Sand dune ecosystems are one of the areas most affected by the introduction of invasive species which represents a threat for biodiversity conservation. Their invasion patterns and spread may depend on their salinity tolerance, besides other factors. To test this hypothesis, we investigated the effects of salt stress on seed germination and on the activity of antioxidant enzymes (catalase, CAT; ascorbate peroxidase, APX; peroxidase, POX; and glutathione reductase, GR) in two legume species, an invasive, Acacia longifolia (Andrews.) Willd., and a native, Ulex europaeus (L.), very common in the sand dunes of the coast of Portugal. Salt stress was induced by adding NaCl at different concentrations, 0, 50, 100 and 200 mM, for 15 days. Results showed that the highest germination percentages were obtained in distilled water (control) and that, with increasing salt concentration, seed germination was delayed and decreased in both species. Inhibition of germination was higher in the native species, only 3% of seeds germinated at 100 mM and no seeds germinated at 200 mM NaCl. In the invasive species, the reduction was higher at 200 mM NaCl (16%). Considering the coefficient of germination velocity, a decrease in both species with increasing NaCl concentration was observed. The CAT and GR activities decreased in A. longifolia with increasing salinity. In turn, APX activity significantly increased as NaCl concentration increased while the POX activities declined at the highest NaCl concentration. On the other hand, at 50 mM NaCl lower activity of CAT and APX and higher GR and POX were found in U. europaeus. In both species, protein content increased as NaCl concentration increased. In addition, it seems that APX activities play an essential role in the scavenging reactive oxygen species (ROS). These results suggest that the seeds of the invasive legume A. longifolia are more tolerant to salinity than the native legume U. europaeus, and seem better equipped to handle the physiological stress of high salinity, which may contribute to its invasive ability in sand dunes.  相似文献

Arbuscular mycorrhizal colonization was measured in the most abundant plant species of the Paraguaná Peninsula, northwestern Venezuela. These plant species included: Acacia tortuosa, Argusia gnaphalodes, Croton punctatus, Croton rhamnifolius, Egletes prostrata, Melochia tomentosa, Panicum vaginatum, Scaevola plumieri, Sporobolus virginicus, Suriana maritima, Leptothrium rigidum, and Fimbristylis cymosa. Mycorrhizal colonization was assessed using the Trouvelot et al. (1986) method that allows for simultaneous evaluation of frequency of colonization (%F), intensity of colonization (%M), and the proportion of arbuscules (%A) and vesicles (%V) present in the roots. Average frequency of colonization was 69%. The highest frequency of colonization was around 92% in C. rhamnifolius and A. tortuosa; in the other species, it varied from 49 to 86%. L. rigidum and F. cymosa were considered nonmycorrhizal because its colonization was very scarce and at all times appeared without arbuscules. Average intensity of colonization was 7%. The highest intensity of colonization was 18% in C. rhamnifolius. In the other species, it varied from 3 to 15%. Paspalum vaginatum, A. gnaphalodes, M. tomentosa, and S. maritima had their fungal structures tightly packed in modified little ovoid roots. In general, frequency of AM colonization was high and similar to those reported for other tropical ecosystems, whereas the intensity of AM colonization was low and similar to values obtained in analogous studies in disturbed ecosystems.  相似文献

An emerging paradigm in sustainable biotechnique is the use of mutualists to enhance plant growth and secondary metabolism. Our objective was to determine impact of two groups of fungal mutualists on growth and phytochemistry of Echinacea purpurea. Growth, development, and phytochemical concentration were measured in greenhouse-grown 12-week-old plants colonized by arbuscular mycorrhizal fungi (AMF) (Rhizophagus intraradices and Gigaspora margarita) or the endophytic entomopathogen, Beauveria bassiana. In one experiment, all measured growth parameters were increased in mycorrhizal plants. Biomass of AMF-colonized plants was over 13-fold greater than non-mycorrhizal controls receiving the same levels of phosphorous, and over 4-fold greater than non-mycorrhizal controls given additional phosphorous. Endophytic colonization by B. bassiana had minor effects on growth. Colonization by AMF and B. bassiana alone or in combination altered concentrations of phytochemicals (pigments, polyphenolics, alkylamides, and terpenes). Mycorrhizal plants produced up to 4.6-fold higher concentration of polyphenolics. Specific alkylamides increased 1.7 fold in plants colonized only with B. bassiana and up to a 2.4-fold increase in plants colonized by both mutualists. Changes in other phytochemical classes were related to differences in plant size induced by AMF. Phytochemical content (concentration × biomass) was increased up to 30-fold in mycorrhizal plants. Phytochemical relationships to plant biomass were confirmed in a second experiment in which non-mycorrhizal plants were fertilized to produce biomass equivalent to that of mycorrhizal plants. Based on this study, mycorrhizal colonization of E. purpurea enhances phytochemical content; this has major implications for the natural product industries and growers of E. purpurea.  相似文献

