SYNOPSIS. The intraflagellar structure (IFS) of the flagella of Trypanosoma brucei was examined on the basis of ultrathin sections in various planes. The IFS is composed of filaments approximately 50 A thick. These filaments seem to be identical with the protofilaments found earlier to be the basic elements of the contractile flagellar fibrils. The fibrillar system is firmly connected with the IFS and the latter is attached to the flagellar membrane by filaments. The lattice-like appearance of the IFS is caused by longitudinal and oblique filaments running in different planes. The structure of this network is discussed in detail. The IFS may serve as an abutment for the contractile flagellar fibrils.  相似文献

Summary The fine structure of the myofibers of Notoplana acticola as studied by electron microscopy indicates that they are composed of thick myofilaments about 200 Å wide with tapering ends and thin myofilaments about 50 Å wide, arranged alongside each other parallel to the long axis of the cell. There is no orderly transverse arrangement of filaments; instead they appear staggered in the fiber. In cross sections 6 to 10 thin filaments form an orbit around one thick filament with possible cross-linkage between the two types of filaments.Dense bodies are associated with the sarcolemma and with the sarcoplasmic reticulum, and appear to serve as attachments for the thin filaments. Dense bodies are compared to elements forming a fragmented Z-disc.Mitochondria, situated in the periphery or the center of fibers, are associated with granules interpreted as glycogen.The sarcoplasmic reticulum consists of: sacs or cisternae in close proximity to the sarcolemma, longitudinal tubular elements between and parallel to the myofilaments, and a tubular network around the filaments. There is no well-defined sarcolemmal-derived transverse tubule system as described in striated muscles. It is hypothesized that in these muscles, the functional equivalent of the T system may be the area of sarcolemma in contact with the cisternae of the sarcoplasmic reticulum.This work was supported by Grant No. GM 10292 from the U. Summary Campaniform sensilla associated with filiform hairs comprise an important receptor type of the multimodal sensory system of the cerci of crickets and cockroaches. Their axon projections were investigated using iontophoretic cobalt injection into single sensilla.In crickets (Gryllus bimaculatus, Acheta domestica), six different types of cereal campaniform sensilla projections can be distinguished on the basis of their axonal arborizations and terminations. Typically, a proportion of cereal campaniform sensilla, associated with long filiform hairs, give rise to axons that ascend as through fibres from the terminal ganglion to reach the sixth abdominal ganglion. Cereal campaniform sensilla associated with clavate hairs have projections restricted to the terminal ganglion alone.Whereas in crickets axons of cercal campaniform sensilla invade only certain segmental neuropils in the terminal ganglion, in cockroaches (Periplaneta americana) axons from cercal campaniform sensilla branch in every segmental neuropil. A proportion of cereal campaniform sensilla in this species also gives rise to through fibres to the fifth abdominal ganglion.We discuss morphological and functional interpretations of differences between crickets and cockroaches and consider the significance of this type of receptor in the context of previous studies of the cercal system.  相似文献

Rat oocytes in the meiotic prophase are studied by means of classical techniques of electron microscopy, preferential staining methods for DNA and RNA and specific enzymatic hydrolysis. The axial cores in leptotene and the lateral arms in the pachytene synaptonemal complex are composed by fibrils that keep a positive contrast after the application of the ethylenediaminetetraacetic acid staining method. They disappear with RNAse treatment, which reveals the presence of chromatin fibrils in the zone occupied by the cores. Preferential staining for DNA corroborates this evidence. Medial arm and lateral-medial fibrils are formed by ribonucleoproteic filaments that form bridges between pairing homologues in the zygotene. In the advanced pachytene stage, the RNA becomes scarce in these structures. No DNA can be detected either in the lateral-medial fibrils or in the medial arm. During diplotene the synaptonemal complex loses its individually and the synaptic space becomes wider and irregular. At the same time, loss of chromatin and a large increase of RNA-containing particles occur. These processes lead to the typical interphasic arrangement of nuclear components seen in the dictyate stage.  相似文献

