Uptake ofmyo-inositol by astrocytes in hypertonic medium (440 mosm/kg H2O) was increased near 3-fold after incubation for 24 hours, which continued for 72 hours, as compared with the uptake by cells cultured in isotonic medium (38 nmoles/mg protein).myo-Inositol uptake by astrocytes cultured in hypotonic medium (180 mosm/kg H2O) for periods up to 72 hours was reduced by 74% to 8 to 10 nmoles/mg protein. Astrocytes incubated in either hypotonic or hypertonic medium for 24 hours and then placed in isotonic medium reversed the initial down- or up-regulation of uptake. Activation of chronic RVD and RVI correlates with regulation ofmyo-inositol uptake. A 30 to 40 mosm/kg H2O deviation from physiological osmolality can influencemyo-inositol homeostasis. The intracellular content ofmyo-inositol in astrocytes in isotonic medium was 25.6 ± 1.3 g/mg protein (28 mM). This level ofmyo-inositol is sufficient for this compound to function as an osmoregulator in primary astrocytes and it is likely to contribute to the maintenance of brain volume.  相似文献

The reactions of the starfish Asterias amurensis and Patiria pectinifera that live in Vostok Bay at the salinity of 32–33 to a salinity decrease were studied under laboratory conditions. The lower limits of the desalination tolerance range of A. amurensis and P. pectinifera were, respectively, 24 and 20. A. amurensis proved to be less resistant to desalination. Under experimental conditions, all specimens of this species survived the salinity of 22, while those of P. pectinifera tolerated 18. At the same time, A. amurensis responded more actively than P. pectinifera to unfavorable changes in the environment. Turned to their dorsal side and exposed to a salinity of 16 to 32, the former reverted to the normal position within a shorter time than the latter. Being a more euryhaline species, P. pectinifera endured a salinity decrease to 6 or 8 over, respectively, 21 or 28 h. However, only 30–40% of all specimens could recover locomotory activity 12 or 8.5 h after being placed into water of normal salinity.  相似文献

The Passeridae are characterized by possessing a neomorphic preglossale bone in the tongue and a uniquely enlarged M. hypoglossus anterior forming part of the seed-cup used to orient and hold seeds in position when they are being husked. The pale rock finch (Carpospiza brachydactyla), which had been suggested in recent years to be allied to the Fringillidae rather than to the Passeridae, also possesses these unique derived features of the passerine finches and hence is clearly a member of the latter group. Further, the combination of the neomorphic preglossale bone and the large M. hypoglossus anterior provide strong support for the independent evolution of granivorous feeding in the Passeridae and against the placement of these birds with any members of the Ploceidae.Communicated by F. The reproductive structures ofDeplanchea tetraphylla (Bignoniaceae) exhibit a significant number of unusual features: inflorescence with an apical platform; flowers yellow, short-tubed, strongly zygomorphic; mouth closed through lateral compression; stamens and style long-exserted, erect or slightly reclined; nectar dark brown, exposed in the spoon-shaped lowermost corolla lobe and apparently acting also as a visual cue. These features suggest a highly elaborate syndrome for bird pollination: the birds (probably lorikeets) perch on the inflorescence platform and bend downwards to take up the exposed nectar, thus touching the exserted anthers and stigmas with the throat or breast. The likely evolution of this syndrome by additive steps, effecting a change from head up to head down position of the pollinator, can be traced from the floral structure of the remaining four species of the genus.  相似文献

The seed morphology of 151 species of Southern AfricanOrchidoideae (Orchideae andDiseae; sensuDressler 1981) was studied by means of scanning electron microscopy. Two different seed types were found. (1) In the majority of species the seeds are minute and fusiform. The seed coat is made up of comparatively few concave and elongate testa cells with straight or slightly undulate and generally unthickened anticlinal cell walls. The seed type was here termed Satyrium-type. While most species are very similar in the ornamentation of the periclinal walls of their seed coat, considerable variation was found inHolothrix where two distinct groups can be recognized in this respect. (2) A remarkably different seed type was observed inDisa uniflora and three apparently closely related species (Disa uniflora-type), where large balloon-like seeds occur. Their seed coat consists of convex cells with undulate anticlinal walls. It is suggested that this seed type is a derived condition and has evolved in adaptation to the specialized habitat alongside streams. The possibility of hydrochory in these four species is briefly discussed.  相似文献

