Photoregulation of chalcone synthase (CHS) mRNA accumulation was analysed in parsley (Petroselinum crispum) and mustard (Sinapis alba) plants at different developmental stages. In both species, CHS mRNA accumulation in young etiolated seedlings was primarily under phytochrome control. In leaves of adult re-etiolated plants, a UV-B photoreceptor was predominantly involved in photocontrol. The reduced red light control in mature leaves was not due to the absence of immunoreactive phytochrome. The apparent dependence of photoreceptor usage on the developmental state of the cell or organism was in accordance with observations on the photoregulation of fusion constructs between CHS promoters from parsley or mustard and the β-glucuronidase reporter gene (GUS). When tested in the parsley protoplast transient expression system, both constructs yielded the same type of photoregulation as observed for the endogenous CHS gene.  相似文献

Host plant relations of the monophagous weevil Ceutorhynchus constrictus Marsh. (Coleoptera: Curculionidae: Ceutorhynchinae) feeding on garlic mustard, Alliaria petiolata (Bieb.) Cavara & Grande (Cruciferae) were studied in the laboratory. Most other crucifers were rejected in choice tests using garlic mustard as a reference plant, but Brassica nigra, Sinapis alba and Thlaspi arvense were as acceptable as the host plant. Flowering plants of Descurainia sophia were acceptable while young plants of this species were not. The most important feeding stimulants in extracts of garlic mustard were uncharged, water soluble compounds. The most abundant glucosinolate in garlic mustard, sinigrin, was a feeding stimulant, too. However, the feeding stimulatory activity of sinigrin was only expressed in the presence of still unidentified uncharged compounds from garlic mustard leaves. Host plant relations in monophagous crucifer-feeding insects is discussed in relation to the distinctness of glucosinolate patterns found in their host plants.
Zusammenfassung Ceutorhynchus constrictus Marsh. (Coleoptera: Chrysomelidae: Ceutorhynchinae) ist ein monophager Rüsselkäfer, der an Knoblauchhederich frisst. Das Wirtswahl-Verhalten dieses Käfers ist im Labor untersucht worden. Die meisten Crucifiren waren im Wahlversuche nicht akzeptiert, wenn Knoblauchhederich als Vergleichspflanze vorhanden war. Von Brassica nigra, Sinapis alba, und Thlaspi arvense wurden im Vergleich gleiche Mengen verzehrt wie von der Wirtspflanze. Blühende Descurainia sophia Pflanzen wurden, im Gegensatz zu Jungpflanzen der gleichen Art, angenommen. Die wichtichsten Phagostimulanten in Extrakten von Knoblauchhederich-Blättern waren ungeladene, wasserlösliche Substanzen. Das häufigste Glukosinolat im Knoblauchhederich, Sinigrin, war auch ein Phagostimulant. Doch war die phagostimulierende Wirkung von Sinigrin nur in Kombinationen mit noch nicht identifizierten, ungeladenen Substanzen aus Knoblauchhederich-Blätter nachweisbar. Wirtspfanzen-Beziehungen von monophagen Insekten werden diskutiert im Zusammenhang mit der Eigenart des Glukosinolat-Inhaltes ihrer Wirtspflanzen.

The lack of availability of sources of resistance against Alternaria brassicicola within the family Brassicaceae has made oilseed mustard plants a target for one of the most damaging and widespread fungal diseases, Alternaria black spot. Of the other non‐host‐resistant/tolerant plants, Sinapis alba, white mustard, is considered to be the most important apart from Arabidopsis. To understand the defence response of S. alba upon incompatible interaction with this pathogen, a functional genomic approach using cDNA amplified fragment length polymorphism was performed. The highly reproducible bands, found to be either more amplified or uniquely present in infected S. alba plants compared with non‐infected plants, were further subjected to comparative reverse Northern analysis in the incompatible white mustard (S. alba) and compatible India mustard (Brassica juncea L.) plants. The suppression of 46% of the genes in the compatible background indicates the possibility of effective and specific recognition of Alternaria in S. alba. Analysis of the 118 genes up‐regulated specifically in infected S. alba compared with B. juncea showed that 98 genes have similarity to proteins such as receptor‐like protein kinase genes, genes involved with calcium‐mediated signalling and salicylic acid‐dependent genes as well as other genes of known function in Arabidopsis. The apparent expression profile data were further confirmed for selected genes by quantitative real‐time polymerase chain reaction analysis. Classification of these genes on the basis of their induction pattern in Arabidopsis indicates that the expression profile of several of these genes was distinct in S. alba compared with B. juncea.  相似文献

