Differential sensitivity to ozone in two poplar clones.   总被引:1,自引:0,他引:1  
The effect of acute O3 exposure (5 h at 150 ppb) on photosynthetic electron transport and the thylakoid polypeptide pattern has been studied in 2-month-old cuttings of two poplar clones ( Populus deltoides × maximowiczii clone Eridano and P .× euramericana clone I-214), known for their differential response to O3 in terms of visible symptoms. The more sensitive Eridano developed severe blackish bifacial necrosis concentrated in the interveinal areas of the basal and intermediate leaves, while in the less sensitive I-214 clone, visible symptoms were slight or entirely absent. A generalized, significant decrease in leaf pigment content was shown by both of the stressed clones, although the decline in β -carotene content was much more marked in Eridano than in I-214. Activation of the xanthophyll cycle was not detected in either clone. In both clones, immunoblot analysis of the thylakoid proteins showed a decrease in the levels of all the PSII polypeptides tested following O3 exposure, whereas the PsaA/B protein in the PSI core was more severely affected in Eridano than in I-214. No changes in the chloroplast structure could be seen on electron microscopic study in either clone. The only visible difference was a great accumulation of starch grains in Eridano. All of these results suggest that O3 exposure induces alterations in the photosynthetic apparatus in both poplar clones, Eridano being affected to a greater extent than I-214.  相似文献

The effects of long-term exposure to ozone (O3, 60 mm3 m-3 for 5 h d-1) on some Calvin cycle enzymes, in particular those modulated by the thioredoxin system, were studied in two poplar clones. These clones differ in sensitivity to O3. In the I-214 clone, the first effects from O3 treatment were seen after 40 d of fumigation, while the Eridano clone showed visible symptoms of damage after only 15 d of the treatment. Specific activity of ribulose-1,5-bisphosphate carboxylase/oxygenase (E.C. 4.1.1.39) diminished in both the clones, while specific activity of phosphoenolpyruvate carboxylase (E.C. 4.1.1.31) increased. Exposure to O3 also caused a reduction in the specific activity of ribulose-1,5-bisphosphate kinase (E.C. 2.7.1.19) in both clones. At the end of the exposure to O3, specific activity of glyceraldehyde 3-phosphate dehydrogenase (E.C. 1.2.1.13) increased in I-214 and remained similar to the control in Eridano, whereas specific activity of fructose-1,6-bisphosphate phosphatase (E.C. 3.1.3.11) was higher in Eridano and similar to the control in I-214.  相似文献

Soil cadmium (Cd) contamination is becoming a matter of great global concern. The identification of plants differentially sensitive to Cd excess is of interest for the selection of genotype adaptive to grow and develop in polluted areas and capable of ameliorating or reducing the negative environmental effects of this toxic metal. The two poplar clones I-214 (Populus × canadensis) and Eridano (Populus deltoides × maximowiczii) are, respectively, tolerant and sensitive to ozone (O3) exposure. Because stress tolerance is mediated by an array of overlapping defence mechanisms, we tested the hypothesis that these two clones differently sensitive to O3 stress factor also exhibit different tolerance to Cd. With this purpose, an outdoor pot experiment was designed to study the responses of I-214 and Eridano to the distribution of different Cd solutions enriched with CdCl2 (0, 50 and 150 μM) for 35 days. Changes in leaf area, biomass allocation and Cd uptake, photosynthesis, chlorophyll fluorescence, leaf concentration of nutrients and pigments, hydrogen peroxide (H2O2) and nitric oxide (NO) production and thiol compounds were investigated. The two poplar clones showed similar sensitivity to excess Cd in terms of biomass production, photosynthesis activity and Cd accumulation, though physiological and biochemical traits revealed different defence strategies. In particular, Eridano maintained in any Cd treatment the number of its constitutively wider blade leaves, while the number of I-214 leaves (with lower size) was reduced. H2O2 increased 4.5- and 13-fold in I-214 leaves after the lowest (L) and highest (H) Cd treatments, respectively, revealing the induction of oxidative burst. NO, constitutively higher in I-214 than Eridano, progressively increased in both clones with the enhancement of Cd concentration in the substrate. I-214 showed a more elevated antioxidative capacity (GSH/GSSG) and higher photochemical efficiency of PSII (Fv/Fm) and de-epoxidation degree of xantophylls-cycle (DEPS). The glutathione pool was not affected by Cd treatment in both clones, while non-protein thiols and phytochelatins were reduced at L Cd treatment in I-214. Overall, these two clones presented high adaptability to Cd stress and are both suitable to develop and growth in environments contaminated with this metal, thus being promising for their potential use in phytoremediation programmes.  相似文献

