Efforts to mitigate amphibian declines are hindered by a lack of information about basic aspects of their biology and demography. The effective to census population size ratio (N e /N c ) is one of the most important parameters for the management of wildlife populations because it combines information on population abundance and genetic diversity and helps predict population viability in the long term. Few studies have calculated this ratio in amphibians, which sometimes show low ratios, associated with a higher extinction risk. Here we integrate field-based (capture-mark-recapture studies, egg string counts) and molecular approaches (estimation of the effective number of breeders (N b ) and the effective population size (N e ) based on genotypes from larval cohorts and candidate parents) to produce the first estimates of the N e /N c and N b /N c ratios in two amphibians, the Iberian ribbed newt Pleurodeles waltl and the western spadefoot Pelobates cultripes. Additionally, we investigate sex-biased dispersal in both species based on direct (field observations) and indirect (genetic) evidence. Both species showed similar ratios, slightly lower in Pleurodeles (0.21–0.24) than in Pelobates (0.25–0.30). Observed displacement rates were low in both species (P. waltl?=?0.51%; P. cultripes?=?1.23%). We found no evidence for sex-biased dispersal in P. cultripes, but both direct and indirect evidences suggest a tendency for female-biased dispersal in P. waltl. We discuss differences in the genetic estimates of N e and N b provided by three inference methods and the implications of our findings for the management of these species, characteristic of Mediterranean wetlands in the Iberian Peninsula and listed as Near Threatened.  相似文献

Background

A frequent observation in molecular evolution is that amino-acid substitution rates show an index of dispersion (that is, ratio of variance to mean) substantially larger than one. This observation has been termed the overdispersed molecular clock. On the basis of in silico protein-evolution experiments, Bastolla and coworkers recently proposed an explanation for this observation: Proteins drift in neutral space, and can temporarily get trapped in regions of substantially reduced neutrality. In these regions, substitution rates are suppressed, which results in an overall substitution process that is not Poissonian. However, the simulation method of Bastolla et al. is representative only for cases in which the product of mutation rate μ and population size Ne is small. How the substitution process behaves when μNe is large is not known.

Results

Here, I study the behavior of the molecular clock in in silico protein evolution as a function of mutation rate and population size. I find that the index of dispersion decays with increasing μNe, and approaches 1 for large μNe . This observation can be explained with the selective pressure for mutational robustness, which is effective when μNe is large. This pressure keeps the population out of low-neutrality traps, and thus steadies the ticking of the molecular clock.

Conclusions

The molecular clock in neutral protein evolution can fall into two distinct regimes, a strongly overdispersed one for small μNe, and a mostly Poissonian one for large μNe. The former is relevant for the majority of organisms in the plant and animal kingdom, and the latter may be relevant for RNA viruses.

Through stochastic simulations, estimates of breeding values accuracies and response to selection were assessed under traditional pedigree-based and genomic-based evaluation methods. More specifically, several key parameters such as the trait’s heritability (0.2 and 0.6), the number of QTLs underlying the trait (100 to 200), and the marker density (1 to 10 SNPs/cM) were evaluated. Additionally, impact of two contrasting mating designs (partial diallel vs. single-pair mating) was investigated. Response to selection was then assessed in a seed production population (seed orchard consisting of unrelated selections) for different effective population sizes (Ne?=?5 to 25). The simulated candidate population comprised a fixed size of 2050 individuals with fast linkage disequilibrium decay, generally found in forest tree populations. Following the genetic/genomic evaluation, top-ranked individuals were selected to meeting the predetermined effective population size in target production population. The combination of low h2, high Ne, and dense marker coverage resulted at maximum relative genomic prediction efficiency and the most efficient exploitation of the Mendelian sampling term (within-family additive genetic variance). Since genomic prediction of breeding values constitutes the methodological foundation of genomic selection, our results can be used to address important questions when similar scenarios are considered.  相似文献

In territorial species, observed density dependence is often manifest in lowered reproductive output at high population density where individuals have fewer resources or are forced to inhabit low-quality territories. The Arctic fox (Vulpes lagopus) in Iceland is territorial throughout the year and feeds mostly on birds, since lemmings are absent from the country. Thus, the population does not exhibit short-term population cycles that are evident in most of the species’ geographical range. The population has, however, gone through a major long-term fluctuation in population size. Because of the stability in hunting effort and reliable hunting records since 1958, the total number of adult foxes killed annually can be used as an index of population size (N t ). An index of carrying capacity (K) from population growth data for five separate time blocks during 1958–2007 revealed considerable variation in K and allowed a novel definition of population density in terms of K, or N t /K. Correlation analysis suggested that the reproductive rate was largely determined by the proportion of territorial foxes in the population. Variation in litter size and cub mortality was, on the other hand, related to climatic variation. Thus, Arctic foxes in Iceland engage in typical contest competition but can adapt their territory sizes in response to both temporal and spatial variation in carrying capacity, resulting in surprisingly little variation in litter size.  相似文献