The chromosomes of Chinese hamster cells were examined with the electron microscope and the following observations were made concerning the structure and organization of the kinetochore. — The kinetochore consists of a dense core 200–300 Å in diameter surrounded hy a less dense zone 200–600 Å wide. The dense core consists of a pair of axial fibrils 50–80 Å in diameter which may be coiled together in a cohelical manner. The less dense zone about the axial elements is composed of numerous microfibrils which loop out at right angles to the axial fibrils. Together the structures comprise a lampbrush-like filament which extends along the surface of each chromatid. Some sections suggested that two such filaments may be present on each chromatid. The fine structure of kinetochores associated with spindle filaments was essentially the same as those free of filaments. The structure and organization of the kinetochore of these mammalian cells was compared to that of lampbrush chromosomes of certain amphibian oöcytes, dipteran polytene chromosome puffs, and the synaptinemal complex seen during meiotic prophase.The authors also wish to thank Dr. Arthur Cole of the Department of Physics for the use of his electron microscope facilities and for his helpful criticism.  相似文献

Summary The fine structures of the neurons and neuropils of the magnocellular supraoptic nucleus and the parvocellular nuclei of the rostral hypothalamus, including the suprachiasmatic and medial, lateral and periventricular preoptic nuclei, and the neuronal apparatus of the organum vasculosum laminae terminalis, have been examined in the male White-crowned Sparrow, Zonotrichia leucophrys gambelii, by correlated light and electron microscopy.The magnocellular supraoptic nucleus is characterized by large neurosecretory perikarya which contain a well developed Golgi complex and densecored granules 1,500–2,200 Å in diameter. The neuropil displays axons, dendrites and glial fibers. Some axonal profiles contain dense-cored vesicles 800–1,000 Å in diameter and clear vesicles 500 Å in diameter. Axo-somatic and axo-dendritic synapses are conspicuous in this nuclear region.The suprachiasmatic nucleus is characterized by an accumulation of small neurons with moderately developed cellular organelles and some dense-cored granules, approximately 1,000 Å in diameter. The profiles of axons within the neuropil contain dense-cored granules 800–1,000 Å in diameter and clear vesicles 500 Å in diameter.The neurons of the medial preoptic nucleus are relatively large and exhibit well developed cellular organelles and dense-cored granules 1,300 to 1,500 Å in diameter. Granular materials are formed within the Golgi complex. The medial preoptic nucleus is rich in secretory perikarya.Occasionally, neurons with granules 1,500–2,200 Å in diameter are encountered in the lateral preoptic and periventricular preoptic nuclei. They may be considered as scattered elements of the magnocellular (supraoptic and paraventricular) system.The organum vasculosum laminae terminalis consists of three layers, i.e., ependymal, internal and external zones, and exhibits a vascular arrangement similar to that of the median eminence. Models of sexual selection suggest that females should prefer to mate with older males because old age is evidence of heritable high viability. In a longitudinal analysis, we demonstrate that male field crickets (Gryllus campestris) alter their calling song with age. Carrier frequency, a calling song character related to growing condition and the main song component under female preference, changed towards higher sexual attractiveness with age. Body mass decreased slightly with age, while chirp rate, an indicator of current condition, remained stable. By choosing males singing at a low frequency, female field crickets would base their mate choice decision on a sexual trait that indicates superior growing conditions as juvenile and on viability, i.e. enhanced current condition as adult.  相似文献