The photosynthetic response was studied in two clones ( Populus deltoides × maximowiczii Eridano and Populus × euramericana I‐214), known for their differential response to ozone (O3) in terms of visible symptoms, when exposed to O3 (60 nl l−1 5 h day−1, 7 and 15 days). The photosynthetic ability was tested using gas exchange and chlorophyll fluorescence analysis. O3 caused a decrease in the CO2 assimilation rate at light saturation level in mature leaves of both clones. Alterations of Chl fluorescence parameters, in particular the Fv/Fm ratio and non‐photochemical quenching were also observed. The effects were similar for both clones and it could not be concluded that differential effects on electron transport capacity were responsible for the observed reduction in photosynthesis. The reduction of photosynthetic rate in Eridano was due mainly to a reduced mesophyll activity, as evidenced by the increase in intercellular CO2 concentration and the minimal changes in stomatal conductance. In contrast, in I‐214, stomatal effects were primarily responsible, although effects on the mesophyll cannot be excluded. Data obtained indicate that the effects observed at the mesophyll level may be attributed to indirect effects caused by membrane disorders.  相似文献

Isoprene is the most important nonmethane hydrocarbon emitted by plants. The role of isoprene in the plant is not entirely understood but there is evidence that it might have a protective role against different oxidative stresses originating from heat shock and/or exposure to ozone (O(3)). Thus, plants under stress conditions might benefit by constitutively high or by higher stress-induced isoprene emission rates. In this study, measurements are presented of isoprene emission from aspen (Populus tremuloides) trees grown in the field for several years under elevated CO(2) and O(3). Two aspen clones were investigated: the O(3)-tolerant 271 and the O(3)-sensitive 42E. Isoprene emission decreased significantly both under elevated CO(2) and under elevated O(3) in the O(3)-sensitive clone, but only slightly in the O(3)-tolerant clone. This study demonstrates that long-term-adapted plants are not able to respond to O(3) stress by increasing their isoprene emission rates. However, O(3)-tolerant clones have the capacity to maintain higher amounts of isoprene emission. It is suggested that tolerance to O(3) is explained by a combination of different factors; while the reduction of O(3) uptake is likely to be the most important, the capacity to maintain higher amounts of isoprene is an important factor in strengthening this character.  相似文献

The role of ethylene (ET) signaling in the responses of two hybrid aspen (Populus tremula L. x P. tremuloides Michx.) clones to chronic ozone (O(3); 75 nL L(-1)) was investigated. The hormonal responses differed between the clones; the O(3)-sensitive clone 51 had higher ET evolution than the tolerant clone 200 during the exposure, whereas the free salicylic acid concentration in clone 200 was higher than in clone 51. The cellular redox status, measured as glutathione redox balance, did not differ between the clones suggesting that the O(3) lesions were not a result of deficient antioxidative capacity. The buildup of salicylic acid during chronic O(3) exposure might have prevented the up-regulation of ET biosynthesis in clone 200. Blocking of ET perception with 1-methylcyclopropene protected both clones from the decrease in net photosynthesis during chronic exposure to O(3). After a pretreatment with low O(3) for 9 d, an acute 1.5-fold O(3) elevation caused necrosis in the O(3)-sensitive clone 51, which increased substantially when ET perception was blocked. The results suggest that in hybrid aspen, ET signaling had a dual role depending on the severity of the stress. ET accelerated leaf senescence under low O(3), but under acute O(3) elevation, ET signaling seemed to be required for protection from necrotic cell death.  相似文献

