The zinc ion coordination of vincristine was studied by polarography; the analogous calcium ion coordination process was studied potentiometrically by a calcium ion selective electrode. In both cases, complexes of 1:1 composition were formed. The formation constant of the calcium complex was found to be 1g K = 3.27 ± 0.1. On the basis of the substitution of zinc in its vincristine complex by calcium and magnesium ions respectively, the ratio of the corresponding stability constants could be estimated as KZn:KCa (and KZn:KMg) ∼ 105−3 × 104. The complex formation processes proved to be pH-independent in the pH range 3.4–5.5, indicating that the metal ions are coordinated by the unprotonated oxygen donor atoms of vincristine.  相似文献

1. The properties of 3,5-di-tert-butyl-4-hydroxybenzylidenemalononitrile (SF 6847) were studied chemically and spectroscopically. Two molecular species of SF6847 were identified: the undissociated form (SFH; ?363, 10 mM?1) and the dissociated form (SF?; ?454, 35 mM?1). The pKa value of the molecule was determined to be 6.9.2. On the basis of these properties the interactions of SF6847 with liposomes and valinomycin · K+ were studied. The partition constants of SFH (Knp and SF? (K?p) to liposomes were determined separately; Knp was 56 mM?1 and was independent of the pH of the medium, whereas K?p dependend greatly on the pH, being 1.2 mM?1 at pH 7.0 and 2.9 mM?1 at pH 8.0. Using these values, the partition constant of total SF6847 (Kp) was calculated and found to be essentially the same as that calculated from the kinetics of proton uptake. It was concluded that the amount of SF? bound to liposomes is rate limiting for proton uptake.3. The effects of membrane potential on partition constants were studied. The K?p decreased greatly upon generation of a membrane potential negative inside the liposomes but increased upon generation of a membrane potential positive inside the liposomes.4. The interaction of SF6847 with valinomycin in aqueous solution and in liposomes was demonstrated only in the presence of potassium ion. Potassium ion could not be replaced by sodium ion. Evidence was obtained for the formation of the ternary complex valinomycin · K+ · SF? in liposomes and in hexane. It was concluded that SF? became more soluble in the liposomal membranes on formation of this ternary complex. All these results support our proposed mechanism for the proton uptake cycle (Yamaguchi, A. and Anraku, Y. (1978) Biochim. Biophys. Acta 501, 136–149).  相似文献

The stability constants of magnesium and lithium complexes of 1,4,7,10-tetraazacyclododecane (cyclen) have been determined in 0.5 M KNO 3 at 25 °C by means of potentiometric titration, as KMgL = 1.77 × 10 2 and KLi L < 10 −2, respectively. Ab initio calculations on the protonated species of the cyclen ligand have been performed in order to obtain a better understanding of experimental protonation constants, and to compare them to previous calculations on the investigated metal complexes.  相似文献

Arsenazo III forms a 1:1 complex with calcium. The affinity constant of arsenazo III for calcium (pKCa) has been determined by titrating purified arsenazo III with standard calcium solutions. The method of evaluation used allows one to determine correct pKCa values even in the presence of micromolar amounts of contaminating calcium. The pKCa is influenced by the following factors: (a) in the neutral pH range the apparent pKCa increases strongly with pH; (b) alkali ions bind weakly to arsenazo III and millimolar concentrations cause a decrease in the apparent pKCa; (c) the magnesium affinity of arsenazo III, although much lower than the calcium affinity, increases strongly with pH in the neutral range (at pH 7.0 the calcium affinity of arsenazo III is not appreciably altered by up to 2 mm magnesium); (d) strontium and barium form weaker complexes with arsenazo III than calcium, but much stronger complexes than magnesium; (e) the apparent pKCa decreases with increasing buffer concentration in the millimolar range. The pKCa of arsenazo III is so high that, unless the arsenazo III concentration greatly exceeds the calcium concentration, a considerable fraction of the total arsenazo III is in the calcium complexed form. Because of this, arsenazo III responds nonlinearly to all but the lowest calcium concentrations; however, quantitation of the calcium concentration can readily be done from the mass action law provided that the pKCa is determined under the actual experimental conditions. Arsenazo III is a reliable calcium indicator if the experimental conditions, particularly pH, are well controlled.  相似文献

