Summary The effects of abscisic acid (ABA) on the orientation and cold stability of cortical microtubules (MTs) in epidermal cells of epicotyls of the dwarf pea,Pisum sativum L. cv. Little Marvel, were examined by immunofluorescence microscopy. The effect of ABA on the elongation of epicotyls and on the orientation of cortical MTs was opposite to that of gibberellin A3 (GA3). Treatment with ABA, which reduced the promotion of epicotyl elongation by GA3, eliminated the GA3-induced predominance of transverse MTs and resulted in a predominance of longitudinal MTs. The effect of ABA on the cold stability of cortical MTs was also opposite to that of GA3. ABA increased the cold stability of MTs, while GA3 decreased it. The predominance of longitudinal MTs brought about by ABA may have some relationship to ABA-induced inhibition of the elongation of the epicotyl. Actinomycin D inhibited the elongation of epicotyl segmentsfrom azuki bean seedlings that was induced by simultaneous treatmentwith IAA and GA3. The drug also inhibited the elongation ofthe segments that was caused by IAA alone when it was appliedtogether with IAA. When the segments were pretreated with GA3and then incubated with IAA, GA3 promoted the elongation causedby IAA and brought about a predominance of transverse corticalmicrotubules (MTs) in the epidermal cells of the segments. Thechange in the arrangement of MTs caused by pretreatment withGA3 was evident 1 h after the start of subsequent incubationwith IAA when the effect of pretreatment with GA3 on the elongationhad not yet become apparent. Pretreatment with GA3 did not causeany change in the arrangement of MTs when GA3-pretreated segmentswere not incubated subsequently with IAA. Although actinomycinD applied before treatment with IAA did not inhibit the IAA-inducedelongation, the drug diminished the promotion of the elongationcaused by pretreatment with GA3 and prevented GA3 from bringingabout a predominance of transverse MTs when the drug was appliedduring the pretreatment with GA3. GA3-induced synthesis of mRNA seems to be involved in the promotionby GA3 of IAA-induced elongation and in the GA3-induced rearrangementof cortical MTs. Young seedlings of various citrus species give rise, after decapitation,to adventitious buds at the cut end of the epicotyl. With suchtreatment, more buds formed on seedlings of sweet orange, sourorange, and grapefruit than of lemon, mandarin, and calamondin.Anatomical observations at the cut stem end show formation ofbuds occurring from tissues close to the cambium. Growth regulatorsapplied at the cut epicotyl end modified the patterns of budformation. 6-Benzylaminopurine (BAP) promoted bud formation;indole-3-butyric acid (IBA), abscisic acid (ABA), and gibberellicacid (GA3) inhibited it. BAP reversed the inhibitory effectof GA3. Gibberellic acid caused a typical elongation of thesprout axis. The use of decapitated seedlings as a tool forstudying stem differentiation in citrus is suggested.  相似文献

Longitudinal microtubules are predominant in epidermal cellsof the 3rd internodes of dwarf pea (Pisum sativum L. cv. LittleMarvel) seedlings. In more than 50% of the cells, cortical microtubulesare running parallel to the cell axis. GA3 promotes elongation of the internodes and gives rise toa predominance of transverse microtubules. In more than 60%of the GA3-treatd cells, cortical microtubules are running transverseto the cell axis. Longitudinal microtubules in the GA3-untreated cells are resistantto low-temperature treatment, but transverse microtubules inthe GA3-treated cells are sensitive to it. Longitudinal microtubulesare present in GA3-treated epidermal cells with low frequency.They are resistant to low-temperature treatment. Longitudinal, oblique and transverse microtubules are presentwith almost the same frequency in epidermal cells of the 3rdinternodes of tall pea (cv. Early Alaska) seedlings. GA3 promoteselongation of the internodes also in tall pea seedlings, butit does not alter the direction of cortical microtubules sodistinctly as it does