Synchronized oscillatory activity at the gamma frequency (30-70 Hz) is thought to be important for information processing in many sensory systems. Here, I used patch-clamp recordings in neuron pairs in rat olfactory bulb slices to assess the mechanisms underlying such "gamma" activity in the olfactory system. Patterned electrical stimulation of afferents that mimicked a natural odor stimulus elicited rapidly synchronized spikes (lag < or = 5 ms) in mitral cells, along with oscillatory activity at the gamma (approximately 50 Hz) frequency. Analysis of coupling potentials, combined with dendritic sectioning, indicated that mitral cell synchrony was mainly driven by inhibitory postsynaptic potentials (IPSPs) imposed by GABAergic granule cells. Recordings in granule cell pairs indicated that granule cells were themselves synchronized by their excitatory inputs from mitral cells, providing a means to coordinate GABA release. These results demonstrate that rapid synchrony can emerge in a network through the precise back-and-forth interplay between neuronal populations. 相似文献

Leaked-integrate-and-fire coupled oscillators (LIFs) were used as a model of electrophysiological activity. The activity of these oscillators determines the speech rhythm, which is governed by the square-law map with inhibition as a controlling parameter. Regular rhythms of convulsive repetitions at early stuttering are changed, however, by a mixture of repetitions and neurotic pauses. This mixture is a "stumbling block" for clinicians. Due to delays, only inhibitory LIFs are capable to create the synchronic activity in-phase or in-anti-phase at medial or at low coupling. This activity has the form of slow oscillations damping to the background level. Splashes of the activity above or below the level lead to neurotic disorders or to convulsive repetitions. It is known from the experimental data that at different cerebellar neurons there are voltage-dependent Ca2+ channels, NMDA receptors, metabotropic glutamate and GABAB receptors. This receptor arrangement ensures that activation of excitatory and inhibitory input results in changes in activity of protein kinases and phosphatases and subsequent modification of synaptic efficacy. The mechanism of synaptic plasticity is advanced that in accordance with the known experimental data concerning the modification of excitatory and inhibitory inputs to Purkinje cells, granule cells, and deep cerebellar nuclei cells. The mechanism is based on a postulate that phosphorylation/dephosphorylation of AMPA (GABAA) receptors on cerebellar cells causes the LTP/LTD of excitatory (LTD/LTP of inhibitory) transmission. It is assumed that modification rules for Purkinje cells, granule cells, and deep cerebellar nuclei cells, wherein cGMP-dependent protein kinase G is involved in synaptic plasticity, are distinct from those of hippocampal/neocortical cells, wherein cAMP-dependent protein kinase A is involved in synaptic plasticity, since cGMP (cAMP) concentration decreases (increases) with Ca2+ rise. 相似文献

Neuronal function depends on the properties of the synaptic inputs the neuron receive and on its intrinsic responsive properties. However, the conditions for synaptic integration and activation of intrinsic responses may to a large extent depend on the level of background synaptic input. In this respect, the deep cerebellar nuclear (DCN) neurons are of particular interest: they feature a massive background synaptic input and an intrinsic, postinhibitory rebound depolarization with profound effects on the synaptic integration. Using in vivo whole cell patch clamp recordings from DCN cells in the cat, we find that the background of Purkinje cell input provides a tonic inhibitory synaptic noise in the DCN cell. Under these conditions, individual Purkinje cells appear to have a near negligible influence on the DCN cell and clear-cut rebounds are difficult to induce. Peripheral input that drives the simple spike output of the afferent PCs to the DCN cell generates a relatively strong DCN cell inhibition, but do not induce rebounds. In contrast, synchronized climbing fiber activation, which leads to a synchronized input from a large number of Purkinje cells, can induce profound rebound responses. In light of what is known about climbing fiber activation under behaviour, the present findings suggest that DCN cell rebound responses may be an unusual event. Our results also suggest that cortical modulation of DCN cell output require a substantial co-modulation of a large proportion of the PCs that innervate the cell, which is a possible rationale for the existence of the cerebellar microcomplex. 相似文献