The interaction between Na+ and K+ ions and benzo-15-crown-5 and dibenzo-30-crown-10 in CH3 CNH2O mixtures has been studied spectrophotometrically, at 25 °C and at ionic strength of 0.015. The formation constants for 1:1 complexes at different weight percentages of acetonitrile in water were determined. In all solvent mixtures used, the stabilities of the complexes vary in the order DB30C10-K+ > B15C5-K+ > B15C5-Na+. A linear relationship is observed between log Kf of the three complexes and the mole fraction of acetonitrile. The solvent dependence of the stability constants is most notable with B15C5-K+ and the least with DB30C10-K+ complex.  相似文献

The synthesis of three tetrapeptides, Gly-Pro-Gly-Tyr. Gly-Pro-Tyr-Gly. and Tyr-Pro-Gly-Gly, are described. All contain proline as the second amino acid subunit to act as a break point in metal complex formation.The proton and copper(II) complex formation constants have been measured at 22°C and l = 0.10 mol dm?3 (KNO3). The copper(II) complexes have also been studied spectrophotometrically over the pH range of 6–11 by absorption spectroscopy (800–200 nm), circular dichroism spectroscopy, and electron paramagnetic resonance spectroscopy. The experimental data have been combined to determine the complex species present as a function of pH and the coordination centers used.  相似文献

Methylmercury(II) and mercury(II) complexes of imidazole (1), 1-methylimidazole (2), and the 1,3-dimethylimidazolium ion (3) have been prepared in aqueous or ethanolic solution. Elemental analysis and 1H nmr spectroscopy have been used to characterize the complexes. The MeHg (Me = methyl) binding sites have been identified as N1, N3 (1), N3, C2 (2), and C2 (3). Reaction with HgO leads to the formation of Hg-bridged complexes of the type Im-Hg-Im, (Im = imidazole), where bonding occurs through N1 (1) and C2 (3); the latter is also formed as a result of symmetrization of the C2-bound MeHg complex. The formation of the C2-bound (carbene) complexes is discussed in terms of the increased acidity of the C2 proton resulting from coordination of an electrophilic species at N3. Based on electrostatic considerations, there appears to be a “minimum degree of activation” required before C2 bonding can occur, which explains the lack of this coordination mode in 1. 199Hg-1H spin-spin coupling (4J) is observed for C-bound mercury, but not for N-bound mercury, which is interpreted in terms of a decreased ligand exchange rate in the former case, due to the greater stability of the Hg-C bond. 2J coupling constants measured in (CD3)2SO for a number of MeHg complexes of heterocyclic ligands (including the imidazoles of the present study) correlate well with the ligand pKa (25°C, aqueous solution), according to 2J = ?3.88 pKa + 248.5. Results in the present work are discussed in relation to our previous work with nucleosides. The significance of the results to biological systems is considered.  相似文献

