In cattle, a gregarious species, the social group influences individual stress responses to fear-eliciting situations. As handling can be stressful for farm animals, it can be hypothesised that social partners modify individual responses to handling. The present experiment investigated the effect of the presence or absence of social partners on behavioural reactions of beef calves in a handling test. At the age of 10 months, 38 calves from two breeds (Salers and Limousine) were individually subjected to the docility test, once while in visual contact with four familiar peers, and once in the absence of peers, following a crossover design. The docility test procedure included physical separation from peers (30 s; period 1), exposition to a stationary human (30 s; period 2), and handling by human (30 s-2.5 min, according to the success in handling; period 3). In absence of human (period 1), calves in visual contact with their peers spent more time motionless than when peers were totally absent (P<0.001). The social environment also influenced the duration of handling (period 3); the human required more time to successfully handle calves when peers were present (P<0.05). In conclusion, the presence of peers affects individual calves' reactions to the docility test.  相似文献

The nitrogen uptake and growth capabilities of the potentially harmful, raphidophycean flagellate Heterosigma akashiwo (Hada) Sournia were examined in unialgal batch cultures (strain CCMP 1912). Growth rates as a function of three nitrogen substrates (ammonium, nitrate and urea) were determined at saturating and sub-saturating photosynthetic photon flux densities (PPFDs). At saturating PPFD (110 μE m−2 s−1), the growth rate of H. akashiwo was slightly greater for cells grown on NH4+ (0.89 d−1) compared to cells grown on NO3 or urea, which had identical growth rates (0.82 d−1). At sub-saturating PPFD (40 μE m−2 s−1), both urea- and NH4+-grown cells grew faster than NO3-grown cells (0.61, 0.57 and 0.46 d−1, respectively). The N uptake kinetic parameters were investigated using exponentially growing batch cultures of H. akashiwo and the 15N-tracer technique. Maximum specific uptake rates (Vmax) for unialgal cultures grown at 15 °C and saturating PPFD (110 μE m−2 s−1) were 28.0, 18.0 and 2.89 × 10−3 h−1 for NH4+, NO3 and urea, respectively. The traditional measure of nutrient affinity—the half saturation constants (Ks) were similar for NH4+ and NO3 (1.44 and 1.47 μg-at N L−1), but substantially lower for urea (0.42 μg-at N L−1). Whereas the α parameter (α = Vmax/Ks), which is considered a more robust indicator for substrate affinity when substrate concentrations are low (<Ks), were 19.4, 12.2 and 6.88 × 10−3 h−1/(μg-at N L−1) for NH4+, NO3 and urea, respectively. These laboratory results demonstrate that at both saturating and sub-saturating N concentrations, N uptake preference follows the order: NH4+ > NO3 > urea, and suggests that natural blooms of H. akashiwo may be initiated or maintained by any of the three nitrogen substrates examined.  相似文献

Background  

Sixteen, spring-born, single suckled, castrated male calves of Limousin × Holstein-Friesian and Simmental × Holstein-Friesian dams respectively, were used to investigate the effect of weaning on total leukocyte and differential counts, neutrophil functional activity, lymphocyte immunophenotypes, and acute phase protein response. Calves grazed with their dams until the end of the grazing season when they were housed in a slatted floor shed. On the day of housing, calves were assigned to a treatment, (i) abruptly weaned (W: n = 8) or (ii) non-weaned (controls) (C: n = 8). Weaned calves were housed in pens without their dams, whereas non-weaned (control) calves were housed with their dams. Blood was collected on day -7, 0 (housing), 2, 7, and 14 to determine total leukocyte and differential counts and concentration of fibrinogen and haptoglobin. Lymphocyte immunophenotypes were characterised using selected surface antigens (CD4+, CD8+, WC1+ (γδ T cells), MHC Class II+ lymphocytes), and the functional activities of neutrophils (surface expression of L-selectin (CD62L), phagocytic and oxidative burst activity) were investigated using flow cytometry.  相似文献

