Chlorella vulgaris Beyerinck (Emerson's strain), fails to grow in the dark even when sugars are provided. This phenomenon was clearly demonstrated in the alga, C. vulgaris, for which the growth rate in darkness on a glucose medium remained constant for 2 days and then declined to approach zero. Pigment concentrations also declined in darkness. Changes in flow rate of 1% CO2-in-air from zero to 7 ml per minute caused a progressive increase in the dark growth rate over a 5-day period, but did not maintain growth in the dark. Rates above 7 ml per minute produced no changes in growth rates.

White light intensities below the compensation point of the alga maintained heterotrophic growth. The saturation value for this response was 0.8 μw/cm2. White light also initiated growth in nongrowing cultures transferred from darkness to light.

The action spectrum for heterotrophic growth indicated a porphyrin as the active pigment. Light in the 425 mμ region was 4 times as effective as white light in stimulating heterotrophic growth. A secondary peak of growth stimulation occurred in the 575 mμ region.

The respiration of glucose by the alga was stimulated by low intensities of white light. This response was not immediate, but was clearly present after the third day of incubation.

Malonate and cyanide were inhibitory to growth of C. vulgaris on inorganic medium or glucose medium under 300 ft-c of white light. These data suggested that succinic dehydrogenase and cytochrome oxidase systems were present.

Substances inhibitory to growth were excreted into the medium under dark-growth conditions, and 2 of these substances were indentified as formic and acetic acids.

The evidence suggested that respiration of glucose cannot proceed for an extended period of time in darkness. The reason for this is postulated to be the lack of a cytochrome or a cytochrome precursor.

Three strains of Caldariomyces fumago were immobilized in 4% k-carrageenan and tested for semicontinuous production of chloroperoxidase (CPO). Over an 80-day period, growing in defined medium, C. fumago strains CMI 89362 and ATCC 11925 produced enzyme concentrations of 99 and 71 mg/liter, respectively, during six production periods of 12 to 14 days, while C. fumago DAOM 137632 produced only 24 mg of CPO per liter during six growth periods of 10 days. CPO production was unaffected by various regimens of washing between transfers. Mycelial growth was primarily restricted to the head surface, and bead size increased linearly with time. Attempts to restrict growth but maintain CPO production were unsuccessful. Pigment production, fructose utilization, and pH change in the immobilized cell cultures compared closely with the growth characteristics of free cell cultures. By using an airlift tower fermentor with an external loop run with continuous medium replacement of 20 ml/h (D = 0.016), strain CMI 89362 in bead form produced CPO at 40 mg/liter for 11 days.  相似文献

Growth of Prasinocladus marinus (Cienk.) Waern was maximal in media of 35‰ salinity, showing a five-fold increase over a 7-day period. At lower salinities (10–27.5‰) growth varied between 55–90% and at 45‰ was 85% of that in ‘normal’ sea water. Yield of extracellular carbohydrate was higher than in ‘normal’ sea water in lowered salinities and at 45‰ over a 42-day period. Larger quantities of extracellular carbohydrate were obtained at all stages of growth in media of 15‰, compared with those of 35‰ and 45‰. Whilst growth forms were markedly different at low salinities, DNA measurements indicated that similar quantities of cell material were present. Mannitol and glucose were the principal carbohydrates identified in intracellular material, and glucose in the extracellular products. Chloroplast pigment synthesis appears unaffected by growth at 15‰, 35‰, and 45‰.  相似文献

Highly chlorophyllous photomixotrophic callus was visually selected from callus originating from soybean (Glycine max (L.) Merr. var. Corsoy) cotyledon. Suspension cultures initiated from this callus became photoautotrophic under continuous light with an atmosphere of 5% CO2 (balance air). Dry weight increases of 1000 to 1400% in the 2-week subculture period have been observed. The cellular Chl content ranged from 4.4 to 5.9 micrograms per milligram dry weight which is about 75 to 90% of the Chl content in soybean leaves under equivalent illumination (300 micro-Einsteins per square meter per second).