Protein–protein interactions play central roles in physiological and pathological processes. The bases of the mechanisms of drug action are relevant to the discovery of new therapeutic targets. This work focuses on understanding the interactions in protein–protein–ligands complexes, using proteins calmodulin (CaM), human calcium/calmodulin‐dependent 3′,5′‐cyclic nucleotide phosphodiesterase 1A active human (PDE1A), and myosin light chain kinase (MLCK) and ligands αII–spectrin peptide (αII–spec), and two inhibitors of CaM (chlorpromazine (CPZ) and malbrancheamide (MBC)). The interaction was monitored with a fluorescent biosensor of CaM (hCaM M124C–mBBr). The results showed changes in the affinity of CPZ and MBC depending on the CaM–protein complex under analysis. For the Ca2+–CaM, Ca2+–CaM–PDE1A, and Ca2+–CaM–MLCK complexes, CPZ apparent dissociation constants (Kds) were 1.11, 0.28, and 0.55 μM, respectively; and for MBC Kds were 1.43, 1.10, and 0.61 μM, respectively. In competition experiments the addition of calmodulin binding peptide 1 (αII–spec) to Ca2+hCaM M124C–mBBr quenched the fluorescence (Kd = 2.55 ± 1.75 pM) and the later addition of MBC (up to 16 μM) did not affect the fluorescent signal. Instead, the additions of αII–spec to a preformed Ca2+hCaM M124C–mBBr–MBC complex modified the fluorescent signal. However, MBC was able to displace the PDE1A and MLCK from its complex with Ca2+–CaM. In addition, docking studies were performed for all complexes with both ligands showing an excellent correlation with experimental data. These experiments may help to explain why in vivo many CaM drugs target prefer only a subset of the Ca2+–CaM regulated proteins and adds to the understanding of molecular interactions between protein complexes and small ligands. Metal ion and substrate binding to carbamate kinase from Streptococcus faecalis was studied by nuclear magnetic resonance (NMR) and electron paramagnetic resonance using Mn2+ as the paramagnetic probe. The enzyme binds Mn2+ weakly (KD = 0.45 ± 0.05 mm) with a stoichiometry of one per two subunits. However, in the presence of nucleotides, tighter binding of Mn2+ was observed with KD = 44 ± 4 μm in the presence of ADP and KD = 23 ± 4 μm with ATP present. Proton relaxation rate enhancement studies were conducted on water molecules interacting with ternary enzyme-Mn2+-nucleotide and binary enzyme-Mn2+ complexes. Mn2+ bound to carbamate kinase enhances the proton relaxation rate of water giving a binary enhancement value of ?b = 9.3 ± 0.4. When enzyme-Mn2+ was titrated with ADP or ATP, a bell-shaped titration curve was obtained typical of many other enzyme-Mn2+-nucleotide ternary complexes. Computer fits to the titration data gave ternary enhancement values of ?tADP = 14 ± 1 and ?tATP = 19 ± 1. The dissociation constants for Mn-ADP and Mn-ATP binding to carbamate kinase were also obtained from these data analyses and are K1 = 2.5 ± 0.5 μm and K1 = 50 ± 8 μm, respectively. Therefore, these data demonstrate the formation of a ternary enzyme-metal-nucleotide bridge complex at the nucleotide substrate site of carbamate kinase. Distance measurements were conducted by NMR techniques with 13C-enriched carbamate and demonstrate that carbamate is 4–8 Å from enzyme-bound Mn2+. Thus carbamate binds near the metal-nucleotide substrate site of carbamate kinase.  相似文献

The pyruvate dehydrogenase complex was isolated from the mitochondria of broccoli florets and shown to be similar in its reaction mechanism to the complexes from other sources. Three families of parallel lines were obtained for the initial velocity patterns, indicating a multisite ping-pong mechanism. The apparent Km values obtained were 321 ± 18, 148 ± 13, and 7.2 ± 0.51 μm for pyruvate, NAD+, and CoA, respectively. Product inhibition studies using acetyl-CoA and NADH yielded results which were in agreement with those predicted by the multisite ping-pong mechanism. Acetyl-CoA and NADH were found to be competitive inhibitors versus CoA and NAD+, respectively. All other substrate-product combinations showed uncompetitive inhibition patterns, except for acetyl-CoA versus NAD+. Among various metabolites tested, only hydroxypyruvate (Ki = 0.11 mM) and glyoxylate (Ki = 3.27 mM) were found to be capable of inhibiting the broccoli enzyme to a significant degree. Initial velocity patterns using Mg2+? or Ca2+-thiamine pyrophosphate and pyruvate as the variable substrate were found to be consistent with an equilibrium ordered mechanism where Mg? or Ca-thiamine pyrophosphate bind first, with dissociation constants of 33.8 and 3 μm, respectively. The Mg- or Ca-thiamine pyrophosphate complexes also dissociated rapidly from the enzyme complex.  相似文献

(1) The effects of calmodulin binding on the rates of Ca2+-dependent phosphorylation and dephosphorylation of the red-cell Ca2+ pump, have been tested in membranes stripped of endogenous calmodulin or recombined with purified calmodulin. (2) In Mg2+-containing media, phosphorylation and dephosphorylation rates are accelerated by a large factor (at 0°C), but the steady-state level of phosphoenzyme is unaffected by calmodulin binding (at 0°C and 37°C). In Mg2+-free media, slower rates of phosphoenzyme formation and hydrolysis are observed, but both rates and the steady-state phosphoenzyme level are raised following calmodulin binding. (3) At 37°C and 0°C, the rate of (Ca2+ + Mg2+)-ATPase activity is stimulated maximally by 6–7-fold, following calmodulin binding. At 37°C the apparent Ca2+ affinity for sustaining ATP hydrolysis is raised at least 20-fold, Km(Ca) ? 10 μM (—calmodulin) and Km(Ca) < 0.5 μM (+ calmodulin), but at 0°C the apparent Ca2+ affinity is very high in calmodulin-stripped membranes and little or no effect of calmodulin is observed (Km(Ca) ? 3–4 · 10-8 M). (Ca2+ + Mg2+)-ATPase activity in calmodulin activated membranes and at saturating ATP levels, is sharply inhibited by addition of calcium in the range 50–2000 μM. (4) A systematic study of the effects of the nucleotide species MgATP, CaATP and free ATP on (Ca2+ + Mg2+)-ATPase activity in calmodulin-activated membranes reveals: (a) In the 1–10 μmolar concentration range MgATP, CaATP and free ATP appear to sustain (Ca2+ + Mg2+)-ATPase activity equally effectively. (b) In the range 100–2000 μM, MgATP accelerates ATP hydrolysis (Km(MgATP) ? 360 μM), and CaATP is an inhibitor (Ki(CaATP) ? 165 μM), probably competing with MgATP fo the regulatory site. (5) The results suggest that calmodulin binding alters the conformational state of the Ca2+- pump active site, producing a high (Ca2+ + Mg2+)-ATPase activity, high Ca2+ affinity and regulation of activity by MgATP.  相似文献