White mustard (Sinapis alba L.) plants, susceptible to 2,4-D, and scentless mayweed (Tripleurospermum maritimum L.) plants, resistant to 2,4-D, markedly differ in the distribution and metabolism of 2,4-D. When 2,4-D-14C was applied onto the leaf, radioactivity was found in mustard after 8 days in the entire plant, whereas in scentless mayweed radioactivity occurred mainly in the leaf onto which it was applied and in another one or two leaves, with the roots showing only traces of radioactivity. The two plant species differed both in the character and in the number and amount of metabolites detected in aqueous and ether fractions. The metabolite with Rf 0.63 -0.65, soluble in ethylether (39% of the total radioactivity applied), and that with Rf 0.35 -0.36 (23% of the total radioactivity) were present exclusively in the roots of scentless mayweed plants. The Rf values established indicate that the former metabolite may be a conjugate with the amino acids alanine and valine and the latter a hydroxylated derivate of 2,4-D. In the shoots of both plants, 2,4-D-14C was metabolized to identical metabolites with Rf 0.59 -0.61, which occurred in mustard plants only in trace amounts. Free 2,4-D-14C occurred in the shoots of both plants and in considerable amounts also in mustard roots; it could not be demonstrated in the roots of scentless mayweed. The two species did not differ in the uptake rate or in the amount of absorbed 2,4-D-14C.  相似文献

Thioglucoside glucohydrolase (EC 3.2.3.1; myrosinase) hydrolyses glucosinolates and thereby liberates glucose and sulphur and nitrogen compounds. To examine the hypothesis that the myrosinase-glucosinolate system is influenced by environmental factors, the effect of sulphate on the expression of myrosinases was examined. On examining different plant organs at various stages, it was observed that sulphate induces a differential expression of myrosinase polypeptides in plants ofSinapis alba L. (white mustard). Specific myrosinase polypeptides, dependent on sulphate in the growth medium, were detected on immunoblots. Without sulphate a maximum of three polypeptides was detected in buds, two in cotyledons and one in stems and roots. In plants cultured on medium with sulphate up to four polypeptides could be observed in cotyledons, five polypeptides in buds, two in stems and one in roots. Expression of myrosinases was, in general, high in plants cultured on a medium supplemented with sulphate. In floweringS. alba plants, sulphate-starved plants showed a higher expression of myrosinase in cotyledons and stems compared to plants fed with sulphate. Sulphate-fed plants had a high expression in inflorescences and roots. The organ- and time-specific induction of the myrosinase expression is discussed in relation to sulphate metabolism and availability of sulphate under normal conditions of cultivation and in relation to protection of Brassicaceae species. This is the first evidence for a specific induction of individual myrosinase proteins.  相似文献

Fragments of chloroplast DNA from mustard (Sinapis alba L.) seedlings, generated by the restriction endonuclease Eco RI, were used to assess the frequency of complementary sequences in mustard RNA by DNA/RNA hybridization. A pronounced increase in hybridization to a single DNA fragment was found with RNA from seedlings irradiated with continuous far-red light, compared to RNA from dark-grown seedlings.  相似文献

Corn stunt spiroplasma (CSS) was transmitted by the leafhopper vector Euscelidius variegatus (Kirschbaum) and produced symptoms on four dicotyledonous plant species, Sinapis alba L. (mustard), Pisum sativitm L. (pea), Raphanus sativus L. (radish) and Spinacia oleracea L. (spinach). The vectors became infective by microinjection with a broth culture of CSS. This insect also acquired CSS from infected mustard plants and transmitted it to healthy ones.  相似文献

