Spawning Atlantic salmon, Salmo salar (L.), brown trout, Salmo trutta (L.) and rainbow trout, Salmo gairdneri (Richardson) were held in tanks for four weeks and then injected with filtrates from lesions of salmon naturally infected with UDN. In some cases prednisolone acetate was included in the inoculum. Water temperatures were recorded daily and the time of appearance of the lesions was noted. Passage experiments using extracts of these lesions were carried out.  相似文献

The commonly applied classification systems of fish gonad maturity divide the maturation process into certain stages. However, the scales do not entirely reflect the continuity of the maturation process. Based on light microscope observations, the paper describes a comprehensive pattern of testicular transformations during maturation. The study was carried out on precocious underyearling and 1-year-old males of sea trout (Salmo trutta m. trutta L.), 1-year-old males of salmon (Salmo salar L.), and males of brown trout (Salmo trutta m. fario L.) aged from 7 months to 4 years. A total of 821 gonads collected during all seasons of the year were examined. The fish were fixed in Bouin's fluid. Histological slides of the mid-part of the gonad were made using the standard paraffin technique. The 3-6 microm sections were stained with Heidenhain haematoxylin. Histological changes of testes during maturation were similar in the three species studied. Immature and resting gonads contained type A spermatogonia in lobules only. The appearance of cystic structures containing type B spermatogonia in the lobules signalled the beginning of the sexual cycle in male gonads. Type B spermatogonia underwent synchronous mitotic divisions resulting in an increase in the total number of spermatogonia. As the spermatogenesis continued, the gonads showed a gradual increase in the number of cysts containing cells at all the spermatogenetic stages: type B spermatogonia, primary and secondary spermatocytes, spermatids, and spermatozoa. The well-formed spermatozoa were released to the lobule lumen once the Sertoli cells and spermatozoa connections broke up and the cyst disappeared. This was a continuous process observed throughout the spawning season. The spermatozoa were moved to the efferent duct. While some of the germ cells were completing spermatogenesis, the lobules contained less and less cysts with type B spermatogonia, primary and secondary spermatocytes, and spermatids; eventually all the cells completed spermatogenesis. At the end of maturation, vacuoles, up to 18.9 microm in final diameter (brown trout), appeared in the Sertoli cells. The vacuoles were visible in the lobule wall epithelium for a prolonged period of time. In most salmonid individuals examined, the reproductive cycles were observed to overlap. In some fish, the preparation for another cycle began very early, i.e., at the and of preceding spermatogenesis, which had not been observed before. The diel rhythm of emergence from the gravel and displacement downstream has been studied in three salmonoid species: brown trout ( Salmo trutta L.), Atlantic salmon ( S. saiar L.) and grayling ( Thymallus thymallus L.). Grayling emerged in the morning but delayed downstream displacement until the night, while the Salmo species emerged just before downstream displacement chiefly at night.  相似文献

By means of electron microscopy, we studied the development of pollen grain walls in Calendula officinalis L., Dimorphotheca aurantiaca DC., and Cichorium intybus L. (Asteraceae). As a reference, we studied the plants from the families Schisandraceae (Schisandra chinensis (Turcz.) Baill.), Lauraceae (Persea americana Mill.), Boraginaceae (Borago officinalis L.), and Cycadaceae (Encephalartos altensteinii Lehm.). In Asteraceae, we revealed two successively initiated layers of glycocalyx that form outer and inner layers of the ectexine. The formation of endexine is contributed by plasma membrane and small vesicles. Glycocalyx in the plants from the families Schisandraceae, Lauraceae, Boraginaceae, and Cycadaceae was found to consist of radially arranged helical cylindrical units, which are receptors of sporopollenin deposition. It is assumed that the receptor-independent accumulation of sporopollenin is also possible.  相似文献

