In Experiment 1, the influence of exogenous GH on steroid secretion by granulosa and theca interna cells recovered from small (1-3 mm), medium (4-6 mm) and large (8-12 mm) follicles was tested. In the second experiment, theca cells (Tc) and granulosa cells (Gc) obtained from large follicles were cultured separately or in two types, Tc/Gc co-culture, where both types of cells were mixed in one well or Gc and Tc were separated by cell culture membrane inserts. In the third experiment, the influence of GH on the morphology of Gc and Tc cells and activity of Delta(5),3beta-hydroxysteroid dehydrogenase (3beta-HSD) was studied. Cells were grown in the control medium (M199+5% of calf serum) or supplemented with 100 ng/ml GH. Testosterone (10(-7) M) was added as the aromatase substrate to granulosa cells cultures. The media were assayed after 48 h of culture for progesterone and oestradiol by RIA. GH added to the culture media had no effect on oestradiol and progesterone secretion by granulosa cells isolated from small and medium follicles while it stimulated both oestradiol and progesterone secretion by Gc isolated from large preovulatory follicles. A stimulatory effect on oestradiol secretion by Tc isolated from all size follicles was observed. GH did not stimulate progesterone secretion by Tc isolated from small follicles but stimulated progesterone secretion by Tc isolated from medium and large preovulatory follicles. Both co-culture systems exhibited synergistic effect on oestradiol secretion. The stimulatory effect on progesterone secretion under the influence of GH was observed in Gc cultured alone and Tc cultured alone. In contrast, the secretion of progesterone was attenuated in both co-culture systems and the addition of GH further augmented this attenuation. A statistically significant increase in oestradiol secretion was observed in all culture conditions. The addition of GH to the culture medium stimulated the activity of 3beta-HSD compared with the control culture from both types of cells. In conclusion, the present studies indicate that there are direct and follicular development stage dependent actions of GH on steroidogenesis of porcine follicular cells.  相似文献

The aim of the present research is to ascertain whether in larval Xenopus laevis nerve-independence for the regeneration of early stage limbs and nerve-dependence of late stage limbs observed in a previous work (Filoni and Paglialunga, '90) is related to extrinsic (systemic) factors or to intrinsic changes taking place in the limb cells themselves during development. In this paper the regenerative capacity of early and late stage hindlimbs under the same extrinsic conditions, insofar as both are grafted onto the denervated hindlimbs of host larvae at the same developmental stage, is studied. All the grafted limbs are amputated after the host larvae have reached stage 57-58 (according to Nieuwkoop and Faber, '56). In experiment I, the grafted limb is amputated at stage 52, at the thigh level; in experiment II, the grafted limb is amputated at stage 54-55, at the tarsalia level; in experiment III the grafted limb is amputated at stage 57, at the tarsalia level. In all three experiments, together with the grafted limb, also the host limb is amputated at the tarsalia level. The results show that while grafted limbs amputated at stages 52 and 54-55 regenerate in the absence of nerves, grafted limbs amputated at stage 57 cannot. The failure of late stage grafted limbs to regenerate cannot be explained in terms of an immune-type inhibiting reaction since it has been observed also in denervated autografted limbs and in the host limbs. Since all the grafted limbs are in the same environmental conditions, the results show that in larval Xenopus laevis nerve-independence for regeneration of early stage limbs and nerve-dependence of late stage limbs are not related to factors extrinsic to the limb but to intrinsic changes taking place in the limb cells themselves during development.  相似文献

The experiments have been performed on 24 guinea pigs (48 labyrinthes), 6 of them--control and 18 have been subjected to noise (one octava) with the average geometrical in diaposone of 2,000 Hz, at intensity level 100 dB. Six animals are subjected to a single effect for 6 h and 12--to repeated effect during 6 days running, 4-6 h daily. Isolation of both the vestibular and the cochlear parts of the membranous labyrinth is performed simultaneously. This gives a possibility to study all the receptors of the internal ear as a whole. Certain ultrastructural changes in all the vestibular receptors both at a single and repeated effects are revealed. Dilatation of the granular endoplasmic reticulum is observed, situating mainly in the basal part of the cell; swelling of mitochondria is accompanied with a sharp clearance of their matrix. In some mitochondria there is a local destruction of their external and internal membranes. Moreover, in cytoplasm of the receptory cells sharply osmiophilic fibrillar structures are revealed, they resemble crists and are arranged in bundles. In some cells they are not numerous and localize mainly in mitochondria, in others--their number is greater, and they localize not only in mitochondria, but in the surrounding cytoplasm, too. Similar structures are observed in some preganglionic myelin fibers. These phenomena can be considered as development of calcification processes. The changes described, evidently, form the basis of the vestibular disorders under the noise effect.  相似文献

