A simple method for determining bioethanol content in gasoline containing bioethanol (denoted as E-gasoline in this study) is urgently required. Liquid scintillation counting (LSC) was employed based on the principle that 14C exists in bioethanol but not in synthetic ethanol. Bioethanol was extracted in two steps by water from E-gasoline containing 3% (E3) or 10% (E10) bioethanol. The 14C radioactivity was measured by LSC and converted to the amount of bioethanol. The bioethanol content in E-gasoline was determined precisely from the partition coefficient in the extraction and the amount of bioethanol in the water phases: 2.98 ± 0.10% for E3 and 10.0 ± 0.1% for E10 (means ± SD; n = 3). It appears that this method can be used to determine bioethanol content in E-gasoline quickly and easily.  相似文献

Eleven published and unpublished methods for extraction of chloride from plant tissue were compared, using a homogeneous sample of dried, ground maize leaves previously labelled with36Cl. The most effective method of extraction of Cl, estimated by liquid scintillation counting of36Cl, was with hot water. However, six other methods, including either cold water extraction, or dry-ashing at 500°C of plant material previously treated to give an alkaline pH, gave36Cl values that were not statistically different from that obtained with hot water extraction. The lowest estimates of36Cl content were obtained either by wet digestion in nitric/acetic acid or by dry-ashing following Mg(NO3)2 treatment of the plant material.  相似文献

During the phytoplankton succession in 1984 and 1985, the effect of fluctuating light on algal photosynthesis (incorporation of 14C, acidified water sample) was studied in the northern Baltic. Bottles were mounted on moving racks that mimicked vertical transport caused by Langmuir circulations in the trophogenic layer. Assuming that the photoinhibition observed near the surface in fixed-depth incubations (from 1 to 8% of integral photosynthesis) was avoided in cycled samples, vertical cycling conducted around noon resulted in on average 10% lower photosynthesis than fixed-depth incubations (n = 17). This difference lies within the 5% confidence limits of the measurement, and hence it was concluded that the lack of short-term fluctuations in light associated with the vertical circulation of natural phytoplankton communities does not seriously bias conventional in situ 14CO2 fixation measurements performed at fixed depths in the study area.  相似文献

Experiments were carried out to define the effect of mixing upon primary productivity. The method compares the production of a water column with 14C-labelled samples (i) along a classic vertical profile and (ii) with a rotating system moving between the surface and the bottom of the pond, which was 0·5 m deep. In all the experiments, photosynthesis was best in the moving flask. The effect of mixing is particularly important at sunrise and sunset hours: the vertical photosynthetic budget may be over 2·5 times that of homogeneous non-turbulent water. These results have clear implications for monitoring intensive outdoor algal cultures.  相似文献

One- and two-dimensional1H- and13C-NMR spectra of lipid extracts fromUlva rigida, Gracilaria longa, Fucus virsoides andCodium tomentosum collected in the northern Adriatic Sea allowed screening of the content of fatty acid chains, carotenoids, free and acylated cholesterol and chlorophylls. The carotenoid-to-polyunsaturated fatty acid molar ratio was taken as a comparison parameter in samples ofUlva rigida collected in differentloci and seasons; the value was markedly higher in samples from the Lagoon of Venice than from marine coastal waters. The total cholesterol concentration was evaluated by1H-NMR spectroscopy and similar values were found for all species. Two-dimensional heterocorrelated NMR spectroscopy was shown to give characteristic fingerprints of the lipid extracts from algal samples as regards the content in chlorophylls, unsaturated fatty acids and carotenoids.author for correspondence  相似文献

A comparison has been made of the use of the spectrophotometer and liquid scintillation counter for the colorimetric quantification of protein. The attenuation of photon detection from sealed miniature 14C radioactive standards enabled protein quantification over a concentration range far in excess of that achievable by the use of a spectrophotometer. Accuracy of quantification was high over the entire range of protein concentration. The ability of the technique to provide quick and accurate protein estimations is discussed.  相似文献