Analysis of genetic datasets can be particularly useful in providing guidelines for conservation management of understudied species targeted by commercial activities. Here we used population genetic approaches to inform on the conservation status of the Neotropical long-nose pirá catfish, Conorhynchos conirostris. Pirá is a large migratory fish endemic to the São Francisco River Basin (SFRB). It is an evolutionarily divergent and relict species, being the sole representative of an incertae sedis family. The species is considered locally extinct in the upper and lower SFRB, listed as vulnerable on the IUCN Red List and as endangered on the Brazilian Red List (ICMBIO). Fishing prohibition has received severe criticism from middle SFRB fisheries that claim that this understudied species is relatively abundant in that region. We used information from 13 microsatellite markers and COI mitochondrial sequences to clarify the genetic diversity of this enigmatic species in the middle SFRB, to estimate contemporary effective population size (Ne), and to assess its conservation status. Results from bottleneck analyses indicated that the species has experienced recent reductions in population size, which is consistent with small estimates of contemporary Ne. The predicted amount of heterozygosity loss (Ht) in t generations ranged from 0.1152 (for an estimated Ne of 26.4; t?=?100) to 0.7573 (for an estimated Ne of 169.9; t?=?10). Our study supports the conservation status proposed by the ICMBIO to the remaining pirá population. Moreover, we highlight the need for demographic data and the re-assessment of the current IUCN classification for this evolutionary relict lineage.  相似文献

The specific features of mating females and males of different morphs collected from a population of the wasp P. dominulus in Southern Ukraine were studied. Data analysis of mating individuals in experiments indicated a random mate choice by size and assortative mating by coloration. The most actively copulating males in the experiments had light-colored mesopleuron, sternite, and fore and middle coxae. They were similar in the color pattern and size to resident males, which defend their territories in the habitat. Most P. dominulus females mated only once. In their coloration, those females represented the majority of the individuals in the population. The color variability of the foundresses was shown to change in a cyclic pattern. The possibility that such variability may result from selective or assortative mating is discussed. The color and pattern traits of P. dominulus females and males could serve as markers of their reproductive strategies and their reproductive success on the whole.  相似文献

As an initial contribution to understanding the adaptive value of behavioral and life-history strategies, the life cycle and mating behavior of an unstudied species of tephritid fruit fly in the genus Rhagoletis are characterized for the first time. Over a 9-month fruiting period, a small proportion of Solanum appendiculatum Dunal (< 10 %) was found to be infested with a single larva of Rhagoletis solanophaga (Hernández & Frías). The average duration of R. solanophaga lifecycle (c.a. 140 days from egg laying to death of adults) exceeded the three month fruitless period. Additionally, R. solanophaga is capable of exploiting Solanaceous plants in at least two genera. These features could have selected for a non-diapausing species of Rhagoletis, a genus where most species are univoltine. Nevertheless, some individuals in the population became dormant. As other members of the genus, R. solanophaga exhibited a resource defense mating system with forced copulations and multiple mating. Both males and females could be highly promiscuous and individual mating success exhibited a wide range of outcomes. Regardless of mating success, mated females stored similar amounts of sperm in two spherical spermathecae. Long copulations were observed, perhaps functioning as a form of mate guarding with probable disadvantages for females. We outline hypotheses and opportunities for future comparative studies examining sperm competition and mate guarding.  相似文献

Bactrocera carambolae and B. dorsalis (Diptera: Tephritidae) are extremely destructive pests of fruits and vegetables in the Asia-Pacific region. Earlier reports have described that B. carambolae and B. dorsalis, respectively, use mainly star fruit and mango, suggesting a certain level of host partitioning which can be ascribed neither to differences in larval food qualities nor host-specific parasitoid mortality. This study specifically examined reproductive interference (antagonistic sexual interaction) between B. carambolae and B. dorsalis as a potential factor strongly affecting their host partitioning. We observed mating behaviors, especially interspecific courtships and mating, by cohabiting the conspecific and heterospecific pairs together. Consequently, we quantified their effects on the reproductive success of females. Males of both species frequently courted their own females, but they also courted females of other species. Courtship refusal by females was not selective in males of either species. This incomplete discrimination of both sexes led to frequent occurrences of interspecific sexual interactions in both species, but only B. carambolae females showed reduced reproductive success. These results suggest that B. dorsalis, superior in reproductive interference, can occupy high-quality mango, whereas B. carambolae, inferior in reproductive interference, must use low-quality star fruit.  相似文献