No growth can be observed in the dark in sucrose-lacking medium or in the presence of 0.5 micromolar 3-(3,4-dichlorophenyl)-1,1-dimethylurea, a concentration which does not inhibit heterotrophic growth (on sucrose). Photoautotrophic growth has an absolute requirement for elevated CO2 concentrations (>1%). During the 14-day subculture period, growth (fresh weight and dry weight) is logarithmic. Photosynthesis quickly increases after day 4, reaching a peak of 83 micromoles CO2 incorporated per milligram Chl per hour while dark respiration decreases 90% from day 2 to day 6. The pH of the growth medium quickly drops from 7.0 to 4.5 before slowly increasing to 5.0 by day 14. At this pH range and light intensity (200-300 microEinsteins per square meter per second), no O2 evolution could be detected although at high pH and light intensity O2 evolution was recorded.

The effects of selenium (0.01, 0.5, 1, 5 and 10 mg/liter) on the growth and ultrastructure of the microalga Dunaliella salina were investigated following its transfer into clean water. Selenium concentrations of 5 and 10 mg/liter were toxic to D. salina, and reinoculation of microalga into clean water did not prevent it from total mortality. When reinoculated from medium with 0.01 mg Se/liter, the cell population density of D. salina was restored in 14 days. The number of ultrastructural alterations in cells was the same as in the control, while the excretory activity of microalga between days 4 and 10 of this experiment was higher. Cell population growth of D. salina transferred from 0.5 and 1 mg Se/liter was lower than in the control. No ultrastructural defects were observed in microalga reinoculated from medium with a selenium concentration of 0.5 mg/liter and the excretion level corresponded to that at 0.01 mg/liter. Various types of ultrastructural damage were found in microalga from medium with 1 mg Se/liter, which was previously reported to be threshold for D. salina; however, the number of cell injuries decreased with increasing time in clean medium. Excretory activity was decreased at the beginning of experiment; but after 7 days, it was restored to the control level. Though there were no ultrastructural alterations in microalgal cells from medium with 0.5 mg Se/liter, we assume that they had molecular defects that could inhibit the cell population growth. The study of microalgae following their reinoculation from medium containing toxicants into clean medium can be a useful method for evaluating algal survival after toxic exposure.  相似文献

Photosynthesis is a fundamental process sustaining heterotrophic organisms at all trophic levels. Some mixotrophs can retain functional chloroplasts from food (kleptoplasty), and it is hypothesized that carbon acquired through kleptoplasty may enhance trophic energy transfer through increased host growth efficiency. Sacoglossan sea slugs are the only known metazoans capable of kleptoplasty, but the relative fitness contributions of heterotrophy through grazing, and phototrophy via kleptoplasts, are not well understood. Fitness benefits (i.e. increased survival or growth) of kleptoplasty in sacoglossans are commonly studied in ecologically unrealistic conditions under extended periods of complete darkness and/or starvation. We compared the growth efficiency of the sacoglossan Elysia viridis with access to algal diets providing kleptoplasts of differing functionality under ecologically relevant light conditions. Individuals fed Codium fragile, which provide highly functional kleptoplasts, nearly doubled their growth efficiency under high compared to low light. In contrast, individuals fed Cladophora rupestris, which provided kleptoplasts of limited functionality, showed no difference in growth efficiency between light treatments. Slugs feeding on Codium, but not on Cladophora, showed higher relative electron transport rates (rETR) in high compared to low light. Furthermore, there were no differences in the consumption rates of the slugs between different light treatments, and only small differences in nutritional traits of algal diets, indicating that the increased growth efficiency of E. viridis feeding on Codium was due to retention of functional kleptoplasts. Our results show that functional kleptoplasts from Codium can provide sacoglossan sea slugs with fitness advantages through photosynthesis.  相似文献

