Studies of the effect of pentobarbital anesthesia on the radiation response have been performed using early generation isotransplants of three spontaneous tumors of the C3H mouse: a mammary carcinoma (MCaIV), a fibrosarcoma (FSaII), and a squamous cell carcinoma (SCCVII). The enhancement ratio of pentobarbital [ER(PB)] for TCD50 as the end point was greater than or equal to 1 for all conditions tested. The ER(PB) for O2 3 ATA conditions and two equal doses was 1.46, 1.72, and 2.21 for MCaIV, FSaII, and SCCVII, respectively. The ER(PB) using MCaIV was the same for O2 and carbogen at 1 or 3 ATA. Also, tumor size of MCaIV did not significantly affect the ER(PB) for O2 3 ATA conditions. Further, with the two-dose protocol the anesthesia and the hyperbaric oxygen needed to be used at the second dose; condition at the first dose was not critical. For fractionated irradiation of MCaIV (10 and 15 equal doses) the ER(PB) was smaller than for two-dose treatment; also the effect was less for intratumor temperature of 35 degrees C than 26-27 degrees C. There was no effect of the anesthesia on the acute response of normal skin of the leg. Lung damage by hyperbaric oxygen was not an important factor in these results. Additionally, ERs were computed for O2 at 3 ATA. This ER(O2 3 ATA) was larger for anesthesized than conscious mice. The ER(O2 3 ATA) for MCaIV was high (greater than 1.5) even for radiation given in 10 or 15 equal doses.  相似文献

The role of protein kinase C (PKC) in the carcinogenesis of human breast tissue has been studied at the molecular level for more than two decades. In this study, we employed Western blotting to determine the presence of PKC isoforms in cancerous and normal breast tissues. The results indicate significant expression of a conventional PKC (PKCα) and two atypical PKCs (PKC ζ and λ/ι) in both breast tumors and adjacent normal breast tissue. For the α,ζ and λ/ι isoforms, the expression of individual isoforms was higher in the breast tumors than in the adjacent normal breast tissue. Although the correlation coefficient was low, significant linear correlation was found among the activities of the isoforms. The data suggest a potential new direction in cancer chemotherapy, namely the blockage of the signal transduction pathway of specific PKC isoforms.  相似文献

In this study, laryngeal flow fields are investigated and compared in normal larynx and models of larynx with unilateral vocal fold paralysis (UVFP). In paralytic models, three fixed initial glottal gaps are considered to understand the positive or probable negative impacts of surgical operation on unilaterally paralytic larynx, by which the paralyzed vocal fold is brought closer to the mid-plane. Various features of the flow fields have been discussed in detail including glottal gap width, glottal flow rate, glottal exit pressure pattern and glottal jet evolution. The numerical solution of fluid-structure interaction is carried out using ANSYS, and the results confirm some of the favorable effects of surgery on the patient’s larynx. It is also shown that by tightening the glottal gap, some of the problems caused by the presence of a motionless vocal fold, such as leakage through glottal gap in the closure phase resulting in breathy voice can be moderated, although some of the symptoms of this disorder remain relatively unchanged.  相似文献

We studied the expression level and cell-specific expression patterns of 5alpha-reductase (5alpha-R) types 1 and 2 iso-enzymes in human hyperplastic and malignant prostate tissue by semi-quantitative RT-PCR and in situ hybridisation analyses. In situ hybridisation established that 5alpha-R1 mRNA is preferentially expressed by epithelial cells and little expressed by stromal cells whereas 5alpha-R2 mRNA is expressed by both epithelium and stroma. Semi-quantitative RT-PCR has been performed on total RNA from different zones of normal prostate, BPH tissues and liver. We found that 5alpha-R1 and 5alpha-R2 mRNAs expression was near the same in all zones of normal prostate. In BPH tissue, 5alpha-R1 and 5alpha-R2 mRNAs expression was slightly but significantly increased, when it was compared to the levels recorded for normal prostate. In cancer samples, 5alpha-R1 mRNA expression was higher than in normal and hyperplastic prostate but the level of 5alpha-R2 mRNA was not statistically different from that observed in the different zones of normal prostate. In liver, 5alpha-R2 mRNA level was similar to that measured in BPH but 5alpha-R1 mRNA expression was ten times higher. The increase observed in 5alpha-R isoenzymes expression in BPH tissue could play an important role in the pathogenesis and/or maintenance of the disease.  相似文献

We examined laryngeal resistance (Rla) in six normal subjects in control and four kinds of loaded breathing: hypercapnia, chest strapping, added external resistance, and inhaled methacholine. Rla was measured with a low-frequency sound methed (Sekizawa et al., J. Appl. Physiol. 55: 591-597, 1983). In control and the four kinds of loaded breathing, changes in Rla were tightly coupled with ventilation and Rla decreased during inspiration and increased during expiration. Hypercapnia and chest strapping significantly decreased Rla in both inspiration and expiration in all subjects. Added external resistance decreased inspiratory Rla in all subjects, but decreased expiratory Rla in three subjects, did not change it in two subjects, and increased it in one subject. Inhaled methacholine increased Rla in both inspiration and expiration in all subjects. The present study suggests that although laryngeal movement is tightly coupled with ventilation, laryngeal aperture may be determined by the complex competition of dilating and constricting mechanisms associated with the activity of the respiratory center and neural reflexes from the airway.  相似文献