This study presents a comprehensive analysis ofnitrogen (N) cycling in a second-order forestedstream in southern Michigan that has moderatelyhigh concentrations of ammonium (mean,16 g N/L) and nitrate (17 g N/L). Awhole-stream 15NH4 + addition wasperformed for 6 weeks in June and July, and thetracer 15N was measured downstream inammonium, nitrate, and detrital and livingbiomass. Ancillary measurements includedbiomass of organic matter, algae, bacteria andfungi, nutrient concentrations, hydrauliccharacteristics, whole-stream metabolism, andnutrient limitation assays. The resultsprovide insights into the heterotrophic natureof woodland streams and reveal the rates atwhich biological processes alter nitrogentransport through stream systems.Ammonium uptake lengths were 766–1349 m anduptake rates were 41–60 g N m–2min–1. Nitrate uptake could not bedetected. Nitrification rates were estimatedfrom the downstream increase in15N-enriched nitrate using a simulationmodel. The ammonium was removed bynitrification (57% of total uptake),heterotrophic bacteria and fungi associatedwith detritus (29%), and epilithic algae(14%). Growth of algae was likely limited bylight rather than nutrients, and dissolvedO2 revealed that the stream metabolism washeterotrophic overall (P:R = 0.2). Incubationsof detritus in darkened chambers showed thatuptake of 15N was mostly heterotrophic.Microbial N in detritus and algal N inepilithon appeared to reach isotopic steadystate with the dissolved ammonium, but theisotopic enrichment of the bulk detritus andepilithon did not approach that of ammonium,probably due to a large fraction of organic Nin the bulk samples that was not turning over. The actively cycling fraction of total N inorganic compartments was estimated from theisotopic enrichment, assuming uptake ofammonium but not nitrate, to be 23% forepilithon, 1% for fine benthic organic matter,5% for small woody debris, and 7% for leaves. These percentages agree with independentestimates of epilithic algal biomass, whichwere based on carbon:chlorophyll ratios in bulksamples and in algal fractions separated bydensity-gradient centrifugation in colloidalsilica, and of microbial N in the detritus,which were based on N released by chloroformfumigations.  相似文献

The aim of this study was to examine whether flux through the pathways of carbohydrate oxidation is accurately reflected in the pattern of 14CO2 release from positionally labelled [14C]substrates in conventional radiolabel feeding studies. Heterotrophic cell suspension cultures of Arabidopsis thaliana were used for this work. The presence of an alkaline trap to capture metabolically generated 14CO2 had no significant effect on the ratio of 14CO2 release from specifically labelled [14C]substrates, or on the metabolism of [U-14C]glucose by the cells. Although the amount of 14CO2 captured in a conventional time-course study was only about half of that released from a sample acidified at an equivalent time point, the ratios of 14CO2 released from different positionally labelled [14C]glucose and [1-14C]gluconate were the same in untreated and acidified samples. Less than 5% of radioactivity supplied to the growth medium as [14C]bicarbonate was incorporated into acid-stable compounds, and there was no evidence for appreciable reassimilation of 14CO2 generated intracellularly during oxidation of [1-14C]gluconate by the cells. It is concluded that the ratio of label captured from specifically labelled [14C]glucose is a valid and convenient measure of the relative rates of oxidation of the different positional carbon atoms within the supplied respiratory substrate. However, it is argued that failure to compensate for the incomplete absorption of 14CO2 by an alkaline trap may distort estimates of respiration that rely on an absolute measure of the amount of 14CO2 generated by metabolism.  相似文献

Summary Two liquid scintillation fluors, the first using a 2-methoxyethanol/toluene solvent, the second a Triton X-100/toluene solvent, are discussed. Data presented indicate that the technique using the 2-methoxyethanol/toluene solvent produces higher d.p.m. than does solid support counting. The Triton X-100/toluene solvent fluor is suggested for measuring aqueous samples such as algal excretion products and 14C stock solutions.  相似文献

以淡水生态系统常见的5种微藻为研究对象,通过稳态条件下单一培养的方式获取各微藻在氮素或磷素缺乏条件下对应的生长特征参数和R*值,同时将5种微藻在养分资源脉冲供给的方式下进行混合培养,进而检测养分资源脉冲供给对几种微藻种间竞争的影响作用,并与基于稳态条件下所预测的微藻种间竞争结果进行比较。研究结果显示,在稳态条件下具有最小R*值的纤细角星鼓藻在与其它微藻的种间竞争过程中始终保持优势地位,而其余4种微藻在混合培养状态下的相对比重亦与其稳态条件下的R*值紧密相关。然而,资源脉冲供给条件下的竞争优势种纤细角星鼓藻能与其它种群数量较小的微藻共存。此外,在氮素和磷素的不同供给比例情况下,对应微藻群落的结构也会发生相应的变化。实验养分资源脉冲作用下多种微藻的共存现象与自然水体中的观测现象相一致,显示了资源脉冲可能是维持生物多样性水平的一个重要机制。  相似文献