Growth medium components and cultivation conditions for the extremely thermophilic bacterium Thermotoga neapolitana were optimized. A defined marine salts medium was formulated. Trace amounts of iron stimulated growth of T. neapolitana, while zinc inhibited growth at concentrations exceeding 11.1 μM. Other trace metals had no effect on its growth. Of the vitamins tested, only biotin was required for optimal growth. A defined mineral medium containing 5 g of carbohydrates per liter as the carbon source and 0.5 g of cysteine per liter as the sulfur source and reductant supported growth. Growth was stimulated by inclusion of vitamin-free Casamino Acids. Elemental sulfur, cystine, and dimethyl disulfide in the growth medium enhanced growth. Elemental sulfur and cystine relieved growth inhibition by hydrogen. T. neapolitana formed colonies in 2 days on plates of complex medium solidified with gellan gum and in 4 days on defined medium. The efficiency of plating was determined when growing cultures were sampled both aerobically and anaerobically and plated under aerobic and anaerobic conditions. Mean plating efficiencies were improved by sampling the growing cultures under strictly anaerobic conditions. Little or no improvement was obtained by inoculating plates inside an anaerobic chamber. Plating efficiencies of approximately 80% were obtained. Polycarbonate jars with aluminum lids withstood repeated incubation at 77°C without significant deterioration of the anaerobic seal and provided the most consistent results.  相似文献

In vitro growth of explant (juice vesicle or albedo tissues) cultures from citron (Citrus medica), lemon (C. limon), grapefruit (C. paradisi), sweet orange (C. sinensis), and mandarin (C. reticulata) fruits was stimulated by addition of orange juice (10% v/v optimum) to a basal medium containing Murashige and Skoog salts, 50 grams per liter sucrose, 100 milligrams per liter myo-inositol, 5 milligrams per liter thiamine·HCl, 2 milligrams per liter 2,4-dichlorophenoxyacetic acid and 0.5 milligrams per liter kinetin. In analyzing this effect of orange juice on citron explant cultures, we failed to obtain increased yields by addition of appropriate concentrations of citric acid to the basal medium but obtained growth stimulation when the medium was supplemented with juice from an “acidless” orange variety (cv. Lima). These facts suggest that some component(s) other than citric acid is involved. Addition of the inorganic ash corresponding to 10% (v/v) orange juice to the basal medium had no effect on yields. Similarly, the stimulatory effect of orange juice could not be explained based on its content of sucrose or of organic growth factors already present in the basal medium.  相似文献

The short term effects of increased levels of CO2 on gas exchange of leaves of bigtooth aspen (Populus grandidentata Michx.) were studied at the University of Michigan Biological Station, Pellston, MI. Leaf gas exchange was measured in situ in the upper half of the canopy, 12 to 14 meters above ground. In 1900 microliters per liter CO2, maximum CO2 exchange rate (CER) in saturating light was increased by 151% relative to CER in 320 microliters per liter CO2. The temperature optimum for CER shifted from 25°C in 320 microliters per liter CO2 to 37°C in 1900 microliters per liter CO2. In saturating light, increasing CO2 level over the range 60 to 1900 microliters per liter increased CER, decreased stomatal conductance, and increased leaf water use efficiency. The initial slope of the CO2 response curve of CER was not significantly different at 20 and 30°C leaf temperatures, although the slope did decline significantly during leaf senescence. In 1900 microliters per liter CO2, CER increased with increasing light. The light saturation point and maximum CER were higher in 30°C than in 20°C, although there was little effect of temperature in low light. The experimental results are consistent with patterns seen in laboratory studies of other C3 species and define the parameters required by some models of aspen CER in the field.  相似文献

The effectiveness of iron compounds on growth inhibition of Sphaerotilus species was compared. In this study, two strains of Sphaerotilus were tested with different iron concentrations in a synthetic sewage (S-medium) as formulated by Lackey and Wattie (Sewage Works J. 12:669-684, 1940). For both strains, >80% inhibition of the maximum respiration rate was obtained by the following levels of soluble iron concentrations at pH 6.0: iron citrate, 20 mg/liter as Fe; iron cysteine, 5 mg/liter as Fe; and ferrous sulfate, 10 mg/liter as Fe. At a pH of 6.7 with iron citrate (20 mg/liter as Fe), inhibition of both strains was in excess of 50%. Insoluble iron compounds, such as iron hydroxides and ferrous carbonate, were found to be much less effective than the soluble iron compounds as inhibitors of these two strains. Aged iron hydroxide (500 mg/liter as Fe) produced a 70% inhibition in the maximum respiration rate while fresh iron hydroxide (52 mg/liter as Fe) and ferrous carbonate (500 mg/liter as Fe) produced a 20% inhibition. Chemical analyses of the iron-inhibited Sphaerotilus strains showed a close relationship between the inhibition of the organism's growth and the amount of iron sorbed by the organism.  相似文献

