The uv absorption of Na-hyaluronate (NaHA) films and the refractive indices, water content, and swelling of LiHA films have been measured as a function of relative humidity. Three peaks are observed in the uv absorption of NaHA (at about 250, 310, and 330 nm) for water content above 10 water molecules per disaccharide. The absorptivity of the 250, 310, and 330 nm peaks increase as the water content increases, indicating a change in the electronic properties of the HA molecule. The refractive indices, water content, and swelling of LiHA films are used to determine the optical polarizability via the Lorentz–Lorenz relation. The polarizability of LiHA is found to have a similar dependence on water content as NaHA, though the changes observed are larger in magnitude. © 1994 John Wiley & Sons, Inc.  相似文献

目的:用水溶性的碳化二亚胺(EDC)作为交联剂对透明质酸(HA)进行化学修饰,研究交联产物(HA-EDC)的物化性质和微观结构。方法:制备HA与EDC的交联产物,对该交联产物进行流变性和热学性能的研究,并对其进行红外光谱、拉曼光谱以及核磁共振的微观结构研究。结果:反应时间和交联剂添加量的不同可以改变交联反应的交联度。不同摩尔比的EDC和HA的交联产物有不同的溶胀性。光谱分析得到交联产物的N-酰脲的结构。结论:可以用碳化二亚胺来对透明质酸进行化学修饰,得到具有很强吸水性并且吸水性可以控制的交联产物,交联反应过程中,产物的结构从不稳定的O-异酰脲转变为稳定的N-酰脲。  相似文献

Sodium hyaluronate (NaHA) is an ultrahigh molecular weight polysaccharide that is found in body tissues, synovial fluid, the vitreous humor, and the umbilical cord, and the size characterization of NaHA is important in pharmaceutical applications. On-line field-flow fractionation/multiangle light scattering/differential refractive index (FlFFF/MALS/DRI) has been applied for the study of degradation efficiency of sodium hyaluronate (NaHA). A NaHA raw sample was degraded by different chemical or physical methods and the degraded NaHA samples were separated using field-programming FlFFF, in which separation is achieved by differences in diffusion coefficients or hydrodynamic diameters. Separation was followed by serial detection using MALS and DRI. Molecular weight distribution (MWD) and information relating to the radius of gyration of the NaHA samples were examined for the raw and degraded NaHA samples. Samples studied include: two different products of ultrasonic degradation, two products of alkaline degradation, and four different products of enzymatic degradation. While alkaline degradation showed a moderate degradation compared to ultrasonic and enzymatic methods in reducing average MW, the latter two degradation methods showed significant changes in average molecular weight and in conformation of NaHA.  相似文献

The extensional viscosity and the steady shear viscosity of sodium type hyaluronan (NaHA) in water with sodium chloride and/or sucrose and in DMSO solvent were measured. The extensional viscosities for HA in aqueous solution (0.05, 0.1, 0.3 w/v%) were constant at lower extensional rates, and then became strain thinning above a critical extensional rate. However, on adding sodium chloride, the extensional viscosity decreased and became strain thickening at higher extensional rates. Sodium ions shield the electrostatic repulsion between carboxyl residues of HA molecules and constrict the coil dimensions. The strain thickening of HA solution in the presence of sodium chloride at higher extension rates is due to the coil stretching. The addition of sucrose increased the extensional viscosity and shifted the critical extensional rate to lower strain rates. With increasing strain (shear) rates, extensional (shear) viscosities for HA aqueous solutions remained constant up to a critical extension (shear) rate; but they showed no plateau and decreased linearly in DMSO. It is clear that molecular interaction of HA in DMSO is stronger than that in aqueous solution. This should be attributed to the different conformations of HA in DMSO and in aqueous solutions.  相似文献

A new fluorometric method for the determination of carbodiimides with trans-aconitic acid as a reagent has been developed. With the present method, as little as 50 pmol of carbodiimides can be determined. The relative standard deviations obtained with 25 nmol of dicyclohexylcarbodiimide (DCC), diisopropylcarbodiimide (DIC), and 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide HCl (EDC) were 1.35, 1.41, and 2.72% (n = 10), respectively.  相似文献

