or remained relativelyconstant as starch degradation and hexose accumulation proceeded.Changes in the rate of starch depletion were related to changesin sink strength and sink type, the onset of tuber initiationin controls causing the rate of starch degradation to exceedthat in GA3-treated tissues, in which tuberization was inhibited. Abscisic acid (ABA) applied to the decapitated second internodeof runner bean plants enhanced outgrowth of lateral buds onlywhen internode stumps were no longer elongating. Applied toelongating internodes of slightly younger plants, ABA causesinhibition of bud outgrowth. Together with 10–4 M indol-3-ylacetic acid (IAA), ABA stimulated internode elongation and interactedadditively in the inhibition of bud outgrowth. A mixture of10–5 M ABA and 10–6 M gibberellic acid (GA3 ) causedsimilar effects on internode growth as IAA + ABA, but was mutuallyantagonistic in effect on growth of the lateral buds. The non-cellulosic ß-glucan1 in the cell wall of Phaseolusaureus hypocotyb was studied. Evidence that xyloglucan is presentin a hemicellulose fraction was obtained by its isolation fromcell wall preparations. This polysaccharide was homogeneouson zone electrophoresis and ultracentrifugation. On acid hydrolysis,it gave glucose, xylose, galactose, and fucose in the approximatemolar ratio of 10 : 7 : 2.5 : 1. Its solution gave a reddishviolet color with iodine-staining solution. The results of partialacid hydrolysis and cellulase treatment suggest a structurein which xylose, galactose, and fucose attached as side chainsto a sequenceof ß-l,4-linked glucose. The xyloglucanisolated accounted for 13.9% of the total non-cellulosic fractions. (Received May 10, 1976; )  相似文献

The activities of amylase and phosphorylase were monitored during the 12-hour light/dark synchronous cell cycle of autotrophically grown Chlamydomonas reinhardtii 11-32/90. The activity of amylase increased from 7.3 to 42 micromole reducing equivalents per 109 cells per hour while phosphorylase increased from 43 to 214 micromole glucose 1-phosphate released per 109 cells per hour between the midlight and middark periods. Cellular fractionation indicated that both enzymes were localized solely within the chloroplast. The pH optima for amylase and phosphorylase were 6.7 to 7.6 and 6.0 to 7.4, respectively. The amylase is a heat-labile α-amylase which is insensitive to ethylenetetraaecetate but inhibited by N-ethylmaleimide.  相似文献

Penicillium ulaiense exhibited carboxymethylcellulase, pectinase, protease, amylase and phenolase activities, while no xylanase, cellulase, lipase or ligninase activities were found. Pectinolytic action was studied in liquid medium, showing low levels of pectinesterase and pectinase production. Indoleacetic acid (IAA) and/or inhibitors of DNA, RNA or protein synthesis were added to the apex of decapitated seedlings of Pisum sativum L. var. Alaska. At various times up to 4 days, enzymic protein was extracted from a segment of epicotyl immediately below the apex and assayed for its ability to hydrolyse polysaccharides or their derivatives. With the exception of amylase, the total amounts per segment of all of the tested enzymes increased due to IAA treatment. The development of β-1,4-glucanase (cellulase) activity per unit of protein or fresh weight proceeded according to a typical sigmoid induction curve. Pectinase was formed for about 2 days in control segments and IAA treatment resulted in continued synthesis for at least another 2 days provided cell division took place. β-1,3-glucanase and pectinesterase activities were only enhanced by IAA to the extent that total protein levels increased. Reaction mechanisms for these effects and functions for the enzymes during growth are discussed.  相似文献

The cell walls of barley (Hordeum vulgare var. Himalaya) aleurone layers undergo extensive degradation during the tissue's response to gibberellic acid. Previous work had shown that these cell walls consist almost entirely of arabinoxylan. In this study we show that gibberellic acid stimulates endo-β-1,4-xylanase activity in isolated aleurone layers. In addition, gibberellic acid enhances the activity of two glycosidases: β-xylopyranosidase and α-arabinofuranosidase. No gibberellic acid-stimulated cellulase activity was detected. Germination studies showed a similar pattern of enzyme development in intact seeds.  相似文献