The submerged aquatic plant Isoetes howellii Engelmann possesses Crassulacean acid metabolism (CAM) comparable to that known from terrestrial CAM plants. Infrared gas analysis of submerged leaves showed Isoetes was capable of net CO2 uptake in both light and dark. CO2 uptake rates were a function of CO2 levels in the medium. At 2,500 microliters CO2 per liter (gas phase, equivalent to 1.79 milligrams per liter aqueous phase), Isoetes leaves showed continuous uptake in both the light and dark. At this CO2 level, photosynthetic rates were light saturated at about 10% full sunlight and were about 3-fold greater than dark CO2 uptake rates. In the dark, CO2 uptake rates were also a function of length of time in the night period. Measurements of dark CO2 uptake showed that, at both 2,500 and 500 microliters CO2 per liter, rates declined during the night period. At the higher CO2 level, dark CO2 uptake rates at 0600 h were 75% less than at 1800 h. At 500 microliters CO2 per liter, net CO2 uptake in the dark at 1800 h was replaced by net CO2 evolution in the dark at 0600 h. At both CO2 levels, the overnight decline in net CO2 uptake was marked by periodic bursts of accelerated CO2 uptake. CO2 uptake in the light was similar at 1% and 21% O2, and this held for leaves intact as well as leaves split longitudinally. Estimating the contribution of light versus dark CO2 uptake to the total carbon gain is complicated by the diurnal flux in CO2 availability under field conditions.  相似文献

Although most upwelling regions are marked by strong fluctuations in water temperature, few studies have examined how episodic cold-water events affect the physiology and ecology of benthic marine invertebrates. I tested the hypothesis that upwelling-related variation in water temperature regulates the feeding, growth, and energetics of two rocky intertidal predators, the sea star Pisaster ochraceus (Brandt, 1835) and the whelk Nucella canaliculata (Duclos, 1832). Sea stars and whelks were maintained in laboratory tanks at a constant 9 °C, a constant 12 °C, and a treatment that simulated the Oregon coast upwelling regime by cycling between 14-day periods of 12 and 9 °C. Early in the experiments, sea stars and whelks held at 9 °C consumed about 30% fewer mussels (Mytilus trossulus) than those in warmer tanks. Despite lower consumption by whelks in colder tanks, 9 and 12 °C individuals attained the same final size. Similarly, sea stars in 9 °C tanks showed greater growth per gram of mussel tissue consumed than individuals held at 12 °C. These results suggest that reduced consumption under colder conditions was balanced by reduced metabolic costs. Moreover, there appeared to be an energetic advantage to living in the temperature regime characteristic of intermittent upwelling. Sea stars alternately exposed to 12 and 9 °C had a significantly higher growth rate, conversion efficiency, and storage of reserves in the pyloric caeca than individuals in the constant 12 °C tanks. Whelks maintained under fluctuating temperatures tended to grow faster than those held at constant 12 or 9 °C, although this trend was not statistically significant (p=0.069). These results suggest that benthic consumers experiencing cyclic temperatures may feed intensely during periods of warmer water while benefiting from reduced metabolic costs during cold-water intrusions. Because the fecundity of Pisaster and Nucella is a function of energy stored during the upwelling season, interannual variability in upwelling patterns could alter the reproductive output of these species.  相似文献

