One of the crucial events in the pathogenesis of neurodegenerative disorders linked with dementia-like Alzheimer’s Disease (AD) is the disturbance in neurotransmission based on progressive deficit of neuromediators that is manifested by marked decrease in cognitive behavior, loss of memory and inability to learn as a result of impairment in synaptic plasticity of neurons.In this study we have used a new complex of proteoglycans of embryonic genesis (PEG) created by Prof. L. Mkrtchyan, as a possible therapeutic approach that can rescue neurons from further degeneration caused by beta-amyloid (Aβ). We attempt to reveal the biochemical (determination of neuroactive amino acids such as glutamate, GABA, taurine, glycine and aspartate) changes and behavior on Y-maze and avoidance/exploratory activity on elevated plus-maze task in rats’ brain after modeling Alzheimer’s disease by i.c.v. injection of Aβ25–35. Furthermore, in this study we analyzed the neuroprotective properties of PEG.Under the influence of PEG the concentration of all investigated amino acids both in cerebral cortex and hippocampus (except striatum changes) increased. In the present study we demonstrated that bilateral i.c.v. injection of aggregated Aβ25–35 in dosage 30 nmol/rat resulted in impairment in spatial alternation behavior. Both preliminary (single) and double injection of PEG showed constant improvement of spatial memory after the first trial up to 90 days after i.c.v. injection of aggregated Aβ25–35.Our findings suggest that proteoglycans of embryonic genesis in neurodegenerative state show an expressed regulatory–protective effect.  相似文献

Influence of propranolol (5 mg/kg i.p.) on rhythmic metrazol activity (RMA) was studied in 11 male albino rats with chronically implanted cortical as well as subcortical (thalamic ventrobasal complex and dorsal hippocampus) electrodes. Metrazol was injected subcutaneously at a low dose of 30 mg/kg. Another group of five rats was used to study the action of propranolol on spontaneously appearing episodes of rhythmic spikes as well as on vigilance. The incidence of both rhythmic metrazol activity and episodes was significantly increased by propranolol. The latency to the first burst of rhythmic metrazol activity and to the maximum of its incidence was significantly shortened by propranolol. Propranolol also increased the incidence of relaxed wakefulness and delayed the appearance of slow wave sleep. The facilitation of RMA and spontaneous episodes could not be explained only by the increased amount of relaxed wakefulness, i.e. the vigilance level necessary for these two phenomena, because these changes did not coincide in time. In two animals, the combination of propranolol and metrazol led to the appearance of ictal activity which was sometimes accompanied by partial clonic convulsions. This phenomenon was never seen after metrazol alone (30 mg/kg s.c.) in control recordings. Propranolol was found to potentiate the possible models of human absences--spontaneous episodes, RMA and minimal clonic seizures.  相似文献

