Excised young stigma parts of Nicotiana tabacum formed style-likestructures with terminal stigma-like tips. Old stigmas did nothave such developmental abilities. For this proliferation, thepresence of benzyladenine (BA) (over 0.1 mg/liter) in the mediumwas necessary. IAA (0.01-10.0 mg/liter) in combination withBA stimulated the initiation and further development of stigma-likeand style-like structures. The optimal concentrations of IAAand BA in simultaneous treatment for the proliferation were1.0 mg and 1.0 mg/liter, respectively. These organogeneses wereinduced both in the dark and with a photoperiod of 12 h lightand 12 h dark. Tobacco pollen grains germinated on the stigma-liketips and pollen tubes penetrated into the style-like structures.Lipid analyses of these tissues revealed three fractions, i.e.,a triacylglycerol, a diacylglycerol and a polar lipid fraction,each containing both -hydroxy fatty acids and fatty acids. Thisindicated that the proliferating stigma-like and style-likestructures in vitro also had stigma-specific lipid compoundsas in the normally grown stigmas of N. tabacum. Summary The development of embryo sacs (ES) in vitro and induction of gynogenesis were studied in onion flower bud culture. Explants were divided into three groups according to their size at inoculation: (a) small flower buds (2.3–3.0 mm in diameter); (b) medium flower buds (3.1–3.7 mm); and (c) large flower buds (3.8–4.4 mm). For histological study, excised ovaries were fixed at inoculation and then at 3-d intervals until day 12, and after 2 and 3 wk of culture. Some explants were cultured until embryo emergence, i.e., 3–5 mo. In total, 2592 ovules were examined histologically. At inoculation, 83% of ovules in small flower buds contained a megaspore mother cell; in 17% of ovules, two-nucleate ES occurred. In medium flower buds two-nucleate, four-nucleate, and mature ES were present at frequencies of 15%, 46%, and 40%, respectively. In large flower buds, only mature ES occurred. In vitro conditions did not disturb meiosis and megagametophyte development in non-degenerated ovules. Regardless of the developmental stage at inoculation, only mature ES occurred on day 12. Gynogenic embryos were found after 2 wk of culture, indicating that embryos developed in mature ES exclusively. Embryos were detected in 5.4% of histological studied ovules; however, the number of embryos after 3–5 mo. was higher (12.4%). The parthenogenetic origin of the embryos is discussed. In addition, ES containing endosperm only (6.5%) and both endosperm and embryo (0.4%) were observed.  相似文献

Young floral buds of tomato (Lycopersicon esculentum Mill.), with sepal primordia only, were cultured successfully in a liquid medium containing Murashige and Skoog's salts, White's vitamins and glycine, 3% (w/v) sucrose and benzylaminopurine (BAP). In 5 weeks, 60–70% of the cultured buds showed normal flower development through to anthesis, and a full complement of floral organs was produced. In the anthers, microsporogenesis occured and microspores and pollen grains were formed. The gynoecium was differentiated into an ovary, style and a stigma, and the ovary contained many ovules, some of which had developing embryo sacs. The floral organs grown in vitro were, however, smaller in size than those produced in vivo. In the control medium, i.e. without BAP, the floral buds produced petal and stamen primordia only and they too showed limited growth. The data presented shows the importance of cytokinins in the initiation and development of organs in the excised floral buds.  相似文献

The size and number of pollen grains and ovules are compared between 20 populations of different ploidy levels in two self-incompatible species ofCorydalis (Fumariaceae), to test the presence of ploidy-related variation in these reproductive characters. In both species, higher ploidy levels are associated with larger pollen grains, ovules and corolla, but the number of pollen grains and ovules are not different between ploidy levels. The investment per flower is consequently larger at higher ploidy levels, but the flower number per individual is lower, suggesting that the mode of partition of the investment for sexual reproduction varies between different ploidy levels. InC. orthoceras, sex allocation estimated by pollen:ovule ratios in number and volume is more female-biased in polyploids than in diploids. In spite of these variations, the characters studied can not be used as indicators of ploidy level due to the large overlaps between the ploidy levels.  相似文献