Radioresistant TN-368 lepidopteran insect cells were examined with respect to their sensitivity to the chemical agents methyl methanesulfonate (MMS), N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), propane sultone (PS), mitomycin C (MMC), and 4-nitroquinoline 1-oxide (4NQO). Based on survival ability, the TN-368 cells were more resistant than most mammalian cells to each of these agents. Concentrations of these agents which reduce survival to about 10% were used to assess recovery ability assayed by colony formation in liquid-holding and split-dose experiments. Liquid-holding experiments were performed by exposing cells in the plateau phase of growth for 1 h to 8 mM MMS, 50 microM MNNG, 9 mM PS, 110 microM MMC, or 175 microM 4NQO, removing the drug and incubating cells in spent medium for 6 h, and plating for colony formation. Split-dose experiments were performed by exposing exponentially growing cells to the above drug concentrations for 1 h, incubating in fresh medium for 6 h, exposing the cells to the agent for an additional hour, and plating. The TN-368 cells were able to significantly recover from MMS, MNNG and PS in both types of experiment. Recovery from 4NQO was observed in liquid-holding experiments and not assessed in split-dose experiments. In all cases where recovery was observed, survival enhancement was approximately 2-fold. Recovery from MMC (a cross-linking agent) exposure was not observed in either type of experiment. In addition, recovery from 8-methoxypsoralen plus UVA light (PUVA), another cross-linking treatment, was not observed. These studies indicate that DNA-DNA and/or DNA-protein crosslinking may be important molecular lesions causing death in the lepidopteran cells and that these cells may have some difficulty in repairing such damage.  相似文献

Genetic instability, provoked by exogenous mutagens, is well linked to initiation of cancer. However, even in unstressed cells, DNA undergoes a plethora of spontaneous alterations provoked by its inherent chemical instability and the intracellular milieu. Base excision repair (BER) is the major cellular pathway responsible for repair of these lesions, and as deficiency in BER activity results in DNA damage it has been proposed that it may trigger the development of sporadic cancers. Nevertheless, experimental evidence for this model remains inconsistent and elusive. Here, we performed a proteomic analysis of BER deficient human cells using stable isotope labelling with amino acids in cell culture (SILAC), and demonstrate that BER deficiency, which induces genetic instability, results in dramatic changes in gene expression, resembling changes found in many cancers. We observed profound alterations in tissue homeostasis, serine biosynthesis, and one-carbon- and amino acid metabolism, all of which have been identified as cancer cell ‘hallmarks’. For the first time, this study describes gene expression changes characteristic for cells deficient in repair of endogenous DNA lesions by BER. These expression changes resemble those observed in cancer cells, suggesting that genetically unstable BER deficient cells may be a source of pre-cancerous cells.  相似文献

The electron microscopic study of the material demonstrates that after injection of staphylococcal toxin in combination with high temperature, in the bone marrow cells of the animals certain ultrastructural changes are observed already from the first day of the experiment and they progressively increase. Especially essential changes are observed in the cells of erythroid and granulocytic sprouts. In elements of lymphoid and megakaryocytic sprouts morphological changes are manifested to a less degree. Previous studies have shown variable relationships between dispersal rate and ecosystem functioning, but the reasons for and mechanisms behind variable dispersal rate – functioning patterns are currently unknown. In this study we used six bacterial lake water communities in a laboratory experiment in order to investigate how dispersal among communities influences community productivity by evaluating three different mechanisms: 1) changes in taxonomic diversity, 2) changes in phylogenetic diversity or 3) changes in the composition of functional traits. The experiment was conducted in two phases; (A) a dialysis bag experiment where the dispersal rate among six communities was manipulated and the subsequent change in bacterial diversity and growth rate was recorded, and (B) a regrowth experiment where we manipulated available resources to study how well a taxon grows on certain organic carbon resources, i.e. their functional traits. From experiment (B) we could thus estimate changes in functional traits in communities in experiment (A). Bacterial production was affected by dispersal, but not consistently among lakes. Neither change in taxonomic or phylogenetic diversity with dispersal could explain the observed dispersal – productivity relationships. Instead, changes in trait composition with dispersal, especially the communities’ ability to use p-coumaric acid, an aromatic compound, could explain the observed dispersal – productivity relationships. Changes in this trait caused by dispersal seemed especially important for bacterial productivity in waters with a high aromaticity of the organic matter pool. We conclude that the effect of dispersal on bacterial communities can affect ecosystem functioning in different ways, through changes in functional key-traits which are important for the local environment.  相似文献