THE introduction of liquid scintillation counting techniques to the study of aquatic productivity1–3 has stimulated interest in the accurate determination of low rates of algal photosynthesis, such as occur in tropical waters or in deep living phytoplankton populations. Attention is also being directed to the measurement of soluble extracellular products of photosynthesis4. Our studies have revealed some potential sources of interference in the widely used 14C technique when used to study low rates: (1) retention of NaH14C03 by filters; (2) passive adsorption of radioactivity by filters and/or seston and (3) soluble and insoluble radioactive impurities in commercially available NaH14C03.  相似文献

The effect of filamentous algae invasion into Zostera marina meadows on water quality, sediment sulfur pools and sulfide invasion into plant tissues was studied experimentally. Sulfide invasion was assessed through analysis of sulfur isotopic composition (δ34S) and total sulfur (TS) concentrations in plant tissues. The algal mats (5 and 10 cm thickness) depleted oxygen in the mats and increased the pools of sulfides in the sediments. Plants exposed to algal mats had δ34S signals closer to the δ34S of sediment sulfide, whereas plants with no mats present had δ34S signals closer to the δ34S of seawater sulfate, indicating a higher sulfide invasion in plants exposed to algal mats. The δ34S varied between the plant tissues with the leaves having more positive δ34S signals than roots and rhizomes, indicating that sulfide was invading into the roots and moved to the other tissues through the lacunae. TS concentrations were higher in plants exposed to algal mats suggesting that sulfur derived from sediment sulfide accumulated in the plants. Fsulfide showed that up to 50% of the sulfides in the plants were derived from sedimentary sulfides. The combined effect of water column anoxia in the lower parts of the meadow and high sulfide invasion into the plants lead to significantly reduced growth rates after 3 weeks and the below-ground tissues showed signs of degradation suggesting that algal mats invasion in to Zostera marina meadows can result in seagrass decline.  相似文献

The fate of lignin in water and sediment of the Garonne river (France) and of a pond in its floodplain was examined using specifically labeled [14C-lignin] lignocelluloses. No significant differences appeared in the mineralization rate of alder, poplar or willow [14C-lignin] in running water samples. Conversion of total radioactivity to 14CO2 ranged between 18.7% and 24.4% after 120 days of incubation. Degree of 14C-labeled lignin mineralization in standing water and sediments was clearly lower, especially in submerged sediments, and was correlated with oxygen supply. After 60 days of incubation 3.3% to 7.9% of the 14C-labeled lignin was recovered in water samples as dissolved organic carbon originating from microbial metabolism. In water extracts from sediment the percentage of dissolved organic 14C was only 0.4% to 1.3% of the applied activity. In the humic fraction extracted from sediments it did not exceed 4.4% which was much lower than in soils. No significant difference appeared between river and pond conditions for humic substances formation.  相似文献

Short-term 14C-fixation (4 h) Selenastrum capricornutum algal toxicity tests were conducted with Cd (n=8), Zn (n=9) and suspended sediment aqueous elutriates (n=28) and the results were compared to those obtained in a 48 h population growth test. In order to provide more realistic experimental conditions, toxicity tests were carried out in prefiltered nutrient-spiked Lake Geneva water. The population growth inhibition test was significantly more sensitive than the14 C-fixation test for Cd (median EC50-4h and EC50-48h values of 600 and 118 μg L-1, respectively) whereas no significant difference was measured for Zn toxicity (median EC50-4h and EC50-48h values of 97 and 96 μg L-1, respectively). With suspended sediment aqueous elutriates, the relative sensitivity of the two different end points is sample dependent, with ratios of the EC25 for the14 C-fixation: population growth test ranging from <0.26 to >53.3. Elutriate toxicity shows no apparent relationship between the acute and chronic test, indicating that population growth inhibition cannot be derived directly or predicted from14 C-fixation. Both tests with their specific advantages and limitations provide valuable complementary information to measure the impact of single toxicants or complex mixtures on aquatic plants. The algal communities of the Flower Garden Banks National Marine Sanctuary have not been comprehensively evaluated and only a few dominant macroalgal species have been reported. This study utilizes both destructive and non‐destructive sampling techniques to characterize and taxonomically identify the ‘algal mat’ community structure. The East and West Flower Garden Banks are located on the outer continental shelf approximately 200 km off the Texas‐Louisiana coastline. The average depth of both banks is 100 m with the crest approximately 20 m from the surface. Harvest and photogrammetric samples were collected during two extended cruises to the Flower Garden Banks in October 1998 and March 1999. Forty, 0.25‐m2 quadrats of standing stock material were randomly collected along with one hundred sixty‐one 0.25 m2 photo‐quadrats from an average depth of 27 m. Photo‐transparencies were projected to an actual size grid with 25 random points. Four thousand twenty‐five transparency points were evaluated and used to calculate percent composition of algal cover. Harvest samples were used to characterize the “algal mat” composition, species richness, abundance, and biomass. Forty‐two species were identified from the samples representing 14 Orders. The “red algal mat” was the dominant algal coverage comprising 38.4% of all photogrammetric samples. This mat was primarily composed of members from the Order Ceramiales. Centroceras, Ceramium, and Polysiphonia comprised 33.4% of the mat, Anotrichium and Hypoglossum, 22.4%.  相似文献

