Synthetic routes are described to the d-mannopentaoside methyl 3-O-(3,6-di-O-α-d-mannopyranosyl-α-d-mannopyranosyl)-6-O-α-d-mannopyranosyl-α-d-mannopyranoside, and the d-mannohexaoside methyl 3-O-(3,6-di-O-α-d-mannopyranosyl-α-d-mannopyranosyl)-6-O-(2-O-α-d-mannopyranosyl-α-d-mannopyranosyl)-α- d-mannopyranoside, formed in a regio- and stereo-controlled way by employing the properly protected d-mannobioside methyl 2,4-di-O-benzyl-3-O-(2,4-di-O-benzyl-α-d-mannopyranosyl)-α-d-mannopyranoside and d-mannotrioside methyl 2,4-di-O-benzyl-3-O-(2,4-di-O-benzyl-α-d-mannopyranosyl)-6-O-(3,4,6-tri-O-benzyl-α-d- mannopyranosyl)-α-d-mannopyranoside as key intermediates.  相似文献

The lipopolysaccharide isolated from the cells of Shigella boydii type 8 bacteria gave precipitin bands against homologous antisera on Ouchterlony plates, whereas the carbohydrate-containing fractions obtained from it did not. One of the fractions was obtained in major proportion and contained 23.5% of sugars. A structure was assigned to the carbohydrate chain in this material by using the results of methylation, periodate oxidation, and deamination studies.  相似文献

Conformation of the antifreeze glycoprotein of polar fish   总被引:5,自引:0,他引:5  
High-field proton and 13C NMR spectroscopy has been used to test and refine the recent proposal, based on vacuum uv circular dichroism results, of a threefold left-handed helical conformation for antifreeze glycoprotein (AFGP). Partial assignment of the protons of the glycotripeptide repeating unit has been made by comparison with spectra of model compounds, by selective decoupling, and by measurements of nuclear Overhauser effect (nOe). At 40 degrees C, AFGP fraction 8 (Mr 2600) shows 2-Hz linewidths which broaden at lower temperature. Neither 1H nor 13C chemical shifts depend strongly on temperature, suggesting no abrupt conformational transition. The nOe between alanine alpha and beta protons vary with temperature and with field strength, from small positive enhancements at 50 degrees C and 80 MHz to large negative effects at 3 degrees C and 300 MHz, indicating a substantial change of rotational correlation time with temperature. The higher-molecular-weight fraction 1-4 shows negative nOe at all temperatures. The CD spectra of fraction 1-4 show bands characteristic of the polyproline II structure at both 3 and 50 degrees C, while those bands in fraction 8 are weaker at 50 than 3 degrees C. The 1H nOe, the 13C T1, and CD data are interpreted as indicating that AFGP fraction 8 is an extended "rod-like" conformation at low temperature which becomes a flexible coil at high temperature, while fraction 1-4 is a flexible rod with sufficient segmental mobility to eliminate any long-range order.  相似文献

A heptasaccharide was released from the plant cell-wall, pectic polysaccharide rhamnogalacturonan II by selective acid hydrolysis of the glycosidic linkages of apiosyl residues. The heptasaccharide was purified to homogeneity by gel filtration and anion-exchange chromatography. Some of the heptasaccharide molecules were found to be mono- and some di-O-acetylated, but the location of the acetic ester groups was not determined. The heptasaccharide was found to have the following structure, where AceA = an aceryl (3-C-carboxy-5-deoxy-l-xylosyl) residue, and Api = an apiose residue.