Tactile contact is often used to improve the human–foal relationship, in particular during sensitive periods such as weaning. However, the method used to provide the contact (forced or unforced) may affect subsequent reactions to humans. The aim of our study was to compare the effect of forced and unforced handling at weaning on horses’ behaviour towards humans and handling.A total of 23 Anglo-Arabian foals received individual human contact at weaning. Constrained-handled foals (forced human contact; FC, n = 8) were stroked while being restrained (i.e., held by the halter without being led) so that human contact was forced. Unconstrained-handled foals (unforced human contact; UC, n = 7) were stroked without any restraint and could thus avoid contact if they so wanted. Each individual was handled 5 min twice a day for 14 consecutive days. Control foals were not handled (no contact; NC, n = 8). Foals’ reactions towards a human (e.g. proximity and contact seeking), and their manageability (fitting of a halter) were then assessed. The animals from the different treatments were then mixed and tested again 4 months later. Non-parametric Mann–Whitney U-tests were used to compare experimental groups.Only forced human contact reduced fear reactions toward humans (e.g. shorter latency to approach a passive human, P < 0.01; shorter latency to touch the foal, P < 0.05) in the short-term (immediately after the handling sessions) and increased foals’ manageability in a familiar environment (shorter latency to be placed a halter on and less defences displayed, P < 0.05). It had no effect in an unfamiliar environment. These effects did not last longer than 4 months.Forced human contact thus appears to be more efficient in improving human–horse relationships and subsequent handling than unforced human contact. However, the lack of effect on reactivity towards humans in an unfamiliar environment, and the lack of long-term effects suggest that this method needs to be improved to be more effective. Some of our results suggest that horses could perceive human contact (i.e., stroking) as positive, however further studies are needed to confirm this hypothesis.  相似文献

Negative emotional states in humans are associated with a negative (pessimistic) response bias towards ambiguous cues in judgement tasks. Every mammalian young is eventually weaned; this period of increasing nutritional and social independence from the dam is associated with a pronounced behavioural response, especially when weaning is abrupt as commonly occurs in farm animals. The aim of the current study was to test the effect of separation from the cow on the responses of dairy calves in a judgement task. Thirteen Holstein calves were reared with their dams and trained to discriminate between red and white colours displayed on a computer monitor. These colours predicted reward or punishment outcomes using a go/no-go task. A reward was provided when calves approached the white screen and calves were punished with a timeout when they approached the red screen. Calves were then tested with non-reinforced ambiguous probes (screen colours intermediate to the two training colours). “GO” responses to these probes averaged (± SE) 72±3.6 % before separation but declined to 62±3.6 % after separation from the dam. This bias was similar to that shown by calves experiencing pain in the hours after hot-iron dehorning. These results provide the first evidence of a pessimistic judgement bias in animals following maternal separation and are indicative of low mood.  相似文献

Weaning of beef calves is a stressful event that negatively impacts health and performance. A variety of interventions have been proposed to reduce stress and improve gains following weaning. This study used 288 7- to 8-month-old calves from two separate locations, to examine four different weaning strategies, as well as the impact of shipment. Calves were blocked by weight and sex, and then randomly assigned to one of four treatments: abrupt weaning (AW), where calves were separated from the dam on day 0 (D0) and allowed no further contact with the dam; fence line (FL), where calves were weaned on D0 but had fence line contact with dams for 7 days; nose flap (NF), where on day -6 calves received a nose flap that interferes with suckling, then had the flap removed and were weaned from the dam on D0; and intermittent separation (SEP), where calves were removed from dams for 24-h intervals on day -13 and day -6, then weaned on D0, but allowed fence line contact with the dam for 7 days. Each treatment group was further divided into two subgroups, one of which was shipped early (D0 for AW, day 7 for others) or shipped later (day 28). Body weight and sickness were recorded for all groups. Results showed a negative impact on gain for early shipping compared to later shipping, and poorer gain in AW calves than most other treatments. Results of the analyses of morbidity were inconclusive. This study found that delayed shipment following FL weaning improves performance under common management conditions for the US cow–calf industry.  相似文献