The present study was initiated to determine if algal dietary fibers (DF) bind the carcinogen N-[methyl-14C]-nitrosodimethylamine (DMNA) in vitro and how bioaccumulation of orally-given carcinogen is affected by a diet containing algal DF. Eight kinds of algal DF, including powdered fronds of Laminaria religiosa (LRP) and agar (from Gracilaria verrucosa) were used in the in vitro test. Cellulose powder (CP) was used as a control DF. In vitro binding rates of DMNA by CP, LRP and agar were 0.28%, 0.65% and 0.21% of the initial dose, respectively. Rats fed a diet containing 2% LRP or 2% agar were examined at 3 or 24 h after dosing. There was reduced retention of the orally-ingested DMNA in the liver, possibly because of reduced DMNA-absorption from the intestinal tract earlier than 3 h after dosing. Binding rates of DMNA by algae were neither related to the DF values nor to the extent of reduction of DMNA-absorption from the intestinal tract.  相似文献

In crop carbon budget studies losses of root material during storage and washing of samples may cause considerable errors. To correct data from field experiments where rhizosphere C fluxes in wheat and barley were determined by14C pulse-labelling at different development stages, experiments were performed to quantify losses of14C from roots during washing. Losses of14C from wheat roots grown on nutrient solution and stored in different ways, decreased from on average 45% of total14C content 8 days after labelling to 27% after 21 days. This decrease was probably related to the incorporation of14C into structural compounds. During washing of oven-dried soil cores of held-grown wheat and barley 3 weeks after labelling, different size classes of losses of14C from the roots increased substantially with the development stage of the crop at labelling. The 0.3–0.6 mm size class increased from 5% of the14C in roots > 0.3 mm in young plants to 25% at ripening, and the < 0.3 mm size class increased from 8 to 41% of total14C content. The latter size class was, however, determined by washing handpicked roots and may therefore partly consist of adhering exudates, mucilages and microorganisms. The effect of development stage on root washing losses was attributed to root senescence which increases the fragility of roots. Thus, especially at the rate development stages root washing losses caused a severe underestimation of the root14C content. Measurements of the uptake and loss of 4C in the light and in the dark in the Tasman and Coral Seas have revealed methodological problems with the estimation of productivity in these waters. Rates of productivity estimated without replication, time series incubations and dark controls frequently overestimated the true rates of autotrophic production. The data showed unexpectedly high rates of both uptake and loss in the dark in oligotrophic waters. In oligotrophic oceanic waters, dark incorporation of 14C sometimes equalled the uptake of 14C in the light bottle. Rapid uptake of isotope in the dark controls appeared to be the result of rapid bacterial growth and metabolism. This problem was exacerbated by agitation of the sample before or during the incubation. Tropical samples were particularly susceptible to problems arising from the agitation of the samples. Latitudinal gradients of dark uptake and loss were revealed in these incubations. The loss of label during 8–12 hours in the dark (after 12 hr in the light) was as high as 50% in subtropical waters. The loss was frequently unmeasurable (< 10%) in temperate waters. The time course of 14C uptake indicated active grazing in the bottles and suggested that most of the nighttime losses of label were due to grazing by microheterotrophs. Respiratory losses appeared to be small. Calculated values of the assimilation number (or photosynthetic capacity) which did not correct for dark 14C uptake were too high to be biochemically realistic. The errors were due to the heterotrophic uptake of label and the lack of dark controls. Rapid release of 14C in the dark after incubation in the light meant that the estimate of productivity was dependant on the trophic state of the sample and on the period of incubation.  相似文献

A method is presented for separation of double- or triple-labeled liquid scintillation samples based on the partial decay of one of the nuclides. Validity of the procedure has been demonstrated with the following combinations of nuclides commonly encountered in laboratory situations; 131I-14C, 32P-14C, and 203Hg-14C, Separation of all nuclide pairs agrees favorably with predieted values except when the activity ratios exceed 103. The determinations are independent of quenching and nuclide spectra and greatly augment the number of β-β and β-γ combinations resolved by liquid scintillation spectrometry. The utility of this method for separation of triple-labeled samples is discussed.  相似文献