A survey is made of the occurrence, nature and frequency of satellited chromosomes in the agamospermous genusTaraxacum. Species belonging to the 10 sections thought to be most primitive in the genus lack satellited chromosomes. In most other sections, a characteristic satellited chromosome is seen with a large euchromatic region distal to the presumed nucleolar oraniser region (NOR). In sections of a precursor type, there is always one chromosome of this Taraxacum type per haploid genome. In sections thought to be of an advanced type the number of such satellited chromosomes is very unstable, sometimes even within the same tissue. In sectionHamata, two such satellited chromosomes are invariably found in triploids. This finding strongly supports the integrity of this section, suggests that the species of the section are monophyletic, and have evolved from a single ancestor subsequent to the occurrence of obligate agamospermy. In three sections of the genus, satellited chromosomes of the conventional type with a very small distal euchromatic region distal to the NOR are reported for the first time in the genus.  相似文献

Summary The acrosome in spermatozoa from the caput epididymidis of the Australian Brush-tailed possum, Trichosurus vulpecula, typically forms a cup-like structure, sitting on the anterior third of the dorsal surface of the nucleus. The base of the acrosomal cup is narrowly separated from the nuclear surface, while the body of the cup projects voluminously away from the nucleus.During epididymal transit these pronounced marginal extensions of the acrosome are retracted towards the nucleus, and the electron dense acrosomal material undergoes a process of compaction within the plasma membrane of the head to produce the convex ovate form of the definitive acrosome. During this process a variety of bizarre forms of the acrosome are produced before its final configuration is attained.The authors would like to thank Dr. D.J.H. A new species of aploactinid, Cocotropus keramaensis, is described based on a single specimen collected off the Kerama Islands, southern Japan. This new species is separable from other known congeners in the following combination of characters: XII, 10 dorsal fin rays; II, 7 anal fin rays; 13 pectoral fin rays; 26 vertebrae; 4+8=12 gill rakers; five distinct preopercular spines; upper jaw longer than the lachrymal; single papilla on the posterior portion of the maxilla; outer edge of the lower jaw with four distinct papillae; first sensory pore of the lower jaw on both sides fused; anterior tip of the isthmus reaching to the fifth sensory pore of the lower jaw; and anterior four dorsal spines anterior to the third neural spine.  相似文献

Summary We have investigated the central connections of the classical olfactory system in the weakly electric fish Gnathonemus petersii using HRP and cobalt labelling techniques. The olfactory bulb projects bilaterally via the medial and lateral olfactory tracts to restricted areas of the telencephalon, namely to its rostromedial, lateral and posterior medial parts. The most extensive telencephalic target is the posterior terminal field, which arcs around the lateral forebrain bundle at levels posterior to the anterior commissure. Projections to the contralateral hemisphere cross in the ventral telencephalon rostral to the anterior commissure and via the posterior dorsal part of the anterior commissure; endings are also present within the anterior commissure. Bilateral projections to the preoptic area, to the nucleus posterior tuberis and to an area in the thalamus are apparent. In all cases, contralateral projections are less extensive than those on the side ipsilateral to the injected bulb. A projection via the medial olfactory tract can be followed to the contralateral bulb. Following injections into the olfactory bulb, retrogradely labelled neurons are found in the contralateral bulb and in six telencephalic areas; they are also present in the periventricular diencephalon and in an area lateral to the nucleus posterior tuberis. The present results support the suggestion that a reduction in olfactory input to the telencephalon occurs together with increased telencephalic differentiation in actinopterygian fishes.  相似文献