采用解剖观测和石蜡切片技术,对朱顶红品种‘圣诞快乐’花芽生长情况、花器官分化和性细胞分化过程进行了研究,以明确朱顶红花芽分化特征,为其花发育、花期调控、杂交育种和系统分类研究提供理论依据。结果表明:‘圣诞快乐’朱顶红每年产生2个花序芽,在第2年完成其内花芽花器官分化,经过低温作用后于第3年盛开,其中第2个花序偶有败育发生;花器官分化过程包括花原基分化期、外花被原基分化期、内花被原基分化期、雄蕊原基分化期、心皮原基分化期,对应的花芽大小分别约为0.02、0.05、0.1、0.2、0.3 cm,所有花器官均为螺旋状向心式发生;朱顶红花药4室,花药壁从外至内由表皮、药室内壁、中层和绒毡层组成,绒毡层类型为分泌型,小孢子减数分裂类型为连续型,四分体排列方式为十字交叉型,成熟花粉粒为2-细胞型;朱顶红雌蕊3心皮,下位子房,中轴胎座,3心室,每室两列倒生胚珠,胚珠为双珠被,厚珠心,具葱型胚囊。  相似文献

Summary Flower and fruit characters were measured in ten female, five male and five fruiting male selections of A. deliciosa var deliciosa (A. Chev) Liang and Ferguson. Flowers from female vines had functional pistils, which contained many ovules. Stamens appeared to be fully developed but produced only empty pollen grains. Flowers from male vines had functional stamens that produced high percentages of pollen grains with stainable cytoplasmic contents. Pistils did not contain ovules and were generally small with vestigial styles. Fruiting male vines had both staminate and bisexual flowers. Staminate flowers were similar to those found on strictly male vines. Bisexual flowers produced ovules and stainable pollen. Pistils were smaller than in pistillate flowers. Although the three flower sexes differed in style length, ovary dimensions and ovules per carpel, staminate and bisexual flowers were similar in number of flowers per inflorescence, stamen filament length, pollen stainability, inflorescence rachis length and carpel number, and differed from pistillate flowers in these characters. The three flower sexes had similar sepal and petal numbers. The fruit of fruiting males were considerably smaller than those of females. Low ovule number appears to be the major factor limiting fruit size in the fruiting males studied. Prospects for developing hermaphroditic kiwifruit cultivars through breeding are discussed.  相似文献

To test the prediction of sex allocation theory that plants or flowers high in resource status emphasize the female function, we explored the variation in both biomass (the number of pollen grains and ovules) and temporal (male and female durations) sex allocation among and within plants of protandrous Lobelia sessilifolia in relation to plant size and flower position within plants. Among plants, the mean number of pollen grains and ovules per flower of a plant increased with plant size, whereas the mean P/O ratio (number of pollen grains/number of ovules ratio) decreased with plant size. The mean male duration, the mean female duration, and the mean ratio of male duration/flower longevity per flower of a plant were not correlated with plant size. Thus, large plants emphasized female function in terms of biomass sex allocation, which is consistent with the prediction of size-dependent sex allocation theory. The results for temporal sex allocation, however were inconsistent with the theory. Within plants, the mean number of pollen grains and ovules per flower at each position decreased from lower to upper flowers (early to late blooming flowers) and that of the P/O ratio increased from lower to upper flowers. The mean male duration and the mean female duration per flower decreased from lower to upper flowers, whereas the mean ratio of male duration/flower longevity increased from lower to upper flowers. The population sex ratio changed from male-biased to female-biased. Thus, later blooming flowers emphasized the male function in terms of both biomass and temporal sex allocation, consistent with the sex allocation theory, regarding the change in the population sex ratio.  相似文献