Seven innate immune parameters were investigated in 64 full-sib families (the offspring of 64 sires and 45 dams) from two year-classes of farmed rohu carp (Labeo rohita). Survival rates were also available from Aeromonas hydrophila infection (aeromoniasis) recorded in controlled challenge tests on a different sample of individuals from the same families. Due to strong confounding between the animal additive genetic effect and the family effects (common environmental + non-additive genetic), reliable additive (co)variance components and hence heritabilities and genetic correlations could not be obtained for the investigated parameters. Therefore, estimates of the association of challenge test survival with the studied immune parameters were obtained as product moment correlations between family least square means. These correlations revealed statistically significant (p < 0.05) negative correlations of survival with bacterial agglutination titre (−0.48), serum haemolysin titre (−0.29) and haemagglutination titre (−0.34); and significant positive correlation with ceruloplasmin level (0.51). The correlations of survival to aeromoniasis with myeloperoxidase activity, superoxide production and lysozyme activity were found to be not significantly different from zero (p > 0.05). Assuming that the negatively correlated candidate traits are not favourable as indirect selection criteria, the results suggest that ceruloplasmin level could potentially be a marker for resistance to aeromoniasis in rohu. The use of this immune parameter as an indirect selection criterion for increased resistance to aeromoniasis in rohu will, however, require that the parameter shows significant additive genetic variation and a significant genetic correlation with survival. Further studies are therefore needed to obtain a reliable heritability estimate for ceruloplasmin and its genetic correlation with survival from aeromoniasis.  相似文献

The effect of handling and hand-feeding of calves with or without their dams present on the calves’ subsequent response to humans was investigated using 5 groups of 10 Danish Friesian calves, each of which received a different treatment. The treatments were: calf separated from its dam immediately after birth, housed in a single pen and handled (group SH) or not handled (group S) calf adjacent to cow and handled (group CH) or not handled (C) for the first 4 days after calving. The fifth group was housed together with their dams for the first 4 days of life and then moved to single pens and handled there for the next 4 days (group C-SH). The handling treatment was carried out three times daily for 4 days and consisted of 6 min of hand-feeding with milk from a teat-bucket and patting, stroking and talking to the calf. After separation, all calves were housed in single pens. Human contact was minimised except during the treatment periods.

The approach behaviour of each calf to an unknown person was tested at days 20, 40 and 55 in their home pen. Flight distance was determined at day 55. The test at day 55 was carried out in a large single pen where the calves had been housed for 24 h prior to the test. The latency to interact (in seconds) with the person was significantly shorter (P<0.001) in all three tests for SH (13±5, 29±7 and 44±8, at days 20, 40 and 55, respectively) and C-SH (28±6, 47±17 and 70±17) compared to S (240±25, 238±33 and 173±5), C (240±28, 202±36 and 167±13) and CH (233±36, 271±29 and 128±19). The results of the latter three groups did not differ significantly. Treatment also affected the position and the orientation of the calf in the pen during the tests. Calves in SH and C-SH stood at the front of the pen and faced the person more often than calves in S, C and CH. Again, there were no significant differences between the results of the latter three groups.

The results indicate that the presence of the dam limits the effect of the handling treatment on young calves’ motivation to interact with humans. It is likely that primary socialization occurs with the dam, preventing a secondary socialization developing with humans, until the calf is isolated from the dam.  相似文献