Effects of sodium chloride (NaCl), guanidine hydrochloride (GuHCl), or sucrose on the viscoelasticity of sodium hyaluronate (NaHA) solutions were studied. NaCl and GuHCl decreased both storage and loss moduli, while sucrose increased both moduli. The critical concentration C* was determined as an inflection point in the plot of zero shear specific viscosity vs concentration for NaHA solutions with and without NaCl, GuHCl, or sucrose. It is suggested that sodium ions or guanidinium ions shield the electrostatic repulsion of NaHA molecules, hence reduce the coil dimension, and C* shifted to higher concentrations. However, sucrose enhances the entanglement coupling between NaHA molecules and retards the disentanglement of molecular chains or promotes to create hydrogen bonds, and then C* for NaHA solutions with sucrose shifts to lower concentrations. This is in agreement with the results of light scattering measurements in the presence of 0.2M NaCl. Both the radius of gyration and hydrodynamic radius of NaHA were reduced in dilute solutions by the addition of sucrose, and added sucrose enhances the interaction between NaHA monomer units. In the case of concentrated NaHA solution, such interactions result to increase the storage and loss moduli because of the enhancement of temporary network formation. © 1999 John Wiley & Sons, Inc. The aim of this work was to investigate the rheological properties of different saline solutions of sodium hyaluronate (NaHA) with special interest for medical applications. The experimental results were compared with literature data for commercial ophthalmic viscosurgical devices (OVDs) used in cataract surgery. We offer some tools to tailor the rheological behavior of OVDs for different purposes. We have investigated to which extent surgical requirements can be fulfilled by adjusting either the molecular weight of NaHA or its concentration, parameters that are in some respects equivalent but not in others. Furthermore, we demonstrate that moduli and complex viscosities of NaHA saline solutions are adequately falling on master curves, using either empirical or calculated shift factors, the latest ones being based on a modified Rouse model.  相似文献

The focus of this work is on a comparative rheological characterization of sodium hyaluronate (NaHA) samples from fermentative production (NaHA-1, Mw = 1.7 × 10? g/mol) and from rooster comb (NaHA-2, Mw = 4.6 × 10? g/mol) with synovial fluid (SF) taken post mortem from different patients above 60 years. Steady state shear flow and uniaxial extension experiments were carried out for different concentrations of NaHA in 0.01 M phosphate-buffered saline (PBS) and on several SF samples.The synovial fluid of older patients was found to have a viscoelasticity comparable to that of younger patients investigated in earlier studies. Comparison of steady state shear experiments revealed a comparable progression for the viscosity curves of NaHA-2 and SF. This behavior was also observed in extensional flow, where comparable results for NaHA-2 and different SF samples were obtained.  相似文献

A plasma process for the surface modification of HA powders has been developed. Acrylic acid and acrylic acid/octadiene plasma deposited films onto HA particles have demonstrated to interact with SBF allowing the calcium dissolution-precipitation mechanism. Therefore, a nanostructured composite between HA and a self-assembling peptide scaffold (RAD16-I) has been developed. The differentiation of mESC in this scaffold has been studied, in order to test the osteogenic capacity of the new composite material. We have observed that the mESC can be induced to produce Ca salts (mineralization) in a 3D-microenvironment and moreover, this activity can be enhanced by the presence of HA particles into the nanofiber scaffold.  相似文献

In near weak acid to neutral medium, ethyl violet (EV) can react rapidly with hyaluronic acid (HA) to form a complex, which results in a significant enhancement of resonance Rayleigh scattering (RRS) and an appearance of a new spectrum, and the scattering wavelengths appear at 231, 274, 326, 498 and 640 nm. The maximum scattering wavelength appears at 326 nm. The RRS intensity is directly proportional to the concentration of HA in the range of 0.4-48.0 microg mL(-1). A new method for the determination of trace amounts of HA based on the RRS method has been developed. The method exhibits high sensitivity, and the detection limit for HA is 9.6 x 10(-2) microg mL(-1). This method was applied for determining HA in eyedrops and in sodium hyaluronate injection samples with satisfactory results. Furthermore, the enhancement reasons of RRS and the relationship between RRS spectral characteristics of the HA-EV complex and its absorption spectrum have been discussed.  相似文献