in dwarf pea seedlings. As in dwarf pea seedlings, longitudinal microtubules are resistantto low-temperature treatment, and transverse microtubules aresensitive to it in tall pea seedlings. The relationship between the dwarf habit of growth and corticalmicrotubule (MT) orientation as affected by gibberellin wasexamined using a gibberellin responding dwarf mutant of Zeamays L. (d5). The 4 mm portion of the mesocotyl below the coleoptilarnode of dark-grown seedlings was divided into four 1 mm segments.MTs were observed by means of immunofluorescence microscopyon the tangential surface of the epidermal cells (EP-t), theradial surfaces of epidermal (EP-r) and cortex cells (C-r) inboth normal and d5 seedlings. MT arrangement in EP-t was transverse,oblique and/or longitudinal. In lower regions, cells with transverselyoriented MTs decreased, while those with obliquely/longitudinallyoriented MTs increased. The frequency of the occurrence of transverselyoriented MTs was much higher in normal than in d5. In EP-r almostall the cells of the four regions had transversely orientedMTs. In d5 seedlings only a few percent of cells had obliquelyoriented MTs. In C-r all the cells of the four regions of normalseedlings had transversely oriented MTs, while in d5, 20% ofthe uppermost region cells showed oblique/longitudinal orientationand more in lower regions. Gibberellin A1, (GA1), GA19, and GA20 in phloem exudates andcotyledons of seedlings of Pharbitis nil cv. Violet, grown underdifferent photoperiodic conditions, were qualitatively and semi-quantitativelyanalyzed by a combination of high performance-liquid chromatography(HPLC) and radioimmunoassays (RIA). The levels of GA19 and GA20were higher in cotyledons from plants grown under dark treatment(DT) conditons of 16 h-light/8 h-dark for 6 days followed by8 h-light/16 h-dark for 3 days than in those grown under continuouslight (CL) for 9 days. This relationship was also observed forthe GAs in phloem exudates, although the levels were much lowerthan in the cotyledons. When GAs were applied to the cotyledons,elongation of the epicotyl was promoted more by GA20 than byGA1 or GA19, especially under the CL treatment. Four 13-hydroxygibberellins, gibberellin A1 (GA1), 3-epi-GA1,GA19 and GA20 were identified by full-scan GC/MS in extractsof lettuce seedlings (Lactuca sativa L. cv. Grand Rapids). Theresults suggest that the early-13-hydroxylation biosyntheticpathway to GA1 functions in the lettuce seedlings. It was alsofound that GA1 is active per se in the control of hypocotylelongation in lettuce seedlings. To investigate the relationshipbetween control by light of hypocotyl elongation and levelsof endogenous GAs in lettuce, endogenous levels of GAs werequantified by radioimmunoassay in seedlings that had been grownfor 5 days in the dark (5D) and in those that had been grownfor 4 days in the dark and then under white light for 1 day(4D1L). The endogenous level of GA1 in the upper and lower partsof hypocotyls in 5D seedlings was about four times higher thanthat in 4D1L seedlings. The response of explants (hypocotylsegments with cotyledons) from dark-grown seedlings to GA1 isknown to be similar in the dark and under white light when theexplants are treated with inhibitors of the biosynthesis ofGA. Therefore, the above information suggests that the highlevel of GA1 in hypocotyls of dark-grown seedlings is responsiblefor the rapid elongation of hypocotyl, while irradiation bywhite light decreases the endogenous level of GA1 in the hypocotylswith a resultant decrease in the rate of hypocotyl elongation. Phaseolus multiflorus plants at three stages of developmentwere decapitated either immediately below the apical bud orlower down at a point 1 cm above the insertion of the primaryleaves. Growth regulators in lanolin were applied to the cutstem surface. IAA always inhibited axillary bud elongation anddry-matter accumulation, and enhanced internode dry weight butnot elongation. GA3 applied below the apical bud greatly increasedinternode elongation and dry weight, but