The kinetic properties of a microsomal gill (Na+,K+)-ATPase from the freshwater shrimp, Macrobrachium olfersii, acclimated to 21‰ salinity for 10 days were investigated using the substrate p-nitrophenylphosphate. The enzyme hydrolyzed this substrate obeying cooperative kinetics at a rate of 123.6 ± 4.9 U mg− 1 and K0.5 = 1.31 ± 0.05 mmol L− 1. Stimulation of K+-phosphatase activity by magnesium (Vmax = 125.3 ± 7.5 U mg− 1; K0.5 = 2.09 ± 0.06 mmol L− 1), potassium (Vmax = 134.2 ± 6.7 U mg− 1; K0.5 = 1.33 ± 0.06 mmol L− 1) and ammonium ions (Vmax = 130.1 ± 5.9 U mg− 1; K0.5 = 11.4 ± 0.5 mmol L− 1) was also cooperative. While orthovanadate abolished p-nitrophenylphosphatase activity, ouabain inhibition reached 80% (KI = 304.9 ± 18.3 μmol L− 1). The kinetic parameters estimated differ significantly from those for freshwater-acclimated shrimps, suggesting expression of different isoenzymes during salinity adaptation. Despite the ≈2-fold reduction in K+-phosphatase specific activity, Western blotting analysis revealed similar α-subunit expression in gill tissue from shrimps acclimated to 21‰ salinity or fresh water, although expression of phosphate-hydrolyzing enzymes other than (Na+,K+)-ATPase was stimulated by high salinity acclimation.  相似文献

In this study, two total mixed rations (TMR, based on dry ingredients) consisting (per kg dry matter (DM)) of 300 or 400 g finely chopped hay mixture of grass and alfalfa (H30 versus H40) were compared concerning their effects on dry matter intake (DMI), ruminal fermentation patterns and chewing activity of early-weaned (8 weeks milk-fed) calves. Ten ruminally cannulated male German Holstein calves were randomly assigned to two dietary treatments (n = 5) and observed from an age of 8–15 weeks. One group received the H30 (11.3 MJ metabolizable energy (ME)/kg DM) and the other the H40 (10.7 MJ ME/kg DM) TMR. All calves received grass hay (9.0 MJ ME/kg DM) separately. Water, TMR and hay were offered ad libitum twice daily (08:00 and 16:00 h). Rumen fluid was collected via cannula at an age of 9, 11, 13 and 15 weeks, twice weekly just prior to as well as 1, 3, 5 and 7 h after morning feeding. Chewing activity was recorded by a special head collar. As the calves aged DMI increased rapidly congruent with the recommended range for weaned calves. Because of the differing energy supply, calves receiving the H30 TMR were heavier than calves receiving the H40 TMR (139 kg versus 123 kg, P=0.007). During the trial ruminal pH of all calves were within the target range (6.2 ± 0.5), indicating physiological ruminal fermentation patterns. Daily mean ruminal pH was uninfluenced by treatment, however at an age of 13 and 15 weeks H30 showed a higher short chain fatty acid (SCFA) level than H40 (P=0.098; P=0.036). At an age of 15 weeks H30 showed a critical decrease in ruminal pH (3 h after feeding: 5.7) corresponding to a higher ruminal SCFA concentration (148.2 mmol/L, P=0.007). Chewing activity was well developed at an early age due to an increasing DMI after weaning. At an age of 15 weeks chewing activity (per day: 613–743 min total chewing; 358–418 min rumination) was similar to that of adult cows. In summary, feeding a dry TMR consisting per kg DM of 300–400 g hay to early-weaned calves can be recommended for a successful calf rearing up to an age of 15 weeks.  相似文献