Membrane vesicles isolated from wild-type and dicyclohexylcarbodiimide-resistant strains of Escherichia coli exhibit identical respiration-dependent transport activities, and in both cases, this activity is abolished by extraction of the vesicles with 1.0 M guanidine-HCl. Transport activity of extracted wild-type vesicles is completely restored by exposing the vesicles to lipophilic or water-soluble carbodiimides, while transport activity of the mutant vesicles is not restored by exposure to lipophilic carbodiimides. Strikingly, however, complete reactivation of transport in mutant vesicles is observed with water-soluble carbodiimides. Similarly, the Ca2+, Mg2+-stimulated ATPase activity of wild-type vesicles is inhibited by both classes of carbodiimides, while the ATPase activity of mutant vesicles is inhibited by water-soluble carbodiimides, but resistant to inhibition by lipophilic carbodiimides. The carbodiimide-reactive component of the membraneous Ca2+, Mg2+-stimulated ATPase complex in wildtype vesicles is readily labeled with N,N'-dicyclohexyl[14C]-carbodiimide, while the analogous component in mutant vesicles is not reactive. Alternatively, when vesicles are treated with 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide [14C]methiodide, a water-soluble carbodiimide, the carbodiimide-reactive component is labeled to a similar degree in both preparations. The results suggest that the altered carbodiimide-reactive proteolipid in the dicyclohexylcarbodiimide-resistant mutant is specifically defective in its ability to react with lipophilic carbodiimides. In addition, these and other findings indicate that the increase in proton permeability observed on extraction of isolated membrane vesicles with chaotropic agents is due exclusively to an effect on the carbodiimide-reactive component of the Ca2+, Mg2+-stimulated ATPase complex.  相似文献

Water deficit is the major yield‐limiting factor for sugarcane crop production that can be enhanced by inoculating with plant growth promoting bacteria (PGPB) combined with humic substances. The aim of this work was to examine changes to the metabolic profile and antioxidant enzyme activity of sugarcane treated with PGPB and humic acid (HA) after drought and then rehydration. The drought was imposed by withholding irrigation for 21 days thereby measuring enzyme activity, metabolic profile and photosynthetic rate 1 week after rehydratation. Growth of plants treated with HA, PGPB and with both treatments combined (PGPB + HA) was higher than control plants. The antioxidant enzymes superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) activities remained higher after rehydration only in plants treated with HA. Plants treated with HA and PGPB + HA exhibited increased transpiration, stomatal conductance and net photosynthesis than plants treated with PGPB. The PGPB‐treated plants exhibited drought resistance that resembled ‘delayed stress onset’, which is a new term for preserving water in the plants tissues. Water preservation in plants treated with PGPB was corroborated by higher relative water content (RWC) than control plants at the end of the drought period. Plants treated with HA + PGPB exhibited the highest water potential after rehydration and high RWC. Osmotic adjustment in the other treatments (control, HA and PGPB) was indicated by a new pattern of metabolic response after rehydration, including generally enhanced carbohydrates and proteins and specific changes induced by HA‐enhancing aromatic compounds, whereas PGPB exhibited enhanced fatty acids and other aliphatic H species. Humic acids assist with drought stress recovery by inducing antioxidant enzyme activity whereas PGPB induced preservation of leaf water potential and RWC by closing stomata efficiently, resulting in plant water preservation.  相似文献