Summary Treatment of young thalli ofSphacelaria rigidula with 0.04 g of nocodazole (Nz) per ml for up to 36 h affects microtubules (Mts) only slightly, but blocks a large number of mitotic cells in metaphase, without disruption of the metaphase plate. Higher concentrations of Nz (0.1 g/ml) depolymerize interphase Mts. Only a few perinuclear and some short Mts resist and remain associated with the centrosomes. Fragmented Mts or groups of Mts sometimes remain in the apical dome. After treatment with 0.1 g of Nz per ml, prometaphase cells are blocked at metaphase, while post-metaphase cells become binuclear, due to the failure of cytokinesis. With anticentrin immunofluorescence, a positive centrin signal is always observed in the centrosome area. Centrosome duplication is not affected by Nz, but separation is disturbed. After recovering for 2–4 h, most of the blocked metaphases proceed normally. In such cells duplicated centrosomes are seen in different stages of separation. In some cells independent aster-like microtubule configurations appear in the apical dome, occasionally displaying centrin at their centre. During recovery various configurations of bimitosis or multipolar mitosis were found. The multipolar spindles may share common centrosomes. Up to four centrosomes may accompany each nucleus. In some 24 h treated cells, as well as in cells recovering for 2 h, the centrin-positive structure is rod-like, extending in opposite directions from the usual position to the poles. Electron microscopical examination of thin sections revealed that the growth pattern of the apical cells is disrupted after Nz treatment. The observations show that: (a) the Mt cytoskeleton is involved in maintaining the polarity and growth pattern of apical cells, (b) mitosis is blocked by low concentrations of Nz without significant depolymerization of Mts, (c) the centrosome cycle is independent of the nuclear cycle, (d) centrosome separation and differentiation are disturbed by Nz treatment, (e) during recovery from Nz treatment, centrosomal material that may have separated from the centrosomes, as well as Mt fragments that resisted depolymerization, may operate as Mt nucleation centres.Abbreviations DIC differential interference contrast - EM electron microscope - Mt microtubule - MTOC microtubule-organizing center - Nz nocodazole - NBBC nucleus-basal body connector  相似文献

Summary The spermatozoon of the holothurian Leptosynapta clarki has a small circular head measuring about 3.0 at the greatest diameter, a midpiece containing a single mitochondrion and a tail flagellum measuring between 35 and 45 in length. The acrosomal region contains a granule measuring 0.7 in diameter which consists of electron dense material arranged in concentric lamellae. Five concentric very electron dense lamellae alternate with areas of much less electron dense material in the central region of the granule. This granule rests in an anterior nuclear depression.The nucleus is circular in shape and contains one or two unbound vacuoles which frequently contain a fine granular material. Posteriorly the nucleus is bounded by a large mitochondrion and an occasional Golgi complex. The proximal centriole which contains a lateral arm of dense material lies in a deep fossa projecting into the nucleus. The distal centriole lies posteriorly in the mitochondrial mass and gives rise to nine satellite projections and their Y-shaped connective extensions.The tail contains the 9 + 2 tubule arrangement and tapers at its distal end.This investigation was supported by a National Research Council grant to F. A new rust fungus,Spumula serispora, is described. The morphology of the teliospores and the telia was studied. In the teliospores ofS. serispora andS. quadrifida, the type species of the genus, sterile cells could be found, which are comparable to apical cells of otherRaveneliaceae. Similar sterile cells were also present in the telial heads ofRavenelia texensis. The ontogeny of the teliospores ofS. serispora and the teliospore heads ofRavenelia texensis was studied and compared. In both species the ontogeny of the spores heads was similar. Summary The role of the paraventricular nucleus (PVN) and biogenic amines (BA) in regulating the level of corticoids in the serum of osmotically stressed mallard ducks (Anas platyrhynchos) was analyzed employing three experimental approaches: 1) pharmacologic alteration of central BA levels, 2) microscopic evaluation of BA distribution, and 3) placement of electrolytic lesions into the PVN. Reserpine and -methyl-p-tyrosine (mpt), agents that decrease the amount of BA's in the central nervous system, produced a fivefold increase in the concentration of serum corticoids. Conversely, pargyline and amphetamine, agents that increase the functional pool of BA's, prevented the rise in serum corticoid concentration normally observed in birds challenged with an intraperitoneal injection of hypertonic saline. When the topographic distribution of BA's was analyzed in the brains of osmotically stressed and nonstressed ducks distinct changes in the intensity of catecholamine (CA) fluorescence were observed in only one location, the PVN of the hypothalamus. Additionally, electrolytic lesions stereotaxically placed in the PVN blocked the osmotic stress-induced rise in serum corticoid concentration. These data therefore indicate that the PVN in the mallard duck plays some role in regulating the observed stress-induced rise in serum corticoid concentration, and that this regulatory function is probably inhibited by catecholamines.This research was supported by research grant No. GB 33321 from the National Science Foundation. Summary The relationship between the inhibition of repair of radiation-induced DNA damage and the inhibition of recovery from radiation-induced potentially lethal damage (PLD) by hypertonic treatment was compared in 9L/Ro rat brain tumor cells. Fed plateau phase cultures were-irradiated with 1500 rad and then immediately treated for 20 min with a 37° C isotonic (0.15 M) or hypertonic (0.50 M) salt solution. The kinetics of repair of radiation-induced DNA damage as assayed using alkaline filter elution were compared to those of recovery from radiation-induced PLD as assayed by colony formation. Hypertonic treatment of unirradiated cells produced neither DNA damage nor cell kill. Post-irradiation hypertonic treatment inhibited both DNA repair and PLD recovery, while post-irradiation isotonic treatment inhibited neither phenomenon. However, by 2 h after irradiation, the amount of DNA damage remaining after a 20 min hypertonic treatment was equivalent to that remaining after a 20 min isotonic treatment. In contrast, cell survival after hypertonic treatment remained 2 logs lower than after isotonic treatment even at times up to 24 h. These results suggest that the repair of radiation-induced DNA damageper se is not causally related to recovery from radiation-induced PLD. However, the data are consistent with the time of DNA repair as an important parameter in determining cell survival and, therefore, tend to support the hypothesis that imbalances in sets of competing biochemical or metabolic processes determine survival rather than the presence of a single class of unrepaired DNA lesions.  相似文献

