Mutations controlling the resistance to 6-mercaptopurine (6-M) and the ability to multiply in a medium with a low concentration of glucose (“glucose-independent” mutants) were induced in cultured Chinese hamster cells by N-nitrosomethylurea (NMU), 5-bromodeoxyuridine (BUdR), UV and X-rays. The chemical agents were found to be very active in induction of mutations to 6-M resistance (NMU and BUdR) and mutations of “glucose independence” (NMU). These agents increase the yield of mutations as compared to the spontaneous mutation rate by about two orders of magnitude. The induced rate of 6-M-resistant mutations by X-rays was 2.0 ? 10−7 per viable cell per roentgen. BUdR approximately equally increases the cell's sensitivity to both inactivating and mutagenic action of X-rays. The maximum induction of mutations to 6-M resistance by UV was observed at 100 erg/mm2. This dose leads to 1 16-fold increase of the mutation frequency as compared to the spontaneous rate. Further increase of the UV dose up to 200 erg/mm2 resulted in a lower yield of mutations per dose unit. The highest yield of mutations to 6-M resistance induced by NMU, BUdR and X-rays was observed if cells were plated in selective medium several generations after the mutagenic treatment. The maximum yield of mutations to 6-M resistance induced by UV and of glucose-independence induced by NMU was recorded if cells were transferred to selective media immediately after treatment. The kinetics of expression of mutations and the decline of their number observed after prolonged incubation of treated cells in nonselective conditions are discussed.  相似文献

The Na+ and K+ content of non-metabolizing yeast cells was determined before and after monochromatic ultraviolet (UV) irradiation. UV facilitated the uptake of Na+ into and the loss of K+ from the cells (net ion flux); the effect is greatest for the shortest wavelength employed (239 mµ) and is partly dependent upon the presence of oxygen. The UV effect on net ion flux persists for at least 90 minutes during which tests were made and it occurs following dosages which are without measurable effect on colony formation. The UV effect on net ion flux is decreased by acidity and promoted by alkalinity. Addition of calcium ions in sufficient amount prevents the usual net ion flux changes observed in irradiated yeast. Increase in concentration gradient between the inside and the outside of the cell increases the net ion flux of irradiated yeast, Na+ uptake leading K+ loss in all cases. UV appears to act by disorganizing the constituents of the cell surface, permitting K+ to leave the cell in exchange for Na+. At low intensities of UV this ionic exchange approaches equivalence, but at higher intensities more Na+ is taken up than K+ is lost. Some evidence suggests that the Na+ in excess over that exchanged for K+ is adsorbed to charged groups produced by the photochemical effect of UV on the cell surface.  相似文献

Ultraviolet (UV) A signals (320–400 nm) are important in mate choice in numerous species. The sensitivity for UV signals is not only assumed to be costly, but also expected to be a function of the prevailing ecological conditions. Generally, those signals are favored by selection that efficiently reach the receiver. A decisive factor for color signaling is the lighting environment, especially in aquatic habitats, as the visibility of signals, and thus costs and benefits, are instantaneously influenced by it. Although ecological aspects of color signal evolution are relatively well-studied, there is little data on specific effects of environmental UV-light conditions on signaling at these shorter wavelengths. We studied wild-caught gravid female 3-spined sticklebacks Gasterosteus aculeatus of 2 photic habitat types (tea-stained and clear-water lakes), possessing great variation in their UV transmission. In 2 treatments, tea-stained and clear-water, preferences for males viewed under UV-present (UV+) and UV-absent (UV–) conditions were tested. A preference for males under UV+ conditions was found for females from both habitat types, thus stressing the significance of UV signals in stickleback’s mate choice decisions. However, females from both habitat types showed the most pronounced preferences for males under UV+ conditions under clear-water test conditions. Moreover, reflectance measurements revealed that the carotenoid-based orange-red breeding coloration in wild-caught males of both habitat types differed significantly in color intensity (higher in clear-water males) and hue (more red shifted in clear-water males) while no significant differences in UV coloration were found. The differential reflection patterns in longer wavelengths suggest that sticklebacks of both habitat types have adapted to the respective water conditions. Adaptations of UV signals in a sexual context to ambient light conditions in both behavior and coloration seem less evident.  相似文献

