Existing techniques for analysing the organization of behavioural events are discussed. Cluster analysis, mutual replaceability, ‘melody’ detection and Markov analyses are examined in order to discover the validity and usefulness of the concepts searched for by each technique, and the mathematical and statistical background of the techniques is scrutinized. Some new techniques, designed to search for and detect possible important sequential organization patterns, are described in the light of the previous criticisms. Of these techniques, the Pre-post-state Histogram (PPSH)—a multi-order Markov graphical analysis—would appear to be the most important for future work, especially when used in conjunction with the other techniques described.  相似文献

A + U-Rich elements (AREs) have been extensively investigated as cis-acting determinants of rapid mRNA turnover. Recently, a number of RNA-binding proteins interacting with AREs have been described. This article presents strategies and techniques used by our laboratory to identify and characterize a family of ARE-binding proteins collectively termed AUF1. However, these techniques may be applied to the study of any protein displaying sequence-specific RNA binding activity. The techniques described here include the purification of native AUF1 from cultured cells as well as the preparation of recombinant AUF1 proteins using a bacterial expression system. Analyses of RNA-protein interactions are also described, including the use of gel mobility shift assays with synthetic RNA probes to monitor specific RNA binding activity in cell extracts or with recombinant proteins. Variations of this technique are also described to evaluate the RNA binding affinity of recombinant proteins and the use of specific RNA competitors to assess RNA determinants of protein binding specificity. Other techniques presented include the identification of specific proteins in RNA:protein complexes using antibody supershifts and the estimation of molecular weights of RNA-binding proteins by UV crosslinking. Results of individual experiments are presented as examples of some techniques. Throughout the article, suggestions are included to avoid commonly encountered problems and to assist in the optimization of these techniques for the study of other RNA-binding proteins.  相似文献

Micro- and nanofabrication techniques have revolutionized the pharmaceutical and medical fields as they offer the possibility for highly reproducible mass-fabrication of systems with complex geometries and functionalities, including novel drug delivery systems and bionsensors. The principal micro- and nanofabrication techniques are described, including photolithography, soft lithography, film deposition, etching, bonding, molecular self assembly, electrically induced nanopatterning, rapid prototyping, and electron, X-ray, colloidal monolayer, and focused ion beam lithography. Application of these techniques for the fabrication of drug delivery and biosensing systems including injectable, implantable, transdermal, and mucoadhesive devices is described.  相似文献

Several simple techniques of sensory assessment are described and the conditions for their satisfactory performance stated. The manner in which these experiments can be used to provide open-ended practical exercises for classes is discussed and its advantages as a relatively cheap and simple method of teaching experimentation is described. Many techniques for surgically sterilizing dogs and cats have been described; each technique offers advantages and disadvantages to both the patient and surgeon. Techniques that have been described include traditional midline ovariohysterectomy, lateral flank ovariohysterectomy, castration, early age gonadectomy, ovariectomy, laparoscopic ovariohysterectomy and ovariectomy, and vasectomy. Regardless of the technique selected, strict adherence to sound surgical technique and asepsis is mandatory for good surgical outcome with minimal complications. Various techniques for the management of upper extremity contour deformities have been suggested since aesthetic brachioplasty was first described. Such deformities are commonplace with aging, after normal weight loss, and especially after massive weight loss such as is seen following bariatric surgery. Despite the multiplicity of procedures described for the correction of these deformities, there are still problems associated with current brachioplasty techniques, including incorrectly placed incisions, widened hypertrophic scars, and postoperative contour deformities. In addition, postoperative skin laxity and ptosis in the axillary region are frequently encountered in the more extreme deformities. The authors present their technique for upper extremity brachioplasty. This technique is suitable for patients with severe brachial ptosis and skin laxity, with relatively little lipomatous tissue, which may extend from the olecranon to the chest wall. The described surgical approach provides excellent overall extremity contour with favorable scars while simultaneously addressing axillary contour deformities.  相似文献