Summary The two types of scales of the coccolithophorid Hymenomonas (= Cricosphaera) carterae have been studied with the negative staining technique. Both types of scales, including the non-mineralized circular ones and the larger, mineralized elliptical ones are made up of a highly ordered network of radial and concentric fibrils. Both fibrils are ribbon shaped and have dimensions of 10–22×30–75 Å in cross-sections. The margin of the scales is composed of two closely associated fibrillar ribbons which are observed occasionally in edge view. In the elliptical scales, the periphery of the fibrillar network often is obscured by a deposition of the coccolith calcium carbonate. The substructure of the fibrils, the occurrence of kinking sites, and the fibrillar resistance to alkaline and weak acid treatments strongly resemble the scales of Pleurochrysis scherffelii. In relation to the recent findings of Brown and co-workers (Brown et al., 1969, 1970) on scale formation processes, it is hypothesized that the fibrillar network of the scales of both species is structurally and chemically identical, and that the fibrillar material consists of a cellulose-like polysaccharide.Supported by the Deutsche Forschungsgemeinschaft (to W. Summary In three species of Teleosts (Tinea tinea L., Leuciscus cephalus cabeda Risso, Epinephelus guaza L.) a round strongly argentophilic body of considerable size occurs in the cytoplasm of the nervus terminalis ganglion cells. In Tinea, surgical interruption of functional connections of the ganglion cells does not produce any apparent change either in the number or in the size of these cytoplasmic bodies.Electron microscopical observations show that the neurofibrillar body is made up of densely packed and irregularly arranged bundles. In cross section, each of these bundles appears to be composed of neurofilaments (100 Å in thickness) and neurotubules (diameter: 300 Å). Each tubule is surrounded by 9–10 filaments equi-distant from one another, and at a distance of 30–40 Å from the central tubule.The authors are indebted to Prof. G. Summary Electron microscope examination of the plasma membrane of chick embryo fibroblasts cultured in vitro revealed the presence of a single osmiophilic layer about 90 Å thick and a substructure composed of ovoid sub-units associated with an amorphous component. These ovoid sub-units measured approximately 112 Å along the major axis and 75 Å along the minor axis and were composed of a central core, approximately 30 Å by 60 Å, surrounded by a peripheral component.Examination of other membranous components of these cells revealed a similar ovoid subunit structure in a single layered membrane. Differences in thickness and in the sizes of ovoid sub-units were seen in these membranes. The ergastoplasmic membranes, the outer nuclear membranes, the outer mitochondrial and the Golgi membranes were found to be the thinnest.These varied in thickness from approximately 75 Å to 80 Å. The thickest membranes seen were the inner nuclear membranes. These were approximately 100 Å thick. The dimensions of the ovoid sub-units corresponded with differences in the thickness of the various membranes. These findings support the concept of a particulate substructure of cell membranes.This work was aided by Research Grant PH 5593 from the National Science Foundation. Summary The primary spermatocytes of an Opilionid, Acanthopachylus aculeatus show (at least as a rule) two bodies which in longitudinal sections appear as integrated of up to 12 dark bands each about 300 Å wide, interlaced with one another by a curtain of thin filaments. The same bodies appear in transversal sections as formed by an hexagonal lattice the nodal points of which are the cross sections of the dark bands. The study of spermatocytes at early prophase demonstrated that at the inception these bodies are formed by association of several structures comparable to those observed in the axis of paired autosomes and called synaptinemal complexes.The findings in this species are compared with their similar in other species, particularly with species of Gryllidae.This investigation was supported by United States Public Health Service Research Grant GM 08337 from the Research Grants Branch, Division of Medical Sciences, and partly by Grant RF 61034 from The Rockefeller Foundation.  相似文献

Summary Electron microscope studies on collagen from rat-tail tendon using the negative staining technique indicate the presence of fine filaments 15–30 Å in diameter within the fibres. The banding of the fibrils was only slightly affected by aminoacetylnitrile bisulphate, but an increased amount of fine fibrous material was present in preparations from experimental animals. It is suggested that this material represents a soluble form of collagen which is known to predominate after administration of the drug. Despite the fact that this would be a chemically abnormal collagen, its structure by electron microscopy corresponds remarkably well with the form suggested from X-ray diffraction and physico-chemical studies on normal soluble collagen.The author would like to thank Mr. J. Nearest neighbor analyses of the field crickets Gryllus integer, G. veletis, and Teleogryllus oceanicus demonstrated that calling ♂♂ were aggregated. Broadcasts of conspecific song to calling ♂♂ indicated that attraction of neighboring ♂♂ maintained inter-male distances. Broadcasts of G. integer song through aggregated and isolated loudspeakers showed that the total number of crickets and parasitoid flies, Euphasiopteryx ochracea, attracted to aggregated loudspeakers was greater than that to an isolate. The average number of attracted crickets and flies in an aggregation was comparable to the isolated total.  相似文献