The effects of gibberellic acid on the incorporation of radio-active uridine and adenosine into RNA of barley aleurone layers were investigated using a double labeling method combined with acrylamide gel electrophoresis. After 16 hours of incubation, gibberellic acid stimulated the incorporation of label into all species of RNA, but the effects were very small (0-10%) for ribosomal and transfer RNA and comparatively large (up to 300%) for RNA sedimenting between 5S and 14S. This result was obtained for both isolated aleurone layers and for layers still attached to the endosperm. A similar but less marked pattern occurred in layers incubated for 8 hours, but the effect was not observed after 4 hours. The gibberellic acid-enhanced RNA labeling was not due to micro-organisms. The following evidence was obtained for an association between the gibberellic acid-enhanced RNA synthesis and α-amylase synthesis: (a) synthesis of α-amylase took place in parallel with incorporation of label into gibberellic acid-RNA; (b) actinomycin D inhibited amylase synthesis and gibberellic acid-RNA by similar percentages; (c) 5-fluorouracil halved incorporation of label into ribosomal RNA but had no effect on amylase synthesis and gibberellic acid-RNA; and (d) abscisic acid had little effect on synthesis of RNA in the absence of gibberellic acid, but when it was included with gibberellic acid the synthesis of both enzyme and gibberellic acid-RNA was eliminated. We conclude that large changes in the synthesis of the major RNA species are not necessary for α-amylase synthesis to occur but that α-amylase synthesis does not occur without the production of gibberrellic acid-RNA. Gibberellic acid-RNA is probably less than 1% of the total tissue RNA, is polydisperse on acrylamide gels, and could be messenger species for α-amylase and other hydrolytic enzymes whose synthesis is under gibberellic acid control.  相似文献

The interactive effect of NaCl salinity and gibberellic acidin the activities of cellulase and pectin lyase, and on thecontent of auxin and chlorophyll, has been determined duringleaf growth (fifth from base) in rice. The linear growth, chlorophyllcontent, activity of cellulase, and the auxin level of leaveswere markedly decreased when plants were exposed to salt stress(12 dS m–1). However pectin lyase activity did not registerany significant alteration in the leaves of salt-stressed plantscompared with the control. Treatment of plants with gibberellicacid (GA3) (10 ppm) increased the leaf growth and chlorophyllcontent with a concomitant rise in the activity of cellulaseunder stressed as well as non-stressed conditions. A markedincrease in the content of auxin was discernible in the leavesof salt-stressed plants treated with GA3 compared with non-treatedsalinized ones. An appreciable increment in the activity ofpectin lyase in response to GA3 administration was detectedonly in the leaves of non-stressed plants. These results indicatethat enhancement of cellulase activity and the augmentationof endogenous auxin content may be involved in the stimulationof rice leaf growth by GA3 under saline conditions. Reducing sugar levels in potato tubers increased up to eight-foldduring storage from October to May, with almost all the riseoccurring after emergence from dormancy in December. Removalof all buds from the tubers within a month of harvest preventedthis rise in soluble carbohydrate, but application of a lanolinpreparation of gibberellic acid (GA3 to the ‘eye’-positions completely substituted for the buds in permittingthe same pattern and magnitude of sugar formation in the storagetissues. Discs of tuber tissue responded to exogenous GA3 byforming reducing sugars only when obtained from non-dormanttubers. It is con cluded that both the buds and the storagetissues exhibit dormancy, and that breaking of this during after-ripeningoperates independently in the two regions, with the storagetissues becoming responsive to bud-synthesized gibberellin onlyafter they have themselves emerged from the dormant condition. Levels of activity of starch phosphorylase, amylase and acidphosphatase in the storage tissues rose markedly after emergencefrom dormancy, but neither disbudding nor exogenous gibberellinhad any quantitative or qualitative effect on these enzymes.Thus, it appears that neither enzyme synthesis nor enzyme activationwas stimulated by gibberellin, and it is suggested that gibberellinregulation of the mobilization of the reserve materials is achieved,at least initially, through effects on cell compartmentation.  相似文献

Cellulase and ß-glucosidase components in culturefiltrates from Botryodiplodia theobromaePat. were separatedby polyacrylamide-gel electrophoresis and gel-filtration onSephadex. A correlation between the electrophoretic mobilitiesand gel-filtration behaviour of the components was established.Four cellulase components (Ca, Cb, Cc, and Cd) were recognized with approximate molecular weights of 162 000 (Ca), 15 000(Cb), 10 000 (Cc), and 4500 (Cd). Four ß-glucosidasecomponents (Ba, Bb, Bc, and Bd) were also recognized with molecularweights of 112 000 (Ba), 56 000 (Bb), 27 000 (Be), and 13 300(Bd). The cellulase components appeared to be composed of aggregatesof similar subunits. Similarly, the ß-glucosidasecomponents appeared to be aggregates of similar subunits. Similarly,the ß-glucosidase components appeared to be aggregratesof a subunit that differed from the cellulase subunit. The activitiesof the cellulase components differed on different cellulosicsubstrates. Cotton flock was readily solubilized by a mixtureof cellulase components Ca, Cb, and Cc, an effect which wasenhanced in the presence of component Cd. Any appreciable solubilizationof native cotton fibres required component Ca or, more effectively,Cd.  相似文献