Knowledge of linkage disequilibrium (LD) is important for effective genome-wide association studies and accurate genomic prediction. Chinese Merino (Xinjiang type) is well-known fine wool sheep breed. However, the extent of LD across the genome remains unexplored. In this study, we calculated autosomal LD based on genome-wide SNPs of 635 Chinese Merino (Xinjiang type) sheep by Illumina Ovine SNP50 BeadChip. A moderate level of LD (r 2?≥?0.25) across the whole genome was observed at short distances of 0–10 kb. Further, the ancestral effective population size (N e ) was analyzed by extent of LD and found that N e increased with the increase of generations and declined rapidly within the most recent 50 generations, which is consistent with the history of Chinese Merino sheep breeding, initiated in 1971. We also noted that even when the effective population size was estimated across different single chromosomes, N e only ranged from 140.36 to 183.33 at five generations in the past, exhibiting a rapid decrease compared with that at ten generations in the past. These results indicated that the genetic diversity in Chinese Merino sheep recently decreased and proper protective measures should be taken to maintain the diversity. Our datasets provided essential genetic information to track molecular variations which potentially contribute to phenotypic variation in Chinese Merino sheep.  相似文献

Using the capillary electrophoresis of ISSR DNA fragments, the genetic structure of the adventitious populations of Helix pomatia L. (Mollusca, Pulmonata) inhabiting the territory of Eastern Europe was studied. A significant decrease in genetic heterogeneity (He = 0.095 ± 0.002) and population differentiation level (Fst = 0.074) was revealed in comparison with similar data obtained using the ISSR method with electrophoretic detection in agarose gel and allozymes in polyacrylamide gel. Similar to other methods, an effect of spatial isolation (RM =–0.476) was found on the basis of regression analysis between genetic and geographical distances. The effective population size, calculated with the help of M. Slatkin’s model, turned out to be the largest of the previously calculated values (Ne = 41.8), which indicates the high viability of the studied groups of Roman snails in newly developed territories.  相似文献

Sexual selection is believed to play a major role in speciation processes of cichlid fishes in the East African Great Lakes, in particular Lakes Malawi and Victoria, by driving diversification in male nuptial coloration. In Lake Tanganyika, a high rate of color pattern diversification is found in the genus Tropheus, where more than 100, mostly allopatric, color morphs have been described. Whether color pattern differentiation in Tropheus could follow from sexual selection as well, has not yet been conclusively shown. Unlike typical sexually selected species, Tropheus are sexually monomorphic, establish temporary pair bonds and spawn monogamously. Variance in mating success among individuals would still allow for sexual selection, but due to their high population census sizes, parentage data from natural populations are difficult to obtain. Simulations designed according to existing data on mating behavior in Tropheus suggested that variance in male reproductive success can be substantial and lead to levels of opportunity for selection, which are similar to values estimated, for example, from natural populations of sexually dimorphic songbirds. Variance in male success was mostly affected by the way females perceived and reacted to differences between males, and to a lesser extent by the duration of components of the reproductive cycles of males and females. These results indicate that future work on the importance of sexual selection in Tropheus should concentrate on mate choice cues and female mate choice behavior, but does not depend critically on the acquisition of more detailed life history data.  相似文献

The endangered fountain darter Etheostoma fonticola is found only in the Comal and San Marcos rivers in the Guadalupe River basin in central Texas, USA. Comal River fountain darters were believed to be extirpated following a severe drought in the 1950s and were reintroduced in the early 1970s using 457 darters from the San Marcos River. In this study we used 23 microsatellite loci to describe and evaluate the genetic diversity, population structure and effective population size (N e) of fountain darters. We also evaluated the genetic effect of the Comal River reintroduction and the influence of low-head dams (dams) on dispersal in both rivers. Bayesian analysis of individual genotypes and Analysis of Molecular Variation supported two distinct populations concordant with the two rivers. Estimates of N e were much smaller (<10 %) than census size (N c) in both rivers but did not indicate the populations are at risk of an immediate and rapid loss of genetic diversity. Coalescent-based estimates of the genetically effective number of founders (Nf) for the Comal River averaged about 49 darters and, together with the indices of genetic diversity and the bottleneck test (heterozygosity excess) results, were consistent with a founder event following the reintroduction in the Comal River. Finally, our results regarding the influence of dams on fountain darter dispersal were equivocal and did not support a conclusion. We recommend this issue be examined further as part of the fountain darter recovery program.  相似文献