Crystal structure of a lysozyme-tetrasaccharide lactone complex   总被引:6,自引:0,他引:6  
The binding of a proposed transition-state analogue, the δ-lactone derived from tetra-N-acetylchitotetraose, to lysozyme in the crystal at pH 2.6 has been studied by X-ray diffraction techniques to a resolution of 2.5 Å. The tetrasaccharide lactone is bound in sites A, B, C, D with sugar residues located in sites A, B and C in similar positions to those observed previously in the complex with tri-N-acetylchitotriose. Analysis of the electron density map for site D, by direct model-building and with a computer model-building programme, indicates that the δ-lactone ring is in a conformation close to a sofa or a boat which brings the hydroxymethyl group C(6)O(6) axial. These studies provide support for the role of strain in the proposed mechanism of lysozyme catalysis. The orientation of the lactone group in site D is slightly different from that originally derived by hypothetical model-building.  相似文献

Pressure-area curves of monolayer films were measured for phosphatidylcholines (PC) in which the 1-position was occupied by palmitic acid and the 2-positions were occupied respectively by: 20 : 0, 20 : 1n9, 20 : 2n6, 20 : 3n3, 20 : 3n6, 20 : 3n9, 20 : 4n6 or 20 : 5n3 fatty acids. The interactions of these PC with cholesterol or desmosterol were studied.Fully saturated PC (16 : 0–20 : 0) displayed a relatively small molecular area. The presence of one double bond greatly increased the molecular area, but a second double bond resulted in only a small additional increase in area. A third double bond caused a further large expansion in area, but the presence of a fourth or fifth double bond had little additional effect.Condensation of molecular area was observed with all sterol/PC mixed films. Approximately equimolar mixtures of sterols and unsaturated PC condensed maximally, but 16 : 0–20 : 0 PC condensed most in mixtures containing 20–30 mol% of either sterol.The extent of condensation varied with surface pressure. The pressure at which maximum condensation occurred depended upon the structure of the PC and was always 20 dyn/cm or lower. The pressure at which maximum condensation with cholesterol occurred increased with increasing unsaturation of the PC.  相似文献

A new cytochrome P-450 model that simulates the unusual spectral and substrate-oxidation properties of cytochrome P-450 is proposed. The complex, consisting of glutathione(GSH), hemin and pyridine(py), exhibits similar optical and EPR spectra to cytochrome P-450 in ferric low-spin state. On omission of py, a ferric high-spin state was produced. On exposure of the GSH-hemin-py complex to CO, a characteristic absorption band appeared at 450nm, like that typical of cytochrome P-450. Two types of spectral changes were observed when aminopyrine or phenobarbital (Type I) and aniline or quinoline (Type II) were added to the GSH-hemin complex. Hydroxylation, dealkylation and aromatic methyl migration activities were observed with the GSH-hemin complex.  相似文献

An antiserum against material shed into serum-free medium by MCF-7 human mammary carcinoma cells (anti-SFM II) disrupts cell-cell interactions in murine mammary tumor epithelial cells (MMTE). We now report purification of an 80 kd glycoprotein (GP80) from SFM of MCF-7 mammary carcinoma cells that blocks the activity of anti-SFM II. Anti-SFM II also inhibits compaction of eight-cell mouse embryos, and purified GP80 blocks this reaction. An antiserum against purified GP80 (anti-GP80) has all adhesion-disrupting activities displayed by anti-SFM II. It recognizes one band at 80 kd in SFM and a 120 kd band in detergent extracts of epithelial but not fibroblastic cells. In immunofluorescence studies it is restricted to sites of cell-cell interaction in cultured epithelial cells. Thus a cell surface glycoprotein of 120 kd, the medium form of which is approximately 80 kd, which is neither species nor tissue specific, is expressed at early stages of mammalian development and is found on epithelia.  相似文献

Two acidic proteins from the 50 S subunit of Bacillus stearothermophilus ribosomes, namely B-L13 (homologous to Escherichia coli protein L7L12) and B-L8, form a complex. Radioactive B-L13, added to ribosomes before dissociation, does not appear in the complex after electrophoresis, so the (B-L13 · B-L8) complex must exist in the ribosome before dissociation. Digestion of B. stearothermophilus ribosomes with polyacrylamide-bound trypsin causes the appearance of new B-L8 and B-L13 spots on two-dimensional polyacrylamide gel electrophoresis, in a pattern which suggests that single molecules of B-L13 are being sequentially cleaved from a four-to-one complex of B-L13 and B-L8.  相似文献