Species–environment relationships were studied between the occurrence of 13 fish and lamprey species and 9 mainly map‐based environmental variables of Finnish boreal small streams. A self‐organizing map (SOM) analysis showed strong relationships between the fish species and environmental variables in a single model (explained variance 55.9%). Besides basic environmental variables such as altitude, catchment size, and mean temperature, land cover variables were also explored. A logistic regression analysis indicated that the occurrence probability of brown trout, Salmo trutta L., decreased with an increasing percentage of peatland ditch drainage in the upper catchment. Ninespine stickleback, Pungitius pungitius (L.), and three‐spined stickleback, Gasterosteus aculeatus L., seemed to benefit from urban areas in the upper catchment. Discovered relationships between fish species occurrence and land‐use attributes are encouraging for the development of fish‐based bioassessment for small streams. The presented ordination of the fish species in the mean temperature gradient will help in predicting fish community responses to climate change.  相似文献

Eleven bacterial strains were isolated from the gastrointestinal tract of four fish species, Atlantic salmon (Salmo salar L.), Arctic charr (Salvelinus alpinus L.), Atlantic cod (Gadus morhua L.) and wolffish (Anarhichas lupus L.). All the strains were Gram-positive rods, non-sporing, catalase and oxidase-negative, able to grow at pH 9.0 but not on acetate containing media (pH < or = 5.4), and were fermentative. They had a high content of oleic acid (18:1 n-9) in cellular lipid, and were found to belong to the genus Carnobacterium by phenotypic criteria. The eleven carnobacteria strains were further identified on the basis of 16S rDNA sequence analysis and AFLP(TM) fingerprinting.  相似文献

A putative reservoir of functional plasma membrane proteins, the secretory vesicle identified by latent alkaline phosphatase and tetranectin, has previously been demonstrated based on indirect evidence (Borregaard, N., Miller, L. J., and Springer, T. A. (1987) Science 237, 1204-1206; Borregaard, N., Christensen, L., Bjerrum, O. W., Birgens, H. S., and Clemmesen, I. (1990) J. Clin. Invest. 85, 408-416). Difficulties in separating plasma membranes from this entity by density gradient centrifugation has prohibited discriminative dynamic and quantitative studies of secretory vesicles and plasma membranes. By combining density centrifugation with free flow electrophoresis we overcame this obstacle. Freshly prepared unperturbed human neutrophils were subjected to nitrogen cavitation followed by density centrifugation on Percoll gradients. Light membrane fractions containing plasma membranes and secretory vesicles were applied to high voltage free flow electrophoresis on an Elphor VaP 22. Plasma membrane vesicles, identified by HLA class I antigen mixed enzyme-linked immunosorbent assay (Bjerrum, O. W., and Borregaard, N. (1990) Scand. J. Immunol. 31, 305-313) and 125I applied to cells before cavitation, were clearly separated from secretory vesicles. Electron microscopy revealed a morphology typical of plasma membranes in the former fraction and a population of vesicles with markedly different appearance in the latter. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis profiles demonstrated distinct differences in protein patterns between the two fractions. Superoxide generating capacity induced by sodium dodecyl sulfate and cytosol, an entity traditionally ascribed to the plasma membrane, was largely confined to fractions containing secretory vesicles. Thus, the majority of membrane-bound NADPH oxidase components of light membranes of human neutrophils colocalize with secretory vesicles.  相似文献

The resistance to low tensions of dissolved oxygen over periods ranging from less than a day to a week was determined for rainbow trout ( Salmo gairdnerii Richardson), perch ( Perca fluviatilis L.), roach ( Rutilus rutilus (L.)), and mirror carp, a domestic form of the common carp ( Cyprinus carpio L.) at 10°, 16° and 20°C., tench ( Tinea tinea (L.)) at 10° and 16°C., dace ( Leuciscus leuciscus (L.)) at 16° and 20°C., chub ( Squalius cephalus (L.)) at 20°C., and bleak ( Alburnus alburnus (L.)) at 16°C.
At each temperature period of survival decreased with fall in oxygen tension.
Rise in temperature between 10° and 20°C. reduced resistance to lack of oxygen of all species except rainbow trout, in which there was no significant difference between resistance at 16° and 20°C. Rise in temperature between 10° and 16°C. did, however, lower resistance considerably.
Tench were the most resistant to lack of oxygen and, in general, trout were the most susceptible. At 20°C. mirror carp required a higher tension of oxygen to enable them to survive for a week than did rainbow trout, but for shorter periods could withstand much lower tensions than rainbow trout; at 10° and 16°C. their resistance was intermediate between that of tench and rainbow trout.  相似文献

