To remediate indoor fungal growth, understanding the moisture relations of common indoor fungi is crucial. Indoor moisture conditions are commonly quantified by the relative humidity (RH). RH is a major determinant of the availability of water in porous indoor surfaces that fungi grow on. The influence of steady‐state RH on growth is well understood. Typically, however, the indoor RH constantly changes so that fungi have to endure frequent periods of alternating low and high RH. Knowledge of how common indoor fungi survive and are affected by the low‐RH periods is limited. In particular, the specific effects of a drop in RH on the growth of the mycelium remain unclear. In this work, video microscopy was used to monitor hyphal growth of Penicillium rubens on gypsum substrates under controlled dynamic humidity conditions. The effect of a single period of low RH (RH = 50–90%) interrupting favourable conditions (RH = 97%) was tested. It was found that hyphal tips ceased to extend when exposed to any tested decrease in RH. However, new hyphal growth always emerges, seemingly from the old mycelium, suggesting that this indoor fungus does not rely only on conidia to survive the humidity patterns considered. These findings are a fundamental step in unravelling the effect of RH on indoor fungal growth.  相似文献

The industrialization of traditional processes relies on the scientific ability to understand the empirical evidence associated with traditional knowledge. Cork manufacturing includes one operation known as stabilization, where humid cork slabs are extensively colonized by fungi. The implications of fungal growth on the chemical quality of cork through the analysis of putative fungal metabolites have already been investigated. However, the effect of fungal growth on the mechanical properties of cork remains unexplored. This study investigated the effect of cork colonization on the integrity of the cork cell walls and their mechanical performance. Fungal colonization of cork by Chrysonilia sitophila, Mucor plumbeus Penicillium glabrum, P. olsonii, and Trichoderma longibrachiatum was investigated by microscopy. Growth occurred primarily on the surface of the cork pieces, but mycelium extended deeper into the cork layers, mostly via lenticular channels and by hyphal penetration of the cork cell wall.In this first report on cork decay in which specific correlation between fungal colonization and mechanical proprieties of the cork has been investigated, all colonizing fungi except C. sitophila, reduced cork strength, markedly altering its viscoelastic behaviour and reducing its Young's modulus.  相似文献

毛乌素沙地克隆植物沙鞭生长对AM真菌生态分布的影响   总被引:1,自引:0,他引:1  
从毛乌素沙地沙鞭群落间隔空地沙鞭根状茎延伸方向设置样地,2007年5月、7月和10月在0-10、10-20、20-30、30-40和40-50 cm共5个土层分别采集土壤样品,研究了AM真菌时空分布及沙鞭侵入间隔空地前后对AM真菌分布的影响。结果表明,沙鞭能与AM真菌形成良好共生关系,菌根结构为中间型(I-型),AM真菌定殖率和孢子密度有明显的时空分布规律,并与土壤因子密切相关。随采样时间后延AM真菌定殖率先增后降,最大值出现在7月份;孢子密度先降后增,最大值出现在10月份。土壤采样深度对AM真菌定殖和孢子密度有显著影响,AM真菌最高定殖率和最大孢子密度均出现在0-20 cm浅土层。孢子密度仅与泡囊定殖率显著正相关。土壤速效N与菌丝和总定殖率极显著负相关;土壤温度与泡囊、丛枝、菌丝和总定殖率显著和极显著正相关,与孢子密度显著负相关;土壤湿度与丛枝、菌丝和总定殖率显著正相关,与孢子密度显著负相关。克隆植物沙鞭的生长对土壤AM真菌活动和分布有很大影响,沙鞭入侵前后样地中AM真菌孢子密度和定殖率变化显著。  相似文献

