Acute myocardial infarction due to an acute type A aortic dissection involving the left main coronary artery

We report a case of anterior myocardial infarction due to a Stanford type A aortic dissection involving the left main trunk of the coronary artery. Acute myocardial infarction due to extension of an acute Stanford type A aortic dissection is an infrequent but devastating situation. In our case a spontaneous aortocoronary dissection involving the Valsalva sinus and the ascending aorta with a history of hypertension is the most plausible cause. Emergent aortic replacement and revascularisation was performed. (Neth Heart J 2007;15:263-4.)

Comparison of the tryptophan synthetase alpha-subunits of several species of Enterobacteriaceae

Bacteriol. 91:1819-1826. 1966.-The tryptophan synthetase alpha subunits of Escherichia coli K-12, E. coli B, Shigella dysenteriae, Salmonella typhimurium, and Aerobacter aerogenes have been purified and their structures compared. Each of these alpha subunits exhibits a sedimentation coefficient of about 2.7S. Peptide patterns of trypsin plus chymotrypsin digests of the alpha subunits have indicated that all of the alpha subunits have peptide regions in common. The patterns of E. coli K-12, E. coli B, and S. dysenteriae alpha subunits appear to be nearly identical, whereas the alpha subunits from S. typhimurium and A. aerogenes differ from those of E. coli and from each other. It has also been shown that the E. coli structural gene for the alpha subunit is translated identically in E. coli and S. typhimurium.

THE CHLAMYDOMONAS GENOME PROJECT

San Francisco, CA 94083-051 USA; ³Department of Botany, Iowa State University, Ames, IA 50011-1020 USA The Chlamydomonas genome project involves 1) sequencing of cDNAs isolated from cells exposed to various environmental conditions, 2) construction of a high density DNA microarray, 3) construction of genomic contigs that nucleate around specific physical and genetic markers, 4) generation of a complete chloroplast genome sequence and analyses of chloroplast gene expression, and 5) placement of the genomic information on the network in a user friendly format. The aspects of the project emphasized by our group at Stanford involves the generation of normalized cDNA libraries, sequencing of the cDNAs to generate both contigs and unigene families and the use of this information to construct a high density cDNA microarray. I will discuss the techniques involved in securing cDNA sequence information and the ways in which that information is assembled and analyzed. I will also discuss the use of the different cDNA libraries to identifying differentially expressed genes (by in silico subtractions) and strategies for constructing high-density cDNA microarrays. Finally, if completed, the first global expression studies, and the use of the microarrays to elucidate global expression in mutant strains will be discussed.

Fluorescent method for the detection of excreted ribonuclease around bacterial colonies

Bacteriol. 92:1469-1472. 1966.-A test for the release of extracellular ribonuclease by Bacillus subtilis colonles was developed. The method consists of incorporating acridine orange and ribonucleic acid into nutrient agar plates and viewing the grown bacterial colonies under ultraviolet light. Regions of ribonuclease secretion appear as dark halos around the colonies on a green fluorescent background. The theoretical basis and the utility of this test are discussed.     

Influence of glutamic acid on the endogenous respiration of Bacillus subtilis

Bacteriol. 91:546-550. 1966.-Amino acids serve as the major initial endogenous substrate for Bacillus subtilis. The endogenous activity of freshly harvested washed cells is high and falls off rapidly with time of shaking at 30 C to lower but still significant levels. The rate of O(2) consumption after the addition of glutamic acid also decreases as the cells age, but more slowly than noted for endogenous respiration. When cells were fed glutamate as soon as possible after harvesting, an apparent stimulation of endogenous respiration was noted. However, endogenous activity was inhibited if the cell suspensions were shaken for at least 1 hr before addition of the glutamate. Similar results were obtained with glycerol or glucose as exogenous substrates. Variation in rates of respiration with age of the cells, inherent instability of B. subtilis, and possible utilization of substances initially excreted by the cells appear to account for the variations noted regarding the influence of an exogenous substrate on endogenous respiration.