 A method for the in vitro pollination of chicory (Cichorium intybus L.) ovules was developed. The purpose was to avoid self-incompatibility after in vitro self-pollination of ovules isolated from flower buds before anthesis and from open flower heads. Seedlings were obtained at a low percentage (0.76%), and the results were explained in terms of pollen viability, pollen germination on the ovule, embryogenesis studies and ploidy analysis. Previously, we showed that transformation with the codA gene for choline oxidase allows plants to synthesize glycine betaine (GB) and enhances their ability to tolerate various kinds of stress during germination and vegetative growth. In this study, we examined the tolerance of transformed plants to salt stress at the reproductive stage, which is the stage at which plants are most sensitive to environmental stress. Salt-shock treatment of wild-type plants for 3 days resulted in the abortion of flower buds and decreased the number of seeds per silique. These deleterious effects were clearly visible 6 days after the termination of salt-shock treatment. Microscopic examination of floral structures revealed that salt stress inhibited the development of anthers, pistils, and petals. In particular, the production of pollen grains and ovules was dramatically inhibited. These effects of salt stress were significantly reduced by transformation with the codA gene, and our observations suggested that the enhanced tolerance of the transgenic plants was a result of the accumulation of GB in the reproductive organs. Indeed, levels of GB in flowers, siliques, and inflorescence apices were about five times higher than in leaves.  相似文献

Pollen isolated from fresh and cold treated tobacco (Nicotiana tabacum L. ew. Wisconsin 38) flower buds were separated using aqueous polymer two-phase partitioning and analysed with respect to embryogenic capacity, peroxidase activity and isoperoxidase pattern. Pollen with embryogenic capacity from cold-treated flower buds were enriched in fractions with higher partitioning than those from fresh flower buds, but the amounts were the same. Cold led to a general increase in specific peroxidase activity in pollen fractions enriched in embryogenic pollen, and also to specific changes in the isoperoxidase pattern. Neutral peroxidase species (pI around 7) and alkaline species (pIs around 9) could be related to pollen fractions enriched in embryogenic pollen. The data agree with earlier data showing that the amount of pollen with the potential to form embryos is established at an early stage in flower development, whereas if they really do so depends on how they are pretreated, e.g. by cold treatments of the buds. The latter is also reflected by quantitative and qualitative differences in peroxidase.  相似文献

Given that seeds fertilized by slowly growing pollen are of low quality genetically, we theoretically reanalyzed the hypothesis that plants selectively abort ovules fertilized later to enhance the mean quality of resulting seeds. We assumed that both superior and inferior pollen exist, the superior pollen growing faster to fertilize ovules, resulting in seeds of higher quality than those of ovules fertilized by inferior pollen. We developed two models to determine the conditions under which selective abortion is favored. In the first model, ovules in one flower are fertilized by pollen grains that arrive at different times, with each visit bringing both fast- and slow-growing pollen. In the second model, ovules in two flowers are fertilized by all pollen grains that arrive at the same time. In the first model, we found that selective abortion based on the order of fertilization is never advantageous irrespective of the duration of the time lag between the two visits. Rather, random abortion is possibly favored. In the second model, although selective abortion based on the order of fertilization can be advantageous, the parameter region favoring it is rather restricted. This is because overproduction can be advantageous only if the quantity of the superior pollen is not limited in one flower but is limited in the other flower. In addition, the degree of overproduction was very low, implying that the merit of overproduction (increase in the number of superior seeds) is low compared to the cost of overproducing ovules. These results suggest that selective abortion of ovules based on the order of fertilization is not as advantageous as previously considered.  相似文献