The relationships of mycorrhizal fungal respiration and productivity to climate and atmospheric chemistry remain under characterized. We quantified mycorrhizal sporocarp and hyphal respiration, as well as growing season net hyphal production, under ambient and elevated carbon dioxide (CO2) and ozone (O3) in relation to natural temperature and moisture variation. Hyphal respiration did not respond significantly to elevated CO2 and O3. Sporocarp respiration was affected by temperature and moisture content while hyphal respiratory response to temperature was undetected over the narrower range of soil temperatures captured. Hyphal respiration comprised 31 % of soil respiration, and the ratio of hyphal respiration to soil respiration declined with elevated CO2. Hyphal biomass was reduced under all treatments though not statistically significant. Given the large fraction of soil respiration represented by mycorrhizal fungi and its sensitivity to climate, a small change in fungal respiration could strongly affect carbon budgets and cycling under climate change.  相似文献

Secondary air filters in the air-handling units on four floors of a multi-story office building with a history of fungal colonization of insulation within the air distribution system were examined for the presence of growing fungi and production of volatile organic compounds. Fungal mycelium and conidia of Cladosporium and Penicillium spp. were observed on insulation from all floors and both sides of the air filters from one floor. Lower concentrations of volatile organics were released from air filter medium colonized with fungi as compared with noncolonized filter medium. However, the volatiles from the colonized filter medium included fungal metabolites such as acetone and a carbonyl sulfide-like compound that were not released from noncolonized filter medium. Biological control of Botrytis cinerea by Clonostachys rosea is an alternative to chemical control of rose Botrytis blight in greenhouses. Environmental conditions affect the colonization of senescing and dead tissues by both fungi. The contribution of microclimatic variables to debris colonization/sporulation by both fungi was estimated by path coefficient analysis. We monitored daily values of: maximum, average, and minimum temperatures (T max, T ave, and T min), and relative humidity (RHmax, RHave, and RHmin); accumulated rainfall; vapour pressure deficit average; hours with RH?>?90% (RH90); and average temperature during RH90 (T ave90). Association of variables accumulated between the first and seventh day before sampling explained colonization/sporulation variation: R 2=0.81–0.86 for B. cinerea and 0.91–0.96 for C. rosea. RHmax and RH90 were the main factors that directly favoured colonization/sporulation of both fungi. Colonization/sporulation negatively correlated with RHmin, T min, and T ave for B. cinerea and T min, T ave, and T ave90 for C. rosea. The antagonist can suppress B. cinerea colonization/sporulation on rose debris under a wide range of environmental conditions.  相似文献

Polyamines are known to strongly stimulate hyphal growth in arbuscular mycorrhizal fungi. The effect of the polyamines putrescine, spermidine and spermine on spore germination, hyphal elongation and branching by the AM fungus Glomus etunicatum was investigated in this study. The effect of spermine on infection and the development of the host and of daughter spores was further investigated using the dual monoaxenic culture system comprised of Gl. etunicatum fungal cultures in Ri T-DNA transformed carrot hairy roots. Spermidine and spermine showed positive effects on germination and all three polyamines significantly promoted hyphal growth. Hyphal branching was also strongly stimulated by treatment with polyamines, such as an increase in the number of branches. Infection during the early stages of the in vitro co-culture life cycle was enhanced in the presence of spermine, and daughter spores appeared at earlier timepoints compared to the control. Our results demonstrate that polyamines stimulate germination and hyphal branching in the early stage of AM fungal colonization. Moreover, results from the investigations conducted in the fungus-root co-culture suggest that polyamines may be involved in establishing the symbiotic relationship between root and fungus.  相似文献