This experiment was designed to study the effects of avoiding calf contact and genotype on the metabolic, behavioural and reproductive traits of beef cows during lactation. Fifty-two multiparous cows, 25 Parda de Montaña (PA) and 27 Pirenaica (PI), fed at maintenance were assigned from the day after calving to twice-daily nursing (2× 30-min sessions at 0800 and 1600 h) either with fence contact with their calves (partial contact, PC) or without visual, tactile and olfactory contact (non-contact, NC) between suckling periods. Blood samples were collected to analyse metabolites (triglycerides, cholesterol, non-esterified fatty acids (NEFA), β-hydroxybutyrate and urea) and progesterone at different intervals. Cow–calf behaviour was monitored on weeks 4, 9 and 15 of lactation. Cow activity at oestrus was recorded through collars. Cows from both treatments and breeds showed similar live-weight gains during the first three months post-partum (P > 0.10). Milk yield and calf gains were not affected by treatment (P > 0.10) but they were greater in PA than in PI (P < 0.05). Plasma triglycerides and urea in the cows were not affected either by calf contact, breed or week post-partum (P > 0.10). Plasma cholesterol increased from week 6 post-partum onwards in PA cows (P < 0.05) while this rise was delayed to week 7 of lactation in PI breed (P < 0.05). Plasma NEFA was greater in blood from PC–PA cows than in the rest of groups (P < 0.05), and these metabolites were greater on week 1 and lower on week 11 than in rest of samplings (P < 0.05). Plasma β-hydroxybutyrate was not affected by either calf contact or breed (P > 0.10), but it was greatest on weeks 1–3 than in the rest of lactation (P < 0.05). Cows with PC calves took less time to first contact after they entered the barn than their NC counterparts (P < 0.05). Dams from both contact treatments nursed their offspring and remained in close proximity for similar time within suckling periods (P > 0.10). PA cows devoted more time than PI ones to lick their young (P < 0.001). There were no differences throughout lactation in any of the studied maternal behaviours (P > 0.10). Calf contact and breed did not affect the interval to first post-partum ovulation or oestrus in these cows (P > 0.10). Under twice-daily nursing conditions, the limitation of visual, tactile and olfactory contact with calves did not trigger different maternal or reproductive traits in these breeds but only a slightly higher mobilisation of body fat substrates in Parda de Montaña compared to Pirenaica.  相似文献

Low temperatures increase mortality of calves but we know little of calves’ abilities to thermoregulate. To examine effects of milk feeding level on thermoregulatory behaviour, 27 Holstein calves were housed for 3 d after birth in individual pens fitted with heat lamps at one end to provide a thermal gradient. They were fed milk at a high (30% BW) or low (8% BW) daily allowance. Daily temperatures in the barn were 6.27 ± 3.12 °C, range: −5.46 °C to 16.63 °C. Temperature loggers were fitted to each calf to record ambient temperatures chosen by the calf. Video cameras recorded the calf's distance from the heat lamp and lying posture (on sternum or side, with legs extended or contracted) for 24 h/d, and records were scored every 20 min. Calves spent most time in the portion of the pen with heat lamps (Proc Mixed P < 0.05) but there was no effect of feeding level (P > 0.10). Calves tended to be closer to the heat lamp during the coldest periods of the day than during the warmer periods (P < 0.10). The difference between the temperature recorded in the barn and the temperature recorded on the calf's back was correlated with distance from the heat lamp (r = 0.32, P = 0.01) showing that the temperature logger on the calf reflected the ambient temperature chosen. Time spent in different lying postures was not affected by feeding level or barn temperature (P > 0.10). Young calves show a preference for warmer environments but this preference does not interact with the amount of milk fed. Temperature recorders attached to the calves can measure calves’ thermal preferences. Calves did not thermoregulate through changes in resting posture.  相似文献