The microtubule (MT) arrangement in Closterium acerosum cells was observed by indirect immunofluorescence microscopy both during and following cell division, and during cell expansion without cell division. (During the division period, some cells of this alga divide whereas other cells expand in their middle region without division.) Before septum formation, all cells had a ring-like MT bundle (MT ring) in their middle. Both septum formation and expansion without cell division occurred at the position of this ring. During the periods of division, short, hair-like MTs appeared around the nucleus in some of the cells, in addition to the MT ring. In dividing cells, spindle MTs appeared as the chromosomes were condensed. During the early stages of expansion of the semicells, after cell division, the spindle MTs assumed a radial arrangement, moved, and settled in a position between the daughter chloroplasts. These MTs disappeared about 1.5 h after septum formation. As the new semicells were growing, wall MTs appeared, arranged transversely along the expanding wall. These transverse MTs disappeared gradually 4–5 h after septum formation, and only an MT ring remained near the boundary between the new and old semicells. The MT ring was present until the next cell division or expansion without cell division. During the latter course of development, transverse wall MTs were present only at the band-like expanding region. At the earlier stage of expansion without cell division, the short, hair-like MTs remained around the nucleus, but as time passed, both the hair-like MTs and, somewhat later, the transverse ones disappeared and only the MT rings remained. The remaining MT ring was not always positioned at the boundary between the expanding and the old cell region. Summary Neurons in cultures of central nervous tissue exhibited marked structural changes when exposed to hypertonic solutions. Cellular reactions were described in living neurons as well as after fixation and staining in preparations observed with both the light and electron microscope. The structures involved in these changes were mainly the nucleolus, the nucleus and the Nissl substance.Nucleolus In living neurons, observed with phase contrast optics, the nucleolus became invisible in hypertonic medium. This change occurred within a few seconds, and it was reversible when the cells were brought back to isotonic solutions. Fixation of the cells while exposed to hypertonic solution caused the nucleolus to reappear as a granular body. In stained preparations it appeared as a more irregular body in contrast to the smoothly outlined nucleolus in normal cells. In electron microscopic preparations of neurons which were fixed while exposed to hypertonic solutions the nucleolus was visible only as nucleolar shadow, overlaid by a few small irregular bodies of higher electron density than other nuclear contents.Nucleus The nuclear membrane of living neurons exposed to hypertonic media lost much of its sharp definition and became rather hazy in outline. The nuclear diameter increased about 10% in hypertonic medium, and the nuclear space became somewhat denser when observed with the phase contrast microscope. In Nissl stained preparations the nuclear space was filled with many small granular or rod-shaped bodies in contrast to the clear vesicular appearance of the nuclei of untreated cells. In electron microscopic preparations the nuclear space exhibited a spotty appearance due to the presence of electron dense and light areas.Nissl Substance In living neurons immersed in hypertonic solutions the Nissl substance showed a slight increase in phase density, especially after repeated changes between hypertonic and isotonic solutions. Sometimes a distinct striation in the Nissl substance appeared. In Nissl stained preparations there was no marked change observed in comparison with normal cells. However, in the electron microscope, the Nissl substance of hypertonically treated cells exhibited a marked structural change. The membrane-bound spaces of the endoplasmic reticulum assumed a rather precise orientation parallel to the cell membrane so that in extreme cases a concentric arrangement of endoplasmic cisternae was observed. The normal arrangement of ribosomal granules in rosettes and clusters became disturbed and the granules were more uniformly distributed.The cells as whole units showed a distinct shrinkage in hypertonic solution which may account for the more crowded appearance of various organelles such as mitochondria and Golgi complexes. There was also a marked increase in agranular reticulum profiles and small membrane bound vesicles in treated cells. The karyotypes of three taxa from thePhleum alpinum group of sect.Phleum (P. alpinum subsp.rhaeticum, 2n = 14,P. commutatum, 2n = 28, and informally namedP. commutatum, 2n = 14) were investigated by Giemsa C-banding. The overall similarity of diploid genomes suggests thatP. alpinum subsp.rhaeticum andP. commutatum are closely related — their karyotypes vary only with respect to their average amounts of telomeric heterochromatin. TheP. commutatum genome contains less telomeric heterochromatin than the genome ofP. alpinum subsp.rhaeticum, but in theP. alpinum group as a whole almost fluent transition between low (1.5%) and high (25.5%) amounts of telomeric heterochromatin was observed among populations. In the karyotype of tetraploidP. commutatum, seven distinguishable chromosome types were observed. Each of them is represented at somatic metaphase by four chromosomes. C-band structure of karyotype and average amount of telomeric heterochromatin suggest that this taxon has originated from hybridization between two related diploid forms of theP. commutatum — P. alpinum complex.  相似文献