We used the intracellular recording method to study the effect of a group of nerves in the visceral complex on the activity of a pacemaking giantneuron located in the peripheral part of the visceral ganglion in a mollusk. Single excitations of the left and right pallial, the intestinal, and the anal nerves with electrical stimuli evoked similar responses, consisting of phases of rapid depolarization (duration 100 msec, amplitude 3–5 mV) and slower hyperpolarization (duration 400 msec, amplitude 5–8 mV). The excitation also had an aftereffect, which was expressed in inhibition of the background activity of the pacemaker for several seconds. The most interesting of the functional characteristics of that response was the effects of summation. With rhythmic excitation by stimuli of low frequency (0.5–1 c/sec) the result of summation was general hyperpolarization of the neuron and the appearance of giant inhibitory postsynaptic potentials (IPSP's) with an amplitude of 12–16 mV. With higher frequency of excitation (2–3 c/sec and upward) we observed depolarization replacing the hyperpolarization of the neuron, but IPSP's of large amplitude were absent. At the end of rhythmic excitation prolonged inhibition of the pacemaker's activity, lasting some minutes, occurred in all cases. This article discusses the possible mechanisms of that type of prolonged inhibition of the pacemaker's activity, the origin of the phases in biphasic responses, and the reasons for differences in the course of summation of biphasic postsynaptic potentials.M. A rapid reduction of cyclic nucleotide phosphodiesterase activity occurs after the replating of confluent cultures of BHK 21 c/13 fibroblasts into fresh medium. This reduction in activity depends on the density to which the cultures are reseeded and the concentration of serum in the medium. Enzyme activity in BHK cells is restored after 24 to 48 hours if cells are diluted into medium containing 10% fetal calf serum or 0.5% fetal calf serum supplemented with insulin (10(-6)M), but not into 0.5% serum alone. The restoration in enzyme activity is blocked by cycloheximide or Actinomycin D. When BHK cells become quiescent by maintanance in 0.5% serum conditions for 48 hours, a rapid (15--60 minutes) increase in cyclic AMP phosphodiesterase activity occurs when 10% serum is added to the cultures. Enzyme activity is increased even further after 24 to 48 hours in the 10% serum. Cycloheximide or Actinomycin D do not affect the rapid increase in enzyme activity in response to serum, but completely inhibit the long term increase. In contrast to serum, insulin (10(-8) to 10(-6)M) has no short term effect, but does increase enzyme activity after 24 to 48 hours to levels comparable to those seen with addition of 10% serum. As is the case with serum, this long term effect of insulin on enzyme activity is prevented by inhibitors of protein and RNA synthesis. Kinetic analyses of cyclic AMP phosphodiesterase activity in homogenates of quiescent BHK cells indicate the presence of only high Km (congruent to 20 muM) enzyme activity. Addition of serum or insulin to quiescent cells results in the appearance of apparent low Km enzyme activity in homogenates. Sucrose gradient analysis of BHK cells displays two forms of cyclic AMP phosphodiesterase enzyme activity: a 3--4 S form and 5--6 S form. In quiescent cells, the 5--6 S form greatly predominates relative to the 3--4 S form. Addition of serum to quiescent cells results in a rapid appearance of increased 3--4 S form enzyme activity. Insulin also increases the activity of this higher affinity 3--4 S enzyme form after 24 to 48 hours in culture. The functional significance of short and long term regulation of cyclic nucleotide phosphodiesterase(s) in cells is discussed.  相似文献

A simple method for the isolation of single ascospores of the fission yeast Schizosaccharomyces pombe was examined. Single spores in the 7-day-old sporulating culture of a homothallic strain were separated from remaining vegetative cells by isopycnic centrifugation in the linear gradient from 10 to 60% of Urografin solution at 700 X g for 20 min. Protein content of isolated spores was very low as compared with that of vegetative cells. The isolated spores germinated through the following steps when cultured in a liquid medium at 25--35 degrees C; loss of refractility (darkening) under a phase-contrast microscope, spherical growth (swelling), emergence of germ tubes, elongation of germ tubes, cell plate formation, and cell separation. The absorbance at 650 nm of the spore suspension initially decreased, accompanied by darkening of spores, and then increased with spherical growth. The germination rate of isolated spores reached almost 100%.  相似文献

Amyloid-induced aggregation and precipitation of soluble proteins were investigated in vitro using the amyloid fibrils of the beta(25--35) peptide, a cytotoxic fragment of the Alzheimer's beta-peptide at positions 25--35. The aggregation rate of firefly luciferase was found to be modulated by both a chaperone molecule DnaK and the beta(25--35) amyloid, but their effects were opposite in direction. The amyloid fibril drastically facilitated the luciferase aggregation, which may define a kind of anti-chaperone activity. The effect of the beta(25--35) amyloid to promote protein aggregation and precipitation was further demonstrated for a wide variety of target proteins. The amount of coprecipitation was well correlated with the predicted isoelectric point of the target proteins, indicating that the interaction between the beta(25--35) amyloid and the target was driven by an electrostatic force between them. This view was confirmed by the experiments using an electrically neutral mutant peptide, beta(25--35)KA. It was also found that clustering of the beta(25--35) peptide to form amyloid and the conformation of the target protein are additional factors that determine the strength of the amyloid-protein interaction. Spectroscopic and electron microscopic methods have revealed that the proteins coprecipitated with the beta(25--35) amyloid formed amorphous aggregates deposited together with the amyloid fibrils. The conformation of protein molecules left in the residual soluble fraction was also damaged in the amyloid-containing solution. As a summary, this study has proposed a scheme for events related to the nonspecific amyloid-protein interaction, which may play substantial roles in in vivo conditions.  相似文献