Failed transfer of passive immunity (FTPI) in dairy calves – which is often due to the low amount of colostrum provided within a few hours after birth – remains a crucial issue. Enabling dairy calves to nurse colostrum from their dams could be useful in increasing intake and thus avoiding FTPI, but further potential effects on the health and welfare of both calves and dams should also be considered. In this study, 107 calf-dam pairs from two Italian dairy farms were alternately assigned to one of the following colostrum provision methods (CPMs): ‘hand-fed method’ (HFM) – the calf was separated from the dam immediately after birth and colostrum was provided by nipple-bottle (n = 50); ‘nursing method’ (NM) – the calf nursed colostrum from the dam for the first 12 h of life without farmer assistance (n = 30); and ‘mixed method’ (MM) – the nursing calf received a supplementary colostrum meal by nipple-bottle (n = 27). Serum of calves (1 to 5 days of age) and samples of their first colostrum meal were analysed by electrophoresis to assess immunoglobulin (Ig) concentration. Additionally, behavioural indicators of separation distress (calf and dam vocalisations; calf refusal of the first meal after separation; undesirable dam behaviour at milking) in the following 24 h were recorded as binary variables (Yes/No), and the health status of calves (disease occurrence and mortality) and dams (postpartum disorders and mastitis occurrence) were monitored for the first 3 months of life and 7 days after parturition, respectively. The lowest FTPI occurrence (calf serum Ig concentration <10.0 g/l) was found in the MM (11.1%) and the HFM (22.0%) compared with the NM (60.0%) (P<0.05), and the highest percentage of calves with optimal transfer of passive immunity (serum Ig concentration ≥16.0 g/l) was observed in the MM (55.6%). The lowest calf–dam separation distress was observed in the HFM (P<0.05). The highest calf disease occurrence was recorded in the HFM (64.0%) and the lowest in the NM (33.3%), with an intermediate value for the MM (44.4%) (P<0.05). No effect of the CPM was observed on dam health or calf mortality (P>0.05). The results of this study indicated that providing calves with a supplementary colostrum meal in addition to nursing from the dam (MM) is truly effective in maximizing passive immunity transfer. Anyway, specific strategies should be studied to minimise calf-dam separation distress.  相似文献

Clonal cultures of Alexandrium species collected from a shrimp pond on the northern coast of Vietnam were established and morphologically identified as Alexandrium minutum. Nucleotide sequences of domains 1 and 2 of the large subunit ribosomal (LSU) rRNA gene showed high sequence similarity to A. minutum isolates from Malaysia. Paralytic shellfish toxin profile of the clones was characterized by the dominance of GTX4, GTX1, and NEO. GTX3, GTX2, and dcSTX were also present in trace amount. Toxin content varied among the strains and growth stages, ranged from 3.0 to 12.5 fmol cell−1. In addition to these known toxin components, a new gonyautoxin derivative was detected by HPLC, eluting between GTX4 and GTX1. The peak of this compound disappeared under non-oxidizing HPLC condition but unchanged either after treated with 0.05 M ammonium phosphate/10% mercaptoethanol or 0.1N HCl hydrolysis. LCMS ion scanning showed a parental ion of [M + H]+ at m/z 396, [M − SO3]+ at m/z 316, and [M − SO4]+ at m/z 298. Based on these results, the derivative was identified as deoxy-GTX4-12ol, and this represents the first report of this toxin analogue.  相似文献

Preference for NH4+ or NO3 nutrition by the perennial legume Sesbania sesban (L.) Merr. was assessed by supplying plants with NH4+ and NO3 alone or mixed at equal concentrations (0.5 mM) in hydroponic culture. In addition, growth responses of S. sesban to NH4+ and NO3 nutrition and the effects on root nodulation and nutrient and mineral composition of the plant tissues were evaluated in a hydroponic setup at a range of external concentration of NH4+ and NO3 (0, 0.1, 0.2, 0.5, 2 and 5 mM). Seedlings of S. sesban grew equally well when supplied with either NH4+ or NO3 alone or mixed and had high relative growth rates (RGRs) ranging between 0.19 and 0.21 d−1. When larger plants of S. sesban were supplied with NH4+ or NO3 alone, the RGRs and shoot elongation rates were not affected by the external concentration of inorganic N. At external N concentrations up to 0.5 mM nodulation occurred and contributed to the N nutrition through fixation of gaseous N2 from the atmosphere. For both NH4+ and NO3-fed plants the N concentration in the plant tissues, particularly water-extractable NO3, increased at high supply concentrations, and concentrations of mineral cations generally decreased. It is concluded that S. sesban can grow without an external inorganic N supply by fixing atmospheric N2 gas via root nodules. Also, S. sesban grows well on both NH4+ and NO3 as the external N source and the plant can tolerate relatively high concentrations of NH4+. This wide ecological amplitude concerning N nutrition makes S. sesban very useful as a N2-fixing fallow crop in N deficient areas and also a candidate species for use in constructed wetland systems for the treatment of NH4+ rich waters.  相似文献