Plots containing Lolium perenne L., Trifolium repens L. or a mixture of both plant species were exposed to elevated atmospheric CO2 (eCO2) for 10 consecutive seasons using free‐air CO2 enrichment technology at ETH Zürich, Switzerland. The CO2 treatment was crossed with a two‐level nitrogen (N) fertilization treatment. In the tenth year, soil samples were collected on three occasions through the growing season to assess the impact of eCO2 and N fertilization on mycorrhizal fungal abundance. Soil moisture content, which varied with harvest date, was linked to the vegetation type and was higher under eCO2. Root weight density was affected by vegetation type: lower for clover, higher for grass. Root weight density was stimulated by eCO2 and decreased by high N fertilization. The percent root length colonized by mycorrhizal fungi was lowest in the clover plots and highest in the grass plots. High N significantly decreased root length colonized. There was no overall effect of eCO2 on root length colonized; however, there was a significant eCO2× N interaction: eCO2 increased root length colonized at high N, but decreased root length colonized at low N. Extraradical mycorrhizal hyphal density was linked to soil moisture content. Extraradical mycorrhizal hyphal density was not affected by eCO2 or high N individually, but as for root length colonized, there was a significant eCO2× N interaction: eCO2 increased extraradical mycorrhizal hyphal density at low N but not at high N. These environmental effects on root colonization and external mycorrhizal hyphae were independent of soil moisture content and root weight density. This field study demonstrated a significant mediating effect of N fertilization on the response of arbuscular mycorrhizal fungi to eCO2 irrespective of any change in root biomass.  相似文献

Summary  In the present study, the influence of Bacillus subtilis JA on arbuscular mycorrhizal fungi (AMF) was evaluated by either pot culture or in vitro conditions, respectively. Under the pot culture conditions, the inoculation of B. subtilis JA decreased the frequency (% F) of the root colonization by indigenous arbuscular mycorrhizal fungi and the shoot dry weight of maize (Zea mays L.), but had no apparent effect on the intensity (% I) of AM fungal root colonization. The unknown volatile emitted from the B. subtilis JA in vitro significantly inhibited spore germination and the hyphal growth in the dual-compartment experiments. Moreover, the data from the direct interaction between B. subtilis JA and Glomus etunicatum showed that soluble antifungal lipopeptides influenced the development of AMF. Therefore, the application of antifungal Bacillus strains should take the compatibility with the indigenous beneficial fungi into consideration.  相似文献

Summary Materials being used or considered for use in space flight were examined for their susceptibility to fungal colonization. The materials included soft goods (clothing) and insulation and fabrication products such as Velcro® attachments and elastic cord binders. Materials were exposed for at least 28 days in a highhumidity chamber colonized with over 50 species of fungi, including those species recommended for determining recalcitrance of materials to fungal biodegradation. At least nine of 25 products demonstrated extensive microscopic colonization by fungi, mostly byAcremonium obclavatum. Challenge procedures that rely on observations with the unaided eye, or 40×magnification of growth by a restricted number of fungal species with a cellulosic substrate as a positive control, are insufficient for determining the resistance of synthetic substrates to fungal colonization.  相似文献

于陕北黄土沟壑采煤沉陷区内布设试验小区,对黄花菜(Hemerocallis citrina Baroni)接种丛枝菌根真菌(arbuscular mycorrhizal fungi,AMF)—摩西管柄囊霉菌,通过测定黄花菜光合性能、植株生长、抗逆性、土壤养分含量、根际微生物数量等,揭示AM真菌对黄花菜生长和土壤养分的影响。结果表明,黄花菜种植3—5个月后,接种AM真菌显著提高了黄花菜株高、冠幅及其根系菌根侵染率、菌丝密度。与不接种对照区相比,接种AM真菌后黄花菜叶片的光合速率、可溶性糖含量和过氧化氢酶活性分别提高了51%、12%、79%。接种AM真菌处理区黄花菜根际土壤的电导率、有机质、碱解氮和速效钾含量等均显著高于对照区,细菌数量和磷酸酶活性的菌根贡献率分别达77%和24%。表明采煤沉陷区扰动土壤接种AM真菌具有增强土壤微生物活性、改善土壤肥力和提高黄花菜植株抗逆性的作用,对促进陕北黄土沟壑采煤沉陷区经济作物生长和提高土壤质量具有重要现实生态意义。  相似文献