The morphology of Paramecium bursaria cells, both originally possessing 2 micronuclei (MI), or one MI, has been studied after a local UV irradiation of the germ nuclei. Elimination of one of the MI in bimicronuclear cells usually did not lead to general cytological damage in the UV progeny. Irradiation of MI in the unimicronuclear cells resulted in the following: producing of amicronuclear (MI-) UV subclones, 86% of which appeared to be non-viable. The MI- cells are characterized by the appearance of somatic deciliation and the loss of mobility, disorganization structures of the oral apparatus, the feeding process and the cell division. It is remarkable that the macronucleus (MA) of cells from those cultures displays fragmentation and aberrant division. The structure of MA was changed. Dimensions of the MI- ciliates decreased (by 1.7--1.8 times in average); sometimes these cells had 3--5 contractile vacuoles with one pore. These data suggest that the MI has important role during the vegetative phase of the cell life. Possible asexual cellular functions of the MI and the role of the break of nuclear interactions in the appearance of the morphological damage are discussed.  相似文献

构建表达Tat与人酸性成纤维细胞生长因子(acidic fibroblast growth factor, aFGF)融合蛋白的重组质粒pET3c–Tat-aFGF14-154和pET3c-Tat-aFGF27-154,并转化大肠杆菌BL21(DE3)中。37℃,l mmol/L的IPTG诱导4 hrs后,获得分子量约为18 kDa的融合蛋白,表达量分别占菌体总蛋白的42%和24%。利用阳离子交换(CM-Sepharose FF)、肝素亲和层析(Heparin-Sepharose CL-6B)以及分子筛(Sephadex G-25)联用的方法可获得纯度大于95%的目的蛋白,得率分别为93和78 mg/L。Tat-aFGF14-154及其对照蛋白aFGF14-154对Balb/c 3T3细胞有明显促细胞增殖的作用,最佳作用浓度分别为1280 ng/ml和160 ng/ml。而Tat-aFGF27-154 及其对照蛋白aFGF27-154则几乎没有促细胞增殖活性。免疫荧光分析结果表明,Tat-aFGF14-154及Tat-aFGF27-154均能穿过PC12、Balbc/3T3、HaCaT和海马神经元细胞的细胞膜,并主要定位于细胞浆中。此外,四种重组蛋白均能降低Aβ25-35对大鼠海马神经元细胞的毒性,在剂量为1000 ng/ml时,Tat-aFGF14-154、Tat-aFGF27-154、aFGF14-154及aFGF27-154细胞存活率分别为90.66%、81.87%、85.71%和78.02%,而Aβ25-35模型组的存活率仅为56.87%。  相似文献

In vitro, the sigma(s) subunit of RNA polymerase (RNAP), RpoS, recognizes nearly identical -35 and -10 promoter consensus sequences as the vegetative sigma70. In vivo, promoter selectivity of RNAP holoenzyme containing either sigma(s) (Esigma(s)) or sigma70 (Esigma70) seems to be achieved by the differential ability of the two holoenzymes to tolerate deviations from the promoter consensus sequence. In this study, we suggest that many natural sigma(s)-dependent promoters possess a -35 element, a feature that has been considered as not conserved among sigma(s)-dependent promoters. These -35 hexamers are mostly non-optimally spaced from the -10 region, but nevertheless functional. A +/- 2 bp deviation from the optimal spacer length of 17 bp or the complete absence of a -35 consensus sequence decreases overall promoter activity, but at the same time favours Esigma(s) in its competition with Esigma70 for promoter recognition. On the other hand, the reduction of promoter activity due to shifting of the -35 element can be counterbalanced by an activity-stimulating feature such as A/T-richness of the spacer region without compromising Esigma(s) selectivity. Based on mutational analysis of sigma(s), we suggest a role of regions 2.5 and 4 of sigma(s) in sensing sub-optimally located -35 elements.  相似文献