Various materials were mixed with suspensions of Serratia marcescens and other organisms. Samples were removed and frozen at intervals after mixing; the number of cells that survived both freeze-drying and exposure to air varied rhythmically as a function of time between mixing and freezing. When assayed before or immediately after drying there were essentially no fluctuations. The response was evident only when these dried samples were exposed to air. In a typical experiment, the number of cells surviving in the sample frozen 30 sec after adding propyl gallate was at least 10 times that in samples frozen either 20 sec earlier or 20 sec later. Other "peaks" in survival were observed at approximately 125 and 450 sec, but the times at which the peaks were observed were not consistent from one experiment to the next. Although we have been unable to control or predict the time at which maxima in resistance occur, we have shown that the phenomenon does occur with Escherichia coli and Saccharomyces cerevisiae as well as with S. marcescens. Furthermore, a rhythmic response also was obtained after a change in pH or cell concentration. It appears that microorganisms respond physiologically and synchronously to changes in their environment, and some of these responses have survival value.  相似文献

本实验用氚标记环磷酸腺苷(~3H-cAMP)及碘标记环一磷酸鸟苷(I~(125)-cAGP)的放射免疫法,测定了体外培养S_(180)-V系细胞周期中悬浮的M期细胞和其贴瓶细胞与均相细胞内cAMP及cGMP的含量,并对相应的细胞形态的变化做了扫描电镜观察。此外,对这三种群体细胞体积进行了测量,据此对这些细胞内cAMP及cGMP浓度进行了换算。实验结果表明:M期细胞体积的平均值接近其它时相细胞平均细胞体积的2倍,M期细胞的cAMP含量明显低于贴瓶细胞和均相细胞内cAMP含量,而cGMP含量略有提高,但差异不显著,但cAMP/cGMP比值与贴瓶细胞(非M期的其它时相细胞)及均相细胞(包括M期时相的混合细胞)相比,显著降低,相差2—3倍。用扫描电镜对上述测定的各类细胞组份进行了对应的观察,从其外形的变化表明,S_(180)-V系细胞的骨架系统很不发达,在cANP含量低的漂浮组(M期细胞)比贴瓶细胞的骨骼系统更不发达.  相似文献

Juxtaglomerular complex (JGA) and kidney medullar interstitial cells (IC) in rats in acute water and salt loadings were studied. The animals were killed 1 and 2 hours later after water 0.9% and 2% natrium chloride per os administration in quantity of 5%-7% per total body weight. Plasma osmolality and plasma renin activity (PRA) was determined. In light microscopy the granulation of the JGA-cells was in accordance with PRA in all the experiment. IC were not altered in water loading. Marked diminishing of lipid granulation of the IC in all zones of papilla at 2% salt loading was observed. In electron microscopy changes in distribution and density of lipid granules of the IC in all zones of the renal papilla were noted. The lesions showed the dystrophic and necrobiotic changes in some IC, especially in central zone of papilla.  相似文献

Degeneration of the tactile cells in epithelium of the cat sinuous hairs after sectioning the infraorbital nerve manifests itself as cytoplasmic vacuolization and induration with electron opaque bodies in it, changes in nuclear configuration and in chromatin density. At all stages of the experiment the tactile cells were practically observed demonstrating various degenerative signs. On the 45th day after sectioning the tactile cells disappeared completely. Structural changes in the tactile cells were preceded by degeneration and complete disappearance of tactile menisci from epithelium of the sinuous hairs. Perhaps, sensitive innervation is necessary to maintain differential status of the tactile cells in epithelium.  相似文献