The effects of salicylic acid (SA) on ethylene biosynthesis in detached rice leaves were investigated. SA at pH 3.5 effectively inhibited ethylene production within 2 h of its application. It inhibited the conversion of ACC to ethylene, but did not affect the levels of ACC and conjugated ACC. Summary Results of experiments with four poplar clones and various chemical fertilizers in a nursery in southern Greece are presented. At the end of the first growth period the heights of the four clones, without fertilizers, decreased in the order of I-214>I-262>cv. campeator > black poplar 1/64 with significant differences only between black poplar 1/64 and the rest of the clones.Of the fertilizer nutrients N, P, K and Mg only N improved heights of all clones significantly and especially of the clone I-214. One hundred and 200 kg of P fertilizer per ha had minimal or negative effect on height increase of all clones.Ammonium sulfate, ammonium nitrate and potassium nitrate all at 400 kg N per ha were found equally effective in improving height growth of the clone I-214 but ammonium nitrate is the N fertilizer of choice by its higher N content and relatively lower price.Ammonium nitrate at 200 kg N per ha, in two or three equal dosages, during the first growth period, June–July, gave the maximum height increase for two consecutive years of the clone I-214. Six hundred kgs, of N per ha reduced height increase of the same clone and increased losses of N, as NO3 , in drainage water.  相似文献

以美洲黑杨与大青杨的27个杂种无性系1年生盆栽扦插苗为材料,以美洲黑杨无性系I-69为对照,对其生长性状、扦插成活率以及主要光合生理指标进行遗传变异分析,并利用分析结果对杂种无性系进行初步选择.结果表明:(1)27个杂种无性系间苗期生长性状及光合生理等12个性状指标差异极显著,无性系重复力均在0.7以上,最高达0.987,均受强遗传控制.(2)结合生长性状分析不同无性系的净光合速率,发现具有较高净光合速率的无性系,同样具有较大生长量和较高成活率.(3)经主成分分析和聚类分析,初选出7个生长快、成活率较高、光合作用能力较强的无性系(编号依次为135、141、153、155、165、177和193),7个无性系苗高超对照10%~33%,地径超对照11%~24%,成活率超对照30%~48%,净光合速率超对照7%~29%,且干形优美,适应性强,可作为进一步选育的材料.  相似文献

Information on plant responses to combined ozone and cadmium stresses are scarce and limited to herbaceous species. In this research, two poplar clones (I-214 and Eridano), differently sensitive to O3, were grown for 5 weeks in pots supplied with 0, 53.5, and 160.5 mg(Cd) kg?1 (soil d.m.) and then exposed to 15-d O3 fumigation (0.06 mm3 dm?3, 5 h a day). The effects of the two stressors, alone or in combination, on Cd, Ca, Fe, and Zn accumulation in above-nad below-ground organs, photosynthesis, leaf pigments, and accumulation of H2O2 and NO were investigated. Cadmium induced a reduction in stomatal conductance and a significant accumulation of H2O2 and NO in both clones nad negatively affected the carotenoid content in I-214. Ozone, on the other hand, counteracted Cd accumulation in the above-ground organs and significantly increased the xanthophyll de-epoxidation state indicating photoinhibition in O3-treated plants. Surprisingly, O3 alone or in combination with Cd decreased H2O2 accumulation in I-214. The NO production was generally stimulated by Cd, whereas it decreased following O3 exposure in I-214. The overall data indicate that Cd and O3 induced clone specific responses. Moreover, when they were applied in combination, antagonistic rather than synergistic effects were observed.  相似文献