Summary Post-irradiation DNA degradation in P. mirabilis rec + strains after UV irradiation is found to be more extensive in starvation buffer than in growth medium. In growth medium restriction of protein synthesis, but not DNA synthesis, largely prevents the expression of breakdown limitation. By the addition of chloramphenicol during post-irradiation incubation in growth medium the expression of break-down limitation was followed and found to occur 20 to 40 min after UV irradiation. Pre-irradiation by a low dose of UV leads after a corresponding time of post-irradiation incubation to breakdown limitation even in starvation buffer after a second UV exposure.Post-irradiation DNA degradation is presumed to be initiated at the sites of DNA lesions which arise at replication points damaged by UV. While pre-starvation restricts the efficiency of postirradiation DNA degradation by the reduction of the number of replication points active at the time of irradiation, caffeine as well as 2,4-dinitrophenol inhibit DNA degradation even in rec - cells probably by the interference with nicking or exonucleoltytic events initiated at those sites in the absence of breakdown limitation.Breakdown limitation is postulated to be due to inducible derepression of REC-functions which lead to the protection and, probably, repair of DNA lesions arising at the replication points following UV exposure.  相似文献

The human cell line IFr is a variant with an increased resistance to cell proliferation inhibition (CPI) by human interferon (HuIFN)-α, established from RSa cells with unusually high-sensitivity to CPI. IFr cells were later found to have increased resistance to the cell-killing effects of far-ultraviolet (UV) irradiation. Here, in cell lysates extracted from UV-irradiated IFr cells but not in those from irradiated RSa cells, fibrinolytic protease activity was found to be elevated promptly and transiently after irradiation. Treatment of IFr cells with HuIFN-α alone also resulted in the elevation of protease activity, but not that of RSa cells. Both the activity elevated after UV irradiation and after HuIFN-α treatment was inhibited to the greatest extent by antipain in vitro. Moreover, the refractoriness of IFr cells to UV cell-killing and to HuIFN-α CPI was suppressed by culturing with medium containing antipain immediately after UV irradiation or during HuIFN-α exposure. In similarly treated RSa cells, there was no modulation of UV- or HuIFN-α-susceptibility. These comparative characteristics between the two cell lines suggested that antipain-sensitive proteases and/or cellular functions may be involved in increased resistance to UV and HuIFN-α of IFr cells. © 1996 Wiley-Liss, Inc.  相似文献

Abstract Modification of the ‘intracellular concentration of reduced glutathione’ (IC-GSH) affected the response of cultured rose cells (Rosa damascena) to ultraviolet radiation (UV)-induced leakage of K+. High IC-GSH induced by incubation of cells in 10 mol m?3 GSH (IC-GSH increased linearly with time from 20 to about 600 μmol g?1 in 61.2 ks) caused cells to become significantly less sensitive to UV. Low IC-GSH induced by treatment with 1 mol m?3 buthionine sulphoximine (BSO) plus 1 mol m?3 diethylmaleate (DEM) (IC-GSH decreased from 20 to about 3 μg g?1 in 61.2 ks) reduced, rather than increased, the UV-sensitivity of the cells. However, treatment with DEM also induced a large transient K+ leakage; and treatment with BSO induced a slight leakage. The K+ leaked was recovered by 3.24 ks. Following K+ recovery, the DEM-treated cells showed almost complete insensitivity to UV, and BSO-treated cells showed a slightly reduced sensitivity to UV. These results are in agreement with our previous findings that other treatments (heat, cycloheximide, UV), which also cause a transient leakage of K+, also reduce the induction of K+ leakage by a subsequent UV treatment. We conclude that high IC-GSH may play a role in protecting plant cells from UV-induced K+ leakage. Increased UV-sensitivity with low ICGSH was not observed, we believe, because of the transient K+ leakage, though the mechanism of reduced sensitivity to UV induced by transient leakage of K+ is not known at this time. Treatment with UV did not reduce the IC-GSH, showing that this is not the mechanism by which UV induces K+ leakage.  相似文献

In the course of an experiment involving UV irradiations of polar germ cells in eggs, an interesting mutant was obtained. The male in which it was observed was heterozygous for six chromosome III genes. A test-cross of this male to the stock homozygous for the six chromosome III genes produced progeny among which there was a large deficit of wild parental class and the presence of crossovers was observed.  相似文献

Among polar bears (Ursus maritimus), fitness is dependent on body size through males’ abilities to win mates, females’ abilities to provide for their young and all bears’ abilities to survive increasingly longer fasting periods caused by climate change. In the Western Hudson Bay subpopulation (near Churchill, Manitoba, Canada), polar bears have declined in body size and condition, but nothing is known about the genetic underpinnings of body size variation, which may be subject to natural selection. Here, we combine a 4449‐individual pedigree and an array of 5,433 single nucleotide polymorphisms (SNPs) to provide the first quantitative genetic study of polar bears. We used animal models to estimate heritability (h2) among polar bears handled between 1966 and 2011, obtaining h2 estimates of 0.34–0.48 for strictly skeletal traits and 0.18 for axillary girth (which is also dependent on fatness). We genotyped 859 individuals with the SNP array to test for marker–trait association and combined p‐values over genetic pathways using gene‐set analysis. Variation in all traits appeared to be polygenic, but we detected one region of moderately large effect size in body length near a putative noncoding RNA in an unannotated region of the genome. Gene‐set analysis suggested that variation in body length was associated with genes in the regulatory cascade of cyclin expression, which has previously been associated with body size in mice. A greater understanding of the genetic architecture of body size variation will be valuable in understanding the potential for adaptation in polar bear populations challenged by climate change.  相似文献