The biological activity of the catholyte and anolyte of double distilled water was studied in experiments on the germination of wheat grains in the period from March to May. The activity of the solutions, which was characterized by a growth index, was high early in this period, then decreased almost to zero in the middle of the period, and then increased to about the initial value by the end of the period. Throughout, the efficiency of the anolyte of double distilled water generally exceeded the efficiency of the catholyte. Early and late in the period, the stimulatory effect of the anolyte exceeded that of the catholyte by a factor of 5–5.5. The changes in the biological activity of the catholyte and anolyte of double distilled water were also compared with the changes in the biological activity of the catholyte of nutrient medium M9. The stimulatory effect of the catholyte of the nutrient medium was evaluated from the change in the growth of E. coli cells. Early in the period at a cultivation temperature of 20°C, the stimulatory effect determined from the increase in the optical density of the cell suspension in the experiment with respect to a reference value was 55–60%. Next, the stimulatory effect decreased almost to zero in the middle of the period and increased to approximately initial value by the end of the period. It was assumed that the physicochemical mechanisms of action of the catholyte and anolyte of double distilled water on the wheat seed germination and of the catholyte of the nutrient medium on E. coli cell growth are of different nature.  相似文献

Clava multicornis Forskål (Cnidaria, Hydrozoa) from the North Sea was cultured under a variety of environmental conditions, and quantitative aspects of the following processes examined: food intake, growth, oxygen consumption, losses of material, and food conversion. The experiments were conducted in sea water (salinity 32 ‰) at different constant temperature levels (6°, 11° and 16° C) and different daily food rations. The polyps were fed living larvae of the brine shrimpArtemia salina. Daily rations ranged from 2.3 % (6° C) to 19.0 % (16° C) of the dry weight of the polyp colonies. The food ration essential for minimum growth increased with the test temperature. The calorific value of theArtemia larvae was 5854 cal per g organic dry substance. The calorific values of the colonies ofClava multicornis increased at all 3 test temperatures with ascending daily food rations; they ranged from 5367 to 6003 cal per g organic dry substance. Colony growth was determined in 3 different ways: by measuring the increase in polyp number, the length increase of all polyps of a given colony, and the increase of the dry weight of the organic substance of a given colony. Growth was exponential in all 3 cases. The lowest test temperature, or small daily rations, caused slow growth; the highest temperature, or large daily rations, resulted in rapid growth. Oxygen consumption of individual colonies was measured at 16° C and 3 different daily rations; the colonies showed the same intensity of respiration at all 3 daily rations. A colony of 1.5 mg organic dry substance respired 0.107 ml oxygen per 24 hours, a colony of 5.0 mg, 0.269 ml oxygen. At 11° and 16° C gonophores developed well and were counted; at 6° C no gonophores were observed. The amount of the excrement discharged byC. multicornis at 16° C increased from 26.0 % of the food eaten (minimum daily ration) to 39.3% (maximum daily ration). Gross efficiency increased with falling temperature and rising daily ration. At 16° C, net efficiency increased with rising daily ration. On the basis of the data obtained for gross efficiency, oxygen consumption and excrementation, an energy budget was made up.  相似文献

Two mutants of Clostridium thermocellum were isolated after UV light mutagenesis. Mutant A1, selected as asporogenous, exhibited a fermentation pattern similar to that of the wild type. However, at pH 6.5, the mutant degraded 12% more cellulose than did the wild type, leading to enhanced ethanol production. Mutant 647, selected as ethanol tolerant, was able to grow in medium containing 4% ethanol. During the early stage of the exponential growth phase, ethanol was produced as the main product, up to a concentration of about 9 g/liter. After 3 days of culture, 48.3 g (89% of the initial amount) of degraded cellulose per liter was fermented into 12.7 g of ethanol per liter.  相似文献

Upon exposure to 2°C, the leaves and crowns of rye (Secale cereale L. cv `Puma') and wheat (Triticum aestivum L. cv `Norstar' and `Cappelle') increased in cold hardiness, whereas little change in root cold hardiness was observed. Both root and shoot growth were severely reduced in cold-hardened Norstar wheat plants frozen to −11°C or lower and transplanted to soil. In contrast, shoot growth of plants grown in a nutrient agar medium and subjected to the same hardening and freezing conditions was not affected by freezing temperatures of −20°C while root growth was reduced at −15°C. Thus, it was apparent that lack of root development limited the ability of plants to survive freezing under natural conditions.

Generally, the temperatures at which 50% of the plants were killed as determined by the conductivity method were lower than those obtained by regrowth. A simple explanation for this difference is that the majority of cells in the crown are still alive while a small portion of the cells which are critical for regrowth are injured or killed.