An assay procedure for thyroid hormone receptor activity which used nitrocellulose membrane filters was developed. Receptor proteins, extracted from washed rat liver nuclei with a 0.4 M NaCl solution, were incubated with 125I-labeled thyroid hormone (T3), and filtered on the cellulose ester membranes under suction at 2 degrees C. The filters were subsequently washed with cold buffer and counted for 125I radioactivity. The method allowed an accurate estimation of the receptor activity, satisfying a linear relationship between the activity and the receptor protein concentrations. The usefulness of this filter-binding method became evident when it was compared with the conventional procedure that employs Sephadex G-25 columns. For practical application to routine assays, various filtration conditions were examined, and a standard procedure was established. Using this technique, the isolated receptors were determined to possess an apparent Kd of 1.38 X 10(-10) M and a pH optimum of T3 binding at 8.2-8.4.  相似文献

The transport kinetics of the influenza virus hemagglutinin from its site of synthesis to the apical plasma membrane of Madin-Darby canine kidney cells, a polarized epithelial cell line, were studied by a sensitive tryptic assay. Hemagglutinin acquired terminal sugars, as judged by sensitivity to endo-β-N-acetylglucosaminidase H, 10–15 min after synthesis, and first appeared on the apical domain 15 min later. None of the pulse-labeled hemagglutinin accumulated on the basolateral domain. At 20°C, terminal glycosylation continued, but no hemagglutinin was detected on the cell surface within 2 hr. If the incubation temperature was raised from 20°C to 37°C, hemagglutinin was quickly externalized, demonstrating that the inhibition at low temperature was reversible.  相似文献

A new covalent chromatography system utilizing Activated Thiol Sepharose 4B was employed to quantitate the content of thionein chains synthesized in a polysomal cell-free system. Liver polysomes from zinc injected rats directed the translation of more thionein-like polypeptide chains than polysomes from control rats. The increase was similar to the stimulation in MT synthesis in vivo following a zinc injection. This evidence supports the concept that metallothionein synthesis is regulated by changes in the pool of translatable thionein mRNA.  相似文献

The soluble, lignin-carbohydrate complex (LCC) from the rumen fluid of steers fed a diet of pure spear grass (Heteropogon contortus) has been purified by gel filtration. The purified LCC contained 7.4% of carbohydrate which, on hydrolysis, gave d-glucose, d-xylose, l-arabinose, l-rhamnose, and traces of d-galactose and d-mannose. The structure of the LCC was examined by methylation analysis, using g.l.c.-m.s. for the unequivocal classification of the sugar derivatives. d-Glucose, d-xylose, and l-rhamnose were shown to be glycosidically linked to lignin. Some of the d-glucosyl residues carry other (1→4)-linked d-glucose units, and some of the d-xylosyl residues bear other (1→4)-linked d-xylose units and (1→3)-linked l-arabinofuranosyl groups. The major carbohydrate component is a single d-glucopyranosyl group. The LCC was subjected to various chemical treatments in an investigation of the chemical nature of the bonding between lignin and the carbohydrates. d-Glucose could be enzymically hydrolyzed from the LCC, but only with a very high concentration of β-d-glucosidase. The presence of lignin in rumen LCC has been confirmed by nitrobenzene oxidation, vanillin and syringaldehyde being identified by g.l.c.-m.s. as oxidation products from both the original spear grass and the LCC.  相似文献

The secondary structure of the protein moiety of the α1-acid glycoprotein (orosomucoid) was evaluated from its primary structure by using the Lim and Chou and Fasman predictions, and the corresponding dichroic spectrum was calculated. The experimental dichroic spectrum of the whole glycoprotein was compared with the summation of (i) the calculated dichroic spectrum due to the protein moiety and (ii) the experimental dichroic spectrum of the carbohydrate moiety. The results are in good agreement with the fact that the carbohydrate moities do not produce any pertubation of the protein conformation. In addition, we observed that four out of five glycan chains are linked to Asn residues which are situated either in a reverse β-turn or in regions where charged and polar residues are numerous, that is, on the outside of the protein.  相似文献