This paper presents data showing that the sympathetic autonomic areas of the cat thoracolumbar spinal cord contain nerve terminals and fibres with immunoreactivity for at least seven neuropeptides. The distribution in the intermediolateral cell column of the terminals and fibres which contain enkephalin-, neuropeptide Y-, neurotensin-, substance P-, and neurophysin II-like immunoreactivity (ENK, NPY, NT, SP, and NP2, respectively) suggests that these peptides are involved in more generalized functions of the autonomic nervous system. On the other hand, peaks in density of immunoreactivity at certain levels suggest that different levels of influence of sympathetic preganglionic neurons by the various peptides may occur along the length of the thoracolumbar cord. The distribution of terminals and fibres containing somatostatin- and oxytocin-like immunoreactivity (SS and OXY) suggests that these peptides may be part of specific pathways to particular sympathetic preganglionic neurons. The possible sources of the terminals and fibres containing ENK, NPY, NT, SS, and SP include the spinal cord and supraspinal areas, whereas the source of these structures with OXY and NP2 is most likely supraspinal. The data suggest that coexistence of peptides and interactions between structures containing different neuropeptides occur in the spinal autonomic areas. It is speculated that neuropeptides have an important role to play in the regulation of the cardiovascular division of the autonomic nervous system. 相似文献

A modified system of Hodgkin-Huxley equations was used to describe transmembrane ionic currents during fixed changes of membrane potential and generation of action potentials in the soma of mollusk giant neurons. In the guinea-pig colon, acetylcholine (ACh) release from intrinsic cholinergic motor neurons is inhibited by adrenoceptors, opioid and muscarinic receptors. Chronic sympathetic denervation resulted in supersensitivity to the inhibitory effect of DAMGO (mu-opioid agonist) on ACh release and on the peristaltic reflex. After chronic treatment with naltrexone (NTX) supersensitivity to DAMGO and subsensitivity to UK14,304 (alpha2-adrenoceptor agonist) developed for both functional parameters. The facilitatory effect of scopolamine on ACh release remained unchanged after chronic NTX treatment, whereas it was potentiated after chronic sympathetic denervation. These data suggest the existence of a functional interaction between different inhibitory pathways modulating cholinergic motor neurons in the guinea-pig colon. Namely, chronic manipulation of an inhibitory pathway may entail adaptive sensitivity changes in another inhibitory pathway so that homeostasis can be maintained. 相似文献

In order to gain insight into sensory processing modulating reproductive behavioral and endocrine changes, we have aimed at identifying afferent pathways to neurons synthesizing luteinizing hormone-releasing hormone (LHRH, also known as gonadotropin-releasing hormone [GnRH]), a key neurohormone of reproduction. Injection of conditional pseudorabies virus into the brain of an LHRH::CRE mouse line led to the identification of neuronal networks connected to LHRH neurons. Remarkably, and in contrast to established notions on the nature of LHRH neuronal inputs, our data identify major olfactory projection pathways originating from a discrete population of olfactory sensory neurons but fail to document any synaptic connectivity with the vomeronasal system. Accordingly, chemosensory modulation of LHRH neuronal activity and mating behavior are dramatically impaired in absence of olfactory function, while they appear unaffected in mouse mutants lacking vomeronasal signaling. Further visualization of afferents to LHRH neurons across the brain offers a unique opportunity to uncover complex polysynaptic circuits modulating reproduction and fertility. 相似文献

Postsynaptic processes induced by glutamate, GABA, and dopamine in dendritic spines of inhibitory striatal neurones, were studied. Some functional features were revealed in striatal neurones activation of two protein kinases, cAMP-dependent PKA and cGMP-dependent PKG; presence of calcium/calmodulin-independent adenylate cyclase; bidirectional changes of the cAMP concentration with dopamine. Rise of the cGMP concentration in striatum seems to be a result of activation of the membrane-bound guanylate cyclase via the GABAb receptors. The findings suggest that the active protein kinases/phosphatases ratio is affected by calcium influx through the NMDA-channels. 相似文献