In this study we investigated the ability of Chara intermedia to acclimate to different irradiances (i.e. “low-light” (LL): 20–30 μmol photons m−2 s−1 and “high-light” (HL): 180–200 μmol photons m−2 s−1) and light qualities (white, yellow and green), using morphological, photosynthesis, chlorophyll fluorescence and pigment analysis.Relative growth rates increased with increasing irradiance from 0.016 ± 0.003 (LL) to 0.024 ± 0.005 (HL) g g−1 d−1 fresh weight and were independent of light quality. A growth-based branch orientation towards high-light functioning as a mechanism to protect the plant from excessive light was confirmed. It was shown that the receptor responsible for the morphological reaction is sensitive to blue-light.C. intermedia showed higher oxygen evolution (up to 10.5 (HL) vs. 4.5 (LL) nmol O2 mg Chl−1 s−1), photochemical and energy-dependent Chl fluorescence quenching and a lower Fv/Fm after acclimation to HL. With respect to qP, the acclimation of the photosynthetic apparatus depended on light quality and needed the blue part of the spectrum for full development. In addition, pigment composition was influenced by light and the Chl a/Car and Antheraxanthin (A) + Zeaxanthin (Z)/Violaxanthin (V) + A + Z (DES) ratios revealed the expected acclimation behaviour in favour of carotenoid protection under HL (i.e. decrease of Chl a/Car from 3.41 ± 0.48 to 2.30 ± 0.35 and increase of DES from 0.39 ± 0.05 to 0.87 ± 0.03), while the Chl a/Chl b ratios were not significantly affected. This research describes four X-ray structures of Vibrio harveyi chitinase A and its catalytically inactive mutant (E315M) in the presence and absence of substrates. The overall structure of chitinase A is that of a typical family-18 glycosyl hydrolase comprising three distinct domains: (i) the amino-terminal chitin-binding domain; (ii) the main catalytic (α/β)8 TIM-barrel domain; and (iii) the small (α + β) insertion domain. The catalytic cleft of chitinase A has a long, deep groove, which contains six chitooligosaccharide ring-binding subsites (−4)(−3)(−2)(−1)(+1)(+2). The binding cleft of the ligand-free E315M is partially blocked by the C-terminal (His)6-tag. Structures of E315M-chitooligosaccharide complexes display a linear conformation of pentaNAG, but a bent conformation of hexaNAG. Analysis of the final 2Fo − Fc omit map of E315M-NAG6 reveals the existence of the linear conformation of the hexaNAG at a lower occupancy with respect to the bent conformation. These crystallographic data provide evidence that the interacting sugars undergo conformational changes prior to hydrolysis by the wild-type enzyme.  相似文献

A 42-day experiment was conducted to compare the effects of various levels of sodium selenite (SS) and Se-enriched yeast (SY) on chicken productivity, carcass traits, and breast Se concentration. Six hundred 1-day-old Cobb 500 broiler chicks were placed on 1 of 6 experimental treatments. The treatments consisted of feeding a diet without Se supplementation (basal diet) or basal diet with 0.6 mg/kg supplemented Se supplied by SS, SY, or a mix of the two (0.45 SS + 0.15 SY; 0.3 SS + 0.3 SY; 0.15 SS + 0.45 SY). Chicks in all Se-supplemented treatments had significantly higher final body weight and eviscerated weight than those on the basal diet (P < 0,05) and no significant differences were observed among selenium source (P < 0.05). Also, chicks in all Se-supplemented treatments had significantly higher Se contents in breast tissue than the control group (P < 0.05). Replacing SS by SY in the broiler diets resulted in increased concentrations of Se in the breast (P < 0.01). Strong correlations were found between breast Se concentrations and the level of SY supplementation of the broiler diet (r = 0.992). The results from this experiment indicate that SY is a superior source of selenium for the production of selenized meat, and can be used, without any detrimental effect on chicken performance, for adding nutritional value to broiler meat and thus safely improving human selenium intake.  相似文献