The poplar-and-willow borer, Cryptorhynchus lapathi (L.) (Coleoptera: Curculionidae), is a wood-boring pest of economic importance in irrigated hybrid poplar (Populus spp.) farms in eastern Washington and Oregon. There is no practical insecticide control tactic against either the larval or adult stage of C. lapathi. To assess variability in C. lapathi toward clone preference, we initiated a no-choice study on 180 caged trees that consisted of five clones in a randomized complete block design. C. lapathi was significantly more successful at establishing a population in two clones with Populus trichocarpa X P. deltoides (TxD) parentage (female x male) than in either of two clones with P. deltoides x P. nigra (DxN) parentage (female x male), or a single clone with P. deltoides x P. maximowiczii (DxM) parentage (female X male). There were no significant differences in the rate of weevil development among infested clones, with the exception of DxM trees. Larvae in DxM clones developed on average to the fourth size grouping and those in the two TxD clones developed on average to the fifth size grouping, and this difference was significant. These results corroborate our general damage surveys conducted in the field. Our findings provide growers with the option to choose less susceptible varieties when replanting.  相似文献

Ethylene production was severely inhibited in climacteric applefruits treated with either low O2(O2 :1–3%, CO2: 0%)or high CO2(O2:15–21%, CO2:10–20%) for 4, 10, and 15 days, respectively. In treatment with low O2 for 10 or 15days, 1-aminocyclopropane-1-carboxylic acid (ACC) was accumulated in large quantity,but in the fruit treated with high CO2, the content of ACC was slightly lower than thatin control fruit. When the fruit was turned to air after 4-days treatment with low O2, ethylene production and ACC content were able to recover to control level. But in the fruits withlow O2 for 10 or 15 days ethylene production was 100 times lower, and ACC contentwas much higher than those in control even 30-35 days after they were turned to air. It appears that inhibitory effect of the low O2 treatment for longer than 10 days on theconversion of ACC to ethylene was irreversible. In the case of high CO2 treatment fordifferent periods ethylene production obviously decreased at first, then followed by arapid increase. When the fruits were turned to air ethylene production in the treat-ment for 4 days was able to recover to control level, but it remained much lower in tre-atment for 10 or 15 days than that in control throughout the period of recovery. However, ACC contents in both treatments were slightly lower than those in control during a few days after the removal of treatment, but increased slightly after 2-3 weeks.Furthemore, the ability of flesh discs to convert exogenous ACC to ethylene was redu-ced remarkably by the treatment of fruits with either low O2 or high CO2 for longerthan 10 days.  相似文献

Ethylene biosynthesis in leaf discs of tobacco ( Nicotiana tabacum L. cv. Xanthi), as measured by the conversion of L-[3,4-14C]-methionine to 14C2H4, was markedly inhibited by exogenous ethylene. This inhibition was accompanied by a decrease in total (free + conjugated) content of 1-aminocyclopropane-1-carboxylic acid (ACC), most of which appeared in its conjugated inactive form. The autoinhibitory effect of ethylene was reversible and could be relieved by Ag+. The Ag+-treated leaf discs, with or without ethylene, contained only free ACC at an increased level. The results suggest that in tobacco leaves, the autoinhibition of ethylene production resulted from reduction in the availability of free ACC, through both suppression of ACC formation and increased ACC conjugation.  相似文献

The successful adoption of water recycling strategies in many arid regions will require crops able to tolerate poor-quality waters. We evaluated different clones for salt and boron (B) tolerance within each of seven genetically distinct genomic groups (e.g., deltoides, deltoides x nigra, trichocarpa x deltoides, trichocarpa x deltoides x maximowizcii, trichocarpa x deltoides x nigra, trichocarpa x nigra, trichocarpa x maximowizcii). During each evaluation period, different clones within each of the groups were irrigated with high sodium chloride (NaCl) salinity (i.e., 10-30 dS m(-1)) and B (i.e., 10 mg L(-1)) water up to a maximum of 150 days, for a 4-year testing period under micro-field plot conditions. Excessive accumulation (up to 6%) of chloride (Cl) likely caused toxicity symptoms (necrosis of the leaves) observed in the less tolerant clones, while leaf B concentrations rarely exceeded 300 mg kg(-1) DM in any clone. Increased soil salinity likely hindered the uptake of B by the clones. Our results show that a wide range of selected Populus clones, of parentage trichocarpa x nigra, followed by deltoides x nigra show potential salt and B tolerance as young trees to recycled waters high in salinity and B.  相似文献