The tropical coastal dunes in central Gulf of Mexico have been stabilizing over the last decades resulting in reduced substrate mobility, and promoting primary succession. We describe changes in species richness and diversity in dune vegetation during 20?years. Our questions: (a) Do species richness and diversity increase over time as predicted by models of ecological succession or do they show a hump-backed manner similar to the observations in temperate coastal dunes?, (b) What is the interaction between vegetation cover and diversity and species richness?, (c) Is there a relationship between species diversity and succession rate and does succession rate change over time?, and (d) How do plant functional types change during succession? In order to answer these questions, we set 140 4?×?4?m permanent plots in a mobile dune area and monitored vegetation cover and species richness from 1991 to 2011. In time, diversity increased in a logistic manner toward an asymptotic value once vegetation cover surpassed 60?%. Species richness increased in a humped-back shape, also reaching a maximum peak at 60?% vegetation cover. The succession rate of diversity was measured by the Euclidean distance, and showed a significant humped-back relation, meaning that it was slower in early and late successional stages. The study supports the intermediate disturbance theory. The conservation of coastal dunes vegetation should focus on all, species-poor and species-rich habitats that help to maintain the ecological integrity of these ecosystems. The understanding of community dynamics and diversity patterns becomes an essential component of coastal dune management and conservation.  相似文献

Bacterial diversity in the rhizosphere of beach morning glory (Calystegia soldanella) and wild rye (Elymus mollis), two of the major plant species inhabiting the coastal sane dune in Tae-An, Korea, was studied by the analysis of community 16S rRNA gene clones. The amplified rDNA restriction analysis (ARDRA) of the clones using HaeIII exhibited significant differences in the community composition between the two plant species as well as regional differences, but also identified a specific ARDRA pattern that was most common among the clones regardless of plant species. Subsequent sequence analysis indicated that the pattern was that of Lysobacter spp., which is a member of the family Xanthomonadaceae, class Gamma proteobacteria. The Lysobacter clones comprised 50.6% of the clones derived from C. soldanella and 62.5% of those from E. mollis. Other minor patterns included those of Pseudomonas spp., species of Rhizobium, Chryseobacterium spp. and Pantoea spp. among C. soldanella clones, and Pseudomonas sp. and Aeromonas hydrophila among E. mollis clones. It is not yet clear what kind of roles Lysobacter plays in association with sand dune plants, but its universal presence in the rhizosphere, together with the potential of this taxon for antagonistic activity against plant pathogens, suggests that Lysobacter might form a symbiotic relationship with its host plants.  相似文献

Coastal dunes provide a variety of cultural and ecological ecosystem services to local communities. However, most of these ecosystems are globally threatened by anthropic factors. Chile is home to important dune ecosystems such as the emblematic Ritoque coastal dunes located in the hotspot for biodiversity conservation in central Chile where there is a delicate relationship between conservation and human development interests, among local communities and authorities. Here, we performed a choice experiment study to determined willingness to implement conservation measures in the Ritoque dunes. In particular, we assessed residents’ preferences for improvements of the current state of local ecosystem services such as scenic beauty, biodiversity and archaeological sites. Residents showed preferences for improvements in Ritoque coastal dunes’ ecosystem services, and a high willingness to pay for protecting biodiversity and archeological sites, rather than for recreational infrastructure. Our study highlights the importance of place-based research that specifically addresses residents and stakeholders’ conservation priorities, which could be used to design and implement effective conservation management strategies for coastal ecosystems and coastal cities near them.  相似文献