The occurrence of beet mild yellowing virus (BMYV) on feeding- and sugar-beet in Czechoslovakia has been proved. The virus was transmitted by aphidMyzus persicae (Sulz.) on indicator plantsSinapis alba L.,Capsella bursa-pastoris Medik, andClaytonia perfoliata Donn and from these plants back to sugar-beet cv. ‘Dobrovická A.’ A weed plantRaphanus raphanistrum L. was identified as a new natural host plant of BMYV. Rhizomania caused by Beet necrotic yellow vein virus (BNYVV) is one of the most devastating sugar beet diseases. Sugar beet plants engineered to express a 0.4 kb inverted repeat construct based on the BNYVV replicase gene accumulated the transgene mRNA to similar levels in leaves and roots, whereas accumulation of the transgene-homologous siRNA was more pronounced in roots. The roots expressed high levels of resistance to BNYVV transmitted by the vector, Polymyxa betae. Resistance to BNYVV was not decreased following co-infection of the plants with Beet soil borne virus and Beet virus Q that share the same vector with BNYVV. Similarly, co-infection with the aphid-transmitted Beet mild yellowing virus, Beet yellows virus (BYV), or with all of the aforementioned viruses did not affect the resistance to BNYVV, while they accumulated in roots. These viruses are common in most of the sugar beet growing areas in Europe and world wide. However, there was a competitive interaction between BYV and BMYV in sugar beet leaves, as infection with BYV decreased the titres of BMYV. Other interactions between the viruses studied were not observed. The results suggest that the engineered resistance to BNYVV expressed in the sugar beets of this study is efficient in roots and not readily compromised following infection of the plants with heterologous viruses.  相似文献

The qualitative and quantitative composition of sterols in the free form and esterified to fatty acids was studied in seed oils from Brassica napus, B. campestris, B..iuncea, B. nigra, Sinapis alba and S. aruefisis (Brassica kaber). Sitosterol, followed by campesterol, predominated in both the free and the esterified sterols. The free sterols were richer in brassicasterol (ca 10–20%) than the steryl esters (3–10%). Small amounts of δ5-avenasterol and δ7-stigmastenol were also found in the Brassica oils, often more in the esterified than in the free form. The quantity of sterols was studied only in Brassica campestris, which had ca 0.3 % in the free as well as in the esterified form. In Sinapis alba, ca 10% of the sterols in the free form and 20 % in the esterified sterols were δ5-avenasterol. This compared to only a few per cent in both the free and esterified sterols in the Brassica oils. Similarly, ca 2 % of cholesterol was found among the sterols of Sinapis alba but only traces in the Brassica oils. The similarity of sterol compositions among the cultivated brassicas and wild mustard (Sinapis arvensis), and the specific characteristics of the sterols in white mustard (Sinapis alba) adds further weight to the suggestion that wild mustard should be treated as Brassica kaber and strengthens the generic separation of Sinapis alba.  相似文献

When examining plants of white mustard (Sinapis alba L.), a significant relationship between the size of the roots and the yield was found, both in a pot experiment and in a field one. The yield was influenced by the size of the root system during and after flowering time. Especially the number of seeds has increased, and the average weight of seeds showed the same tendencies after the flowering time. The productivity of plants with an above-average yield was increasing more quickly than the size of their root system. From the beginning of the vegetation till the end of flowering these most efficient plants showed the lowest permitivity dispersion and the highest dispersion after the flowering time. The dispersion has increased towards the end of vegetation.  相似文献

Abstract Phytochrome-mediated anthocyanin synthesis of the mustard seedling (Sinapis alba L.) was investigated. Light pre-treated and dark-grown seedlings differing in responsiveness and level of phytochrome (Ptot) were compared. The data obtained support the traditional view that a seedling measures the amount of Pfr. The alternative view that a plant measures the Pfr/Ptot ratio does not seem to be compatible with the data obtained with the mustard seedling.  相似文献

The cabbage stem flea beetle (CSFB), Psylliodes chrysocephala L. (Coleoptera: Chrysomelidae), is one of the most important pests in European winter oilseed rape production. Adult beetles feed on young leaves whereas larvae mine within the petioles and stems. Larval infestation can cause significant crop damage. In this study, the host quality for CSFB of four oilseed rape (Brassica napus L.) cultivars and seven other brassicaceous species with different glucosinolate (GSL) profiles was assessed under controlled conditions. Larval instar weights and mortality were measured after 14 and 21 days of feeding in the petioles of test plants. To study the impact of GSL on the performance of larvae, the GSL contents in petioles from non-infested and infested plants were analysed before, and 21 days after, the start of larval infestation. Larval performance was not significantly different between the four cultivars of oilseed rape, but differed considerably among the other brassicaceous species tested. In comparison to the weight of larvae in the standard B. napus cv. Robust, the larval weight was higher in turnip rape (Brassica rapa L. var. silvestris) and significantly reduced in white mustard (Sinapis alba L.), oil radish (Raphanus sativa L. var. oleiformis), and cabbage (Brassica oleracea L. convar. capitata var. alba). The duration of larval development increased in white mustard and oilseed radish. The GSL profiles of the petioles showed little difference between non-infested and infested plants of oilseed rape whereas the content of aliphatic GSL increased in the infested turnip rape plants. In contrast, the aliphatic and benzenic GSL decreased in infested Indian rape (B. rapa subsp. dichotoma Roxb.). Larval weight was not correlated with the total GSL content of plants, neither before infestation nor 21 days after. Larval weight was positively correlated with progoitrin and 4-hydroxyglucobrassicin. White mustard, which provides inferior host quality for larval development, has the potential to introduce insect resistance into high-yielding oilseed rape cultivars in breeding programmes.  相似文献