A study was conducted to compare astaxanthin binding ability of solubilized muscle proteins of Atlantic salmon (Salmo salar L.), haddock (Melanogrammus aeglefinus L.) and Atlantic halibut (Hippoglossus hippoglossus L.). Muscle proteins of juvenile Atlantic salmon, haddock and halibut were solubilized by sequential extraction of muscle tissue using low ionic strength solutions. Electrophoretic protein profiles of the six solubilized fractions from these species were similar. Each solubilized fraction from the three species was examined for its relative astaxanthin binding capacity. The amount of bound astaxanthin was significantly different (P < 0.05) among the six fractions of each species. Significant differences in astaxanthin binding were only found for fractions A and E among the species. The amount of bound astaxanthin in various fractions of each species showed a good correlation (R2 = 0.80–0.92) with the ANS (8-anilino-1-naphthalenesulfonate) fluorescence intensity of those fractions. The pattern and extent of astaxanthin binding to the muscle proteins of juvenile salmon, haddock and halibut is comparable to that reported previously for adult Atlantic salmon [Saha, M.R., Ross, N.W., Gill, T.A., Olsen, R.E., Lall, S.P., 2005. Development of a method to assess binding of astaxanthin to Atlantic salmon S. salar L. muscle proteins. Aquacult. Res. 36, 336–343.]. These combined observations suggest that the carotenoid binding capacity of the muscle proteins of salmon is not the limiting factor in the deposition of carotenoid in their flesh.  相似文献

Experiments show that the first of a series of replicate electric fishings can cause a decrease in catchability so that the second and subsequent catches are made from what is, effectively, a reduced population. Depletion (DeLury) estimates can be thus seriously low. Species affected are roach Rutilus rutilus (L.), gudgeon Gobio gobio (L.), rudd Scardinius erythophthalmus (L.) and, from other published data, probably rainbow trout Salmo gairdneri Richardson and eels Anguilla anguilla (L.). Being caught does not, apparently, cause fish to become less catchable. The reason why an electric field causes a proportion of a population to become uncatchable is obscure but it may be that refractory behaviour is involved; the effect lasts for between 3 and 24 h. A method is given for adjusting DeLury estimates for decreased catchability.  相似文献

Human KB carcinoma cells resistant to high levels of colchicine, vinblastine, vincristine, adriamycin, and actinomycin D exhibit reduced accumulation of these structurally unrelated chemotherapeutic agents (Akiyama, S.-I., Fojo, A., Hanover, J. A., Pastan, I., and Gottesman, M. M. (1985) Somatic Cell Mol. Genet. 11, 117-126; Fojo, A., Akiyama, S.-I., Gottesman, M. M., and Pastan, I. (1985) Cancer Res. 45, 3002-3007). To examine the mechanism of reduced drug accumulation in these cells, we measured [3H]vinblastine ([3H]VBL) binding to membrane vesicles made from drug-sensitive (KB-3-1), drug-resistant (KB-C4), and revertant (KB-R1) cells. Membrane vesicles from KB-C4 cells bound up to 8-fold more [3H]VBL than vesicles from the parental KB-3-1 or revertant KB-R1 cell lines. No difference in binding of [3H]dexamethasone, to which the cells are equally sensitive, was observed. The difference in [3H]VBL binding by vesicles from resistant and sensitive cells was eliminated by the addition of 10 micrograms/ml verapamil, which is known to reverse the multidrug-resistance phenotype. Drug binding by KB-C4 vesicles was osmotically insensitive, temperature-dependent, and trypsin-sensitive. Binding of [3H]VBL by KB-C4 vesicles was inhibited by vinblastine, vincristine, and daunomycin (in decreasing order). Dexamethasone at 100 microM, colchicine at 100 microM, and actinomycin D at 100 microM did not significantly inhibit [3H]VBL accumulation. No significant differences in tubulin content were detected among vesicles from sensitive and resistant cells. These data demonstrate that membrane vesicles from multiply drug-resistant cells bind increased amounts of vinblastine.  相似文献