Benefits arising from facultative mutualisms between ants and plants vary with the identity of the ant partner. Invasive and native ants are both attracted to plants that offer extrafloral nectar, but few studies have compared their abilities to displace herbivores and benefit plants. Yellow crazy ants Anoplolepis gracilipes have invaded eucalypt woodlands of Arnhem Land, northern Australia, where they displace the native dominant weaver ant Oecophylla smaragdina. We compared the plant defense services provided by A. gracilipes and O. smaragdina ants on trees with (Acacia lamprocarpa) and without (Eucalyptus tetrodonta) extrafloral nectar rewards through surrogate herbivore (termite) addition experiments and surveys of herbivore damage. Anoplolepis gracilipes were more likely than O. smaragdina to discover termites on A. lamprocarpa, but the likelihood of termite discovery on E. tetrodonta did not vary with ant species. Anoplolepis gracilipes were also more thorough in their attacks of termites, recruited 3.4–4 times more workers to termites, and were 3.4 times quicker at discovering termites on A. lamprocarpa than were O. smaragdina. Discovery of termites by other predators did not vary significantly between trees in A. gracilipes and O. smaragdina sites. Herbivory scores did not reflect the foliage patrolling pattern by the ants. Old A. lamprocarpa leaves and both new and old leaves and branches on E. tetrodonta in A. gracilipes sites had higher chewing herbivory scores than their counterparts in O. smaragdina sites. Our results reveal that the more aggressive and efficient foliar patrolling by A. gracilipes does not translate to increased plant protection. Ant invasions can disrupt native ant–plant mutualisms despite invasive ants possessing many traits associated with effective plant guarding.  相似文献

The aim of this research was to link vegetation characteristics, such as spatial and temporal distribution, and environmental variables, with land cover information derived from remotely sensed satellite images of the Eastern Mediterranean coastal wetlands of Turkey. The research method was based on (i) recording land cover characteristics by means of a vegetation indicator, and (ii) classifying and mapping coastal wetlands utilizing a Landsat Thematic Mapper (TM) image of Çukurova Deltas in Turkey. Vegetation characteristics of various habitats, such as sand dunes, salt marshes, salty plains and afforestation areas, were identified by field surveys. A Landsat TM image of 4 July 1993 was pre-processed and then classified using the Maximum Likelihood (ML) algorithm and Artificial Neural Networks (ANN). As a result of this supervised classification, the land cover types were classified with a largest accuracy of 90.2% by ANN. The classified satellite sensor imagery was linked to vegetation and bird census data, which were available through literature in a Geographical Information System (GIS) environment to determine the spatial distribution of plant and bird biodiversity in this coastal wetland. The resulting data provide an important baseline for further investigations such as monitoring, change detections and designing conservation policies in this coastal ecosystem.  相似文献

The apoptosis program of physiological cell death elicits a range of non-phlogistic homeostatic mechanisms—“recognition, response and removal”—that regulate the microenvironments of normal and diseased tissues via multiple modalities operating over short and long distances. The molecular mechanisms mediate intercellular signaling through direct contact with neighboring cells, release of soluble factors and production of membrane-delimited fragments (apoptotic bodies, blebs and microparticles) that allow for interaction with host cells over long distances. These processes effect the selective recruitment of mononuclear phagocytes and the specific activation of both phagocytic and non-phagocytic cells. While much evidence is available concerning the mechanisms underlying the recognition and responses of phagocytes that culminate in the engulfment and removal of apoptotic cell bodies, relatively little is yet known about the non-phagocytic cellular responses to the apoptosis program. These responses regulate inflammatory and immune cell activation as well as cell fate decisions of proliferation, differentiation and death. Here, we review current knowledge of these processes, considering especially how apoptotic cells condition the microenvironments of normal and malignant tissues. We also discuss how apoptotic cells that persist in the absence of phagocytic clearance exert inhibitory effects over their viable neighbors, paying particular attention to the specific case of cell cultures and highlighting how new cell-corpse-clearance devices—Dead-Cert® Nanoparticles—can significantly improve the efficacy of cell cultures through effective removal of non-viable cells in the absence of phagocytes in vitro.  相似文献