The present study was undertaken to examine the effect of administered recombinant mouse leptin on the recovery of regressed ovary in fasted ducks. Twenty-eight ducks were divided into five groups: fed ad libitum (control; n = 5), fasted control (FC; n = 5), fasted + low dose of leptin (F + L; n = 5), fasted + medium dose of leptin (F + M; n = 5) and fasted + high dose of leptin (F + H; n = 3). All four fasted groups were fasted for 2 days and then ad libitum and the ducks were treated with leptin at doses of 0 (control and FC), 50 (F + L), 250 (F + M) and 1000 (F + H) μg/kg body weight/day on day 3–5. Results showed that a moderate dose of leptin (250 μg/kg body weight/day) injected during the re-feeding period: (i) promoted the recovery of the regressed ovary as evidenced by an increase in ovary weight and recovery of yellow hierarchical follicles; (ii) elevated the plasma 17β-estradiol (E2) level; (iii) increased the mRNA levels of ovary follicle-stimulating hormone receptor (FSHR), luteinizing hormone receptor (LHR) and estrogen receptor-β (ER-β). Furthermore, the results also showed that a high dose of leptin (1000 μg/kg body weight/day) may have a negative effect on the recovery of the regressed ovary. In conclusion, this study indicates that, in ducks, leptin may be involved in the recovery of the regressed ovary caused by 2 days of fasting. This effect may be related to increased plasma E2 levels and stimulation of the mRNA levels of ovarian FSHR, LHR and especially ER-β.  相似文献

Suckling and nutrition are generally recognized as two major factors controlling the duration of the postpartum anovulatory period. In the present study, the effect of premature weaning and suckling restriction with nose plates (NPs) on cow and calf performance was evaluated. The study was conducted over 2 years; primiparous Hereford cows, weighing (mean ± S.E.M.) 344 ± 3.5 kg and with 4.1 ± 0.05 units of body condition score (BCS) (scale 1–8 [Vizcarra, J.A., Ibañez, W., Orcasberro, R., 1986. Repetibilidad y reproductibilidad de dos escalas para estimar la condición corporal de vacas Hereford. Investigaciones Agronómicas 7 (1), 45–47]) at calving, remained with their calves until 72.5 ± 1.2 days postpartum (day 0). They were then assigned to one of three treatments: (i) calves with free access to their dams and ad libitum suckling (S, n = 29); (ii) calves fitted with NPs for 14 days, but remained with their dams (NP, n = 29), and (iii) calves that were weaned from their dams (W, n = 28). All cows were anestrus at the time treatments commenced (day 0). All cows were blood sampled twice weekly from 1 week before the beginning of the experiment until the end of the mating period (day 74) for progesterone analysis. The mating period began on day 14. Cows in W treatment had ovulations earlier (P < 0.05) than those in NP and S groups. Cows in the NP group had longer (P < 0.05) intervals between the first progesterone increase and normal luteal phase than cows in the other two treatments groups (23.3 ± 3.2 vs. 6.5 ± 3.2 and 5.2 ± 3.3 days for NP, S and W cows, respectively). Fifty per cent of the cows with NP had a short cycle (7 days) but there was a group of cows that had longer (P < 0.05) intervals (66 days) between first progesterone increase and normal estrous activity. In the NP group, 8 of 29 cows had a short luteal phase and then a normal one; for 9 of these 29 cows progesterone concentrations remained low for 6 weeks from the beginning of the treatment; and for 12 of these 29 cows progesterone concentrations initially increased after treatment initiation, but these animals became anestrus thereafter. Short-term suckling restriction with NPs led to a variable response in primiparous cows of moderate body condition under range conditions.  相似文献