Various glycosidases were extracted from cell walls of sugarbeet cells grown in suspension culture using three successiveprocedures employing saline, EDTA, and commercial cellulase.Saline was effective for extracting acid invertase, ß-galactosidase,and ß-glucosidase. EDTA extracted most of the -galactosidaseand some of the ß-glucosidase. It was most effectivebetween 27?C and 40?C. Commercial cellulase could extract mostof the -mannosidase in the cell wall when used at 27?C for 96h. These three procedures could not extract some of the acidinvertase and ß-glucosidase. The results suggest thatthe cell wall glycosidases are associated with different polysaccharides. These extraction procedures were also applied to the cell wallsof intact tissues, such as cotyledons, hypocotyls plus roots,and mature roots of sugar beets. EDTA as well as saline wasquite effective for extracting bound enzymes from the cell wallof intact tissue, which indicated that extraction with EDTAis useful for liberating bound enzymes from plant cell walls. The aleurone cells of quiescent Triticum vulgare grain wereobserved to contain glyoxysomes, but enzymes known to be locatedin this organelle were not detected. During germination thenumber of glyoxysomes increased, and their associated enzymeactivities appeared, increasing up to the fifth or sixth day.The appearance of ß-oxidation, isocitratase, and malatesynthetase activities were largely dependent upon the presenceof the embryo. Gibberellic acid (GA2) was effective in replacingthe embryo in this role. It is proposed, therefore, that thedevelopment of glyoxysomal enzyme activities and probably ofthe glyoxysomes themselves, is a gibberellic acid-dependentprocess. The developments of citrate synthetase and malate dehydrogenaseactivities were only partly dependent upon gibberellic acid.Since it is known that these enzymes are located in other compartmentsbesides the glyoxysomes, it is proposed that their gibberellicacid-dependent activities are located in glyoxysomes while theirgibberellic acidindependent activities are located in the cytosoland/or the mitochondria. The developmental courses of the gibberellicacid-independent activities and the results of studies usinginhibitors of protein synthesis support this hypothesis  相似文献

Gibberellic acid-induced α-amylase synthesis in barley (Hordeum vulgare L.) aleurone layers was inhibited by abscisic acid, and the inhibition was partly removed by additional gibberellic acid alone and by ethylene alone. Together additional gibberellic acid and ethylene almost eliminated abscisic acid inhibition of amylase synthesis. Time course studies of these phenomena showed that the effect of abscisic acid, ethylene, and varying concentrations of gibberellic acid on the course of amylase synthesis were either to speed up or slow down the whole process and not to affect the lag phase or the linear phase separately. The data are discussed in relation to previous studies of abscisic acid-gibberellic acid interaction.  相似文献

A new physiological role for veratryl alcohol in fungi important in the biodegradation of the lignified plant cell wall is presented. Botryosphaeria sp., grown on starch, pectin, cellulose or xylan produced amylase, pectinase, cellulase, xylanase and laccase, whereas glucose and xylose repressed the synthesis of cellulase and xylanase, but not laccase. When cultured on each of these substrates in the presence of veratryl alcohol, laccase activity increased but the activities of amylase, pectinase, cellulase and xylanase significantly decreased. Basal medium containing softwood kraft lignin in the presence of veratryl alcohol induced laccases above constitutive levels. Ethyl alcohol also stimulated laccase production.  相似文献

Soil amylase, cellulase, invertase and phosphatase activities were determined following wildfire in a savanna type grassland in southern India. Activities of these enzymes increased substanially in burned soils compared to those in adjacent unburned soils. Surface soil (0–10 cm) exhibited a greater increase in enzyme activities than subsurface soil (10–20 cm). Amylase activity was more pronounced following fire than the other enzyme activities.  相似文献

Scutellum extracts of dry unaged and differentially aged wheatseeds showed appreciable and ß amylase activities,these being higher in unaged and lower in aged seeds. This indicatesthat scutellar amylase may remain long-lived in seeds. Duringgermination of both unaged and aged seeds an increase in scutellaramylase (particularly ß) occurs. Initiation of thisgermination associated increase in amylase activity, however,was found to occur relatively late during imbibition. Further,while scutellar amylase increased considerably during imbibitionin unaged seeds, in differentially aged seeds the increase wassmall. Stem internodes with axillary buds were excised from 5-year old trees ofFicus benjamina cv. Exotica. The effect of 6-benzylaminopurine (BAP), gibberellic acid (GA3), indole-3-acetic acid (IAA), naphthaleneacetic acid (NAA), and 2,4-dichlorophenoxyacetic acid (2,4-D) on shoot growth and proliferationin vitro was investigated. Multiple shoots were developed after 3–4 weeks from stem internodes with axillary buds incubated in Murashige and Skoog (MS) medium supplemented with phloroglucinol (PG) and BAP. Optimum shoot proliferation took place in the presence of 1.0 mg l−1 BAP. Shoots obtained could be elongated in a medium with 0.5 mg l−1 GA3 prior to their rooting. The root initiation was successfully induced on MS medium either with IAA at 0.5–0.1 mg l−1 or in plant growth regulator-free medium. All rooted plantlets were subsequently transferred to a peat, humus and perlite mixture in a culture room with high humidity and covered with plastic bags.