Aflatoxins are highly carcinogenic mycotoxins frequently produced by Aspergillus flavus. Contamination of maize with aflatoxins imposes both economic and health burdens in many regions. Identification of the most important etiologic agents of contamination is complicated by mixed infections and varying aflatoxin-producing potential of fungal species and individuals. In order to know the potential importance of an isolate to cause a contamination event, the ability of the isolate to produce aflatoxins on the living host must be determined. Aflatoxin production in vitro (synthetic and natural media) was contrasted with in vivo (viable maize kernels) in order to determine ability of in vitro techniques to predict the relative importance of causal agents to maize contamination events. Several media types and fermentation techniques (aerated, non-aerated, fermentation volume) were compared. There was no correlation between aflatoxin production in viable maize and production in any of the tested liquid fermentation media using any of the fermentation techniques. Isolates that produced aflatoxins on viable maize frequently failed to produce detectable (limit of detection = 1 ppb) aflatoxin concentrations in synthetic media. Aflatoxin production on autoclaved maize kernels was highly correlated with production on viable maize kernels. The results have important implications for researchers seeking to either identify causal agents of contamination events or characterize atoxigenic isolates for biological control.  相似文献

Identifying and quantifying the factors that contribute to the potential misclassification of the ecological status of water bodies is a major challenge of the Water Framework Directive (WFD). The present study compiles extensive biomonitoring data from a range of macrophyte-based classification methods developed by several European countries. The data reflect spatial and temporal variation as well as inter-observer variation. Uncertainty analysis identified that factors related to the spatial scale of sampling generally contributed most to the uncertainty in classifying water bodies to their ecological status, reflecting the high horizontal and depth-related heterogeneity displayed by macrophyte communities. In contrast, the uncertainty associated with temporal variation was low. In addition, inter-observer variation, where assessed, did not contribute much to overall uncertainty, indicating that these methods are easily transferable and insensitive to observer error. The study, therefore, suggests that macrophyte-based sampling schemes should prioritize large spatial replication over temporal replication to maximize the effectiveness and reliability of water body classification within the WFD. We encourage conducting similar uncertainty analyses for new/additional ecological indicators to optimize sampling schemes and improve the reliability of classification of ecological status.  相似文献

The probability of, and time to, fixation of a mutation in a population has traditionally been studied by the classic Wright–Fisher model where population size is constant. Recent theoretical expansions have covered fluctuating populations in various ways but have not incorporated models of how the environment fluctuates in combination with different levels of density-compensation affecting fecundity. We tested the hypothesis that the probability of, and time to, fixation of neutral, advantageous and deleterious mutations is dependent on how the environment fluctuates over time, and on the level of density-compensation. We found that fixation probabilities and times were dependent on the pattern of autocorrelation of carrying capacity over time and interacted with density-compensation. The pattern found was most pronounced at small population sizes. The patterns differed greatly depending on whether the mutation was neutral, advantageous, or disadvantageous. The results indicate that the degree of mismatch between carrying capacity and population size is a key factor, rather than population size per se, and that effective population sizes can be very low also when the census population size is far above the carrying capacity. This study highlights the need for explicit population dynamic models and models for environmental fluctuations for the understanding of the dynamics of genes in populations.  相似文献

This article analyses the morphological history of the outer Thames seabed, covering over 3,000 km2 from Aldeburgh (Suffolk), to Southend-on-Sea (Essex) and Margate (Kent). The region has been depicted on bathymetric charts since the sixteenth century, and has been formally charted since the eighteenth century. Charts published since the early 1800s incorporate sufficient grid reference or ground control detail for georectification onto a common coordinate system (British National Grid). The morphological history of the outer seabed was thus reconstructed through the digitisation and interpolation of soundings onto a regular grid (3D surface). The evolution of seabed morphology was examined using transects, bathymetric change maps and spatial statistics. The results show considerable spatial variability in seabed behaviour. Within the central Thames, banks have experienced significant depth changes can be associated with lateral shifts in individual banks. Some of the outer banks in this region exhibit progressive elongation. Shifts in bank position across the Suffolk shoreface appear to be more subtle, and there is evidence here of both onshore and offshore migration. There is no clear evidence of any regionally coherent response to large-scale historical forcing such as sea-level rise.  相似文献