Metals bind to the bacterial cell wall, yet the binding mechanisms and affinity constants are not fully understood. The cell wall of gram positive bacteria is characterized by a thick layer of peptidoglycan and anionic teichoic acids anchored in the cytoplasmic membrane as lipoteichoic acid or covalently bound to the cell wall as wall teichoic acid. The polyphosphate groups of teichoic acid provide one-half of the metal binding sites for calcium and magnesium, which contradicts previous reports that calcium binding is 100 % dependent on teichoic acid. The remaining binding sites are formed with the carboxyl units of peptidoglycan. In this work we report equilibrium association constants and total metal binding capacities for the interaction of calcium and magnesium ions with the bacterial cell wall. Metal binding is much stronger than previously reported. Curvature of Scatchard plots from the binding data and the resulting two regions of binding affinity suggest the presence of negative cooperative binding, which means that the binding affinity decreases as more ions become bound to the sample. For Ca2+, Region I has a KA = (1.0 ± 0.2) × 106 M?1 and Region II has a KA = (0.075 ± 0.058) × 106 M?1. For Mg2+, KA1 = (1.5 ± 0.1) × 106 and KA2 = (0.17 ± 0.10) × 106. A binding capacity (η) is reported for both regions. However, since binding is still occurring in Region II, the total binding capacity is denoted by η2, which are 0.70 ± 0.04 and 0.67 ± 0.03 µmol/mg for Ca2+ and Mg2+ respectively. These data contradict the current paradigm of only a single metal affinity value that is constant over a range of concentrations. We also find that measurement of equilibrium binding constants is highly sample dependent. This suggests a role for diffusion of metals through heterogeneous cell wall fragments. As a result, we are able to reconcile many contradictory theories that describe binding affinity and the binding mode of divalent metal cations.  相似文献

Artificial pH gradients across tonoplast vesicles isolated from storage tissue of red beet (Beta vulgaris L.) were used to study the kinetics of a Ca2+/H+ antiport across this membrane. Ca2+-dependent H+ fluxes were measured by the pH-dependent fluorescence quenching of acridine orange. ΔpH-dependent Ca2+ influx was measured radiometrically. Both H+ efflux and Ca2+ influx displayed saturation kinetics and an identical dependence on external calcium with apparent Km values of 43.9 and 41.7 micromolar, respectively. Calcium influx was unaffected by an excess of Mg2+ but was inhibited by La3+ > Mn2+ > Cd2+. The apparent Km for external calcium was greatly affected (5-fold) by internal pH in the range of 6.0 to 6.5 and a transmembrane effect of internal proton binding on the affinity for external calcium is suggested.  相似文献

Dissociation and alkali complex formation equilibria of nitrilotris(methylenephosphonic acid) (NTMP, H6L) have been studied by dilatometric, potentiometric and 31P NMR-controlled titrations. Dilatometry indicated the formation of alkali complexes ML (M=Li, Na, K, Rb, Cs) at high pH with a stability decreasing from Li to Cs. An efficient combination of potentiometric and NMR methods confirmed two types of alkali metal complexes MHL and ML. Stability constants for the equilibria following M+ + HL5− ? MHL4− and M+ + L6− ? ML5−, respectively, were determined: logKNaHL=1.08(0.07), logKKHL=0.86(0.08), logKNaL=2.24(0.03). Systematic errors are introduced by using alkali metal hydroxides as titrants for routine potentiometric determinations of dissociation constants pKa5app and pKa6app. Correction formulae were derived to convert actual dissociation constants pKa into apparent dissociation constants pKaapp (or vice versa). The actual dissociation constants were found: pKa5(H2L4− ? H+ + HL5−)=7.47(0.03) and pKa6(HL5− ? H+ + L6−)=14.1(0.1). The anisotropy of 31P chemical shifts of salts MnH6 − nL (M=Li, Na, n=0-5) is more sensitive towards titration (n) than isotropic solution state chemical shifts.  相似文献