Summary The structural relationship between the Sertoli cell and the developing spermatid was studied with the electron microscope. In the contact area of the Sertoli cell with the anterior part of the developing spermatid, a filamentous structure is observed. This structure consists of fine tubular filaments about 100 Å in diameter associated with dense material. The submicroscopic organization of terminal chromosome regions of Drosophila hydei polytene chromosomes is described. A compact region composed of tightly packed fibrils of 100 to 125 Å diameter embedded in an amorphous material is located at each of the chromosome ends of the 5 long chromosome arms. From this compact region, sometimes containing cavities, fibrils extend onto the nearest normal band region. The diameter of the extending fibrils is 100–125 Å, 200–250 Å or 400 Å. Pronase digestion of fixed and squashed chromosomes reduced the electron density of the amorphous matrix in the compact regions but failed to affect the diameter of the fibrils. The extending fibrils, however, showed a decrease in diameter after pronase digestion. The most frequently observed diameter values were 100–125 Å. — The volume of the terminal structures, including the compact region as well as the extending fibrils, is characteristically different for the various elements of the karyotype. Chromosome 2 displays the largest terminal structure, whereas chromosome 4 only occasionally shows the presence of compact regions. — End to end association of the long chromosome arms involves the fusion of the compact terminal structures. The non-random distribution of end to end association seems to be correlated with the volume of the terminal structures. Chromosome 2 which contains the largest compact terminal region is more frequently involved in end to end associations than any other chromosome arm. — The terminal regions show replication of DNA. They belong to the group of regions which display a discontinuous labeling pattern along the chromosomes, representing a late phase of the replication cycle. — The unique structural organization of the terminal chromosome regions, which is never observed at any other location of the genome supports the idea that they are morphological manifestations of the postulated telomeres.  相似文献

ABSTRACT. The precision of auditory lateralization was determined behaviourally for the cricket, Gryllus bimaculatus L. A forced-choice Y-maze test was devised in which the cricket, on entering the test arena, could not — in contrast to free phonotactic approaches — change its walking direction until after it had passed through a narrow wire-mesh tunnel. For a sound frequency of 4.7 kHz, matching the species' calling frequency, the minimum audible angle for correct side discrimination was 15°. For stimulus angles smaller than 15° from the longitudinal body axis, the crickets walked randomly to either side; stimulus angles greater than 25° resulted in all crickets turning correctly. These data reveal a sharply tuned lateral sensitivity for the auditory pathway of crickets, with an optimum at the species' calling frequency of 4.7 kHz (when compared with 3.5 and 6.0 kHz). The results for the forced-choice test are compared with the walking pattern during free phonotactic approaches, in order to determine the possible strategy underlying the acoustic orientation behaviour of the cricket.  相似文献

In the cricket Gryllus bimaculatus, it is demonstrated that the medial septum in the prothoracic trachea of the auditory system plays an important role in shaping the directional sensitivity of the ear.After perforation of the medial septum, the directional characteristic of intact animals, showing a mean right-to-left difference in sensitivity of 14 dB, becomes more omnidirectional with a mean right-to-left difference of only 7 dB. Correspondingly, the rate of change in auditory sensitivity for a sound source moving from frontal to contralateral is reduced to 0.78 dB/10° versus 1.5 dB/10° in intact animals (Figs. 2, 3).A computer simulation of phonotaxis based on these findings predicts a reduction in phonotactic performance in animals with a perforated septum. This prediction is in good quantitative agreement with experimental data (Fig. 4) and emphasizes the importance of an intact septum for effective phonotaxis in crickets.  相似文献