The literature on short scar mastopexy was reviewed, with a focus on the different techniques. Currently four techniques have been described: the periareolar, the vertical, the inverted-T, and the L-shaped scar. The different techniques were evaluated with regard to patient selection, operative techniques, scar length, and complications. A large number of techniques have been published for minimal ptosis, whereas for significant ptosis, the number of surgical options is limited. It is evident that limited scar techniques can be applied to all grades of ptosis, but there is no one technique that can satisfactorily correct all degrees of ptosis. Plastic surgeons should weigh the advantages and limitations of each technique to correctly address breast ptosis. This article reviews an algorithmic approach to correct all degrees of ptosis with mastopexy.  相似文献

Two distribution-free permutation techniques are described for the analysis of ecological data. These methods are completely data dependent and provide analyses for the commonly-encountered completely-randomized and randomized-block designs in a multivariate framework. Several protein analysis techniques are described in this review to give insight into the potential applications for research. Protein analysis can be performed in several ways. All techniques are derived from the same general principle, the migration of charged particles in an electrical field. Electrophoresis of biomolecules, like proteins, provides the possibility to identify and characterise the molecules based upon different chemical properties. Immobilisation of the proteins after electrophoresis on paper is necessary to allow easy handling of the materials (blotting). These techniques also provide information on the state of a protein, whether it is activated or inactivated. This review describes molecular cytogenetic techniques for detection and characterization of genetic aberrations associated with human disease. The techniques of fluorescence in situ hybridization, primed in situ labeling and comparative genome hybridization are described, as are probes for repeated sequences, whole chromosomes and specific loci. There are a variety of techniques that can be used to enhance or improve the nasal tip. These techniques often use suture techniques and invisible grafts to achieve the desired result. The former methods have been well described throughout the literature. Among the latter techniques, the columellar strut remains a popular and effective form of an invisible graft in rhinoplasty. The purpose of this article is to define the role of the columellar strut graft, describe how to perform it correctly in rhinoplasty, provide a clinical algorithm for its application, and detail a 15-year retrospective analysis of the senior author's (R.J.R.) experience. Previous references to the importance of the columellar strut graft in rhinoplasty have been described; however, none has formally defined its singular importance in both primary and secondary open rhinoplasty. This article details the role of the columellar strut and its relationship to nasal tip projection and lower lateral cartilage symmetry with an explanation of methods for improving each. Surface plasmon optical techniques are described as sensitive tools that allow for the on-line characterization of supramolecular biofunctional architectures at solid/solution interfaces. After a short introduction into the fundamentals of surface plasmon optics the observation of the build up of a functional bio-interface by the self-assembly process of long chain thiolates at an Au surface is described. Criteria are developed for tailoring the SAM architectures optimized for maximum protein binding from solution by specific bio-recognition reactions. SPM is employed to image the selective binding of streptavidin to a functionalized SAM laterally patterned by UV-photolithographic techniques.  相似文献

A new method for the preparation of pancreatic acini is described. The method is simple and much more rapid than previously described techniques, the time required for preparation of pancreatic acini being 20 min from removal of the pancreas. Acini prepared with this method perform in a superior manner when stimulated by either caerulein or secretin. In this review, tagging techniques with reagents used for ultraviolet-visible (UV-Vis), fluorescent (FL), chemiluminescent (CL) and electrochemical detection (ED) for higher carboxylic acids in HPLC are evaluated in terms of the tagging reactions, handling, flexibility, stability of the reagents and the corresponding derivatives, sensitivity and selectivity. Emphasis is given to the applications of these tagging techniques to biologically important carboxylic acids of relatively high molecular mass including free fatty acids, prostaglandins, leukotrienes and thromboxanes etc. Some typical examples are described. Although RIA and GC-MS are powerful techniques for the highly sensitive determination of carboxylic acids, tagging for these techniques is not included in this review because recent progress in tagging methods has been mainly concerned with HPLC detection.  相似文献

Hyperpolarized NMR is a promising approach to address the sensitivity limits of conventional NMR metabolomics approaches, which currently fails to detect minute metabolite concentrations in biological samples. This review describes how tremendous signal enhancement offered by dissolution-dynamic nuclear polarization and parahydrogen-based techniques can be fully exploited for molecular omics sciences. Recent developments, including the combination of hyperpolarization techniques with fast multi-dimensional NMR implementation and quantitative workflows are described, and a comprehensive comparison of existing hyperpolarization techniques is proposed. High-throughput, sensitivity, resolution and other relevant challenges that should be tackled for a general application of hyperpolarized NMR in metabolomics are discussed.  相似文献