Influenza A viruses possess two virion surface proteins, hemagglutinin (HA) and neuraminidase (NA). The HA binds to sialyloligosaccharide viral receptors, while the NA removes sialic acids from the host cell and viral sialyloligosaccarides. Alterations of the HA occur during adaptation of influenza viruses to new host species, as in the 1957 and 1968 influenza pandemics. To gain a better understanding of the contributions of the HA and possibly the NA to this process, we generated cell lines expressing reduced levels of the influenza virus receptor determinant, sialic acid, by selecting Madin-Darby canine kidney cells resistant to a lectin specific for sialic acid linked to galactose by alpha(2-3) or alpha(2-6) linkages. One of these cell lines had less than 1/10 as much N-acetylneuraminic acid as its parent cell line. When serially passaged in this cell line, human H3N2 viruses lost sialidase activity due to a large internal deletion in the NA gene, without alteration of the HA gene. These findings indicate that NA mutations can contribute to the adaptation of influenza A virus to new host environments and hence may play a role in the transmission of virus across species.  相似文献

Engineering of hyaluronic acid (HA) biosynthetic pathway in recombinant Escherichia coli as production host is reported in this work. A hyaluronic acid synthase (HAS) gene, sphasA, from Sreptococcus pyogenes with the start codon gtg to atg mutant, was expressed in recombinant E. coli with or without the genes ugd, galF and glmU, which are analogs of hasB, hasC and hasD from Streptococcus, respectively, encoding UDP-glucose 6-dehygrogenase, Glucose-1-P uridyltransferase, and N-acetyl glucosamine uridyltransferase enzymes in the HA biosynthetic pathway. The single, double and triple organized artificial operons of sphasA, ugd, galF and glmU were designed and constructed using the inducible plasmid backbone of pMBAD. Only the triple expression recombinant, Top10/pMBAD-spABC, generated a relatively high titer of HA (approximately 48 mg/l at 48 h), indicating that both of the enzymes encoded by ugd and galF are essential for HA biosynthesis. A new gene of ssehasA with identical protein sequence of seHAS from Streptococcus equisimilis, was artificially synthesized after substituting all of the rare codons in the natural sehasA. The HA titer at 24 h flask culture increased to approximately 190 mg/l in sseAB and 160 mg/l in sseABC, respectively. Sorbitol could be used as another carbon source for HA accumulation, and the metabolic pathway for HA synthesis in a recombinant E. coli was presented. The concentration of Mg(2+) cofactor of HA synthase was optimized and a cell growth inhibition phenomenon was observed during HA accumulation. Molecular weight (MW) measurements revealed that the mean MW of HA produced from the recombinant E. coli under different conditions ranges from approximately 3.5x10(5) to 1.9x10(6)Da, indicating that the recombinant E. coli can be used as a potential host candidate for industrial production of HA.  相似文献

2009年全球性爆发的H1N1病毒已经导致213个国家和地区受到感染, 有16 226人死亡。病毒与宿主细胞表面受体的结合是病毒感染不可缺少的第一步, 从而导致病毒膜与宿主细胞膜的融合。血凝素(Hemagglutinin, HA)就是介导这种受体结合与膜融合的病毒蛋白, 受体结合位点(Receptor binding sites, RBSs)位于HA蛋白三聚体中每个单体的球形头部, 主要由190位螺旋(190~198aa)、130位环(135~138aa)和220位环(221~228)3个二级结构域组成。文章收集了1918~2009年间1 221株H1N1病毒株的HA1序列(长度为327个氨基酸残基), 通过序列比对、各位点氨基酸残基的熵值以及3D结构模拟等生物信息学研究。结果显示不同宿主的不同病毒RBSs具有不同的熵值, 而且不同宿主的病毒HA1其RBSs具有不同的优势序列。3D结构模拟也显示了H1N1不同HA1之间在190位螺旋构象上的细微差异。该研究揭示了不同HA1上RBSs的一些新的特征, 为进一步探讨病毒感染的机理提供了新的信息  相似文献