Ultrafiltration is an attractive downstream processing technique for concentrating enzymes and could be considered the primary step of purification. However, the efficiency of this process is often limited by protein fouling and shear-induced enzyme inactivation, which decreases permeate flux and results in the loss of enzyme activity. Although the rejection of phytase was higher than 99%, the loss of the enzyme activity was 14% during operation, indicating that the shear forces generated in the filter have significant influences on the enzyme activity. Two preparations using glycerol (25% and 35%, v/v) as a cryo-protecting agent at different temperatures were studied. The preparation containing 35% glycerol retained 70% of the initial enzyme activity at 70 °C after 1 h and had more than 3 and 6 months storage half-life at 29 °C and 4 °C, respectively.  相似文献

1. In a tethered cockroach (Periplaneta americana) whose wings have been cut back to stumps, it is possible to elicit brief sequences of flight-like activity by puffing wind on the animal's body. 2. During such brief sequences, rhythmic bursts of action potentials coming from the thorax at the wingbeat frequency, descend the abdominal nerve cord to the last abdominal ganglion (A6). This descending rhythm is often accompanied by an ascending rhythm (Fig. 2). 3. Intracellular recording during flight-like activity from identified ascending giant interneurons, and from some unidentified descending axons in the abdominal nerve cord, shows that: (a) ventral giant interneurons (vGIs) remain silent (Fig. 3); (b) dorsal giant interneurons (dGIs) are activated at the onset of the flight-like activity and remain active sporadically throughout the flight sequence (Fig.4); (c) some descending axons in the abdominal nerve cord show rhythmic activity phase-locked to the flight rhythm (Fig. 5). 4. Also during such brief sequences, the cercal nerves, running from the cerci (paired, posterior, wind sensitive appendages) to the last abdominal ganglion, show rhythmic activity at the wingbeat frequency (Fig. 6). This includes activity of some motor axons controlling vibratory cercal movements and of some sensory axons. 5. More prolonged flight sequences were elicited in cockroaches whose wings were not cut and which flew in front of a wind tunnel (Fig. 1B). 6. In these more prolonged flight sequences, the number of ascending spikes per burst was greater than that seen in the wingless preparation (Fig. 8; compare to Fig. 2). Recordings from both ventral and dorsal GIs show that: in spite of the ongoing wind from both the tunnel and the beating wings, which is far above threshold for the vGIs in a resting cockroach, the vGIs are entirely silent during flight. Moreover, the vGIs response to strong wind puffs that normally evoke maximal GI responses is reduced by a mean of 86% during flight (Fig. 9). The dGIs are active in a strong rhythm (Figs. 11 and 12). 7. Three sources appear to contribute to the ascending dGI rhythm (1) the axons carrying the rhythmic descending bursts; (2) the rhythmic sensory activity resulting from the cercal vibration; and (3) the sensory activity resulting from rhythmic wind gusts produced by the wingbeat and detected by the cerci. The contribution of each source has been tested alone while removing the other two (Figs. 13 and 14). In laboratory experiments, oysters (Crassostrea virginica) were fed Alexandrium fundyense (strain CB501) vegetative cells or resting cysts (from strains CB501 and GMT25) produced from laboratory cultures. The toxicity per cyst was 1.7 pg STXequiv/cyst and for vegetative cells 3.9 pg STXequiv/cell. The toxic, resting cysts and vegetative cells were removed from suspension in the experimental containers within about 4 h. Oysters fed toxic vegetative cells digested 72% of cells ingested, and 28% survived gut passage by forming temporary cysts. Toxin levels of oysters fed vegetative cells averaged 27 μg STXequiv/100 g meat. Resting cysts added to the experimental containers adhered to the walls so that only 40% of the cysts added were available to the oysters during the experiment. Of the cysts that were ingested, approximately 59% were digested, and oysters accumulated toxins (an average of 1.2 μg STXequiv/100 g meat), showing that consumption of resting cysts can cause toxicity in oysters. Direct consumption of resting cysts, thus, may explain shellfish toxicity in areas without known blooms, but with toxic resting cysts in the sediment. These results suggest a possible role of toxic cysts in mediating time-lags between surface blooms and appearance of toxicity in benthic grazers, and the possible role of benthic grazers in controlling seed populations, except in anoxic areas, which can serve as cyst “refuges” from grazing mortality.  相似文献