simultaneously reducedbud elongation and dry-weight increase. Application of GA3 1cm above the buds had no effect on bud elongation in the youngestplants, but enhanced their elongation in the two older groups.IAA always antagonized GA3-enhancement of internode extensiongrowth, whereas its effects on GA3-enhanced dry-matter accumulationdepended on the stage of internode development. Bud elongationwas greater in plants treated with GA3+IAA than in plants treatedonly with IAA, except in the youngest plants decapitated immediatelybelow the apical bud, where GA3 caused a slight increase inIAA-induced bud inhibition. GA3 increased inhibition of buddry weight by IAA in the two youngest groups of plants, butslightly reduced it in the oldest plants. No simple compensatorygrowth relationship existed between internode and buds. It wasconcluded that, (1) auxin appears to be the principal growthhormone concerned in correlative inhibition, and (2) availabilityof gibberellin to internode and buds is of importance as a modifyingfactor in auxin-regulated apical dominance by virtue of itslocal effects on growth in the internode and in the buds.  相似文献

Abscisic acid (ABA) applied to the decapitated second internodeof runner bean plants enhanced outgrowth of lateral buds onlywhen internode stumps were no longer elongating. Applied toelongating internodes of slightly younger plants, ABA causesinhibition of bud outgrowth. Together with 10–4 M indol-3-ylacetic acid (IAA), ABA stimulated internode elongation and interactedadditively in the inhibition of bud outgrowth. A mixture of10–5 M ABA and 10–6 M gibberellic acid (GA3 ) causedsimilar effects on internode growth as IAA + ABA, but was mutuallyantagonistic in effect on growth of the lateral buds. Effects of GA3 on the stability of cortical microtubules (MTs)were studied in mesocotyl cells of etiolated maize seedlings.Propyzamide, an MT-disrupting agent specific for plant tubulin,disrupted cortical MTs in cells in the upper regions of mesocotylsof GA3-untreated seedlings and caused swelling of the cells.GA3 prevented propyzamide from disrupting MTs and from causingsuch swelling. Chilling of mesocotyls at 4°C for 60 minbrought about the disruption of cortical MTs in cells in theupper regions of mesocotyls of GA3-untreated seedlings, butnot in cells in corresponding regions of GA3-treated seedlings,suggesting that treatment with GA3 increased the stability ofcortical MTs in maize mesocotyl cells. Cortical MTs in protoplastsisolated from mesocotyls of GA3-untreated seedlings failed towithstand chilling at 0°C for 90 min, while those in protoplastsisolated from mesocotyls of GA3-treated seedlings withstoodchilling successfully. Summary In epidermal cells of azuki bean (Vigna angularis) epicotyl segments, that were sequentially treated with an auxin-free solution and an auxin solution, cortical microtubules changed their orientation from longitudinal to transverse. The effects of light irradiation on the arrangement of corticalmicrotubules (MTs) were examined in etiolated A vena mesocotylsand coleoptiles, and in Pisum epicotyls. Elongation of A venamesocotyls ceased as a result of irradiation with white lightwithin 1 h. The predominant orientation of MTs became more longitudinalwithin 1 h in epidermal cells and changed from transverse tooblique, after the elongation ceased, in parenchymal cells.Irradiation with red and with blue light also caused cessationof cell elongation and the same changes in the orientation ofMTs. Elongation of Avena coleoptiles ceased as a result of irradiationwith white light within 24 h. The predominant orientation ofMTs became more longitudinal in epidermal cells and changedfrom transverse to oblique in parenchymal cells. The changein orientation of MTs in epidermal cells preceded that in parenchymalcells. In Pisum epicotyls, elongation ceased as a result ofirradiation with white light within 1 h. Although the orientationsof MTs in epidermal cells did not show any remarkable change,those in parenchymal cells changed from transverse to obliqueafter