Conductance and equilibrium dialysis studies are reported for the aqueous systems (native calf thymus) DNA-CaCl2 and DNA-MgCl2 at various pH values and ionic strengths at 25 °C. Discontinuities occur in the conductance curves at mole ratios of Ca2+ and Mg2+ to nucleate phosphorus of 0.125, 0.30, and 0.50. The dialysis results show the formation of complexes of stoichiometry 0.50 and 1.00 mol Ca2+ or Mg2+/mol nucleate phosphorus (2:1 and 1:1 complexes), the latter only in neutral or alkaline solutions, in agreement with the conductance discontinuity at 0.50. The other discontinuities may be due to preferential binding in the formation of the 2:1 complex. Binding constants for the 2:1 complexes are evaluated. Absorption-temperature profiles have been determined for “native” and dialysed DNA in the presence of NaCl, CaCl2, and MgCl2. For dialysed DNA at 26 ° C and 260 nm the decrease of absorbance with increased salt concentration was halted for MgCl2 and CaCl2 at a concentration corresponding to the formation of the 2:1 complex. The absorbance of “native” DNA did not decrease. Tm and the reciprocal of the hypochromic rise (1h) increased linearly with log (salt concn). Values of Tm were the same at 230, 260, and 280 nm, but h was greater at 230 and 280 than at 260 nm, which may be due to the existence of alternating blocks of (A + T) and (G ? C) pairs. The entropy of transition was in the order Ca > Mg ? Na.  相似文献

The rates of formation and dissociation of concanavalin A with some 4-methylumbelliferyl and p-nitrophenyl derivatives of α- and β-D-mannopyranosides and glucopyranosides were measured by fluorescence and spectral stopped-flow methods. All process examined were uniphasic. The second-order formation rate constants varied only from 6.8 · 104 to 12.8 · 104 M?. s?1, whereas the first-order dissociation rate constants ranged from 4.1. to 220 s?1, all at ph 5.0, I = 0.3 M, and 25°C. Dissociation rates thus controlled the value of binding constant. The effect of temperature on these reactions was examined, from which enthalpies and entropies of activation and of reaction could be calculated. The effects of pH at 25°C on the reaction rates of 4-methylumbelliferyl α-D-mannopyranoside and 4-methylumbelliferyl α-D-glucopyranoside with concanavalin A were examined. The value of the binding constant Kap (derived from the kinetics) at any pH could be related to the intrinsic binding constant K by the expression Kap = KaK(Ka + [H+])?1. The values of Ka, the ionization constant of the protein segment responsive to sugar binding, were 3 · 10?4 M and 1 · 10?4 M for 4-methylumbelliferyl α-D-mannopyranoside and 4-methylumbelliferyl α-D-glucopyranoside, respectively. The binding constant of p-nitrophenyl α-D-mannopyranoside is surprisingly much less sensitive to a pH change from 5.0 to 2.7. Ionic strength had little effect on the binding characteristics of 4-methylumbelliferyl α-D-mannopyranoside to concanavalin A at pH 5.2 and 25°C.  相似文献