Sexually selected ornaments and signals are costly to maintain if they are maladaptive in nonreproductive contexts. The jumping spider Cosmophasis umbratica exhibits distinct sexual dichromatism with males displaying elaborate UV body markings that signal male quality. Female C. umbratica respond favorably to UV‐reflecting males and ignore males that have their UV masked. However, Portia labiata, a UV‐sensitive spider‐eating specialist and a natural predator of C. umbratica, is known to use UV reflectance as a cue when hunting prey. We investigated the cost of these UV signals in C. umbratica in terms of their predation risk. Under experimental conditions, three choice scenarios were presented to P. labiata individuals. Choices by P. labiata were made between male C. umbratica with and without the UV signal; a UV‐reflecting male and non‐UV‐reflecting female; and a UV‐masked male and female. The presence and absence of UV signals was manipulated using an optical filter. Portia labiata exhibited a strong bias toward UV+ individuals. These results suggest the sexually selected trait of UV reflectance increases the visibility of males to UV‐sensitive predators. The extent of this male‐specific UV signal then is potentially moderated by predation pressure. Interestingly though, P. labiata still preferred males to females irrespective of whether UV reflectance was present or not. This suggests P. labiata can switch cues when conditions to detect UV reflectance are not optimal.  相似文献

UV-irradiation induces an exponential increase in the frequency of mutation from asparagine requirement to asparagine non-requirement in Jensen sarcoma cells grown in vitro. The corrected mutation frequency increases from the spontaneous rate of 5.1·10?6 per cell to 1248·10?6 per cell with a dose of 180 erg/mm2 of 254 nm UV A substantial increase was oberved even without correction for survivors, and no significant difference was observed in the UV sensitivity of asparagine-requiring and non-requiring Jensen clones. When Jensen cells were plated at low densities in a feeder layer of LMTK-cells inactivated by HAT medium, an increase in the cloning ability of the former was observed as compared to appropriate controls without the feeder layer, but the increase was constant over all doses of UV tested. Revertants are stable and possess measurable asparagine synthetase.It is concluded that UV is an extremely effective mutagen in this system.  相似文献

8-Azaguanine (8AG)-resistant mutations induced by X-rays, ultraviolet radiation (UV) and a chemical carcinogen, 4-hydroxyaminoquinoline 1-oxide (4-HAQO) were examined during the cell cycle of synchronized HeLa S3 cells. Mutants induced by 400 R of X-rays occurred in a higher frequency in the X-ray sensitive G1-S boundary phase than in the X-ray-resistant G2, S and early g2 phases. 8AG-resistant mutants induced by treatment with 10?5 M 4-HAQO for 20 min appeared in a higher frequency in the early to middle S phases than in the other phases. In the case of UV, however, we found no significant difference in the induced mutation frequencies the cell cyle, because the mutation frequencies induced by the UV doses (0–20 Jm2) used were too low for detection of the difference. These results suggest that there is a close correlation between the critical damage induced in DNA molecule(s) at the DNA-synthetic phase in the cell cycle and mutagenesis, because mitotic cells have a low mutability in spite of their high radio-sensitivity.  相似文献

UV‐induced synthesis/accumulation of photoprotective pigments and antioxidant activity were investigated in the hot‐spring cyanobacterium Leptolyngbya cf. fragilis. The results indicated that UV radiation may induce biosynthesis of carotenoids, allophycocyanin, phycoerythrin, and scytonemin while phycocyanin degrades in response to longtime UV radiation. Moreover, pigment composition of L. cf. fragilis was significantly altered with increasing UV radiation times, probably due to destruction and resynthesis of accessory pigments as an adaptation strategy to UV stress. The in vitro antioxidant analysis of different extracts of UV treated cyanobacteria exhibited concentration‐dependent antioxidant activity. Ethyl acetate extract of 72 h UV treatment showed maximum total antioxidant activity (IC50 = 71.73 ± 5.3 μg mL?1) followed by ethyl acetate control (non‐UV irradiated) extract (IC50 = 109.43 ± 2.76 μg mL?1). This is the first report for the UV‐induced synthesis of photoprotective pigments and their antioxidant activity in L. cf. fragilis.  相似文献