cell elongation ceased. The change in orientation of MTs and the cessation of cell elongationof A vena mesocotyls induced by white-light irradiation wereboth significantly retarded by treatment with IAA. A synthetic brassinosteroid, 22,23(S,S)-homobrassinolide (hBR),was examined for its interaction with IAA and GA3 in the elongationof hypocotyl sections of light-grown cucumber (Cucumis salivusL. cv. Aonagajibai) seedlings. hBR alone was less active thanIAA. Its optimal concentration was around 10 µM and thelowest effective concentration between 10 and 100 µM,which is more than 100 times higher than that of brassinolide.hBR was more active in sections from younger seedlings. Itsgrowth-promoting effect was negated or greatly reduced by inhibitorsof auxin-induced elongation such as p-chlorophenoxyisobutyricacid and kinetin. hBR acted synergistically with IAA and 2,4-Dbut not with GA3 showing only an additive effect. Sequentialtreatment of sections with hBR and then with IAA also resultedin synergistic enhancement of auxininduced elongation, but whenthe order of treatment was reversed, hBR was inactive. The synergisticeffect was obtained with 1 h pretreatment with hBR and couldbe reduced by subsequent washing with water. There was no sequentialinteraction between hBR and GA3. The synergistic pretreatmenteffects of hBR and GA3 were simply additive to each other. Amembrane-bound ATPase inhibitor, dicyclohexylcarbodiimide, inhibitedthe hBR-induced elongation, but did not affect GA3-induced elongation.The findings led to the conclusion that brassinosteroids enhanceauxin action and possess growth-promoting activity which isindependent of that of gibberellin. The levels of endogenous gibberellin A1 and A19 (GA1 and GA19)and abscisic acid (ABA) in three rice (Oryza sativa L.) cultivars,Nihonbare (normal), Tan-ginbozu (semid-warf) and Tong-il (semi-dwarf),were measured at various stages of internode elongation andear development. GA19 was the main GA in Nihonbare and Tong-ilthroughout the life cycle but was not detected in Tan-ginbozu.The levels of GA1 in the ears of all three cultivars were lowand reached their maxima after anthesis. Similarly, the earsof all three cultivars contained high ABA levels which peakedafter anthesis. Shoots contained low quantities of ABA throughoutthe life cycle. Evidence that the synthesis of GA3 is involved in the growthof floral orga'ns of Pharbitis nil is presented. GAs in floralorgans at different developmental stages were surveyed usingTLC followed by the bioassay with two dwarf rice seedlings,‘Tanginbozu’ and ‘Waito-C’. The amountof GAs in the petal and stamen increased rapidly after the petalemerged from calyx, reached a maximum 12 hr before anthesis,then declined markedly thereafter. The GA content in the calyxremained unchanged before and after anthesis, and that in thepistil increased after anthesis. Pharbitis flowers containedat least two active GAs, one of which was probably GA3, theother appeared to be GA19. GA3 was detected in relatively largeamounts in both the petal and stamen during their rapid elongation.In the calyx, which showed little increase in fresh weight duringrapid flower growth, GA9 was the dominant GA. Exogenously suppliedGA3 promoted elongation of sections in excised young filaments.Sucrose was necessary for definite growth promotion by GA3.GA19 had little effect on filament elongation, and IAA was ratherinhibitive. Microfibrils (MFs) on the inner surface of the walls of Avenacoleoptile and mesocotyl cells and of Pisum epicotyl cells wereexamined by a replica method. In the elongating epidermis ofthese three organs, cells having MFs that were transverse, obliqueor longitudinal to the elongation axis were intermingled. Inthe elongating parenchymal tissues, all cells deposited MFstransversely. In non-elongating cells of Avena coleoptiles andPisum epicotyls, the orientation of MFs on the inner wall surfaceof both epidermal and parenchymal cells was more longitudinalthan in elongating cells. These observations on the orientationsof MFs are