Previous study showed that nitrogen content in lignite humic acids (HA) increased significantly during lignite biodegradation. In this paper we evaluated the factors responsible for the increased level of N in HA and the formation of new nitrogen compound following microbial degradation. When the ammonium sulfate concentration in lignite medium was < 0.5%, the N-content in HA was higher than that in the crude lignite humic acid (cHA); when the ammonium sulfate concentration was ≥ 0.5%, both the biodegraded humic acid (bHA) N-content and the content of bHA in lignite increased significantly, but at 2.0% no increase was observed. This indicated that HA incorporated N existing in the lignite medium, and more HA can incorporate more N with the increase of bHA amount in lignite during microbial degradation. CP/MAS 15N NMR analysis showed that the N incorporated into HA during biotransformation was in the form of free or ionized NH2-groups in amino acids and sugars, as well as NH4 +. We propose nitrogen can be incorporated into HA biotically and abiotically. The high N content bHA has a potential application in agriculture since N is essential for plant growth.  相似文献

A new approach to create chimeric genes by directed exchange of oligonucleotide fragments was developed. By oligonucleotide-directed mutagenesis a few deletion mutants of the influenza virus hemagglutinin (HA) gene were obtained. These variants of HA gene contain unique restriction sites in DNA regions coding for the A and B epitopes of the HA molecule. The obtained special vectors may be used for cloning DNA fragments coding for new amino acid sequences in internal sites of the HA gene.  相似文献

Hyaluronic acid (HA) hydrogel beads were prepared by photopolymerization of methacrylated HA and N-vinylpyrrolidone using alginate as a temporal spherical mold. Various fabrication conditions for preparing the hydrogel beads, such as the concentration of methacrylated HA and UV irradiation time, were optimized to control swelling properties and enzymatic degradability. A new concept for cell encapsulation is proposed in this paper. Viable cells were directly injected into the hydrogel beads using a microinjection technique. When bovine articular chondrocytes were injected into HA hydrogel beads and cultivated for 1 week, the cells could proliferate well within the HA beads. HA hydrogel beads could be potentially used as injectable cell delivery vehicles for regenerating tissue defects.  相似文献

The complete amino acid sequences of the hemagglutinin (HA) glycoprotein of three equine-2 influenza viruses from tropical Africa are presented in comparison with that of a well characterized European equine-2 virus (Suffolk/89) and a consensus sequence from the database. The sequences of the tropical African viruses were deduced from the complete nucleotide sequences of their HA genes reported earlier. Mutational changes in the nucleotide sequences resulted in amino acid changes in the HA which led to the introduction of a new asparagine-linked (N-linked) glycosylation site in two viruses. This new glycosylation site enhanced the infectivity of these viruses as investigated by plaque assay, virus titration in embryonated chicken eggs and tunicamycin treatment. The role of N-linked glycosylation of influenza virus HA glycoprotein in virus infectivity, antigenicity and immunogenicity is discussed in the light of the results of our previous and present investigations.  相似文献

Glutamate and gamma-aminobutyric acid (GABA) are major excitatory and inhibitory retinal neurotransmitters. The balance between these signals is a key principle of organization at retinal level. Although glutamate-induced excitotoxicity could mediate retinal ganglion cell death in glaucoma, the GABAergic system was not previously examined in this disease. The aim of this work was to study the retinal GABAergic activity in eyes with ocular hypertension induced by hyaluronic acid (HA). For this purpose, weekly injections of HA were performed unilaterally in the rat anterior chamber, whereas the contralateral eye was injected with saline solution. At 3 weeks of treatment with HA, GABA turnover rate, glutamic acid decarboxylase activity, and both glutamate- and high K(+)-induced GABA release significantly decreased, whereas GABA uptake increased in HA-treated eyes. The binding of t-butylbicyclophosphorothionate (TBPS) to GABA(A)/benzodiazepine Cl(-) channels significantly increased in eyes injected with HA as compared with vehicle-injected eyes. Changes in GABA uptake and TBPS binding persisted at 6 weeks of treatment with HA. These results indicate a dysfunction of the retinal GABAergic activity in hypertensive eyes, which could suggest the involvement of GABA in glaucomatous neuropathy.  相似文献