Striatin family proteins are key regulators in signalling pathways in fungi and animals. These scaffold proteins contain four conserved domains: a caveolin‐binding domain, a coiled‐coil motif and a calmodulin‐binding domain at the N‐terminus, and a WD‐repeat domain at the C‐terminus. Fungal striatin orthologues are associated with sexual development, hyphal growth and plant pathogenesis. In Fusarium verticillioides, the striatin orthologue Fsr1 promotes virulence in the maize stalk. The relationship between fungal striatins and pathogenicity remains largely unexplored. In this study, we demonstrate that the Colletotrichum graminicola striatin orthologue Str1 is required for full stalk rot and leaf blight virulence in maize. Pathogenicity assays show that the striatin mutant strain (Δstr1) produces functional appressoria, but infection and colonization are attenuated. Additional phenotypes of the Δstr1 mutant include reduced radial growth and compromised hyphal fusion. In comparison with the wild‐type, Δstr1 also shows a defect in sexual development and produces fewer and shorter conidia. Together with the fact that F. verticillioides fsr1 can complement Δstr1, our results indicate that C. graminicola Str1 shares five phenotypes with striatin orthologues in other fungal species: hyphal growth, hyphal fusion, conidiation, sexual development and virulence. We propose that fungal striatins, like mammalian striatins, act as scaffolding molecules that cross‐link multiple signal transduction pathways.  相似文献

We investigated the colonization and diversity of arbuscular mycorrhizal (AM) fungi associated with 24 moss species belonging to 16 families in China. AM fungal structures, i.e. spores, vesicles, hyphal coils (including intracellular hyphae), or intercellular nonseptate hyphae, were found in 21 moss species. AM fungal structures (vesicles, hyphal coils, and intercellular nonseptate hyphae) were present in tissues of 14 moss species, and spores and nonseptate hyphae on the surface of gametophytes occurred in 15 species. AM fungal structures were present in 11 of the 12 saxicolous moss species and in six of the ten terricolous moss species, but absent in two epixylous moss species. AM fungal structures were only observed in moss stem and leaf tissues, but not in rhizoids. A total of 15 AM fungal taxa were isolated based on trap culture with clover, using 13 moss species as inocula. Of these AM fungi, 11 belonged to Glomus, two to Acaulospora, one to Gigaspora, and one to Paraglomus. Our results suggest that AM fungal structures commonly occur in most mosses and that diverse AM fungi, particularly Glomus species, are associated with mosses.  相似文献

Chaetomium globosum Kunze:Fr is a dermatophytic, dematiaceous fungus that is ubiquitous in soils, grows readily on cellulolytic materials, and is commonly found on water-damaged building materials. Chlorate affects nitrogen metabolism in fungi and is used to study compatibility among anamorphic fungi by inducing nit mutants. The effect of chlorate toxicity on C. globosum was investigated by amending a modified malt extract agar (MEA), oat agar, and carboxymethyl cellulose agar (CMC) with various levels of potassium chlorate (KClO3). C. globosum perithecia production was almost completely inhibited (90–100?%) at low levels of KClO3 (0.1?mM) in amended MEA. Inhibition of perithecia production was also observed on oat agar and CMC at 1?and 10?mM, respectively. However, hyphal growth in MEA was only inhibited 20?% by 0.1–100?mM KClO3 concentrations. Hyphal growth was never completely inhibited at the highest levels tested (200?mM). Higher levels of KClO3 were needed on gypsum board to inhibit perithecia synthesis. In additional experiments, KClO3 did not inhibit C. globosum, Fusarium oxysporum, Aspergillus niger, Penicillum expansum, and airborne fungal spore germination. The various fungal spores were not inhibited by KClO3 at 1–100?mM levels. These results suggest that C. globosum perithecia synthesis is more sensitive to chlorate toxicity than are hyphal growth and spore germination. This research provides basic information that furthers our understanding about perithecia formation and may help in developing control methods for fungal growth on building materials.  相似文献