Results are presented from laboratory modeling of the dynamics of space cyclotron masers. A selfoscillatory mode of cyclotron instability in the nonequilibrium plasma of an ECR discharge in a magnetic mirror trap is found. The plasma comprises two electron populations: the background population with a density of N e ~ 1013–1014 cm?3 and temperature of T e ≈ 300 eV and the energetic population with a density of N e ~ 1010 cm?3 and temperature of T e ≈ 10 keV. Quasi-periodic pulsed precipitation of energetic electrons from the trap, accompanied by microwave bursts at frequencies below the electron gyrofrequency in the center of the trap, is detected. The study of the microwave plasma emission and the energetic electrons precipitated from the trap shows that the precipitation is related to the excitation of whistler-mode waves propagating nearly parallel to the trap axis. The observed instability has much in common with phenomena in space magnetic traps, such as radiation belts of magnetized planets and solar coronal loops. The experimental results demonstrate the opportunity of laboratory modeling of space cyclotron masers. The main tasks and possibilities of such modeling are discussed.  相似文献

Clonal structure in clonal plants can affect sexual reproduction. Individual ramets can decrease reproduction if their neighbors are ramets of the same genet due to inbreeding depression or self-incompatibility. We assessed ramet reproductive success in the partial self-incompatible Ferocactus robustus (Cactaceae) as a function of floral display size in focal ramets and floral display size and clonal structure of their reproductive neighborhoods. Ramets were labeled, sized in number of stems, mapped and genetically identified through RAPD markers in one population. A pollen dispersal area of 15-m radius was established for each ramet to determine the clonal diversity in the neighborhoods. Flower production and fruit set were counted on a monthly basis during one reproductive season as a surrogate of ramet fitness. We expected a decrease in individual ramet reproductive success as a function of the number of reproductive ramets of the same genet in the neighborhood. A total of 272 sampled ramets revealed 116 multilocus genotypes, showing high clonal diversity in the population (G/N = 0.43, D = 0.98). Clonal diversity of neighborhoods ranged from 0.06 to 1 and fruit set varied from 0 to 76.9%. Individual ramet reproductive success was influenced by (1) mate availability, (2) floral display size of a genet within the reproductive neighborhood, and (3) the proportion of distinguishable genotypes. Floral display size of genets and ramets coupled with the genetic diversity within the reproductive neighborhood determines the low sexual reproduction in F. robustus.  相似文献

Roads can substantially impact the population connectivity of a wide range of terrestrial vertebrates, often resulting in loss of genetic diversity and an increase of spatial genetic structure. We studied the Western Diamond-backed Rattlesnake (Crotalus atrox), a large and abundant venomous predator, to test the hypothesis that a large and relatively new roadway in Arizona (Interstate Highway I-10) is a barrier that impacts gene flow and population genetics via habitat fragmentation. Based on 72 C. atrox sampled from three specific sampling sites (“subpopulations”) on both the west and east corridors of I-10, we used 30 nuclear microsatellite DNA loci and three mitochondrial DNA genes (2615 bp) to assess genetic diversity and structure, estimate effective population size (N e ), and describe patterns of gene flow. We found no evidence for loss of genetic diversity or a decrease in N e between the three subpopulations. Our microsatellite analysis showed that two subpopulations in close proximity (4 km), but separated by I-10, showed greater levels of genetic differentiation than two subpopulations that were separated by a greater distance (7 km) and not by I-10 or any other obvious barriers. Mitochondrial DNA analyses showed no significant genetic differentiation nor any indication of historically impeded gene flow. Tajima’s D and mismatch distribution tests revealed that demographic expansion is occurring in the overall population (all three subpopulations). Bayesian clustering and spatial genetic autocorrelation analyses of microsatellite data showed resistance to gene flow at the approximate location of I-10. Simulations that investigated gene flow between the subpopulations (with and without a highway barrier present) were consistent with our molecular results. We conclude that I-10 has reduced gene flow in a population of an important reptilian predator of the Sonoran Desert in southern Arizona and make conservation recommendations for reversing this trend.  相似文献