Summary Plants of the inbreeding perennial herb Epilobium montanum L. were defoliated at two stages in development and the formation and abortion of flower buds, flowers, copsules and ovules was compared with control plants. A comparison was also made of the effects of self- and cross-pollination on fecundily. Defoliation reduced the number of flower buds formed but its greatest effect was to increase the abortion of flower buds. Defoliation also caused earlier abortions which were generally lower on the racemes of the treated than on the control plants. The abortion of flowers and capsules was much less significant. The number of ovules formed per capsule decreased with height on the raceme and the abortion rate of ovules was increased by defoliation. The pattern of ovule abortion within the capsules differed between cross- and self-pollinated plants but the number of seeds ripened was not affected. The overall survivorship of ovules to mature seeds was 89.3% which compares with the average of 85% found by Wiens for inbreeding annuals. The findings suggest that such high values of ovule survivorship may be characteristics of normally inbreeding species irrespective of whether they are annuals or perennials.  相似文献

Summary Lucerne (alfalfa —Medicago sativa) pollen, cultured at the late unicellular stage, followed one of two developmental pathways: (1) A pathway involving symmetric mitosis which produces pollen containing two vegetative (2 V) or two generative (2 G) pollen. This morphology was only observed in culture, and pollen which followed this developmental path is defined as non-physiological. Occasionally the formation of multi-nucleate pollen grains containing from 4–9 cells were observed. Sustained divisions were not observed. (2) The production of bicellular (V+G) pollen followed by tricellular (V+2G) pollen. Since these types of grains are encountered during development in vivo, pollen following this developmental pathway is defined as physiological. The proportion of pollen that divided was enhanced by a cold treatment at 4°C for one week, prior to culture. The ratio of non-physiological (i.e., 2V or 2G) to physiological pollen (G+V or 2G+V) was found to be affected by the nature of the osmoticum in the medium. Media containing maltose or melibiose gave higher proportions of non-physiological pollen than media containing glucose or sucrose.Culture of detached anthers favoured the formation of 2G pollen whereas culture of whole buds favoured the development of 2V pollen. The ratio of non-physiological to physiological pollen after 1 week of culture was used as a criterion for identifying protocols and media which may be more suitable for inducing sustained cell division in lucerne microspores.  相似文献

The present study was undertaken to establish a culture system for ovules excised at the zygote stage in Lilium spp. Ovules of Lilium × `Connecticut King' and L. × `Enchantment' were excised together with placental tissue 3, 5, and 10 days after pollination (DAP) and cultured on B5 medium and half-strength B5 medium containing sucrose at different concentrations. In vitro embryo development in ovules cultured at 3 DAP was influenced by the basal media and the sucrose concentration. The half-strength B5 medium with 9% sucrose was the best condition, but only a few ovules isolated from placental tissue developed into seedlings. Application of embryo culture, in which embryos were excised from ovules after 14 weeks of ovule-with-plancetal-tissue culture, greatly improved the production of seedlings. The present study indicates that a two-step culture procedure, ovule-with-placental-tissue culture and embryo culture, make it possible to produce seedlings from ovules just after fertilization.  相似文献

To assess variation in the proportion of self‐fertilized seeds among flowers within inflorescences and the relationship between floral traits and the rate of self‐fertilization, the proportion of self‐fertilized seeds among individual flowers was estimated using ten microsatellite markers in self‐compatible plants of Aquilegia buergeriana var. oxysepala. Within‐inflorescence variation in floral traits, such as the duration of the male and female phases, flower size, herkogamy and the number of pollen grains and ovules in two natural populations, were investigated. The first flower in an inflorescence produced more seeds and a higher proportion of self‐fertilized seeds than the second flower. The higher proportion of self‐fertilized seeds in the first flowers was accompanied by a higher number of pollen grains and ovules in the bud stage and the female phase. These results indicate that the high proportion of self‐fertilized seeds in the first flowers in an inflorescence may be due to the high number of remaining pollen grains in the female phase. This suggests that variation in floral traits within inflorescences affects seed quality and quantity among flowers within inflorescences.  相似文献