We previously described the isolation and preliminary characterization of a Chinese hamster ovary cell mutant, termed G.7.1, that carried a temperature-sensitive, conditional-lethal lesion affecting the acidification of vesicles in crude cellular extracts (Marnell, M. H., Mathis, L. S., Stookey, M., Shia, S.-P., Stone, D. K., and Draper, R. K. (1984) J. Cell Biol. 99, 1907-1916). In the present report, we have separated lysosomal vesicles from more buoyant nonlysosomal vesicles by centrifuging cell extracts with Percoll and correlated the acidification defect with nonlysosomal vesicles, including endosomes, but not with secondary lysosomes. Moreover, the acidification of nonlysosomal vesicles prepared from mutant cells grown at the permissive temperature was more sensitive to thermal inactivation than similar vesicles from parental cells, implying that a heat-sensitive component is a normal resident of nonlysosomal vesicles in the mutant. This heat-sensitive component is apparently not associated with lysosomes, or if it is, it does not inhibit lysosomal acidification at the nonpermissive temperature. We also found that the transferrin-mediated uptake of iron is inhibited by 50% in the mutant cells at the nonpermissive temperature and that the inhibition cannot be accounted for by reduced binding or internalization of transferrin.  相似文献

Dehydrins are characterized by the consensus KIKEKLPG amino acid sequence found near the carboxy terminus, and usually repeated from one to many times within the protein. A synthetic peptide containing this consensus sequence was used to produce specific antibodies that recognize dehydrins in a wide range of plants. This range covered two families of monocots, viz. Gramineae (Hordeum vulgare L., Triticum aestivum L., Zea mays L., Oryza sativa L.) and Liliaceae (Allium sativa L.), and five families of dicots, Malvaceae (Gossypium hirsutum L.), Solanaceae (Lycopersicon esculentum L.), Brassicaceae (Raphanus sativus L.), Fabaceae (Vigna unguiculata L.), and Cucurbitaceae (Cucumis sativus L.). Two families of gymnosperms, Pinaceae (Pinus edulis Engelm.) and Ginkgoaceae (Ginkgo biloba L.), were also included. For several plants in which dehydrin cDNA and genomic clones have previously been characterized, it now appears that the dehydrin family of proteins is larger, and the regulation of dehydrin expression much more complex, than earlier studies have shown.  相似文献

We caught smolts of Arctic charr, Salvelinus alpinus (L.), Atlantic salmon, Salmo salar L., and brown trout, Salmo trutta L., in a trap situated at the mouth of the river Halselva (70° N 23° E), northern Norway during a 5-year period. Salmon and charr were the first to leave freshwater at the end of May, while most trout left freshwater about 14 days later. Whereas the midnight sun shines continuously during the downstream migration period, the light intensity has a diel intensity pattern. The majority of the descending migrants were recorded during the night. The number of descending fish was relatively low at water temperatures below 3°C. The increase in water level was largely caused by snowmelt and thus correlated with lower water temperatures. The number of migrants of all three species increased with increasing water level and decreased with increasing water temperature, with the exception of trout, which increased with water temperature. Notably, the increase in number of migrants was also correlated with the increase in water level the following day, indicating that fish movements represent an early response to a later spate. There was no significant relationship between the number of migrants and the daily change in water level or temperature. The three species were highly synchronised in their daily number of migrants. The strongest synchronisation was found between Arctic charr and Atlantic salmon, followed by Arctic charr and brown trout.  相似文献