Extracts from developing seeds, seedings, and different organsof mature plants of Sinapis alba L. were tested for the presenceof myrosinase. Three different patterns of isoenzymes were distinguishedby isoelectric focusing. One was found in developing seeds andseedlings, a second in mature leaves, stems, flowers, and podsand a third in roots. Sections of all the parts tested were examined for the presenceof myrosin cells by light microscopy. Developing seeds and seedlingsshowed a good correlation between the appearance of myrosinaseand the presence of myrosin cells. No myrosin cells were foundin root or flower parts although they showed myrosinase activity.  相似文献

In roots, the “hidden half” of all land plants, gravity is an important signal that determines the direction of growth in the soil. Hence, positive gravitropism has been studied in detail. However, since the 19th century, the response of roots toward unilateral light has also been analyzed. Based on studies on white mustard (Sinapis alba) seedlings, botanists have concluded that all roots are negatively phototropic. This “Sinapis-dogma” was refuted in a seminal study on root phototropism published a century ago, where it was shown that less then half of the 166 plant species investigated behave like S. alba, whereas 53% displayed no phototropic response at all. Here we summarize the history of research on root phototropism, discuss this phenomenon with reference to unpublished data on garden cress (Lepidium sativum) seedlings, and describe the effects of blue light on the negative bending response in Thale cress (Arabidopsis thaliana). The ecological significance of root phototropism is discussed and the relationships between gravi- and phototropism are outlined, with respect to the starch-statolith-theory of gravity perception. Finally, we present an integrative model of gravi- and blue light perception in the root tip of Arabidopsis seedlings. This hypothesis is based on our current view of the starch-statolith-concept and light sensing via the cytoplasmic red/blue light photoreceptor phytochrome A and the plasma membrane-associated blue light receptor phototropin-1. Open questions and possible research agendas for the future are summarized.  相似文献

Summary Sexual and somatic hybrid plants have been produced between Sinapis alba L. (white mustard) and Brassica napus L. (oil-seed rape), with the aim to transfer resistance to the beet cyst nematode Heterodera schachtii Schm. (BCN) from white mustard into the oil-seed rape gene pool. Only crosses between diploid accessions of S. alba (2n = 24, Sa1Sa1) as the pistillate parent and several B. napus accessions (2n = 38, AACC) yielded hybrid plants with 31 chromosomes. Crosses between tetraploid accessions of S. alba (2n = 48, Sa1Sa1Sa1Sa1) and B. napus were unsuccessful. Somatic hybrid plants were also obtained between a diploid accession of S. alba and B. napus. These hybrids were mitotically unstable, the number of chromosomes ranging from 56 to more than 90. Analysis of total DNA using a pea rDNA probe confirmed the hybrid nature of the sexual hybrids, whereas for the somatic hybrids a pattern identical to that of B. napus was obtained. Using chloroplast (cp) and mitochondrial (mt) DNA sequences, we found that all of the sexual F1 hybrids and somatic hybrids contained cpDNA and mtDNA of the S. alba parent. No recombinant mtDNA or cpDNA pattern was observed. Three BC1 plants were obtained when sexual hybrids were back-crossed with B. napus. Backcrossing of somatic hybrids with B. napus was not successful. Three sexual hybrids and one BC1 plant, the latter obtained from a cross between a sexual hybrid and B. napus, were found to show a high level of BCN resistance. The level of BCN resistance of the somatic hybrids was in general high, but varied between cuttings from the same plant. Results from cytological studies of chromosome association at meiotic metaphase I in the sexual hybrids suggest partial homology between chromosomes of the AC and Sa1 genomes and thus their potential for gene exchange.  相似文献