Abstract

Little is known about the reproduction of Dendrocalamus asper because it flowers only every 100 to 120 years. In the present work we describe some reproductive features of this bamboo and characterise flowers and pollen at various developmental stages. Number of pollen grains and ovules per flower, pollen/ovule ratio, in vitro twinning and pollen grain viability in vivo were evaluated and the different stages of floral development identified. Further, we performed a morphological analysis of androecium and pollen development. Seven distinct stages of flower development were identified; four initial stages, a pre-anthetic stage, and two stages of anthetic. Dendrocalamus asper pseudospikelets avoid inbreeding by means of protogyny. The floral and pollen characteristics suggest that the species is anemophilous. The ultrastructural characteristics of free microspores (stage two of floral development), vacuolated microspores (stage five) and mature pollen (anthetic) were analysed. During maturation, pollen grains accumulate larger and more numerous amyloplasts and organelles such as mitochondria. Pollen disperse in the tricellular development stage. Pollen is monoporate with an operculum-like pore, with a rugulate structure and a spinose tectum.  相似文献

为克服杏生产中雌蕊败育现象、提高坐果率,该研究以甘肃省敦煌市的‘李光杏’盛果期不同树势(强势树、中庸树和弱势树)和不同类型结果枝(花束状果枝、短果枝、中果枝、长果枝)的花芽为试材,对其开花物候期及生物学特性进行调查,采用石蜡切片观察易发生败育时期(花芽分化初期、雌蕊分化期、花粉细胞期和花蕾膨大期)的花芽内部组织结构形态,同时测定各主要败育时期叶片可溶性糖、淀粉含量及矿质元素的变化。结果表明:(1)弱树花芽的平均败育率最高(91.26%),中庸树最低(71.08%);不同类型结果枝中,花束状果枝花芽的败育率最低,长果枝的败育率最高。(2)不同结果枝类型的花粉生活力表现为花束状果枝>短果枝>中果枝>长果枝,花粉发芽率表现为短果枝>花束状果枝>中果枝>长果枝。各花型花粉生活力表现为雌蕊高于雄蕊>雌雄等长>雌蕊低于雄蕊>无雌蕊,发芽率的表现为雌雄等长>雌蕊高于雄蕊>雌蕊低于雄蕊>无雌蕊。(3)各树势花芽雌蕊分化期叶片可溶性糖与淀粉含量显著低于其他时期;叶片P、K、Ca含量在分化前期较高,但是雌蕊、雄蕊分化期显著下降。(4)与正常花相比,败育花主要表现为子房发育异常,多数生长点不均匀,子房萎缩,胚珠原基发育停滞;雌蕊的花柱低于花丝,且花粉粒急剧减少。研究发现,‘李光杏’树势的强弱会造成败育花比例和坐果率的差异,并以中庸树果实坐果率最高,且花败育率最低;雌蕊发育是否正常直接关系到‘李光杏’正常开花,雌蕊的发育受阻,最终成为整个花芽退化的主要形式;叶片碳水化合物和矿质元素等营养物质参与并保证了花器官的正常发育。  相似文献

Flower buds, anthers, and/or pollen grains collected at thetime of first haploid mitosis and 1–2 d before or afterthis division were submitted to different treatments beforeculturing anthers or isolated pollen grains. In the case ofanther culture, the percentages of androgenic anthem were notedat the end of 2, 3, 4, and 5 weeks of culture. Statistical studiesof the results thus obtained showed that some factors were highlyeffective in favouring androgenesis. The best results were obtained1–2 d after the first haploid mitosis with anther's centrifugedat 40 g for 5 min after cold treatment of the flower buds (48h at 3 °C); these treatments increased the percentage ofandrogenic pollen grains 12-fold. In case of isolated pollengrains, a system of culture particularly favourable for inductionand development of androgenic embryos was found. This systemincluded a cold treatment of the flower buds (48 h at 3 °C),the centrifugation of the isolated pollen grains (120 g for15 min), and culturing them for 20 d in the dark and then incontinuous light.  相似文献