A trade-off between flight capability and reproduction is well known in adult females of the wing-dimorphic cricket Velarifictorus ornatus, but it is not clear whether such a trade-off exists in adult males of the species. In the present study, we investigated sexual maturation time, mating frequency, and the fertilization success of spermatophores after sequential mating in long-winged (LW) and short-winged (SW) adult males of V. ornatus to evaluate the potential reproductive advantage of the SW over the LW male morph. We found that the SW males of V. ornatus attained sexual maturity earlier and produced heavier spermatophores during the early stage after adult emergence than their LW counterparts. Additionally, within a 24-h mating period, the SW males showed a higher mating frequency, greater spermatophore weight, and shorter intermating time interval compared with their LW counterparts. Although females copulated with the two male morphs produced eggs of similar size, fertilization success by SW males was significantly higher than by the LW males. These results provide support for a trade-off between dispersal capability and reproduction success in wing-dimorphic males of V. ornatus.  相似文献

Background

Intra-specific variation in sperm length influences male reproductive success in several species of insects. In males of the malaria vector Anopheles gambiae, sperm length is highly variable but the significance of this variation is unknown. Understanding what determines the reproductive success of male mosquitoes is critical for controlling malaria, and in particular for replacing natural populations with transgenic, malaria-resistant mosquitoes.

Methods

A laboratory population of A. gambiae males was tested for intra-specific variation in sperm length. A full-sib quantitative genetic design was used to test for a genetic component of sperm length in A. gambiae males and estimate its heritability. This study also tested for a relationship between sperm length and male reproductive success in A. gambiae. Male reproductive success was measured as the proportions of inseminated and ovipositing females.

Results

There was intra-specific variation of sperm length in A. gambiae. There was no significant genetic variation in sperm length and its heritability was low (h2 = 0.18) compared to other insects. Sperm length was correlated with male body size (measured as wing length). Males with short sperm had significantly higher reproductive success than males with long sperm and this was independent of body size.

Conclusion

This is the first study to demonstrate intra-specific variation in sperm length in A. gambiae and that males with short sperm have higher reproductive success. That sperm length influences female oviposition is important for any strategy considering the release of transgenic males.

Comparing the ratio of effective number of breeders (N b ) to adult population size (N) among closely related coexisting species can provide insights into the role of life history on N b /N ratios and inform conservation programs towards limiting the loss of evolutionary potential in natural populations. We estimated N b and N in two coexisting salmonid fishes (Brook trout and Atlantic salmon) for 3–4 consecutive years in two small, adjacent streams in Newfoundland, Canada, using mark-recapture (N), linkage disequilibrium (N b(LD)), and sibship frequency approaches (N b(Sib) ). We found that N b /N ratios were about 20-fold greater in Atlantic salmon than in brook trout (mean 0.20, range 0.06–0.56 vs. mean 0.02, range 0.01–0.05, respectively). This difference was consistent across N b estimators. In addition, we found that removing migrants reduced N b : the strength of the effect was weak for N b(LD) and much stronger for N b(Sib). Our results highlight the importance of subtle ecological differences and gene flow in shaping N b /N. They also provide some evidence that the linkage between demographic and evolutionary processes varies between closely related taxa and suggest that a more complete understanding of the N b /N range across various species is an important component of conservation genetics and management.  相似文献

We investigated factors which may determine realised paternity following multiple mating in a promiscuous ladybird Harmonia axyridis (Pallas) (Coleoptera: Coccinellidae), namely the mating order and ability of males of different morphs to fertilize females. In experiments using males of two colour morphs, we found that the first male had a reproductive advantage over the second male (sperm precedence P2 = 0.38). Males of the melanic morph spectabilis had a reproductive advantage over males of the semi-melanic morph axyridis (P2 = 0.35). Analysis of paternity using microsatellites applied to young larvae, which avoids possible differential mortality of progeny, confirmed the precedence of the first male and higher paternity rate of colour morphs that are more frequent in the wild. Hatchability of eggs increased from 65% after the first mating to 85% after the second mating delayed by five days. Average duration of mating was 157 min and did not differ between the first and second mating and between the colour morphs of males.  相似文献

Females of some polygynous fish species display female nuptial signals (FNSs), advertising spawning readiness to potential mates. Halichoeres margaritaceus, a polygynous coral reef fish, reportedly display red belly color and a unique bobbing behavior as FNSs advertising spawning readiness. In this study, I examined whether FNSs were observed in an H. margaritaceus population in Australia. Females in this study displayed red belly color as an FNS, although at a lower intensity than that reported in a Japanese population. Contrary to observations made in the Japanese population, bobbing behavior performed by females in this study showed no temporal trend as spawning approached, indicating bobbing behavior did not function as an FNS in the Australian population. The role of local selective pressures on FNS intensity and lifetime reproductive benefits H. margaritaceus may potentially gain in utilizing FNSs are briefly discussed.  相似文献