 The growth and mineral nutrition responses of seedlings of two provenances of Afzelia africana Sm. from Senegal and Burkina Faso, inoculated with four ectomycorrhizal (ECM) fungi (Scleroderma spp. and an unidentified isolate) from the same regions were assessed in a pot experiment in a savanna ECM-propagule-free soil deficient in NPK. There was little variation in the ability of the different fungal species to colonize roots of either provenance of A. africana or to produce external hyphal in soil. Root colonization by ECM fungi and their hyphal development were not related to mineral nutrition or ECM dependency. Differences in P, N, Mg and Ca concentrations in the leaves of inoculated and non-inoculated Afzelia seedlings were not always associated with production of biomass. Only leaf K concentration increased in both provenances after ECM inoculation. However, the Burkina Faso provenance responded better to inoculation with the two fungal isolates than the Senegal provenance in terms of biomass production. This was due to stimulation of root dry weight of the Burkina Faso provenance. Therefore, the hypothesis arises that non-nutritional rather than nutritional effects explain the contribution of ECM inoculation to the growth of A. africana seedlings. 采用分室培养系统,模拟正常水分和干旱胁迫两种环境条件,探讨不同丛枝菌根真菌(arbuscular mycorrhizal fungi,AMF)对紫花苜蓿(Medicago sativa L.)生长和土壤水稳性团聚体的影响.试验条件下,Glomus intraradices对苜蓿根系的侵染率均显著高于Acaulospora scrobiculata和Diversispora spurcum接种处理.正常水分条件下,供试AM真菌均能显著提高植株生物量及磷浓度.干旱胁迫显著抑制了植株生长和菌根共生体发育,总体上菌根共生体对植株生长没有明显影响,接种D.spurcum甚至趋于降低植株生物量;同时,仅有G.intraradices显著提高了植株磷浓度.AM真菌主要影响到>2mm的水稳性团聚体数量,以G.intraradices作用效果最为显著.在菌丝室中,G.intraradices显著提高了总球囊霉素含量.研究表明AM真菌对土壤大团聚体形成具有积极作用,而菌根效应因土壤水分条件和不同菌种而异,干旱胁迫下仅有G.intraradices对土壤结构和植物生长表现出显著积极作用.在应用菌根技术治理退化土壤时,需要选用抗逆性强共生效率高的菌株,对于不同AM真菌抗逆性差异的生物学与遗传学基础尚需进一步研究.  相似文献

Natural gypsum (NG) and phosphogypsum (PG) were tested for resistance to fungal growth based on standard test ASTM D 3273-86, with the recommended mixture of three fungal species, and using the same test modified by the use of a Cladosporium sp. A, isolated from a gypsum plaster ceiling. In the standard test little growth occurred on any of the test specimens. However, abundant fungal growth was produced by the Cladosporium sp. A on phosphogypsum, which was much more susceptible than natural gypsum. Phosphogypsum heated to 600°C to destroy organic residues was resistant to growth of Cladosporium sp. A, as well as other fungi isolated from phosphogypsum panels stored in the environment for 2 years: Cladosporium sp. B, Aspergillus niger and Trichoderma sp. Phosphogypsum moulded in Petri dishes was susceptible to growth of a wide range of fungi, although Fusarium sp. and Rhizopus sp. caused practically no discoloration of the substrate. Six biocides were separately incorporated into the phosphogypsum at concentrations recommended by the suppliers and test specimens incubated on Sabouraud agar inoculated with various fungal isolates. The biocide 2-N-octyl-4-isothiazolin-3-one was the most efficient compound. It prevented the growth of the fungi most likely to cause health problems in buildings, but not that of Helminthosporium sp., isolated from powdered phosphogypsum in the factory. This was the most resistant fungus showing growth on all biocide-containing specimens.  相似文献