Recent studies indicate that dopamine neurons in the ventral tegmental area (VTA) and substantia nigra pars compacta (SNc) convey distinct signals. To explore this difference, we comprehensively identified each area's monosynaptic inputs using the rabies virus. We show that dopamine neurons in both areas integrate inputs from a more diverse collection of areas than previously thought, including autonomic, motor, and somatosensory areas. SNc and VTA dopamine neurons receive contrasting excitatory inputs: the former from the somatosensory/motor cortex and subthalamic nucleus, which may explain their short-latency responses to salient events; and the latter from the lateral hypothalamus, which may explain their involvement in value coding. We demonstrate that neurons in the striatum that project directly to dopamine neurons form patches in both the dorsal and ventral striatum, whereas those projecting to GABAergic neurons are distributed in the matrix compartment. Neuron-type-specific connectivity lays a foundation for studying how dopamine neurons compute outputs. 相似文献

Convergence of both afferents from the PC and saccular macula, and those from the PC and utricular macula on single vestibular neurons was noted by use of intercellular recording from vestibular neurons. Vestibular neurons were classified VO neurons (vestibulo-ocular proper neurons), VOS (Vestibulo-oculo-spinal neurons sending axon collaterals both to the extraocular motoneuron pools and to the spinal cord), VS neurons (vestibulospinal proper neurons) and V neurons (vestibular neurons without axons to the oculomotor nuclei or the spinal cord) on the basis of whether or not they responded antidromically to stimulation of the oculomotor nuclei and the spinal cord. Of the total 143 vestibular neurons recorded in the series of experiments on convergence of the PC and saccular afferents, 47 neurons (33%) were received inputs from both the PC and saccular nerves. Twenty-six of the 47 convergent neurons were identified as having the nature of VS neurons. Half (13/26) of those were activated monosynaptically from both the PC and saccular nerves. Only one saccular-activated neuron without PC inputs sent an axon to the oculomotor nuclei. In the other series of experiments on the convergence of the PC and utricular afferents, 41 (37%) of 111 vestibular neurons were proved to converge on inputs from both nerves. The majority (35/41) of the neurons received monosynaptic inputs from the PC nerve and polysynaptic EPSP-IPSP sequences from the utricular nerve, or vice versa. The ratio of PC-otolith convergent neurons among utricular-activated neurons (41/54, 76%) was higher than that among saccular activated neurons (47/88, 53%). The percentage of utricular alone neurons without PC inputs (13/111, 12%) was less than that of the saccular alone without PC inputs (41/145, 28%). In conclusion, the convergence of canal and otolith inputs likely contribute mainly to vestibulospinal reflexes including the vestibulocollic reflex, by sending inputs to the neck and other muscles during head inclination which creates the combined stimuli of angular and linear acceleration. 相似文献

Summary Plasmid maintenance allows the strain JM109 of Escherichia coli to grow in a minimal defined medium (M9). JM109 carrying no plasmid can hardly grow in M9 whereas JM109 carrying one, two and three plasmids have a clear metabolic advantage over the untransformed strain. In a complex medium like LB (Luria-Bertani Broth) all strains grow well and despite the number of plasmids carried by the host maximum specific growth rates are not severely affected. Our results suggest that the glucose metabolism is an essential factor contributing to this behavior. 相似文献