The mechanism by which metalloporphyrins escape from mitochondria has been studied in isolated rat-liver mitochondria using Co-deuteroporphyrin as the model compound. During the first 10--15 min of incubation the efflux is about 10% of the total amount of Co-deuteroporphyrin synthesized. The efflux then increases to a second steady-state leve of 25--35% after 30--45 min of incubation. The efflux is inversely correlated to the energy state of the mitochondria. Globin at concentrations less than 0.4 mumol/l enhances the efflux of Co-deuteroporphyrin, but has no effect on the degree of energy coupling or on the rate of Co-deuteroporphyrin synthesis. The effect of globin can be competitively inhibited by adding haemin. Haemin (0.5--1.0 mumol/l) when added to the medium in the absence of globin reduces the efflux of Co-deuteroporphyrin by 20--30%, but has no effect on the metal-chelatase activity. Neither albumin nor haemoglobin increases the efflux of Co-deuteroporphyrin from intact mitochondria. The results suggest that the efflux of metalloporphyrin is regulated in part by the energy state of the mitochondria and in part by the presence of metalloporphyrin-binding ligants and unattached haemin in the incubation medium.  相似文献

Enzymes from fish and aquatic invertebrates have recently been characterized and their study has led to the emergence of some new applications of these classes of enzymes. However, very little is known about lipases from mollusks. A lipolytic activity was located in the marine snail digestive glands (hepatopancreas), from which a marine snail digestive lipase named mSDL was purified. Pure mSDL has a molecular mass of about 70 kDa as determined by SDS/PAGE analysis. Unlike the known digestive lipases while acting at 37 °C, the mSDL displayed its maximal activity on long and short-chain triacylglycerols at 50 °C. A specific activity of 400 U/mg and 100 U/mg was obtained with TC4 or olive oil as substrate respectively. Only 25% of the maximal activity was measured at 37 °C. Interestingly, neither colipase, nor bile salts were detected in the marine snail hepatopancreas, which suggests that colipase evolved in invertebrates simultaneously with the appearance of an exocrine pancreas and a true liver which produces bile salts. No similarity was found between the N-terminal amino acids sequence of the mSDL and those of the known digestive lipases. Altogether, these results suggest that the mSDL is a member of a new group of digestive lipases belonging to invertebrates.  相似文献

Effect of phenobarbital (PhB, 20 and/or 40 mg/kg) on epileptic ECoG phenomena induced by metrazol was studied in acute experiments in rats aged 7, 12, 18, 25 and 90 days. Fractionated administration of metrazol (20 mg/kg i.p. each 300 s) was used to quantify the effects of PhB. First signs of metrazol action (sharp elements and/or rhythmic metrazol activity) were not reliably influenced by PhB. On the contrary, the latency of the first EEG seizures as well as of the first generalized EEG seizures was prolonged and thus a dose necessary for their elicitation was increased in all age groups. These differences reached statistical significance in 12-, 18- and 25-day-old rats. A lack of effect of PhB against the rhythmic metrazol activity supports the adequacy of this activity as a model of human absences. Differences between the development of antiepileptic and hypnotic effects of PhB (described earlier) suggest two different mechanisms of action.  相似文献