Nuclear pore complexes (NPCs) conduct selective, bidirectional transport across the nuclear envelope. The NPC passageway is lined by intrinsically disordered proteins that contain hydrophobic phenylalanine-glycine (FG) motifs, known as FG nucleoporins (FG nups), that play the key role in the NPC transport mechanism. Cohesive interactions among the FG nups, which arise from the combination of hydrophobic, electrostatic, and other forces, have been hypothesized to control the morphology of the assemblies of FG nups in the NPC, as well as their permeability with respect to the transport proteins. However, the role of FG nup cohesiveness is still vigorously debated. Using coarse-grained polymer theory and numerical simulations, we study the effects of cohesiveness on the selective permeability of in vitro FG nup assemblies in different geometries that have served as proxies for the morphological and transport properties of the NPC. We show that in high-density FG nup assemblies, increase in cohesiveness leads to the decrease in their permeability, in accordance with the accepted view. On the other hand, the permeability of low-density assemblies is a nonmonotonic function of the cohesiveness, and a moderate increase in cohesiveness can enhance permeability. The density- and cohesiveness-dependent effects on permeability are explained by considering the free-energy cost associated with penetrating the FG nup assemblies. We discuss the implications of these findings for the organization and function of the NPC.  相似文献

Acacia species in arid environments are thought to only establish in years of above-average rainfall, so should exhibit cohorted or pulsed recruitment. I studied population demography of Acacia erioloba Meyer in semi-arid savanna in the Kimberley area (mean annual precipitation = 425 ± 132 mm), South Africa, to establish whether they recruit episodically. This species was found to have a sapling bank at the sites, indicating that even though cohorted recruitment probably occurs, it is not the primary factor limiting recruitment to larger size classes. A. erioloba saplings given supplementary water and protection from herbivory showed significantly less height growth than saplings given water but not protected from herbivores, and grass within exclusion plots was taller and denser than outside of exclusion plots. The generalized linear model, although finding a significant difference between watered and protected and watered and unprotected saplings, explained only 16% of variation in growth, demonstrating the importance of factors other than competition from grass when rainfall is above-average. Average height increase across all treatments for a growing season was small (43.1 ± 30.81 mm), suggesting most investment is belowground. A. erioloba sapling below surface stem diameter correlated positively with aboveground growth, indicating larger, and presumably older, individuals grow faster. When grass was removed around saplings, growth rates were not significantly greater than for saplings surrounded by grass, over a growing season. Thus, in these study sites, pulsed events allow seedlings to establish, and then saplings slowly accumulate over many growing seasons within the grass sward, owing to slow aboveground growth combined with competition from grass in absence of grazers. Release events, e.g., heavy grazing combined with good rainfall, may allow mass release, giving the impression of cohorted or pulsed recruitment.  相似文献

A dextranase gene from Penicillium minioluteum (strain IMI068219) has been cloned, sequenced and expressed in Saccharomyces cerevisiae via fusion of the DNA segment encoding the mature dextranase protein with α-factor signal sequence, and insertion into the GAL1–controlled expression vector pYES2/CT. Galactose-induced expression yielded extracellular dextranase activity of 0.63 units/ml and cell-associated dextranase activity of 0.48 units/ml, after 24 h incubation. The dextranase construct was introduced into a strain of S. cerevisiae expressing the human cytochrome P450 3A4 (CYP3A4) and the cognate reductase, which was then used to develop a microplate toxicity bioassay. Toxicity was signalled as inhibition of dextranase activity, assayed fluorimetrically. This novel bioassay was assessed using six economically significant mycotoxins.  相似文献