Radioiodination of penicillin X (p-hydroxybenzylpenicillin) with 125INa, using the chloramine-T method, is simple and almost quantitative. The product thus obtained can be used without further purification for the penicillin-binding proteins (PBPs) assay. The chemical structure of 125I-penicillin X is very close to that of penicillin G, and the affinity of the two compounds for the PBPs are very similar. 125I-penicillin X can then advantageously replace [14C]penicillin G in these experiments, as its specific radioactivity is 2000 Ci/mol, in our preparations, instead of about 50 Ci/mol; thus, the experimental procedure is simplified and the autoradiography exposure time is reduced to 12-48 h.  相似文献

Periodate oxidation of LPG-1 established that N-acetylneuraminic acid residues are linked preponderantly α-(2→3) to D-galactose residues. The resistance of 2-acetamido-2-deoxyD-galactose residues to periodate oxidation suggests that they are linked at either O-3 or O-4 to D-galactose residues. After treatment of LPG-I with alkaline sulfite, ≈80% of 2-acetamido-2-deoxygalactose was recovered as the sulfonic acid derivative. The Gal→GalNAc disaccharide released from sialic-acid-free LPG-I by digestion with endo-2-acetamido-2-deoxy-α-D-galactosidase (which suggests an α-D-GalNAc→-L-Ser or -L-Thr linkage) gave a high color-yield in the Morgan—Elson reaction, indicating that 2-acetamido-2-deoxy-D-galactose residues are linked at C-3 to D-galactose residues. The migration of the released Gal-GalNAc disaccharide was the same as that of a standard sample of O-β-D-galactosyl-(1→3)-2-acetamido-2-deoxy-D-galactose. Treatment of sialic acid-free LPG-I with Streptococcus pneumoniae β-D-galactosidase, which hydrolyzes only galactosides linked β-D-(1→4) gave no free D-galactose, whereas treatment of LPG-I with bovine testes β-D-galactosidase released > 90% of D-galactose. These results provide evidence for β-D-Galp-(1→3)-α-D-GalNAcp-(1→3)-L-Ser or -L-Thr and α-NeuAc-(2→3)-β-D-Galp-(1→3)-α-D- GalNAcp-(1→3)-L-Ser or -L-Thr structures. The sensitivity of the methods used and the recovery of constituents following treatment of LPG-I do not rule out the occurrence of small amounts of other tri- or tetra-saccharide chains.  相似文献

The seeds of Anthocephalus indicus contain a water-soluble polysaccharide composed of D-xylose, D-mannose, and D-glucose in the molar ratios 1:3:5. Methylation analysis afforded 2,3,4-tri-O-methyl-D-xylose, 2,3,6,-tri-O-methyl-D-mannose, 2,3,6-tri-O-methyl-D-glucose, 2,3-di-O-methyl-D-glucose, and 2,3,4,6-tetra-O-methyl-D-glucose in the molar ratios 7:21:12:15:8. Periodate oxidation and methylation data indicated 22.5% and 21.9% of end groups, respectively. The above findings, together with the results of partial hydrolysis with acid, indicate the polysaccharide to consist of a linear chain of (1→4)-linked β-D-mannosyl and β-D-glucosyl residues to which α-D-xylosyl and β-D-glucosyl groups are attached by (1→6)-linkages.  相似文献

A unit gravity sedimentation chamber with a flow deflector of special design is described. The flow deflector consists of a rotational-symmetric Perspex body fitted in the top cone of the chamber. This deflector allows both the undisturbed introduction of a buffered step gradient and the undisturbed application of an extremely thin sample layer (0.28 mm) as well as the undisturbed fractionation of the contents of the chamber. The operational quality of the method is substantiated by the separation of : (i) murine leukemia G1,S- and G2,M-phase cells; (ii) human lymphocytes from monocytes; (iii) diploid and tetraploid rat liver nuclei; and (iv) isolated chromosomes from Chinese hamster into different size classes.  相似文献