Abstract

Advances on plant–fungal interactions reveal that root symbiotic fungi actively modulate host growth, resistance response and secondary metabolism. Artemisia annua has been widely recognized as an important medicinal plant for artemisinin production, yet little is known about the fungal consortium associated with roots of A. annua. In this article, microscopic and culture-dependant methods were used to evaluate the identity and taxonomic affinities of root symbiotic fungi. Morphological evidence confirmed that arbuscular mycorrhizal fungi were dominant fungal group in naturally regenerated roots, but low colonization frequency in planted roots. Dark septate endophytes (DSEs) were easily found, which were characterized with dark pigmented hypha and a sclerotium-like structure in root cortex, and other endophytic fungi also occurred. A total of 36 isolates were recovered. Combined morphological and molecular identification (based on ITS sequences) determined 21 fungal taxa (genotype), which were placed into numerous lineages of Ascomycota. The best BLAST match indicated that almost half of total taxa were closely related to undescribed fungi, some of them may act as novel DSEs but experimental data were warranted. Interestingly, remarkable difference of fungal community associated with two types of roots was examined and no culturable fungi overlapped. Our findings provide some additional evidence that DSEs and other root endophytes may be as common as mycorrhizal fungi. Recovered fungi as raw materials for bioassay of endophytes-mediated promotion of artemisinin content in A. annua will be conducted in further research.  相似文献

Phytoremediation is the use of selected plants to decontaminate polluted environments. Arbuscular mycorrhizal fungi (AMF) may potentially be useful for phytoremediation, but it is not known how petroleum hydrocarbons influence AMF spore germination and hyphal growth. To address this question, germination of spores and germ tube growth of Glomus intraradices Schenck and Smith and Glomus aggregatum Schenck and Smith were assessed in soil contaminated with up to 3% (w/v) of F2 diesel oil or HAGO reference oil. Hyphal growth, colonization and progeny spore production were assessed in vitro using transformed root cultures of Daucus carota and G. intraradices spores in a F2 diesel contaminated medium. In addition, extraradical hyphal growth of G. intraradices colonizing Daucus carota in the presence of F2 diesel was studied. Neither F2 diesel nor HAGO reference oil affected spore germination or germ tube growth in soil. However, in the presence of plant roots, germ tube growth of G. intraradices was reduced and delayed in the presence of F2 diesel and root colonization was not detected. Hyphal growth of pre-colonized carrot roots by G. intraradices was reduced and delayed in F2 contaminated medium compared to controls. F2 diesel did not inhibit spore germination of these AMF species but did reduce colonization, germ tube and hyphal growth. These results suggest that AMF inoculum can be established in petroleum-contaminated sites. However, it may prove beneficial to plant pre-colonized plants to increase the probability of sufficient AMF colonization and growth. The likely mechanism(s) of petroleum toxicity in this plant-microbe system was discussed.  相似文献

We examined the effect of two levels of soil pH (5 and 6) on the ability (effectiveness) of ectomycorrhizal fungi to increase the growth of Eucalyptus globulus Labill. at a deficient supply of P. Plants were inoculated with one of six fungal isolates [Laccaria laccata (Scop. ex Fr.) Berk. and Br. (isolates A and B), Pisolithus tinctorius (Pers.) Coker and Couch (isolates A and B), Descolea maculata Bough. and Mal. and Setchelliogaster sp. nov.] and were grown in a P-deficient sand, in pots, in a temperature-controlled glasshouse. Seedlings were harvested 89 days after planting and were assessed for dry matter production, tissue P concentrations, ectomycorrhizal colonization of roots and hyphal development in soil.Uninoculated plants had less than 5% of their fine root length colonized by ectomycorrhizal fungi. In contrast, inoculated plants had 30% or greater of their fine root length ectomycorrhizal. Inoculation increased the uptake of P and growth of plants for all isolates and at both levels of soil pH, although growth responses to inoculation were greater at pH 6, particularly for the two L. laccata isolates. Isolates which colonized roots most extensively increased plant growth to the greatest extent. D. maculata was the most effective fungal isolate at pH 5, and both D. maculata and L. laccata A were most effective at pH 6. The effects of soil pH on plant growth were also related to some extent to the effects of soil pH on colonized root length. Growth responses to inoculation were related less well to hyphal development in soil. The L. laccata isolates formed more hyphae in soil (on a per pot, per m of fine root, and per m of colonized fine root basis) than other fungal isolates, but were not always more effective in increasing plant grown.  相似文献