Summary Male Wistar-Kyoto rats were given either tap water (control) or 3%-alanine (taurine-depleted) for three weeks. To prepare for the kidney function studies, the animals were then implanted with femoral vessels and bladder catheters. Two days after surgery, each rat was given an intravenous infusion of saline at the rate of 50l/min and urine samples were collected at specific time intervals. An isotonic saline solution (0.9% NaCl) was infused for determination of baseline parameters and was followed by the infusion of a hypotonic saline solution (0.45% NaCl). Two days later, the infusion protocol was repeated in the same animals; however, a hypertonic saline solution (1.8% NaCl) was substituted for the hypotonic saline solution. Renal excretion of fluid and sodium increased in the control, but not taurine-depleted, rats during the hypotonic saline infusion. Interestingly, diuretic and natriuretic responses were similar between the groups during hypertonic saline infusion. The results suggest that taurine-depletion in rats affects renal excretory responses to a hypotonic, but not a hypertonic, saline solution.  相似文献

The seed storage globulins from sixHelianthus and four hybrids were studied using mono and bidimensional gel SDS electrophoresis (+ 2 mercaptoethanol). The polypeptide composition of each subunit was determined. Different pairs are specifically expressed according to the species studied. Three typical patterns were discriminated. All the studied species exhibit five subunits: two of them are expressed in all the species (11 and 22). The subunit corresponding to the 11 pair is present inH. petiolaris and in the three populations ofH. annuus studied. The 2b2 pair is common toH. annuus andH. argophyllus. H. petiolaris presents two specific 2a2 and 44 pairs andH. annuus a specific 33 pair. InH. argophyllus 11 33 or 44 are never observed but are replaced by 13 and 31 pairs. Some globulins, poorly represented, are of forms but present chains of higher molecular weights (in the range 54–56 kDa). Expressing variations in the banding patterns between these species by the use of a similarity index reveals complete identity between the three populations ofH. annuus. Identity between the twoH. petiolaris studied is also observed.H. annuus andH. argophyllus appear to be closer to each other thanH. petiolaris concerning the seed storage globulins.  相似文献

Summary Five isolatedZea mays sperm cells, taken from the same population as used for a previous morphometric study, were serially ultrathin sectioned and computer-reconstructed to yield three-dimensional images as well as quantitative data. All cells were found to be essentially spherical and to contain a full complement of cell constituents except plastids and microtubules. The nuclei of three cells were highly curved into a C or V shape while the other two nuclei were not curved, but were more spherical to disc shaped. The three curved nuclei were heterochromatic in appearance, the other two were more euchromatic. Mitochondria were closely associated with the nuclei, were predominately in the form of large, variously shaped complexes, and ranged in number from 7 to approximately 74 per cell. Dictyosomes tended not to be close to the nucleus and ranged in number from 6 to 23 per cell. The endoplasmic reticulum was similarly not typically associated with the nucleus, and varied from extensive sheet-like areas to small membranous whorls. In addition to confirming the findings of previous studies on isolated corn sperm cells and providing new three-dimensional and distribution data, the results of the present work underscore the existence of a high degree of morphological variability amongZea mays sperm cells of a population.Abbreviations ER endoplasmic reticulum - SD standard deviation  相似文献