Summary The ultrastructure of the specialized nauplius eye of three species of the copepod genusSapphirina was investigated. The gross morphology described earlier (Elofsson, 1966a) was confirmed. The ventral cup is covered by a red pigment and the lateral cups by a red and a black pigment. The ultrastructural configuration of the pigment granules was found to differ in the two kinds of pigment cells. The black pigment cell, moreover, contains a large number of transversely banded fibrils and is able to produce reflecting crystals. The pigment granules of the black pigment cell show a variation in electron density. An intimate connexion exists between the black pigment cell and large retinula cells in the lateral cups, indicating an exchange of material. The tapetal cells present in all three cups form crystal platelets contained in two sets of membranes. It is suggested that the ventral cup and part of the lateral cups function as thePecten-eye (Land, 1965). The rhabdomeres of the retinula cells are composed of microvilli measuring 400 Å. The orientation of these seems to exclude polarotactic behaviour. The ventral cup and the four small cells of the lateral cups contain some retinula cells with microvilli arranged parallel to the incoming light. The retinula cells further develop an intricate system of membrane-invaginations penetrating deep into the cell and associated with numerous mitochondria. Retinula cells of the ventral cup and part of the lateral cups contain clear portions filled with granular material only. Retinula and other cells contain attenuated mitochondria with parallel tubuli. The proximal lens in front of each lateral cup consists of one cell. A development from the conjunctival cells is suggested. Summary A variety of human tendons have been studied at the electron microscope level. The fibers of these tendons are composed of collagen fibrils that average 1,750 Å and 600 Å in diameter. A third population that measures 100 Å in diameter may represent immature collagen or filaments that are incorporated into tendon elastic fibers. The larger collagen fibrils vary in ratio with respect to one another, and are connected by interfibrillar bridges which in some cases appear to extend through the substance of the fibril. The collagen fibrils of the paratenon are less-well organized than those of the tendon proper and average 600 Å in diameter. Tendons that exhibit the property of lateral stretch (plantaris and palmaris) were compared at the ultrastructural level with tendons that do not have this property. No differences between the two tendon types could be determined in normal or spread preparations, indicating that the differences in physical characteristics are a result of fiber rather than fibril organization.Supported by Edward G. Summary Investigations were performed by light and electron microscope on the submicroscopic structure of the epithelium of Corti's organ in the white rat.Morphological and structural differences between the inner hair cells and the outer hair cells are revealed.The inner hair cells are closely inter-related with the inner supporting cells and have a polyhedral shape, whereas the outer hair cells look like cylinders and are surrounded by an intraepithelial fluid.The structural peculiarities consist of differences in the dimensions of the hairs, in the arrangement of cytoplasmic organoids and in the aspect of the receptoneural junction. In both sensory hair cells 4 zones of different structure can be distinguished from the surface inwards: apical zone, intermediate zone, perinuclear zone and receptoneuronal junction. The functional value of these different zones is discussed and compared with what has been demonstrated in other receptors.The pillar cells and the Deiters' cells are supporting cells which have a filamentous skeleton, composed of submicroscopic individual filaments. These filaments have a diameter of about 215 Å and present some analogies with the tonofilaments of the stratified squamous epithelium. The filaments are arranged differently in the pillar cells and in the Deiters' cells. Possible functional differences between these patterns are discussed.The reticular membrane is not an extracellular cuticle. It consists of intracellular cementitious material (like the terminal bars of the epithelial cells).The Hensen's and Claudius cells, the Böttcher's cells, the inner supporting cells, the inner and outer spiral sulcus cells are regular prismatic cells with few endoplasmic organoids and without filaments.This work is dedicated to the memory of the late Prof. L.