Under the influence of alkyl hydroxybenzene (C6-AHB) added to cell suspensions at a concentration of (1-5) x 10(-3) M, the cells of Saccharomyces cerevisiae, Micrococcus luteus, and Thioalkalivibrio versutus underwent dramatic changes in the ultrastructural organization of cell membranes, cytoplasm, and inclusions. In yeast suspension, the first changes were observed after 15 min in the structure of slit-like invaginations in the cytoplasmic membrane (CM): they were shortened and thickened. In the subsequent 30 to 60 min, CM ruptures were formed in the regions devoid of intramembrane protein particles and in the slit-like invaginations. After 24 h, complete disintegration of the intracellular membrane structures and conglomeration of the ribosomal part of the cytoplasm occurred. Similar changes were observed on the exposure of gram-positive and gram-negative bacteria to AHB. However, the cell wall in all the microorganisms studied was not destroyed, and in Micrococcus luteus it was even thickened. These mummified forms were preserved as morphologically intact but nonviable cells for more than three years of observations. By their ultrastructural characteristics, these mummified forms of microorganisms were similar to the fossilized microorganisms discovered by us in fibrous kerite. The concept of micromummies was formulated. AHB are supposed to play an important role in the process of fossilization of microorganisms in nature.  相似文献

Since the beneficial effects of low-dose radiation (0.01 Gy) are usually observed in normal cells, we investigated whether the adaptive response was induced by low-dose radiation in neoplastic cells of different origin as well as in normal cells. Cell lines used in this experiment were as follows: mouse lymphocytes (NL); L929 cells established from mouse connective tissue; primary mouse keratinocytes (PK); line 308 from mouse papilloma; X-ray sensitive lymphoma cells, L5178Y-S and EL-4 cells from mouse lymphoma. The adaptive response was determined by cell survival and apoptosis. The involvement of apoptosis in the adaptive response was examined by ELISA and TUNEL assay. Adaptive response was induced by pretreatment with low-dose radiation of 0.01 Gy in normal cells such as NL, L929, and PK, but not in L5178Y-S, EL-4, and line 308 cells. In addition, the reduction of apoptosis by pretreatment with low-dose radiation was observed in NL, L929, and PK, but not in L5178Y-S, EL-4, and line 308 cells. These results suggested that the adaptive response could be induced by pretreatment with low-dose radiation and the phenomena were observed in normal cells, not in neoplastic cells. In addition, pretreatment with low-dose radiation reduced apoptosis, suggesting that an anti-apoptotic pathway may be involved in the adaptive response. Summary The acute toxicity of lectin ML I from the toxic drug, mistletoe, was demonstrated in previous experiments. Because the reason for this extremely high toxicity is not yet clear, mice were studied histochemically at different times after treatment with various doses of ML I, ML I A or ML I B chain separately, or recombinations of ML I A and ML I B. Various plasma membrane-associated hydrolases as well as Golgi apparatus-and endoplasmic reticulum-linked hydrolases, peroxisomal and extraperoxisomal oxidases, lysosomal hydrolases, mitochondrial dehydrogenases, the cytoskeletal proteins keratin and vimentin as well as iron, glycogen and lipids were analysed in all organs and tissues of female mice. Irrespective of the dose, a clear-cut response was only observed in the liver. After ML I treatment, glycogen disappeared completely from all hepatocytes, and this effect did not depend on the ML I-concentration and exposure time. The increase in activity of Golgiassociated thiamine pyrophosphatase in hepatocytes and of non-specific alkaline phosphatase in the sinusoidal endothelial cells depended on the applied ML I concentration and the time of treatment. Doses of 600 or 900 ng ML I/kg drastically increased the phosphatase activities. These clear-cut changes of glycogen and enzyme activities were not observed after administration of the ML I B chain alone, and less so when the mice were treated only with the ML I A chain, or were treated with a recombination of ML I A and ML I B even at concentrations higher than that of ML I. The glycogen as well as thiamine pyrophosphatase and non-specific alkaline phosphatase responses after ML I administration were highly reproducible. None of the other investigated cell constituents responded to treatment with ML I, irrespective of the organs, tissues and cells analysed. This was also true for mice treated with ML I concentrations higher than the LD50, after which the animals died with 2–3 days. In conclusion, application of ML I induces clear-cut liver cell changes, which are not observed after treatment with individual constituents of the ML I molecule either singly or in recombination. Using a sterol auxotroph of the LM cell mouse fibroblast, we demonstrate that relatively few cholesterol analogues can substitute for cholesterol as a growth factor. The auxotroph grows normally on desmosterol and trans-22-dehydrocholesterol and at reduced rates on dihydrocholesterol, campesterol, and 22,23-dihydrobrassicasterol. It does not grow with beta-sitosterol, stigmasterol, ergosterol, or cis-22-dehydrocholesterol when the sterol is present as sole supplement but does grow at normal rates when the analogue is supplied with suboptimal amounts of cholesterol. Two contrasting types of membrane lipid changes are observed in cells grown on cholesterol analogues. In cells grown with dihydrocholesterol, a marked increase in desaturation and elongation of fatty acids is noted. Conversely, when cells are grown with cis-22-dehydrocholesterol, desaturation and elongation of fatty acids are severely curtailed. Cells grown on alkyl sterols respond like cells grown on cis-22-dehydrocholesterol but in a less pronounced fashion. The effects of sterol substitution in mammalian cells versus in lower eukaryotes are compared, and an explanation for the secondary changes in fatty acid composition in terms of phospholipid phase behavior is suggested.  相似文献