Subthreshold aortic nerve (AN) inputs to neurons receiving a monosynaptic AN-evoked input (MSNs: respond to each of two AN stimuli separated by 5 ms) and neurons receiving a polysynaptic AN input (PSNs) in the nucleus of the solitary tract (NTS) were identified in anesthetized rats. In extracellular recordings from 24 MSNs and 49 PSNs, 12% of MSNs and 29% of PSNs only responded to AN stimulation during the application of excitatory amino acids. In intracellular recordings from 24 MSNs and 22 PSNs, 12% of MSNs and 14% of PSNs responded to AN stimulation with excitatory postsynaptic potentials that did not evoke action potential discharge. Reductions in arterial pressure produced minimal changes in the spontaneous discharge of suprathreshold AN-evoked neurons, suggesting that these neurons receive excitatory inputs from nonbaroreceptor sources. The results suggest that some baroreflex-related NTS neurons exist in a "reserve state and can be changed to an active state or vice versa. This will change the number of neurons involved in baroreflex circuits and provides a novel mechanism for regulating baroreflex function independently of alterations in peripheral afferent input. 相似文献

The role of excitatory amino acid (EAA) receptors in the rostral ventrolateral medulla (RVLM) in maintaining resting sympathetic vasomotor tone remains unclear. It has been proposed that EAA receptors in the RVLM mediate excitatory inputs both to presympathetic neurons and to interneurons in the caudal ventrolateral medulla (CVLM), which then provide a counterbalancing inhibition of RVLM presympathetic neurons. In this study, we tested this hypothesis by determining the effect of blockade of EAA receptors in the RVLM on mean arterial pressure (MAP), heart rate (HR), and renal sympathetic nerve activity (RSNA), after inhibition of CVLM neurons. In anesthetized rats, bilateral injections of muscimol in the CVLM increased MAP, HR, and RSNA. Subsequent bilateral injections of kynurenic acid (Kyn, 2.7 nmol) in the RVLM caused a modest reduction of approximately 20 mmHg in the MAP but had no effect, when compared with the effect of vehicle injection alone, on HR or RSNA. By approximately 50 min after the injections of Kyn or vehicle in the RVLM, the MAP had stabilized at a level close to its original baseline level, but the HR and RSNA stabilized at levels above baseline. The results indicate that removal of tonic EAA drive to RVLM neurons has little effect on the tonic activity of RVLM presympathetic neurons, even when inputs from the CVLM are blocked. Thus the tonic activity of RVLM presympathetic neurons under these conditions is dependent on excitatory synaptic inputs mediated by non-EAA receptors and/or the autoactivity of these neurons. 相似文献

In computer-experiments using the interactive program, CRONA, and using data from natural experiments that measured the reversal potential of slow (long-term) inhibitory postsynaptic potentials (slow IPSPs), we determined the probable location of the region of potassium-conducting synapses that are responsible for their generation. Parameters such as the geometric dimensions of neuronal dendritic branches and the intracellular concentration of K+ were studied for their effect on the determination of this region. It is concluded that these synaptic inputs are non-somatic, and that allowing for the variability of the initial parameters they probably lie on the apical dendrites at a distance between 110 and 460 µm from the soma.Dnepropetrovsk State University. Impairments in certain cognitive functions, such as working memory, are core features of schizophrenia. Convergent findings indicate that a deficiency in signalling through the TrkB neurotrophin receptor leads to reduced GABA (gamma-aminobutyric acid) synthesis in the parvalbumin-containing subpopulation of inhibitory GABA neurons in the dorsolateral prefrontal cortex of individuals with schizophrenia. Despite both pre- and postsynaptic compensatory responses, the resulting alteration in perisomatic inhibition of pyramidal neurons contributes to a diminished capacity for the gamma-frequency synchronized neuronal activity that is required for working memory function. These findings reveal specific targets for therapeutic interventions to improve cognitive function in individuals with schizophrenia. 相似文献

Researchers studied the convergence of the vertical posterior semicircular canal (PC), saccular nerves (SAC), utricular nerves (UT), and horizontal semicircular canal nerves (HC) on single vestibular neurons. The vestibular neurons were categorized by their innervating targets. Vestibular neurons were classified as vestibulospinal proper neurons (VS), vestibulo-ocular proper neurons (VO), vestibulo-oculo-spinal neurons sending axon collaterals to the extraocular motoneuron pools and spinal cord (VOS), and vestibular nucleus neurons without axons to the oculomotor nuclei or the spinal cord (V).