compatible with those our previously reported observationson the orientations of microtubules (MT) (Iwata and Hogetsu1988). Disruption of MTs of Avena coleoptiles by treatment withamiprophosmethyl caused changes in the orientation of depositionof MFs. These results support the idea that MFs are usuallyco-aligned with MTs in organ cells and that the orientationof MFs is controlled by MTs. The averaged direction of MFs, visualized under polarized light,showed a clear difference between the epidermal and inner-tissuecell walls in the elongating regions of the three organs. Inalmost all elongating and non-elongating epidermal cells, theaveraged direction of MFs was longitudinal, while it was transversein all inner-tissue cells. Prohexadione calcium (BX-112) is a novel plant growth regulatorthat inhibits the late stages of the biosynthesis of gibberellinsin plants. Fourteen kinds of gjbberellin, helminthosporol and'helminthosporic acid were applied simultaneously with BX-112to rice seedlings (Oryza sativa L. ), and their growth-promotingactivities in terms of shoot elongation were examined. The growth-promotingactivities of GA1, GA4, GA18, GA22, GA23, GA38, helminthosporoland helminthosporic acid were not inhibited by BX-112, but thoseof GA5, GA9, GA15, GA19, GA20, GA31, GA44 and GA53 were inhibited.These results suggest that 3ß-hydroxylation is animportant and necessary step in the biosynthesis of gibberellinsthat promote shoot elongation in rice seedlings. The weak promotionof shoot elongation by GA22 in the presence of BX-112 suggeststhat the effect of a hydroxyl group at C-18 of GA22 might beable to mimic the effect of the 3ß-hydroxyl groupof GA1. Helminthosporol and helminthosporic acid may promotethe shoot elongation of rice by mimicking physiologically activegibberellins and not by stimulating their biosynthesis. 1Part I is the previous paper by Nakayama et al. The biological activity of a cell wall-derived xyloglucan nonasaccharide(XG9) was investigated using a bioassay with entire pea epicotyls(Pisum sativum cv. Progress). The xyloglucan fragment was foundto inhibit gibberellic acid-induced elongation of etiolatedpea epicotyls with maximum inhibition at concentrations rangingfrom 10–11 to 10–9M. Growth of etiolated epicotylsin the absence of exogenously applied GA3 was also inhibitedby XG9 in the same concentration range. A cell wall-derivedheptasaccharide (XG7) lacking the fucosyl-galactosyl-side chainshowed no inhibitory effect in the pea epicotyl bioassay withand without exogenous GA3. Furthermore, the biological activityof a synthetic pentasaccharide (XG5), containing the fucosylgalactosyl-sidechain which is necessary for the biological activity was investigatedin the same bioassay. Compared to XG9 the pentasaccharide hada similar inhibitory activity on GA3-promoted elongation aswell as on the endogenous growth in the absence of exogenouslyapplied GA3, but did not exhibit a distinct concentration optimum. Effects of ions on the orientation of cortical micro-lubules(MTs) in Spirogyra cells were studied. After depo-lymerizalionwith amiprophos-methyl (APM), MTs were allowed to reorganizein NaCI solutions of various concentrations. As the concentrationof NaCI increased, the frequency of cells that had oblique MTsincreased. When cells in NaCI solution were transferred intoartificial pond water (APW) and incubated for 6 h, all the MTschanged to become transverse to the longitudinal axis of thecell. KC1 and MgCl2 also had effects on the orientation of MTs.However, NH4Cl, CaCl2;, CoCl2, and Co(NO3)2 did not show anyeffect. These results suggest that Na+, K+, and Mg2+have effectson MT orientation and that NH+4, Ca2+, Co2+, Cl, andNO3 have little effect. When MTs were reorganized ineither NaCl or KCl solutions, all the oblique MTs were organizedinto an S-helix. In contrast, some of the oblique MTs were foundas a Z-helix in the cells incubated in MgCl2 or mannitol solutions.These results suggest that effects of Na+ and K+ on the orientationof MTs are not the same as those of Mg2+ and mannitol.