Inducing hens to molt increases egg quality, egg production and extends the productive life of hens. It has been previously demonstrated that melengestrol acetate (MGA), an orally active progestin, decreased gonadotropic support for the ovary, which decreased the steroidogenic support for the oviduct and resulted in the cessation of lay. Estradiol produced by the theca cells of small follicles stimulates the production of the yolk proteins vitellogenin II and apolipoprotein II by the liver and supports the oviductal epithelial cells. The objective of the present experiment was to determine gene expression for yolk proteins and oviductal epithelial cell turn-over in response to a MGA-induced molt. Hy-Line W-36 laying hens were fed either 0 or 8mg MGA per day for 28 days in a balanced diet and then returned to a standard layer ration until day 36. Four birds per treatment on days 1, 8, 16, 28 and 36 were euthanized and the liver was removed and snap frozen in liquid nitrogen until RNA was extracted. Expression of vitellogenin II and apolipoprotein II genes was determined using real-time RT-PCR. A portion of the magnum was removed to determine proliferation and programmed cell death for secretory and ciliated luminal epithelium. Vitellogenin II and apolipoprotein II gene expression was reduced in hens fed 8mg MGA compared to those fed 0mg MGA. There was no effect of day on gene expression of either yolk protein. Cell proliferation was increased in the ciliated epithelial cells of the oviduct in hens receiving 8mg MGA compared to those receiving 0mg. However, programmed cell death of the ciliated epithelial cells was not different between controls and MGA treatment. Programmed cell death and proliferation increased in the secretory epithelial cells in hens receiving 8mg MGA compared to controls. Therefore, utilizing MGA as an alternative method to induce molt results in some, but not all, of the physiological changes previously described for hens molted by feed withdrawal. These findings lead us to suggest that some of the observed physiological changes resulting from feed withdrawal are required to increase egg quality and egg production following molt and other changes are not necessary, but are just a result of nutrient deprivation.  相似文献

The aim of the study was to follow morphology of the prolactin producing cells in growing female rats with evoked "early androgen syndrome". The experiment was carried out on 3, 6 and 12-week-old animals. At the age of 3 weeks no changes in morphology of the prolactin cells were observed as compared to that of control animals. In 6 and 12-week-old animals the significant differences between androgenized and control animals were found. The prolactin cells differed both from those characteristic for normal females and normal males. The main characteristic features were: significantly smaller number of cells than in normal females, their stronger fluorescence and presence of large and giant prolactin cells similar to the so called "pregnancy cells". Possible factors responsible for the described above changes are discussed.  相似文献

In many dinoflagellates, it has been a challenging task to study the qualitative and quantitative processes leading to encystment because gametes are often not morphologically distinguishable from other vegetative cells. We examined whether sexual differentiation is accompanied by changes in cell surface glycoprotein properties that are reflected in the binding patterns of complementary lectins. Labeling percentages of nine different fluorescein isothiocyanate (FITC)-conjugated lectins were analyzed together with cell and cyst abundance in N-deplete and f/2 control cultures of Scrippsiella lachrymosa Lewis throughout an encystment experiment. Although labeling varied between lectins and treatments and considerable changes occurred through time, no direct correlation was observed between glycoconjugate properties and sexual life cycle processes. A conspicuous decrease in labeling of lectins that are complementary to amino sugars (in particular, with WGA, a lectin that is complementary to N-acetylglucosamine) was observed in the low nitrogen treatment, suggesting a link between the nutrient status of a cell and expression of surface carbohydrates. Presumably, the reduction of N-acetylglucosamine residues was an early indication of N stress in cell populations. Labeling experiments with phosphate-limited cells showed that the decrease in WGA-complementary amino-sugar residues was not specific for N stress, but appeared to be a general response to nutrient limitation. Our findings confirm that glycoconjugate composition of dinoflagellate cells can change depending on their physiological state, which has to be considered when applying lectins for taxonomic differentiation of dinoflagellate species.  相似文献