Summary Epidermal cells of normal fin web and of the epidermal papilloma of the flathead sole, Hippoglossoides elassodon, were compared by light and electron microscopy. The predominant cells of normal epidermis are squamous and are characterized by complex microvillous interdigitations between adjacent cells, numerous desmosome-like structures, terminal bars, prominent cytoplasmic filaments, and blunt, broad-based microvilli on the outer (free) surfaces of the superficial cells. The less numerous mucous cells of normal epidermis possess abundant ergastoplasm and mucus-filled vacuoles, but lack desmosome-like structures and cytoplasmic filaments.In contrast, in the epidermal papilloma, mucous cells are absent; and the tumor cells are without microvilli, desmosome-like structures, cytoplasmic filaments, and conspicuous ergastoplasm. The papilloma cells are in addition characterized by certain features not seen in normal epidermis, including prominent nuclear pores, very large nucleoli, nuclear dense bodies, vesicular mitochondria with few cristae and dense internal granules, and three types of distinctive cytoplasmic particles of possible viral nature.This investigation was supported by Public Health Service research grant CA 08158 from the National Cancer Institute, and American Cancer Society Institutional Grant No. IN-53 E.We are indebted to Miss Mary Bens for her indispensable technical assistance.  相似文献

在研究胰岛素(Ins)、地塞米松(Dex)和甲基异丁基黄嘌呤(Mix)对脂肪细胞分化过程中PAI-1基因表达的影响基础上,为进一步探讨Ins、Dex调控PAI-1基因转录表达的调控机制,应用DNA重组技术,构建含萤光素酶(luciferase)报告基因和PAI-1启动子不同长度片段的嵌合质粒,转染3T3-L1前脂肪细胞并测定报告基因荧光素酶的活性.结果表明,小鼠PAI-1基因起动子-690至-850碱基序列之间有一个Dex的正调控元件.用计算机软件进行分析发现:Dex顺式元件位于PAI-1启动子的-750至-770碱基序列.其组成为:5′ GGTAACCTCTGTTCTCAT 3′.同时还发现在PAI-1启动子的-720至-740碱基序列中,存在一个C/EBPs的结合元件5′CCAAT3′并用凝胶电泳迁移实验对这些元件进行了鉴定.表明Dex正是通过激活转录因子(糖皮质激素受体,GR)和C/EBPα一起与各自的顺式元件结合来促进PAI-1基因的表达.  相似文献

Human proliferative and secretory endometrium from normal women and from menorrhagic patients was maintained in culture for up to a 24 h in the presence of [3H]-arachidonic acid (3H-AA). This prostaglandin (PG) precursor was incorporated into endometrial neutral lipids and phospholipids in a time-dependent manner. Uptake of 3H-AA into phospholipids was significantly higher in normal secretory endometrium than in normal proliferative endometrium. However, this increased uptake of 3H-AA into phospholipids between the 2 phases of the cycle did not occur in menorrhagic endometrium. In contrast, uptake of 3H-AA into neutral lipids (especially triglyceride) was approximately 2-fold higher in menorrhagic endometrium compared to normal endometrium at both stages of the cycle, particularly during the proliferative phase. Abnormalities apparently exist in menorrhagic endometrium in the uptake processes which control arachidonic acid (AA) turnover. These abnormalities may be responsible, in part for abnormal PG production by menorrhagic endometrium.  相似文献

The activities of alkaline phosphatase and phosphoamino acid phosphatases were measured in normal and cancerous regions of the human larynx. For each larynx, alkaline phosphatase and phosphotyrosine phosphatase activities were higher in the tumor than in the corresponding normal tissue. Phosphothreonine and phosphoserine phosphatase activities were relatively low and there were no consistent trends. The increased alkaline phosphatase activity in the tumors supports histological observations that ossification of cartilage seems to occur at the site of invasion; the phosphatase acting on phosphotyrosine could serve as a regulator of cell differentiation during tumorigenesis.  相似文献

Androgen-dependent induction of mk1, true tissue kallikrein, in submandibular gland was studied in C3H/HeN and ICR mice and their F1 progeny. By injection of 5alpha-dihydrotestosterone (DHT), total esteroproteinase activities of female mice were increased to the level of male mice in both C3H/HeN and ICR strains. The mk1 content measured by the radioimmunoassay with anti-mk1 antiserum was decreased in ICR mice, but markedly increased in C3H/HeN mice after DHT injection. We examined the kallikrein isozyme pattern in SMG of two strains using isoelectric focusing. Female ICR mice expressed mainly mk1, mk13 and mk22, and slight mk9. Female C3H/HeN mice expressed mk1, mk9 and pI 6.6-kallikrein. Injection of DHT did not induce any additional kallikrein isozyme in C3H/HeN mice. Furthermore, we made an F1(C3H/HeN) mouse expressing mk13 and mk22 by mating (female C3H/HeN x male ICR). F1(C3H/HeN); these mice showed an androgen response similar to that observed in the ICR mice: mk1 induction in F1(C3H/HeN) mice was decreased by injection of DHT. We suggest the possibility that androgen-dependent mk1 biosynthesis might interact with the expression of other kallikrein isozymes.  相似文献