The blue throat feathers of male bluethroats (Luscinia s. svecica) show a reflectance peak in the ultraviolet (UV) waveband (320 to 400 nm). The throat is actively displayed during courtship, suggesting a role for sexual selection on an ultraviolet signal. Indeed, a recent aviary experiment demonstrated that females discriminated against males with artificially reduced UV reflectance (Andersson and Amundsen 1997). Here, we report the results of a similar experimental manipulation applied on free-ranging males. UV-reduced (UVR) males had a lower success in attracting mates, as judged from a significantly later start of egg laying, compared with control (C) males. UVR males also spent significantly less time advertising for additional mates when their own mate was fertile, and they had a lower success in achieving extra-pair fertilizations. Furthermore, UVR males tended to guard their mates more closely and lose more paternity in their own brood than C males did. We conclude that the treatment affected both social and extra-pair mate choice. This is the first experimental evidence that UV signalling influences male mating success in free-ranging birds.  相似文献

Catalase zymograms performed on hemolysates of horse red cells revealed three phenotypes each composed of a single zone of activity designated F, M and S in decreasing order of migration towards the anode. It is proposed that these phenotypes are controlled by a pair of alleles, CatF and CatS, such that the genotypes CatFCatF, CatFCatS and CatSCatS give rise respectively to phenotypes F, M and S. Earlier studies have shown that Cosmophasis umbratica, a jumpingspider from Singapore, is sexually dimorphic in the reflectanceof ultraviolet (UV) (males, but not females, have UV-reflectingmarkings). Here we present the first experimental evidence thatthe male's UV markings influence mate choice of C. umbraticafemales. When presented with males whose appearance was manipulatedby the use of a UV-blocking filter, females spent more timewatching UV+ males (i.e., males with UV present) and less timewatching UV– males (UV absent). We also manipulated thelevels of male brightness by using 2 UV-transmitting neutraldensity filters and showed that UV reflectance was used specificallyfor hue discrimination instead of being used for detecting differencesin brightness alone. This is not only the first strong evidenceof UV influence on female mate-choice decisions for a spiderbut also the best experimental demonstration of color vision,whatever the wavelength.  相似文献

The ability of posttreatment exposure to non-toxic concentrations of thymidine (TdR) to enhance the lethal effects of a number of alkylating agents, X-rays and UV and the lethal and mutagenic effects of N′-ethyl-N-nitrosourea (ENU) and N-methyl-N-nitrosourea (MNU) has been examined in V79 cell lines. TdR posttreatment enhanced the cytotoxic effects of ethyl methanesulphonate (EMS), MNU and ENU but not of UV or X-rays and increased both the spontaneous and MNU- and ENU-induced frequencies of azaguanine resistant (AZR) mutants. No significant effect of TdR on the spontaneous frequency of thioguanine resistant (TGR) mutants was demonstrated but the frequency of MNU-induced mutants to TGR was enhanced. The effects on expression of both potentially lethal and premutagenic damage were reversed by addition of an equimolar concentration of deoxycytidine (dCdR). The enhancement in spontaneous and induced mutant frequency (IMF) at the HGPRT locus appears to be due to an alteration in the selective efficiency of purine analogous due to alteration in growth kinetics of cells exposed to TdR or treated with alkylated agents or posttreated with thymidine after alkylation damage and not to an alteration in the miscoding potential of alkylated bases.  相似文献

Some fish, including the guppy, have the ability to perceive ultraviolet (UV) wavelengths. Female guppies prefer to associate with males that are viewed under light conditions that include UV-A, in preference to conditions lacking these wavelengths. We used reflexion spectrophotometry to show that male guppies reflect UV light from both their structural (purple, green and white) and pigment (orange) colour patches and that males differ in the levels of UV light reflected. Varying components of UV may affect both the brightness and hue of particular colour patches. This may produce nonspectral colours that are visible to the guppy but that are outside human perception. We used video analysis to quantify male reflexion in the UV and visible wavebands. Male guppies with high and low UV reflexion, but similar human-visible coloration and area of coloration, were paired for use in mate choice experiments. Female guppies shown pairs of males differing in their levels of UV reflexion had no preference for either high or low UV reflexion. This suggests that UV reflexion does not provide particularly significant information relating to male quality or influence female preference in this population of guppies. A technique for transplantation of cytoplasm between Drosophila eggs is described. Polar cytoplasm of newly laid eggs was first made ineffective for the determination of germ cells by UV irradiation. The sterility which results from this UV irradiation could be prevented by the injection of polar cytoplasm, but not by the injection of anterior cytoplasm from unirradiated donor eggs. The results provide the first demonstration of transplantation of agents causing determination in an insect and also provide a bioassay for these agents.Microscopical observations of UV irradiated eggs with and without transplanted polar cytoplasm revealed that UV irradiation delays the migration of cleavage nuclei into the posterior periplasm and prevents cytoplasmic protrusions at the posterior pole from becoming isolated from the periplasm to form pole cells. Transplantation of polar cytoplasm repairs these defects.  相似文献