During the periods 1956–1963 and 1967–1970 traps were operated to catch upstream- and downstream-migrating sea trout, Salmo trutta L. A total of 15 788 sea trout were tagged, using Carlin tags. The number of recaptures made in the traps was 4481, of which 1796 were recaptured more than once.
The distribution of the 2122 recaptures in the sea provides a picture of the sea-migration pattern. Of the sea recaptures, 52.8% were reported as within a distance of 3 km from the river mouth, compared to 0.7% more than 80 km away. All the different size-groups of sea trout were represented among both the long-distance and the short-distance migrants. The results of this study of sea trout migrations are discussed in relation to the published results for Atlantic salmon, Salmo salar L., and sea charr, Salvelinus alpinus (L.), from the same river.
The four highest values recorded for mean distance of daily travel away from the river were 20, 8, 8 and 6km day−1 by smolts and 6, 6, 5 and 5km day−1 by larger-sized sea trout.
Recaptures of tagged sea trout in rivers other than the Vardnes totalled 506, of which 306 had been tagged as smolts. The calculated minimum percentage of stray is 15.5%. The proportion of sea trout from the Vardnes river that actually spawn in other rivers is not known. No significant difference in length distribution was found between the sea trout caught in the Vardnes river and those caught in other rivers. An hypothesis concerning the selective advantages of straying by anadromous salmonids living in small rivers is discussed.  相似文献

Band 3, the erythrocyte anion transporter, has been shown to transfer between human erythrocytes and sonicated vesicles (Newton, A. C., Cook, S. L., and Huestis, W. H. (1983) Biochemistry 22, 6110-6117). Functional band 3 becomes associated with dimyristoylphosphatidylcholine vesicles incubated with human red blood cells. Proteolytic degradation patterns reveal that the transporter is transferred to the vesicles in native orientation. In erythrocytes, native band 3 is degraded on the exoplasmic membrane face by chymotrypsin and on the cytoplasmic surface by trypsin (Cabantchik, Z. I., and Rothstein, A. (1974) J. Membr. Biol. 15, 227-248; Jennings, M. L., Anderson, M. P., and Monaghan, R. (1986) J. Biol. Chem. 261, 9002-9010). Band 3 in intact protein-vesicle complexes is degraded by exogenous chymotrypsin but not by trypsin. In contrast, trypsin entrapped in the lumen of the vesicles proteolyses the vesicle-bound band 3 quantitatively. Band 3 remaining in the membranes of vesicle-treated cells and in cell fragments is not degraded detectably by vesicle-entrapped trypsin. These observations indicate that band 3 is unlikely to transfer between cell and vesicle membranes via a water-soluble form or to adhere nonspecifically to the vesicle surface; the aqueous contents of vesicles and cells (or membrane fragments) are not pooled during cell-vesicle incubations, hence no cell-vesicle fusion occurs; and the band 3 associated with the sonicated vesicle fraction is inserted in the vesicle bilayer in native orientation, with its cytoplasmic segment contacting the aqueous contents of the vesicle lumen.  相似文献

Otoliths of Atlantic salmon, Salmo salar L., are more slender than the otoliths of brown trout, Salmo trutta L. Discriminant analysis on otolith measurements of juvenile Atlantic salmon and brown trout from four river systems revealed a discriminant function which distinguished more than 94% of the cases. This function was tested by using data from a fifth river with cohabiting Atlantic salmon and brown trout: all Atlantic salmon and 91 % of the brown trout were correctly classified.  相似文献

Phloem parenchyma and companion cells from four species of legumes, Phaseolus vulgaris L., Melilotus alba Desr., Desmodium canadense, L. and Dolichos lablab L., were examined electron microscopically to estabish the presence, structure and development of P-protein. P-protein components consisting of granular, fibrillar, tubular or crystalline structures were found in parenchyma cells of all species and in companion cells of M. alba. The earliest stages of P-protein formation were closely associated with dictyosome cisternae, dictyosome vesicles and/or spiny vesicles. After their formation, the P-protein bodies were frequently transformed into one or more structurally different components. Although these components appeared to be develop-mentally related, their specific associations and transformations differed in each species examined.  相似文献