The interactions between cytochrome P450 2B4 (d-2B4), NADPH:cytochrome P450 reductase and cytochrome b5 have been investigated in the monomeric reconstituted P450 2B4-containing monooxygenase system in the presence of a substrate (7-pentoxyresorufin) and an electron donor, NADPH. Each partner was immobilized via its amino groups on the carboxymethyldextran biochip surface of the optical biosensor IAsys+. Such mode immobilization was not accompanied by any loss of activities of the immobilized proteins. The formation of binary d-Fp/d-2B4 complexes was registered. The association/dissociation rate constants (kon/koff) were (0.013 ± 0.005) × 106 M?1 s?1/0.05 ± 0.02 s?1, and dissociation constant (KD) was (0.26 ± 0.13) × 10?6 M. Comparison of kon, koff and KD values for d-Fp/d-2B4 complexes formed under hydroxylation (O-dealkylation) with corresponding constants obtained for the oxidized proteins of (0.10 ± 0.03) × 106 M?1 s?1/(0.14 ± 0.06) s?1, and (0.71 ± 0.37) × 10?6 M, respectively shows that the decrease in kon and an insignificant decrease in KD are associated with the increase of complex lifetime during transition from the oxidized to hydroxylation conditions. Complex formation between d-Fp and d-b5 was not registered in both hydroxylation conditions and in the case of oxidized forms of these proteins. In both cases formation of the ternary d-Fp/d-2B4/d-b5 complexes occurred.  相似文献

The Ca2+-activated channel of intermediate-conductance (KCa3.1) is a target for antisickling and immunosuppressant agents. Many small peptides isolated from animal venoms inhibit KCa3.1 with nanomolar affinities and are promising drug scaffolds. Although the inhibitory effect of peptide toxins on KCa3.1 has been examined extensively, the structural basis of toxin-channel recognition has not been understood in detail. Here, the binding modes of two selected scorpion toxins, charybdotoxin (ChTx) and OSK1, to human KCa3.1 are examined in atomic detail using molecular dynamics (MD) simulations. Employing a homology model of KCa3.1, we first determine conduction properties of the channel using Brownian dynamics and ascertain that the simulated results are in accord with experiment. The model structures of ChTx-KCa3.1 and OSK1-KCa3.1 complexes are then constructed using MD simulations biased with distance restraints. The ChTx-KCa3.1 complex predicted from biased MD is consistent with the crystal structure of ChTx bound to a voltage-gated K+ channel. The dissociation constants (Kd) for the binding of both ChTx and OSK1 to KCa3.1 determined experimentally are reproduced within fivefold using potential of mean force calculations. Making use of the knowledge we gained by studying the ChTx-KCa3.1 complex, we attempt to enhance the binding affinity of the toxin by carrying out a theoretical mutagenesis. A mutant toxin, in which the positions of two amino acid residues are interchanged, exhibits a 35-fold lower Kd value for KCa3.1 than that of the wild-type. This study provides insight into the key molecular determinants for the high-affinity binding of peptide toxins to KCa3.1, and demonstrates the power of computational methods in the design of novel toxins.  相似文献

Calciphorin, the putative mitochondrial calcium ionophore from rat liver mitochondria, exhibits the inherent properties of the mitochondrial calcium transport system and is similar to the calf heart preparation reported earlier. The protein has a strong selectivity for Ca2+, and has a Kd for Ca2+ of 56.5 ± 6.6 μM and 13.9 ± 2.1 μM in organic extraction and flow dialysis experiments, respectively. Reduction of the contaminating lipids from 23 ± 6.5 to 1.73 ± 0. moles per mole protein does not alter the affinities, Ca2+/protein soichiometry or selectivity for Ca2+.  相似文献

The binding of Mn2+, Ca2+, and rare earth ions to apoconcanavalin A has been studied by water proton relaxation enhancement, electron paramagnetic resonance spectroscopy, and fluorescence spectroscopy. An electron paramagnetic resonance and water proton relaxation rate study of the titration of apoconcanavalin A with Mn2+ gives evidence of two equivalent binding sites per monomer with KD = 50 μm ± 4 μm. When a similar Mn2+ titration of apoconcanavalin A is performed in the presence of Ca2+ ion, very little free Mn2+ is detected by electron paramagnetic resonance until the two Mn2+ binding sites per monomer are filled. The substitution of a rare earth ion for Ca2+ ion in the above experiment often resulted in a slight displacement of Mn2+ from the transition metal site as detected by electron paramagnetic resonance. A water proton relaxation rate study of the titration of apoconcanavalin A with Gd3+ reflects two binding sites with a KD = 40 μm ± 4 μm and two with a KD = 200 μm ± 50 μm. The fluorescence emission spectrum of concanavalin A (λem = 340 nm) is slightly quenched by the addition of Tb3+ while Tb3+ fluorescence is greatly enhanced. A fluorometric titration of apoconcanavalin A with Tb3+ also reflects two sites with a KD = 40 μm ± 15 μm and two with a KD = 270 μm ± 50 μm.  相似文献