The dorsal hippocampus was electrically stimulated in unanaesthetized, unrestrained rats with a cobalt-gelatin rod in their cortex. The significance of the hippocampus in the elicitation of both physiological spontaneous rhythmic activity (episodic activity of 8--9/sec frequency bound, in rats, to a state of quiet wakefulness, and "sleep spindles") and pathological rhythmic activity of the self-sustained after-discharge (SSAD) type was determined from the aspect of the EEG and behavioural characteristics. 1. Single electrical pulses (0.1 msec, 1--10 V, 0.3/sec) elicited an evoked potential bilaterally in the somatosensory cortex. Elicitation of rhythmic after-activity (of the type of episodes or sleep spindles) was observed only in some cases in which an adequately strong stimulus was used. 2. Repeated series of rhythmic electrical stimuli following each other at short intervals (2--3 min) led to the formation of SSAD in about one third of the cases and at all stimulation frequencies (3-15/sec), although low frequencies (3--4/sec) were the least effective. The character of the SSAD and simultaneous behavioural phenomena differed fundamentally from those evoked by electrical stimulation of the thalamus (Chocholová et al. 1977). The development of paroxysmal after-activity was signalled by responses of a more or less distinct "recruiting" character during stimulation. On the basis of a comparison of electrographic and behavioural manifestations after electrical stimulation of the thalamus and hippocampus, the possibility of both thalamic and extrathalamic projection from the hippocampus to the cortical region is considered.  相似文献

Rhythmic fluctuations (Piper rhythm) have been resolved in surface electromyography (sEMG) recordings. For a long time this periodic signal could only be detected in isometric contractions, where the innervation time is much longer than the oscillation period. However, recently a periodic signal could also be resolved in cyclic movements like running, using non-linear scaled wavelets.The purpose of this study was to characterise the rhythm for different running velocities. A rhythmic signal (the Piper rhythm) was found in the EMG signal of both bellies of the gastrocnemius muscle with a frequency range of 20–35 Hz. A decrease of the Piper frequency could be observed for all of the 13 subjects when increasing the running speed from 1.3 to 4.9 m/s. The time and frequency analysis indicated that the origin of the periodic signal is from the activation of the muscle and is not likely resulting from muscle vibrations. No correlation between the medial and the lateral part of the muscle bellies could be found indicating that the two muscle compartments are fine tuned by two distinct processes. According to the literature, the signal might be the manifestation of a cortical rhythm, which has been shown for isokinetic movements.  相似文献

Rhythmic activity of the spike-and-wave type was induced by administration of metrazol (25 or 50 mg/kg s.c.) in male rats with implanted cortical electrodes. The animals were deprived of water and then allowed to lick water from a tube. Under control conditions they licked for 3-4 min without an interruption. Rhythmic metrazol activity deranged the licking; when this pathologic activity represented about one third of the time, the licking was fully blocked. We conclude that the impairment of highly motivated behaviour confirms the adequacy of the rhythmic metrazol activity as a model of human primary generalized seizures of the absence type.  相似文献

Two media were developed which specifically allow the cultivation of Bacillus thuringiensis while it is in the vegetative as opposed to the spore form. Using these media B. thuringiensis was shown conclusively for the first time to exist in an active form on the phylloplane. The profile of its appearance in vegetative and spore form was followed over a growing season on clover (Trifolium hybridum) in the field. Three simultaneous and sudden rises and declines of both spore and vegetative cell densities were observed. The most common other spore-former on these leaves was Bacillus cereus but the fluctuations in appearance of these two very closely related species were not co-incident. Using specific PCR primers a considerable diversity of cry toxin gene types was found in isolates that had been recovered in vegetative form ('vegetative isolates') with the majority possessing multiple delta-endotoxin genes while some had only one of those tested. Bioassays against a lepidopteran insect of purified delta-endotoxins showed that they were no more potent than those from a laboratory-adapted strain. PCR primers for an internal region of the vip3A gene produced amplification in 70% of the vegetative isolates compared to 25% of the laboratory-adapted strains tested.  相似文献