Suspension cultures of Norstar wheat grown in B-5 liquid medium supplemented with 3 milligrams per liter of 2,4-dichlorophenoxyacetic acid could be cold hardened to the same levels as soil growth plants. These cultures produce roots when transferred to the same growth medium supplemented with a low rate of 2,4-dichlorophenoxyacetic acid (<1 milligram per liter). When frozen to −15°C regrowth of cultures was 50% of the control, whereas the percentage of calli with root development was reduced 50% in cultures frozen to −11°C. These results suggest that freezing affects root morphogenesis rather than just killing the cells responsible for root regeneration.

In order to reduce the cost of Bacillus thuringiensis bioinsecticide-process production, a new medium only composed of starch, soya bean and diluted sea water, providing particularly 7.5 g/l NaCl, was optimized. The results obtained with cultures carried out into 1000 ml shake flasks showed 7% improvement of delta-endotoxin production in such cheap medium compared to that containing a mixture of pure minerals instead of sea water. Interestingly, this new formulated medium was shown to be efficient for delta-endotoxin production by several B. thuringiensis strains exhibiting various larvicidal activities towards either Diptera or Lepidoptera. Moreover, the use of such medium for large scale production of bioinsecticides was also evidenced in an automated full controlled 3 l fermenter. So that it was shown that sea water did not affect such requirements and have positive effect on growth, sporulation and delta-endotoxin synthesis. This should contribute to a significant reduction of the cost of B. thuringiensis bioinsecticide production and utilisation.  相似文献

The growth of marine diatoms Phaeodactylum tricornutum was investigated on a medium with artificial sea water under artificial and natural light. The maximum specific growth rate was 0.7 day–1, the productivity was 0.8 g/L day, and the maximum biomass was 3.86 g/L under artificial light in laboratory conditions. In the conditions of Crimea, the maximum productivity of P. tricornutum amounted to 6 g/m2 day under natural light in an outdoor photobioreactor (pool). The results of cultivation of P. tricornutum in a pool with artificial seawater under natural light may serve as a basis for developing technologies for the industrial cultivation of algae.  相似文献

Candida ingens, a pellicle-forming yeast utilizing volatile fatty acids, grew over a pH range of 4.1 to 6.0 on nonsterile supernatants from anaerobically fermented pig wastes; growth was inconsistent between pH 4.1 and 4.6. When ambient temperature above the pellicle was 21°C and the temperature of the medium was 29 to 32°C, a pH range of 4.8 to 5.0 gave yields of 1.90 to 3.31 g of dry matter per liter, and 0.059 to 0.065 mol of volatile fatty acids was utilized per liter. There was no advantage in utilization of volatile fatty acids and yield of dry matter in keeping the pH constant during a 24-h growth period. C. ingens grew at pH 4.8 and 5.0 when both ambient and medium temperatures were 30°C. When ambient temperature was 10°C, maximum yield and utilization of volatile fatty acids occurred at a medium temperature of 28 to 30°C.  相似文献

The effect of the component concentrations of a synthetic medium on acetone and butanol fermentation by Clostridium acetobutylicum ATCC 824 was investigated. Cell growth was dependent on the presence of Mg, Fe, and K in the medium. Mg and Mn had deleterious effects when in excess. Ammonium acetate in excess caused acid fermentation. The metabolism was composed of two phases: an acid phase and a solvent one. Low concentrations of glucose allowed the first phase only. The theoretical ratio of the conversion of glucose to solvents, which was 28 to 33%, was obtained with the following medium: MgSO4, 50 to 200 mg/liter; MnSO4, 0 to 20 mg/liter; KCl, 0.015 to 8 g/liter (an equivalent concentration of K+ was supplied in the form of KH2PO4 and K2HPO4); FeSO4, 1 to 50 mg/liter; ammonium acetate, 1.1 to 2.2 g/liter; para-aminobenzoic acid, 1 mg/liter; biotin, 0.01 mg/liter; glucose, 20 to 60 g/liter.  相似文献