Studies on the glutamine synthetases (GS, EC 6.3.1.2) of green (GS2) and etiolated leaves (GSet) ofSinapis alba L. (cv. Steinacher) revealed striking similarities between the respective enzyme proteins. The enzymes showed corresponding chromatographic properties, both on dimethylaminoethyl-Sephacel and on hydroxylapatite columns. The purified GS proteins were also identical with regard to the molecular weight of their subunits. Isoelectrofocusing of pure GSet yielded two distinct polypeptide bands in the pH 5.6 region of the gels. This pattern corresponded to the two strong bands of GS2. Two charge variants of GS polypeptides could be detected by Western-blot analysis of the soluble protein of green leaves using antibodies against mustard GS2. In immunoprecipitation experiments, the holoenzymes of GS2 and GSet were recognized with identical affinities by this antiserum. We conclude that strong similarities exist between the proteins of the GS enzymes in green and etiolated leaves of mustard. Most probably only one GS form, namely the plastidic enzyme, can be found in the epigeal organs ofSinapis. The polypeptides of the GS2 subunits showed no differences in the hydrophobicity of the polypeptide chains. Neither glucosyl nor mannosyl residues could be detected. Dedicated to Professor Dr. H. A study was conducted to determine whether the cuticles in two genera of the family Cruciferae are effective barriers to infection by Rhizoctonia solani, and whether differences in cuticle and epidermal cell wall thickness and morphology of epicuticular wax exist between resistant and susceptible cultivars. As Canola/rapeseed (Brassica napus) and mustard (Sinapis alba) plants develop from 1 to 3 weeks of age, they become increasingly resistant to R. solani AG2-1 seedling root rot. Seven-day-old seedlings of S. alba cultivars are invariably more resistant than B. napus cultivars. Brassica napus cultivars do not show an obvious cuticle layer at 1 week but at 3 weeks the presence of a cuticle is seen through autofluorescence with a concomitant increase in resistance to R. solani. Removal of the cuticle from 3-week-old hypocotyls by chloroform treatment results in a decrease in cuticular autofluorescence and a significant increase in disease severity in both resistant and susceptible cultivars. Three-week-old plants of S. alba have a much lower percent disease rating and a significantly (p=0.05) thicker cuticle layer than similar-age plants of B. napus. The results suggest that the cuticle plays an important role in the resistance of S. alba and older plants of B. napus to infection by R. solani.  相似文献

Seeds of Sinapis alba L. were germinated in darkness for 3 dand a part of the etiolated seedlings were transferred to long-dayconditions for 6 weeks. Myrosinase solutions were prepared forcotyledons, hypocotyls, primary roots, leaves, stems, inflorescences,and seeds, and used to hydrolyse sinigrin. Glucose, one of the cleavage products, was determined by fourdifferent spectrophotometric methods, and their usefulness forcalculations of myrosinase activity in crude plant extractsis considered. Specific activity was calculated in relations to protein, andit was found to be about 30% higher in seedlings and also higherin seeds than in adult plants. Of the organs, those with thehighest activity were the hypocotyls and the stems. The differentparts of the plant contained different numbers of isoenzymes,as shown by polyacrylamide gel electrophoresis. Seedlings yieldedtwo, and adult plants four or five isoenzymes.  相似文献

The beet western yellows virus (BWYV) was identified in sugar beet plants with leaf yellowing symptoms. When transmitted toSinapis alba L. the virus isolate caused severe symptoms of yellowing and violetting of the interveinal leaf tissue of this plant. By aphidsMyzus persicae (Sulz.) the virus isolate was transmitted toLactuca sativa L.,Raphanus sativus L. var.radicula Pers.,Baphanus sativus L. ssp.sativus L. ap., and toBrassica oleracea L. var.gemmifera DC. InLactuca sativa plants the virus induces a yellowing along with thickenning and brittleness of leaves and with mild dwarfing of the plants. InBaphanus sativus var.radicula andBaphanus sativus ssp.sativus plants it brings about a yellowing of the leaf margins with a change in consistency as was the case in lettuce, and inBrassica oleracea var.gemmifera it causes violet spots on the lower leaf sides. The transmission was proved in repeated experiments by a backtransmission to beet andSinapis alba and further transmission from beet toSinapis alba. The transmission of the virus isolate toVicia faba L.,Datura stramonium L., andPetunia hybrida hort. was unsuccessful. In the course of transmissions the isolate properties did not change. In its host range the virus resembles the Duffus’ strain 3 BWYV, isolated from beet in the U.S.A. This is the first characteristic of an Europian BWYV isolate, as obtained from naturally infected beet plants.  相似文献
